Synthesis and Reactivity in Inorganic Metal-Organic and Nano-Metal Chemistry

Abstract:Four new types of iodo-bridged binuclear platinum complexes [Pt2AI5] (A = dimethylamine, pyridine, 2-methylpyridine, and o-toluidine) (a-d) have been synthesized and characterized by elemental analysis, conductivity, thermal analysis, IR, 1H NMR, and mass spectra techniques. The cytotoxicity was investigated by MTT assay. The results indicate that complexes (a-d) have certain cytotoxicity against Bel-7402, HCT-8 and Hela cell lines. The complex a, c, and d display increased cytotoxicity than cisplatin against HCT-8, Bel-7402 and Hela cell lines, respectively. The results suggest that these complexes have selectivity against the tested carcinoma cell lines, with the amine ligand having an important effect on the cytotoxicity.

Abstract:Two novel complexes were prepared and showed remarkable anticancer activity for HeLa cells. Fluorescence and UV–vis spectroscopy showed that these complexes interacted with DNA through the embedding methods. Traditional platinum (Pt)‐based drugs have limited use in cancer treatment because of their toxicity, which generates the need for new Pt complexes with effective anticancer activity. Two potent prospective anticancer small molecule drug complexes, PtLCl (1) and PdLCl (2) (L:4‐[2,2′:6′,2′‐terpyridin‐4′‐yl]phenol), were synthesized via a low‐temperature solvothermal method. The structures of the complexes are characterized by a four‐coordinated rhombus plane, according to single‐crystal X‐ray diffraction studies. The complexes 1 and 2 were further characterized using several techniques including powder X‐ray diffraction, infrared spectroscopy, and cyclic voltammetry. Cytotoxicity was assessed using flow cytometry and morphological observations of HeLa cells. The findings demonstrate that both complexes were highly effective at inducing cell death, and the tumor inhibition rate of 1 was greater than that of 2, with total tumor inhibition of 92.3% and 72.3%, respectively, after 24 h. In addition, fluorescence and UV–vis spectroscopy showed that these complexes interacted with DNA through the embedding methods. The four‐coordinated planar structure and π–π stacking could prevent DNA transfer and replication, which has the potential for more effective cancer cell eradication through intercalation. Therefore, these two complexes are promising candidates for the development of new Pt‐ and Pd‐based anticancer drugs.

Abstract:Five new tetradentate ligands and their corresponding palladium complexes, [Pd(L)]Cl2 (L = N,N′-dimethyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-diethyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-dipropyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-ditert-butyl-1,10-phenanthroline-2,9-dimathanamine, N,N′-dicyclohexyl-1,10-phenanthroline-2,9-dimathanamine) have been synthesized. The ligands and their complexes have been characterized by elemental analysis, IR, and 1H NMR. The complexes have been assayed for antitumor activity in vitro against the mouse leukemia L1210 and the mouse liver carcinoma Bel7402 cell lines. The results showed that the activities of these complexes are significantly dependent on the nature of the alkyl groups on the coordinated amine moieties, and three of these palladium complexes showed lower ID50 values against the two cell lines than cisplatin.
Synthesis and Biological Activity of Binuclear Platinum Complexes Containing Two Monofunctional Cis-[Pt(Nh3)(2)Cl](+) Units Bridged by 4,4 '-Dipyridyl Selenides or Sulfides

Abstract:The synthesis of four binuclear platinum complexes of general formula ¿cis-[Pt(NH3)2Cl]2(L)¿ (NO3)2 [L is 4,4'-dipyridyl sulfide (compound I), 4,4'-dipyridyl selenide (compound II), bis(3-methyl-4-pyridyl) sulfide (compound III) or bis(3-methyl-4-pyridyl) selenide (compound IV)] has been achieved. These compounds have been characterized by elemental analysis, IR, 1H-NMR, 13C-NMR and 195Pt-NMR spectroscopy. The above dinuclear platinum complexes have been assayed for antitumor activity in vitro against the cisplatin-sensitive L1210 (the mice leukemia) and cisplatin-insensitive HCT8 (the human coloadenocarcinoma) cell lines. The compounds show IC50 values comparable to or higher than cisplatin against L1210 cell line; however, they have lower IC50 values than cisplatin against the cisplatin-insensitive HCT8 cell line. The complexes containing S exhibit a cytotoxicity against the two cancer cell lines superior to the Se analogues. DNA binding studies indicate the compound I possibly interacts with DNA nonintercalatively. The mode of DNA binding of the dinuclear Pt(II) complexes bridged by 4,4'-dipyridyl selenides or sulfides may be different to that of the aliphatic diaminebridged dinuclear Pt(II) complexes reported previously.

