Abstract:Background-Preeclampsia is a prevalent life-threatening hypertensive disorder of pregnancy. The circulating antiangiogenic factor, soluble endoglin (sEng), is elevated in the blood circulation of women with preeclampsia and contributes to disease pathology; however, the underlying mechanisms responsible for its induction in preeclampsia are unknown.Methods and Results-Here, we discovered that a circulating autoantibody, the angiotensin receptor agonistic autoantibody (AT(1)-AA), stimulates sEng production via AT(1) angiotensin receptor activation in pregnant mice but not in nonpregnant mice. We subsequently demonstrated that the placenta is a major source contributing to sEng induction in vivo and that AT(1)-AA-injected pregnant mice display impaired placental angiogenesis. Using drug screening, we identified tumor necrosis factor-alpha as a circulating factor increased in the serum of autoantibody-injected pregnant mice contributing to AT(1)-AA-mediated sEng induction in human umbilical vascular endothelial cells. Subsequently, among all the drugs screened, we found that hemin, an inducer of heme oxygenase, functions as a break to control AT1-AA-mediated sEng induction by suppressing tumor necrosis factor-alpha signaling in human umbilical vascular endothelial cells. Finally, we demonstrated that the AT1-AA-mediated decreased angiogenesis seen in human placenta villous explants was attenuated by tumor necrosis factor-alpha-neutralizing antibodies, soluble tumor necrosis factor-alpha receptors, and hemin by abolishing both sEng and soluble fms-like tyrosine kinase-1 induction.Conclusions-Our findings demonstrate that AT1-AA-mediated tumor necrosis factor-alpha induction, by overcoming its negative regulator, heme oxygenase-1, is a key underlying mechanism responsible for impaired placental angiogenesis by inducing both sEng and soluble fms-like tyrosine kinase-1 secretion from human villous explants. Our results provide important new targets for diagnosis and therapeutic intervention in the management of preeclampsia. (Circulation. 2010;121:436-444.)

Abstract:Pre-eclampsia is a complication of pregnancy characterized by hypertension and proteinuria that results in maternal and neonatal mortality and morbidity.Although the etiology remains unclear,it seems to be initiated by placental factors that enter the maternal circulation and may cause endothelial dysfunction.Recently,anti-angiogenic factors such as endoglin,the soluble form of vascular endothelial growth factor receptor(sFlt-1) as well as the placental growth factor have been found to be associated with the onset of pre-eclampsia.This review will summarize our current understanding of molecular structure of sFlt-1,signal pathways,with an emphasis on its role in the pathogenesis of preeclampsia and regulative factors.
Estradiol Promotes Trophoblast Viability and Invasion by Activating SGK1

Abstract:Background: 17 beta-Estradiol (E2) is a critical regulator of trophoblast function during pregnancy. Serum-and glucocorticoid-inducible kinase (SGK1) has been shown to regulate specific cellular targets downstream of E2. However, whether and how SGK1 directly mediates the regulatory effects of E2 on trophoblasts functions remain unknown. Methods: SGK1 expression in human villous samples and serum E2 levels were measured in women with early pregnancy loss (EPL) and healthy pregnant women. The effect of E2 on SGK1 regulation was assessed using luciferase reporter gene assay and Chromatin Immunoprecipitation assay. The mediation of regulatory effects of E2 by SGK1 on trophoblast functions including cell viability, invasion and related signaling molecules such as B cell leukemia/lymphoma 6, E-cadherin, matrix metalloproteinase 2, alpha-ENaC, vascular endothelial growth factor, and the phosphorylation status of FOXO1 and AKT were evaluated in HTR8/SVneo cells transfected with SGK1 knockdown plasmid with/without E2 treatment. Results: SGK1 protein levels in human villous samples and serum E2 levels were decreased in patients with EPL compared to controls. E2 (10 nM) increased SGK1 promoter activity directly through estrogen receptor. E2-activated SGK1 enhanced cell viability, invasion and downstream targets in trophoblast cells. SGK1 knockdown abrogated the above responses to E2 treatment. Conclusions: SGK1 mediates the effects of E2 on trophoblast viability and invasion, suggesting that SGK1 acts as a key node in regulating the cross-talk at the feto-maternal interface during the development of placenta and might be a potential therapeutic target for EPL.
Placental growth factor mediates pathological uterine angiogenesis by activating the NFAT5-SGK1 signaling axis in the endometrium: implications for preeclampsia development

