Abstract:Considering that the current bioproduction of 1,3-propanediol is mainly carried out under strict anaerobic conditions, which requires airtight production equipment and large amount of nitrogen to maintain the oxygen free conditions in the fermentation system, and the production procedure demands quite complex operation technique which leads to high cost, an endure-oxygen Klebsiella pneumoniae strain ZU-05 was used to produce 1,3-propanediol from glycerol under non-strict anaerobic conditions, and the main fermentation parameters were optimized. Under the non-strict anaerobic condition with 20 g/L glycerol, 6 g/L corn syrup, 8 g/L glucose and inoculation volume of 10% at pH 6.7, 37℃, the utilizing rate of glycerol and yield of 1,3-Propanediol were 95 % and 51% within 60 h, respectively. The results are meaningful for simplifying the procedure and increasing the profit of production from glycerol to 1,3-propanediol.
Inactivation of hydrogenase-3 leads to enhancement of 1,3-propanediol and 2,3-butanediol production by Klebsiella pneumoniae

Abstract:Klebsiella pneumoniae can use glucose or glycerol as carbon sources to produce 1,3-propanediol or 2,3-butanediol, respectively. In the metabolism of Klebsiella pneumoniae , hydrogenase-3 is responsible for H 2 production from formic acid, but it is not directly related to the synthesis pathways for 1,3-propanediol and 2,3-butanediol. In the first part of this research, hycEFG , which encodes subunits of the enzyme hydrogenase-3, was knocked out, so K. pneumoniae Δ hycEFG lost the ability to produce H 2 during cultivation using glycerol as a carbon source. As a consequence, the concentration of 1,3-propanediol increased and the substrate (glycerol) conversion ratio reached 0.587 mol/mol. Then, K. pneumoniae Δ ldhA Δ hycEFG was constructed to erase lactic acid synthesis which led to the further increase of 1,3-propanediol concentration. A substrate (glycerol) conversion ratio of 0.628 mol/mol in batch conditions was achieved, which was higher compared to the wild type strain (0.545 mol/mol). Furthermore, since adhE encodes an alcohol dehydrogenase that catalyzes ethanol production from acetaldehyde, K. pneumoniae Δ ldhA Δ adhE Δ hycEFG was constructed to prevent ethanol production. Contrary to expectations, this did not lead to a further increase, but to a decrease in 1,3-propanediol production. In the second part of this research, glucose was used as the carbon source to produce 2,3-butanediol. Knocking out hycEFG had distinct positive effect on 2,3-butanediol production. Especially in K. pneumoniae ΔldhAΔadhEΔhycEFG , a substrate (glucose) conversion ratio of 0.730 mol/mol was reached, which is higher compared to wild type strain (0.504 mol/mol). This work suggests that the inactivation of hydrogenase-3 may have a global effect on the metabolic regulation of K. pneumoniae , leading to the improvement of the production of two industrially important bulk chemicals, 1,3-propanediol and 2,3-butanediol.

Decrease of 3-hydroxypropionaldehyde accumulation in 1,3-propanediol production by over-expressing dha T gene in Klebsiella pneumoniae TUAC01

Abstract:Glycerol can be biologically converted to 1,3-propanediol, a key raw material required for the synthesis of polytrimethylene terephthalate and other polyester fibers. In 1,3-propanediol synthesis pathway, 3-hydroxypropionaldehyde (3-HPA) was an inhibitory intermediary metabolite. The accumulation of 3-HPA in broth would cause an irreversible cessation of the fermentation process. With the object of reducing 3-HPA level in the fermentation broth, dha T gene which encodes 1,3-propanediol oxidoreductase (PDOR) was cloned and over expressed in 1,3-propanediol producing bacterium Klebsiella pneumoniae TUAC01. dha T gene was linked downstream of the ptac promoter in an expressing vector pDK6 to form plasmid pDK- dha T. The newly formed pDK- dha T was transformed to K. pneumoniae TUAC01. Under the inducement of IPTG, PDOR was over-expressed when the constructed strain was cultured on an LB medium or a fermentation medium. A 5 L scale-up fermentation experiment was done to test the 3-HPA accumulation in broth, with the initial substrate glycerol 30 g/L; the peak levels of 3-HPA in broth were 7.55 and 1.49 mmol/L for control host strain and the constructed strain, respectively. In 50 g/L initial glycerol experiment, the peak level of 3-HPA in broth was 12.57 and 2.02 mmol/l for the control host strain and the constructed strain, respectively. Thus the fermentation cessation caused by the toxicity of 3-HPA was alleviated in the constructed strain.
Regulation of 3-Hydroxypropionaldehyde Accumulation in Klebsiella Pneumoniae by Overexpression of Dhat and Dhad Genes

