Abstract:Brain tissues are surrounded by two tightly adhering thin membranes known as the pia-arachnoid complex (PAC), which is pivotal in regulating brain mechanical response upon mechanical impact. Despite the crucial role of PAC as a structural damper protecting the brain, its mechanical contribution has received minimal attention. In this work, the mechanical contribution of PAC on brain tissues against mechanical loading is characterized by using a custom-built indentation apparatus. The indentation responses of the isolated and PAC-overlaid brains are quantitatively compared at different length scales and strain rates. Results show that PAC substantially affects the indentation response of brain tissues at micro- and macro-scales and provides better protection against mechanical impact at a relatively small (μm) length scale. The modulus of the PAC-overlaid brain shows a threefold stiffening at the microscale compared with that of the isolated brain (with instantaneous shear modulus distribution means of 0.85 ± 0.14 kPa versus 2.64 ± 0.43 kPa at the strain rate of 0.64 s-1 and 1.40 ± 0.31 kPa versus 4.02 ± 0.51 at 1.27 s-1). These findings indicate that PAC seriously affects the mechanical response of brain tissues, especially at the microscale, and may have important implications for the studies of brain injury.

Abstract:Traumatic brain injury (TBI) has become a recent focus of biomedical research with a growing international effort targeting material characterization of brain tissue and simulations of trauma using computer models of the head and brain to try to elucidate the mechanisms and pathogenesis of TBI. The meninges, a collagenous protective tri-layer, which encloses the entire brain and spinal cord has been largely overlooked in these material characterization studies. This has resulted in a lack of accurate constitutive data for the cranial meninges, particularly under dynamic conditions such as those experienced during head impacts. The work presented here addresses this lack of data by providing for the first time, in situ large deformation material properties of the porcine dura-arachnoid mater composite under dynamic indentation. It is demonstrated that this tissue is substantially stiffer (shear modulus, μ=19.10±8.55kPa) and relaxes at a slower rate (τ1=0.034±0.008s, τ2=0.336±0.077s) than the underlying brain tissue (μ=6.97±2.26kPa, τ1=0.021±0.007s, τ2=0.199±0.036s), reducing the magnitudes of stress by 250% and 65% for strains that arise during indentation-type deformations in adolescent brains. Statement of significance: We present the first mechanical analysis of the protective capacity of the cranial meninges using in situ micro-indentation techniques. Force-relaxation tests are performed on in situ meninges and cortex tissue, under large strain dynamic micro-indentation. A quasi-linear viscoelastic model is used subsequently, providing time-dependent mechanical properties of these neural tissues under loading conditions comparable to what is experienced in TBI. The reported data highlights the large differences in mechanical properties between these two tissues. Finite element simulations of the indentation experiments are also performed to investigate the protective capacity of the meninges. These simulations show that the meninges protect the underlying brain tissue by reducing the overall magnitude of stress by 250% and up to 65% for strains.

