Abstract:Bacterial infections occur frequently during the implantation of medical devices, and functional coating is one of the effective means to prevent and remove biofilms. In this study, three different hydrophilic polyoxazolines with carboxyl groups (aPOx: PT1, PT2 and PT3) and bactericidal silver nanoparticles (AgNPs) were synthesized successfully, and an aPOx-AgNP multilayer film was prepared by electrostatic layer-by-layer self-assembly. The effect of charge density and assembly solution concentration was explored, and the optimal self-assembly parameters were established (PT2 1 mg/mL and AgNPs 3 mg/mL). The hydrophilicity of the surface can be enhanced to resist protein adhesion if the outermost layer is aPOx, and AgNPs can be loaded to kill bacteria, thereby realizing the bactericidal anti-adhesion potential integration of the aPOx-AgNP multilayer film. In addition, the aPOx-AgNP multilayer film was found to have the characteristic of intelligent and efficient pH-responsive silver release, which is expected to be used as a targeted anti-biofilm surface of implantable medical devices.
Photoactive MOF-Derived Bimetallic Silver and Cobalt Nanocomposite with Enhanced Antibacterial Activity

Abstract:Conventional antibiotic-based treatment of bacterial infections remains one of the most difficult challenges in medicine because of the threat of multidrug resistance caused by indiscriminate abuse. To solve these problems, it is essential to develop an effective antibacterial agent that can be used at a small dose while minimizing the occurrence of multiple resistance. Metal-organic frameworks (MOFs), which are hyper-porous hybrid materials containing metal ions linked by organic ligands, have recently attracted attention because of their strong antibacterial activity through metal-ion release, unlike conventional antibiotics. In this study, we developed a photoactive MOF-derived cobalt-silver bimetallic nanocomposite (Ag@CoMOF) by simply depositing silver nanoparticles on a cobalt-based MOF through nanoscale galvanic replacement. The nanocomposite structure continuously releases antibacterial metal ions (i.e., Ag and Co ions) in the aqueous phase and exhibits a strong photothermal conversion effect of Ag nanoparticles, accompanied by a rapid temperature increase of 25-80 °C under near-infrared (NIR) irradiation. Using this MOF-based bimetallic nanocomposite, superior antibacterial activities were achieved by 22.1-fold for Escherichia coli and 18.3-fold for Bacillus subtilis enhanced inhibition of bacterial growth in a liquid culture environment compared with the generally used chemical antibiotics. In addition, we confirmed the synergistic enhancement of the antibacterial ability of the bimetallic nanocomposite induced by NIR-triggered photothermal heating and bacterial membrane disruption even when using a small amount of the nanocomposites. We envision that this novel antibacterial agent using MOF-based nanostructures will replace traditional antibiotics to circumvent multidrug resistance and present a new approach to antibiotic development.
Surface-Catalyzed Nitric Oxide Release via a Metal Organic Framework Enhances Antibacterial Surface Effects

Abstract:It has been previously demonstrated that metal nanoparticles embedded into polymeric materials doped with nitric oxide (NO) donor compounds can accelerate the release rate of NO for therapeutic applications. Despite the advantages of elevated NO surface flux for eradicating opportunistic bacteria in the initial hours of application, metal nanoparticles can often trigger a secondary biocidal effect through leaching that can lead to unfavorable cytotoxic responses from host cells. Alternatively, copper-based metal organic frameworks (MOFs) have been shown to stabilize Cu2+/1+ via coordination while demonstrating longer-term catalytic performance compared to their salt counterparts. Herein, the practical application of MOFs in NO-releasing polymeric substrates with an embedded NO donor compound was investigated for the first time. By developing composite thermoplastic silicon polycarbonate polyurethane (TSPCU) scaffolds, the catalytic effects achievable via intrapolymeric interactions between an MOF and NO donor compound were investigated using the water-stable copper-based MOF H3[(Cu4Cl)3(BTTri)8-(H2O)12]·72H2O (CuBTTri) and the NO donor S-nitroso-N-acetyl-penicillamine (SNAP). By creating a multifunctional triple-layered composite scaffold with CuBTTri and SNAP, the surface flux of NO from catalyzed SNAP decomposition was found tunable based on the variable weight percent CuBTTri incorporation. The tunable NO surface fluxes were found to elicit different cytotoxic responses in human cell lines, enabling application-specific tailoring. Challenging the TSPCU-NO-MOF composites against 24 h bacterial growth models, the enhanced NO release was found to elicit over 99% reduction in adhered and over 95% reduction in planktonic methicillin-resistant Staphylococcus aureus, with similar results observed for Escherichia coli. These results indicate that the combination of embedded MOFs and NO donors can be used as a highly efficacious tool for the early prevention of biofilm formation on medical devices.
Metal-Organic Framework/Ag-Based Hybrid Nanoagents for Rapid and Synergistic Bacterial Eradication