Abstract:3-Hydroxypropionaldehyde (3-HPA), an important intermediary metabolite of 1,3-propanediol (PDO) production, would be toxic to the cell growth and led to the abnormal cessation of the fermentation process. In this study, the dhaD gene encoding glycerol dehydrogenase (GDH) and dhaT gene encoding 1,3-propanediol oxidoreductase (PDOR) were overexpressed in Klebsiella pneumoniae ACCC 10082 to decrease the 3-HPA accumulation and increase the coenzyme NADH supply. By the construction of pTD plasmid, GDH and PDOR were both overexpressed and their enzyme activities were increased by 2.6- and 3.2-fold, respectively. The enzyme activity ratio of PDOR/GDHt (glycerol dehydratase) also was increased. On the other hand, NADH production was enhanced and the ratio of NADH/NAD+ exceeded 1 after the inducement of IPTG for the constructed strain. The two factors enhanced the transformation of 3-HPA to PDO. In the batch and fed-batch fermentation by the constructed strain, the peak of 3-HPA accumulation reduced by 52.2% and 33.3%, respectively, compared with the control. The PDO concentration and yield reached 59.2g/L and 0.48mol/mol, respectively. Furthermore, the fed-batch fermentation process appeared easier to be regulated. This work is considered helpful for the further understanding on the PDO metabolic mechanism of K. pneumoniae and also useful for the PDO fermentation in a large-scale bioreactor.
Construction of polyhydroxybutyrate pathway in Klebsiella pneumoniae]

Abstract:1,3-propanediol production with the byproduct of biodiesel production is important to increase the economic benefit of biodiesel industry. Accumulation of 3-hydroxypropionaldehyde is one of the key problems in the 1,3-propanediol fermentation process, leading to the cell death and the fermentation abnormal ceasing. Different from the traditional way of reducing the accumulation of the 3-hydroxypropionaldehyde, we introduced the polyhydroxybutyrate pathway into the Klebsiella pneumoniae for the first time to enhance the tolerance of K. pneumoniae to 3-hydroxypropionaldehyde, at the same time, to improve the 1,3-propanediol production. Plasmid pDK containing phbC, phbA, phbB gene was constructed and transformed into K. pneumoniae successfully. PHB was detected in the engineered K. pneumoniae after IPTG induction and its content enhanced with the IPTG concentration increasing. The optimized IPTG concentration was 0.5 mmol/L. The constructed K. pneumoniae could produce 1,3-propanediol normally, at the same time accumulate polyhydroxybutyrate. With the constructed strain, the fermentation proceeds normally with the initial glucose was 70 g/L which the wild type strain stopped growing and the fermentation was ceasing; 1,3-propanediol concentration and yield reached 31.3 g/L and 43.9% at 72 h. Our work is helpful for the deep understanding of 1,3-propanediol metabolic mechanism of Klebsiella pneumoniae, and also provides a new way for strain optimization of Klebsiella pneumoniae.
Effects of Aldehyde Dehydrogenase Gene Knockout on 1,3-Propanediol Production by Klebsiella Pneumoniae

Abstract:Glycerol can be transformed to 1,3-propanediol (1,3-PD) by Klebsiella pneumoniae with accompanying ethanol accumulation.Aldehyde dehydrogenase (ALDH) is one of the key enzymes for biosynthesis of ethanol, which competes for reducing equivalents NADH with 1,3-propanediol dehydrogenase which catalyzes biosynthesis of 1,3-PD.Based on metabolic analysis, one reasonable method to improve 1,3-PD yield is to inhibit the catalysis of ALDH so that ethanol production could be restrained. In this paper, a homologous recombination vector pUCAT was constructed, in which the ALDH gene of K.pneumoniae was disrupted by inserting tetracycline resistance gene (Tcr).The amplified DNA fragment of 5′ALDH-Tcr-3′ALDH from pUCAT was used to transform K.pneumoniae M5aL and the ALDH gene knockout recombinants were obtained.Comparing with those of the wild type K.pneumoniae M5aL, the ALDH activity of the recombinants were undetected, cell growth was inhibited obviously, ethanol yields were decreased by 43%—53%, 1,3-PD yields and molar conversions from glycerol to 1,3-PD were increased by 27%—42% and 19%—24% respectively.