Abstract:Abstract Background The pia arachnoid complex (PAC) is a cerebrospinal fluid-filled tissue conglomerate that surrounds the brain and spinal cord. Pia mater adheres directly to the surface of the brain while the arachnoid mater adheres to the deep surface of the dura mater. Collagen fibers, known as subarachnoid trabeculae (SAT) fibers, and microvascular structure lie intermediately to the pia and arachnoid meninges. Due to its structural role, alterations to the biomechanical properties of the PAC may change surface stress loading in traumatic brain injury (TBI) caused by sub-concussive hits. The aim of this study was to quantify the mechanical and morphological properties of ovine PAC. Methods Ovine brain samples (n = 10) were removed from the skull and tissue was harvested within 30 min post-mortem. To access the PAC, ovine skulls were split medially from the occipital region down the nasal bone on the superior and inferior aspects of the skull. A template was used to remove arachnoid samples from the left and right sides of the frontal and occipital regions of the brain. 10 ex-vivo samples were tested with uniaxial tension at 2 mm s −1 , average strain rate of 0.59 s −1 , until failure at < 5 h post extraction. The force and displacement data were acquired at 100 Hz. PAC tissue collagen fiber microstructure was characterized using second-harmonic generation (SHG) imaging on a subset of n = 4 stained tissue samples. To differentiate transverse blood vessels from SAT by visualization of cell nuclei and endothelial cells, samples were stained with DAPI and anti-von Willebrand Factor, respectively. The Mooney-Rivlin model for average stress–strain curve fit was used to model PAC material properties. Results The elastic modulus, ultimate stress, and ultimate strain were found to be 7.7 ± 3.0, 2.7 ± 0.76 MPa, and 0.60 ± 0.13, respectively. No statistical significance was found across brain dissection locations in terms of biomechanical properties. SHG images were post-processed to obtain average SAT fiber intersection density, concentration, porosity, tortuosity, segment length, orientation, radial counts, and diameter as 0.23, 26.14, 73.86%, 1.07 ± 0.28, 17.33 ± 15.25 μm, 84.66 ± 49.18°, 8.15%, 3.46 ± 1.62 μm, respectively. Conclusion For the sizes, strain, and strain rates tested, our results suggest that ovine PAC mechanical behavior is isotropic, and that the Mooney-Rivlin model is an appropriate curve-fitting constitutive equation for obtaining material parameters of PAC tissues.

Abstract:The cerebral meninges, made up of the dura, arachnoid, and pia mater, is a tri-layer membrane that surrounds the brain and the spinal cord and has an important function in protecting the brain from injury. Understanding its mechanical behavior is important to ensure the accuracy of finite element (FE) head model simulations which are commonly used in the study of traumatic brain injury (TBI). Mechanical characterization of freshly excised porcine dura-arachnoid mater (DAM) was achieved using uniaxial tensile testing and bulge inflation testing, highlighting the dependency of the identified parameters on the testing method. Experimental data was fit to the Ogden hyperelastic material model with best fit material parameters of μ = 450 ± 190 kPa and α = 16.55 ± 3.16 for uniaxial testing, and μ = 234 ± 193 kPa and α = 8.19 ± 3.29 for bulge inflation testing. The average ultimate tensile strength of the DAM was 6.91 ± 2.00 MPa (uniaxial), and the rupture stress at burst was 2.08 ± 0.41 MPa (inflation). A structural analysis using small angle light scattering (SALS) revealed that while local regions of highly aligned fibers exist, globally, there is no preferred orientation of fibers and the cerebral DAM can be considered to be structurally isotropic. This confirms the results of the uniaxial mechanical testing which found that there was no statistical difference between samples tested in the longitudinal and transversal direction (p = 0.13 for μ, p = 0.87 for α). A finite element simulation of a craniotomy procedure following brain swelling revealed that the mechanical properties of the meninges are important for predicting accurate stress and strain fields in the brain and meninges. Indeed, a simulation using a common linear elastic representation of the meninges was compared to the present material properties (Ogden model) and the intracranial pressure was found to differ by a factor of 3. The current study has provided researchers with primary experimental data on the mechanical behavior of the meninges which will further improve the accuracy of FE head models used in TBI.

Abstract:Tuning the parameters of a PID controller is very important in PID control. Ziegler and Nichols proposed the well-known Ziegler-Nichols method to tune the coefficients of a PID controller. This tuning method is very simple, but cannot guarantee to be always effective. For this reason, this paper investigates the design of self tuning for a PID controller. The controller includes two parts: conventional PID controller and fuzzy logic control (FLC) part, which has self tuning capabilities in set point tracking performance. The proportional, integral and derivate (KP, KI, KD) gains in a system can be self-tuned on-line with the output of the system under control. The conventional PI controller (speed controller) in the Chopper-Fed DC Motor Drive is replaced by the self tuning PID controller, to make them more general and to achieve minimum steady-state error, also to improve the other dynamic behavior (overshoot). Computer Simulation is conducted to demonstrate its performance and results show that the proposed design is success over the conventional PID controller.

