Abstract:Background: Recent studies have demonstrated that changes in brain information processing during anesthetic-nduced loss of consciousness (LOC) might be influenced by phase-amplitude coupling (PAC) in electroencephalogram (EEG). However, most anesthesia research on PAC typically focuses on delta and alpha oscillations. Studies of spatial-frequency characteristics by PAC for EEG may yield additional insights into understanding the impaired information processing under anesthesia unconsciousness and provide potential improvements in anesthesia monitoring. Objective: Considering different frequency bands of EEG represent neural activities on different spatial scales, we hypothesized that functional coupling simultaneously appears in multiple frequency bands and specific brain regions during anesthesia unconsciousness. In this paper, PAC analysis on whole-brain EEG besides delta and alpha oscillations was investigated to understand the influence of multiple cross-frequency coordination coupling on information processing during the loss and recovery of consciousness. Method: EEG data from fifteen patients without cognitive diseases (7 males/8 females, aged 43.8 +/- 13.4 years, weighing 63.3 +/- 14.9 kilograms) undergoing lower limb surgery and sevoflurane anesthesia was recorded. To investigate the spatial-frequency characteristics of EEG source signals during loss and recovery of consciousness, the time-resolved PAC (tPAC) was calculated to reflect cross-frequency coordination in different frequency bands (delta, theta, alpha, beta, gamma) and different functional regions (Visual, Limbic, Dorsal attention, Ventral attention, Default, Somatomotor, Control, Salience networks). Furthermore, different patterns (peak-max and trough-max) of PAC were examined by constructing phase-amplitude histograms using phase bins to investigate the different information processing during LOC. The multivariate analysis of variance (MANOVA) and trend analysis were used for statistical analysis. Results: Theta-alpha and alpha-beta PAC were observed during sevoflurane-induced LOC, which significantly changed during loss and recovery of consciousness (E-4,E-70 = 16.553, p < 0.001 for theta-alpha PAC and F-4,F-70 = 12.446, p < 0.001 for alpha-beta PAC, MANOVA test). Simultaneously, PAC was distributed in specific functional regions, i.e., Visual, Limbic, Default, Somatomotor, etc. Furthermore, peak-max patterns of theta-alpha PAC were observed while alpha-beta PAC showed trough-max patterns and vice versa. Conclusion: Theta-alpha and alpha-beta PAC observed in specific brain regions represent information processing on multiple spatial scales, and the opposite patterns of PAC indicate opposite information processing on multiple spatial scales during LOC. Our study demonstrates the regulation of local-global information processing during sevoflurane-induced LOC. It suggests the utility of evaluating the balance of functional integration and segregation in monitoring anesthetized states.

Abstract:Phase-amplitude coupling (PAC) is the association of the amplitude of a high-frequency oscillation with the phase of a low-frequency oscillation. In neuroscience, this relationship provides a mechanism by which neural activity might be coordinated between distant regions. The dangers and pitfalls of assessing PAC with commonly used statistical measures have been well-documented. The limitations of these measures include: (1) response to non-oscillatory, high-frequency, broad-band activity, (2) response to high-frequency components of the low-frequency oscillation, (3) adhoc selection of analysis frequency-intervals, and (4) reliance upon data shuffling to assess statistical significance. Objective. To address issues (1)–(4) by introducing a nonparametric multitaper estimator of PAC. Approach. In this work, a multitaper PAC estimator is proposed that addresses these issues. Specifically, issue (1) is addressed by replacing the analytic signal envelope estimator computed using the Hilbert transform with a multitaper estimator that down-weights non-sinusoidal activity using a classical, multitaper super-resolution technique. Issue (2) is addressed by replacing coherence between the low-frequency and high-frequency components in a standard PAC estimator with multitaper partial coherence, while issue (3) is addressed with a physical argument regarding meaningful neural oscillation. Finally, asymptotic statistical assessment of the multitaper estimator is introduced to address issue (4). Main results. Multitaper estimates of PAC are introduced. Their efficacy is demonstrated in simulation and on human intracranial recordings obtained from epileptic patients. Significance. This work facilitates a more informative statistical assessment of PAC, a phenomena exhibited by many neural systems, and provides a basis upon which further nonparametric multitaper-related methods can be developed.

