Abstract:During normal ripening the dry weight of the total cell wall material decreased, while water-soluble pectin content increased extensively with the decrease in amounts of ionically associated and covalently bound pectins and cellulose. However, hemicellulose content did not change significantly. Low temperature stress slowed down degradation of pectin and cellulose, causing accumulation of high molecular weight pectin and increase in juice viscosity. Intermittent warming resulted in solubilization and depolymerization of pectins and cellulose, thus leading to cell wall metabolism associated with fruit ripening. Radiolabeling test using ~(14)C-sucrose indicated that cell wall synthesis continued during the degradation of cell wall, and increased during the onset of fruit ripening. Fruit senescence was directly associated with reduced cell wall synthesis. The higher amount of cell wall material in chilling-injured fruit than that in normal fruit was not due to an increase in cell wall synthesis but rather to reduced degradation of cell wall material.

Abstract:The effects of nano-ZnO-based low-density polyethylene (NZLDPE)-packaging on chilling tolerance and pectin metabolism in postharvest "Hujingmilu" peaches were investigated. Peaches packed in low-density polyethylene (LDPE) and NZLDPE were stored at 2 degrees C for 40 d. Compared with the control, both packages alleviated the development of chilling injury, showed higher fruit firmness with lower browning index, electrolyte leakage, relative viscosity, and decay rate. NZLDPE-packaging performed better than LDPE-packaging because of the rapid formation of low O-2 and high CO2. Moreover, NZLDPE-packaging inhibited pectin esterase and enhanced polygalacturonase and beta-galactosidase, leading to the promotion of alkali soluble-pectin and water-soluble pectin, and the decrease of chelater-soluble pectin. These effects were of great benefit to the maintaining of cell wall structure and the degradation of calcium-pectate gel, which finally alleviated the chilling injury and therefore maintained good quality during chilling stress.

Abstract:Fruit of jujube (Ziziphus jujube Mill. cv. Dongzao), known as a high-quality fresh edible variety, is popular among consumers on account of its bright color, crispy texture and abundant nutrients. However, the softening of jujube fruit after harvest seriously shortens its shelf life. To examine possible treatments to reduce softening rate, we evaluated the effect on fruit quality attributes of an elevated atmospheric O2 concentration during cold storage, ultimately unraveling its biological effect on jujube cell wall metabolism and antioxidant level. Our results demonstrated that an elevated O2 atmosphere during cold storage induced both the activity of antioxidant enzymes and relative expression levels of related genes, promoting the scavenging of hydrogen peroxide and superoxide anion. By the end of storage, the activities of superoxide dismutase, peroxidase, and catalase in elevated O2-treated jujubes were 1.9, 1.3, and 1.1 times, respectively, those in the control group. In addition, elevated O2 concentration inhibited the activities of polygalacturonase, pectin methyl esterase, cellulase and the relative abundance of related transcripts, thus delaying the degradation of cellulose and hemicelluloses and reducing the conversion of insoluble pectin to water-soluble pectin and ionic-soluble pectin. These changes presumably contribute to the maintenance of the cell wall components and cell microstructure. In conclusion, an elevated atmospheric O2 concentration of 60 % during cold storage promoted antioxidant capacity and inhibited cell wall metabolism in postharvest jujube fruit, reducing postharvest softening.

Abstract:In order to explore the effectiveness of nano-SiOx/chitosan complex(NSCC) on postharvest Huanghua pear fruit(Pyrus pyrifolia Nakai),fruits were stored at 20 ℃ after coating with 1% NSCC.The effect of NSCC coating on the ripening and cell wall metabolism in fruit was investigated.Results showed that NSCC coating not only significantly delayed the onset of climacteric ethylene production and respiration in fruit,and also significantly decreased the activities of pectinesterase(PE),polygalacturonase(PG) and cellulase during storage.NSCC coating also delayed the depolymerization of propectin,cellulose and inhibited the increase of water soluble pectin(WSP),maintained fruit firmness.Application of NSCC can extend 10 days of the storage life of Huanghua pear fruit.
Effect of heat conditioning and intermittent warming on chilling injury and pectolytic enzymes in peaches

