Abstract:Early social isolation (SI) increased open field activity and anxiety‐like behavior on the successive alleys test in female rats. SI and single prolonged stress synergistically increased approach latency in the novelty suppressed feeding test. Complex effects of stress history were observed across a 3‐day fear conditioning paradigm while no group differences were observed in home cage ethanol consumption, regardless of prior ethanol exposure. These data contribute to the growing knowledge of stress history on stress‐ and ethanol‐related behaviors. Background Exposure to stress during childhood and adolescence is a risk factor for alcohol use disorder (AUD) and comorbid conditions, including posttraumatic stress disorder (PTSD). We previously established an adolescent social isolation (SI) model that leads to the emergence of a wide range of behavioral risk factors for AUD, including increased anxiety‐like behavior, locomotor activity, and ethanol consumption in male and female rats. Here, we sought to test the hypothesis that SI may increase vulnerability to single prolonged stress (SPS), a rodent model of PTSD. Methods Female Long Evans rats (n = 8/group) were either single‐housed or group‐housed (GH) (4/cage) on postnatal day 21. One week later, rats underwent testing in the open field test (OFT), elevated plus‐maze (EPM), and successive alleys test (SAT). Following initial behavioral testing, a subset of SI/GH rats were exposed to SPS. All rats were then tested on the novelty‐suppressed feeding test (NSFT) followed by fear conditioning and home cage two‐bottle choice to assess ethanol consumption. Results SI significantly increased activity in the OFT and anxiety‐like behavior on the SAT, but not the EPM. While SI and SPS alone had no effect on the NSFT, exposure to both stressors significantly increased approach latency. Complex effects of stress history were observed across a 3‐day fear conditioning paradigm and no group differences were observed with home cage ethanol consumption, regardless of prior ethanol exposure. Conclusions The results from this study provide novel evidence that SI interacts with SPS in female rats to influence behavior in assays of unconditioned anxiety‐like behavior (NSFT) and conditioned fear. Surprisingly, stress exposure had no effect on home cage ethanol consumption. Ultimately, these models provide useful avenues to examine the interaction between stressful experiences, alcohol exposure, biological sex, and the neurobiological adaptations underlying potential risk factors for psychiatric conditions.

Abstract:Background: In humans, early life stress (ELS) is associated with an increased risk for developing both alcohol use disorder (AUD) and post-traumatic stress disorder (PTSD). We previously used an infant footshock model that produces stress-enhanced fear learning (SEFL) in rats and mice and increases aversion-resistant alcohol drinking in rats to explore this shared predisposition. The goal of the current study was to extend this model of comorbid PTSD and AUD to male and female C57BL/6J mice. Methods: Acute ELS was induced using 15 footshocks on postnatal day (PND) 17. In adulthood, alcohol drinking behavior was tested in one of three two-bottle choice drinking paradigms. In continuous access, mice were given 24 h access to 5% or 10% ethanol and water for five consecutive drinking sessions each. In limited access drinking in the dark, mice were given 2 h of access to 15% ethanol and water across 15 sessions 3 h into the dark cycle. In intermittent access, mice were presented with 20% ethanol and water Monday, Wednesday, and Friday, for four consecutive weeks. In a fifth week of intermittent access drinking, increasing concentrations of quinine (10 mg/L, 100 mg/L, and 200 mg/L) were added to the ethanol to test aversion-resistant drinking. Intermittent access drinking was tested with and without a period of adolescent drinking (PND 35). Results: Infant footshock did not alter drinking in the continuous or limited access tasks. Adult consumption and preference were lower in the intermittent access task when adolescent drinking was included and there were ELS-induced differences in consumption of quinine-adulterated ethanol in females. Conclusions: Our results demonstrate that infant footshock followed by a period of adolescent drinking is a viable model of comorbid PTSD and AUD in rats and mice.