Power and Phase Properties of Oscillatory Neural Responses in the Presence of Background Activity

Abstract:Natural sensory inputs, such as speech and music, are often rhythmic. Recent studies have consistently demonstrated that these rhythmic stimuli cause the phase of oscillatory, i.e. rhythmic, neural activity, recorded as local field potential (LFP), electroencephalography (EEG) or magnetoencephalography (MEG), to synchronize with the stimulus. This phase synchronization, when not accompanied by any increase of response power, has been hypothesized to be the result of phase resetting of ongoing, spontaneous, neural oscillations measurable by LFP, EEG, or MEG. In this article, however, we argue that this same phenomenon can be easily explained without any phase resetting, and where the stimulus-synchronized activity is generated independently of background neural oscillations. It is demonstrated with a simple (but general) stochastic model that, purely due to statistical properties, phase synchronization, as measured by 'inter-trial phase coherence', is much more sensitive to stimulus-synchronized neural activity than is power. These results question the usefulness of analyzing the power and phase of stimulus-synchronized activity as separate and complementary measures; particularly in the case of attempting to demonstrate whether stimulus-synchronized neural activity is generated by phase resetting of ongoing neural oscillations.
Addressing Pitfalls in Phase-Amplitude Coupling Analysis with an Extended Modulation Index Toolbox

Abstract:Phase-amplitude coupling (PAC) is proposed to play an essential role in coordinating the processing of information on local and global scales. In recent years, the methods able to reveal trustworthy PAC has gained considerable interest. However, the intrinsic features of some signals can lead to the identification of spurious or waveform-dependent coupling. This prompted us to develop an easily accessible tool that could be used to differentiate spurious from authentic PAC. Here, we propose a new tool for more reliable detection of PAC named the Extended Modulation Index (eMI) based on the classical Modulation Index measure of coupling. eMI is suitable both for continuous and epoched data and allows estimation of the statistical significance of each pair of frequencies for phase and for amplitude in the whole comodulogram in the framework of extreme value statistics. We compared eMI with the reference PAC measures-direct PAC estimator (a modification of Mean Vector Length) and standard Modulation Index. All three methods were tested using computer-simulated data and actual local field potential recordings from freely moving rats. All methods exhibited similar properties in terms of sensitivity and specificity of PAC detection. eMI proved to be more selective in the dimension of frequency for phase. One of the novelty's offered by eMI is a heuristic algorithm for classification of PAC as Reliable or Ambiguous. It relies on analysis of the relation between the spectral properties of the signal and the detected coupling. Moreover, eMI generates visualizations that support further evaluation of the coupling properties. It also introduces the concept of the polar phase-histogram to study phase relations of coupled slow and fast oscillations. We discuss the extent to which eMI addresses the known problems of interpreting PAC. The Matlab® toolbox implementing eMI framework, and the two reference PAC estimators is freely available as EEGLAB plugin at https://github.com/GabrielaJurkiewicz/ePAC .

Abstract:Bandpass filters play a core role in ECoG signal processing. Commonly used frequency bands such as alpha, beta, and gamma bands can reflect the normal rhythm of the brain. However, the universally predefined bands might not be optimal for a specific task. Especially the gamma band usually covers a wide frequency span (i.e., 30-200 Hz) which can be too coarse to capture features that appear in narrow bands. An ideal option is to find the optimal frequency bands for specific tasks in real-time and dynamically. To tackle this problem, we propose an adaptive band filter that selects the useful frequency band in a data-driven way. Specifically, we leverage the phase-amplitude coupling (PAC) of the coupled working mechanism of synchronizing neuron and pyramidal neurons in neuronal oscillations, in which the phase of slower oscillations modulates the amplitude of faster ones, to help locate the fine frequency bands from the gamma range, in a task-specific and individual-specific way. Thus, the information can be more precisely extracted from ECoG signals to improve neural decoding performance. Based on this, an end-to-end decoder (PACNet) is proposed to construct a neural decoding application with adaptive filter banks in a uniform framework. Experiments show that PACNet can improve neural decoding performance universally with different tasks.

