Abstract:Background: The conditions of space flight have a significant effect on the physiological processes in the human body, yet the molecular mechanisms driving physiological changes remain unknown. Methods: Blood samples of 18 Russian cosmonauts who had conducted long-duration missions to the International Space Station were collected 30 days before launch and on the first and seventh days after landing. Results: A panel of 125 proteins in the blood plasma was quantitated by a well-established and highly regarded targeted mass spectrometry approach. This method involves the monitoring of multiple reactions in conjunction with stable isotope-labeled standards at the University of Victoria - Genome BC Proteomics Centre. Conclusions: Reduction of circulating plasma volume during space flight and activation of fluid retention at the final stage of the flight affect the changes in plasma protein concentrations present in the first days after landing. Using an ANOVA approach, it was revealed that only 1 protein (S100A9) reliably responded to space flight conditions. This protein plays an important role in the functioning of the endothelium and can serve as a marker for activation of inflammatory reactions. Concentrations of the proteins of complement, coagulation cascades, and acute phase reactants increase in the blood of cosmonauts as measured the first day after landing. Most of these proteins' concentrations continue to increase by the 7th day after space flight. Similar dynamics are observed for proteases and their inhibitors. Thus, there is a shift in proteolytic blood systems, which is necessary for the restoration of muscle tissue and maintenance of oncotic homeostasis.

Abstract:Maintenance of astronaut health during spaceflight will require monitoring and potentially modulating their microbiomes. However, documenting microbial shifts during spaceflight has been difficult due to mission constraints that lead to limited sampling and profiling. Here we executed a six-month longitudinal study to quantify the high-resolution human microbiome response to three days in orbit for four individuals. Using paired metagenomics and metatranscriptomics alongside single-nuclei immune cell profiling, we characterized time-dependent, multikingdom microbiome changes across 750 samples and 10 body sites before, during and after spaceflight at eight timepoints. We found that most alterations were transient across body sites; for example, viruses increased in skin sites mostly during flight. However, longer-term shifts were observed in the oral microbiome, including increased plaque-associated bacteria (for example, Fusobacteriota ), which correlated with immune cell gene expression. Further, microbial genes associated with phage activity, toxin–antitoxin systems and stress response were enriched across multiple body sites. In total, this study reveals in-depth characterization of microbiome and immune response shifts experienced by astronauts during short-term spaceflight and the associated changes to the living environment, which can help guide future missions, spacecraft design and space habitat planning.

Abstract:Spaceflight entails a variety of environmental and psychological stressors that may have long-term physiological and genomic consequences. Metabolomics, an approach that investigates the terminal metabolic outputs of complex physiological alterations, considers the dynamic state of the human body and allows the identification and quantification of down-stream metabolites linked to up-stream physiological and genomic regulation by stress. Employing a metabolomics-based approach, this study investigated longitudinal metabolic perturbations of male ( n = 40) and female ( n = 11) astronauts on 4–6-month missions to the International Space Station (ISS). Proton nuclear magnetic resonance ( 1 H-NMR) spectroscopy followed by univariate, multivariate and machine learning analyses were used on blood serum to examine sex-specific metabolic changes at various time points throughout the astronauts' missions, and the metabolic effects of long-duration space travel. Space travel resulted in sex-specific changes in energy metabolism, bone mineral and muscle regulation, immunity, as well as macromolecule maintenance and synthesis. Additionally, metabolic signatures suggest differential metabolic responses—especially during the recovery period—with females requiring more time to adjust to return to Earth. These findings provide insight into the perturbations in glucose and amino acid metabolism and macromolecule biosynthesis that result from the stressors of long-duration spaceflight. Metabolomic biomarkers may provide a viable approach to predicting and diagnosing health risks associated with prolonged space travel and other physiological challenges on Earth.

Characterization of a spontaneously occurring arthritis in male DBA/1 mice.

Abstract:OBJECTIVE We studied the incidence, severity, histopathologic features, and status of infection in a spontaneous polyarthritis which occurs in male DBA/1 mice. METHODS Over a 25-week period, the arthritis was evaluated macroscopically in naive mice of different strains. The histopathology was evaluated at different phases of the disease. Virologic and bacteriologic investigations were performed. RESULTS The arthritis occurs in approximately 80% of aging male DBA/1 mice. The disease is chronic and destructive, and the affected joints are characterized by synovial lining proliferation and infiltration of inflammatory cells, mainly mononuclear. The susceptibility appears to be dependent on both sex-linked and non-sex-linked genes, since female DBA/1 mice are not affected and since male BALB/c housed in the same colony are only marginally affected. CONCLUSION This first description of a spontaneous arthritis in aging male DBA/1 mice should provide new opportunities to study general features of a chronic and intermittent arthritis in a well-characterized strain, in which no generalized aberrations of the immune system have been described.

