Abstract:Glutamate uptake and metabolism was studied in cerebral cortical astrocytes. The expression of the astrocytic glutamate transporter GLAST was found to be stimulated by extracellular glutamate through activation of kainate receptors on the astrocytes. Energy metabolism and ammonia homeostasis are two important aspects of glutamate handling in astrocytes. It is well known that glutamate transport into astrocytes and glutamine formation are energy consuming processes. Furthermore, ammonia is required for glutamine production. On the other hand, glutamate metabolism through the tricarboxylic acid cycle is an energy and ammonia producing pathway. In the present study it was shown that at an extracellular glutamate concentration of 0.5 mM, high energy phosphates were reduced, and more than 50% of the glutamate carbon skeleton entered the tricarboxylic acid cycle to yield products like lactate, aspartate, and additionally glutamate and glutamine derived from tricarboxylic acid cycle intermediates. Entry into the cycle was not affected by the transaminase inhibitor aminooxyacetic acid, indicating that deamination is the major route for 2-oxoglutarate formation from glutamate. Synthesis of glutamate from 2-oxoglutarate, however, proceeded via transamination. In an earlier study it was shown that at glutamate concentrations at and below 0.2 mM, glutamine appears to be the major product and entry of glutamate into the tricarboxylic acid cycle is decreased 70% by aminooxyacetic acid. In an attempt to unify the above mentioned results, it is suggested that availability of ammonia and energy demands are major factors determining the metabolic fate of glutamate in astrocytes.

Abstract:CASE DESCRIPTION A 5-year-old 38.3-kg (84.5-lb) mixed-breed dog was examined because of acute onset of lethargy and anorexia. Four days later, a raised, firm, warm 15 × 10-cm lesion was detected in the right caudal paralumbar area. CLINICAL FINDINGS Cephalexin treatment yielded a poor response. Formalin-fixed tissue and fluid samples from the cystic areas of the lesion were submitted for cytologic and histologic examinations, routine bacterial and mycobacterial culture, and genus identification and 16S partial sequencing via PCR assays. Cytologic examination revealed chronic pyogranulomatous inflammation. Histologic examination by use of routine, Giemsa, silver, acid-fast, and modified acid-fast stains revealed multifocal nodular granulomatous panniculitis without identifiable organisms. Mycobacteria were initially identified via PCR assay and mycobacterial culture within 3 days. Mycobaterium goodii was speciated by use of partial 16S RNA sequence analysis. TREATMENT AND OUTCOME The lesion resolved after long-term treatment with a combination of rifampin and clarithromycin and insertion of a Penrose drain. There has been no recurrence of the condition. CLINICAL RELEVANCE M goodii is an environmental rapidly growing mycobacterium and is a zoonotic pathogen. Infections have not been previously reported in domestic animals in North America, although there are rare reports of infection in humans associated with surgery, especially surgical implants. Domestic animals are a potential sentinel for this non-tuberculous mycobacterial infection in humans, although lack of speciation in infections of domestic animals likely underestimates the potential public health importance of this pathogenic organism. Current microbiological molecular methods allow for a rapid and inexpensive diagnosis.

Abstract:Dynamic interconnect holograms are designed by the use of a simulated annealing algorithm and written to a 128 × 128 pixel ferroelectric spatial light modulator that is used in a binary-phase mode. Dynamic holograms are used to implement a 2 × 2 crossbar with single-mode fiber inputs and outputs, which function with as high as 27 dB of isolation between output ports. The principle is extended to two-dimensional interconnection holograms, and arbitrary fan-out to as high as 64 points is demonstrated with good performance.Images of interconnection holograms are transferred from the spatial light modulator to an optically addressed spatial light modulator that is used in a binary-phase mode. The addition of a fixed array generator computer-generated hologram permits replication of the hologram image, thus creating a larger hologram with a high space-bandwidth product on the optically addressed spatial light modulator.Results of a preliminary experiment are presented.

Abstract:Purpose – Despite the use in companies of policy and control mechanisms to tackle cyberloafing, these practices are still popular among employees. The purpose of this paper is to suggest that control systems alone are unable to deter cyberloafing because they are eventually perceived as a sort of “ineffectual dog that may bark a lot, but ultimately does not bite.” Instead, control systems are only expected to deter cyberloafing if employees view them as leading to punitive consequences.Design/methodology/approach – First, given the easy visibility of cyberloafing activities, the paper proposes a design for control systems that not only includes perceptions of organizational control (monitoring), but also perceptions of the supervisor's physical proximity (proximity). Data are collected from university administration and services personnel, whose main working tool is the computer.