Abstract:After menstruation the uterine spiral arteries are repaired through angiogenesis. This process is tightly regulated by the paracrine communication between endometrial stromal cells (EnSCs) and endothelial cells. Any molecular aberration in these processes can lead to complications in pregnancy including miscarriage or preeclampsia (PE). Placental growth factor (PlGF) is a known contributing factor for pathological angiogenesis but the mechanisms remain poorly understood. In this study, we investigated whether PlGF contributes to pathological uterine angiogenesis by disrupting EnSCs and endothelial paracrine communication. We observed that PlGF mediates a tonicity-independent activation of nuclear factor of activated T cells 5 (NFAT5) in EnSCs. NFAT5 activated downstream targets including SGK1, HIF-1α and VEGF-A. In depth characterization of PlGF - conditioned medium (CM) from EnSCs using mass spectrometry and ELISA methods revealed low VEGF-A and an abundance of extracellular matrix organization associated proteins. Secreted factors in PlGF-CM impeded normal angiogenic cues in endothelial cells (HUVECs) by downregulating Notch-VEGF signaling. Interestingly, PlGF-CM failed to support human placental (BeWo) cell invasion through HUVEC monolayer. Inhibition of SGK1 in EnSCs improved angiogenic effects in HUVECs and promoted BeWo invasion, revealing SGK1 as a key intermediate player modulating PlGF mediated anti-angiogenic signaling. Taken together, perturbed PlGF-NFAT5-SGK1 signaling in the endometrium can contribute to pathological uterine angiogenesis by negatively regulating EnSCs-endothelial crosstalk resulting in poor quality vessels in the uterine microenvironment. Taken together the signaling may impact on normal trophoblast invasion and thus placentation and, may be associated with an increased risk of complications such as PE.

Abstract:Preeclampsia is a syndrome characterized by hypertension during pregnancy, which is a leading cause of morbidity and mortality in both mother and newborn in developing countries. Some advances have increased the understanding of pathophysiology of this disease. For example, reduced utero-placental blood flow associated with impaired trophoblast invasion may lead to a hypoxic placenta that releases harmful materials into the maternal and feto-placental circulation and impairs endothelial function. Identification of these harmful materials is one of the hot topics in the literature, since these provide potential biomarkers. Certainty, such knowledge will help us to understand the miscommunication between mother and fetus. In this review we highlight how placental extracellular vesicles and their cargo, such as small RNAs (i.e., microRNAs), might be involved in endothelial dysfunction, and then in the angiogenesis process, during preeclampsia. Currently only a few reports have addressed the potential role of endothelial regulatory miRNA in the impaired angiogenesis in preeclampsia. One of the main limitations in this area is the variability of the analyses performed in the current literature. This includes variability in the size of the particles analyzed, and broad variation in the exosomes considered. The quantity of microRNA targets genes suggest that practically all endothelial cell metabolic functions might be impaired. More studies are required to investigate mechanisms underlying miRNA released from placenta upon endothelial function involved in the angiogenenic process.

Abstract:Objectives. To verify the hypothesis that vascular endothelial growth factor (VEGF) attenuates N omega-Nitro-L-arginine Methyl Ester (L-NAME)-induced preeclampsia-like manifestations in rats. Study design. Forty pregnant Wistar rats were randomly divided into four groups: control, preeclampsia model, VEGF treatment, and VEGF prophylactic. On day 5 of gestation, L-NAME was injected subcutaneously in rats of the preeclampsia model, VEGF treatment, and VEGF prophylactic groups. VEGF was given after the occurrence of hypertension and proteinuria in the VEGF treatment group and from day 5 in the VEGF prophylactic group. Blood pressure was monitored and urine protein was assayed. Blood platelet was counted, and serum nitric oxide metabolites, endothelin-1, 6-keto-PGF-1 alpha, and TXB(2) were determined. Results. Blood pressure increased significantly on day 8 of gestation in the preeclampsia model and VEGF treatment groups compared with control (p < 0.05 for both) and remained elevated through the pregnancy in the preeclampsia model group. Blood pressure was significantly decreased after the administration of VEGF in the VEGF treatment group (p < 0.05). There was no significant difference in blood pressure between the VEGF prophylactic group and control (p < 0.05). Urine protein, platelet count, serum nitric oxide metabolites, endothelin-1, 6-keto-PGF-1, and TXB2 were significantly different between control and the preeclampsia model group (p < 0.05), but not between control and the VEGF treatment or VEGF prophylactic groups (p > 0.05 for all). Conclusion. VEGF attenuates L-NAME-induced preeclampsia-like manifestations in rats, suggesting the important role of VEGF in preeclampsia and providing a potential strategy for the prevention and treatment of preeclampsia.
Decreased FGF23 inhibits placental angiogenesis via the ERK1/2-EGR-1 signaling pathway in preeclampsia