Effects of isolation stress and voluntary ethanol exposure during adolescence on ethanol and nicotine co-use in adulthood using male rats

Abstract:Background: Alcohol use in adolescence may increase susceptibility to substance use disorders (SUDs) in adulthood. This study determined if voluntary ethanol (EtOH) consumption during adolescence, combined with social isolation, alters the trajectory of EtOH and nicotine intake during adulthood, as well as activating brain neuroinflammation. Methods: Adolescent male isolate- and group-housed rats were given 0.2 % saccharin/20 % EtOH (Sacc/EtOH) or water using intermittent 2-bottle choice; controls were given water in both bottles (n=17-20 per group). Some rats from each group (n=5-6) were euthanized one week later to measure autoradiographic [3H]PK-11195 binding, an indicator of microglial reactivity, and the remainder (n=11-14 per group) were tested in adulthood in 2-bottle choice, followed by nicotine self-administration using an incremental fixed ratio (FR) schedule with Sacc/EtOH and water concurrently available. Results: Isolation housing increased adolescent intake of Sacc/EtOH, but the increase did not produce an observable neuroimmunological response in brain. Adolescent EtOH exposure decreased adult intake of both Sacc/EtOH and unsweetened EtOH, with isolate-housed rats showing a greater effect than group-housed rats. In the co-use model, a cross-price economic demand analysis revealed a substitutional relationship between Sacc/EtOH and nicotine, but no effect of adolescent Sacc/EtOH exposure. Compared to group-housed rats, isolate-housed rats were more sensitive to the changing price of nicotine and showed greater substitutability of Sacc/EtOH for nicotine. Conclusion: The current results suggest that adolescent EtOH exposure per se, with or without isolation stress, does not likely explain the enhanced risk for either alcohol or nicotine use later in life.

Abstract:Social stress exposure heightens the risk of substance abuse disorder development, especially when endured during adolescence, influencing long-term mental health. This study investigates early-life stress's potential to confer resilience against later-life stressors. To investigate this hypothesis, we examined the impact of a single social defeat (SD) incident during adolescent mice's lives on subsequent voluntary ethanol consumption following repeated adult social stress exposure. Half of the adolescent mice experienced SD at postnatal day 28. Three weeks later (postnatal day 49), defeated groups encountered four confrontations with aggressive residents every 72 hours, while control groups were exposed to non-resident exploration. A day after the last SD, defeated mice were classified as resilient or susceptible based on their response to a social interaction test (SIT), a model for depressive behavior. To assess ethanol consumption during young adulthood, researchers used the 'drinking in the dark' and oral ethanol self-administration paradigms. Stress inoculation (IS) slightly increased resilient animals in the SIT. In mice without IS exposure during adolescence, susceptible defeated mice displayed higher ethanol consumption and motivation than control and resilient mice. IS in adolescence effectively counteracted this effect, as IS-SD groups, whether resilient or susceptible, showed no increase in ethanol intake. These groups also exhibited similar motivation to control, measured by the progressive ratio. Notably, elevated IL-6 levels seen in SD-S mice were absent in IS-exposed mice. Additionally, IS-exposed groups had lower prefrontal cortex IL-6 and CX3CL1 levels. These findings support the hypothesis that IS, induced by moderate-intensity stress during adolescence, can enhance resilience to more severe stressors in adulthood.

Abstract:The purpose of this study was to evaluate the effects of early life stress on the vulnerability to ethanol consumption in adolescence. To this aim, mice were separated from their mothers for 12 hours/day on postnatal days 8 and 12. Emotional behavior (light-dark box, elevated plus maze and tail suspension tests) and pre-attentional deficit (pre-pulse inhibition) were evaluated in adolescent maternal separated (MS) mice. Alterations of the corticotropin-releasing factor (CRF), glucocorticoid receptor (NR3C1), tyrosine hydroxylase (TH), mu-opioid receptor (MOr), brain-derived neurotrophic factor (BDNF), neuronal nuclei (NeuN), microtubule-associated protein 2 (MAP2) and neurofilament heavy (NF200)-immunoreactive fibers were studied in the paraventricular nucleus of the hypothalamus (PVN), ventral tegmental area (VTA), nucleus accumbens (NAc) or hippocampus (HIP). The effects of maternal separation (alone or in combination with additional stressful stimuli) on ethanol consumption during adolescence were evaluated using the oral ethanol self-administration paradigm. MS mice presented mood-related alterations and pre-attentional deficit. Increased CRF, MOr and TH, and reduced BDNF, NR3C1, NeuN, MAP2 and NF200-immunoreactive fibers were observed in the PVN, NAc and HIP of adolescent MS mice. In the oral ethanol self-administration test, adolescent MS mice presented higher ethanol consumption and motivation. Exposure to additional new stressful stimuli during adolescence significantly increased the vulnerability to ethanol consumption induced by maternal separation. These results clearly demonstrated that exposure to early life stress increased the vulnerability to ethanol consumption, potentiated the effects of stressful stimuli exposure during adolescence on ethanol consumption and modified the expression of key targets involved in the response to stress, ethanol reinforcing properties and cognitive processes.

