Abstract:In recent years, point-of-care testing has played an important role in immunoassay, biochemical analysis, and molecular diagnosis, especially in low-resource settings. Among various point-of-care-testing platforms, microfluidic chips have many outstanding advantages. Microfluidic chip applies the technology of miniaturizing conventional laboratory which enables the whole biochemical process including reagent loading, reaction, separation, and detection on the microchip. As a result, microfluidic platform has become a hotspot of research in the fields of food safety, health care, and environmental monitoring in the past few decades. Here, the state-of-the-art application of microfluidics in immunoassay in the past decade will be reviewed. According to different driving forces of fluid, microfluidic platform is divided into two parts: passive manipulation and active manipulation. In passive manipulation, we focus on the capillary-driven microfluidics, while in active manipulation, we introduce pressure microfluidics, centrifugal microfluidics, electric microfluidics, optofluidics, magnetic microfluidics, and digital microfluidics. Additionally, within the introduction of each platform, innovation of the methods used and their corresponding performance improvement will be discussed. Ultimately, the shortcomings of different platforms and approaches for improvement will be proposed.

Abstract:This report deals with the pathology, diagnosis and therapy of cryptorchidism from the andrological point of view. Hypothalamic-pituitary gonadal failure and a reduced transformation of gonocytes are the key factors in fertility disorders. The aim of all well-timed therapy must be to improve the fertility potential of the sexually mature male. At present, the determination through biopsy of the fertility index during orchiopexy offers the only possibility of prospective diagnosis in this respect. In the future, it will be necessary to have a standardized procedure in order to guarantee the long-term effect of correctly timed orchiopexy on fertility.

Abstract:Biochemical analysis is crucial in understanding mechanism of life activities and giving biological insights into life process. Considering the explosive development of microfluidics over the past decades, in this review, we summarized recent advances in biochemical analysis on microfluidic platforms. Highlight was put on the integrated technologies such as optical, electrical, acoustic and magnetic techniques. Focus also was given on relative applications in biomimetics, drug screening, biomolecular detection, single cell and stem cell analysis.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:Nowadays, biochemical analysis such as cell analysis, disease diagnosis and treatment, environmental monitoring and food safety are gaining increasing attention. Optically integrated microfluidic technology exhibits the merits of high sensitivity, fast detection, and great selectivity, in which flexible optical detection techniques could be easily adopted according to the unique design of microfluidic structure and screening analysis of various scales could be readily realized based on the precisely-controlled fluidics, achieving the high throughput analysis, miniaturized reagent consumption and reduced assay expenditure. In this review, the integration of microfluidic chips with optical detection techniques (UV–Vis absorption, fluorescence, infrared, surface plasmon resonance, chemiluminescence and optical tweezers), and its application in biochemical analysis are introduced, and the future development prospects of optically integrated microfluidic chips are prospected.

Abstract:In this review, we present recent advancements and novel developments in fluidic systems for applied analytical purposes in chemistry, biochemistry, and life science in general that employ and reflect the full benefits of microfluidics. A staggering rise in publications related to integrated, all-in-one microfluidic chips capable of separation, reaction, and detection have been observed, all of which realise the principal of micro total analysis systems or lab-on-a-chip. These integrated chips actively adopt the scaling law concepts, utilising the highly developed fabrication techniques. Their aim is to multi-functionalise and fully automate devices believed to assist the future advancements of point-of-care, clinical, and medical diagnostics.

Abstract:As a kind of facile tool, microfluidic paper-based analytical devices (μPADs) have been widely used in analytical and biomedical fields. However, because we lack the ability to control the continuous perfusion of these devices, they are not generally used in fields that require continuous flow, especially biofluidics fields such as cell culturing, drug screening, and organs on chips. In this study, we designed a novel, low-cost, and compact platform that can be used to control the continuous perfusion of μPADs. As most of the parts of this platform can be created using a three-dimensional (3D) desktop printer, our platform can be easily duplicated by other researchers. We demonstrate that with our system, μPADs can be promising paper-based biofluidic platforms for cell culturing and drug screening.

