Abstract:Two hundred sixteen early-weaned piglets (initial BW 5.8 +/- 1.1 kg) were used to evaluate the effects of microbial phytase (Natuphos (R)) on growth performance and the digestibility of phosphorus (P), crude protein (CP) and organic matter (OM) in maize-soyabean meal based diets. Pigs were allotted to three treatments with eight pigs per pen and nine pens per treatment. Pigs were phase-fed in two, two-week periods each. Three dietary total P levels combination (DM basis) for pigs in phase 1 and 2 were: 1. control, 0.8 and 6.9 g P/kg diet (1.2 g P/kg diet from dicalcium phosphate - DCP) without phytase, 2. 50% DCP reduction of the control + phytase (50% DCP reduction + phytase), 7.5 and 6.3 g P/kg diet (0.6 g P/kg diet from DCP) and 3. without supplementation + phytase (no DCP + phytase), 6.9 and 5.8 g P/kg diet, respectively. Phytase was added to the phytase-supplemented diets at a level of 500 phytase units/kg of feed for both phases. Chromic oxide (2.5 g/kg) was added to the diets as an indigestible marker to determine coefficient of total tract apparent digestibility (CTTAD) of feed. There were no differences (P > 0.05) in overall average daily gain (295, 311 and 297 g/d) and feed/gain ratio (1.47, 1.46 and 1.46) for the control, 50% DCP reduction + phytase and no DCP + phytase diets, respectively. The CTTAD of organic P in the diets with added phytase was significantly higher (P < 0.01) than in the control diet in both phase 1 (0.43 and 0.40 vs 0.32) and 2 (0.42 and 0.36 vs 0.28). The CTTAD of organic matter (OM) was also improved by phytase in phase 2. However, the CTTAD of inorganic P and CP was not influenced by phytase supplementation. Serum inorganic phosphorus concentration (119 mg/L) in pigs fed the diet with 50% DCP reduction + phytase was higher (P < 0.05) than in pigs fed the control diet (101 mg/L) and the diet without DCP supplementation + phytase (105 mg/L) in phase 2 and was higher on d 1 after weaning than in phase 1 and 2. In summary, phytase supplementation of reduced inorganic phosphorus diets, fed to young pigs, maintained performance and improved the digestibility of organic P and decreased the excretion of faecal phosphorus.

Abstract:Myo-inositol is a breakdown product of phytate produced in the gut through the action of phytase. Although the effect of phytase-released phosphorus (P) on growth performance of animals has been well characterized, there is still little understanding of the effect of myo-inositol. The first objective of this study was to determine the effects of added myo-inositol to a phytate-rich low-protein diet on growth performance and apparent total tract digestibility (ATTD) in growing piglets. The second objective was to determine whether myo-inositol could directly affect intestinal epithelial cell proliferation and function for which we used intestinal porcine epithelial cells (IPEC-J2). A total of 128 weanling piglets were allotted to four dietary treatments consisting of eight replicates per treatment and four piglets per replicate in a randomized complete block design for 4 wk. The four experimental diets comprised the positive control [PC; 20% crude protein (CP)], negative control (NC; 17% CP), negative control plus 2.0g/kg myo-inositol (NC+INO; 17% CP), and negative control plus 3000FTU/kg phytase (NC+PHY; 17% CP). Average daily feed intake (ADFI), average daily gain (ADG), and gain-feed ratio (G: F) were recorded. Phytase supplementation in the protein-deficient NC diet increased the G:F ratio (P < 0.05) without myo-inositol effects on growth performance. Phosphorus digestibility in the phytase-supplemented group increased compared to the PC, NC, and NC+INO groups, whereas plasma myo-inositol concentration was significantly higher (P < 0.05) in the NC+INO group. Due to the lack of myo-inositol effect on growth performance, an additional in vitro study was conducted to determine the direct effect of myo-inositol on the intestinal epithelium that might not be reflected in growth performance. Myo-inositol increased the mRNA abundance of selected nutrient transporters in a concentration-dependent manner (P < 0.05). Myo-inositol also enhanced barrier integrity in the IPEC-J2 monolayer by increasing the transepithelial electrical resistance (TEER) with reduced paracellular permeability of FITC-dextran (P < 0.05). In conclusion, despite the lack of myo-inositol effect on animal performance, the in vitro data indicate that myo-inositol may directly regulate gut barrier integrity. Addition of myo-inositol to pig diets at levels that enhance intestinal epithelial cell function may result in effects on growth performance and gut health of pigs.

