Abstract:The purpose of this study was to investigate how pre-hypoxia exposure affected the mitochondrial structure and bioenergetic function of large yellow croaker in responding to Cu stress. Fish were acclimated to normoxia and 3.0 mg DO L−1 for 48 h, then subjected to 0 and 120 μg Cu L−1 for another 48 h. Hypoxic acclimation did not affect mitochondrial ultrastructure and reactive oxygen species (ROS), but reduced oxidative phosphorylation (OXPHOS) efficiency. Cu exposure impaired mitochondrial ultrastructure, increased ROS generation and inhibited OXPHOS efficiency. Compared with Cu exposure alone, hypoxic acclimation plus Cu exposure reduced ROS production and improved OXPHOS efficiency by enhancing mitochondrial respiratory control ratio, mitochondrial membrane potential, and activities and gene expressions of electron transport chain enzymes. In conclusion, hypoxic acclimation improved the mitochondrial energy metabolism of large yellow croaker under Cu stress, facilitating our understanding of the molecular mechanisms regarding adaptive responses of hypoxia-acclimated fish under Cu stress.

Abstract:Alkali Aggregate Reaction (AAR) and Delayed Ettringite Formation (DEF) cause expansion of the affected concrete that generally leads to cracking and decrease of its mechanical properties. As a consequence, these pathologies raise severe problems in terms of serviceability, sustainable operation of the concrete works and structural integrity. To manage with considered suffering structures, it is necessary to provide predictive models able to re-assess their mechanical state. In this paper, we describe the RGIB model developed with this objective and we compare its predictions to the results of experimental tests specifically performed to validate it. The objective is to emphasize which processes are well taken into account and which improvements could be implemented.

Abstract:Ammonia is considered to be the major chemical pollutant causing fish poisoning in aquaculture. This research aimed to evaluate the impact of acute ammonia exposure on the large yellow croaker's meat quality, gill morphology, liver oxidative stress, and hematological parameters. The fish were exposed to total ammonia nitrogen concentrations of 0, 2.96, 5.92, and 8.87 mg/L for 48 h, respectively. The findings demonstrated that all ammonia-exposed fish had higher liver lactate dehydrogenase and glutamic oxalate transaminase activities. The glucose, blood urea nitrogen, and creatinine levels in 8.87 mg/L total ammonia nitrogen (TAN) were higher than other samples. The total protein, albumin, and triglyceride levels in serum decreased significantly in ammonia-exposed samples. After 48 h of ammonia exposure, superoxide dismutase activities showed a 76.1%, 118.0%, and 156.8% increase when fish were exposed to 2.96, 5.92, and 8.87 mg/L TAN, respectively. Catalase activities and glutathione contents were considerably higher (p < 0.05) in all ammonia-treated samples compared to 0 mg/L TAN. The ammonia-treated gill lamellae become thicker, shorter, and curved. Additionally, the ammonia exposure resulted in the accumulation of free amino acids and the loss of nucleotides. The inosine monophosphate and adenosine monophosphate contents in the flesh were decreased after 12 h of exposure to 2.96, 5.92, and 8.87 mg/L ammonia compared to the control group. Overall, large yellow croakers exposed to ammonia for 6 h presented not only changes in serum composition but also oxidative stress, liver and gill tissue damage and flesh quality deterioration.

Reactive oxygen species (ROS) homeostasis is essential for the maintenance of normal physiological conditions in an organism. Direct evidence that environmental hypoxia leads to ROS overproduction is scarce in marine fish. Furthermore, the sources of ROS overproduction in marine fish under hypoxic stress are poorly known. In this study, we investigated the effects of hypoxia on redox homeostasis in L. crocea and the impact of impaired redox homeostasis on fish. We first confirmed that hypoxia drove ROS production mainly via the mitochondrial electron transport chain and NADPH oxidase complex pathways in L. crocea and its cell line (large yellow croaker fry (LYCF) cells). We subsequently detected a marked increase in the antioxidant systems of the fish. However, imbalance between the pro-oxidation and antioxidation systems ultimately led to excessive ROS and oxidative stress. Cell viability showed a remarkable decrease while oxidative indicators, such as malondialdehyde, protein carbonylation, and 8-hydroxy-2 deoxyguanosine, showed a significant increase after hypoxia, accompanied by tissue damage. N-acetylcysteine (NAC) reduced ROS levels, alleviated oxidative damage, and improved cell viability in vitro. Appropriate uptake of ROS scavengers (e.g., NAC and elamipretide Szeto-Schiller-31) and inhibitors (e.g., apocynin, diphenylene iodonium, and 5-hydroxydecanoate) may be effective at overcoming hypoxic toxicity. Waterborne zinc induced lobe-dependent effect on oxidative stress and energy metabolism in hepatopancreas of Larimichthys crocea