Abstract:Microfluidic-based point-of-care diagnostics offer several unique advantages over existing bioanalytical solutions, such as automation, miniaturisation, and integration of sensors to rapidly detect on-site specific biomarkers. It is important to highlight that a microfluidic POC system needs to perform a number of steps, including sample preparation, nucleic acid extraction, amplification, and detection. Each of these stages involves mixing and elution to go from sample to result. To address these complex sample preparation procedures, a vast number of different approaches have been developed to solve the problem of reagent storage and delivery. However, to date, no universal method has been proposed that can be applied as a working solution for all cases. Herein, both current self-contained (stored within the chip) and off-chip (stored in a separate device and brought together at the point of use) are reviewed, and their merits and limitations are discussed. This review focuses on reagent storage devices that could be integrated with microfluidic devices, discussing further issues or merits of these storage solutions in two different sections: direct on-chip storage and external storage with their application devices. Furthermore, the different microvalves and micropumps are considered to provide guidelines for designing appropriate integrated microfluidic point-of-care devices.

Abstract:As a powerful tool, microfluidics has been widely used in bioanalysis due to the advantages of high portability, fewer sample requirements and rapid detection time. To achieve selective and sensitive detection, enzymes are commonly used as biorecognition elements. However, enzymes used in bioanalysis still suffer from high cost and low stability. Therefore, emerging nanozymes (nanomaterials with enzyme-like characteristics) have been developed to replace enzymes to address their shortcomings. The microfluidic devices based on nanozymes have attracted a lot of interest and shown great promise in bioanalysis via a simple, rapid, and portable approach. This review focuses on recent advances in typical nanozymes and nanozymes involved in cascaded reactions used in microfluidic bioanalysis. Abstract:The field of microfluidics has exploded in the past decade, particularly in the area of chemical and biochemical analysis systems. Borrowing technology from the solid-state electronics industry and the production of microprocessor chips, researchers working with glass, silicon, and polymer substrates have fabricated macroscale laboratory components in miniaturized formats. These devices pump nanoliter volumes of liquid through micrometer-scale channels and perform complex chemical reactions and separations. The detection of reaction products is typically done fluorescently with off-chip optical components, and the analysis time from start to finish can be significantly shorter than that of conventional techniques. In this review we describe these microfluidic analysis systems, from the original continuous flow systems relying on electroosmotic pumping for liquid motion to the large diversity of microarray chips currently in use to the newer droplet-based devices and segmented flow systems. Although not currently widespread, microfluidic systems have the potential to become ubiquitous.

Abstract:The emergence of nanofluidic lab‐on‐a‐chip systems heralds new opportunities in molecular biosensing. This review provides a holistic overview of the principles and construction of functional nanofluidic devices, focusing on their applications in single‐molecule analysis, ultrasensitive detection, and subcellular analysis of single cells. The review aims to provide the inspiration and guidance to accelerate the clinical translation of nanofluidic technologies. Biosensing plays a vital role in healthcare monitoring, disease detection, and treatment planning. In recent years, nanofluidic technology has been increasingly explored to be developed into lab‐on‐a‐chip biosensing systems. Given now the possibility of fabricating geometrically defined nanometric channels that are commensurate with the size of many biomolecules, nanofluidic‐based devices are likely to become a key technology for the analysis of various clinical biomarkers, including DNA (deoxyribonucleic acid) and proteins in liquid biopsies. This review summarizes the fundamentals and technological advances of nanofluidics from the purview of single‐molecule analysis, detection of low‐abundance molecules, and single‐cell analysis at the subcellular level. The extreme confinement and dominant surface charge effects in nanochannels provide unique advantages to nanofluidic devices for the manipulation and transport of target biomarkers. When coupled to a microfluidic network to facilitate sample introduction, integrated micro‐nanofluidic biosensing devices are proving to be more sensitive and specific in molecular analysis compared to conventional assays in many cases. Based on recent progress in nanofluidics and current clinical trends, the review concludes with a discussion of near‐term challenges and future directions for the development of nanofluidic‐based biosensing systems toward enabling a new wave of lab‐on‐a‐chip technology for personalized and preventive medicine.

Abstract:In the past two decades, Micro Fluidic Systems (MFS) have emerged as a powerful tool for biosensing, particularly in enriching and purifying molecules and cells in biological samples. Compared with conventional sensing techniques, distinctive advantages of using MFS for biomedicine include ultra-high sensitivity, higher throughput, in-situ monitoring and lower cost. This review aims to summarize the recent advancements in two major types of micro fluidic systems, continuous and discrete MFS, as well as their biomedical applications. The state-of-the-art of active and passive mechanisms of fluid manipulation for mixing, separation, purification and concentration will also be elaborated. Future trends of using MFS in detection at molecular or cellular level, especially in stem cell therapy, tissue engineering and regenerative medicine, are also prospected.