Abstract:An experiment was conducted to test two hypotheses: 1) reducing dietary Ca and P reduces gastric pH and diarrhea in weanling pigs; 2) negative effects of low Ca and P on pig growth performance may be overcome if phytase is added to the diets. A total of 320 weanling pigs (6.35 ± 0.87 kg) were allotted to eight corn-soybean meal-based diets in a randomized complete block design with five pigs per pen. Two phase 1 (days 1 to 14) control diets containing 100 or 50% of total Ca and digestible P relative to the requirement, and six diets in which 500, 2,000, or 16,000 units of phytase/kg feed (FTU) were added to each control diet were formulated. Phytase was assumed to release 0.16% total Ca and 0.11% digestible P. Common diets were fed in phases 2 (days 15 to 27) and 3 (days 28 to 42). Growth performance data were recorded within each phase. Data for fecal scores and gastrointestinal pH were recorded for phase 1. Colon content (day 14), the right femur (days 14 and 42), and blood samples (days -1, 14, 27, and 42) were collected from one pig per pen. In phase 1, reducing Ca and P did not reduce gastric pH or fecal score, but pigs fed the 50% diets had reduced (P < 0.05) average daily gain (ADG) and average daily feed intake (ADFI) compared with pigs fed the 100% diets. In both 50% and 100% diets, phytase above 500 FTU increased (P < 0.05) gain:feed ratio (G:F) and tended (P < 0.10) to reduce gastric pH of pigs. From days 1 to 42, pigs fed the 50% diets tended (P < 0.10) to have reduced ADG and ADFI compared with pigs fed the 100% diets, but among the 100% diets, pigs tended (P < 0.10) to have a linear increase in G:F as phytase level increased. Pigs fed the 50% diets had reduced (P < 0.05) concentrations of inositol phosphate esters (IP) in the colon and reduced bone ash (days 14 and 42) compared with pigs fed the 100% diets. Phytase did not affect bone ash or most blood metabolites. Concentrations of IP in the colon decreased, whereas plasma inositol increased (d 14; P < 0.05) in pigs fed diets with phytase (≥ 500 FTU). In pigs fed the 100% diets, IP in the colon linearly decreased (P < 0.05), but plasma inositol linearly increased (P < 0.05) with increasing levels of phytase. In conclusion, reducing Ca and P in diets for weanling pigs did not influence gastric pH or fecal score, but compromised growth performance and bone ash. However, regardless of dietary Ca and P, high doses of phytase increased phytate degradation and inositol absorption, which consequently increased G:F of pigs.

Abstract:This study was carried out to determine the effects of phytase on the growth performance, bone mineralization, digestion and metabolization of nutrientsubstance in Avian broilers. Each of 400, 1 day old male Avian broilers and 400, 1 day old female Avian broilers was randomly divided into 5 groups (Group I, available phosphorus-adequate group without phytase; Group II, available phosphorus-deficient group without phytase; Group III, available phosphorus-deficient group with 250 U kg(-1) phytase; Group IV, available phosphorus-deficient group with 500 U kg(-1) phytase; Group V, available phosphorus-deficient group with 750 U kg(-1) phytase). Each group included 4 replicates, each with 20 broilers. The results showed that the effect of phytase on the growth permance in male broilers was more obvious than that of female. Dry tibia weights of male and female broilers fed with Group II and III diets were decreased compared with Group I; Group IV and V were increased compared with Group I. Tibia phosphorus contents of male and female broilers fed with Group IT-V were decreased compared with Group I. Tibia calcium contents of male broilers fed with Group II and V diets were increased by compared with Group I; Group III and IV were decreased compared with Group I. Tibia calcium contents of female broilers fed with Group II, IV and V diets were increased compared with Group I; Group III was decreased compared with Group I. Apparent digestibility coefficients of total phosphorus of Group II-V in male and female broilers were increased compared with Group I. Phosphorus concentrations of faeces of Group II-V in male and female broilers were decreased compared with Group I. Apparent digestibility coefficients of calcium Group II-V in male and female broilers were increased compared with Group I. The result showed that phytase added to available phosphorus-deficient broiler diet could improve the growth performance, bone mineralization as well as calcium and phosphorus availability.
Effects of phytase on apparent digestibility of organic phosphorus and nutrients in maize–soya bean meal based diets for sows