Abstract:About 86% peach fruit (Prunus persica, cv. Baifeng) becamewoolly during a ripening period of 4 d at 20 ℃ following 29 d of cold storage at 0 ℃. Conditioning at 35 ℃ for 48 h prior to cold storage or warming at 20 ℃ for 24 h every 9 d of cold storage was highly effective in alleviating woolliness, although low sensory quality or flesh softening also occurred. Browning was not always accompanied with the development of woolliness, but retention of flesh firmness was a typical woolliness symptom. Extractable juice content was highly negatively correlated with the incidence of woolliness. Endo-PG activity in fruit cold-stored immediately and continuously after harvest was low throughout storage and the subsequent ripening period. Both heat conditioning and intermittent warming enhanced PE, endo-PG and exo-PG activities, with the second intermittent warming being the most effective. Chilling injury development appeared to be associated with reduced endo-PG activity.
Molecular Mechanisms Through Which Short-Term Cold Storage Improves the Nutritional Quality and Sensory Characteristics of Postharvest Sweet Potato Tuberous Roots: A Transcriptomic Study

Abstract:Sweet potato (Ipomoea batatas (L.) Lam.) is a commercially relevant food crop with high demand worldwide. This species belongs to the Convolvulaceae family and is native to tropical and subtropical regions. Storage temperature and time can adversely affect tuberous roots’ quality and nutritional profile. Therefore, this study evaluates the effect of storage parameters using physicochemical and transcriptome analyses. Freshly harvested tuberous roots (Xingxiang) were stored at 13 °C (control) or 5 °C (cold storage, CS) for 21 d. The results from chilling injury (CI) evaluation demonstrated that there was no significant difference in appearance, internal color, weight, and relative conductivity between tuberous roots stored at 13 and 5 °C for 14 d and indicated that short-term CS for 14 d promoted the accumulation of sucrose, chlorogenic acid, and amino acids with no CI symptoms development. This, in turn, improved sweetness, antioxidant capacity, and nutritional value of the tuberous roots. Transcriptome analyses revealed that several key genes associated with sucrose, chlorogenic acid, and amino acid biosynthesis were upregulated during short-term CS, including sucrose synthase, sucrose phosphate synthase, phenylalanine ammonia-lyase, 4-coumarate-CoA ligase, hydroxycinnamoyl-CoA quinate hydroxycinnamoyltransferase, serine hydroxymethyltransferase, alanine aminotransferase, arogenate dehydrogenase, and prephenate dehydratase. These results indicated that storage at 5 °C for 14 d could improve the nutritional quality and palatability of sweet potato tuberous roots without compromising their freshness.

Abstract:Persimmon fruit can be stored for 20-25 days and 35-45 days at 12 ℃ and 8 ℃, respectively. If it is to be stored over 50 days, the storage temperature should be 4 ℃ or even lower. Chilling injury symptoms and physiological disorders were observed when the fruit were transferred to normal temperature after storage at 4 ℃ for more than 20 days. In chilling-injured fruit, the primary cell wall and the middle lamella could not be dissolved normally, the fruit juice became more viscous and extractable juice rate was decreased. Respiration rate, ethylene production and skin permeability of chilled persimmon fruit were unusually high.

Abstract:Two cultivars of peaches ('Cangfangzaosheng' and 'Songsenzaosheng') were stored at 2°C, 8°C and 15°C for maximum of 41 days. The nanostructural changes of water-soluble pectin (WSP) and chelate-soluble pectin (CSP) of all cultivars were analyzed qualitatively and quantitatively by atomic force microscopy (AFM). The results show that long linear single pectin chains of peach were detached and the percentage of small width chains increased during storage. This phenomenon was also found with increased storage temperature. Moreover, the widths of pectin molecules of both peach cultivars were similar and very regular. Combined with previous reports, the results indicate that nanostructural degradations of pectin widths might be independent of their chemical content changes.
The Effects of Different Temperature and Humidity Conditions on the Ripening and Cracking of Annona Atemoya Fruit During Storage by Regulating the Conversion of Starch into Soluble Sugars