Abstract:PURPOSE: This study aimed to investigate the expression of fibroblast growth factor 23 (FGF23) in pregnant women with preeclampsia and elucidate its role in promoting placental angiogenesis through the ERK1/2-EGR-1 signaling pathway.METHODS: Serum FGF23 levels were measured by ELISA in healthy pregnant women and patients with preeclampsia during the first, second, and third trimesters of pregnancy. Wound healing, Transwell, and tube formation assays were performed to investigate the effects of FGF23 on cell migration, invasion and tube formation. The expression of vascular endothelial growth factor A (VEGF-A) and its upstream signaling molecules, p-ERK, and EGR-1, in placental tissues was detected by RT-qPCR and western blotting. Additionally, the effect of FGF23 on VEGF-A, p-ERK, and EGR-1 expression was further explored in vitro.RESULTS: Serum FGF23 levels increased with gestational age. During the third trimester, the control group exhibited a more pronounced increase in FGF23 levels than the preeclampsia group. Administering exogenous FGF23 promoted trophoblast cell migration, invasion and enhanced tube formation in vascular endothelial cells. The expression levels of VEGF-A, p-ERK, and EGR-1 in the placental tissues were significantly lower in the preeclampsia group than in the control group. In vitro experiments confirmed that FGF23 up-regulated VEGF-A expression through the p-ERK/EGR-1 signaling pathway.CONCLUSION: The serum level of FGF23 decreased in pregnant women with preeclampsia, inhibiting the ERK1/2-EGR-1 pathway and resulting in decreased expression of VEGF-A, thereby inhibiting placental angiogenesis. This could be a potential mechanism involved in the progression of preeclampsia.

Effect of Vascular Endothelial Growth Factor on Pre-Eclampsia in Pregnant Rats

Abstract:OBJECTIVE:To investigate relationship between the vascular endothelial growth factor (VEGF) and the pathogenesis of pre-eclampsia in pregnant rats.METHODS:Pregnant rats were divided into two groups randomly. Saline solution or L-nitro arginine methyl ester (L-NAME) 125 mg/d was given subcutaneously from day 7 of gestation till establishing pre-eclampsia. Systolic blood pressure, urine protein, platelet count, and weight of pups and placenta were determined. The levels of VEGF in pregnant rats venous serum, placenta and decidual tissue from normal pregnancy and pre-eclampsia rats were detected by ELISA and immunohistochemistry, respectively.RESULT:Pregnant rats which were given L-NAME produced physical signs similar to those of pre-eclampsia, such as increase in systolic blood pressure [(145.3 +/-4.6)mmHg] and urine protein [(814.3 +/-57.5)mg/L], and decrease in platelet count [(467.1 +/-76.3) x 10(9)/L] and weight of pups and placenta. Compared with controls, the intensity of VEGF immunostaining in trophoblast or decidual cells were significantly reduced. The serum levels of VEGF were significantly lower in pre-eclampsia group than in normal pregnancy.CONCLUSION:Decreased serum levels of VEGF and reduced expression of VEGF in placental tissues might in part explain the pathogenesis of pre-eclampsia in pregnant rats.

Abstract:Introduction: Pre-eclampsia (PE) is associated with lowered circulating levels of oestrogen. The effects of oestrogen are normally mediated by the classical oestrogen receptors (ERs). Intriguingly, a new oestrogen receptor, G-protein coupled receptor 30 (GPR30), has been recently found to play an important role in many oestrogenic effects. However, the potential role of GPR30 in PE remains poorly understood. Methods: Placentas, decidua and umbilical cord tissues were collected in 21 PE cases and following 20 uncomplicated pregnancies. Immunohistochemistry was used to detect the expression of GPR30 in the placental tissues. An in vitro cellular hypoxia/reoxygenation (H/R) model was established by using the immortalized human umbilical vein endothelial cell (HUVEC) line. Immunofluorescence was used to detect the expression of GPR30 and p-eNOS (Ser1177) protein. The expression of GPR30, PI3K/Akt pathway and p-eNOS (Ser1177)/eNOS proteins in HUVECs were assessed using Western blotting techniques. An in vitro tube formation assay and a migration assay were used to detect angiogenesis of HUVECs. The effect of GPR agonists and inhibitors was determined. Results: The expression of GPR30 in vascular endothelial cells of placentas, decidua and umbilical cord tissues was lower in women with pre-eclampsia compared to normal pregnant women. The selective GPR30 agonist G1 and a general oestrogen receptors agonist 17-β-estradiol (E2) both exerted a protective effect in hypoxia/reoxygenation (H/R) exposed HUVECs by up regulating NO production though the phosphoinositide 3-Kinase/Akt (PI3K/Akt) pathway. This effect was abolished by the selective GPR30 inhibitor G15. Conclusion: This study suggests that GPR30, as an upstream signaling molecule of PI3K/Akt/eNOS pathway, playing a critical role in the pathogenesis of PE. GPR30 also may be a potential therapeutic target for PE and other pregnancy complications that result from placental endothelial dysfunction.
Chronic Hypoxia Differentially Promotes VEGF- and FGF2-Stimulated Angiogenic Responses in HUAE Cells.