Abstract:Thirty crossbred sows (PIC-22) were used to investigate the effects of phytase on phosphorus utilization and the lactation performance of sows for two dietary inorganic phosphorus contents in maize–soya bean meal diets. The sows were allotted to treatments on day 100 of gestation. The treatment diet (Phytase) contained 5.4g total phosphorus/kg with added phytase (Natuphos®) at 500 Phytase Units (PU)/kg diet and the control diet (Control) contained 7.4g total phosphorus/kg without added phytase. Chromic oxide (2.5g/kg) was added to the diets as a marker to determine apparent digestibility of feed composition. Blood samples were collected from the anterior vena cava at farrowing and at weaning to determine serum inorganic phosphorus and urea nitrogen. The coefficient of total tract apparent digestibility (CTTAD) of total phosphorus, organic phosphorus, crude protein and organic matter in the phytase diet was, respectively, 0.73, 0.61, 0.73 and 0.83, which was significantly higher (P<0.01) than 0.59, 0.28, 0.62 and 0.77 in the control diet. Total phosphorus content in the faeces of sows fed the phytase diet decreased by 27.2% compared to the faeces of sows fed the control diet (P<0.01). The serum inorganic phosphorus concentrations (107 and 103mg/l for control and phytase diets, respectively) at farrowing were higher (P<0.01) than the serum inorganic phosphorus levels (90 and 91mg/l for control and phytase diets) at weaning. The sow body weights and backfat changes, litter sizes, and litter weights at weaning were not influenced by a reduction in inorganic phosphorus and by a concomitant supplementation of phytase.

Abstract:Abstract Global concern about sustainability has led many countries to establish legislation to reduce phosphorus (P) pollution. Phytase application has evolved significantly in the last years, with greater dose rates leading to a greater reduction in inorganic P supplementation and decreased feeding costs. An experiment was conducted to determine the impact of 3 supplementary levels of phytase on pig growth performance, bone mineralization, and mineral excretion (P, Ca, and N) in corn/barley/wheat/SBM based diets containing no inorganic P. Crossbred [(Landrace x Large White) Duroc] pigs [n=137; initial body weight (BWi) = 16.8 ± 4.04 kg) were housed in 24 pens containing 5 or 6 pigs/pen) for 62-day trial. After a 21-day adaptation period, each pen was assigned one of three experimental treatments according to the dietary inclusion dose of the study phytase (Quantum Blue, AB Vista): 500, 800 and 1,500 FTU/kg feed. The feeding program was based on a single diet that was pelleted and formulated according to FEDNA (2013) recommendations. The basal diet was designed with a similar composition of ingredients and nutrients (2,450 kcal/kg net energy, 15.3% crude protein, and 0.23% phytic P) except for the levels of total P and calcium. Feed consumption (per pen) and individual BW of the animals were recorded on days 0, 21, 42, and 62 of the study. On day 62 of the study, pigs were euthanized. Feces and blood samples from 2 pigs per pen were collected on days 0, 42, and 62 of the study. Data were analyzed using a mixed effects model with repeated measures. The statistical model included experimental treatment, initial BW block, study day and their interaction as fixed effects, and pen as a random effect of the fit model platform in JMP 16.0. The addition of 1,500 FTU/kg phytase tended to improve (P = 0.10) feed conversion by 4.3% relative to lower phytase inclusion levels. This production improvement is explained by the combination of greater animal growth (1.3%) and lower feed consumption (3.5%). Tendencies were observed in the net bone weight (10.41, 11.50, and 11.24 g, for 500, 800, and 1,500 FTU/kg, respectively; P = 0.05) and in bone ash content (6.26, 6.88, and 6.71 g for 500, 800, and 1,500 FTU/kg, respectively; P = 0.08), with greater values at 800 and 1,500 FTU/kg compared with 500 FTU/kg. The inclusion of 1,500 FTU/kg phytase significantly (P < 0.05) reduced fecal P and calcium content throughout the study compared with inclusion levels of 500 and 800 FTU/kg. The results of this study demonstrate the potential of adding 1,500 FTU/kg phytase to diets formulated without inorganic P to improve feed conversion, bone mineralization, and environmental sustainability, and could be an economically advantageous solution to reduce feed costs.

