Abstract:Chitosan, obtained from chitin in crustacean exoskeletons, has attracted interest due to its biocompatibility and versatility of application in several industries. Chemical methods are traditional for chitosan production but present environmental challenges. This study comprehensively assesses the microwave-assisted method as a greener alternative compared to conventional deacetylation. Morphological, thermal, and chemical properties, yield, and environmental impacts are evaluated. Experimental results indicate microwave-assisted deacetylation produces chitosan with excellent properties, with a high degree of deacetylation (close to 90%) and yield (above 50%). The Life Cycle Impact Assessment (LCIA) study highlights fossil resource scarcity, climate change, human non-carcinogenic toxicity, and terrestrial ecotoxicity as some of the most relevant environmental impacts, considering the Brazilian scenario. Notably, the difference in reaction time (240 vs 16 min) indicates the substantial advantage of the non-conventional technique in terms of reaction time, also reducing the energy consumption during deacetylation from 382.1 to 8.9 kJ per gram of chitosan produced by conventional and microwave-assisted methods respectively. However, LCIA shows NaOH use during deacetylation as a notable contributor to environmental effects, highlighting the necessity for minimizing its presence. Despite this, microwave integration leads to diminished environmental impact. This research highlights the environmental benefits of microwave-assisted deacetylation for more sustainable chitosan production.

Abstract:A new method for quick chitin isolation from the shells of crab, crayfish and shrimp is described. The main difference between the new method and the conventional method is two sodium hypochlorite (NaClO) treatments for 10 min each before the processes of demineralisation and deproteinisation. After the NaClO treatment, only 15 min is adequate for the demineralisation and 20 min for the deproteinisation processes. Newly extracted chitin from crab, crayfish and shrimp shells and commercial chitin were characterised using FT-IR, TGA, X-ray diffractometry and elemental analysis. From the results, it was observed that the chitins isolated with the new method and the commercial chitin had almost the same physicochemical properties. The advantage of the new method compared to traditional methods is the relatively rapid chitin extraction. When compared to the traditional chitin extraction method, the proposed method appears to be promising regarding its time and energy saving nature.

Study on optimization of ultrasonic wave-assisted extraction of chitin from shrimp shell

Abstract:The factors of demineralization and deproteinization were discussed by the single-factor experiment and orthogonal experiment to choose the optimum extraction process from shrimp shell with ultrasonic wave.The results showed that the best demineralization condition were as follows:room temperature,ultrasonic frequency 60kHz,ultrasonic power 180W,concentration of citric acid 14%,liquid-solid ratio 12:1(mL/g),reaction time 22min;the best deproteinization condition were reaction temperature 60℃,ultrasonic frequency 60kHz,ultrasonic power 100W,concentration of NaOH 5%,liquid-solid ratio 5:1(mL/g),reaction time 20min.The quality parameters of the product extracted in the optimum process were nitrogen content 5.92%,ash content 0.04%,water content 3.8%,yield 21.3%.

Abstract:Chitin is one of the most abundant bioactive biopolymer on earth. It is commercially extracted from seafood processing crustacean shell byproducts by harsh thermochemical treatments. The extraction conditions, the source and pretreatment of raw material significantly affect its quality and bioactivity. In this investigation response surface methodology (RSM) has been applied to optimize and evaluate the interaction of variables for extraction of high quality chitin from shrimp processing raw byproducts. Variables such as, concentration of HCl (%, v/v) 4.5 (for wet) and 4.9 (for dry), reaction time 3 h, solid liquid ratio of HCl (w/v) 1:5.5 (for wet) and 1:7.9 (for dry) with two treatments achieved >98 % demineralization of shrimp byproduct. Variables such as, concentration of NaOH 3.6 % (w/v), reaction time 2.5 h, temperature 69.0 ± 1 °C, solid liquid ratio of NaOH 7.4 (w/v) and two treatments accomplished >98 % deproteinization of demineralized byproducts. Significant (p ≤ 0.05–0.001) interactive effects were observed between different variables. Chitin obtained in these conditions had residual content (%, w/w) of ash <0.4 and protein <0.8 and the degree of N-acetylation was >93 % with purity of >98 %. In conclusion, the optimized conditions by RSM can be applied for large scale preparation of high quality chitin from raw shrimp byproduct.