Abstract:During pregnancy, placental development occurs under physiological hypoxia (~2-8% O2), which is critical for placental angiogenesis. To examine mechanisms of chronic hypoxia governing placental angiogenesis, in this study, we determined if chronic hypoxia enhanced placental angiogenesis using human umbilical cord artery endothelial (HUAE) cells as a model. Methods: After isolation from umbilical cords of normal term pregnant patients (n = 3), HUAE cells were cultured under hypoxia (37°C, 5% CO2, 3% O2) and normoxia (37°C, 5% CO2, 95% air) respectively. After 20-25 days, cell phenotypes were verified. Cell proliferation and migration were evaluated by using the BrdU ELISA kit and BD FluoroBlok Insert system, respectively. After serum-starvation, cells derived under HYPOIXA & NORMOXIA were treated with VEGF or FGF2 for 16 hr under hypoxia & normoxia, respectively (designed as chronic hypoxia & normoxia respectively). Additional cells derived under HYPOXIA & NORMOXIA were pre-exposed to normoxia & hypoxia for 24 hr (acute normoxia & hypoxia), respectively, followed by treating cells in the same fashion. Results: The majority of cells exhibited positive uptake of DiI-Ac-LDL, CD31 expression, and formation of capillary-like tube structures. Both VEGF & FGF2 dose-dependently stimulated cell proliferation and migration under normoxia & hypoxia. As compared with chronic normoxia, chronic hypoxia further enhanced VEGF- & FGF2-stimulated cell proliferation and migration. However, acute hypoxia moderately promoted VEGF-, but not FGF2- stimulated cell proliferation in normoxia-derived cells, whereas acute normoxia did not attenuated VEGF - & FGF2-stimulated cell proliferation in hypoxia-derived cells. Acute hypoxia moderately promoted VEGF - & FGF2-stimulated cell migration in normoxia-derived cells, whereas acute normoxia attenuated VEGF - & FGF2-stimulated cell migration in hypoxia-derived cells. Conclusions: Chronic hypoxia further promotes VEGF- & FGF2-stimulated cell proliferation and migration in HUAE cells. Acute changes in O2 levels differentially modulate VEGF- & FGF2-stimulated cell proliferation and migration. These data suggest that chronic and acute hypoxia differentially modulates different steps of VEGF- & FGF2-induced placental angiogenesis. (poster)

Abstract:Background Preeclampsia is characterized by maternal endothelial activation and placental dysfunction. Imbalance in maternal angiogenic and vasoactive factors has been linked to the pathophysiology. The contribution of the placenta as a source to these factors remains unclear. Furthermore, little is known about fetal angiogenic and vasoactive proteins and the relation between maternal and fetal levels. Objectives We describe Placental growth factor (PGF), soluble Fms-like tyrosine kinase 1 (sFLT1), soluble Endoglin (sENG), as well as Endothelin 1, 2 and 3 (EDN1, EDN2 and EDN3) in five vessels in healthy pregnancies, early onset (EOPE) and late onset (LOPE) preeclampsia. Specifically, we aimed to 1) compare protein abundance in vessels at the maternal-fetal interface between EOPE, LOPE and healthy pregnancies, 2) describe placental uptake and release of proteins, and 3) describe protein abundance in the maternal vs fetal circulations. Methods Samples were collected from the maternal radial artery, uterine vein and antecubital vein as well as the fetal umbilical vein and artery in 75 healthy and 37 preeclamptic mother-fetus pairs (including 19 EOPE and 18 LOPE), during scheduled cesarean delivery. This method allows estimation of placental release and uptake of proteins by calculation of venoarterial differences on each side of the placenta. The microarray based SomaScan assay quantified the proteins. Results Abundance of sFLT1 and EDN1 was higher in the maternal vessels in preeclampsia as compared to healthy pregnancies, with highest abundance in EOPE. PGF was lower in the maternal vessels in EOPE as compared to both healthy and LOPE. Maternal EDN2 was higher in preeclampsia, with LOPE having the highest abundance. Our model confirmed placental release of PGF and sFLT1 to the maternal circulation in all groups. The placenta released sFLT1 into the fetal circulation in healthy and LOPE pregnancies. Fetal EDN1 and sFLT1 were higher in early onset preeclampsia, whereas sENG and EDN3 were lower in both preeclampsia groups, compared to healthy. Across groups, abundances of PGF, sFLT1 and EDN3 were higher in the maternal artery as compared to the fetal umbilical vein, whereas EDN2 was lower. Conclusion Increasing abundance of maternal sFLT1 and EDN1 across the groups healthy, LOPE and EOPE combined with a positive correlation may suggest that these proteins are associated with the pathophysiology and severity of the disease. Elevated EDN1 in the fetal circulation in EOPE represents a novel finding. Long term effects of altered protein abundance in preeclampsia on fetal development and health remain unknown. Further investigation of these proteins' involvement in the pathophysiology and as treatment targets is warranted.