Abstract:Abstract This study aimed to determine the efficacy of a bacterial 6-phytase (Buttiauxella spp.) supplemented beyond traditional dose levels based on jejunal mucosa-associated microbiota, apparent ileal digestibility (AID), intestinal health and bone parameters, and growth performance of nursery pigs. Seventy-two weaned pigs (36 barrows and 36 gilts at 21 d of age with 5.8 ± 0.5 kg BW) were allotted to six treatments based on randomized complete block design with sex and initial BW as blocks and fed in three dietary phases (phase 1 for 14 d, phase 2 for 10 d and phase 3 for 14 d). The treatments included a negative control (NC) diet without phytase formulated meeting nutrient requirements by NRC and the other five treatments deficient in calcium (Ca) and phosphorus (P) by 0.12% with increasing levels of a bacterial 6-phytase (0, 500, 1,000, 2,000, and 5,000 FTU/kg feed). Titanium dioxide (0.4%) was added to phase 3 diets as an indigestible marker to measure AID of nutrients. On d 45, all pigs were euthanized to collect ileal digesta to measure AID, the third metacarpus to measure bone parameters, and jejunal mucosa to evaluate intestinal health and microbiota. Data were analyzed using the MIXED procedure for polynomial contrasts and the NLMIXED procedure for broken line analysis using the SAS 9.4. Broken line analysis demonstrated that 948 FTU/kg feed increased (P < 0.05) the ADG and the bone P content. Increasing phytase supplementation increased (linear, P < 0.05) AID of CP, bone P and ash content. Increasing phytase supplementation reduced (P < 0.05) the fecal score during phases 2 and 3. Broken line analysis demonstrated that 1,889 FTU/kg feed increased (P < 0.05) bone breaking strength. Increasing phytase supplementation (PC vs. Phy) increased (P < 0.05) AID of ether extract (EE) and P. The supplementation of phytase at 2,000 FTU/ kg feed tended (P = 0.087) to reduce the relative abundance of Prevotellaceae. In conclusion, the supplementation of a bacterial 6-phytase beyond traditional dose levels improved bone breaking strength, bone ash and P content, AID of CP, EE, and P, and growth performance of nursery pigs with reduced relative abundance of Bacteroidetes specifically Prevotellaceae in the jejunal mucosa. Supplementation of a bacterial 6-phytase between 1,000 and 2,000 FTU/kg feed provided benefits associated with growth performance and bone parameters of nursery pigs.

Effects of microbial phytase on standardized total tract digestibility of phosphorus in feed phosphates fed to growing pigs

Abstract:An experiment was conducted to test the hypothesis that the apparent total tract digestibility (ATTD) and the standardized total tract digestibility (STTD) of P in feed phosphates are increased by microbial phytase when fed to growing pigs. Monocalcium phosphate (MCP), monosodium phosphate (MSP), and magnesium phosphate (MgP) from volcanic deposits were used in the experiment. Three corn-soybean meal based diets that contained 0, 500, or 4,000 units of microbial phytase (FTU), but no feed phosphates, were formulated. Nine additional diets were formulated by adding each of the three feed phosphates to the three basal diets. A P-free diet was also formulated to estimate the basal endogenous loss of P, and therefore, 13 diets were used in the experiment. A total of 117 growing barrows (initial body weight: 15.56 ± 1.68 kg) were allotted to the 13 diets with 9 pigs per diet. Pigs were housed individually in metabolism crates equipped with a feeder and a nipple drinker. Installation of a screen floor under the slatted floor allowed for collection of feces. Diets were fed for 10 d, with the initial 5 d being a period of adaptation to the diet followed by a collection period of 4 d. During the experiment, pigs were fed equal amounts of feed twice daily at 0800 and 1600 h. Results indicated that the ATTD and STTD of P in all diets increased with the inclusion of 500 or 4,000 FTU, but the ATTD and STTD of P in the feed phosphates were not affected by the inclusion of phytase. This indicates that the increases in ATTD and STTD of P that were observed in the mixed diets when phytase was used were due to the release of P from phytate in corn and soybean meal and not from an increase in digestibility of P in feed phosphates. However, MgP had a lower (P < 0.05) ATTD and STTD of P than MCP and MSP. In conclusion, microbial phytase does not increase the digestibility of P in MCP, MSP, or MGP, but the digestibility of P in MgP is less than in MCP and MSP.
161 Impact of insoluble dietary fiber on the efficacy of the phytase enzyme in nursery pigs