Abstract:The molecular mechanisms associated with spaceflight‐induced biological adaptations that may affect many healthy tissue functions remain poorly understood. In this study, we analyzed temporal changes in the serum proteome of six astronauts during prolonged spaceflight missions using quantitative comprehensive proteome analysis performed with the data‐independent acquisition method of mass spectrometry (DIA‐MS). All six astronauts participated in a spaceflight mission for approximately 6 months and showed a decreasing trend in T‐scores at almost all sites where dual‐energy X‐ray absorptiometry scans were performed. DIA‐MS successfully identified 624 nonredundant proteins in sera and further quantitative analysis for each sampling point provided information on serum protein profiles closely related to several time points before (pre‐), during (in‐), and after (post‐) spaceflight. Changes in serum protein levels between spaceflight and on the ground suggest that abnormalities in bone metabolism are induced in astronauts during spaceflight. Furthermore, changes in the proteomic profile occurring during spaceflight suggest that serum levels of bone metabolism‐related proteins, namely ALPL, COL1A1, SPP1, and POSTN, could serve as highly responsive indicators of bone metabolism status in spaceflight missions. This study will allow us to accelerate research to improve our understanding of the molecular mechanisms of biological adaptations associated with prolonged spaceflight.

Abstract:Over the course of a mission to the International Space Station (ISS) crew members are exposed to a number of stressors that can potentially alter the composition of their microbiomes and may have a negative impact on astronauts' health. Here we investigated the impact of long-term space exploration on the microbiome of nine astronauts that spent six to twelve months in the ISS. We present evidence showing that the microbial communities of the gastrointestinal tract, skin, nose and tongue change during the space mission. The composition of the intestinal microbiota became more similar across astronauts in space, mostly due to a drop in the abundance of a few bacterial taxa, some of which were also correlated with changes in the cytokine profile of crewmembers. Alterations in the skin microbiome that might contribute to the high frequency of skin rashes/hypersensitivity episodes experienced by astronauts in space were also observed. The results from this study demonstrate that the composition of the astronauts' microbiome is altered during space travel. The impact of those changes on crew health warrants further investigation before humans embark on long-duration voyages into outer space.

Characterization of two species of Fc fragments obtained from rat IgG2a by prolonged papain digestion.

Abstract:Prolonged papain digestion of rat IgG2a produced two molecular species of Fc fragments, termed Fc(I) and Fc(II). Studies by gel filtration chromatography and polyacrylamide gel electrophoresis in SDS/urea indicated that the two subunit polypeptide chains in each Fc preparation were associated by non-covalent bonds only. By analytical ultracentrifugation Fc(I) was found to have a m.w. of 47,100 and a sedimentation coefficient of 4.08S. Fc(II) had a m.w. of 39,800 and a sedimentation coefficient of 3.83S. The m.w. for the subunit chains of Fc(I) and Fc(II) were 25,300 and 20,300, respectively, as determined by analytical ultracentrifugation under dissociating conditions. Calculation of the frictional coefficient ratios indicated that both Fc fragments possessed compact globular structures. The difference in size between these two Fc fragments probably was due to a loss of some carboxy-terminal residues in Fc(II). Both Fc fragments possessed nearly identical amino-terminal amino acid sequences. Papain cleavage occurred primarily between residues 233/234 and 234/235. The carbohydrate compositions of the two species of Fc fragments were similar. It was concluded that under acid and reducing conditions papain cleavage of rat IgG2a occurred to the carboxy-terminal side of the hinge region. Prolonged papain digestion led to secondary attack in the carboxy-terminal end of the CGAMMA3 domain at an unidentified site, or sites, producing a stable second species of Fc fragments.
Correlation Between Invariable Blood Proteins and Heart Rate Variability in Long-Duration Space Flights