Abstract:Phase-amplitude coupling (PAC) describes the interaction of two separate frequencies in which the lower frequency phase acts as a carrier frequency of the higher frequency amplitude. It is a means of carrying integrated streams of information between micro- and macroscale systems in the brain, allowing for coordinated activity of separate brain regions. A beta–gamma PAC increase over the sensorimotor cortex has been observed consistently in people with Parkinson's disease (PD). Its cause is attributed to neural entrainment in the basal ganglia, caused by pathological degeneration characteristic of PD. Disruptions in this phenomenon in PD patients have been observed in the resting state as well as during movement recordings and have reliably distinguished patients from healthy participants. The changes can be detected non-invasively with the electroencephalogram (EEG). They correspond to the severity of the motor symptoms and the medication status of people with PD. Furthermore, a medication-induced decrease in PAC in PD correlates with the alleviation of motor symptoms measured by assessment scales. A beta–gamma PAC increase has, therefore, been explored as a possible means of quantifying motor pathology in PD. The application of this parameter to closed-loop deep brain stimulation could serve as a self-adaptation measure of such treatment, responding to fluctuations of motor symptom severity in PD. Furthermore, phase-dependent stimulation provides a new precise method for modulating PAC increases in the cortex. This review offers a comprehensive synthesis of the current EEG-based evidence on PAC fluctuations in PD, explores the potential practical utility of this biomarker, and provides recommendations for future research.

Abstract:Introduction: Cross frequency coupling (CFC) between electrophysiological signals in the brain is a long-studied phenomenon and its abnormalities have been observed in conditions such as Parkinson's disease and epilepsy. More recently, CFC has been observed in stomach-brain electrophysiologic studies and thus becomes an enticing possible target for diseases involving aberrations of the gut-brain axis. However, current methods of detecting coupling, specifically phase-amplitude coupling (PAC), do not attempt to capture the phase and amplitude statistical relationships. Methods: In this paper, we first demonstrate a method of modeling these joint statistics with a flexible parametric approach, where we model the conditional distribution of amplitude given phase using a gamma distributed generalized linear model (GLM) with a Fourier basis of regressors. We perform model selection with minimum description length (MDL) principle, demonstrate a method for assessing goodness-of-fit (GOF), and showcase the efficacy of this approach in multiple electroencephalography (EEG) datasets. Secondly, we showcase how we can utilize the mutual information, which operates on the joint distribution, as a canonical measure of coupling, as it is non-zero and non-negative if and only if the phase and amplitude are not statistically independent. In addition, we build off of previous work by Martinez-Cancino et al., and Voytek et al., and show that the information density, evaluated using our method along the given sample path, is a promising measure of time-resolved PAC. Results: Using synthetically generated gut-brain coupled signals, we demonstrate that our method outperforms the existing gold-standard methods for detectable low-levels of phase-amplitude coupling through receiver operating characteristic (ROC) curve analysis. To validate our method, we test on invasive EEG recordings by generating comodulograms, and compare our method to the gold standard PAC measure, Modulation Index, demonstrating comparable performance in exploratory analysis. Furthermore, to showcase its use in joint gut-brain electrophysiology data, we generate topoplots of simultaneous high-density EEG and electrgastrography recordings and reproduce seminal work by Richter et al. that demonstrated the existence of gut-brain PAC. Using simulated data, we validate our method for different types of time-varying coupling and then demonstrate its performance to track time-varying PAC in sleep spindle EEG and mismatch negativity (MMN) datasets. Conclusions: Our new measure of PAC using Gamma GLMs and mutual information demonstrates a promising new way to compute PAC values using the full joint distribution on amplitude and phase. Our measure outperforms the most common existing measures of PAC, and show promising results in identifying time varying PAC in electrophysiological datasets. In addition, we provide for using our method with multiple comparisons and show that our measure potentially has more statistical power in electrophysiologic recordings using simultaneous gut-brain datasets.

