Abstract:Modern drug delivery systems are designed for targeted controlled slow drug release. Up to now polymer based hydrogels have been applied in dentistry, which systems can affect the rate of the release due to their structure. Recently, intensive research for other methods is performed all over the world in order to improve the effectiveness of delivery systems. Nanotechnology is one of the most dynamically developing disciplines and is a powerful tool to increase the bioavailability of drugs. The aim of this work is to synthesise biocompatible nanoparticles by free radical initiated copolymerization of the monomers, 2-hydroxyethyl methacrylate (HEMA) and polyethyleneglycol dimethacrylate (PEGDMA) in aqueous solution, which can support the formation of nanoparticles that can be used as a drug delivery system for dental applications. The polymer-based nanoparticles were prepared via micellar polymerisation, which resulted a well dispersible white powder material. The size of particles was determined by Dynamic Laser Light Scattering (DLS) and Scanning Electron Microscopy (SEM). The size of particles is in range of 50-180 nm, measured by SEM. These values are commensurable with the results obtained by DLS experiments, where two size ranges were observed, as 40 +/- 15 nm and 180 +/- 30 nm. The nanoparticles are suitable for incorporation into a hydrogel matrix and to design new drug delivery devices for dental applications.
Cold tolerance in Campylobacter jejuni and its impact on food safety

Abstract:Campylobacter jejuni is a major cause of foodborne illnesses worldwide and is primarily transmitted to humans through contaminated poultry meat. To control this pathogen, it is critical to understand its cold tolerance because poultry products are usually distributed in the cold chain. However, there is limited information regarding how this thermotolerant, microaerophilic pathogen can survive in cold and aerobic environments in the poultry cold chain. In this study, we investigated the cold tolerance of C. jejuni by measuring the viability of 90 C. jejuni strains isolated from retail raw chicken at 4 °C under aerobic and microaerobic conditions. Despite the microaerophilic nature of C. jejuni, under aerobic conditions, C. jejuni exhibited higher viability at 4 °C and required an extended inactivation time compared to microaerobic conditions. Some strains were highly tolerant to refrigeration temperatures and exhibited increased survival at 4 °C. These cold-tolerant strains mostly belonged to multilocus sequence typing (MLST) clonal complex (CC)-21 and CC-443, indicating that cold tolerance is associated with the phylogeny of C. jejuni. Notably, cold-tolerant strains had an increased probability of illness and were more likely to cause human infections due to their extended survival on refrigerated chicken meat compared to those sensitive to cold stress. Furthermore, the majority of cold-tolerant strains exhibited elevated aerotolerance, indicating that cold tolerance is related to aerotolerance. These findings suggest that refrigeration of chicken meat under aerobic conditions may not be effective at controlling C. jejuni and that cold-tolerant C. jejuni can pose an increased risk to food safety.
The risk of aerotolerant Campylobacter jejuni strains in poultry meat distribution and storage

Abstract:Poultry meat is a major vector for Campylobacter jejuni foodborne illness. Since C. jejuni is microaerophillic, the cell counts gradually decreased during distribution and display under aerobic condition. However, if the pathogen can resist to aerobic condition, it may cause serious problem in food safety. This study determined the aerotolerance of C. jejuni isolated from poultry and the risk of the aerotolerant C. jejuni strains. Fourteen C. jejuni strains isolated from poultry were subjected to aerobic condition in a shaking incubator at 500 rpm and 37 °C. The cell counts of C. jejuni strains were enumerated on modified CCDA-Preston at 0, 24, 72 and 120 h, and the strains, having reduction less than 2 Log CFU/mL for 120 h were determined as aerotolerant C. jejuni strain. Non-aerotolerant and aerotolerant C. jejuni strains were then incubated at 4 °C under aerobic condition to compare the growth between aerotolerant and non-aerotolerant strains. In addition, transcriptomes for virulence and stress response genes (cadF, cdtB, ciaB, clpP and htrB) were compared between non-aerotolerant and aerotolerant C. jejuni strains. Among 14 C. jejuni strains, seven strains (50%) showed less than 2-Log CFU/mL reduction at 37 °C after 24 h, and five strains of them still showed less than 2-Log CFU/mL reduction after 48 h. Especially, C. jejuni SMFM2015-Du7 and C. jejuni SMFM2014-Du16 were still survived after 120 h under aerobic condition, which were then determined as aerotolerant C. jejuni. However, at 4 °C under aerobic condition, there were no significant differences in the reduction of C. jejuni cell counts and virulence gene expressions between non-aerotolerant (C. jejuni SMFM2014-Du8) and aerotolerant strains (C. jejuni SMFM2015-Du7 and C. jejuni SMFM2014-Du16) except for stress response gene (htrB). These results indicate that there are aerotolerant C. jejuni strains, but their risk is similar to non-aerotolerant C. jejuni strains at 4 °C under aerobic condition, which is distribution and storage condition for poultry meat. Therefore, additional food safety management for the aerotolerant C. jejuni is not necessary.