Exosomal microRNA‐302a promotes trophoblast migration and proliferation, and represses angiogenesis by regulating the expression levels of VEGFA in preeclampsia

Abstract:The global morbidity rate of preeclampsia (PE) is 3‑7, and 10‑20% of maternal deaths are associated with PE. However, the mechanism of its pathogenesis remains unknown. The aim of the present study was to examine the relationship between microRNA‑302a (miR‑302a) and PE. Firstly, the relative expression levels of miR‑302a in placental tissues from patients with PE and normal controls were analyzed using reverse transcription‑quantitative PCR. miR‑302a expression was upregulated in PE tissues, particularly in severe PE. Subsequently, HTR‑8/SVneo cells were transfected with miR‑302a vectors to overexpress miR‑302a. The overexpression of miR‑302a markedly promoted cell proliferation, colony formation, migration and invasion in vitro. Subsequently, the present study examined the function of exosomes secreted by HTR‑8/SVneo cells transfected with miR‑302a vectors. Compared with the negative control vector group, miR‑302a expression was markedly increased in exosomes in the miR‑302a overexpression group. Additionally, exosomes with miR‑302a overexpression had repressed HUVEC invasion and ring formation. The luciferase reporter assay indicated that VEGFA was a direct target of miR‑302a, and miR‑302a expression was negatively correlated with VEGFA expression. In conclusion, the present results demonstrated that upregulation of miR‑302a may promote HTR‑8/SVneo cell proliferation, migration and invasion, and repress angiogenesis by targeting VEGFA, indicating that miR‑302a may be a potential target for the development of PE therapies.

AC092100.1 promotes angiogenesis in pre-eclampsia through YTHDC2/VEGFA signaling

Abstract:Aberrant long non-coding RNA (lncRNA) expression has been shown to be involved in the pathological process of pre-eclampsia (PE), yet only a small portion of lncRNAs has been characterized concerning the function and molecular mechanisms involved in PE. This study aimed to investigate the regulatory mechanism of the lncRNA AC092100.1 (AC092100.1) in angiogenesis in PE. In our study, bioinformatics analysis was performed to screen for differentially expressed lncRNAs between normal subjects and PE patients. The levels of AC092100.1 in placental tissues of patients with or without PE were validated using qRT-PCR. The effect of AC092100.1 overexpression on the proliferation, migration, and tube formation of human umbilical vein endothelial cells (HUVECs) was investigated. The binding of AC092100.1 and YT521-B homology domain-containing 2 (YTHDC2) was predicted and verified. The effect of AC092100.1/YTHDC2 on the expression of vascular endothelial growth factor-A (VEGFA) in HUVECs was determined. Finally, a PE mice model was conducted. Fetal mouse growth, the abundance of mesenchymal morphology markers, including hypoxia-inducible factor 1-alpha (HIF-1α), soluble fms-like tyrosine kinase-1 (sFlt-1), soluble endoglin (sEng), Slug, and Vimentin, and endothelial markers, including placental growth factor (PLGF), CD31, and vascular endothelial (VE)-cadherin, in placental tissues were assessed. Here, we found that AC092100.1 was abnormally downregulated in placental tissues from PE patients. We established that AC092100.1 overexpression promoted HUVEC proliferation, migration, and tube formation in vitro. Mechanistically, AC092100.1 induced the accumulation of YTHDC2 and VEGFA through binding to YTHDC2 in HUVECs. Inhibition of YTHDC2 or VEGFA reversed AC092100.1-promoted tube formation. AC092100.1 overexpression contributed to alleviating fetal growth disorder, decreased levels of sEng, HIF-1α, sFlt-1, Slug, and Vimentin, and increased levels of VEGFA, PLGF, CD31, and VE-cadherin in PE mice. Our findings provided evidence supporting the role of the AC092100.1/YTHDC2/VEGFA axis in regulating angiogenesis, which demonstrated a therapeutic pathway for PE targeting angiogenesis.