Abstract:Production of 1,3-propanediol (1,3-PD) from glycerol by Klebsiella pneumoniae is restrained by ethanol formation. The first step in the formation of ethanol from acetyl-CoA is catalyzed by aldehyde dehydrogenase (ALDH), an enzyme that competes with 1,3-PD oxidoreductase for the cofactor NADH. This study aimed to improve the production of 1,3-PD by engineering the ethanol formation pathway. An inactivation mutation of the aldA gene encoding ALDH in K. pneumoniae YMU2 was generated by insertion of a tetracycline resistance marker. Inactivation of ALDH resulted in a nearly abolished ethanol formation but a significantly improved 1,3-PD production. Metabolic flux analysis revealed that a pronounced redistribution of intracellular metabolic flux occurred. The final titer, the productivity of 1,3-PD and the yield of 1,3-PD relative to glycerol of the mutant strain reached 927.6mmolL−1, 14.05mmolL−1h−1 and 0.699molmol−1, respectively, which were much higher than those of the parent strain. In addition, the specific 1,3-PD-producing capability (1,3-PD produced per gram of cells) of the mutant strain was 2-fold that of the parent strain due to a lower growth yield of the mutant. By increasing NADH availability, this study demonstrates an important metabolic engineering approach to improve the efficiency of oxidoreduction-coupled bioprocesses.

Abstract:1,3-Propanediol (1,3-PD) production is favored by high amount of glycerol which also contributes to the lethal 3-hydroxypropionaldehyde (3-HPA) accumulation. The influences of three culture conditions including initial glycerol concentration, stirring rate, and oxidoreduction potential (ORP) regulation on 3-HPA accumulation and 1,3-PD production were investigated. 3-HPA detoxification was available at initial glycerol concentration, aeration rate and stirring rate of up to 40gl−1, 0.5vvm air flow and 250rpm, respectively. Alternatively, the stirring rate of 300rpm was sufficient to fulfill the fermentation with initial 50gglyceroll−1. In addition, 3-HPA accumulation could be avoided at the initial 50gglyceroll−1 under the regulated reducing conditions (−200 and −400mV). However, ORP regulation was not convenient and feasible for the industrial production in that more carbon was distributed to weak organic acids. Consequently, two-stage fed-batch strategy was presented to achieve 3-HPA detoxification and maintain the expected high productivity of 1,3-PD, in which the batch culture stage was conducted at initial glycerol concentration and stirring rate of 40gl−1 and 250rpm, respectively, and the feeding culture stage was performed at stirring rate of 300rpm. 74.07g 1,3-PDl−1 was produced in 24h with the yield and productivity of 0.62molmol−1 and 3.08gl−1h−1.

Abstract:Abstract3-Hydroxypropionic acid (3-HP) is a commercially valuable platform compound. Klebsiella pneumoniae has been concerned as an appropriate host for 3-HP production because of its robust capacity to metabolize glycerol. Glycerol conversion to 3-HP in K. pneumoniae comprises two successive reactions: glycerol dehydratase catalyzes glycerol to 3-hydroxypropionaldehyde (3-HPA); aldehyde dehydrogenase catalyzes 3-HPA to 3-HP. Previous studies focusing on inducible expression of aldehyde dehydrogenase have shown defects of high cost of inducer and low catalytic activity due to inclusion body. Here we show a different strategy that a native promoter in the host K. pneumoniae was used to drive the heterologous expression of aldehyde dehydrogenase gene ald4 from Saccharomyces cerevisiae. The 3-HP yield of the recombinant reached a peak of 4.23 g/L at log phase, but it decreased during later period of fermentation. Except the validation of high activity of ald4, particularly, the 3-HP formation was uncovered to be closely coupled with cell division, and the lacking of NAD and ATP at latter fermentation phase became the bottleneck for cell growth and 3-HP accumulation. Furthermore, 3-HP is postulated to be converted to 3-HPA via feedback inhibition or other metabolite via unknown mechanism. Since glycerol dissimilation is a common mechanism in a variety of bacteria, the expression strategy using native promoter and implications may provide significant insight into the metabolic engineering for 3-HP production.