Abstract:Background: Adolescent brains are highly vulnerable to heavy alcohol exposure. Increased understanding of how alcohol adversely impacts brain maturation may improve treatment outcomes. Objectives: This study characterizes short-term versus long-term effects of ethanol feeding on behavior, frontal lobe glial proteins, and mTOR signaling. Methods: Adolescent rats (8/group) were fed liquid diets containing 26% or 0% ethanol for 2 or 9 weeks, then subjected to novel object recognition (NOR) and open field (OF) tests. Frontal lobes were used for molecular assays. Results: Significant ethanol effects on OF performance occurred in the 2-week model ( p < .0001). Further shifts in OF and NOR performance were unrelated to ethanol exposure in the 9-week models ( p < .05 to p < .0001). Ethanol inhibited MAG1 ( p < .01) and MBP ( p < .0001) after 2 but not 9 weeks. However, both control and ethanol 9-week models had significantly reduced MAG1 ( p < .001–0.0001), MBP ( p < .0001), PDGFRA ( p < .05–0.01), and PLP ( p < .001–0.0001) relative to the 2-week models. GFAP was the only glial protein significantly inhibited by ethanol in both 2- ( p < .01) and 9-week ( p < .05) models. Concerning the mTOR pathway, ethanol reduced IRS-1 ( p < .05) and globally inhibited mTOR ( p < .01 or p < .001) in the 9- but not the 2-week model. Conclusions: Short-term versus long-term ethanol exposures differentially alter neurobehavioral function, glial protein expression, and signaling through IRS-1 and mTOR, which have known roles in myelination during adolescence. These findings suggest that strategies to prevent chronic alcohol-related brain pathology should consider the increased maturation-related vulnerability of adolescent brains.

Abstract:—Alcohol-related mental health problems, including an increased risk of development of anxiety-depressive disorders and alcohol addiction, are often passed down from generation to generation. Currently, there are no studies that consider the effect of prolonged alcohol exposure on the content of neurotransmitter monoamines and neurotransmitter amino acids in the brain structures responsible for the formation of emotional and motivational behavior patterns in the first F1 generation in the absence of direct alcohol exposure to the fetus. The aim of the present work was to study the anxiety behavior and neurochemical changes in the insular cortex and anterior cingulate cortex (ACC) in the intact offspring of rats that consumed ethanol solution for 30 weeks before mating. In the "elevated plus maze" test, females spent more time in open arms compared to males, however, no significant differences in the stress response between the offspring of rats exposed to and not exposed to ethanol were found. In ex vivo experiments, a decrease was observed for the insular cortex in the turnover of extracellular dopamine in animals of both sexes ( p < 0.05) and multidirectional shifts in the total turnover of dopamine in the absence of changes in the content of neurotransmitter amino acids. In the ACC, a decrease in the glycine content ( p < 0.05) was observed only in females. The data obtained indicate that prolonged oral administration of ethanol induces sex-dependent changes in the content of neurotransmitter monoamines and amino acids in the brain structures that determine the formation of emotional and motivational behavior in previously unaffected offspring.

Abstract:Adolescents primarily consume alcohol in binges, which can be particularly harmful to the developing frontal cortex and increase risk for an adult alcohol use disorder. We conducted a study investigating immediate and long lasting changes to the prefrontal cortex (PFC) transcriptome to determine the molecular mechanisms underlying adult ethanol behavioral sensitivity following binge ethanol in adolescence. DBA/2J mice were orally dosed with 4 g/kg ethanol intermittently from day 29 to 42. Adolescent mice were tested for anxiety-like behavior and ethanol sensitivity using the loss of righting reflex task. As adults, mice were tested for cognitive changes using the novel object recognition task, ethanol-induced anxiolysis and ethanol sensitivity. Adolescent binge ethanol altered ethanol sensitivity in young mice and led to lasting memory deficits in the object recognition test and greater ethanol sensitivity in adulthood. Using genomic profiling of transcripts in the PFC, we found that binge ethanol reduced myelin-related gene expression and altered chromatin modifying genes involved in histone demethylation at H3K9 and H3K36. We hypothesize that ethanol's actions on histone methylation may be a switch for future transcriptional changes that underlie the behavioral changes lasting into adulthood.