Abstract:The pia-arachnoid complex (PAC) encasing the brain plays an important role in determining the structural response of the brain due to impact or inertial loading. Some tests about the mechanical properties of the bovine's PAC have been characterized previously. The finite element model can provide useful information about traumatic brain injury, but there is no head finite element model which includes the PAC structure with the true material parameters from tests. The objective of this study is to develop the finite element model of PAC based on the latest test data. Two PAC FE models in which the beam elements and solid elements were used to simulate the trabeculae were developed by using LS-DYNA3D 970. The simulated PAC's stress-strain behaviors of two FE models and experimental results were correlated very well.

Abstract:Finite element head models offer great potential to study brain-related injuries; however, at present may be limited by geometric and material property simplifications required for continuum-level human body models. Specifically, the mechanical properties of the brain tissues are often represented with simplified linear viscoelastic models, or the material properties have been optimized to specific impact cases. In addition, anatomical structures such as the arachnoid complex have been omitted or implemented in a simple lumped manner. Recent material test data for four brain regions at three strain rates in three modes of loading (tension, compression, and shear) was used to fit material parameters for a hyper-viscoelastic constitutive model. The material model was implemented in a contemporary detailed head finite element model. A detailed representation of the arachnoid trabeculae was implemented with mechanical properties based on experimental data. The enhanced head model was assessed by re-creating 11 ex vivo head impact scenarios and comparing the simulation results with experimental data. The hyper-viscoelastic model faithfully captured mechanical properties of the brain tissue in three modes of loading and multiple strain rates. The enhanced head model showed a high level of biofidelity in all re-created impacts in part due to the improved brain-skull interface associated with implementation of the arachnoid trabeculae. The enhanced head model provides an improved predictive capability with material properties based on tissue level data and is positioned to investigate head injury and tissue damage in the future.

Abstract:AbstractThe brain is a complex organ made up of many different functional and structural regions consisting of different types of cells such as neurons and glia, as well as complex anatomical geometries. It is hypothesized that the different regions of the brain exhibit significantly different mechanical properties, which may be attributed to the diversity of cells and anisotropy of neuronal fibers within individual brain regions. The regional dynamic mechanical properties of P56 mouse brain tissue in vitro and in situ at velocities of 0.71–4.28 mm/s, up to a deformation of 70 μm are presented and discussed in the context of traumatic brain injury. The experimental data obtained from micro-indentation measurements were fit to three hyperelastic material models using the inverse Finite Element method. The cerebral cortex elicited a stiffer response than the cerebellum, thalamus and medulla oblongata regions for all velocities. The thalamus was found to be the least sensitive to changes in velocity and the medulla oblongata was most compliant. The results show that different regions of the mouse brain possess significantly different mechanical properties and a significant difference also exists between the in vitro and in situ brain.

Abstract:Background Cerebrospinal fluid leakage through the spinal meninges is difficult to diagnose and treat. Moreover, its underlying mechanism remains unknown. Considering that the dura mater is structurally the strongest and outermost membrane among the three-layered meninges, we hypothesized that a dural mechanical tear would trigger spontaneous cerebrospinal fluid leakage, especially when a traumatic loading event is involved. Thus, accurate biomechanical properties of the dura mater are indispensable for improving computational models, which aid in predicting blunt impact injuries and creating artificial substitutes for transplantation and surgical training. Method We characterized the surface profile of the spinal dura and its mechanical properties (Young's moduli) with a distinction of its inherent anatomical sites ( i.e. , the cervical and lumbar regions as well as the dorsal and ventral sides of the spinal cord). Findings Although the obtained Young's moduli exhibited no considerable difference between the aforementioned anatomical sites, our results suggested that the wrinkles structurally formed along the longitudinal direction would relieve stress concentration on the dural surface under in vivo and supraphysiological conditions, enabling mechanical protection of the dural tissue from a blunt impact force that was externally applied to the spine. Interpretation This study provides fundamental data that can be used for accurately predicting cerebrospinal fluid leakage due to blunt impact trauma.