Rational Proteomic Analysis of a New Domesticated Klebsiella pneumoniae x546 Producing 1,3-Propanediol

Abstract:In order to improve the capability of Klebsiella pneumoniae to produce an important chemical raw material, 1,3-propanediol (1,3-PDO), a new type of K. pneumoniae x546 was obtained by glycerol acclimation and subsequently was used to produce 1,3-PDO. Under the control of pH value using Na + pH neutralizer, the 1,3-PDO yield of K. pneumoniae x546 in a 7.5-L fermenter was 69.35 g/L, which was 1.5-fold higher than the original strain (45.91 g/L). After the addition of betaine, the yield of 1,3-PDO reached up to 74.44 g/L at 24 h, which was 40% shorter than the original fermentation time of 40 h. To study the potential mechanism of the production improvement of 1,3-PDO, the Tandem Mass Tags (TMT) technology was applied to investigate the production of 1,3-PDO in K. pneumoniae . Compared with the control group, 170 up-regulated proteins and 291 down-regulated proteins were identified. Through Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analysis, it was found that some proteins [such as homoserine kinase (ThrB), phosphoribosylglycinamide formyltransferase (PurT), phosphoribosylaminoimidazolesuccinocarboxamide synthase (PurC), etc.] were involved in the fermentation process, whereas some other proteins (such as ProX, ProW, ProV, etc.) played a significant role after the addition of betaine. Moreover, combined with the metabolic network of K. pneumoniae during 1,3-PDO, the proteins in the biosynthesis of 1,3-PDO [such as DhaD, DhaK, lactate dehydrogenase (LDH), BudC, etc.] were analyzed. The process of 1,3-PDO production in K. pneumoniae was explained from the perspective of proteome for the first time, which provided a theoretical basis for genetic engineering modification to improve the yield of 1,3-PDO. Because of the use of Na + pH neutralizer in the fermentation, the subsequent environmental pollution treatment cost was greatly reduced, showing high potential for industry application in the future.

Gene Arrangements in Expression Vector Affect 3-Hydroxypropionic Acid Production in Klebsiella pneumoniae

Abstract:Biosynthesis of 3-hydroxypropionic acid (3-HP) typically involves two sequential reactions catalyzed by glycerol dehydratase (DhaB) and aldehyde dehydrogenase (AldH). Although plasmid-dependent over-expression of the two enzymes is common, systematic investigation of gene arrangement in vector has not been reported. Here we show that gene arrangements have a noticeable influence on 3-HP production. Using Klebsiella pneumoniae as a host, three AldH-coding genes: ald4 from Saccharomyces cerevisiae, aldh from Escherichia coli, and puuC from host K. pneumoniae, were respectively ligated to dhaB. The recombinant Kp/pET-pk-ald4-dhaB (Kp refers to as K. pneumoniae, pk is a native promoter) produced the highest yield of 3-HP in comparison to both Kp/pET-pk-dhaB-ald4 and Kp/pET-pk-dhaB-pk-ald4, suggesting that the preferential expression of AldH can increase 3-HP production. Additionally, when different AldH-coding genes were respectively ligated downstream of dhaB, the recombinant Kp/pET-pk-dhaB-puuC produced more 3-HP than that by Kp/pET-pk-dhaB-aldh or Kp/pET-pk-dhaB-ald4, implying the intrinsic compatibility of native gene puuC with its host. These findings indicate the applicability of native AldH-coding gene and provide insights into strategies for metabolic engineering of multiple genes.

3-Hydroxypropionaldehyde Guided Glycerol Feeding Strategy in Aerobic 1,3-Propanediol Production by Klebsiella Pneumoniae.