Abstract:Two studies of variables affecting voluntary ethanol consumption by adolescent male and female rats are reported. Sprague-Dawley (SD) and spontaneously hypertensive rats (SHRs) were compared in Experiment 1. Starting on postnatal day 30 all had 24-h access to 2%, then 4%, and then 6% ethanol, followed by 1-h access to the 6% until intake stabilized. During the 1-h access SHR females consumed more ethanol than all other groups. In Experiment 2, the same procedure was used to compare SD groups prenatally exposed to nicotine, with controls. Nicotine-exposed females consumed more ethanol during 1-h access than both nicotine-exposed and control males; but after using water intake as a covariate, the differences were not significant. These data show that deprivation conditions need to be considered when generalizing the results of voluntary consumption studies, and that estrogens may be a modulator of addictive behavior.
Sex-dependent effects of acute stress and alcohol exposure during adolescence on mRNA expression of brain signaling systems involved in reward and stress responses in young adult rats

Abstract:Background: Adolescent stress and alcohol exposure increase the risk of maladaptive behaviors and mental disorders in adulthood, with distinct sex-specific differences. Understanding the mechanisms underlying these early events is crucial for developing targeted prevention and treatment strategies. Methods: Male and female Wistar rats were exposed to acute restraint stress and intermittent alcohol during adolescence. We assessed lasting effects on plasma corticosterone (CORT) and adrenocorticotropic hormone (ACTH) levels, and mRNA expression of genes related to corticotropin releasing hormone (CRH), neuropeptide Y (NPY), corticoid, opioid, and arginine vasopressin systems in the amygdala and hypothalamus. Results: The main findings are as follows: (1) blood alcohol concentrations (BAC) increased after the final alcohol administration, but stressed males had lower BAC than non-stressed males; (2) Males gained significantly more weight than females; (3) Stressed females showed higher ACTH levels than non-stressed females, with no changes in males; (4) Stress increased CORT levels in males, while stressed, alcohol-treated females had lower CORT levels than non-stressed females; (5) CRH: Females had lower Crhr1 levels in the amygdala, while alcohol reduced Crhr2 levels in males but not females. Significant interactions among sex, stress, and alcohol were found in the hypothalamus, with distinct patterns between sexes; (6) NPY: In the amygdala, stress reduced Npy and Npy1r levels in males but increased them in females. Alcohol decreased Npy2r levels in males, with varied effects in females. Similar sex-specific patterns were observed in the hypothalamus; (7) Corticoid system: Stress and alcohol had complex, sex-dependent effects on Pomc, Nr3c1, and Nr3c2 in both brain regions; (8) Opioid receptors: Stress and alcohol blunted the elevated expression of Oprm1, Oprd1, and Oprk1 in the amygdala of males and the hypothalamus of females; (8) Vasopressin: Stress and alcohol interacted significantly to affect Avp and Avpr1a expression in the amygdala, with stronger effects in females. In the hypothalamus, alcohol increased Avp levels in females. Conclusions: This study demonstrates that adolescent acute stress and alcohol exposure induce lasting, sex-specific alterations in systems involved in reward and stress responses. These findings emphasize the importance of considering sex differences in the prevention and management of HPA dysfunction and psychiatric disorders.
Early-life inflammation increases ethanol consumption in adolescent male mice

Abstract:Recent studies have demonstrated that stress during the critical windows of development can evoke a cascade of neurological changes that can result in neuropsychiatric disorders later in life. In this study, we examined the effect of early-life inflammation on ethanol consumption in adolescent mice. C57BL/6J mice were assigned to either the control or Lipopolysaccharide (LPS) group on postnatal day 14 (P14). In the latter group, LPS at a dose of 50 μg/kg was injected intraperitoneally. The mice were weaned at P21, and behavior tests were performed at P45. Ethanol consumption was assessed using a two-bottle choice drinking paradigm. Anxiety-like behaviors were assessed by marble burying test (MBT), open field (OF), and elevated plus maze (EPM). Ethanol-induced loss of righting reflex (LORR), hypothermia and ethanol metabolism were assessed to evaluate ethanol intoxication. P14 LPS-injected adolescent male mice exhibited significantly increased ethanol preference and consumption, with a similar taste preference for saccharin and avoidance of quinine. The adolescent male mice showed increased anxiety-like behaviors in the OF and EPM tests, and an increased duration of LORR, without affecting the hypothermic effects of ethanol and ethanol metabolism. Interestingly, these behavioral changes were not obvious in female mice. In conclusion, our data indicate that early-life inflammation may be a risk factor for ethanol consumption in adolescents with greater changes observed in male mice. Significance Statement Our study is the first preclinical model to report the enhancement effect of early-life inflammation on ethanol consumption in adolescent male mice and our findings provide a valuable mouse model to examine the neurobiological mechanisms mediating the long-lasting effects of early-life inflammation on alcohol use disorders vulnerability.