Abstract:We read the article titled “Importance of epicardial adipose tissue thickness measurement in obese adolescents, its relationship with carotid intima-media thickness, and echocardiographic findings” by Boyraz et al1 with great interest. However, we would like to make a few suggestions regarding the analysis and presentation of data. First, in Table I the standard deviation (SD) of some parameters, for example HOMA-IR, LDL-C and triglyceride levels are so large that it renders interpretation of data ambiguous. Such large SD would bring the normal distribution of the data into question, which in turn, would alter the selection of statistical tests. Also, the authors1 indicate that unpaired Student’s t-test was used to evaluate the three groups within the cohort; namely lean, mild-moderate obese and severe obese patients, in pairs. However, in cases with more than three independent groups, the preferred methodology must be an Analysis of Variance (ANOVA) followed by post-hoc analysis if statistical significance is achieved. This further complicates interpretation of the p-values, which we would have preferred to be provided in a separate column next to the data analyzed; rather than at the bottom of the table. Second, the title of Table III is “Tissue Doppler echocardiography measurements in lean, and mildmoderate and severely obese groups”. However, there is no data provided regarding lean patients. Furthermore, instead of measurements, a bunch of “r” correlation coefficients and their respective pvalues are provided. In another words, the title of the table and the contents are irrelevant. Last, although the multivariate regression analysis of the epicardial adipose tissue thickness is provided in Table IV, their correlation with measurements made within groups is not present. We are deducing that Table III was actually aiming to provide the correlations between epicardial adipose thickness and other measured parameters. Is this the case? We believe that Boyraz et al1 selected a great topic for exploration and would like to congratulate them for their efforts in shedding more light on this important matter.

Abstract:Vaccinia virus growth factor (VGF) is encoded by an early gene located near the border between the terminal inverted repeats and the internal unique region of the viral genome. It has been suggested that secreted VGF may serve to stimulate the metabolism of neighboring uninfected cells, thereby increasing the capacity of these cells to support viral replication. In this study, we cloned and sequenced the VGF gene of vaccinia virus strain Tian Tap. The analytical results indicated that VGF of strain Tian Tan possessed the core sequences of Epidermal growth factor superfamily, and more closely related to that of strain WR than to those of other orthopoxvirus strains. Overall deduced amino acid divergence of 4.3% was observed between Tian Tan and WR VGF protein, while divergences ranged from 10% to 20% compared to other orthopoxviruses strains. Most of the amino acid substitutions were found within signal peptide and transmembrane regions, but were not present in secreted ligand domain. It was striking that the amino acid sequence deduced from the ORF TB22L which located within the right terminal region of genome was identical to VGF protein amino acids 67-140, suggesting that ORF TB22L may be derived from the seconed copy of VGF gene of strain Tian Tan.

Abstract:Campylobacter jejuni is one of the most common causes of foodborne human gastroenteritis in the developed world. This bacterium colonizes in the ceca of chickens, spreads throughout the poultry production chain, and contaminates poultry products. Despite numerous on farm intervention strategies and developments in post-harvest antimicrobial treatments, C. jejuni is frequently detected on broiler meat products. This indicates that C. jejuni is evolving over time to overcome the stresses/interventions that are present throughout poultry production and processing. The development of aerotolerance has been reported to be a major survival strategy used by C. jejuni in high oxygen environments. Recent studies have indicated that C. jejuni can enter a viable but non-culturable (VBNC) state or develop biofilm in response to environmental stressors such as refrigeration and freezing stress and aerobic stress. This review provides an overview of different stressors that C. jejuni are exposed to throughout the poultry production chain and the genotypic and phenotypic survival mechanisms, with special attention to aerotolerance, biofilm formation, and development of the VBNC state.

Abstract:Campylobacter spp. constitute a group of microaerophilic bacteria that includes strains that are aerotolerant and capable of surviving in aerobic conditions. Recent studies have shown that aerotolerant strains are highly prevalent in meats, animals, and clinical settings. Changes in growth media and other environmental conditions can affect the aerotolerance of Campylobacter strains and must be considered when studying their aerotolerance in vitro. Polymicrobial interactions and biofilms also play a significant role in the ability of Campylobacter to survive oxygen exposure. Continuous subculturing may foster aerotolerance, and studies have demonstrated a positive correlation between aerotolerance and virulence and between aerotolerance and the ability to survive stressful environmental conditions. Various mechanisms and genetic origins for aerotolerance have been proposed; however, most of the potential genes involved in aerotolerance require further investigation, and many candidate genes remain unidentified. Research is also needed to investigate if there are any clinical implications for Campylobacter aerotolerance. Understanding the aerotolerance of Campylobacter remains an important target for further research, and it will be an important step towards identifying potential targets for intervention against this clinically important food-borne pathogen.