Abstract:The investigation of brain oscillations and connectivity has become an important topic in the recent decade. There are several types of interactions between neuronal oscillations, and one of the most interesting among these interactions is phase-amplitude coupling (PAC). Several methods have been proposed to measure the strength of PAC, including the phase-locking value, circular-linear correlation, and modulation index. In the current study, we compared these traditional PAC methods with simulated electroencephalogram signals. Further, to assess the PAC value at each time point, we also compared two recently established methods, event-related phase-locking value and event-related circular-linear correlation, with our newly proposed event-related modulation index (ERMI). Results indicated that the ERMI has better temporal resolution and is more tolerant to noise than the other two event-related methods, suggesting the advantages of utilizing ERMI in evaluating the strength of PAC within a brain region.

Abstract:Phase-amplitude coupling is a promising construct to study cognitive processes in electroencephalography (EEG) and magnetencephalography (MEG). Due to the novelty of the concept, various measures are used in the literature to calculate phase-amplitude coupling. Here, performance of the three most widely used phase-amplitude coupling measures – phase-locking value (PLV), mean vector length (MVL), and modulation index (MI) – and of the generalized linear modeling cross-frequency coupling (GLM-CFC) method is thoroughly compared with the help of simulated data. We combine advantages of previous reviews and use a realistic data simulation, examine moderators and provide inferential statistics for the comparison of all four indices of phase-amplitude coupling. Our analyses show that all four indices successfully differentiate coupling strength and coupling width when monophasic coupling is present. While the MVL was most sensitive to modulations in coupling strengths and width, only the MI and GLM-CFC can detect biphasic coupling. Coupling values of all four indices were influenced by moderators including data length, signal-to-noise-ratio, and sampling rate when approaching Nyquist frequencies. The MI was most robust against confounding influences of these moderators. Based on our analyses, we recommend the MI for noisy and short data epochs with unknown forms of coupling. For high quality and long data epochs with monophasic coupling and a high signal-to-noise ratio, the use of the MVL is recommended. Ideally, both indices are reported simultaneously for one data set.

Abstract:Phase-amplitude coupling (PAC) between the phase of low-frequency oscillations and the power of high-frequency activity plays a functional role in neuronal computation and information transfer. Traditional Hilbert transform-based PAC methods assume that neuronal activity is narrow-band, sinusoidal, and sustained. However, natural neuronal signals often violate these three assumptions, creating a potential confound for the interpretation of PAC results. In this study, we present a new method, called Tau-Modulation, that does not require these assumptions to be met. We use this method to identify task-relevant neuronal networks in human electrocorticographic signals. Our results show that Tau-Modulation can identify these networks and characterize the strength and frequency of wideband low-frequency coupling with broadband gamma activity. Thus, Tau-Modulation might provide for a robust approach to analyzing neuronal signals and pave the way for new insights on brain functions.

Abstract:Objective. Phase-amplitude coupling (PAC), the coupling of the amplitude of a faster brain rhythm to the phase of a slower brain rhythm, plays a significant role in brain activity and has been implicated in various neurological disorders. For example, in Parkinson's disease, PAC between the beta (13–30 Hz) and gamma (30–100 Hz) rhythms in the motor cortex is exaggerated, while in Alzheimer's disease, PAC between the theta (4–8 Hz) and gamma rhythms is diminished. Modulating PAC (i.e. reducing or enhancing PAC) using brain stimulation could therefore open new therapeutic avenues. However, while it has been previously reported that phase-locked stimulation can increase PAC, it is unclear what the optimal stimulation strategy to modulate PAC might be. Here, we provide a theoretical framework to narrow down the experimental optimisation of stimulation aimed at modulating PAC, which would otherwise rely on trial and error. Approach. We make analytical predictions using a Stuart–Landau model, and confirm these predictions in a more realistic model of coupled neural populations. Main results. Our framework specifies the critical Fourier coefficients of the stimulation waveform which should be tuned to optimally modulate PAC. Depending on the characteristics of the amplitude response curve of the fast population, these components may include the slow frequency, the fast frequency, combinations of these, as well as their harmonics. We also show that the optimal balance of energy between these Fourier components depends on the relative strength of the endogenous slow and fast rhythms, and that the alignment of fast components with the fast rhythm should change throughout the slow cycle. Furthermore, we identify the conditions requiring to phase-lock stimulation to the fast and/or slow rhythms. Significance. Together, our theoretical framework lays the foundation for guiding the development of innovative and more effective brain stimulation aimed at modulating PAC for therapeutic benefit.