EFFECTS OF NICOTINE EXPOSURE DURING ADOLESCENCE ON ETHANOL SELF-ADMINISTRATION IN ADULTHOOD AND nAChR EXPRESSION OF VTA ON RATS

Abstract:Objective:To observe the effects of nicotine exposure during adolescence on ethanol self-administration in adulthood and examine the effects on mRNA expression of some nAChR subunits in VTA;to explore the mechanisms of smoking during adolescence and alcoholism during adulthood.Methods:Wistar rats received a period of 10 day subcutaneous injections of nicotine(1.0 mg·kg-1/day,salt)(N group n=20)or of physiological saline(C group n=19)on PN35.Six rats from each group(CE group n=6,NE group n=6)were randomly selected,and VTA were separated after killed.The total RNA were extracted,mRNA of α4,α5,α7 and β2 nAChR subunit in VTA of each group were measured by real time-PCR.Beginning on PN60,the left rats(NA group n=14,CA group n=12)were induced to establish ethanol two-bottle free-choice self-administration by Samson sucrose fading program.Ethanol consumption,alcohol preference and total fluid intake were measured.Then the same target was measured by the same program.Results:There were no differences in ethanol consumption,alcohol preference and total fluid intake between nicotine and saline treatment during adolescence(P>0.05).The mRNA expressions of each nAChR subunit in VTA showed:(1)There were no differences on mRNA expression of α4,α5,α7 and β2 nAChR subunit between two exposure groups(P>0.05).(2)There were significant differences at different levels on mRNA expression of α4,α5,α7 and β2 nAChR subunits between two alcohol dependent model groups(P<0.05 or P<0.001).α4,α5,and β2 nAChR subunits were up-regulated and α7 nAChR subunit was down-regulated.Factorial ANOVA showed:(1)mRNA expression of α4,α5 was modulated by both factors.(2)β2 was modulated by alcohol dependent model factor.(3)There were interactions between two factors on mRNA expression of α5,α7 and β2 nAChR subunits.Conclusion:(1)The mRNA expressions of α4,α5,α7 and β2 nAChR subunits in VTA can be modulated by nicotine exposure during adolescence.This effect will be found later ethanol intake in Wistar rats during adulthood was not increased by nicotine exposure during adolescence.(2)Both nicotine and ethanol affected on nAChR subtype comprised α4,α5,α7 or β2 subunits in VTA.Maybe it is one of many factors involved in the establishing process of alcohol dependence.

We have previously found that early life sleep disruption impairs social development and alters GABA neurons in the brain of adult prairie voles, a socially monogamous rodent that displays natural ethanol preference in the laboratory. However, it is unclear whether these effects on social behavior are due, in part, to overall anhedonia and/or altered behavioral response to stress. To address this question, litters containing prairie vole pups were sleep disrupted by gentle cage agitation for 7 consecutive days from postnatal days (P) 14 to 21 (early life sleep disruption, or ELSD group) or allowed to sleep undisturbed (Control). Adult voles underwent a 2-bottle choice ethanol drinking procedure integrated with a single session of footshocks. Ethanol intake after footshock was measured as well as c-Fos immunoreactivity in the lateral and central amygdala. ELSD animals showed increased ethanol consumption and increased neural activity in these amygdala regions after footshock compared to control animals. There were no differences in baseline ethanol drinking prior to exposure to a stressor. These results suggest that early life sleep disruption in prairie voles does not produce anhedonia but can have long-lasting effects on stress reactivity. In addition to shaping species-typical social behavior, early life sleep may be important in the development of stress induced ethanol consumption and the activation of limbic pathways associated with stress. Modeling the Multiple Sclerosis Brain Disease Using Agents: What Works and What Doesn't?