Abstract:Recent emerged metal-organic frameworks (MOFs), as superior drug carriers, provide novel strategies to combat pathogenic bacterial infections. Although various antibacterial metal ions can be easily introduced in MOFs for chemical bacterial ablation, such a single-model bactericidal method suffers from high-dose use, limited antibacterial efficiency, and slow sterilization rate. Hence, developing a dual bactericidal system is urgently required. Herein, we report an MOF/Ag-derived nanocomposite with efficient metal-ion-releasing capability and robust photo-to-thermal conversion effect for synergistic sterilization. The MOF-derived nanocarbon consisting of metallic zinc and a graphitic-like carbon framework is first synthesized, and then Ag nanoparticles (AgNPs) are evenly introduced via the displacement reaction between Zn and Ag+. Upon near-infrared irradiation, the fabricated nanoagents can generate massive heat to destroy bacterial membranes. Meanwhile, abundant Zn2+ and Ag+ ions are released to make chemical damage to bacterial intracellular substances. Systematic antibacterial experiments reveal that such dual-antibacterial effort can endow the nanoagents with nearly 100% bactericidal ratio for highly concentrated bacteria at a very low dosage (0.16 mg/mL). Furthermore, the nanoagents exhibit less cytotoxicity, which provides potential possibilities for the applications in the biological field. In vivo assessment indicates that the nanocomposites can realize rapid and safe wound sterilization and are expected to be an alternative to antibiotics. Overall, we present an easily fabricated structure-engineered nanocomposite with chemical and photothermal effects for broad-spectrum bacterial sterilization.

Abstract:A controllable antibacterial Ag@MOF@PDA is constructed by functionalizing cyclodextrin metal–organic frameworks with polydopamine (PDA) and ultrafine Ag NPs. The PDA coating enables its long‐term bactericidal capability by gradual releasing of Ag+, and NIR‐mediated hyperthermia can accelerate its release rate, which realizes a synergistic chemical‐photothermal capacity to combat bacteria and biofilm infections, representing a potential antibiotic‐free alternative in the "post‐antibiotic era." Bacterial infections pose a significant threat to global public health; therefore, the development of novel therapeutics is urgently needed. Herein, a controllable antibacterial nanoplatform utilizing cyclodextrin metal–organic frameworks (CD‐MOFs) as a template to synthesize ultrafine silver nanoparticles (Ag NPs) in their porous structure is constructed. Subsequently, polydopamine (PDA) is encapsulated on the CD‐MOFs' surface via dopamine polymerization to enhance the water stability and enable hyperthermia capacity. The resulting Ag@MOF@PDA generates localized hyperthermia and gradually releases Ag+ to achieve long‐term photothermal‐chemical bactericidal capability. The release rate of Ag+ can be accelerated by NIR‐mediated heating in a controllable manner, quickly reaching the effective concentration and reducing the frequency of medication to avoid potential toxicity. In vitro experiments demonstrate that the combined antibacterial strategy can not only effectively kill both gram‐negative and gram‐positive bacteria, but also directly eradicate mature biofilms. In vivo results confirm that both bacterial‐ and biofilm‐infected wounds treated with a combination of Ag@MOF@PDA and laser exhibit satisfactory recovery with minimal toxicity, displaying a superior therapeutic effect compared to other groups. Together, the results warrant that the Ag@MOF@PDA realizes synergistic antibacterial capacity and controllable release of Ag+ to combat bacterial and biofilm infections, providing a potential antibiotic‐free alternative in the "post‐antibiotic era."