Abstract:Abstract The objective of this experiment was to determine the effect of insoluble fiber on the efficacy of phytase in nursery pig diets. A total of 480 weaned pigs (BW 5.48 ± 0.14 kg; L337 X Camborough, PIC, Hendersonville, TN) were blocked by BW and randomly allotted to 48 pens. A common nutrient-adequate diet was fed from d 0–9. From d9-14, 2 phosphorus [P] deficient basal diets were fed: a low fiber [LF] corn-soybean meal diet (neutral detergent fiber [NDF]=7.5%; standardized total tract digestible [STTD] P = 0.16%) or a higher fiber [HF] corn-soybean meal plus 20% corn bran diet (NDF=15.2%; STTD P = 0.14%). From d 15–35, pigs received 8 dietary treatments (6 pens per treatment): 4 increasing levels of added phytase (0, 109, 218, and 327 phytase units [FTU]/kg; Quantum-Blue-5G, AB-Vista, Wiltshire, UK) applied to either the LF or HF basal diets. Titanium dioxide served as a digestibility marker. Fecal samples were collected on d 32–34. On d 35, one pig per pen was euthanized and fibulas harvested for bone ash assay. Data were analyzed using PROC MIXED of SAS. There were no interactions between fiber and phytase (P > 0.100). Adding phytase increased ADG (0.268, 0.282, 0.291, 0.306 kg/d; P < 0.001) and G:F (0.52, 0.55, 0.56, 0.58; P < 0.001) for the 4 increasing phytase levels, respectively. Fiber decreased ADG (0.282 vs 0.291 kg/d; P = 0.033) and tended to decrease G:F (0.54 vs 0.56; P = 0.097). There were no effects of phytase or fiber on ADFI (P > 0.100). Phytase increased bone ash (P = 0.005), but there was no effect of fiber (P = 0.949). Phytase did not affect the ATTD of DM, NDF, or ADF (P > 0.100) while fiber decreased the ATTD of DM, NDF and ADF (P < 0.001). In conclusion, increasing insoluble fiber did not affect the ability of phytase to improve growth performance and bone mineralization when pigs were fed a P deficient diet.

Abstract:A 42-days study was conducted to evaluate the effects of different dietary types (corn-or wheat-soybean meal-based diet) and phytase (Phy) or a multi-carbohydrase and phytase complex (MCPC) supplementation on growth performance, digestibility of phosphorus (P), intestinal transporter gene expression, plasma indexes, bone parameters, and fecal microbiota in growing pigs. Seventy-two barrows (average initial body weight of 24.70 ± 0.09 kg) with a 2 × 3 factorial arrangement of treatments and main effects of diet type (corn-or wheat-soybean meal-based-diets) and enzyme supplementation (without, with Phy or with MCPC). Each group was designed with 6 replicate pens. The MCPC increased (p < 0.05) average daily gain (ADG) and final body weight (BW). A significant interaction (p = 0.01) was observed between diet type and enzyme supplementation on apparent total tract digestibility (ATTD) of P. The ATTD of P was higher (p < 0.05) in wheat soybean meal-based diets compared to corn-soybean meal-based diets. Compared with the corn-soybean meal-based diet, the relative expression of SLC34A2 and VDR genes in the ileum and SLC34A3 in jejunum of growing pigs fed the wheat-soybean meal based diet was lower (p < 0.05). The MCPC significantly reduced (p < 0.05) the relative expression of TRPV5 and CALB1 genes in the ileum and increased the expression of CALB1 in the duodenum compared to control diet. The phytase increased (p < 0.05) the relative expression of SLC34A1 gene in the duodenum in comparison to control diet and MCPC-supplemented diet. The Ca and P contents in plasma from pigs fed corn-soybean meal-based diet were higher (p < 0.05) than those from pigs fed wheat-soybean meal-based diet, and the parathyroid hormone (PTH) and calcitonin (CT) concentrations were lower (p < 0.05) than those fed wheat-soybean meal-based diet. The content of Ca and P in the femur and the bone strength of pigs in the corn-soybean meal group were significantly higher (p < 0.05) than those in the wheat-soybean meal groups. The phytase increased (p < 0.05) the Ca and P content and bone strength of the femur. Additionally, diet type and both enzymes significantly improved fecal microbial diversity and composition. Taken together, diet type and exogenous enzymes supplementation could differently influence the growth performance, utilization of phosphorus, intestinal transporter gene expression, bone mineralization and microbial diversity and composition in growing pigs.