Biomechanics of vascular areas of the human cranial dura mater

Abstract:Accurate biomechanical properties of the human cranial dura mater are paramount for computational head models, artificial graft developments and biomechanical basic research. Yet, it is unclear whether areas of the dura containing meningeal vessels biomechanically differ from avascular areas. Here, 244 dura mater samples with or without vessels from 32 cadavers were tested in a quasi-static uniaxial tensile testing setup. The thicknesses of the meningeal and periosteal dura in vascular and avascular areas were histologically investigated in 36 samples using van Gieson staining. The elastic modulus of 112 MPa from dura samples containing vessels running transversely was significantly lower than samples with vessels running longitudinally (151 MPa; p < 0.001). The ultimate tensile strength of dura samples with transversely running vessels (11.1 MPa) was significantly lower in comparison to both avascular samples (14.9 MPa; p < 0.001) and samples with a longitudinally running vessel (15.0 MPa; p < 0.001). The maximum force of dura samples with longitudinally running vessels was 37 N (p < 0.001), this was significantly higher compared to the other groups which were 23 N (p < 0.001). The meningeal and periosteal dura layer thicknesses were not statistically different in avascular areas (p > 0.222). However, around the vessels, the meningeal dura layer was significantly thicker compared to the periosteal layer (p ≤ 0.019). The sum of the meningeal and periosteal layers was similar between vascular and avascular areas (p ≥ 0.071). Vascular areas of the human cranial dura mater withstand the same forces as avascular areas when being stretched. When stretched along the vessel, the dura-vessel composite can withstand even higher tensile forces compared to avascular areas. Vascular areas of the cranial dura mater seem to be similar when compared to avascular areas making their separate simulation in computational models non-essential.
Age-dependent changes in material properties of the brain and braincase of the rat

Abstract:Clinical and biomechanical evidence indicates that mechanisms and pathology of head injury in infants and young children may be different from those in adults. Biomechanical computer-based modeling, which can be used to provide insight into the thresholds for traumatic tissue injury, requires data on material properties of the brain, skull, and sutures that are specific for the pediatric population. In this study, brain material properties were determined for rats at postnatal days (PND) 13, 17, 43, and 90, and skull/suture composite (braincase) properties were determined at PND 13, 17, and 43. Controlled 1 mm indentation of a force probe into the brain was used to measure naive, non-preconditioned (NPC) and preconditioned (PC) instantaneous (G(i)) and long-term (G( infinity )) shear moduli of brain tissue both in situ and in vitro. Brains at 13 and 17 PND exhibited statistically indistinguishable shear moduli, as did brains at 43 and 90 PND. However, the immature (average of 13 and 17 PND) rat brain (G(i) = 3336 Pa NPC, 1754 Pa PC; G( infinity )= 786 Pa NPC, 626 Pa PC) was significantly stiffer (p < 0.05) than the mature (average of 43 and 90 PND) brains (G(i) = 1721 Pa NPC, 1232 Pa PC; G( infinity ) = 508 Pa NPC, 398 Pa PC). A "reverse engineering" finite element model approach, which simulated the indentation of the force probe into the intact braincase, was used to estimate the effective elastic moduli of the braincase. Although the skull of older rats was significantly thicker than that of the younger rats, there was no significant age-dependent change in the effective elastic modulus of the braincase (average value = 6.3 MPa). Thus, the increase in structural rigidity of the braincase with age (up to 43 PND) was due to an increase in skull thickness rather than stiffening of the tissue. These observations of a stiffer brain and more compliant braincase in the immature rat compared with the adult rat will aid in the development of age-specific experimental models and in computational head injury simulations. Specifically, these results will assist in the selection of forces to induce comparable mechanical stresses, strains and consequent injury profiles in brain tissues of immature and adult animals.
Mechanical characterization of brain tissue in simple shear at dynamic strain rates