Robust Chiral Metal-Organic Framework Coatings for Self-Activating and Sustainable Biofouling Mitigation

Abstract:Surface coatings are designed to mitigate pervasive biofouling herald, a new era of surface protection in complex biological environments. However, existing strategies are plagued by persistent and recurrent biofilm attachment, despite the use of bactericidal agents. Herein, a chiral metal-organic framework (MOF)-based coating with conformal microstructures to enable a new anti-biofouling mode that involves spontaneous biofilm disassembly followed by bacterial eradication is developed. A facile and universal metal-polyphenol network (MPN) is designed to robustly anchor the MOF nanoarmor of biocidal Cu2+ ions and anti-biofilm d-amino acid ligands to a variety of substrates across different material categories and surface topologies. Incorporating a diverse array of chiral amino acids endows the resultant coatings with widespread signals for biofilm dispersal, facilitating copper-catalyzed chemodynamic reactions and inherent mechano-bactericidal activities. This synergistic mechanism yields unprecedented anti-biofouling efficacy elucidated by RNA-sequencing transcriptomics analysis, enhancing broad-spectrum antibacterial activities, preventing biofilm formation, and destroying mature biofilms. Additionally, the chelation-directed amorphous/crystalline coatings can activate photoluminescent properties to inhibit the settlement of microalgae biofilms. This study provides a distinctive perspective on chirality-enhanced antimicrobial behaviors and pioneers a rational pathway toward developing next-generation anti-biofouling coatings for diverse applications.
Silver‐Infused Porphyrinic Metal–Organic Framework: Surface‐Adaptive, On‐Demand Nanoplatform for Synergistic Bacteria Killing and Wound Disinfection

Abstract:The fabrication of functional nanoplatforms for combating multidrug‐resistant bacteria is of vital importance. Among them, silver nanoparticles (Ag NPs) have shown an antibacterial effect; however, the remainder cores of Ag NPs after use might have a toxic effect on humans. Thus, Ag ions based materials have been fabricated to substitute Ag NPs for antibacterial applications. Nevertheless, the always‐on release state leads to the low biocompatibility, which limits their biomedical applications. In addition, the single effect also restricts their antibacterial ability. Herein, a powerful surface‐adaptive, on‐demand antimicrobial nanoplatform is fabricated by coating hyaluronic acid (HA) on Ag ions loaded photosensitive metal‐organic frameworks to exhibit a strong synergistic effect. The nanoplatform shows good biocompatibility with nontargeted cells, as negatively charged HA can prevent the release of Ag ions. While in the presence of targeted bacteria, the secreted hyaluronidase can degrade HA on the nanoplatform and produce positively charged nanoparticles, which display increased affinity to bacteria and show a strong synergistic antibacterial effect owing to the released Ag ions and generated reactive oxygen species under visible light irradiation. Importantly, due to the outstanding on‐demand antimicrobial performance, the nanoplatform also shows great effects on treating multidrug‐resistant bacteria infected wounds in mice models.
Porphyrin MOF Dots–Based, Function‐Adaptive Nanoplatform for Enhanced Penetration and Photodynamic Eradication of Bacterial Biofilms

Abstract:Recently, antimicrobial photodynamic therapy (aPDT) has been considered as an attractive treatment option for biofilms ablation. However, even very efficient photosensitizers (PSs) still need high light doses and PS concentrations to eliminate biofilms due to the limited penetration and diffusion of PSs in biofilms. Moreover, the hypoxic microenvironment and rapid depletion of oxygen during PDT severely limit their therapeutic effects. Herein, for the first time, a porphyrin‐based metal organic framework (pMOF) dots–based nanoplatform with effective biofilm penetration, self‐oxygen generation, and enhanced photodynamic efficiency is synthesized for bacterial biofilms eradication. The function‐adaptive nanoplatform is composed of pMOF dots encapsulated by human serum albumin–coated manganese dioxide (MnO2). The pH/H2O2‐responsive decomposition of MnO2 in biofilms triggers the release of ultra‐small and positively charged pMOF dots and simultaneously generates O2 in situ to alleviate hypoxia for biofilms. The released pMOF dots with high reactive oxygen species yield can effectively penetrate into biofilms, strongly bind with bacterial cell surface, and ablate bacterial biofilms. Importantly, such a nanoplatform can realize great therapeutic outcomes for treatment of Staphylococcus aureus–infected subcutaneous abscesses in vivo without damage to healthy tissues, which offers a promising strategy for efficient biofilms eradication.