Abstract:A series of novel Pd(II) and Pt(II) complexes, [PdL(2)Cl(2)]·DMF (1), [Pd(2)(L-H)(2)(bpy)Cl(2)]·(H(2)O)(2)·DMF (2), [Pd(2)(L-H)(2)(phen)Cl(2)]·2H(2)O (3), [PtL(2)Cl(2)]·H(2)O (4), [Pt(2)(L-H)(2)(bpy)Cl(2)]·2H(2)O (5), and [Pt(2)(L-H)(2)(phen)Cl(2)]·H(2)O (6), where bpy = 2,2'-bipyridine, phen = 1,10-phenanthroline, and L = 1,3-benzothiazol-2-amine, have been synthesized and characterized. The competitive binding of the complexes to DNA has been investigated by fluorescence spectroscopy. The values of the apparent DNA binding constant, calculated from fluorescence spectral studies, were 3.8 × 10(6) (K(app)(4)), 2.9 × 10(6) (K(app)(1)), 2.4 × 10(6) (K(app)(6)), 2.0 × 10(6) (K(app)(5)), 1.2 × 10(6) (K(app)(3)), and 6.9 × 10(5) (K(app)(2)). The binding parameters for the fluorescence Scatchard plot were also determined. On the basis of the data obtained, it indicates that the six complexes bind to DNA with different binding affinities in the relative order 4 > 1 > 6 > 5 > 3 > 2. Viscosity studies carried out on the interaction of complexes with Fish Sperm DNA (FS-DNA) suggested that all complexes bind by intercalation. Gel electrophoresis assay demonstrates that all the complexes can cleave the pBR 322 plasmid DNA and bind to DNA in a similar mode. The cytotoxic activity of the complexes has been also tested against four different cancer cell lines. The results show that all complexes have activity against KB, AGZY-83a, Hep-G2, and HeLa cells. In general, the Pt(II) complexes were found to be more effective than the isostructural Pd(II) complexes. The mononuclear complexes exhibited excellent activity in comparison with the dinuclear complexes in these four cell lines. Moreover, on the KB cell line (the human oral epithelial carcinoma), the observed result seems quite encouraging for the six complexes with IC(50) values ranging from 1.5 to 8.6 μM. Furthermore, apoptosis assay with hematoxylin-eosin staining shows treatment with the six complexes results in morphological changes of KB cells. The results induce apoptosis in KB cells.

Abstract:Four new benzopyran- platinum (Pt) complexes, [Pt(C1)Cl2]⋅CH3OH (C1-Pt), [Pt(C2)Cl2] (C2-Pt), [Pt(C3)Cl2] (C3-Pt), and [Pt(C4)Cl2] (C4-Pt) with 3-(1H-benzo[d]imidazol-2-yl)-2H-chromen-2-imine (C1), 6-bromo-3-(pyridin-2-yl)-2H-chromen-2-imine (C2), 6-chloro-3-(pyridin-2-yl)-2H-chromen-2-imine (C3), and 5,7-dichloro-3-(pyridin-2-yl)-2H-chromen-2-imine (C4), were first designed and synthesized as mitochondria-targeting antitumor agents. C1-Pt−C4-Pt showed cytotoxic activity against A549 and A549/DDP cancer cells, and most 6-chloro-substituted groups had stronger cytotoxicity than did the benzopyran Pt(II)-substituted ones. Significantly, C3-Pt was much more effective on cisplatin-resistant A549/DDP tumor cells (IC50= 0.15±0.03 μM) than C1-Pt, C2-Pt and C4-Pt. Interestingly, C1-Pt−C4-Pt exhibited much lower cytotoxicity against normal cells (IC50>100 μM) than against A549/DDP tumor cells. Additionally, mechanistic studies showed that C2-Pt and C3-Pt significantly inhibited cell growth in A549/DDP cells in S phase, accumulated more in mitochondria than in nuclei, and caused the loss of mitochondrial membrane potential, concurrently targeting mitochondria.