Abstract:3-Hydroxypropionaldehyde (3-HPA) is a toxic intermediary metabolite in the biological route of 1,3-propanediol biosynthesis from glycerol. 3-HPA accumulated in culture medium would arouse an irreversible cessation of the fermentation process. The role of substrate (glycerol) on 3-HPA accumulation in aerobic fermentation was investigated in this paper. 1,3-Propanediol oxidoreductase and glycerol dehydratase, two key enzyme catalyzing reactions of 3-HPA formation and consumption, were sensitive to high concentration of 3-HPA. When the concentration of 3-HPA increased to a higher level in medium (ac 10 mmol/L), the activity of 1,3-propanediol oxidoreductase in cell decreased correspondingly, which led to decrease of the 3-HPA conversion rate, then the 3-HPA concentration increasing was accelerated furthermore. 3-HPA accumulation in culture medium was triggered by this positive feedback mechanism. In the cell exponential growth phase, the reaction catalyzed by 1,3-propanediol oxidoreductase was the rate limiting step in 1,3-propanediol production. The level of 3-HPA in culture medium could be controlled by the substrate (glycerol) concentration, and lower level of glycerol could avoid 3-HPA accumulating to a high, lethal concentration. In fed batch fermentation, under the condition of initial glycerol concentration 30 g/L, and keeping glycerol concentration lower than 7-8 g/L in cell exponential growth phase, 3-HPA accumulation could not be incurred. Based on this result, a glycerol feeding strategy was set up in fed batch fermentation. Under the optimized condition, 50.1 g/L of 1,3-propanediol was produced in 24 h, and 73.1 g/L of final 1,3-propanediol concentration was obtained in 54 h.
Engineering Glucose-to-Glycerol Pathway in Klebsiella pneumoniae and Boosting 3-Hydroxypropionic Acid Production Through CRISPR Interference

Abstract:The recent decline of the international biodiesel industry has led to decreased production and therefore increased the price of glycerol, which is a major by-product of biodiesel but a substrate for production of 3-hydroxypropionic acid (3-HP), that is, glycerol as a feedstock has no advantage over glucose in price. Hence, we engineered glucose to the glycerol pathway and improved 3-HP production by CRISPR interference (CRISPRi). To begin with, we cloned the genes encoding glycerol 3-phosphate dehydrogenase ( gpd1 ) and glycerol 3-phosphatase ( gpp2 ) from Saccharomyces cerevisiae , which jointly catalyze glucose into glycerol. The genes gpd1 and gpp2 were co-expressed in K. pneumoniae with the dCas9 gene integrated in genome, and this recombinant strain produced 2 g/L glycerol in the shake flask. To minimize the glucose consumption by competing pathways including the EMP pathway, glycerol oxidation pathway, and by-products pathways, we developed an CRISPRi system in aforementioned recombinant K . pneumoniae strain to inhibit the expression of the glyceraldehyde-3-phosphate dehydrogenase gene ( gapA ) and 2,3-butanediol production gene ( budA ), resulting in a bi-functional strain harboring both glucose-to-glycerol pathway and CRISPRi system. Reverse transcription and quantitative PCR (RT-qPCR) results showed that this engineered CRISPRi system transcriptionally inhibited gapA and budA by 82% and 24%, respectively. In shake flask cultivation, this bi-functional strain produced 2.8 g/L glycerol using glucose as the carbon source, which was 46.6% increase compared to the strain without the engineered CRISPRi system. Moreover, this bi-functional strain produced 0.78 g/L 3-HP using glucose as the sole carbon source. In fed-batch cultivation, this bi-functional strain produced 1.77 g/L 3-HP. This study provides insights for co-utilization of distinct carbon sources.

Abstract:The glycerol fed-batch fermentation by Klebsiella pneumoniae CGMCC 1.6366 exhibited the sequential synthesis of products, including acetate, 1,3-propanediol (1,3-PD), 2,3-butanediol, ethanol, succinate, and lactate. The dominant flux distribution was shifted from acetate formation to 1,3-PD formation in early-exponential growth phase and then to lactate synthesis in late-exponential growth phase. The underlying physiological mechanism of the above observations has been investigated via the related enzymes, nucleotide, and intermediary metabolites analysis. The carbon flow shift is dictated by the intrinsic physiological state and enzymatic activity regulation. Especially, the internal redox state could serve as a rate-controlling factor for 1,3-PD production. The q(1,3-PD) formation was the combined outcomes of regulations of glycerol dehydratase activity and internal redox balancing. The q(ethanol)/q(acetate) ratios demonstrated the flexible adaptation mechanism of K. pneumoniae preferring ATP generation in early-exponential growth phase. A low PEP to pyruvate ratio corresponded LDH activity increase, leading to lactate accumulation in stationary phase.
Inhibitory Mechanism of 3-Hydroxypropionaldehyde Accumulation in 1,3-Propanediol Synthesis with Klebsiella Pneumoniae