Abstract:Parkinson's disease is associated with abnormal oscillatory electrical activities of neurons and neuronal ensembles throughout the basal ganglia-thalamocortical network. It has recently been documented in patients with advanced parkinsonism that the amplitude of gamma-band oscillations (50-200 Hz) in electrocorticogram recordings from the primary motor cortex is abnormally coupled to the phase of beta band oscillations within the same signals. It is not known when in the course of the disease the abnormal phase-amplitude coupling (PAC) arises, and whether it is influenced by arousal or prior exposure to dopaminergic medications. To address these issues, we analyzed the relationship between the severity of parkinsonian motor signs and the extent of PAC in a progressive model of parkinsonism, using primates that were not exposed to levodopa prior to testing. PAC was measured in electrocorticogram signals from the primary motor cortex and the supplementary motor area in 3 monkeys that underwent weekly injections of small doses of the neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine, rendering them progressively parkinsonian. We found that parkinsonism was associated with increased coupling between the phase of low-frequency (4-10 Hz) oscillations and the amplitude of oscillations in the high gamma band (50-150 Hz). These changes only reached significance when the animals became fully parkinsonian. The increased PAC was normalized after levodopa treatment. We also found a similar increase in PAC during sleep, even in normal animals. The identified PAC was independent of concomitant changes in spectral power in the 2.9-9.8Hz or 49.8-150.4 Hz ranges. We conclude that PAC is predominately a sign of advanced parkinsonism, and is, thus, not essential for the development of parkinsonism. However, increased PAC appears to correlate with the severity of fully developed parkinsonism.

Abstract:Identifying biomarkers of developing mental disorder is crucial to improving early identification and treatment—a key strategy for reducing the burden of mental disorders. Cross-frequency coupling between two different frequencies of neural oscillations is one such promising measure, believed to reflect synchronization between local and global networks in the brain. Specifically, in adults phase–amplitude coupling (PAC) has been shown to be involved in a range of cognitive processes, including working and long-term memory, attention, language, and fluid intelligence. Evidence suggests that increased PAC mediates both temporary and lasting improvements in working memory elicited by transcranial direct-current stimulation and reductions in depressive symptoms after transcranial magnetic stimulation. Moreover, research has shown that abnormal patterns of PAC are associated with depression and schizophrenia in adults. PAC is believed to be closely related to cortico-cortico white matter (WM) microstructure, which is well established in the literature as a structural mechanism underlying mental health. Some cognitive findings have been replicated in adolescents and abnormal patterns of PAC have also been linked to ADHD in young people. However, currently most research has focused on cross-sectional adult samples. Whereas initial hypotheses suggested that PAC was a state-based measure due to an early focus on cognitive, task-based research, current evidence suggests that PAC has both state-based and stable components. Future longitudinal research focusing on PAC throughout adolescent development could further our understanding of the relationship between mental health and cognition and facilitate the development of new methods for the identification and treatment of youth mental health.

