Abstract:Understanding the mechanism by which patterned gene activity leads to mechanical deformation of cells and tissues to create complex forms is a major challenge for developmental biology. Plants offer advantages for addressing this problem because their cells do not migrate or rearrange during morphogenesis, which simplifies analysis. We synthesize results from experimental analysis and computational modeling to show how mechanical interactions between cellulose fibers translate through wall, cell, and tissue levels to generate complex plant tissue shapes. Genes can modify mechanical properties and stresses at each level, though the values and pattern of stresses differ from one level to the next. The dynamic cellulose network provides elastic resistance to deformation while allowing growth through fiber sliding, which enables morphogenesis while maintaining mechanical strength.

Abstract:Underlying the architectural complexity of plants are diverse cell types that, under the microscope, easily reveal relationships between cell structure and specialized functions. Much less obvious are the mechanisms by which the cellular growth machinery and mechanical properties of the cell interact to dictate cell shape. The recent combined use of mutants, genomic analyses, sophisticated spectroscopies, and live cell imaging is providing new insight into how cytoskeletal systems and cell wall biosynthetic activities are integrated during morphogenesis. The purpose of this review is to discuss the unique geometric properties and physical processes that regulate plant cell expansion, then to overlay on this mechanical system some of the recent discoveries about the protein machines and cellular polymers that regulate cell shape. In the end, we hope to make clear that there are many interesting opportunities to develop testable mathematical models that improve our understanding of how subcellular structures, protein motors, and extracellular polymers can exert effects at spatial scales that span cells, tissues, and organs.

Abstract:<p>Plant morphology emerges from cellular growth and structure. The turgor-driven diffuse growth of a cell can be highly anisotropic: significant longitudinally and negligible radially. Such anisotropy is ensured by cellulose microfibrils (CMF) reinforcing the cell wall in the hoop direction. To maintain the cell's integrity during growth, new wall material including CMF must be continually deposited. We develop a mathematical model representing the cell as a cylindrical pressure vessel and the cell wall as a fibre-reinforced viscous sheet, explicitly including the mechano-sensitive angle of CMF deposition. The model incorporates interactions between turgor, external forces, CMF reorientation during wall extension, and matrix stiffening. Using the model, we reinterpret some recent experimental findings, and reexamine the popular hypothesis of CMF/microtubule alignment. We explore how the handedness of twisting cell growth depends on external torque and intrinsic wall properties, and find that cells twist left-handedly 'by default' in some suitable sense. Overall, this study provides a unified mechanical framework for understanding left- and right-handed twist-growth as seen in many plants.</p>

Abstract:Computational analysis of cell walls Layers of intertwined fibers make up plant cell walls. The various types of fibers respond differently to deformation. Cellulose microfibrils, for example, can stretch or curve, changing their end-to-end length, and can also slide past each other, reorient relative directions, and bundle with neighboring microfibrils. Zhang et al. developed a computational model based on observations of onion skin epidermis that describes how these complex changes in space govern cell wall mechanics. Abstract:Abstract Studies on the mechanics of plant cells usually focus on understanding the effects of turgor pressure and properties of the cell wall (CW). While the functional roles of the underlying cytoskeleton have been studied, the extent to which it contributes to the mechanical properties of cells is not elucidated. Here, we study the contributions of the CW, microtubules (MTs) and actin filaments (AFs), in the mechanical properties of Nicotiana tabacum cells. We use a multiscale biomechanical assay comprised of atomic force microscopy and micro-indentation in solutions that (i) remove MTs and AFs and (ii) alter osmotic pressures in the cells. To compare measurements obtained by the two mechanical tests, we develop two generative statistical models to describe the cell’s behaviour using one or both datasets. Our results illustrate that MTs and AFs contribute significantly to cell stiffness and dissipated energy, while confirming the dominant role of turgor pressure.

Abstract:Plant cell growth is regulated through manipulation of the cell wall network, which consists of oriented cellulose microfibrils embedded within a ground matrix incorporating pectin and hemicellulose components. There remain many unknowns as to how this manipulation occurs. Experiments have shown that cellulose reorients in cell walls as the cell expands, while recent data suggest that growth is controlled by distinct collections of hemicellulose called biomechanical hotspots, which join the cellulose molecule together. The enzymes expansin and Cel12A have both been shown to induce growth of the cell wall; however, while Cel12A's wall-loosening action leads to a reduction in the cell wall strength, expansin's has been shown to increase the strength of the cell wall. In contrast, members of the XTH enzyme family hydrolyse hemicellulose but do not appear to cause wall creep. This experimentally observed behaviour still awaits a full explanation. We derive and analyse a mathematical model for the effective mechanical properties of the evolving cell wall network, incorporating cellulose microfibrils, which reorient with cell growth and are linked via biomechanical hotspots made up of regions of crosslinking hemicellulose. Assuming a visco-elastic response for the cell wall and using a continuum approach, we calculate the total stress resultant of the cell wall for a given overall growth rate. By changing appropriate parameters affecting breakage rate and viscous properties, we provide evidence for the biomechanical hotspot hypothesis and develop mechanistic understanding of the growth-inducing enzymes.