Abstract:Two binuclear 3N-chelated monofunctional PtII complexes, [Pt2L1Cl2]Cl2 (complex III) and [Pt2L2Cl2]Cl2 (complex IV) [L1=3,6,9,16,19,22-hexaazatricyclo[22.2.2.211,14]-triaconta-11,13,24,26(1),27,29-hexaene, L2=3,6,9,17,20,23-hexaazatricyclo[23.3.1.111,15]-triaconta-1(29),11(30),12,14,25,27-hexaene] have been synthesized and structurally characterized. Structural determination revealed that each PtII center was coordinated by one chloride anion and three N atoms from each diethylenediamine motif. The Pt–Cl bonds in complex III are shorter than those found in complex IV. The rigid para- and meta-xylylene groups make the two complexes adopt a rigid boat-like conformation and a flexible twisted chair-like conformation, respectively. Moreover, complex III has higher tendency to bind with each other than complex IV. DNA binding studies demonstrated that complex IV could bind effectively with calf thymus DNA, possibly via platination of N7 of guanine residue, while no obvious DNA binding was observed for complex III. However, complex III displays a comparable cytotoxicity to cisplatin against HeLa cell line, while compound IV does not show any effective cell inhibition at low concentration. Therefore, the rigid spacers in complexes III and IV play a determining role in their anti-cancer activity and DNA binding ability.

Abstract:To develop efficient new antitumor agents, Ni(II), Co(II) and Zn(II) complexes 1 – 3 derived from 2-((2-pyrimidin-2-yl)hydrazono)methyl)quinoline (HL) were prepared and structurally characterized using IR, elemental analysis, UV-Vis, fluorescence spectrometry and single crystal X-ray diffraction analysis. The in vitro cytotoxicity of complexes 1 – 3 against MCF-7, BGC-823, A549, BEL-7402 tumor cell lines and human normal cell line HL-7702 was evaluated using MTT assay. Complexes 1 - 3 display higher cytotoxic activity than cisplatin as well as HL against BGC-823 tumor cell line, with IC 50 values ranging from 5.57 μM to 11.20 μM, and less toxicity toward the normal cell line HL-7702. In particular, complex 3 exhibits the most prominent antiproliferative activity. Complex 3 was found to induce apoptosis of BGC-823 cells greatly in a dose-dependent manner, which was suggested by Annexin V-FITC/PI and AO/EB dual-staining assays. The DNA binding studies, which were performed using UV-Vis, fluorescence and CD spectral analyzes, revealed that complex 3 interacts with CT-DNA mainly via a partial intercalative binding mode. The molecular docking of the interaction of complex 3 with DNA is in accordance with the experimental data.

Abstract:Pt(II) and Pd(II) complexes have emerged as highly potent anticancer drugs, and their efficacy in treating of various types cancer has garnered increasing attention. Herein, we report two trapezoid planar structures of promising complexes with 2-phenyl-1H-imidazo[4,5-f] [1] , [10] phenanthroline as a ligand. The structures of complexes 1 and 2 were studied by a series of characterizations, including single-crystal X-ray diffraction, thermogravimetric analysis and infrared spectroscopy. Fluorescence and UV-vis spectra, revealed the hypochromic phenomenon, which demonstrates the capacity of the complexes 1 and 2 interacting with ct-DNA. Furthermore, the ability of the complexes 1 and 2 to cut DNA was validated by the agar gel electrophoresis experiment. The cytotoxicity of 1 was assessed using flow cytometry analysis and morphology analysis. It is worth noting that, complex 1, had obvious advantages in inducing HeLa cell apoptosis, and the number of apoptotic cells (49.5 %) is significantly higher than that in the control group (1.6 %), after 24 h. Moreover, 1 can generate singlet oxygen and hydroxyl radical to damage cells under light irradiation, thus achieving an excellent to induced HeLa cells apoptosis. Overall, this work illustrated that complexes 1 and 2 are promising candidates for inducing apoptosis in HeLa cells.