Abstract:Size of the nanoparticles plays a very crucial role in their bactericidal activity as smaller size render better perforation via bacterial cell wall and are thus found to be more effective against resistant bacterial infections also. In this regard, fluorescent silver nanoclusters (Ag NCs) having a size less than 2 nm ought to be a potent bactericidal agent against both Gram positive and Gram negative bacterial strains. However, stabilization of such small sized NPs in aqueous medium that is amenable for biological applications is very challenging. Herein, a simple and fast method of synthesis of Ag NCs was proposed using metal–organic framework (MOFs) as scaffold, which was found to be very stable in the ambient conditions. As-synthesized Ag NCs-in-MOF nanocomposite was characterized by UV–Vis spectroscopy, transmission electron microscopy (TEM), and X-ray photoelectron spectroscopy (XPS). The nanocomposite was highly bactericidal against both Gram positive and Gram negative bacterial strains and caused cell wall perforation that was evident from flow cytometry analysis. Results also showed that DNA damage had occurred due to treatment of Ag NCs-in-MOF, ultimately leading to bacterial cell death with considerably lower dose of silver.
Novel coating containing molybdenum oxide nanoparticles to reduce Staphylococcus aureus contamination on inanimate surfaces

Abstract:We previously synthetized molybdenum oxide (MoO3) nanoparticles (NP) and showed their antibacterial activity against a representative collection of the most relevant bacterial species responsible for hospital-acquired infections, including Staphylococcus aureus. The aim of the present study was to prepare and characterize a novel coating with these MoO3 NP, confirm its mechanical stability, and investigate its biocidal effect to reduce S. aureus contamination on inanimate surfaces. In addition, the novel MoO3 NP coating was compared to a silver (Ag) NP coating synthetized by the same procedure. The MoO3 and Ag NP coatings were characterized in terms of their chemical structure by FT-IR, surface morphology by scanning electron microscopy, and mechanical properties by tensile and adhesion tests. The antimicrobial activity of the coatings was tested by following the loss of viability of S. aureus after 6h, 24h, 48h, and 72h exposure. MoO3 and Ag coatings exhibited surfaces of comparable morphologies and both presented elastomeric properties (tensile strength of ~420 kPa, Young's modulus of ~48 kPa, and maximum elongation of ~12%), and excellent (classification of 5B) adhesion to glass, steel and polystyrene surfaces. The two coatings exhibited a good antibacterial activity (R) against S. aureus over time (RMoO3 = 0.2-0.81; RAg = 0.61-2.37), although the effect of the Ag NP coating was more pronounced, especially at 72h (RMoO3 = 0.81 vs RAg = 2.37). Noteworthy, contrary to the Ag NP coating, the MoO3 NP coating was colourless and transparent, avoiding undesired unaesthetic effects. The synthetized coating with NP of MoO3, which has low toxicity to humans, capability of biodegradation, and rapid excretion, can be applied onto most standard materials and therefore is a promising tool to reduce S. aureus contamination on usual inanimate surfaces found in healthcare and community environments.

Abstract:A novel metal–organic framework (MOF) based on Zn(II) ion as metal node was synthesized from tetrazole (HAT) and 1-benzimidazole-5-carboxylic acid (H2Bmic) as well as Zn(NO3)2·6H2O in DMF-water mixed solvent, and its chemical formula is [Zn2(Bmic)2(HAT)2](DMF)3 (1, DMF = N,N-dimethylformamide). The results show that the obtained activated 1a has catalytic activity for aromatic aldehydes cyanation under the existence of the trimethylsilyl cyanide (TMSCN), thus providing trimethylsilyl cyanohydrin in the form of shape along with size selection. In addition, a feasible synergistic mechanism of 1a for cyanosilylation of aldehyde also has been put forward. Furthermore, the treatment effect of the compound against Staphylococcus aureus on burn patients has been evaluated. Firstly, the inhibitory effect of the compound on the Staphylococcus aureus biofilm formation was evaluated in vitro via crystal violet staining experiment. Then, the treatment activity of the compound in vivo was detected by counting the colony-forming units (CFU) numbers in the local tissue of skin infection.