Abstract:The human brain is one of the most complex living structures in the known Universe. It consists of billions of neurons and synapses. Due to its intrinsic complexity, it can be a formidable task to accurately depict brain's structure and functionality. In the past, numerous studies have been conducted on modeling brain disease, structure, and functionality. Some of these studies have employed Agent-based approaches including multiagent-based simulation models as well as brain complex networks. While these models have all been developed using agent-based computing, however, to our best knowledge, none of them have employed the use of Agent-Oriented Software Engineering (AOSE) methodologies in developing the brain or disease model. This is a problem because without due process, developed models can miss out on important requirements. AOSE has the unique capability of merging concepts from multiagent systems, agent-based modeling, artificial intelligence, besides concepts from distributed systems. AOSE involves the various tested software engineering principles in various phases of the model development ranging from analysis, design, implementation, and testing phases. In this paper, we employ the use of three different AOSE methodologies for modeling the Multiple Sclerosis brain disease namely GAIA, TROPOS, and MASE. After developing the models, we further employ the use of Exploratory Agent-based Modeling (EABM) to develop an actual model replicating previous results as a proof of concept. The key objective of this study is to demonstrate and explore the viability and effectiveness of AOSE methodologies in the development of complex brain structure and cognitive process models. Our key finding include demonstration that AOSE methodologies can be considerably helpful in modeling various living complex systems, in general, and the human brain, in particular.
Sex Differences in Contextual Extinction Learning After Single Binge-Like EtOH Exposure in Adolescent C57BL/6J Mice

Abstract:The relationship between chronic heavy drinking and post-traumatic stress disorder (PTSD) is well-documented; however, the impact of more common drinking patterns, such as a single episode leading to a blood alcohol concentration (BAC) of 0.09 g/dL (moderate intoxication), remains underexplored. Given the frequent co-occurrence of PTSD and alcohol misuse, it is essential to understand the biological and behavioral factors driving this comorbidity. We hypothesize that alcohol's immediate sedative effects are coupled with the development of persistent molecular alcohol tolerance, which may disrupt fear extinction learning. To investigate this, we employed a Single Episode Ethanol (SEE) in-vivo exposure to mimic binge-like alcohol consumption over a 6-hour period, following contextual conditioning trials. Extinction trials were conducted 24 hours later to assess the effects on extinction learning. Our findings reveal a significant deficit in fear extinction learning in alcohol-treated adolescent male mice compared to saline-treated controls, with no such effects observed in female adolescent mice. These results suggest that even non-chronic alcohol exposure may contribute to the development of trauma- and stress-related disorders, such as PTSD, in males. Additionally, histological analysis revealed significant alterations in FKBP5, beta-catenin, and GSK-3beta levels in the hippocampus, striatum, and basolateral amygdala of alcohol-treated mice following extinction. The insights gained from this study could reshape our understanding of the risk factors for PTSD and open new avenues for prevention and treatment, targeting the molecular mechanisms that mediate alcohol tolerance.

Abstract:Background: Chronic alcohol drinking increases the risk of alcohol use disorders, causing various neurological disorders. However, the impact of different ethanol levels on a spectrum of behaviors during chronic drinking remains unclear. In this study, we established an intermittent access to ethanol in a two-bottle choice (IA2BC) procedure to explore the dose-dependent effects of ethanol on the behavioral performance of C57BL/6 J mice. Methods: Adult male C57BL/6 J mice were provided voluntary access to different ethanol concentrations (0 %, 5 %, 10 %, and 20 % ethanol) under a 12-week IA2BC paradigm. A battery of behavioral tests was administered to assess alterations in pain threshold, anxiety-like behaviors, locomotor activity, motor coordination, and cognition. Ethanol consumption and preference were monitored during each session. Moreover, the liver, heart, and lung tissues were examined using pathological microscopy. Results: The average (standard deviation) ethanol consumption of mice under the IA2BC paradigm increased dose-dependently to 5.1 (0.2), 8.7 (0.7), and 15.9 (0.8) g/kg/24 h with 5 %, 10 %, and 20 % ethanol, respectively. However, there is no significant difference in ethanol preference among all the ethanol groups. Chronic ethanol drinking caused hyperalgesia, cognitive impairment, and motor incoordination, but caused no changes in body temperature, locomotor activity, or anxiety-like behaviors. Minor histopathological alterations in the liver were detected; however, no major abnormal pathology was observed in the heart or lungs. Conclusion: These findings clarify the link between ethanol dosage and behavioral changes in mice over a 12-week IA2BC paradigm, thereby bridging the knowledge gap regarding the effects of chronic ethanol drinking on neurological disorders.