Abstract:Abstract This study was conducted to determine the efficacy of a bacterial 6-phytase on apparent total tract digestibility (ATTD) of nutrients and retention of phosphorus (P) in nursery and growing pigs. In experiment (Exp.) 1, twelve barrows (initial BW of 8.5 ± 0.3 kg) were individually housed in metabolism crates and were allotted based on a quadruplicate 3 × 3 Latin square design with three treatments and three periods. In Exp. 2, twelve barrows (initial BW of 35.7 ± 1.9 kg) were individually housed in metabolism crates and were allotted following the same experimental design of Exp. 1. The dietary treatments were: positive control (PC): a diet formulated meeting nutrient requirements from NRC (2012); negative control (NC): a diet 0.14% deficient in the Ca and P requirements; and a diet with bacterial 6-phytase at 500 FTU/kg of feed (PHY; Rovabio PhyPlus, Adisseo, France). Each period lasted for eight days, and consisted of a 4-day adaptation period and a 4-day collection period. All pigs were weighed at the end of each period to adjust daily feed allowance. Chromium oxide, at 0.5%, was added as an external marker indicating the start and end of fecal collection. Urine and fecal samples were collected daily and stored until further chemical analysis. Data from both experiments were analyzed using the MIXED procedure of SAS 9.4. Treatments were considered fixed effects, and the individual pig and period were included as random effects. In both experiments, the ATTD of Ca and P, and P retention in PC were greater (P < 0.05) compared with NC. Supplementation of bacterial 6-phytase improved (P < 0.05) ATTD of Ca (45 to 57%) and P (31 to 45%) and P retention (31 to 43%) in nursery pigs, and ATTD of P (44 to 55%) and P retention (42 to 54%) in growing pigs. No differences were detected on the ATTD of DM and CP of nursery and growing pigs among the treatments, and on nitrogen (N) retention for growing pigs. In conclusion, the supplementation of a bacterial 6-phytase improved the ATTD and retention of P in nursery and growing pigs. Results from both experiments indicated that the supplementation of a bacterial 6-phytase in diets for nursery and growing pigs can be a potential alternative to reduce the high levels of inorganic P sources, whereas still increasing the digestibility and utilization of P by nursery and growing pigs.

Over 60% of the total P in cereal grains and oil seeds and their by-products is found as phytate, myo-inositol hexakisphosphate. Phytases are a group of enzymes that initiate the phosphate hydrolysis from phytate and produce myo-inositol and inorganic phosphate by catalyzing the stepwise removal of the phosphate groups. Microbial phytases effectively improve dietary phytate-P utilization and may partially or completely replace inorganic P supplementation in swine and poultry diets. Replacing inorganic P with phytase in these diets can reduce P excretion by up to 50% and, enhance bioavailabilities of Ca, Zn and Fe. The improvement in nitrogen retention and protein and amino acid utilization by dietary phytase supplementation is relatively small and highly variable. The efficacy of supplemental phytase is affected by at least three dietary factors: Ca:P ratio, cholecalciferol and organic adds.