Abstract:Three novel platinum‐based antitumor agents containing biotin and fluorinated pyridine ligands were synthesized by combining the conformational advantages of Pt(IV) precursors and picoplatin, and their physicochemical properties and interactions with DNA were investigated, and the in vitro cytotoxic activities of the complexes were also verified. Picoplatin can effectively circumvent cisplatin‐induced resistance through the "steric hindrance effect" of its structure, but there are still problems such as toxicity and side effects, so the search for safer, more efficient platinum analogs is crucial. Here we report three fluorinated pyridine‐containing Pt(IV) complexes employing biotin as an axial ligand to provide targeting functionality. Lipid solubility studies showed that all the synthesized complexes possessed improved lipid‐water partition coefficients, which are beneficial for transmembrane transport of drugs. The DNA binding activity of the compounds was studied using UV titration, fluorescence titration, and PicoGreen staining, demonstrating the presence of potential intercalation and electrostatic interaction binding modes. It probably attributable to planar π‐π interactions provided by the aromatic pyridine ligands. Meanwhile, molecular docking studies showed that the introduction of axial biotin molecules allowed the complexes to effectively target cancer cells, which was simultaneously confirmed by the results of cellular uptake studies. And another docking result showed that the Pt(IV) complexes binds to DNA by minor groove binding. The synthesized complexes exhibited similar cytotoxicity to cisplatin in five cell lines (SW480, HCT116, HepG2, A549 and LO2), with complex 3 being superior to the parent drug, picoplatin.

Monofunctional Supramolecular Pt(II) Complexes: Synthesis, Single Crystal Structure, Anticancer Activity, E. Coli Growth Retardation and DNA Interaction Study

Abstract:Two new monofunctional mono-metallic trans-Pt(II)(salicylaldimine)(pyridine)·BF4 (C1) and supramolecular dimetallic trans-(Pt(II)(salicylaldimine))2(4,4-bipyridine)·2BF4 (C2) complexes were designed and synthesized through ancillary chloride ligand exchange strategy and structurally characterized by spectroscopic, spectrophotometric and single crystal X-ray analyses. The solid-state structure analyses revealed interactions between the coordination planes, inter-molecular H-bonding, bonding in the ligand hydrogen and BF4 anions and supramolecular interactions with solvent molecules in crystal packing. The in vitro anticancer effect of these complexes was investigated in breast (MCF-7) and liver (HepG2) cancer cells. Both these complexes showed significant anticancer effect comparable to cisplatin. Similarly, the effect of these complexes on Escherichia coli (E. coli) growth retardation was also analyzed and the results revealed stronger growth retardation effect and elongated morphology of bacterial cells in similar fashion as observed for cisplatin. The DNA interaction of C1 and C2 was investigated by gel electrophoresis using pET28 as target DNA. These complexes retarded migration of DNA across the gel showing their interaction with DNA.
Design of a novel Pt(II) complex to reverse cisplatin-induced resistance in lung cancer via a multi-mechanism

Abstract:In order to develop a novel platinum (Pt) complex aiming to overcome cisplatin resistance, we synthesised a series of novel Pt complexes (C1-C6). These Pt complexes displayed potent cytotoxicity activity against resistant lung cancer cells (A549cisR) in vitro and efficiently inhibited tumour growth in vivo. The Pt complexes can target DNA, lead to DNA platination and cause cell cycle arrest in the 5 phase, thus impeding precise DNA synthesis. C6, in particular, induced not only apoptosis but also lethal autophagy in A549cisR cells. In addition, these novel Pt complexes reversed cisplatin-induced resistance via inhibiting the expression of P-glycoprotein and decreasing the level of glutathione in A549cisR cells. Moreover, the ERK pathway was involved in C6-induced overcoming cisplatin resistance.
Synthesis, characterization and anticancer properties: A series of highly selective palladium(II) substituted-terpyridine complexes