Abstract:During plant development, the mechanical properties of the cell walls must be tuned to regulate the growth of the cells. Cell growth involves significant stretching of the cell walls, yet little is known about the mechanical properties of cell walls under such substantial deformation, or how these mechanical properties change to regulate development. Here, we investigated the mechanical behavior of the Arabidopsis leaf epidermal cells being stretched. We found that the mechanical properties arise from the cell wall, which behaves as a fibrous network material. The epidermis exhibited a non-linear stiffening behavior that fell into three regimes. Each regime corresponded to distinct nonlinear behaviors in terms of transverse deformation (i.e., Poisson effect) and unrecoverable deformation (i.e., plasticity). Using a model, we demonstrated that the transition from reorientation and bending-dominated to stretch-dominated deformation modes of cellulose microfibrils cause these nonlinear behaviors. We found the stiffening behavior is more pronounced at later developmental stages. Finally, we show the spiral2-2 mutant has anisotropic mechanical properties, likely contributing to the spiraling of leaves. Our findings reveal the fibrous network nature of cell walls gives a high degree of tunability in mechanical properties, which allows cells to adjust these properties to support proper development.

Abstract:Abstract Plants live in constantly moving fluid, whether air or water. In response to the loads associated with fluid motion, plants bend and twist, often with great amplitude. These large deformations are not found in traditional engineering application and thus necessitate new specialized scientific developments. Studying fluid–structure interaction (FSI) in botany, forestry, and agricultural science is crucial to the optimization of biomass production for food, energy, and construction materials. FSIs are also central in the study of the ecological adaptation of plants to their environment. This review paper surveys the mechanics of FSI on individual plants. I present a short refresher on fluid mechanics then dive into the statics and dynamics of plant–fluid interactions. For every phenomenon considered, I examine the appropriate dimensionless numbers to characterize the problem, discuss the implications of these phenomena on biological processes, and propose future research avenues. I cover the concept of reconfiguration while considering poroelasticity, torsion, chirality, buoyancy, and skin friction. I also assess the dynamical phenomena of wave action, flutter, and vortex-induced vibrations.

Abstract:Plant morphogenesis involves multiple biochemical and physical processes inside the cell wall. With the continuous progress in biomechanics field, extensive studies have elucidated that mechanical forces may be the most direct physical signals that control the morphology of cells and organs. The extensibility of the cell wall is the main restrictive parameter of cell expansion. The control of cell wall mechanical properties largely determines plant cell morphogenesis. Here, we summarize how cell wall modifying proteins modulate the mechanical properties of cell walls and consequently influence plant morphogenesis.

Abstract:Computational modelling has become increasingly important in advancing our understanding of biological systems, necessitating the development of new computational approaches and software. VirtualLeaf, in particular, is a modelling framework for plant tissues that accounts for the biophysical mechanics of plant cell interactions. The plant cell wall plays a pivotal role in plant development and survival, with younger cells generally having thinner, more flexible (primary) walls than older cells. Signalling processes in growth and pathogen infection also affect cell wall stability. This article presents an updated version of VirtualLeaf with improved cell wall mechanics and morphing behaviour. These are crucial for ultimately understanding plant tissue dynamics and essential signalling processes during growth, tissue formation and pathogen defence. The updated version of VirtualLeaf enables detailed modelling of variations in cell wall stability to the level of individual cell wall elements. These improvements lay the groundwork for using VirtualLeaf to address new research questions, including the structural implications of pathogen infection and growth.

Abstract:The plant primary cell wall is a composite material containing stiff cellulose microfibrils that are embedded within a pectin matrix and crosslinked through a network of hemicellulose polymers. This microstructure endows the wall with nonlinear anisotropic mechanical properties and allows enzymatic regulation of expansive cell growth. We present a mathematical model of hemicellulose crosslink dynamics in an expanding cell wall incorporating strain-enhanced breakage and enzyme-mediated crosslink kinetics. The model predicts the characteristic yielding behaviour in the relationship between stress and strain-rate seen experimentally, and suggests how the effective yield and extensibility of the wall depend on microstructural parameters and on the action of enzymes of the XTH and expansin families. The model suggests that the yielding behaviour encapsulated in the classical Lockhart equation can be explained by the strongly nonlinear dependence of crosslink breakage rate on crosslink elongation. The model also demonstrates how enzymes that target crosslink binding can be effective in softening the wall in its pre-yield state, whereas its post-yield extensibility is determined primarily by the pectin matrix.

Abstract:In this paper we present a derivation and multiscale analysis of a mathematical model for plant cell wall biomechanics that takes into account both the microscopic structure of a cell wall coming from the cellulose microfibrils and the chemical reactions between the cell wall’s constituents. Particular attention is paid to the role of pectin and the impact of calcium-pectin cross-linking chemistry on the mechanical properties of the cell wall. We prove the existence and uniqueness of the strongly coupled microscopic problem consisting of the equations of linear elasticity and a system of reaction-diffusion and ordinary differential equations. Using homogenization techniques (two-scale convergence and periodic unfolding methods) we derive a macroscopic model for plant cell wall biomechanics.