Abstract:The design of self-sterilizing surfaces with favorable biocompatibility is acknowledged as an effective approach to deal with the bacterial infections of biomedical devices. In this study, we report an intriguing protocol for the large-scale fabrication of self-sterilizing and biocompatible surface film coatings by using polymer shielded silver nanoparticle loaded oxidized carbon nanotube (AgNPs@oCNT) nano-dispersions. To achieve the antibacterial coatings, the bioinspired positively charged and negatively charged AgNPs@oCNTs were alternately deposited onto substrates by spray-coating assisted layer-by-layer assembly. Then the bacterial inhibitory zones, optical density value monitoring, bacterial killing efficiency and adhesion were investigated; and all the results revealed that the AgNPs@oCNTs thin film coatings exhibited robust and long-term antibacterial activity against both Gram negative and Gram positive bacteria. Moreover, due to the shielding effects of polymer layers, the coatings showed extraordinary blood compatibility and limited toxicity against human umbilical vein endothelial cells. It is believed that the proposed large-scale fabrication of bactericidal, blood and cell compatible AgNPs@oCNT based thin film coatings will have great potential to forward novel operational pathogenic inhibition strategies to avoid undesired bacterial contaminations of biomedical implants or biological devices.STATEMENT OF SIGNIFICANCE: Bacterial infection of medical devices has been considered to be a world-wide clinical threat towards patients' health. In this study, a bioinspired and biocompatible antibacterial coating was prepared via the spray-assisted layer-by-layer (LbL) assembly. The silver nanopartilces loaded oxidized carbon nanotube (AgNPs@oCNT), which were coated by functional polymers (chitosan and synthetic heparin mimicking polymers), were prepared via mussel inspired chemistry; and the spray-assisted assembly process allowed the fast construction on devices. Owing to the antibacterial efficiency of the loaded AgNPs, the coating showed robust bacterial killing activity and resistance towards bacterial adhesion. Moreover, since that the AgNPs were shielded by the polymers, the coating exhibited no clear toxicity at blood or cellular level. Benefiting from the universal and large-scale fabrication advancements of the spray assisted LbL coating; it is believed that the proposed strategy can be applied in designing many other kinds of self-sterilizing biomedical implants and devices.

Enhanced electrochemical expansion of graphite for in situ electrochemical functionalization.

Abstract:An all electrochemical route to functionalized graphene directly from a graphite electrode is described herein obviating the need for defect inducing oxidative or prolonged sonication treatments. Enhanced electrochemical expansion of graphite is achieved by sequential treatment, beginning with the established method of expansion by electrolysis in a Li(+) containing electrolyte, and then with the much larger tetra-n-butylammonium. The result is a hyperexpansion of the graphite basal planes. As a demonstration of the utility of this method, we successfully performed a subsequent in situ electrochemical diazonium functionalization of the hyperexpanded graphite basal planes to give functional graphene sheets. This potential controlled process is more effective than chemical processes and also provides a means of controlling the degree of functionalization. We have further demonstrated that the functionalized graphene could be converted to a pristine low defect form via laser ablation of the funtional groups. As a result, this method presents a potentially scalable approach for graphene circuit patterning.

Abstract:To address the serious threat of bacterial infection to public health, great efforts have been devoted to the development of antimicrobial agents for inhibiting bacterial growth, preventing biofilm formation, and sterilization. Very recently, metal–organic frameworks (MOFs) have emerged as promising materials for various antimicrobial applications owing to their different functions including the controlled/stimulated decomposition of components with bactericidal activity, strong interactions with bacterial membranes, and formation of photogenerated reactive oxygen species (ROS) as well as high loading and sustained releasing capacities for other antimicrobial materials. This review focuses on recent advances in the design, synthesis, and antimicrobial applications of MOF-based materials, which are classified by their roles as component-releasing (metal ions, ligands, or both), photocatalytic, and chelation antimicrobial agents as well as carriers or/and synergistic antimicrobial agents of other functional materials (antibiotics, enzymes, metals/metal oxides, carbon materials, etc.). The constituents, fundamental antimicrobial mechanisms, and evaluation of antimicrobial activities of these materials are highlighted to present the design principles of efficient MOF-based antimicrobial materials. A new MOF@bioactive glass composite reinforced with silver nanoparticles – a new approach to designing antibacterial biomaterials