Abstract:Ten new palladium(II) complexes [PdCl(L1-10)]Cl have been synthesized by the reaction of palladium(II) chloride and ten 4'-(substituted-phenyl)-2,2':6',2''-terpyridine ligands bearing hydrogen(L1), p-hydroxyl(L2), m-hydroxyl (L3), o-hydroxyl (L4), methyl (L5), phenyl (L6), fluoro (L7), chloro (L8), bromo (L9), or iodo (L10). Their structures were confirmed by FT-IR, 1H NMR, elemental analysis and/or single crystal X-ray diffraction analysis. Their in vitro anticancer activities were investigated based on five cell lines, including four cancer cell lines (A549, Eca-109, Bel-7402, MCF-7) and one normal cell line (HL-7702). The results show that these complexes possess a strong killing effect on the cancer cells but a weak proliferative inhibition on the normal cells, implying their high inhibitory selectivity for the proliferation of the cancer cell lines. Flow cytometry characterization reveals that these complexes affect cell proliferation mainly in the G0/G1 phase and induce the late apoptotic of the cells. The quantity of palladium(II) ion in extracted DNA was determined by ICP-MS, which proved that these complexes target genomic DNA. And the strong affinity of the complexes with CT-DNA were confirmed by UV-Vis spectrum and circular dichroism (CD). The possible binding modes of the complexes with DNA were further explored by molecular docking. As the concentration of complexes 1-10 gradually increases, the fluorescence intensity of bovine serum albumin (BSA) decreases by a static quenching mechanism.
Structural Evidence For The Facile Chelate-Ring Opening Reactions Of Novel Platinum(Ii)-Pyridine Carboxamide Complexes

Abstract:The design of Pt(II) complexes with novel structural features is of great relevance in the search for new anticancer agents with innovative chemical and biological properties. In this work we have synthesised and structurally characterised three novel Pt() complexes with a pyridine carboxamide ligand (HL). The X-ray structural analysis shows that complex 1, [Pt(L-N, N')(2)] crystallises in space group C2, while complex 2, [Pt(L-N,N')(L-N)Cl] crystallises in space group P2(1). ESMS and H-1 NMR data show that one of the two ligands in complex 1 undergoes a fast ring-opening reaction to give complex 2 in the presence of Cl-. Complex 2 demonstrated notable activity against human and murine leukemia cells (HL-60 and P388), but rather weak potency against lung adenocarcinoma A-549. However, complex 1 was found to be basically inactive against the above cell lines. This work provides new examples in the design of platinum compounds that can be activated in vivo by biological anions.
A novel benzothiazole-based mononuclear platinum(II) complex displaying potent antiproliferative activity in HepG-2 cells via mitochondrial-mediated apoptosis

