Abstract:Aminopeptidase N (APN) is an important proteinase, and the abnormal level of APN is closely related to many diseases. Thus, the specific detection and tracking of APN activity is of great significance in the fields of clinical diagnosis and drug development. Herein, we designed and synthesized an APN-activated near-infrared (NIR) fluorescent probe ZY-APN and successfully applied it for in vitro and in vivo enzyme detection. Linking L-alanine recognition moiety to the xanthene dye through amide bond endowed ZY-APN with specific response towards APN without being influenced by other bio-species. After reaction with APN, the probe exhibited a notable NIR fluorescence enhancement at 730 nm. With this probe, we could distinguish between normal cells and tumor cells due to the differences of APN activity. Besides, we have realized the visualization of high expression of APN in tumor-bearing mice. Hence, probe ZY-APN may display potential application in the diagnosis and treatment of APN-related diseases, especially cancer.

Abstract:BACKGROUND:C-reactive protein (CRP), one of the classic biomarkers of inflammation, is closely related to infectious inflammation, cardiovascular disease, cancer, and other diseases. Therefore, timely and accurate detection of CRP in human blood is crucial for the discovery, diagnosis, and treatment of the aforementioned diseases. Herein, a novel label-free NIR fluorescence aptasensor with a large Stokes shift based on an AIEE anthracene derivative B and a molybdenum disulfide (MoS2) platform was developed and used for the high sensitivity and specificity detection of CRP. RESULTS:Compound B could emit near-infrared (NIR) fluorescence with a large Stokes shift (190 nm). Notably, this compound could bind with the aptamer of CRP (CRP-Apt) through electrostatic attraction to form a B/CRP-Apt complex, generating an aggregation-induced emission enhancement effect and enhancing the fluorescent intensity of B. B/CRP-Apt could be adsorbed on the surface of MoS2 with the addition of MoS2 to its solution, and the fluorescence of Compound B was quenched. CRP was then added to the above solution. CRP-Apt had a substantially higher affinity for CRP than MoS2. Therefore, B/CRP-Apt detached from the surface of MoS2 and bound to CRP, thereby restoring the fluorescence of B. Experimental results showed a good linear relationship between the fluorescent recovery intensity of B and the concentration of CRP in the concentration range of 0.3-70 ng mL-1, with a limit of detection as low as 0.1 ng mL-1. SIGNIFICANCE AND NOVELTY:The aptasensor integrates the advantages of high sensitivity of NIR fluorescence, high specificity of aptamers, good water-solubility and AIEE effect of Compound B. And it could be applied to the determination of CRP in human serum samples, while most of the reported methods can only determine CRP in spiked human serum samples.

Abstract:Leucine aminopeptidase (LAP), a kind of proteolytic enzyme and an indicator of many kinds of cancer. Developing effective methods for tracking endogenous LAP activity is crucial for LAP-related cancer diagnosis and treatment. In this work, we report a new near-infrared fluorescent probe: CY-P, which is capable of highly effective monitoring of endogenous LAP activity in vitro and in vivo. CY-P was constructed by incorporating a dipeptide (Cys-Leu: Cysteine-Leucine) into a water-soluble NIR-emitting fluorophore (CY-OH: a semi-cyanine scaffold). Upon exposure to LAP, the amide bond in Cys-Leu will be specifically cleaved by LAP, and subsequent an intramolecular cyclization to release the unmasked phenol-based semi-cyanine. In this process, a dramatic fluorescence enhancement at 701 nm was observed, the evaluation of LAP activity is established in terms of the relationship between fluorescence light-up efficiency and LAP activity. In our experiments, CY-P features high feedback towards LAP with various advantages, such as low detection limit (4.9 x 10(-5) U mL(-1)), fast-response (similar to 9 min), good biocompatibility, and NIR emission. As expected, the probe exhibits low cytotoxicity and excellent cell membrane permeability, which is successfully used to monitor endogenous LAP activity in living cancer cells and zebrafish models. Therefore, we anticipate that this newly designed fast LAP detection platform will provide an alternative method for the studies of related diseases. (C) 2021 Elsevier Ltd. All rights reserved.
A Facile Turn-on Chemiluminescence Probe for Sensitive Imaging on Aminopeptidase N Activity.