Abstract:Microfluidic chips have the advantages of small sample size, low detection cost, and short detection time. Combined with optical sensors, electrochemical sensors, and microwave sensors, they can be used to construct sensitive, repeatable, and portable detection systems. Optical biosensors are powerful analytical instruments that can measure biomolecular interactions in real-time and unlabeled manner. The combination of affinity and kinetics can be measured through high sensitivity. many optical biosensors have microfluidic control, making them extremely sensitive. Combining the concept of microfluidic technology, this article briefly elucidates the various cutting-edge research fields of microfluidic technology combined with optical biosensors, such as optical detection methods suitable for microfluidic chips, microfluidic biological immunofluorescence detection, the application of microfluidic technology in optical detection methods. It also prospects the future development direction and the challenges as well as strategies for addressing the problems in the development of the new generation of biological microfluidic sensors were proposed.

Abstract:Cells, as the fundamental structural elements and functional units of life, are necessary to be explored the variation of components and behaviors in the physiological and pathological processes. Benefited from the development of microfluidic culture and fluorescence labeling techniques, chips coupled with fluorescent tools have become a powerful platform for cell analysis. Thus, in this review, we mainly focus on the two aspects of overview: one is the microenvironment simulation on microfluidic systems including the fabrication of the chemical and physical microenvironment, the cell-cell co-culture/cell-extracellular matrix interaction model, and organ/tissue-on-a-chip system; the other is the fluorescence labeling strategies for cellular component detection and behavior observation, introducing from several aspects: nucleic acid-based modification, in situ chemical modification, hydrophobic insertion labeling and genetic-based labeling. At last, the existing achievements and future challenges in fluorescent analysis of cells based on microfluidic chip are discussed.

Abstract:Recent developments in paper‐based electrochemical microfluidics are reviewed through the lens of capillary flow, highlighting how their combination is harnessed to produce simple yet powerful diagnostic tools. Electrochemical detection has the potential to bring quantitative testing to the point of need, but many challenges remain to fully exploit the synergy between electrodes and capillary flow to create integrated devices. Electrochemical paper‐based microfluidics has attracted much attention due to the promise of transforming point‐of‐care diagnostics by facilitating quantitative analysis with low‐cost and portable analyzers. Such devices harness capillary flow to transport samples and reagents, enabling bioassays to be executed passively. Despite exciting demonstrations of capillary‐driven electrochemical tests, conventional methods for fabricating electrodes on paper impede capillary flow, limit fluidic pathways, and constrain accessible device architectures. This account reviews recent developments in paper‐based electroanalytical devices and offers perspective by revisiting key milestones in lateral flow tests and paper‐based microfluidics engineering. The study highlights the benefits associated with electrochemical sensing and discusses how the detection modality can be leveraged to unlock novel functionalities. Particular focus is given to electrofluidic platforms that embed electrodes into paper for enhanced biosensing applications. Together, these innovations pave the way for diagnostic technologies that offer portability, quantitative analysis, and seamless integration with digital healthcare, all without compromising the simplicity of commercially available rapid diagnostic tests.

Abstract:Microfluidics can precisely control and manipulate micro-scale fluids, and are also known as lab-on-a-chip or micro total analysis systems. Microfluidics have huge application potential in biology, chemistry, and medicine, among other fields. Coupled with a suitable detection system, the detection and analysis of small-volume and low-concentration samples can be completed. This paper reviews an optical imaging system combined with microfluidics, including bright-field microscopy, chemiluminescence imaging, spectrum-based microscopy imaging, and fluorescence-based microscopy imaging. At the end of the article, we summarize the advantages and disadvantages of each imaging technology.

Abstract:One of the goals of point-of-care (POC) is a chip-based, miniaturized, portable, self-containing system that allows the assay of proteins, nucleic acids, and cells in complex samples. The integration of nanomaterials and microfluidics can help achieve this goal. This tutorial review outlines the mechanism of assaying biomarkers by gold nanoparticles (AuNPs), and the implementation of AuNPs for microfluidic POC devices. In line with this, we discuss some recent advances in AuNP-coupled microfluidic sensors with enhanced performance. Portable and automated instruments for device operation and signal readout are also included for practical applications of these AuNP-combined microfluidic chips.