Abstract:During severe impact conditions, brain tissue experiences a rapid and complex deformation, which can be seen as a mixture of compression, tension and shear. Moreover, diffuse axonal injury (DAI) occurs in animals and humans when both the strains and strain rates exceed 10% and 10/s, respectively. Knowing the mechanical properties of brain tissue in shear at these strains and strain rates is thus of particular importance, as they can be used in finite element simulations to predict the occurrence of brain injuries under different impact conditions. In this research, an experimental setup was developed to perform simple shear tests on porcine brain tissue at strain rates < 120/s. The maximum measured shear stress at strain rates of 30, 60, 90 and 120/s was 1.15 +/- 0.25 kPa, 1.34 +/- 0.19 kPa, 2.19 +/- 0.225 kPa and 2.52 +/- 0.27 kPa, (mean +/- SD), respectively, at the maximum amount of shear, K = 1. Good agreement of experimental, theoretical (Ogden and MooneyRivlin models) and numerical shear stresses was achieved (p = 0.7866 - 0.9935). Specimen thickness effects (2.0 - 10.0 mm thick specimens) were also analyzed numerically and we found that there is no significant difference (p = 0.9954) in the shear stress magnitudes, indicating a homogeneous deformation of the specimens during simple shear tests. Stress relaxation tests in simple shear were also conducted at different strain magnitudes (10% - 60% strain) with the average rise time of 14 ms. This allowed us to estimate elastic and viscoelastic parameters (mu = 4942.0 Pa and Prony parameters: g1 = 0.520, g2 = 0.3057, tau1 = 0.0264 s, tau2 = 0.011 s) that can be used in FE software to analyze the hyperviscoelastic behavior of brain tissue.

Abstract:Brain mechanics is a topic of deep interest because of the significant role of mechanical cues in both brain function and form. Specifically, capturing the heterogeneous and anisotropic behaviour of cerebral white matter (WM) is extremely challenging and yet the data on WM at a spatial resolution relevant to tissue components are sparse. To investigate the time-dependent mechanical behaviour of WM, and its dependence on local microstructural features when subjected to small deformations, we conducted atomic force microscopy (AFM) stress relaxation experiments on corpus callosum (CC), corona radiata (CR) and fornix (FO) of fresh ovine brain. Our experimental results show a dependency of the tissue mechanical response on axons orientation, with e.g. the stiffness of perpendicular and parallel samples is different in all three regions of WM whereas the relaxation behaviour is different for the CC and FO regions. An inverse modelling approach was adopted to extract Prony series parameters of the tissue components, i.e. axons and extra cellular matrix with its accessory cells, from experimental data. Using a bottom-up approach, we developed analytical and FEA estimates that are in good agreement with our experimental results. Our systematic characterisation of sheep brain WM using a combination of AFM experiments and micromechanical models provide a significant contribution for predicting localised time-dependent mechanics of brain tissue. This information can lead to more accurate computational simulations, therefore aiding the development of surgical robotic solutions for drug delivery and accurate tissue mimics, as well as the determination of criteria for tissue injury and predict brain development and disease progression.

Strain concentration drives the anatomical distribution of injury in acute and chronic traumatic brain injury

Abstract:Brain tissue injury caused by mild traumatic brain injury (mTBI) disproportionately concentrates in the midbrain, cerebellum, mesial temporal lobe, and the interface between cortex and white matter at sulcal depths. The bio-mechanical principles that explain why physical impacts to different parts of the skull translate to common foci of injury concentrated in specific brain structures are unknown. A general and longstanding idea, which has not to date been directly tested in humans, is that different brain regions are differentially susceptible to strain loading. We use Magnetic Resonance Elastography (MRE) in healthy participants to develop whole-brain bio-mechanical vulnerability maps that independently define which regions of the brain exhibit disproportionate strain concentration. We then validate those vulnerability maps in a prospective cohort of mTBI patients, using diffusion MRI data collected at three cross-sectional timepoints after injury: acute, sub-acute, chronic. We show that regions that exhibit high strain, measured with MRE, are also the sites of greatest injury, as measured with diffusion MR in mTBI patients. This was the case in acute, subacute, and chronic subgroups of the mTBI cohort. Follow-on analyses decomposed the biomechanical cause of increased strain by showing it is caused jointly by disproportionately higher levels of energy arriving to high-strain structures, as well as the inability of high-strain structures to effectively disperse that energy. These findings establish a causal mechanism that explains the anatomy of injury in mTBI based on in vivo rheological properties of the human brain.