Abstract:Aminopeptidase N, as a target for drug discovery, shows marked relationships with many diseases, especially liver injury and cancer. Here, we explored a chemiluminescence (CL) probe for sensing APN by tethering the APN-specific substrate group to the ortho-acrylated phenoxy-dioxetane scaffold. In this way, two CL probes (APN-CL and BAPN-CL) were designed with noncapped leucine and butoxy-carbonyl capped leucine as the protecting group to preserve the chemiexcitation energy. The uncovered leucine was demonstrated to be essential for detection of APN activity by comparing the CL intensity of two CL probes. Probe APN-CL was turned on upon APN cleavage, resulting in a high chemiluminescent emission, whereas the chemiexcitation energy of probe BAPN-CL was still restrained even with the high-level APN. The result was further elucidated by molecular docking simulations. Probe APN-CL exhibited a fast response and high sensitivity with a detection limit of 0.068 U/L, and an excellent specificity for the discrimination of APN from biological ions, small molecules, and other proteases commonly found in living system. By virtue of good stability and cell viability, probe APN-CL imaged abnormal levels of APN in tumour cells and tumour-bearing mice. Moreover, this probe APN-CL could be easily used to evaluate APN inhibitors and APN levels in plasma samples from 20 patients. Overall, as a facile and cost-effective probe, APN-CL will be a promising alternative in the early diagnosis of pathologies and for cost-effective screening of inhibitors.
Activatable Near-Infrared Fluorescent Probe for in Vivo Imaging of Fibroblast Activation Protein-Alpha

Abstract:Fibroblast activation protein-alpha (FAPα) is a cell surface glycoprotein which is selectively expressed by tumor-associated fibroblasts in malignant tumors but rarely on normal tissues. FAPα has also been reported to promote tumor growth and invasion and therefore has been of increasing interest as a promising target for designing tumor-targeted drugs and imaging agents. Although medicinal study on FAPα inhibitors has led to the discovery of many FAPα-targeting inhibitors including a drug candidate in a phase II clinical trial, the development of imaging probes to monitor the expression and activity of FAPα in vivo has largely lagged behind. Herein, we report an activatable near-infrared (NIR) fluorescent probe (ANP(FAP)) for in vivo optical imaging of FAPα. The ANP(FAP) consists of a NIR dye (Cy5.5) and a quencher dye (QSY21) which are linked together by a short peptide sequence (KGPGPNQC) specific for FAPα cleavage. Because of the efficient fluorescence resonance energy transfer (FRET) between Cy5.5 and QSY21 in ANP(FAP), high contrast on the NIR fluorescence signal can be achieved after the cleavage of the peptide sequence by FAPα both in vitro and in vivo. In vitro assay on ANP(FAP) indicated the specificity of the probe to FAPα. The in vivo optical imaging using ANP(FAP) showed fast tumor uptake as well as high tumor to background contrast on U87MG tumor models with FAPα expression, while much lower signal and tumor contrast were observed in the C6 tumor without FAPα expression, demonstrating the in vivo targeting specificity of the ANP(FAP). Ex vivo imaging also demonstrated ANP(FAP) had high tumor uptake at 4 h post injection. Collectively, these results indicated that ANP(FAP) could serve as a useful NIR optical probe for early detection of FAPα expressing tumors.

Ultrasensitive Detection of Aminopeptidase N Activity in Urine and Cells with a Ratiometric Fluorescence Probe

Abstract:An ultrasensitive ratiometric fluorescent probe (CVN) has been designed and synthesized by incorporating alanine into the cresyl violet fluorophore. The probe shows ratiometric fluorescence response toward aminopeptidase N (APN) through the increase of fluorescent intensity ratio of 626/575 nm. The sensitivity of the probe is ultrahigh with a detection limit of 33 pg/mL, which can quantify the contents of APN in 500-fold diluted human urine samples. Furthermore, by using ratiometric fluorescence imaging, the probe reveals significantly higher contents of APN in HepG2 cells than those in LO2 cells, which has been further used to distinguish these two types of cells in mixed cocultures. The probe could be of great importance for the APN-related disease diagnosis and pathophysiology elucidation.

Abstract:In situ trapping endogenous leucine aminopeptidase (LAP) in organism is of great significance, but current fluorescent probes for LAP are susceptible to complex experimental conditions because of lacking internal efficient standard. Here, we present HCHMC-S-Leu, a novel near-infrared (NIR) ratiometric fluorescent probe for high effective tracking of endogenous LAP activity via intramolecular S -> N rearrangement. The study mainly focuses on how to exactly modulate the electronic effects and extend the emission wavelength shift for realizing an ideal ratiometric detection of LAP, as well as solving the intrinsic problems of hydmphilicity and short emission wavelength range. By incorporating a dipeptide (cysteinyl-leucine: Cys-Leu) into a fluorescent chlomhydroxyl-merocyanine (CHMC) scaffold with rational design, a novel fluorescent probe HCHMC-S-Leu is screened out. As demonstrated, the new probe exhibits high effective feedback on LAP activity with various advantages, such as favorable hydrophilicity, ideal ratiometric detection of LAP, and NIR emission. Furthermore, imaging of endogenous LAP in living cancer cells and zebrafish models with HCHMC-S-Leu is also successful. This scaffold of NIR ratiometric fluorescent probe represents a new powerful tool for the study of LAP in living systems.