Abstract:Multifunctional materials that combine antimicrobial properties with the ability to stimulate bone formation are needed to overcome the problem of infected bone defects. As a novel approach, a new composite based on bioactive glass nanoparticles in a simple system of SiO 2 –CaO (BG) coated with NH 4 [Cu 3 (μ 3 -OH)(μ 3 -4-carboxypyrazolato) 3 ] (Cu-MOF) with additionally anchored silver nanoparticles (AgNPs) was proposed. Ag@Cu-MOF@BG obtained by the spin coating approach in the form of a disc was characterized using PXRD, ATR-FTIR, XPS, ICP-OES, and TEM. Importantly, the material retained its bioactivity, although ion exchange in the bioactive glass administered as a disc is limited. Hydroxyapatite (HA) formation was identified in TEM images after 7 days of immersion of the composite in a physiological-like buffer (pH 7.4, 37 °C). The Cu and Ag contents of Ag@Cu-MOF@BG were as low as 0.013 and 0.018 wt% respectively, but the slow release of the AgNPs ensured its antibacterial nature. Ag@Cu-MOF@BG exhibited antibacterial activity against all tested bacteria ( E. coli , S. aureus , P. aeruginosa , and K. pneumoniae ) with the diameter of the inhibition zones of their growth between 8 and 10 mm and the reduction index determined to be ≥3. Moreover, the biocompatibility of the new composite has been demonstrated, as shown by cell culture assays with human dermal fibroblasts (HDFs). The results from the migration test also proved that the HDF cell's phenotypic properties were not changed, and the cell adhesion and migration ability were the same as in control indirect assays.

Abstract:Silver nanoparticles (Ag NPs), a commonly used antibacterial nanomaterial, exhibit broad-spectrum antibacterial activity to combat drug-resistant bacteria. However, the Ag NPs often causes a low availability and high toxicity to living bodies due to their easy aggregation and uncontrolled release of Ag+ in the bacterial microenvironment. Here, we report a porous metal−organic framework (MOF)-based Zr-2-amin-1,4-NH2-benzenedicarboxylate@Ag (denoted as UiO-66-NH2-Ag) nanocomposite using an in-situ immobilization strategy where Ag NPs were fixed on the UiO-66-NH2 for improving the dispersion and utilization of Ag NPs. As a result, the reduced use dose of Ag NPs largely improves the biosafety of the UiO-66-NH2-Ag. Meanwhile, after activation by the Ag NPs, the UiO-66-NH2-Ag can act as nanozyme with high peroxidase (POD)-like activity to efficiently catalyze the decomposition of H2O2 to extremely toxic hydroxyl radicals (·OH) in the bacterial microenvironment. Simultaneously, the high POD-like activity synergies with the controllable Ag+ release leads to enhanced reactive oxygen species (ROS) generation, facilitating the death of resistant bacteria. This synergistic antibacterial strategy enables the low concentration (12 μg/mL) of UiO-66-NH2-Ag to achieve highly efficient inactivation of ampicillin-resistant Escherichia coli (AmprE. coli) and endospore-forming Bacillus subtilis (B. subtilis). In vivo results illustrate that the UiO-66-NH2-Ag nanozyme has a safe and accelerated bacteria-infected wound healing.