The Bispectrum and Its Relationship to Phase-Amplitude Coupling

Abstract:Most biological signals are non-Gaussian, reflecting their origins in highly nonlinear physiological systems. A versatile set of techniques for studying non-Gaussian signals relies on the spectral representations of higher moments, known as polyspectra, which describe forms of cross-frequency dependence that do not arise in time-invariant Gaussian signals. The most commonly used of these employ the bispectrum. Recently, other measures of cross-frequency dependence have drawn interest in EEG literature, in particular those which address phase-amplitude coupling (PAC). Here we demonstrate a close relationship between the bispectrum and popular measures of PAC, which we relate to smoothings of the signal bispectrum, making them fundamentally bispectral estimators. Viewed this way, however, conventional PAC measures exhibit some unfavorable qualities, including poor bias properties, lack of correct symmetry and artificial constraints on the spectral range and resolution of the estimate. Moreover, information obscured by smoothing in measures of PAC, but preserved in standard bispectral estimators, may be critical for distinguishing nested oscillations from transient signal features and other non-oscillatory causes of "spurious" PAC. We propose guidelines for gauging the nature and origin of cross-frequency coupling with bispectral statistics. Beyond clarifying the relationship between PAC and the bispectrum, the present work lays out a general framework for the interpretation of the bispectrum, which extends to other higher-order spectra. In particular, this framework holds promise for the detailed identification of signal features related to both nested oscillations and transient phenomena. We conclude with a discussion of some broader theoretical implications of this framework and highlight promising directions for future development.

Abstract:Abstract Objective. Diagnosing epilepsy still requires visual interpretation of electroencephalography (EEG) and magnetoencephalography (MEG) by specialists, which prevents quantification and standardization of diagnosis. Previous studies proposed automated diagnosis by combining various features from EEG and MEG, such as relative power (Power) and functional connectivity (FC). However, the usefulness of interictal phase–amplitude coupling (PAC) in diagnosing epilepsy is still unknown. We hypothesized that resting-state PAC would be different for patients with epilepsy in the interictal state and for healthy participants such that it would improve discrimination between the groups. Approach. We obtained resting-state MEG and magnetic resonance imaging (MRI) in 90 patients with epilepsy during their preoperative evaluation and in 90 healthy participants. We used the cortical currents estimated from MEG and MRI to calculate Power in the δ (1–3 Hz), θ (4–7 Hz), α (8–13 Hz), β (13–30 Hz), low γ (35–55 Hz), and high γ (65–90 Hz) bands and FC in the θ band. PAC was evaluated using the synchronization index (SI) for eight frequency band pairs: the phases of δ, θ, α , and β and the amplitudes of low and high γ . First, we compared the mean SI values for the patients with epilepsy and the healthy participants. Then, using features such as PAC, Power, FC, and features extracted by deep learning (DL) individually or combined, we tested whether PAC improves discrimination accuracy for the two groups. Main results. The mean SI values were significantly different for the patients with epilepsy and the healthy participants. The SI value difference was highest for θ /low γ in the temporal lobe. Discrimination accuracy was the highest, at 90%, using the combination of PAC and DL. Significance. Abnormal PAC characterized the patients with epilepsy in the interictal state compared with the healthy participants, potentially improving the discrimination of epilepsy.

Abstract:Phase-amplitude coupling is a phenomenon observed in several neurological processes, where the phase of one signal modulates the amplitude of another signal with a distinct frequency. The modulation index (MI) is a common technique used to quantify this interaction by assessing the Kullback-Leibler divergence between a uniform distribution and the empirical conditional distribution of amplitudes with respect to the phases of the observed signals. The uniform distribution is an ideal representation that is expected to appear under the absence of coupling. However, it does not reflect the statistical properties of coupling values caused by random chance. In this paper, we propose a statistical framework for evaluating the significance of an observed MI value based on a null hypothesis that a MI value can be entirely explained by chance. Significance is obtained by comparing the value with a reference distribution derived under the null hypothesis of independence (i.e., no coupling) between signals. We derived a closed-form distribution of this null model, resulting in a scaled beta distribution. To validate the efficacy of our proposed framework, we conducted comprehensive Monte Carlo simulations, assessing the significance of MI values under various experimental scenarios, including amplitude modulation, trains of spikes, and sequences of high-frequency oscillations. Furthermore, we corroborated the reliability of our model by comparing its statistical significance thresholds with reported values from other research studies conducted under different experimental settings. Our method offers several advantages such as meta-analysis reliability, simplicity and computational efficiency, as it provides p-values and significance levels without resorting to generating surrogate data through sampling procedures.