Abstract:Cancer, a leading cause of death worldwide, still needs surgery as essential curation in over 80% population. However, complete resection of cancerous tissues remains a great challenge due to the difficulty in tumor margin identification. Fluorescent probe based imaged-guided surgery (IGS) for in-situ local disease focus imaging is thought to be an ideal method for intraoperation tumor identification due to its high spatiotemporal resolution and real-time manner. By targeting tumor microenvironments (TME) or tumor-associated biomolecules, fluorescent probes could "light-up" tumor tissues and navigate surgical operation. Among which, biosensors targeting aminopeptidase N (APN) have attracted intense attention due to its high relevance with progression of tumor. However, most of the probes adopting its natural substrate (L-Alaine) as the recognition unit may be vulnerable toward other widely distributed aminopeptidase such as leukotriene A 4 hydrolase (LTA 4 H) and leucine aminopeptidase (LAP). In order to acquire high specific APN targeted probe for accurate detection of APN activity in biological samples and fulfill high contrast tumor imaging, herein a non-natural amino acid (P-Chloro-L-Phenylalanine, PCPA) based fluorescent probe was firstly reported for APN targeted bioimaging. The acquired named as TMN-PCPA showed high affinity ( K m = 3.27 ± 0.23 μM) and dramatically improved selectivity toward APN than previously reported APN probe. TMN-PCPA was successfully applicated into discrimination tumor bio-samples from normal bio-samples in both cellular and ex vivo level with significantly increased tumor to normal (T/N) ratio, thus showed great potential in intraoperation application such imaged-guided surgery and providing structural basis to high specific APN targeted biosensor construction.

Abstract:We have investigated the low-temperature physical properties of BaTi2Sb2O and Ba1−xNaxTi2Sb2O (x=0.05, 0.1, 0.15, 0.2, 0.25, 0.3) by means of muon spin rotation (μSR) and SQUID magnetometry. Our measurements reveal the absence of magnetic ordering below TDW=58 K in the parent compound. Therefore the phase transition at this temperature observed by magnetometry is most likely due to the formation of a charge-denisty wave (CDW). Upon substitution of barium by sodium in Ba1−xNaxTi2Sb2O we find for x=0.25 superconductivity with a maximum Tc=5.1 K in the magnetization and a bulk Tc,bulk=4.5 K in the μSR measurements. The temperature dependency of the London penetration depth λ−2(T) of the optimally doped compound can be well explained within a conventional weak-coupling scenario in the clean limit.

Abstract:This article discusses the importance of early tumor detection, particularly in liver cancer, and the role of leucine aminopeptidase (LAP) as a potential marker for liver cancer diagnosis and prognosis assessment. The article highlights the limitations of current tumor markers and the need for new markers and multi-marker approaches to improve accuracy. The authors introduce a novel near-infrared fluorescent probe, NTAP, designed for LAP detection. They describe the synthesis of the probe and evaluate its spectral properties, including the LOD was 0.0038 U/mL, and QY was 0.32%. The kinetic properties of NTAP, such as the relationship between LAP concentration (0-0.08 U/mL), reaction time (3 min), and fluorescence excitation spectra (475 nm) and emission spectra (715 nm) are investigated. The article also discusses the stability and selectivity of the probe and its ability to detect LAP in complex samples. Cellular imaging experiments demonstrate the NATP specificity and selectivity in detecting LAP activity and its inhibition. Animal models of liver and lung metastasis are used to evaluate the probe's imaging capabilities, showing its ability to accurately locate and detect metastatic lesions. The article concludes by emphasizing the potential applications of the NTAP probe in early tumor diagnosis, treatment monitoring, and the study of tumor metastasis mechanisms.

It becomes clear that conduction current or polarization current models lead to identical dispersion equations in moving media. The invariance of dispersion equations is defined and discussed. The question of velocity induced wave modes in moving media is considered, and conditions for appearance of these new modes are stated.