Abstract:A novel mononuclear platinum(II) complex, [Pt(L-H)Cl] (1, where L= N-(4-(benzo[d]thiazol-2-yl)phenyl)-2-((2-pyridylmethyl)(2-hydroxyethyl)-amino)acetamide), was obtained by covalently tethering a benzothiazole derivative 2-(4-aminophenyl)benzothiazole to the 2-pyridylmethyl-2-hydroxyethylamine chelating PtII center. In vitro tests indicated that complex 1 displayed excellent antiproliferative activity against the tested cancer cell lines, especially liver cancer HepG-2 and SMMC-7221 cells. Importantly, the complex possessed 4.33-fold higher antiproliferative activity as compared with cisplatin against HepG-2 cells, but was less toxic to the normal cell line L02 with the selectivity index (SI = IC50(L02)/IC50(HepG-2)) value of 8.36 compared to cisplatin (SI, 1.40). The results suggested that 1 might have the potential to act as a candidate for the treatment of hepatocellular carcinoma (HCC). Cellular uptake and distribution studies showed that 1 could effectively pass through the membrane of cells, enter the nuclei and mitochondria, induce the platination of cellular DNA. The interaction of 1 with CT-DNA demonstrated that 1 could effectively bind to DNA in a dual binding mode, i.e., the intercalation of the 2-(4-aminophenyl)benzothiazole unit plus monofunctional platination of the platinum(II) moiety. In addition, Hoechst 33342 staining and flow cytometry analysis illustrated that 1 arrested the cell cycle in HepG-2 cancer cells at G2/M phases, induced mitochondrial membrane depolarization, increased ROS generation, and caused obvious cell apoptosis. Further cellular mechanism studies elucidated that 1 triggered HepG-2 cell apoptosis via the mitochondrial-mediated pathway by upregulating the gene and protein expression levels of Bax, downregulating the gene and protein expression levels of Bcl-2, and activating the caspase cascade.

Abstract:Pyridine-derived Pt(II) complexes Pt1 , [Pt(NMPA)Cl 2 ]; Pt2 , [Pt(NMPA)(H 2 O)](NO 3 ) 2 ; Pt3 , [Pt(NMPA)(DL-pen)](NO 3 ); Pt4 , [Pt(NMPA)(DL‐meth)](NO 3 ) 2 ; Pt5 , [Pt(NMPA)(OX)]; and Pt6 , [Pt(NMPA)(CBDCA)] (where, NMPA = N -methyl-2-(2′-pyridyl)-ethylamine; DL-pen = DL-penicillamine; DL‐meth = DL-methionine; OX = oxalate; CBDCA = 1,1-cyclobutanedicarboxylate) were synthesized and characterized by elemental microanalysis and multi-spectroscopic methods. The electronic structures, molecular electrostatic potential, and natural bond orbital analysis, spectral simulations were studied by DFT and TD-DFT methods. Bio-interaction with CT-DNA was investigated with UV–Vis, competitive fluorescence titration, and viscometric measurements. BSA-complex interactions were probed by spectrophotometric methods. The experimental binding studies with DNA and BSA were strongly validated by molecular docking. ADMET and PASS parameters provided a clear theoretical interpretation of the pharmacokinetic and anticarcinogenic behavior of the synthesized drug candidates. The bio-efficacy and selectivity of the complexes was screened against A549 and HCT116 cell lines by in vitro cytotoxicity assay. A remarkable non-toxic effect on non-tumorigenic cells (HEK-293) was deduced by the NBT assay.

Abstract:The primary objective was to understand more deeply the molecular mechanism underlying different antitumor effects of dinuclear PtII complexes containing aromatic linkers of different length, {[cis-Pt(NH3)2Cl]2(4,4′-methylenedianiline)}2+ (1) and {[cis-Pt(NH3)2Cl]2(α,α′-diamino-p-xylene)}2+ (2). These complexes belong to a new generation of promising polynuclear platinum drugs resistant to decomposition by sulfur nucleophiles which hampers clinical use of bifunctional polynuclear trans PtII complexes hitherto tested. Results obtained with the aid of methods of molecular biophysics and pharmacology reveal differences and new details of DNA modifications by 1 and 2 and recognition of these modifications by cellular components. The results indicate that the unique properties of DNA interstrand cross-links of this class of polynuclear platinum complexes and recognition of these cross-links may play a prevalent role in antitumor effects of these metallodrugs. Moreover, the results show for the first time a strong specific recognition and binding of high-mobility-group-domain proteins, which are known to modulate antitumor effects of clinically used platinum drugs, to DNA modified by a polynuclear platinum complex.