Abstract:Phase-amplitude coupling (PAC) between brain oscillations of different frequencies plays a fundamental role in neural processing, and phase-dependent neuromodulation has emerged as a promising strategy to modulate PAC. In the hippocampus, theta-gamma PAC is critically involved in memory-related functions and information propagation. Computational models provide a valuable platform for investigating the neurobiological mechanisms underlying phase-dependent effects, bypassing the limitations of in vivo and in vitro experiments. In this study, we extended a previously published computational model of the hippocampal CA3 region using the NEURON and Python environments. A closed-loop autoregressive (AR) forward prediction model was employed to sample the network's local field potential (LFP) in real time, enabling the precise calculation of phase-locked stimulus time points. Our results demonstrated the successful delivery of phase-locked current injections to all neuronal populations at both the peak and trough of theta oscillations. Phase-specific alterations in the theta band were observed during stimulation, along with enhanced theta-gamma coupling induced by peak-phase stimulation. Single neuron activity analysis highlighted the critical role of oriens lacunosum-moleculare (OLM) cells in modulating phase-dependent network dynamics. These findings underscore the potential of closed-loop stimulation systems to modulate PAC, with significant implications for the treatment of neurological disorders characterized by abnormal oscillatory activity, such as Alzheimer's disease and other memory-related disorders.

Abstract:Pelvic lymphoceles, occurred in patients undergone radical retropubic prostatectomy, are believed to increase the risk of deep venous thrombosis and so thromboembolic complications. The authors' aim is to evaluate the usefulness of pelvic ultrasonography in the diagnosis and in the possible early treatment of pelvic lymphoceles. Fifty-eight patients undergone radical prostatectomy and pelvic lymphadenectomy, between January '98 and December '99, underwent a pelvic ultrasonography in VII post-operative day. When a symptomatic or large (over 5 cm) lymphocele was found, it was treated with ultrasound (US) guided drainage. In the study, the following parameters were considered: lymphocele occurrence, size, location and treatment and thromboembolic complication. Statistical analysis was carried out with Fisher's exact test. Pelvic US showed a lymphocele in 23 out of 58 (40%) patients. Mean size was 5 cm. Deep venous thrombosis occurred in 4 patients, all with pelvic lymphoceles. No thromboembolic complications were recorded. In our experience, pelvic ultrasound has shown to be an easy e reliable tool for diagnosis and percutaneous treatment of pelvic lymphoceles.
Optimizing real-time phase detection in diverse rhythmic biological signals for phase-specific neuromodulation

Abstract:Closed-loop, phase-specific neurostimulation is a powerful method to modulate ongoing brain activity for clinical and research applications. Phase-specific stimulation relies on estimating the phase of an ongoing oscillation in real time and issuing a control command at a target phase. Phase detection algorithms based on Fast Fourier transform (FFT) are widely used due to their computational efficiency and robustness. However, it is unclear how algorithm performance depends on the spectral properties of the input signal and how algorithm parameters can be optimized. We used offline simulation to evaluate the performance of three algorithms (endpoint-corrected Hilbert Transform, Hilbert Transform and phase mapping) on three rhythmic biological signals with distinct spectral properties (rodent hippocampal theta potential, human EEG alpha and human essential tremor). First, we found that algorithm performance was more strongly influenced by signal amplitude and frequency variation compared with signal to noise ratio. Second, our simulations showed that the size of the data window for phase estimation was critical for the performance of FFT-based algorithms, where the optimal data window corresponds to the period of the oscillation. We validated this prediction with real time phase detection of hippocampal theta oscillations in freely behaving rats performing spatial navigation. Our findings define the relationship between signal properties and algorithm performance and provide a convenient method for optimizing FFT-based phase detection algorithms.