Effect of biofilm formation in a hostile oxidative stress environment on the survival of Campylobacter jejuni recovered from poultry in Iraqi markets

Abstract:Background and Aim: Campylobacter jejuni is a major contributor to bacterial enteritis, a common health problem. The resistance of this microaerophilic bacterium to oxidative stress allows it to thrive under aerobic conditions. This study aimed to investigate whether the capacity of C. jejuni to form biofilms in the presence of oxidative stress contributes to the pathogen’s ability to thrive in agricultural settings as well as in chicken slaughter lines. Materials and Methods: Twenty identified strains originating from chicken samples (eight from caeca contents and 12 from frozen chicken carcasses) were previously isolated and identified according to standard bacteriological protocols, followed by confirmation at the species level using multiplex polymerase chain reaction assay. Crystal violet staining was used to evaluate biofilm formation by these bacteria. Two exposure periods to gaseous ozone (1 and 2 min) were used to assess resistance to oxidative damage. Results: Most of the strong biofilm-forming Campylobacter strains came from imported frozen chicken meat (25%), whereas only 10% came from caeca content. After exposure to gaseous ozone at 600 mg/h for 2 min, strong biofilm-producing strains exhibited a higher survival rate with a limited reduction of up to 3 logs, whereas negative biofilm-producing strains exhibited a limited survival rate with a reduction of 6 logs. Conclusion: Based on our findings, we hypothesized that the presence of C. jejuni strains capable of forming biofilms in poultry farms and/or chicken production facilities triggers a public health alarm as this bacterium seems to be able to adapt more easily to live and thrive in hostile environmental conditions. Abstract:Background: Campylobacter jejuni is a leading cause of bacterial enteritis worldwide. This microaerophilic bacterium can survive in aerobic environments, suggesting it has protective mechanisms against oxidative stress. The clinical C. jejuni Bf strain is characterized by an increased resistance to oxygen. This study aimed to characterize the behavior of the clinical C. jejuni Bf strain under an aerobic atmosphere and in response to ROS-promoter agents. Methods: Growth was studied in both aerobic and microaerobic conditions using classic cultivable methods. Electronic microscopy and mreB gene expression were used to evaluate the morphology of this strain under aerobic conditions. The survival under oxidative stress was tested in the presence of different concentrations of hydrogen peroxide (H2O2) and paraquat (PQ). Results: The results showed that C. jejuni Bf strain can grow aerobically, unlike other strains of C. jejuni tested. Cells of C. jejuni Bf exposed to oxidative stress presented changes in morphology and the gene mreB, responsible for maintaining the bacillary cell morphology, was down-expressed. In aerobically acclimated conditions, C. jejuni Bf exhibited a higher survival rate of 52 % in the presence of H2O2 (1 mM) compared to the reference strain NCTC 11168. Concentrations above 1 mM PQ were lethal for the reference strain but not for C. jejuni Bf. Conclusions: Taken together, these data highlight the resistance to oxidative stress conditions of C. jejuni Bf, indicating that this microorganism seems more adapted to survival in hostile environmental conditions.

Abstract:The activation of the mitogen-activated protein (MAP) kinases by different environmental stresses has been previously observed in several dicot plant species. Here, we report the isolation of a novel MAP kinase in rice that is induced during infection by the blast fungus Magnaporthe grisea or upon mechanical wounding. The gene is designated as BWMK1 for blast- and wound-induced MAP kinase. The cDNA of BWMK1 was isolated from rice leaves challenged by the blast pathogen. Transcripts of the corresponding gene accumulated in rice leaves 4 h after blast inoculation and 30 min after mechanical wounding. This gene encodes a 506 amino acid protein that contains a new dual-phosphorylation activation motif TDY and about 150 unique amino acids on its C terminus. In-gel kinase activity and immunoprecipitation assays confirmed that BWMK1 is a functional MAP kinase. These results show that BWMK1 is a new member of the plant MAP kinase family and may mediate both defense and wound signaling in rice.
Oxygen requirement and tolerance of Campylobacter jejuni