Abstract:A facile, solid-state mechanochemical method was developed for the synthesis of low molecular weight chitosan (LMWC) in one step from chitin and crude shrimp shell powders, in which chitin undergoes simultaneous deacetylation and depolymerisation in the presence of a base catalyst under mechanical milling. The method is advantageous over traditional multi-step methods, featuring enhanced efficiency and significantly reduced environmental impact. Compared to traditional approaches, the base usage is reduced to about 1/10, and the molecular weight of the obtained LMWC product is much more narrowly distributed, with a polydispersity value of only 1.1. The degree of deacetylation and the molecular weight can be adjusted by varying the ball milling parameters. The influences of different types of bases were investigated by solid-state NMR analysis and control experiments, pointing out the critical role of base in both depolymerisation and deacetylation. Base-catalysed mechanochemical transformation of chitin and shrimp shells provides a solvent-free way to effectively valorize shellfishery waste for valuable products.

Abstract:To work out the technical problems of the preparation methods and equipments of chitosan oligosaccharide,the paper creatively proposed the integrated reaction and separation technology which is integrated electrochemical oxidation degradation of chitosan with continuous preparative chromatography separation and extraction of chitosan oligosaccharide.It can be used to obtain chitosan oligosaccharide under high-efficiency,energy-saving and green production.With the numerical simulation and a large number of experiments,it is shown that 120 minutes is the best coupling point between the reaction and the separation.Based on them, an integrated reaction and separation equipment,Coupled/cascade Simulated Moving Bed was developed.The results of practical tests showed that the purity of the product can reach to more than 98%,its energy-savings can be up to 25%and the emissions of elution solvent can be significantly reduced.The method and equipment has a high performance and potential application.

Abstract:Chitin is the second most common polymer after cellulose in earth, existing in the shells of crustaceans like crab and shrimp. Chitosan is a natural amino-polysaccharide derived from chitin, known as one of the most abundant organic materials in nature, it has been widely used in several applications due to its natural origin and exceptional properties such as biocompatibility, biodegradability, non-toxicity, and chelating of metal ions. Chitin and chitosan are characterized by deacetylation degree, one of the most important chemical characteristics that can influence the performance of chitosan in many applications. Chitosan is usually prepared by a thermochemical method, consuming time, energy and reagents. In this review, various methods of chitosan extraction will be approached and compared; the importance of a new method of ecological extraction will be emphasized. Moreover, in order to improve the chitosan functionality, and better control these physicochemical properties, several chemical modifications have been reported. These chemical modifications lead to a broad range of derivatives with a wide range of applications in many fields. Recent examples of the distinct applications, with particular emphasis on environmental applications, have been presented.

Abstract:Extraction and depolymerisation of chitin and chitosan from shrimp waste material was carried out using fish proteases aided process. A high deproteinization level (80 %) was recorded with an Enzyme/Substrate ratio of 10 U/mg. The demineralization of shrimp waste was completely achieved within 6 h at room temperature in HCl 1.25 M, and the residual content of calcium in chitin was below 0.01 %. The degree of N-acetylation, calculated from the 13C CP/MAS-NMR spectrum, was 85 %. The chitin obtained was converted to chitosan by N-deacetylation. X-ray diffraction patterns also indicated two characteristics crystalline peaks approximately at 10° and 20° (2θ). Chitosan was then evaluated in the treatment of unhairing effluents from the tanning industry. A result showed that chitosan as a coagulant has good performance in alkaline pH and at concentration of 0.5 g/L. Within these conditions, chitosan could decrease turbidity value, total suspended solids (89 % at 1.5 g/L), biological oxygen demand (33.3 % at 1.5 g/L) and chemical oxygen demand (58.7 % at 1.5 g/L).

Abstract:Received : 04/11/2016 Accepted :10/06/2017 Abstract: Demineralization (DM) and deproteinization (DP) of shrimp shell Parapenaeus longirostris using Lactobacillus helveticus, depends on the composition of culture medium, and temperature of incubation. In a synthetic medium containing glucose, completely different conditions are required for chitin recovery: 300 g/L of glucose and 35°C for maximal DM (60%) and 80 g/L of glucose and 30°C for a maximum of DP (70%). The use of date’s juice shows that it is possible to extract chitin, in a single step, unlike the simple medium. The richness of the dates juice in mineral elements and the high concentration of reducing sugars (200 g/L) allowed for a maximum activity of proteolytic enzymes favored by a pH maintained constant at around pH 6 for 29h of fermentation, and a significant acidification expressed by a minimum pH of 4.7 which are reflected by rates of 83% and 63% of deproteinization and demineralization, respectively.