Abstract:Normal pregnancy is associated with adaptive hemodynamic, hormonal, and vascular changes, and estrogen (E2) may promote vasodilation during pregnancy; however, the specific E2 receptor (ER) subtype, post-ER signaling mechanism, and vascular bed involved are unclear. We tested whether pregnancy-associated vascular adaptations involve changes in the expression/distribution/activity of distinct ER subtypes in a blood vessel-specific manner. Blood pressure (BP) and plasma E2 were measured in virgin and pregnant (day 19) rats, and the thoracic aorta, carotid artery, mesenteric artery, and renal artery were isolated for measurements of ERα, ERβ, and G protein-coupled receptor 30 [G protein-coupled ER (GPER)] expression and tissue distribution in parallel with relaxation responses to E2 (all ERs) and the specific ER agonist 4,4',4″-(4-propyl-[1H]-pyrazole-1,3,5-triyl)-tris-phenol (PPT; ERα), diarylpropionitrile (DPN; ERβ), and G1 (GPER). BP was slightly lower and plasma E2 was higher in pregnant versus virgin rats. Western blots revealed increased ERα and ERβ in the aorta and mesenteric artery and GPER in the aorta of pregnant versus virgin rats. Immunohistochemistry revealed that the increases in ERs were mainly in the intima and media. In phenylephrine-precontracted vessels, E2 and PPT caused relaxation that was greater in the aorta and mesenteric artery but similar in the carotid and renal artery of pregnant versus virgin rats. DPN- and G1-induced relaxation was greater in the mesenteric and renal artery than in the aorta and carotid artery, and aortic relaxation to G1 was greater in pregnant versus virgin rats. The nitric oxide synthase inhibitor N(ω)-nitro-l-arginine methyl ester with or without the cyclooxygenase inhibitor indomethacin with or without the EDHF blocker tetraethylammonium or endothelium removal reduced E2, PPT, and G1-induced relaxation in the aorta of pregnant rats, suggesting an endothelium-dependent mechanism, but did not affect E2-, PPT-, DPN-, or G1-induced relaxation in other vessels, suggesting endothelium-independent mechanisms. E2, PPT, DPN, and G1 caused relaxation of Ca(2+) entry-dependent KCl contraction, and the effect of PPT was greater in the mesenteric artery of pregnant versus virgin rats. Thus, during pregnancy, an increase in ERα expression in endothelial and vascular smooth muscle layers of the aorta and mesenteric artery is associated with increased ERα-mediated relaxation via endothelium-derived vasodilators and inhibition of Ca(2+) entry into vascular smooth muscle, supporting a role of aortic and mesenteric arterial ERα in pregnancy-associated vasodilation. GPER may contribute to aortic relaxation while enhanced ERβ expression could mediate other genomic vascular effects during pregnancy.