Abstract:The pathogen Campylobacter jejuni is considered a microaerophile yet has been shown to grow in vitro in atmospheres with partial oxygen tension of 21%. To achieve a better understanding of its microaerophily, the oxygen requirement and tolerance of four C. jejuni strains were investigated by measuring their growth under different conditions, by performing bioinformatic analyses and by determining their metronidazole resistance. At high cell densities, C. jejuni showed similar growth under microaerobic and fully aerobic oxygen tensions, but did not grow under oxygen-depleted conditions. At low cell densities, the bacteria grew only under microaerobic conditions. Eighteen genes were identified bioinformatically as potential contributors to the differences in oxygen tolerance between strains. Among them, cj0203, cj0264c, cj0415, cj0425, cj0628, cj0629 and cj0864 were considered the top potential contributors. The oxygen tolerance of the four C. jejuni strains was different, and this tolerance positively correlated with their resistance to metronidazole. This study provided evidence that C. jejuni was an obligate microaerophile. The data indicated that the strains had different oxygen tolerances; it suggested that they could result in phenotypic and physiological differences between strains grown under the same conditions. These differences could modulate the outcome of experiments, and may explain discrepancies in the results between strains.
Quantification of Campylobacter jejuni gene expression after successive stresses mimicking poultry slaughtering steps

Abstract:Broiler meat is considered as the most important source of the foodborne pathogen Campylobacter jejuni. Exposure to stress conditions encountered during the slaughtering process may induce bacterial adaptation mechanisms, and enhance or decrease pathogen resistance to subsequent stress. This adaptation may result from changes in bacterial gene expression. This study aims to accurately quantify the expression of selected C. jejuni genes after stresses inspired from the poultry slaughtering process. RT-qPCR was used to quantify gene expression of 44 genes in three strains after successive heat and cold stresses. Main results indicated that 26 genes out of 44 were differentially expressed following the successive thermal stresses. Three clusters of genes were differentially expressed according to the strain and the stress condition. Up-regulated genes mainly included genes involved in the heat shock response, whereas down-regulated genes belonged to metabolic pathways (such as lipid, amino-acid metabolisms). However, four genes were similarly overexpressed in the three strains; they might represent indicators of the thermal stress response at the species scale. Advances in the molecular understanding of the stress response of pathogenic bacteria, such as Campylobacter, in real-life processing conditions will make it possible to identify technological levers and better mitigate the microbial risk.
Differential carbon source utilization by Campylobacter jejuni 11168 in response to growth temperature variation

Abstract:Campylobacter spp. readily colonize the intestinal tracts of both human and avian species. While most often commensal organisms in birds, campylobacters remain the leading cause of bacterial gastroenteritis in humans. The association of campylobacters with poultry is well established as a primary route for human exposure. The difference in normal core body temperature between chickens (42 degrees C) and humans (37 degrees C) has been suggested to trigger potential colonization or virulence factors and investigators have demonstrated differential gene expression at the two temperatures. Campylobacter spp. exhibit unique nutritional requirements and have been thought to only utilize amino acids and Kreb cycle intermediates as carbon sources for growth. We evaluated the ability of the genome-sequenced strain of Campylobacter jejuni 11168 (GS) to oxidize 190 different substrates as sole carbon sources at 37 degrees C and 42 degrees C using phenotype microarray (PM) technology. Results indicate that the expected amino acids, l-serine, l-aspartic acid, l-asparagine, and l-glutamic acid were utilized in addition to a number of organic acids. In general, oxidation of the substrates was greater at 42 degrees C than at 37 degrees C with a few exceptions. By employing the PM method, we observed a number of potential false-positive reactions for substrates including the triose, dihydroxyacetone; and the pentose sugars, d-xylose, d-ribose, l-lyxose, and d- and l-arabinose. The presence of genes possibly responsible for utilization of pentose sugars is supported by the genomic sequence data, but actual utilization as sole carbon sources for active respiration has not been observed. A better understanding of the metabolic pathways and nutritional requirements of campylobacters could lead to improvements in culture media for detection and isolation of the pathogen and to future intervention methods to reduce human exposure.
Identification of a novel stress resistance mechanism in Campylobacter jejuni

Abstract:Aim: To study stress resistance mechanisms in Campylobacter spp. Methods and results: Campylobacter strains were grown to the appropriate phase in Brucella broth. The cells were diluted into either cell-free spent medium (obtained by filtration of a grown culture) or a freshly prepared medium and the pH reduced to 4.5, a lethal pH value. At suitable time intervals survivors were enumerated on Campylobacter blood free selective agar base. The cell-free spent medium from mid-exponential and stationary phase had a protective effect on acid and thermal stress in Campylobacter jejuni CI 120, a natural isolate. The protective effect of the extracellular compound was not significantly inactivated by boiling, but was inactivated by proteinase. Conclusions: The present study suggests that a protein (or proteins) accumulated by C. jejuni CI 120 during growth may play an active role in the induction of stress responses and that this protein is heat stable. Significance and impact of the study: The results indicate that C. jejuni CI 120, a natural isolate, has the ability to use extracellular signalling mechanisms to induce tolerance to stress factors. This is a major advancement in the understanding of the physiological basis for survival of C. jejuni in the environment.