Abstract:Chitosan, which is a derivative of chitin, is particularly popular due to its biodegradable and renewable nature. However, the properties of chitosan can be inconsistent due to the extraction process and its natural origin, which poses a challenge to its use in composite materials as a matrix. The properties of chitosan can be tuned by controlling the degree of deacetylation (the extent to which acetyl groups are removed from chitin to form chitosan) and molecular weight. This paper presents a detailed study on the extraction and characterization of chitosan from shrimp shells. The structural thermal and mechanical characterization were studied using several techniques: Fourier-transform infrared spectroscopy, X-ray diffraction, thermogravimetric analysis and differential scanning calorimetry. The intrinsic viscosity and deacetylation degree were determined using various methods. The results showed an average degree of deacetylation of 77%. The chitosan films exhibited a high tensile strength of 43.9 MPa and an elongation at break of 3.14%. The thermal analysis revealed that the films had a glass transition temperature of 88 °C and a maximum thermal degradation temperature of 320 °C. The findings of this research could contribute to the development of chitosan-based materials with improved properties, leading to its wider adoption in the future for composite matrix application. The simple and efficient method used for the extraction and purification of chitosan from shrimp shells makes it a cost-effective and eco-friendly alternative to synthetic polymers.

Abstract:Shrimp shells are a key source of chitin, commonly extracted through chemical methods, which may cause minor molecular damage. Nowadays, there is great interest in achieving close to zero protein content in crude chitin in order to use it for high-end markets. Therefore, this study optimized the enzymatic deproteination using two commercial proteases (SEB Pro FL100 and Sea-B Zyme L200) for effective and fast removal of residual protein from Northern shrimp (Pandalus borealis) shell chitin for the first time. The protein content was determined using both the Kjeldahl method and amino acid analysis using gas chromatography–mass spectrometry (GC-MS). The performance of papain (Sea B Zyme L200) was superior to fungal protease (SEB Pro FL100) for this application, and it achieved residual protein content of 2.01%, while the calculated optimum for the latter enzyme was 6.18%. A model was developed using 24 factorial design, and it was predicted that the lowest residual protein content using fungal protease and papain could be achieved at the following conditions: a pH of 4.2 and 7, and an enzyme concentration of 4 and 1.5%, respectively. Thus, the low-protein content obtained using enzymatic deproteination could be an alternative approach to the traditional methods, indicating their potential to produce premium-quality chitin.

Abstract:Chitin, an abundant biopolymer with potential applications in agriculture, medicine, and bioremediation, is conventionally extracted using chemical methods that have environmental disadvantages. This study investigates the extraction of chitin from Litopenaeus vannamei shrimp waste by one-step fermentation using the bacterial strains Pseudomonas aeruginosa QF50 and Serratia sp. QCS23. A total of 4 kg of shrimp waste was treated by fermentation with culture media enriched with different concentrations of glucose (1, 5, and 10%) for 7 days at 25 °C, followed by purification and characterization processes using infrared spectroscopy and X-ray diffraction. The results demonstrated an increase in the yield of crude chitin proportional to the glucose concentration, reaching a maximum of 76.81 ± 7.64% for Pseudomonas aeruginosa QF50 and 71.30 ± 1.16% for Serratia sp. QCS23. Both strains showed high efficiencies in deproteinization (80–87%) and demineralization, with significant improvements especially shown at high glucose concentrations. Structural characterization confirmed the presence of the spectral characteristics of α-chitin, with crystallinity indices of 81% and 71% for chitins obtained with Pseudomonas aeruginosa QF50 and Serratia sp. QCS23, respectively. This study concludes that single-step fermentation with Pseudomonas aeruginosa QF50 and Serratia sp. QCS23 is an effective and sustainable method for the extraction of high-quality chitin from shrimp exoskeleton waste, offering a promising alternative to traditional chemical methods.