Abstract:As a key DNA repair enzyme, human apurinic/apyrimidinic endonuclease 1 (APE1) participates in many crucial cellular processes. It has been recognized as a potential prognostic biomarker and a therapeutic target for many cancer and other diseases. However, no methods are available now for direct quantification of the APE1 activity in subcellular compartments without tedious pre-isolation steps because of the intensive cellular background. In this work, we report a DNA/RNA hybrid-based fluorescent probe which enabled one-step detection of APE1 in the nuclear, cytoplasmic, and mitochondrial extracts of different types of cultured cells without the need of any purification steps. With the noncanonical structure, the hybrid probe showed an exquisite specificity for APE1 and strong resistance against other nucleases. Taking advantage of a substantial acceleration effect of the 3â€™-flanking mismatches on the reaction rates, the hybrid probe showed high sensitivity to the target enzyme (linear working range: 0.02~1.0 U/mL; detection limit: 0.01 U/mL). The mechanism for the rapid digestion of the modified structure by APE1 was clarified via kinetic study and single-molecule fluorescence resonance energy transfer (smFRET) analysis. Significantly varied subcellular distribution of APE1 in different cells were observed by using the probe. The method was simple and robust, which offered an easily accessible molecular tool for high-throughput quantification of the subcellular APE1 activity. It may also facilitate the development of effective APE1-targeted inhibitors as anticancer drugs.

Abstract:Leucine aminopeptidase (LAP) is a vital proteolytic enzyme, and its overexpression is often associated with many physiological diseases, such as liver dysfunction and breast cancer. Therefore, the accurate measurement of LAP concentrations in cells is critical for the diagnosis and prevention of related diseases. Herein, a new ratiometric fluorescent probe, DPP-Leu, based on diketopyrrolopyrrole (DPP) was designed and synthesized for LAP detection based on the specific enzymatic cleavage of the N-terminal leucine residue. The fluorescence intensity ratio of DPP-Leu (I548/I651) showed a remarkable change in the presence of LAP, with a limit of detection of 0.011 U L-1, and DPP-Leu was successfully applied to detect LAP in fetal bovine serum (FBS) and artificial urine. Cell imaging experiments revealed that DPP-Leu could target mitochondria and distinguish tumor cells with high LAP content from normal cells. Importantly, benefiting from the structural transformation of DPP-Leu to the photosensitizer 4 under LAP catalysis, the probe could kill tumor cells under light irradiation without damaging normal cells.

Abstract:Behçet's disease (BD) is a chronic, relapsing, multi-system inflammatory disorder, clinically characterized by recurrent oral and genital ulcers, skin lesions, and uveitis. Other manifestations include arthritis, a positive pathergy test, thrombophlebitis, central nervous system disease and gastrointestinal ulcerations. The majority of affected individuals do not have life-threatening disease, although mortality can be associated with vascular-thrombotic and neurological manifestations. Currently, treatment of BD is symptomatic and empirical, and is tailored according to the severity of clinical features. In the past few years, isolated reports and case-series have been published on adult BD patients suggesting that inhibition of TNF-α is a promising therapeutic approach for severe ocular and various extra-ocular manifestations, including central nervous system involvement. In this study we present our promising experience with Etanercept therapy in juvenile-onset BD patients, characterized by refractory multi-organ involvement.
In situ imaging of aminopeptidase N activity in hepatocellular carcinoma: a migration model for tumour using an activatable two-photon NIR fluorescent probe

Abstract:CD13/aminopeptidase N (APN), which is a zinc-dependent metalloproteinase, plays a vital role in the growth, migration, angiogenesis, and metastasis of tumours. Thus, in situ molecular imaging of endogenous APN levels is considerably significant for investigating APN and its different functions. In this study, a novel two-photon near-infrared (NIR) fluorescence probe DCM-APN was prepared to perform in vitro and in vivo tracking of APN. The N-terminal alanyl site of probe DCM-APN was accurately hydrolysed to the amino group, thereby liberating strong fluorescence owing to the recovery of the Intramolecular Charge Transfer (ICT) effect. By considering its outstanding selectivity, ultra-sensitivity (DL 0.25 ng mL-1) and favourable biocompatibility, the probe DCM-APN was used to distinguish between normal cells (LO2 cells) and cancer cells (HepG-2 and B16/BL6 cells). Furthermore, migration of hepatocellular carcinoma cells was apparently inhibited by ensuring that the APN catalytic cavity was occupied by bestatin. The identification of three-dimensional (3D) fluorescence in cancer tissues was completed under two-photon excitation coupled with lighting up hepatocellular carcinoma tumours in situ; this revealed that probe DCM-APN is an effective tool for detecting APN, thereby assisting in the early diagnosis of tumour in clinical medicine.