Abstract:Atemoya is prone to soft ripening and cracking and has a short storage life at room temperature in summer and autumn in China. To better understand the mechanisms involved in fruit ripening and cracking, the effects of five temperature and humidity treatments (28 degrees C + 50%, 28 degrees C + 70%, 28 degrees C + 95%, 32 degrees C + 95%, 15 degrees C + 95%) were evaluated, and transcriptome analysis of the fruits was performed under storage conditions (stored for 0, 2, 4, and 6 days at 28 and 15 degrees C) in this study. The transcriptome results revealed that starch and sucrose metabolism was the most enriched KEGG pathway, including 485 DEGs. Compared with the 28 degrees C + 95% and 28 degrees C + 70% treatments, which accelerated the fruit ripening and cracking, low-temperature storage (15 degrees C + 95%) inhibited the expression of starch hydrolase genes such as GWD, LSF2, BAM3, BAM9, ISA3, and DPE2, , decreased the activities of BAM and DBE, increased the concentrations of sucrose and trehalose, slowed down the conversion rate of the starch to soluble sugar, decreased the RWC of the pulp, and effectively delayed ripening and cracking. These results provide a physiology and molecular foundation for explaining fruit softening and cracking and extending the shelf life of atemoya.
Hybrid Label-Free Molecular Microscopies for Simultaneous Visualization of Changes in Cell Wall Polysaccharides of Peach at Single- and Multiple-Cell Levels during Postharvest Storage

Abstract:Softening of fruit during the postharvest storage, which is mainly associated with both compositional and spatial changes of polysaccharides within cell wall, affects the texture and quality of fruit. Current research on the fruit softening mechanism lacks an understanding of the overall softening at the cell level. The objective of this work was to investigate the change in the spatial distribution of cell wall polysaccharides in peach flesh cells at both single- and multiple-cell levels in a label-free way during the postharvest storage. Nonmelting peaches (Prunus persica L. Batsch cv.”Zhonghuashoutao”) at commercial maturity were stored at 0 °C and 20 °C. Firmness measurement and chemical analysis were performed at each storage time. In addition, three molecular imaging techniques, namely confocal Raman microspectroscopy (CRM), Fourier transform infrared microspectroscopy (FTIRM), and stimulated Raman scattering microscopy (SRS) were used to visualize changes in the spatial distribution of cell wall polysaccharides of peach fruit in a label-free way during the postharvest storage. The combination of CRM and FTIRM provided complementary spectral information to visualize the spatial changes of cellulose, hemicellulose, and pectin in the cell wall of peach flesh during softening at the single-cell level, and found that the cell wall polysaccharides tended to be concentrated in the cell corner of parenchymal cells at the late stage. Furthermore, SRS, which is an ultrafast Raman imaging technique (approximately three or four orders of magnitude faster than CRM), was used for high-throughput cell wall phenotypes measurement. Different degradation degrees of parenchymal cells during fruit softening were found based on the gray-scale statistical analysis of SRS data. In general, cell wall polysaccharides decreased during softening and tended to be concentrated in the cell corner for most parenchymal cells at the late stage, but there were also some cells not in line with the whole softening trends. The results show that there were differences in the content and spatial changes of cell wall polysaccharides among parenchymal cells of peach fruit during the softening process, and the hybrid use of CRM, FTIRM, and SRS is a promising method for simultaneous visualization of changes in cell wall polysaccharides of peach.