Abstract:Chitosan is a biopolymer material extracted from marine biomass waste such as shrimp and crab shells, which has good biocompatibility and degradability with great potential for application in the field of wastewater treatment and soil remediation. The higher the degree of deacetylation (DD), the better the adsorption performance of chitosan. Chitin deacetylase (CDA) can specifically catalyze the deacetylate of chitin in a green reaction that is environmentally friendly. However, the scarcity of high yielding chitin deacetylase strains has been regarded as the technical bottleneck of chitosan green production. Here, we screened a natural chitin degrading bacterium from coastal mud and identified it as Bacillus cereus ZWT-08 by re-screening the chitin deacetylase activity and degree of deacetylation values. By optimizing the medium conditions and enzyme production process, ZWT-08 was cultured in fermentation medium with 1% (m/V) glucose and yeast extract at pH 6.0, 37°C, and a stirring speed of 180 r/min. After fermenting in 5 L fermenter for 48 h, the deacetylation activity of the supernatant reached 613.25 U/mL. Electron microscopic examination of the chitin substrate in the fermentation medium revealed a marshmallow-like fluffy texture on its structural surface. Meanwhile, 89.29% of the acetyl groups in this chitin substrate were removed by enzymatic digestion of chitin deacetylase produced by ZWT-08, resulting in the preparation of chitosan a degree of deacetylation higher than 90%. As an effective strain for chitosan production, Bacillus cereus ZWT-08 plays a positive role in the bioconversion of chitin and the upgrading of the chitosan industry.

Abstract:Chitin extraction from shrimp shell powder (SSP) using protease-producing microbes is an attractive approach for valorizing shrimp shell waste because it is simple and environmentally friendly. In this study, the protease production and chitin extraction from SSP by Bacillus cereus HMRSC30 were simultaneously optimized using statistical approaches. As a result, fermentation in medium composed of 30 g/L SSP, 0.2 g/L MgSO4 · 7H2O, 3 g/L (NH4)2SO4, 0.5 g/L K2HPO4, and 1.5 g/L KH2PO4 (pH 6.5) for 7 days maximized protease production (197.75 ± 0.33 U/mL) to approximately 1.64-fold compared to unoptimized condition (126.8 ± 0.047 U/mL). This level of enzyme production was enough to achieve 97.42 ± 0.28% deproteinization (DP) but low demineralization (DM) of 53.76 ± 0.21%. The high DM of 90% could be easily accomplished with the post-treatment using 0.4 M HCl and acetic acid. In addition, the study evaluated the possible roadmap to maximize the value of generated products and obtain additional profits from this microbial process. The observation showed the possibility of serving crude chitin as a bio-adsorbent with the highest removal capacity against Coomassie brilliant blue (97.99%), followed by methylene blue (74.42%). The recovered protease exhibited the function to remove egg yolk stain, indicating its potential for use as a detergent in de-staining. The results corroborated the benefits of microbial fermentation by B. cereus HMRSC30 as green process for comprehensive utilization of shrimp shell waste as well as minimizing waste generation along the established process.

Abstract:Chitin has been extracted from six different local sources in Egypt. The obtained chitin was converted into the more useful soluble chitosan by steeping into solutions of NaOH of various concentrations and for extended periods of time, then the alkali chitin was heated in an autoclave which dramatically reduced the time of deacetylation. Chitin from squid pens did not require steeping in sodium hydroxide solution and showed much higher reactivity towards deacetylation in the autoclave that even after 15 min of heating a degree of deacetylation of 90% was achieved. The obtained chitin and chitosan were characterized by spectral analysis, X-ray diffraction and thermo gravimetric analysis.

Abstract:Chitin is the most productive nitrogen-containing polysaccharide in nature with immense potential for transforming into a range of chemicals. However, its dense crystal structure poses a challenge for depolymerization, limiting its applications. To overcome these challenges, a novel series of deep eutectic solvents (DESs) based on benzyltrimethylammonium chloride (TMBAC) as the hydrogen bond acceptor was developed. These TMBAC-based DESs, in combination with lactic acid, oxalic acid, and malic acid as the hydrogen bond donor demonstrated efficient chitin dissolution, achieving a solubility of up to 12% and an 88% recovery rate of regenerated chitin. The regenerated chitin was characterized using XRD, FT-IR, SEM, and 13 C CP-MAS NMR, which indicated the preservation of chitin's chemical structure, a significant decrease in crystallinity, and a reduction in the molecular weight. Furthermore, the enzymatic hydrolysis efficiency of chitin was nearly doubled after treatment with TMBAC-based DESs, surpassing the effectiveness of untreated chitin. This approach holds promise for facilitating subsequent transformation and utilization of chitin.