Abstract:The article analyzes how long-duration space missions' effect on the heart rate variability parameters and invariable blood proteins. The results are discussed taking into correlation between them. Seven Russian cosmonauts took part in the research during their missions to the International Space Station. Samples of dry blood drops were collected as part of the space experiment ''OMICs-DBS'', electrocardiogram samples were collected as part of the space experiment "Cardiovector". We have established a linear relationship between the concentrations of the following proteins: complement C1q subcomponent subunit A (encoded by the C1QA gene), complement C1r subcomponent (encoded by the C1R gene), fibrinogen gamma chain (encoded by the FGG gene),galectin-3 (encoded by the LGALS3 gene), interstitial collagenase or matrix metalloproteinase-1 (encoded by the MMP-1 gene), pigment epithelium-derived factor (encoded by the PEDF gene) and frequency-domain heart rate variability (HRV) parameters at some stages of space flight. Three proteins were associated with of total power parameters, and either positively correlated with the low-frequency (LF) domain as in the case of the C1QA (complement C1q subcomponent subunit A) or negatively - LGALS3, MMP-1 (galectin-3, matrix metalloproteinase-1) correlated with the high-frequency domain (HF). One of the proteins, the PEDF (pigment epithelium-derived factor), positively correlated with the HF, which correspondingly reflected the effect of vagal modulation on the SA node. The Complement C1r subcomponent had positive correlations with both HF and LF. The FGG (fibrinogen gamma chain) was negatively correlated with both individual components of the frequency-domain (HF, ms2 and LF ms2) also its total power. We assume that such statistical relationships reflect the tension of regulatory mechanisms, which is consistent with classical studies of autonomic regulation in space flight.

Abstract:The analysis of exhaled breath condensate (EBC) demonstrates a promising avenue of minimally invasive biopsies for diagnostics. EBC is obtained by cooling exhaled air and collecting the condensation to be utilized for downstream analysis using various analytical methods. The aqueous phase of breath contains a large variety of miscible small compounds including polar electrolytes, amino acids, cytokines, chemokines, peptides, small proteins, metabolites, nucleic acids, and lipids/eicosanoids-however, these analytes are typically present at minuscule levels in EBC, posing a considerable technical challenge. Along with recent improvements in devices for breath collection, the sensitivity and resolution of liquid chromatography coupled to online mass spectrometry-based proteomics has attained subfemtomole sensitivity, vastly enhancing the quality of EBC sample analysis. As a result, proteomics analysis of EBC has been expanding the field of breath biomarker research. We present an au courant overview of the achievements in proteomics of EBC, the advancement of EBC collection devices, and the current and future applications for EBC biomarker analysis.

Abstract:The International Space Station (ISS) allows research to be done in low earth orbit (LEO), offering a uniquely accelerating environment to model disease and study human health. Studies done in microgravity can simulate models of stress, inflammation, aging, immune dysfunction, and pre-malignant transformation of stem cells. The NASA Twins Study suggests that extended periods of spaceflight may affect hematopoietic stem cell and immune cell health and function through their multi-omics approach of one ground-based twin, Mark Kelly, and one on board the ISS, Scott Kelly (Garrett-Bakelman et al., Science, 2019). A recent study also suggests an adaptive response to immune cell activity related to pre- and post-flight long-term missions after transcriptomic analysis of astronaut blood samples (Stratis, et al., Frontiers in Immunology, 2023). These studies emphasize the importance of assessing astronaut hematopoietic stem cell and immune cell function to further understand the mechanisms of cellular and molecular changes associated with time spent in LEO. We designed a longitudinal study to collect (under an IRB-approved protocol) peripheral blood from Axiom Mission 2 (Ax-2) astronauts to assess their hematopoietic stem cell and immune cell health. Timepoints include Launch-45 days, Launch-2 days, inflight, Return+1 day, Return+50 days, and yearly for 5 years. Ax-2 was a 10-day mission which launched on May 21, 2023 and splashed down on May 31, 2023. This was a private crew-tended mission with a sample size of 4 astronauts. As part of the stem cell health and fitness assessment, we selected for CD34+ hematopoietic stem and progenitor cells (HSPCs) from the 4 private astronauts and performed functional survival and self-renewal assays from each timepoint. Preliminary results suggest that the self-renewal capacity of HSPCs decreases inflight, but upon Return+1 day, HSPCs regain their ability to clone themselves at a higher capacity than pre-flight. Furthermore, we collected HSPCs for whole genome sequencing, 10x Genomics single cell sequencing, and whole transcriptome sequencing, to analyze pre-, during, and post-flight genomic and transcriptomic changes. Analyses will be done to identify mutational burden and mutational signatures associated with each timepoint. These mutational signatures will be compared to previously identified C-to-T mutations to assess the pre-malignant status of HSPCs due to short-term spaceflight (Jiang et al., Cell Reports, 2021). In addition to assessing CD34+ HSPC health, we will also profile the immune landscape by sequencing in parallel the CD34- fraction and performing targeted FACS analysis based on initial sequencing results. We also seek to identify markers of inflammation and anti-viral response through analysis of cytokines from biobanked plasma. This investigation has the potential to provide valuable insight into stem cell and immune cell health and fitness associated with short-duration missions in LEO.