Abstract:The plant cell wall is a complex material made of polysaccharides, proteins, ions and water. As an external envelope around the cell it resists the internal turgor pressure. During plant cell growth, the cell wall material must yield to allow the cell to expand in a controlled spatial and temporal pattern. Modeling this behavior has been approached via a variety of techniques ranging from continuum mechanics to atomistic models, whilst at an intermediate scale, mesoscopic models attempt to consider the mechanical behavior of individual polymers and linkages while simplifying molecular structures to relevant properties. In this review an overview is provided over recent modeling approaches focusing on the primary plant cell wall.

Abstract:The control of the cylindrical cell form in plants appears to reside in the orientation of the reinforcing cellulose microfibrils in the side walls. In elongating cells the fibrils are typically transverse. Control of new synthesis of oriented wall texture is shown to be in turn related to the orientation of cytoplasmic elements in the cell periphery. Three properties of these cytoplasmic elements have been deduced from polarization optical properties of treated and normal cell walls. These deduced properties— namely, possession of a long axis and the ability to build microfibrils perpendicular to it, a tendency to cross-bond to make a parallel array, and a sensitivity of this alignment to colchicine—are all well-known properties of mitotic spindle and phragmoplast fibers which form the cross-wall after mitosis. It is proposed that proteins of spindle fiber nature exist in cortical cytoplasm of plant cells and are active in the control of wall texture and cell form.

Abstract:Forces are increasingly recognized as major regulators of cell structure and function, and the mechanical properties of cells are essential to the mechanisms by which cells sense forces, transmit them to the cell interior or to other cells, and transduce them into chemical signals that impact a spectrum of cellular responses. Comparison of the mechanical properties of intact cells with those of the purified cytoskeletal biopolymers that are thought to dominate their elasticity reveal the extent to which the studies of purified systems can account for the mechanical properties of the much more heterogeneous and complex cell. This review summarizes selected aspects of current work on cell mechanics with an emphasis on the structures that are activated in cell-cell contacts, that regulate ion flow across the plasma membrane, and that may sense fluid flow that produces low levels of shear stress.

Abstract:ABSTRACT Rta, the gene product of Kaposi's sarcoma-associated herpesvirus (KSHV) encoded mainly in open reading frame 50 (ORF50), is capable of activating expression of viral lytic cycle genes. What was not demonstrated in previous studies was whether KSHV Rta was competent to initiate the entire viral lytic life cycle including lytic viral DNA replication, late-gene expression with appropriate kinetics, and virus release. In HH-B2, a newly established primary effusion lymphoma (PEL) cell line, KSHV ORF50 behaved as an immediate-early gene and autostimulated its own expression. Expression of late genes, ORF65, and K8.1 induced by KSHV Rta was eliminated by phosphonoacetic acid, an inhibitor of viral DNA polymerase. Transfection of KSHV Rta increased the production of encapsidated DNase-resistant viral DNA from HH-B2 cells. Thus, introduction of an ORF50 expression plasmid is sufficient to drive the lytic cycle to completion in cultured PEL cells.
The impact of microfibril orientations on the biomechanics of plant cell walls and tissues: modelling and simulations

Abstract:It is known that the orientation of cellulose microfibrils within plant cell walls has an important impact on the morphogenesis of plant cells and tissues. Viewing the shape of a plant cell as a square prism or cylinder with the axis aligning with the primary direction of expansion and growth, the orientation of the microfibrils within the cell wall on the sides of the cell is known. However, not much is known about their orientation at the ends of the cell. Here we investigate the impact of the orientation of cellulose microfibrils within a plant cell wall at the ends of the cell by solving the equations of linear elasticity numerically. Three different scenarios for the orientation of the microfibrils are considered. The macroscopic elastic properties of the cell wall are obtained using homogenization theory from the microscopic description of the elastic properties of the cell wall microfibrils and wall matrix. It is found that the orientation of the microfibrils in the upper and lower parts of cell walls do not affect the expansion of the cell in the direction of its axis but do affect its expansion in the lateral directions. The arrangement of the microfibrils in the upper and lower parts of cell walls is especially important in the case of directed forces acting on plant cell walls and tissues.

Abstract:Plant cells are confined by a network of cellulosic walls that imposes rigid control over the selection of division plane orientations, crucial for morphogenesis and genetically regulated. While in animal cells and yeast, the actin cytoskeleton is instrumental in the execution of cytokinesis, in plant cells the microtubule cytoskeleton is taking the lead in spatially controlling and executing cytokinesis by the formation of two unique, plant-specific arrays, the preprophase band (PPB) and the phragmoplast. The formation of microtubule arrays in plant cells is contingent on acentrosomal microtubule nucleation. At the onset of mitosis, the PPB defines the plane of cell division where the partitioning cell wall is later constructed by the cytokinetic phragmoplast, imposing a spatio-temporal relationship between the two processes. Current research progress in the field of plant cell division focuses on identifying and tying the links between early and late events in spatial control of cytokinesis and how microtubule array formation is regulated in plant cells.