Abstract:Many strategies have been used in treatment of cancer, but currently the chemotherapy is still the main force, unfortunately, general agents exhibit inevitable toxicity and inferior selectivity. Therefore, less toxic and high selectivity anticancer reagents have been extensively sought and developed. Here we report a series of Copper (II) complexes CuP1~CuP5 which have good biological activities. Due to the influence of the molecular size, CuP1 displayed the strongest bound with ct-DNA. All of the complexes show positive anticancer activities toward A549, H1975, HepG2 and T47D cancer cells. Additionally, flow cytometry experiments indicated that complexe CuP2 arrest cell cycles at S and G2 phase and mitochondrial membrane potential tests were also performed to explore the mechanism of action. Our findings disclosed that this type of complex represents a potent platform for development of metal anticancer drugs.
Synthesis and Characterization of Trans-Pt(ii)(salicylaldimine)(pyridine/pyridine-4-carbinol)cl Complexes: in Vivo Inhibition of E. Coli Growth and in Vitro Anticancer Activities

Abstract:Three trans-Pt(II) complexes, i.e. Pt(II)(salicylaldimine)(Py)Cl (C1), Pt(II)(salicyl-aldimine)(Py-4-carbinol)Cl (C2), and bi-metallic Pt(II)2(salicylaldimine)(Py)2Cl2 (C3), have been synthesized and their structures have been characterized by NMR, HRMS and X-ray crystal analysis. Bioactivities, especially in vivo inhibition of Escherichia coli growth and in vitro cytotoxicity in MCF-7 and HepG2 human cancer cell lines have been investigated. The results disclosed that all three complexes have dominant retardation effects on E. coli growth and considerable cytotoxicity to both cancer cell lines, which were comparable to those of cisplatin. For E. coli growth inhibition, that of mono-metallic C1 was most prominent than those of polar side chain substituted C2 and bi-metallic C3. Cell cycle studies revealed that their abilities of blocking cell cycle progression in luminal like breast cancer cells caused inhibition of DNA synthesis. Keeping in view their convenient synthesis, stabilities and good cytotoxicities as well as their stronger retardation effects to cell growth in bacteria, this class of Pt(II) complexes could find a possible consideration in anticancer drug discovery.

Abstract:Mitophagy, a selective autophagic process, has emerged as a pathway involved in degrading dysfunctional mitochondria. Herein, new platinum(II)-based chemotherapeutics with mitophagy-targeting properties were proposed. Four novel binuclear anticancer Pt(II) complexes with 4′-substituted-2,2′:6′,2′′-terpyridine derivatives (tpy1–tpy4), i.e., [Pt2(tpy1)(DMSO)2Cl4]CH3OH (tpy1Pt), [Pt(tpy2)Cl][Pt(DMSO)Cl3]CH3COCH3 (tpy2Pt), [Pt(tpy3)Cl][Pt(DMSO)Cl3] (tpy3Pt), and [Pt(tpy4)Cl]ClCH3OH (tpy4Pt), were designed and prepared. Moreover, their potential antitumor mechanism was studied. Tpy1Pt–tpy4Pt exhibited more selective cytotoxicity against cisplatin-resistant SK-OV-3/DDP (SKO3cisR) cancer cells compared with those against ovarian SK-OV-3 (SKO3) cancer cells and normal HL-7702 liver (H702) cells. This selective cytotoxicity of Tpy1Pt–tpy4Pt was better than that of its ligands (i.e., tpy1–tpy4), the clinical drug cisplatin, and cis-Pt(DMSO)2Cl2. The results of various experiments indicated that tpy1Pt and tpy2Pt kill SKO3cisR cancer cells via a mitophagy pathway, which involves the disruption of the mitophagy-related protein expression, dissipation of the mitochondrial membrane potential, elevation of the [Ca2+] and reactive oxygen species levels, promotion of mitochondrial DNA damage, and reduction in the adenosine triphosphate and mitochondrial respiratory chain levels. Furthermore, in vivo experiments indicated that the dinuclear anticancer Pt(II) coordination compound (tpy1Pt) has remarkable therapeutic efficiency (ca. 52.4%) and almost no toxicity. Therefore, the new 4′-substituted-2,2′:6′,2′′-terpyridine Pt(II) coordination compound (tpy1Pt) is a potential candidate for next-generation mitophagy-targeting dinuclear Pt(II)-based anticancer drugs.