Abstract:SUMMARYThe human microbiota encompasses the diverse communities of microorganisms that reside in, on, and around various parts of the human body, such as the skin, nasal passages, and gastrointestinal tract. Although research is ongoing, it is well established that the microbiota exert a substantial influence on the body through the production and modification of metabolites and small molecules. Disruptions in the composition of the microbiota-dysbiosis-have also been linked to various negative health outcomes. As humans embark upon longer-duration space missions, it is important to understand how the conditions of space travel impact the microbiota and, consequently, astronaut health. This article will first characterize the main taxa of the human gut microbiota and their associated metabolites, before discussing potential dysbiosis and negative health consequences. It will also detail the microbial changes observed in astronauts during spaceflight, focusing on gut microbiota composition and pathogenic virulence and survival. Analysis will then turn to how astronaut health may be protected from adverse microbial changes via diet, exercise, and antibiotics before concluding with a discussion of the microbiota of spacecraft and microbial culturing methods in space. The implications of this review are critical, particularly with NASA's ongoing implementation of the Moon to Mars Architecture, which will include weeks or months of living in space and new habitats.
Aging and putative frailty biomarkers are altered by spaceflight

Abstract:Human space exploration poses inherent risks to astronauts' health, leading to molecular changes that can significantly impact their well-being. These alterations encompass genomic instability, mitochondrial dysfunction, increased inflammation, homeostatic dysregulation, and various epigenomic changes. Remarkably, these changes bear similarities to those observed during the aging process on Earth. However, our understanding of the connection between these molecular shifts and disease development in space remains limited. Frailty syndrome, a clinical syndrome associated with biological aging, has not been comprehensively investigated during spaceflight. To bridge this knowledge gap, we leveraged murine data obtained from NASA's GeneLab, along with astronaut data gathered from the JAXA and Inspiration4 missions. Our objective was to assess the presence of biological markers and pathways related to frailty, aging, and sarcopenia within the spaceflight context. Through our analysis, we identified notable changes in gene expression patterns that may be indicative of the development of a frailty-like condition during space missions. These findings suggest that the parallels between spaceflight and the aging process may extend to encompass frailty as well. Consequently, further investigations exploring the utility of a frailty index in monitoring astronaut health appear to be warranted.

Abstract:Abstract Introduction Inhalation is an important route wherethrough occupational exposure can reach and potentially affect pulmonary function. Recently, there is a shift towards using less invasive matrices. We investigated quality and stability of exhaled breath condensate (EBC) as a minimally invasive matrix to identify lung-related biomarkers. Methods EBC and lung function measurements were collected, from twelve healthy participants twice six months apart (T1,T2) using Medivac® TurboDECCS System and spirometry. Cytokines, MMP-1, 2, 7, and 9, VEGF/Flt1, E- and P-selectin, α-amylase activity, proteomics, CysLTs, LTB4, and TXB2 were measured in EBC. This study was approved by the Ethics Committee Research UZ/KU Leuven (S64599). Results Only two out of twenty-four EBC samples exhibited detectable α-amylase activity. Proteomics showed presence of proteins involved in several biological processes, originating from different cellular components, with different molecular functions. Negative associations were shown between forced expiratory flow (FEF) 25% and TXB2(T2), and FEF 75% and forced expiratory volume within the first second (FEV1) and IFNγ(T1). Peak expiratory flow positively associated with CysLTs(T1) and IL6(T2), FEF 25% with CysLTs(T1), FEF 75% with IL8(T1), FEV1 with IL2 and IL6(T2), and forced vital capacity with IL1β, IL2, and IL6(T2). Discussion With only limited saliva contamination, the collection procedure for EBC is robust. Several associations were shown between spirometry parameters and various markers, suggesting the potential of EBC as alternative minimally invasive matrix. Conclusion So far, these results support the usage of EBC to identify biomarkers for lung function. Genome and clonal hematopoiesis stability contrasts with immune, cfDNA, mitochondrial, and telomere length changes during short duration spaceflight