Abstract:Abstract This experiment investigated the effects of protease supplementation to low amino acid (AA) diets containing phytase on pig growth performance, postweaning intestinal health and carcass characteristics. A total of 936 weaned pigs (21 d of age, initial BW 5.87 ± 0.31 kg) were used in a 2 × 2 factorial design comparing the main effects of AA supply [standard feeding program: balanced for all nutrients with adjustment of Ca and P due to inclusion of phytase (2,500 FTU/kg in Phase 1 to 4; 500 FTU/kg in Phase 5 to 9) vs. low AA feeding program: 15% lower standardized ileal digestible lysine with relative reduction of all other essential AA] and protease level (0 vs. 0.0125%). Pens were assigned to dietary treatment according to a randomized complete block design with 26 pigs per pen and nine replicates per dietary treatment. Feed and water were provided on an ad libitum basis for all phases throughout the wean-to-finish period. Feed intake and body weight were determined every 2 wk during nursery period and monthly in the grow-finish period. Intestinal health in the first 17 d was assessed based on lactulose:mannitol ratio (L:M), serum IgA, and pen diarrhea assessment. Overall, pigs fed standard wean-to-finish diets had greater (P < 0.05) ADG and G:F than pigs fed low AA diets. Pig growth performance was not different throughout the wean-to-finish period with or without protease supplementation and with no interaction between AA supply and protease supplementation. There were no differences among dietary treatments for carcass characteristics. No difference was observed for urinary L:M and serum IgA; however, the L:M ratio was approximately 32% lower in pigs fed low AA diets + protease compared with pigs fed standard and low AA diets at d 5 and d 17 postweaning. Pigs fed protease supplemented diets had lower incidence of diarrhea (χ2 < 0.05) compared with pigs fed diets without protease. Results of the experiment indicate that dietary protease supplementation benefits intestinal health of nursery pigs.

Abstract:The aim of this study was to determine how different doses of phytase in diets with extruded soybean seeds and rapeseed meal affected pigs' growth performance, meat quality, bone mineralization, and fatty acid profiles. Sixty pigs were divided into three treatments by sex and body mass. Pigs were divided into starter (25 days), grower (36 days), and finisher (33 days) periods and fed with mash diets. No phytase was used in the control group diet, whereas in Phy1 and Phy2, 100 g and 400 g of phytase per ton of mixture were used, respectively. The feed conversion ratio and meat color were significantly correlated with phytase. Phytase supplementation had no effect on the growth of pigs, but total phosphorus was significantly increased in the bones and meat of the pigs. The enzyme additive reduced the C22:4 n-6 acid content in the meat, whereas other results were not significantly affected. The data suggest that the addition of phytase at a dosage of 100 g/ton to diets with extruded full-fat soya seeds and rapeseed meal can be valuable, as it reduces the FCR and increases the P content in the meat and bones.

Abstract:Animals with intrauterine growth retardation (IUGR) usually undergo injured postnatal growth and development during the early period after birth. Equol (Eq), an isoflavan produced by gut bacteria in response to daidzein intake, has various health benefits. Therefore, the objective of this study was to evaluate whether Eq supplementation can influence the growth performance, redox status, intestinal health and skeletal muscle development of weanling piglets with IUGR. A total of 10 normal-birth-weight (NBW) newborn female piglets and 20 newborn female piglets with IUGR were selected. After weaning at the age of 21 d, 10 NBW piglets and 10 IUGR piglets were allocated to the NBW group and IUGR group, respectively, and offered a basal diet. The other 10 IUGR piglets were allocated to the IUGR + Eq group and offered a basal diet with 50 mg of Eq per kg of diet. The whole trial lasted for 21 d. At the end of the feeding trial, all piglets were sacrificed for the collection of serum, intestinal tissues and skeletal muscles. Supplementation with Eq increased the average daily gain (ADG), average daily feed intake (ADFI), duodenal villus height to crypt depth ratio (V/C), jejunal villus height and V/C, but reduced the duodenal crypt depth in neonatal piglets with IUGR. Meanwhile, Eq supplementation elevated the activities of superoxide dismutase (SOD) and catalase (CAT) in the serum and duodenum and the activity of SOD in the jejunum, but lowered malondialdehyde (MDA) content in the serum, jejunum and ileum of piglets with IUGR. In addition, supplementation with Eq reduced diamine oxidase (DAO) activity and the levels of D-lactate and endotoxin in serum, and the tumor necrosis factor-α (TNF-α) level in jejunum and ileum, whereas the concentration of serum immunoglobulin G (IgG) and the mRNA levels of intestinal barrier-related markers in jejunum and ileum of IUGR piglets were increased. Furthermore, supplementation with Eq elevated the percentage of fast-fibers and was accompanied with higher mRNA expression of myosin heavy chain IIb (MyHC IIb) and lower mRNA levels in MyHC I in the longissimus thoracis (LT) muscle of IUGR piglets. In summary, Eq supplementation can promote antioxidant capacity, maintain intestinal health and facilitate skeletal muscle development, thus resulting in the higher growth performance of IUGR piglets.