Abstract:3-Hydroxypropionaldehyde accumulation may cause the cessation of 1,3-propanediol sustained production with glycerol by Klebsiella pneumoniae. The impeller tip speed shift from higher to lower speed at glycerol excess or the pulsed glycerol feeding could lead to an abrupt increase of the 3-hydroxypropionaldehyde concentration (up to 10 mmol/l) in 10 min. The intracellular consequence of the 3-hydroxypropionaldehyde accumulation has not yet been elucidated. The rapid accumulation of 3-hydroxypropionaldehyde relying on the impeller tip speed shift was employed to investigate the influences of 3-hydroxypropionaldehyde to the activities of nine key enzymes related to glycerol metabolism, CO(2) and O(2) levels in off-gas, cell growth and 1,3-propanediol synthesis. Compared with that at 1.19 mmol/l 3-hydroxypropionaldehyde in broth, the residual enzymatic activities of the nine key enzymes ranged from 9.44 to 74.68% in the cultures at 7.5 mmol/l 3-hydroxypropionaldehyde in broth. The inhibitions of cell growth and the 1,3-propanediol synthesis were unnoticeable at the low level of 3-hydroxypropionaldehyde. By contrast, the CO(2) and O(2) levels changes in off-gas response to the 3-hydroxypropionaldehyde accumulation were less than 15 min. These results suggest that 3-hydroxypropionaldehyde inhibited the growth and metabolism of K. pneumoniae in a more complicated manner.
Enhancing the capability of <i>Klebsiella pneumoniae</i> to produce 1, 3‐propanediol by overexpression and regulation through CRISPR‐dCas9

Abstract:Klebsiella pneumoniae is a common strain of bacterial fermentation to produce 1, 3-propanediol (1, 3-PDO). In general, the production of 1, 3-PDO by wild-type K. pneumoniae is relatively low. Therefore, a new gene manipulation of K. pneumoniae was developed to improve the production of 1, 3-PDO by overexpressing in the reduction pathway and attenuating the by-products in the oxidation pathway. Firstly, dhaB and/or dhaT were overexpressed in the reduction pathway. Considering the cost of IPTG, the constitutive promoter P32 was selected to express the key gene. By comparing K.P. pET28a-P32-dhaT with the original strain, the production of 1, 3-PDO was increased by 19.7%, from 12.97 to 15.53 g l(-1) (in a 250 ml shaker flask). Secondly, three lldD and budC regulatory sites were selected in the by-product pathway, respectively, using the CRISPR-dCas9 system, and the optimal regulatory sites were selected following the 1, 3-PDO production. As a result, the 1, 3-PDO production by K.P. L1-pRH2521 and K.P. B3-pRH2521 reached up to 19.16 and 18.74 g l(-1), which was increased by 47.7% and 44.5% respectively. Overexpressing dhaT and inhibiting expression of lldD and budC were combined to further enhance the ability of K. pneumoniae to produce 1, 3-PDO. The 1, 3-PDO production by K.P. L1-B3-PRH2521-P32-dhaT reached 57.85 g l(-1) in a 7.5 l fermentation tank (with Na+ neutralizer), which is higher than that of the original strain. This is the first time that the 1, 3-PDO production was improved in K. pneumoniae by overexpressing the key gene and attenuating by-product synthesis in the CRISPR-dCas9 system. This study reports an efficient approach to regulate the expression of genes in K. pneumoniae to increase the 1, 3-PDO production, and such a strategy may be useful to modify other strains to produce valuable chemicals.
Production of 3-Hydroxypropionic Acid Via the Propionyl-CoA Pathway Using Recombinant Escherichia Coli Strains.