Abstract:Abstract Background The Inspiration4 (I4) mission, the first all-civilian orbital flight mission, investigated the physiological effects of short-duration spaceflight through a multi-omic approach. Despite advances, there remains much to learn about human adaptation to spaceflight's unique challenges, including microgravity, immune system perturbations, and radiation exposure. Methods To provide a detailed genetics analysis of the mission, we collected dried blood spots pre-, during, and post-flight for DNA extraction. Telomere length was measured by quantitative PCR, while whole genome and cfDNA sequencing provided insight into genomic stability and immune adaptations. A robust bioinformatic pipeline was used for data analysis, including variant calling to assess mutational burden. Result Telomere elongation occurred during spaceflight and shortened after return to Earth. Cell-free DNA analysis revealed increased immune cell signatures post-flight. No significant clonal hematopoiesis of indeterminate potential (CHIP) or whole-genome instability was observed. The long-term gene expression changes across immune cells suggested cellular adaptations to the space environment persisting months post-flight. Conclusion Our findings provide valuable insights into the physiological consequences of short-duration spaceflight, with telomere dynamics and immune cell gene expression adapting to spaceflight and persisting after return to Earth. CHIP sequencing data will serve as a reference point for studying the early development of CHIP in astronauts, an understudied phenomenon as previous studies have focused on career astronauts. This study will serve as a reference point for future commercial and non-commercial spaceflight, low Earth orbit (LEO) missions, and deep-space exploration.

Abstract:According to the existing scenarios of interplanetary missions, the Moon is considered as an intermediate base on the way to deep space. In order to prepare for landing and work on the Moon, it is important to study the applicability of countermeasures in such missions. The paper presents the results of a pilot study performed during a short-term spaceflight (12 days). A new experience of using countermeasure impacts of lower body negative pressure (LBNP) at the early stages of adaptation to microgravity conditions has been gained. To assess the effectiveness of LBNP and changes in human physical performance after the spaceflight, we conducted tests on a treadmill, a bicycle ergometer, and testing with model tasks of on-planetary activity "express test". Regulatory mechanisms of the cardiovascular system proved to be quite effective when creating decompression up to -20 mm Hg, which is less than in preparation for returning to Earth. In the treadmill test, a lower speed was achieved after the spaceflight than before (13 km/h and 15 km/h, respectively) and cardiovascular system response to the change in load was slower. At the same time changes in such physiological parameters as oxygen consumption, respiratory rate and pulmonary ventilation were minimal. In the bicycle ergometer test, peak heart rate values were higher after the spaceflight than before, the physiological value of the standard exercise increased. «Express test» showed the positive dynamics from the first day to the third after returning to Earth: the performance of a dual task, the task of controlling the movement of the non-leading hand, and getting up from the supine position improved. Thus, assessment of the state of gravity-dependent physiological systems after short-term flight indicates the decrease of functional reserves of the organism and the necessity to develop appropriate countermeasures. The study was one of the first steps in the development of new medical control operations at the stage of paradigm shift from orbital flights to deep space exploration.

Abstract:Background: The various types of ionizing radiation and altered gravity in the space environment present a risk to humans during space missions. Changes in the space environment lead to skin diseases, affecting the status of the aviators to fly. Therefore, it is important to explore the molecular-level changes in the skin during space missions. Objectives: Bioinformatics analysis of gene arrays from hair follicle tissue of 10 astronauts was performed to explore changes in gene expression before, during and after space missions. Methods: First, STEM (Short Time-series Expression Miner) software was used to identify the expression patterns of hair follicle genes of astronauts pre-, in- and postflight. Gene Ontology Enrichment Analysis was then performed to explore the gene functions within the module. Protein-protein interaction network analysis was performed on skin-related genes. The transcriptional regulatory network within the module was constructed using the TRRUST database. The circadian rhythm-related genes within the module were screened using the MSigDB (Molecular Signatures Database). Results: Based on differential expression analysis between the two groups, there were 327 differentially expressed genes after the astronauts entered space compared with preflight, and only 54 differentially expressed genes after returning to Earth. This outcome suggests that the expression of most genes can be recovered on return to the ground, but there are a small number of genes whose expression cannot be recovered in a short period of time. Based on time series analysis, 311 genes showed increased expression on entry into space and decreased expression on return to Earth. The genes of this expression pattern were associated with skin development, keratinocyte differentiation and cornification. Ten hub genes were identified as skin-related genes within the module, as well as nine transcription factors and three circadian genes. One hundred and seventy-nine genes decreased in expression after entry into space and increased on return to Earth. By reviewing the literature, we found that four of the genes, CSCD2, HP, CXCR1 and SSTR4, are associated with skin diseases. Conclusions: Through bioinformatics analysis, we found that the space environment affects skin keratinocyte differentiation, leading to skin barrier damage and inflammatory responses, and that this effect was decreased after return to Earth.