Abstract:Research on the mechanical properties of brain tissue has gradually deepened recently. Two indentation protocols were used here to characterize the mechanical properties of cortical tissues. Further, histological staining was used to explore the correlation between the mechanical properties and microstructure on the basis of the density of cell nuclei and proteoglycan content. No significant difference was observed in transient contact stiffness between the cerebral cortex and cerebellar cortex at the depth interval of 0-600 μm under the cortical surface; however, the average shear modulus of the cerebral cortex was higher than that of the cerebellar cortex. The cerebral cortex responded more quickly to the change in load and released stress more thoroughly than the cerebellar cortex. In addition, the density of cell nuclei was related to both the transient contact stiffness and second time constant of cortical tissues. Proteoglycan content had a more significant impact on the shear modulus, second time constant, and stress relaxation rate of cortical tissues. Exploring mechanical properties thoroughly will provide more detailed mechanical information for future brain chip implantation. Alternatively, linking the mechanical properties of cortical tissues to the microstructure can provide basic data for the design and manufacture of substitute materials for brain tissue.

Abstract:Kabuki syndrome, OMIM 147920 (KS) is a disorder characterized by multi‐system abnormalities. These include physical, neurological, endocrine, and autoimmune abnormalities. Multiple autoimmune abnormalities are described in KS such as immune thrombocytopenic purpura (ITP), vitiligo, thyroiditis, hemolytic anemia, and hypogammaglobulinemia. In this report, we describe a patient with KS with sudden onset chorea associated with the presence of anti‐phospholipid antibodies (aPLs) in the serum. Chorea in the presence of aPLs has been well described in the literature both in the presence and absence of antiphospholipid syndrome (APS) and systemic lupus erythematosus (SLE). This report of APL in a patient with KS adds to the list of autoimmune disorders seen in patients with KS and also strengthens the hypothesis that patients with this syndrome have an increased incidence of immune dysregulation. © 2007 Wiley‐Liss, Inc.
Biphasic analysis of rat brain slices under creep indentation shows nonlinear tension-compression behavior

Abstract:Biphasic theory can provide a mechanistic description of deformation and transport phenomena in soft tissues, and has been used to model surgery and drug delivery in the brain for decades. Knowledge of corresponding mechanical properties of the brain is needed to accurately predict tissue deformation and flow transport in these applications. Previously in our group, creep indentation tests were conducted for multiple anatomical regions in acute rat brain tissue slices. In the current study, a biphasic finite element model of creep indentation was developed with which to compare these data. Considering the soft tissue structure of brain, the solid matrix was assumed to be composed of a neo-Hookean ground matrix reinforced by continuously distributed fibers that exhibits tension-compression nonlinearity during deformation. By fixing Poisson's ratio of the ground matrix, Young's modulus, fiber modulus and hydraulic permeability were estimated. Hydraulic permeability was found to be nearly independent of the properties of the solid matrix. Estimated modulus (40 Pa to 1.1 kPa for the ground matrix, 3.2-18.2 kPa for fibers) and hydraulic permeability (1.2-5.5×10-13m4/N s) fell within an acceptable range compared with those in previous studies. Instantaneous indentation depth was dominated by tension provided by fibers, while the tissue response at equilibrium was sensitive to Poisson's ratio. Results of sensitivity analysis also point to the necessity of considering tension-compression nonlinearity in the solid phase when the biphasic material undergoes large creep deformation.