Abstract:Uterine blood flow is elevated during high estrogen states such as pregnancy. We have previously reported that estrogen results in significant vasodilatation and an increase in uterine blood flow during late gestation. However, its role in uterine angiogenesis is less well studied. Moreover, it is unknown if estradiol 17β (E2β) metabolites which are elevated during pregnancy and are synthesized by cytochrome (CYP) P450 1A1 (CYP1A1), 1A2 (CYP1A2), 1B1 (CYP1B1), and catechol-O-methyltransferase (COMT) to form 2-hydroxyestradiol (2-OHE), 4-hydroxyestradiol (4-OHE), 2-methoxestradiol (2-ME), and 4-methoxyestradiol (4-ME) respectively play a role in modulation of uterine angiogenesis and/or blood flow. In the present study, we tested the hypothesis that E2β, its hydroxylated as well as its methoxylated metabolites, will have an angiogenic role in late gestation uterine artery endothelial cells (UAECs) and that the respective enzymes that metabolize E2β are present in these cells. Validated UAECs from late pregnant sheep (day 120-130; term= 147 days; n=5) were treated with vehicle (control, basal media) or increasing concentrations (0.1-100nM) of E2β, 2-OHE, 4-OHE and 2-ME. An In vitro index of cell proliferation was evaluated utilizing the BrdU Proliferation Assay technique following the subsequent 24 hrs of treatment. Furthermore, the presence of the CYP and COMT enzymes was evaluated by immunofluorescent staining and multi-photon confocal imaging. E2βtreatment significantly induced UAEC proliferation (P< 0.0001). Pos-hoc tests revealed maximum response at a physiological E2β concentration of 0.1 nM (P< 0.001, magnitude of response, 2.67 + 0.23 fold of control). Treatment with 2-OHE, and 4-OHE resulted in equivalent significant increases in proliferation (P< 0.0001; maximal responses 1.79 + 0.06 fold of control). In contrast, 2-ME resulted in only a minor increases in UAEC proliferation levels (P= 0.0037). Post hoc tests revealed maximum response at 0.1 nM 2-ME (P < 0.05; maximal response at 1.30 + 0.02 fold of control). Multi photon confocal imaging revealed the presence of CYP1A2, CYP1B1 and COMT, in the cytoplasmic domain of these cells. However, we did not detect CYP1A1. These data demonstrate a potential role of E2β and its metabolites in regulating uterine angiogenesis. These data show for the first time, the presence of estrogen metabolizing enzymes in the uterine artery endothelial cells. Collectively, our findings are suggestive that E2β, its hydroxylated and methoxylated metabolites, may play an important role in maternal uterine adaptation to pregnancy through participation in uterine angiogenesis. Abstract:VEGFR2 (Vascular endothelial growth factor receptor 2) is a central regulator of placental angiogenesis. The study of the VEGFR2 proteome of chorionic villi at term revealed its partners MDMX (Double minute 4 protein) and PICALM (Phosphatidylinositol-binding clathrin assembly protein). Subsequently, the oxytocin receptor (OT-R) and vasopressin V1aR receptor were detected in MDMX and PICALM immunoprecipitations. Immunogold electron microscopy showed VEGFR2 on endothelial cell (EC) nuclei, mitochondria, and Hofbauer cells (HC), tissue-resident macrophages of the placenta. MDMX, PICALM, and V1aR were located on EC plasma membranes, nuclei, and HC nuclei. Unexpectedly, PICALM and OT-R were detected on EC projections into the fetal lumen and OT-R on 20–150 nm clusters therein, prompting the hypothesis that placental exosomes transport OT-R to the fetus and across the blood–brain barrier. Insights on gestational complications were gained by univariable and multivariable regression analyses associating preeclampsia with lower MDMX protein levels in membrane extracts of chorionic villi, and lower MDMX, PICALM, OT-R, and V1aR with spontaneous vaginal deliveries compared to cesarean deliveries before the onset of labor. We found select associations between higher MDMX, PICALM, OT-R protein levels and either gravidity, diabetes, BMI, maternal age, or neonatal weight, and correlations only between PICALM-OT-R ( p < 2.7 × 10 –8 ), PICALM-V1aR ( p < 0.006), and OT-R-V1aR ( p < 0.001). These results offer for exploration new partnerships in metabolic networks, tissue-resident immunity, and labor, notably for HC that predominantly express MDMX.

Abstract:Hypertension, Ahead of Print. BACKGROUND:Preeclampsia is a significant pregnancy disorder with an unknown cause, mainly attributed to impaired spiral arterial remodeling.METHODS:Using RNA sequencing, we identified key genes in placental tissues from healthy individuals and preeclampsia patients. Placenta and plasma samples from pregnant women were collected to detect the expression of TPBG (trophoblast glycoprotein). Pregnant rats were injected with TPBG-carrying adenovirus to detect preeclamptic features. HTR-8/SVneo cells transfected with a TPBG overexpression lentiviral vector were used in cell function experiments. The downstream molecular mechanisms of TPBG were explored using RNA sequencing and single-cell RNA sequencing data. TPBG expression was knocked down in the lipopolysaccharide-induced preeclampsia-like rat model to rescue the preeclampsia features. We also assessed TPBG's potential as an early preeclampsia predictor using clinical plasma samples.RESULTS:TPBG emerged as a crucial differentially expressed gene, expressed specifically in syncytiotrophoblasts and extravillous trophoblasts. Subsequently, we established a rat model with preeclampsia-like phenotypes by intravenously injecting TPBG-expressing adenoviruses, observing impaired spiral arterial remodeling, thus indicating a causal correlation between TPBG overexpression and preeclampsia. Studies with HTR-8/SVneo cells, chorionic villous explants, and transwell assays showed TPBG overexpression disrupts trophoblast/extravillous trophoblast migration/invasion and chemotaxis. Notably, TPBG knockdown alleviated the lipopolysaccharide-induced preeclampsia-like rat model. We enhanced preeclampsia risk prediction in early gestation by combining TPBG expression with established clinical predictors.CONCLUSIONS:These findings are the first to show that TPBG overexpression contributes to preeclampsia development by affecting uterine spiral artery remodeling. We propose TPBG levels in maternal blood as a predictor of preeclampsia risk. The proposed mechanism by which TPBG overexpression contributes to the occurrence of preeclampsia via its disruptive effect on trophoblast and extravillous trophoblast migration/invasion on uterine spiral artery remodeling, thereby increasing the risk of preeclampsia.