Abstract:Chronic obstructive pulmonary disease (COPD) is a type of obstructive lung disease characterized by long-term breathing problems and poor airflow. Exhaled breath condensate (EBC) is now a safe and clinically significant measurement which has a huge potential to measure biomarkers in COPD. Previous studies profiled the pooled EBC samples from COPD or control participants due to technological limitations. In our study, 32 COPD patients and 28 control individuals were enrolled, and their EBC were collected. After matching with sex, age and smoking history, EBC samples of 19 COPD patients and 19 control individuals were analyzed using tandem mass tags (TMTs) quantitative mass spectrometry individually. A total of 257 proteins were identified. Compared with control group, 24 proteins (15 upregulated and 9 downregulated) were differentially expressed in COPD patients. The GO analysis of these differential proteins expressed mostly in the cytoplasm, and the KEGG analysis showed they had a predominant role in inflammatory response. And ACTB, UBC, TUBB and CCT2 involving in cell motility and cytoskeleton played important role in the interaction-net of these proteins. To sum up, we found some proteins might be novel biomarkers of EBC in COPD and TMTs was available to analyze proteomics in individual EBC samples. SIGNIFICANCE: It is still difficult to understand the mechanism of airway inflammation in COPD. Exhaled breath condensate(EBC) might be a great study object, but due to technological limitations, researchers preferred to use pooled EBC samples. This study analyzed individual EBC samples, which would deepen our understanding of the pathogenesis of COPD. And this method can be applied to individual EBC samples for further airway investigations of different purpose and different complexity.

Abstract:Understanding the axis of the human microbiome and physiological homeostasis is an essential task in managing deep-space-travel-associated health risks. The NASA-led Rodent Research 5 mission enabled an ancillary investigation of the gut microbiome, varying exposure to microgravity (flight) relative to ground controls in the context of previously shown bone mineral density (BMD) loss that was observed in these flight groups. We demonstrate elevated abundance of Lactobacillus murinus and Dorea sp. during microgravity exposure relative to ground control through whole-genome sequencing and 16S rRNA analyses. Specific functionally assigned gene clusters of L. murinus and Dorea sp. capable of producing metabolites, lactic acid, leucine/isoleucine, and glutathione are enriched. These metabolites are elevated in the microgravity-exposed host serum as shown by liquid chromatography-tandem mass spectrometry (LC-MS/MS) metabolomic analysis. Along with BMD loss, ELISA reveals increases in osteocalcin and reductions in tartrate-resistant acid phosphatase 5b signifying additional loss of bone homeostasis in flight.

Abstract:The rodent habitat on the International Space Station has provided crucial insights into the impact of spaceflight on mammals, including observation of symptoms characteristic of liver disease, insulin resistance, osteopenia and myopathy. Although these physiological responses can involve the microbiome when observed on Earth, changes in host-microbiota interactions during spaceflight are still being elucidated. Here, NASA GeneLab multiomic data from the Rodent Research 6 mission are used to determine changes to gut microbiota and murine host colon and liver gene expression after 29 and 56-days of spaceflight. Using hybrid amplicon and whole metagenome sequencing analysis, significant spaceflight-associated alterations to 42 microbiome species were identified. These included relative reductions of bacteria associated with bile acid and butyrate metabolism, such as and Functional prediction suggested over-representation of fatty acid and bile acid metabolism, extracellular matrix interactions, and antibiotic resistance genes within the gut microbiome, while host intestinal and hepatic gene expression described corresponding changes to host bile acid and energy metabolism, and immune suppression from spaceflight. Taken together, these changes imply that interactions at the host-gut microbiome interface contribute to spaceflight pathology and highlight how these interactions might critically influence human health and the feasibility of long-duration spaceflight.