Abstract:In this work, we demonstrate a convenient, efficient, and environmentally benign strategy to achieving antimicrobial and antiadhesive purposes using a silver-zwitterion nanocomposite. The synthesis of the nanocomposite relies on loading zwitterionic polymer brushes with Ag+ precursor ions, followed by their in situ reduction to Ag nanoparticle by ultraviolet (UV) irradiation. Both poly(sulfobetaine methacrylate) (pSBMA) and poly(carboxybetaine methacrylate) (pCBMA) have been studied as matrices for the embedding of silver. Well-dispersed silver nanoparticles are embedded into pCBMA matrices. The obtained pCBMA-silver hybrid (CB-Ag) is capable of killing bacteria upon contact and releasing dead bacteria under wet conditions. Results suggest the feasibility of using this nanocomposite system as a robust and reliable antimicrobial and antiadhesive platform for the prevention of microbial colonization.
Ag-doped magnetic metal organic framework as a novel nanostructured material for highly efficient antibacterial activity

Abstract:In the last decades, numerous attempts have been made to prevent microbial pollution spreading, using antibacterial agents. Zeolitic imidazolate framework-8 (ZIF-8) belongs to a subgroup of metal organic frameworks (MOFs) merits of attention due to the zinc ion clusters and its effective antibacterial activity. In this work, Ag-doped magnetic microporous γ-Fe2O3@SiO2@ZIF-8-Ag (FSZ-Ag) was successfully synthesized by a facile methodology in room temperature and used as an antibacterial agent against the growth of the Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus bacteria. Several characterization methods were applied to analyze the properties of the materials, and the results confirmed the accuracy of the synthesis procedure. Silver ions have employed to enhance the efficiency of antibacterial activity. As the results illustrated, FSZ-Ag nanostructured material had superior performance to inactive E. coli and S. aureus in growth inhibition test in liquid media. The best antibacterial activity as minimum inhibitory concentration (MIC) was 100 mg/L of FSZ-Ag against both bacteria. Leaching rates of silver ions showed that 80% of Ag released in the solutions, which was responsible for inhibiting the growth of bacteria. Also, fluorescence microscopy was used to investigate bacterial viability after 20 h contacting FSZ-Ag to distinguish live and dead bacteria by staining with DAPI and PI fluorescence stains. This novel magnetic nanostructured material is an excellent promising candidate to use in biological applications as high potential bactericidal materials.

Abstract:In recent years, humans have been forced to create effective water treatment methods because of the increasing water contamination caused by various industrial activities. This study focuses on synthesizing a nanocomposite consisting of two metal-organic frameworks based on ZIF-8 with effective antibacterial characteristics and water-stable, highly porous UiO-66-NH 2 . After characterizing the synthesized nanocomposite, its application as (i) an adsorbent for methylene blue (MB) removal from a polluted aqueous environment and (ii) an antibacterial agent; was considered. The impact of different factors on the uptake of MB, including dye concentration, adsorption temperature, and the initial pH of the solution, was studied. When the temperature was raised to 45°C, the maximum adsorption capacity of UiO-66-NH 2 @ZIF-8 for MB improved to 215.78 mg/g. Moreover, the pseudo-second-order model accurately described the experimental MB uptake results. According to the thermodynamic analysis, the spontaneous endothermic adsorption process of MB onto the synthesized UiO-66-NH 2 @ZIF-8 was the consequence of an entropy impact instead of an enthalpy change. Additionally, the synthesized nanocomposite exhibited antibacterial efficacy against two different bacteria, gram-negative E. coli and gram-positive S. aureus , with a minimum inhibitory concentration (MIC) of 30 mg/L against both strains. The results indicated that the synthesized MOF@MOF nanocomposite is efficient in dye removal for water treatment and has excellent potential in disinfection applications. Abstract:The growth and spread of multidrug-resistant bacterial species, such as Klebsiella pneumoniae , pose a serious threat to human health and require the development of innovative antibacterial agents. The search for an acceptable, safe, and efficient antibacterial is a matter of significant concern. In the present work, silver-based metal–organic frameworks (Ag-MOFs) showed efficient antibacterial activity against multidrug-resistant K. pneumoniae (KBP 11) with a minimum inhibitory concentration and minimum bactericidal concentration of 10 μg mL −1 . Moreover, the Ag-MOF showed enhanced antibacterial activity compared to silver ions and silver nanoparticles. Our experimental investigation showed that the antibacterial efficacy is attributed to the production of reactive oxygen species and the release of cellular constituents, such as K + ions and proteins. The MOF scaffold enhances the stability and controlled release of silver ions, enabling sustained antibacterial activity and minimizing the risk of bacterial resistance development. Additionally, the MOF class, due to the high surface area and porous nature, enhances the transfer of bacteria into and on the surface of the MOF.