Abstract:Gliogenesis in the spinal cord was studied in chick embryos ranging in age from 3 to 14 days of incubation using H3‐thymidine autoradiography. It has been reported by the author that the neuron production of the spinal cord begins on the third day and ends at the eighth day of incubation. At eight days of incubation glioblasts with small dark round nuclei first appear in the sub‐pial region. Using the selective labeling technique with H3‐thymidine the author demonstrated that no transition exists from pial cells to sub‐pial glioblasts. The glioblasts are produced from the matrix cells by their migration and transformation. These glioblasts mature into neuroglia (oligodendroglia and astrocytes), as development proceeds. The suggestion that the stage of neuroglia differentiation follows that of neuron production in development of the central nervous system has been reconfirmed.

Abstract:BACKGROUND: Preclinical models simulating adolescent substance use leading to increased vulnerability for substance use disorders in adulthood are needed. Here, we utilized a model of alcohol and nicotine co-use to assess adult addiction vulnerability following adolescent alcohol exposure.METHODS: In Experiment 1, adolescent (PND30) male and female Sprague-Dawley rats received 25% ethanol (EtOH) or a control solution via oral gavage every 8 h, for 2 days. In young adulthood, animals were tested with a 2-bottle choice between H20% and 15% EtOH or 0.2% saccharin/15% EtOH, followed by co-use of oral Sacc/EtOH and operant-based i.v. nicotine (0.03 mg/kg/infusion) self-administration. In Experiment 2, adolescents received control gavage, EtOH gavage, or no-gavage, and were tested in young adulthood in a 2-bottle choice between H20% and 15% EtOH, Sacc/EtOH, or 0.2% saccharin.RESULTS: In Experiment 1, the adolescent EtOH gavage reduced adult EtOH consumption in the 2-bottle choice, but not during the co-use phase. During co-use, Sacc/EtOH served as an economic substitute for nicotine. In Experiment 2, the control gavage increased adult EtOH drinking relative to the no-gavage control group, an effect that was mitigated in the EtOH gavage group. In both experiments, treatment group differences in EtOH consumption were largely driven by males.CONCLUSIONS: EtOH administration via oral gavage in adolescence decreased EtOH consumption in adulthood without affecting EtOH and nicotine co-use. Inclusion of a no-gavage control in Experiment 2 revealed that the gavage procedure increased adult EtOH intake and that including EtOH in the gavage buffered against the effect.

Abstract:Key PointsPer occasion, alcohol use is higher among adolescents than adults in both humans and laboratory animals, suggesting that maturational changes in the adolescent brain contribute to the elevation in alcohol consumption seen at this time.Human prospective studies have revealed evidence that certain neural and cognitive or personality associates of heavy adolescent alcohol use may predate and serve as risk factors for such use.There is substantial evidence for notable neural and cognitive consequences of repeated exposure to alcohol during adolescence emerging from both human prospective studies and studies using rodent models. Although dependent measures often differ considerably across species, where comparable measures have been used, evidence for concordance in neural alterations has emerged across species, including disrupted myelination, poorer white matter integrity, alterations in connectivity between frontal and limbic regions and electroencephalographic changes.Studies have revealed that adolescent alcohol exposure is associated with various cognitive and other functional deficits. The consequences that have emerged in human longitudinal work include deficits in verbal learning, attention, and visuospatial and memory tasks, whereas studies using animal models of adolescent alcohol exposure have revealed that such exposure is linked to decreases in cognitive flexibility, behavioural inefficiencies, increases in anxiety-like behaviour, disinhibition, elevated risk-taking, augmented later voluntary consumption and motivation for alcohol, and the persistence of adolescent-like phenotypes into adulthood.In rodent studies, repeated exposure to alcohol during adolescence induces reliable and specific neural alterations that include decreases in cholinergic tone in the basal forebrain, neuroinflammation, disruptions in neurogenesis, epigenetic alterations and the persistence of certain adolescent-like neural characteristics into adulthood.There are numerous critical areas for future work in this area using human longitudinal designs as well as studies in laboratory animals — work that could benefit from an increasing focus on assessment of comparable measures across species. Studies examining functional consequences of adolescent alcohol exposure have largely used different measures to date, with a focus on neuropsychological or cognitive tests in humans contrasting with an emphasis on alterations in risk-taking, anxiety, and later motivation for and sensitivity to alcohol in rodent studies.