Abstract:Strawberry fruit is extremely perishable at least partially because of excessive softening, which can be inhibited by edible chitosan coating during ripening, but the biochemical and molecular mechanisms are not completely understood in the fruit under cold conditions. Here, changes in levels of quality attributes, cell wall constituents, activities and expression of cell wall modifying enzymes, were evaluated in strawberry fruit treated with 1.5% (w/v) chitosan coating and stored at 4 °C for 12 days. Chitosan coating had a significant inhibitory effect on fruit softening during cold storage, rather than other quality attributes. Chitosan coating treated fruit showed the reduction in the content of water-soluble pectin coincident with significant inhibition in activities of polygalacturonase (PG) and pectin methylesterase (PME), and expression of FaPG1and FaPME1, relative to the controls. Activities of these two enzymes and expression of FaPG1 were significantly correlated with the contents of water-soluble pectin. Besides, the decreased link between cellulose and hemicellulose was indicated by the reduced transcript levels of FaXTH1 and FaEXP2, with significantly positive correlation. In conclusion, the effect of chitosan coating on strawberry fruit mainly involves the softening inhibition by metabolic enzymes of pectin, especially PG and PME. The results will provide insights into developing measures to control strawberry fruit softening during cold storage and cold chain logistics.

Abstract:Papaya (Carica papaya L.) is highly susceptible to chilling injury (CI), which primarily causes surface pitting, black scald, susceptibility to fungal infection, and the breakdown of tissue. It developed rapidly at 6 °C but was significantly suppressed at 1 °C, which indicated that the papaya tissue was more solid at a lower temperature. This rare phenomenon is considered to be abnormal CI behavior. The maintenance of fruit tissue integrity during abnormal CI behavior may be related to modifications of the cell wall pectin. The modifications of cell wall pectin were studied during storage at 1 and 6 °C. Storage at 1 °C alleviated the progression of CI and inhibited the degradation of pectin components and cell wall structure of papaya fruit. The increase in water-soluble pectin and the solubilization of cyclohexanediamine tetraacetic acid- (CDTA-) and sodium carbonate (Na2CO3)-soluble pectin was limited at 1 °C. Storage at this temperature limited the detachment of linear backbone chains and minimized the degradation of branching and reticular structure in the pectin molecules. Storage at this temperature also delayed the decrease in the size of pectin backbone and helped to prevent the loss of neutral sugars from the pectin side chains. Thus, alleviation of the degradation of cell wall pectin contributes to the structural integrity of the cell wall and thus helps the maintenance of the tissue integrity of papaya at 1 °C.

Abstract:Mature-green (Prunus salicina Lindl. cv. 'Friar') plums were treated in water at 40 degreesC for 40 min, 45 degreesC for 35 min, 50 degreesC for 30 min or 55 degreesC for 25 min, stored at 0 degreesC for 35 d plus 9 d of ripening at 20 to 25 degreesC. During cold storage, putrescine and cadaverine accumulated most compared to spermine and spermidine, especially in fruits treated at 45 or 50 degreesC, compared to other treatments. Chilling injury (CI) and decay symptoms were severe in fruits of control, 55 and 40 degreesC-treated fruits. Decay Incidence was also high in fruits stored at room temperature. Chilling Injury and decay symptoms were retarded in fruits treated at 45 and 50 degreesC, which also maintained increased polyamines and low ethylene. By the end of their shelf life, only 45 and 50 degreesC-HWD treated fruits were fit for market.

Abstract:This study aimed to elucidate whether 1-methylcyclopropene (1-MCP) treatment delays the fruit softening mechanism associated with the fruit quality of the newly released apple cultivars "Summer King" and "Green Ball" during cold storage. For both cultivars, the fruit treated with 1-MCP exhibited lower internal ethylene concentration, higher firmness, and higher titratable acidity relative to the control fruit, in association with less fruit softening. In addition, the treated fruit significantly delayed fresh weight loss and reduction of soluble solids content, especially in "Green Ball." Moreover, slower degradation of cell wall components (water-soluble pectin, sodium carbonate-soluble pectin, hemicellulose, and cellulose) was also observed in the treated fruit in comparison to the control fruit. Similarly, the enzymatic activities (of polygalacturonase, pectin methylesterase, cellulase, β-galactosidase, and α-L-arabinofuranosidase) that cause cell wall degradation were relatively lower in the treated fruit than in the control fruit for both cultivars, which was further proved by transcriptional analysis of the genes encoding the enzymes. Overall, the results suggested that the usage of 1-MCP is useful to delay fruit softening of the two cultivars during cold storage by delaying the degradation of cell wall components and enzymatic activities of cell wall hydrolysis.