Abstract:Fibroblast activation protein-alpha (FAPα) is a key modulator of the microenvironment in multiple pathologies and is becoming the next pan-cancer target for cancer diagnostics and therapeutics. Chemiluminescence (CL) luminophores are considered as one of the most sensitive families of probes for detection and imaging applications due to their high signal-to-noise ratio. Until now, however, no such effective CL probe was reported for FAPα detection. Herein, we developed a novel CL probe for the detection of endogenous FAPα activity by incorporating FAPα-specific dipeptide substrates (glycine-proline) to the improved Schaap's adamantylidene-dioxetane. In this manner, we designed three CL probes (CFCL, BFCL, and QFCL) with the dipeptide substrate blocked by N-terminal benzyloxycarbonyl, N-tert-butoxycarbonyl or N-quinoline-4-carboxylic acid, respectively, which was used as the masking group to restrain the chemiexcitation energy. Probe CFCL exhibited the optimal specificity for the discrimination of FAPα from dipeptidase IV and prolyl oligopeptidase, which was elucidated by molecular docking simulation. Upon FAPα cleavage, CFCL was turned on for the highly selective and sensitive detection of FAPα with a limit of detection of 0.785 ng/mL. Furthermore, the ability of CFCL to image FAPα was effectively demonstrated in vitro, including various biological samples (plasma and tissue preparations), and in living systems (tumor cells and tumor-bearing mice). Furthermore, this newly established probe could be easily extended to evaluate FAPα inhibitors. Abstract:Aminopeptidases, enzymes with critical roles in human body, are emerging as vital biomarkers for metabolic processes and diseases. Aberrant aminopeptidase levels are often associated with diseases, particularly cancer. Small‐molecule probes, such as fluorescent, fluorescent/photoacoustics, bioluminescent, and chemiluminescent probes, are essential tools in the study of aminopeptidases‐related diseases. The fluorescent probes provide real‐time insights into protein activities, offering high sensitivity in specific locations, and precise spatiotemporal results. Additionally, photoacoustic probes offer signals that are able to penetrate deeper tissues. Bioluminescent and chemiluminescent probes can enhance in vivo imaging abilities by reducing the background. This comprehensive review is focused on small‐molecule probes that respond to four key aminopeptidases: aminopeptidase N, leucine aminopeptidase, Pyroglutamate aminopeptidase 1, and Prolyl Aminopeptidase, and their utilization in imaging tumors and afflicted regions. In this review, the design strategy of small‐molecule probes, the variety of designs from previous studies, and the opportunities of future bioimaging applications are discussed, serving as a roadmap for future research, sparking innovations in aminopeptidase‐responsive probe development, and enhancing our understanding of these enzymes in disease diagnostics and treatment.

A Ratiometric Fluorescent Probe for Monitoring Leucine Aminopeptidase in Living Cells and Zebrafish Model.

Abstract:Leucine aminopeptidase (LAP) is an important cancer-related biomarker, which shows significant overexpression in malignant tumor cells like liver cancer. Developing an effective method to monitor LAP in tumor cells holds great potential for cancer diagnosis, treatment, and management. In this work, we report a novel BODIPY-based fluorescent probe (BODIPY-C-Leu) capable of monitoring LAP in vitro and in vivo in both ratiometric and turn-on model. BODIPY-C-Leu contains an asymmetrical BODIPY dye for fluorescent signaling and a dipeptide (Cys-Leu) as the triggered moiety. Activation occurs by cleavage of the amide bond in dipeptides and subsequently an intramolecular S → N conversion to convert sulfur-substituted BODIPY to amino-substituted BODIPY, resulting in a dramatic fluorescence variation to realize the detection of LAP. Furthermore, we have successfully employed BODIPY-C-Leu to monitor LAP activity in different cancer cells, indicating that HeLa cells have a higher level of LAP activity than A549 cells. Importantly, we demonstrated the capability of the probe for real-time monitoring the drug-induced LAP level changes in zebrafish.

Abstract:Background: Cell-surface proteins, which are closely associated with various physiological and pathological processes, have drawn much attention in drug discovery and disease diagnosis. Thus, wash-free imaging of the target cell-surface protein under its native environment is critical and helpful for early detection and prognostic evaluation of diseases. Results: To minimize the interference from autofluorescence and fit the penetration depth towards tissue samples, we developed a fluorogenic antibody-based probe, Ab-Cy5.5, , which will liberate > 5-fold turn-on near-infrared (NIR) emission in the presence of its target antigen within 10 min. Significance: By taking advantage of the fluorescence-quenched dimeric H-aggregation of Cy5.5, Ab-Cy5.5 with Cy5.5 attached at the N-terminus showed negligible background signal, allowing direct imaging of the target cell- surface protein in both living cells and tissue samples without washing.