Abstract:The objective was to test the hypothesis that supplementation of diets for gestating sows with 25-hydroxycholecalciferol (25-OH-D3) or 1-hydroxycholecalciferol (1-OH-D3) affects serum biomarkers for bone and increases Ca and P balance and the apparent total tract digestibility (ATTD) of gross energy (GE), and the concentrations of digestible energy (DE) and metabolizable energy (ME) in diets without or with microbial phytase. Sixty multiparous sows were allotted to 1 of 6 diets. Diets were formulated using a 3 × 2 factorial with 3 inclusions of supplemental vitamin D metabolite (no metabolite, 25-OH-D3, or 1-OH-D3) and 2 inclusion levels of microbial phytase (0 or 1,000 units). Sows were housed individually in metabolism crates and feces and urine were collected quantitatively. Results indicated that there was no difference in the ATTD of dry matter (DM) and GE and concentration of DE among the 3 diets containing microbial phytase, but the ATTD of DM and GE and concentration of DE was greater (P < 0.05) in diets containing 1-OH-D3 compared with the diet without a vitamin D metabolite if phytase was not used (interaction; P < 0.05). In diets without microbial phytase, ME was greater in diets containing either one of the 2 vitamin D metabolites than in the diet without a vitamin D metabolite, but among diets with microbial phytase, the ME of the 1-OH-D3 diet was less than of the 25-OH-D3 diet (interaction; P < 0.05). No effect of microbial phytase on concentrations of DE and ME was observed. There was no interaction between supplementation of microbial phytase and vitamin D metabolites for Ca and P balances, and regardless of metabolite supplementation, use of microbial phytase increased (P < 0.05) the ATTD and retention of Ca and P. Regardless of dietary phytase, the ATTD and retention of Ca and P increased (P < 0.05) for sows fed a diet containing one of the vitamin D metabolites compared with sows fed the diet without a vitamin D metabolite. Serum biomarkers for bone resorption or bone tissue synthesis were not affected by experimental diets. In conclusion, the ATTD of DM and GE, concentrations of DE and ME, and Ca and P balance in phytase-free diets fed to sows in late gestation were increased by supplementation with 1-OH-D3 or 25-OH-D3, but no differences between the 2 vitamin D metabolites were observed. Supplementation of diets with microbial phytase increased Ca and P balance, but did not affect DE and ME of diets.

Abstract:This experiment was conducted to evaluate the equivalency values of microbial phytase to inorganic phosphorus (Pi) in 22- to 42-d-old broilers fed a corn-soybean meal diet. Ten dietary treatments were designed in which diet 1 was the basal diet [calcium 0.90%, total phosphorus 0.35%, nonphytate phosphorus (NPP) 0.13%] and diet 4 was control (calcium 0.90%, total phosphorus 0.57%, NPP 0.35%). Diets 2 and 3 included 0.20 and 0.27% of NPP, and diets 5 to 10 contained graded levels of microbial phytase (125, 250, 500, 1,000, 2,000, and 4,000 U/kg of diet). Body weight gain, feed intake, phosphorus retained, plasma Pi concentration, tibia ash content, and tibia-breaking strength increased, but plasma calcium concentration decreased with increasing dietary NPP content (from 0.13 to 0.35%) or phytase activity. Microbial phytase caused an increase, whereas dietary NPP caused a decrease in phytate phosphorus (PP) retention. When 4,000 U of phytase was added to the basal diet with 0.13% NPP, 95% PP was released based on tibia ash content, and broiler BW gain and feed intake were equal to those of broilers fed the control diet (NPP 0.35%). Microbial phytase at 125, 250, 500, 1,000, 2,000, and 4,000 U/kg of diet was determined to be equivalent to the addition of 0.035 to 0.208% of Pi from dicalcium phosphate in 22- to 42-d-old broilers. These data suggest that high levels of microbial phytase could release almost all dietary PP and replace all supplemental Pi in growing-finishing broilers.