Abstract:The effects of postharvest β-aminobutyric acid (BABA) treatment on fruit firmness, pectin degrading enzymes, cell wall constituents and microstructural alterations of pericarp in sweet cherry fruit were investigated. BABA significantly delayed the decline of fruit firmness and inhibited the increase of membrane permeability and the accumulation of malondialdehyde in cherries. The BABA-treated fruit exhibited significantly higher contents of water-soluble pectin, CDTA-soluble pectin, Na2CO3-soluble pectin, total pectin, cellulose and hemicellulose than the control during storage. Activities of pectin degrading enzymes including polygalacturonase and pectinmethylesterase were markedly reduced by BABA treatment. Observations by scanning electron microscopy showed BABA maintained smooth cuticle and integrated structure of subepidermal cell in sweet cherry. These results suggest that the delay in fruit senescence by BABA may be due to depressed membrane permeability and malondialdehyde content, reduced activities of polygalacturonase and pectinmethylesterase, enhanced cell wall polysaccharides content, and integrated subepidermal cell structure in sweet cherry.

Abstract:Low temperature storage slows ripening and reduces decay of peach fruit, although storage life is limited due to chilling injury. Intermittent warming (IW) can be used to alleviate fruit chilling injury, however, its effect on aroma remains unknown. Yellow-fleshed peach (Prunus persica L. Batsch cv. Jinxiu) fruit (melting type) was stored at 5 degrees C or exposed to 20 degrees C for 1 d every week during storage. Changes in fruit esters, alcohol acyltransferase (AAT) activity and gene expression were investigated. Flesh browning (FB) was observed on the third day of shelf-life at 20 degrees C after 21 d of storage at 5 degrees C, while no FB was found in IW-treated fruit for up to 28 d. Significant lower ester contents were found in peach fruit with flesh browning. Expression profiles of PpAAT1 were similar to MT activity profiles, both of which increased during shelf-life of fruit treated with IW. As precursors of esters, levels of linoleic and linolenic acids were high in IW-treated peach fruit. Treatment with IW effectively alleviated the loss of aroma-related esters associated with FB, and high levels of AAT activity and PpAAT1 expression in IW-treated peach fruit contributed to the formation of the esters. Abstract:Huanghua pear was used to study on storage effects with different treatments at the low temperature of 0℃.Different preservative,including chitosan composite,1-methylcyclopropene,2,4-dichlorophenoxy-acetic acid and prochloraz iprodione,was used to deal with huanghua pear at the low temperature of 0℃.And decay rate,weight loss rate,respiration rate,firmness,soluble solids,total acid content,pectin content and Vc content were measured every 15 days.The experiments showed that:all the preservative can delay decreasing rate of fruit firmness,soluble solids,total acid,pectin and Vcduring the storage of fruit,inhibit the respiration rate effectively and reduce decay rate and weight loss rate,so the storage time of fruits was extended.And 1-MCP had the best effect.

The characterization of GPH was investigated and its preservation effect with 1-Methylcyclopropene (1-MCP) on apricots storage at low temperature (0 °C) was measured. The results showed that pectin increased the water retention and solubility of hydrogel by crosslinking with γ-PGA, which could regulate the rheological property of γ-PGA hydrogel. The application of GPH (1.5%, v/v) significantly reduced the weight loss, decay rate, and delayed the decline of soluble solids, ascorbic acid and titratable acid content of apricots. Meanwhile, the GPH treatment slowed down the respiration and polygalacturonase activity of apricot, and ensured the firmness and pectin content of apricots. Also, it showed superior effect to the individual hydrogel treatment when GPH were combined with 1-MCP (1 μL/L). On the 60th day of storage, the loss of flavor and nutritional components in GPH/1-MCP group was particularly less. The information indicated that GPH combined with 1-MCP could improve the postharvest preservation effect of apricots, while providing means for better marketability gains.