Abstract:Our study aimed to produce the commercially promising platform chemical 3-hydroxypropionic acid (3-HP) via the propionyl-CoA pathway in genetically engineered Escherichia coli. Recombinant E. coli Ec-P overexpressing propionyl-CoA dehydrogenase (PACD, encoded by the pacd gene from Candida rugosa) under the T7 promoter produced 1.33 mM of 3-HP in a shake flask culture supplemented with 0.5% propionate. When propionate CoA-transferase (PCT, encoded by the pct gene from Megasphaera elsdenii) and 3-hydroxypropionyl-CoA dehydratase (HPCD, encoded by the hpcd gene from Chloroflexus aurantiacus) were expressed along with PACD, the 3-HP titer of the resulting E. coli Ec-PPH strain was improved by 6-fold. The effect of the cultivation conditions on the 3-HP yield from propionate in the Ec-PPH strain was also investigated. When cultured at 30°C with 1% glucose in addition to propionate, 3-HP production by Ec-PPH increased 2-fold and 12-fold compared to the cultivation at 37°C (4.23 mM) or without glucose (0.68 mM). Deletion of the ygfH gene encoding propionyl-CoA: succinate CoA-transferase from Ec-PPH (resulting in the strain Ec-△Y-PPH) led to increase of 3-HP production in shake flask experiments (15.04 mM), whereas the strain Ec-△Y-PPH with deletion of the prpC gene (encoding methylcitrate synthase in the methylcitrate cycle) produced 17.76 mM of 3-HP. The strain Ec-△Y-△P-PPH with both ygfH and prpC genes deleted produced 24.14 mM of 3-HP, thus showing an 18-fold increase in the 3-HP titer in compare to the strain Ec-P.

Abstract:Biosynthesis of 1,3-propanediol attracted much attention recently.The cell growth and biosynthesis of 1,3-propanediol by Klebsiella pneumoniae tended to be ceased in the latter phase of anaerobic culture.The effect of environmental factors was studied in this work.It was indicated that the main by-product ethanol was the key inhibitor.Addition of 4 g/L ethanol in the culture medium would restrain the cell growth and 1,3-propanediol formation.By addition of 8 g/L ethanol,the maximum specific rates of cell growth and 1,3-propanediol biosynthesis,the maximum concentrations of biomass and 1,3-propanediol were decreased by 21.6%,22.1%,59.6% and 33.5% respectively.The inhibition potentials of other by-products on the anaerobic growth of K.pneumoniae were also detected.It was shown that the cell growth was inhibited obviously with 2 g/L acetic acid,yet it would not be affected even with 5 g/L 2,3-butanediol or 10 g/L lactic acid.

Abstract:1,3-propanediol oxidoreductase (PDOR) and formate dehydrogenase (fdh) were co-overexpressed in the wild type Klebsiella pneumoniae to enhance the NADH production and decrease the 3-HPA accumulation which may cause an irreversible cessation of the 1,3-propanediol fermentation process.dhaT gene encoded PDOR from K.pneumoniae and fdh gene encoded fdh from Candida boidinii were both amplified and then overexpressed in K.pneumoniae.For the constructed strain,the activity of PDOR,fdh,and NADH were 2.8,3.5,and 1.3 fold higher than the wild type strain,respectively.In the batch fermentation with initial 50 g/L glycerol,3-HPA concentration decreased by 50.1%,which effectively avoided the irreversible cessation of the fermentation process with strain K.p/pTF.
Improvement of 1,3-Propanediol Production by Recombinant Formate Dehydrogenase in Klebsiella Pneumoniae

Abstract:Biosynthesis of 1,3-propanediol from glycerol by Klebsiella pneumoniae was associated with the consumption of NADH,which was generated/regenerated in the oxidative branch pathway of glycerol in vivo.The availability of NADH would be critical for the final yield of 1,3-propanediol.Formate dehydrogenase gene (fdh) was cloned from Candida boidinii genome and was then transformed into Klebsiella pneumoniae YMU2 by constructed expression vector pMALTM-p2X-fdh to generate recombinant Klebsiella pneumoniae F-1.Compared with K.pneumoniae YMU2,the yield of 1,3-propanediol and production rate of recombinant F-1 were 78.6 g·L-1 and 1.33 g·L-1·h-1,increased by 12.5% and 41.2%,respectively.The metabolic flux of strain YMU2 and recombinant F-1 was analyzed with the formation of main metabolites.