Abstract:Preeclampsia (PE), a severe pregnancy-specific syndrome, is characterized by impaired placental angiogenesis. Although the pathogenesis of this condition remains largely unclear, vascular systemic endothelial injury is thought to be the common contributing factor. Soluble Axl (sAxl), a biomarker of endothelial dysfunction, is known to be abnormally increased in a variety of diseases associated with vascular injury. In a previous study, we found that the plasma levels of sAxl were significantly higher in PE with severe features (sPE) than in pregnant women who did not have PE. The current study aimed to further explore the potential role of sAxl in vascular injury in patients with sPE. We found that the upregulation of sAxl in maternal plasma was positively correlated with the plasma levels of sFlt-1 and negatively correlated with placental NO synthase (eNOS) in women with sPE. Furthermore, elevated levels of sAxl suppressed proliferation and endothelial tube formation and promoted cytotoxicity in human umbilical vein endothelial cells (HUVECs) through the downregulation of p-Akt, p-p70S6K, p-mTOR, and Grb2. Subsequently, we established a pregnant rat model with PE-like characteristics by injecting pregnant rats with an adenovirus expressing sAxl. These rats exhibited a typical PE-like phenotype, including increased blood pressure, proteinuria, and fetal growth restriction, along with abnormal placental and fetal renal morphology. In conclusion, our study demonstrated the role of sAxl in systemic vascular injury through the regulation of the expression of key molecules of angiogenesis and described its potential contribution to the development of sPE.

Abstract:Estradiol-17β (E 2 β) and its metabolites, which are sequentially synthesized by cytochrome P450s and catechol- O -methyltransferase to form 2 and 4-hydroxyestradiol (OHE 2 ) and 2- and 4-methoxestradiol (ME 2 ), are elevated during pregnancy. We investigated whether cytochrome P450s and catechol- O -methyltransferase are expressed in uterine artery endothelial cells (UAECs) and whether E 2 β and its metabolites modulate cell proliferation via ER-α and/or ER-β and play roles in physiological uterine angiogenesis during pregnancy. Cultured ovine UAECs from pregnant and nonpregnant ewes were treated with 0.1 to 100.0 nmol/L of E 2 β, 2-OHE 2 , 4-OHE 2 , 2-ME 2 , and 4-ME 2 . ER-α or ER-β specificity was tested using ICI 182 780, ER-α–specific 1,3-bis(4-hydroxyphenyl)-4-methyl-5-[4-(2-piperidinyleth oxy)phenol]-1H-pyrazole dihydrochloride, ER-β–specific 4-[2-phenyl-5,7-bis(trifluoromethyl)pyrazolo [1,5-a]pyrim idin-3-yl]phenol antagonists and their respective agonists ER-α–specific 4,4′,4"-(4-propyl-[1H]-pyrazole-1,3,5-triyl)trisphenol and ER-β–specific 2,3-bis(4-Hydroxyphenyl)-propionitrile. Angiogenesis was evaluated using 5-bromodeoxyuridine proliferation assay. Using confocal microscopy and Western analyses to determine enzyme location and levels, we observed CYP1A1, CYP1A2, CYP1B1, CYP3A4, and catechol- O -methyltransferase expression in UAECs; however, expressions were similar between nonpregnant UAECs and pregnant UAECs. E 2 β, 2-OHE 2 , 4-OHE 2 , and 4-ME 2 treatments concentration-dependently stimulated proliferation in pregnant UAECs but not in nonpregnant UAECs; 2-ME 2 did not stimulate proliferation in either cell type. Proliferative responses of pregnant UAECs to E 2 β were solely mediated by ER-β, whereas responses to E 2 β metabolites were neither ER-α nor ER-β mediated. We demonstrate an important vascular role for E 2 β, its cytochrome P450- and catechol- O -methyltransferase–derived metabolites, and ER-β in uterine angiogenesis regulation during pregnancy that may be dysfunctional in preeclampsia and other cardiovascular disorders.