Abstract:Chitosan is an extremely valuable polysaccharide and usually obtained from crustacean byproduct. In this work, a new non-animal source chitosan is obtained from fungal precursors (Ganoderma lucidum spore powders, GLSP) using ultrasound-assisted deacetylation (USAD), and the effects of processing parameters (irradiation time, solid-to-liquid ratio, NaOH concentration, and irradiation power) on the DD (degree of deacetylation) of chitosan were investigated. The highest DD value was obtained at irradiation parameters of 20 min, 10% (g:ml) NaOH, 1:25 (g:ml), and 80 W. In addition, the difference in surface morphology, chemical groups, thermal stability, and crystallinity of the resulting chitosan with the treatment of various irradiation times was examined using the SEM, FTIR, TG, and XRD. The results show a new promising herbal source of edible chitosan and the potential material for biomedical applications. Practical applications The chitosan obtained from Ganoderma lucidum spore powders (GLSP) possessed many priorities over the chitosan from shells of crustaceans, which involves a multi-faceted procedure with several economical and consumer related aspects. In the current work, chitosan is obtained from an herbal source GLSP using ultrasound treatment and the effect of processing parameters on chitosan was investigated. The findings show the development of chitosan from an herbal source with potential in functional food applications.
Simultaneous Optimization of Deacetylation Degree and Molar Mass of Chitosan from Shrimp Waste

Abstract:Shrimp waste is a valuable source for chitin extraction and consequently for chitosan preparation. In the process of obtaining chitosan, a determining step is the chitin deacetylation. The main characteristic of chitosan is the degree of deacetylation, which must be as high as possible. The molar mass is another important parameter that defines its utilizations, and according to these, high or low molar masses are required. The present study is an attempt to optimize the deacetylation step to obtain chitosan with a high degree of deacetylation and high or low molar mass. The study was carried out based on experimental data obtained in the frame of a central composite design where three working parameters were considered: NaOH concentration, liquid:solid ratio, and process duration. The regression models defined for the degree of deacetylation (DD) and for the mean molar mass (MM) of chitosan powders were used in the formulation of optimization problems. The objectives considered were simultaneous maximum DD and maximum/minimum MM for the final chitosan samples. For these purposes, multiobjective optimization problems were formulated and solved using genetic algorithms implemented in Matlab®. The multiple optimal solutions represented by trade-offs between the two objectives are presented for each case.

Abstract:The extraction of chitin and chitosan presents challenges due to the complexity of the process and the influence of many variables. This study aimed to optimize chitin and chitosan extraction from Fusarium verticillioides by analyzing many additives and processing variables and modeling their yields using multiple linear regression (MLR) and evolutionary algorithms. FT-IR analysis confirmed the presence of characteristic bands in the extracted samples, and SEM analysis further revealed the microfibrillar appearance of the chitin and the dense, non-porous structure of the chitosan. The Ant Lion Optimizer (ALO) was employed to select significant factors and optimize model parameters. A transformation was applied to capture nonlinear relationships, and the fine-tuned models showed improved predictive power, with p-values of 0.00203 for chitin and 0.00884 for chitosan. Multi-objective optimization (MOO) using the Adaptive Geometry Estimation-based Multi-Objective Evolutionary Algorithm (AGE-MOEA) further identified significant factors for optimal yields, achieving 3 g of Arginine, 100 ml of culture medium volume, 7 to 11 days of incubation time, 0.2 to 1.76 ml of Oligochitin, 1.4 g of FeSO4, 1.5 g of K2HPO4, and 1 g of NaCl. Therefore, the integration of ALO and AGE-MOEA algorithms effectively modeled and optimized chitin and chitosan yields by maximizing biopolymer recovery, enabling significant industrial exploitation.