Abstract:The study aimed to compare differences in oxidative stress and energy metabolism between the left and right lobe of hepatopancreas in large yellow croaker Larimichthys crocea exposed to 0 (control), 20, and 100 μM Zn for 96 h. Tipical biomarkers were examined including the proportion of white hepatopancreas, lipid content, malondialdehyde (MDA) level, glutathione (GSH) content, activity levels of enzymes (Cu/Zn-superoxide dismutase, Cu/Zn-SOD; catalase, CAT; glutathione peroxidase, GPx; glutathione reductase, GR; mitochondrial ATP synthase, F-ATPase; malate dehydrogenase, MDH; succinate dehydrogenase, SDH; hepatic lipase, HTGL; lipoprotein lipase, LPL), mRNA levels of genes encoding these enzymes (sod1, cat, gpx1a, gr, atp5b, mdh, sdh, htgl, and lpl), and gene expression of signaling molecules the NF-E2-related nuclear factor 2 (nrf2) and Kelch-like ECH-associated protein 1 (keap1). A whitish color in the left lobe of hepatopancreas was observed in the control and Zn-exposed fish. Contrarily, the right lobe of hepatopancreas tended towards red with increasing Zn levels. The phenomenon was further confirmed by that lipid content was reduced in the right lobe and was not significantly affected in the left lobe by Zn. The right lobe showed higher energy consumption than the left lobe as reflected by the up-regulation of activity levels of HTGL, LPL, F-ATPase, MDH, and SDH. Lipid peroxidation declined by 20 μM Zn and was unchanged by 100 μM Zn in both lobes, which could be explained by increased activity levels of Cu/Zn-SOD and GPx. However, the magnitude of increase in Cu/Zn-SOD activity was greater in the right lobe than that in the left one. The difference in enzyme activity between two lobes may be involved in changes in mRNA levels of sod1, gr, atp5b, sdh, htgl, lpl, and nrf2, which was further confirmed by positive relationships between enzyme activity and gene expression. Our data also showed positive correlations between nrf2 expression and mRNA levels of its target genes, suggesting that Nrf2 was required for the protracted induction of these genes. Our results demonstrated the potential molecular mechanism of Zn-induced differences between lobes of hepatopancreas, suggesting that the sampling part of hepatopancreas should be considered with caution when assessing metal contamination.
Effect of Copper Sulphate Exposure on the Oxidative Stress, Gill Transcriptome and External Microbiota of Yellow Catfish, Pelteobagrus fulvidraco

Abstract:This study aimed to investigate the potential adverse effects of the practical application of copper sulfate on yellow catfish (Pelteobagrus fulvidraco) and to provide insights into the gill toxicity induced by copper sulphate. Yellow catfish were exposed to a conventional anthelmintic concentration of copper sulphate (0.7 mg/L) for seven days. Oxidative stress biomarkers, transcriptome, and external microbiota of gills were examined using enzymatic assays, RNA-sequencing, and 16S rDNA analysis, respectively. Copper sulphate exposure led to oxidative stress and immunosuppression in the gills, with increased levels of oxidative stress biomarkers and altered expression of immune-related differentially expressed genes (DEGs), such as IL-1β, IL4Rα, and CCL24. Key pathways involved in the response included cytokine–cytokine receptor interaction, NOD-like receptor signaling pathway, and Toll-like receptor signaling pathway. The 16S rDNA analysis revealed copper sulphate altered the diversity and composition of gill microbiota, as evidenced by a significant decrease in the abundance of Bacteroidotas and Bdellovibrionota and a significant increase in the abundance of Proteobacteria. Notably, a substantial 8.5-fold increase in the abundance of Plesiomonas was also observed at the genus level. Our findings demonstrated that copper sulphate induced oxidative stress, immunosuppression, and gill microflora dysbiosis in yellow catfish. These findings highlight the need for sustainable management practices and alternative therapeutic strategies in the aquaculture industry to mitigate the adverse effects of copper sulphate on fish and other aquatic organisms.

Abstract:Dissolved oxygen is an environmental factor that affects fish survival. Aquatic environments suffer from an increasing oxygen deficiency resulting from global warming and environmental pollution. In this study, we combined high-throughput transcriptome analysis with non-targeted liquid chromatography-mass spectrometrybased metabolome sequencing to assess the metabolic response to hypoxia in the liver of Pelteobagrus fulvidraco. Healthy fish with body weight of 12.39 +/- 1.37 g were subjected to hypoxic conditions at a concentration of 1.02 +/- 0.09 mg/L for durations of 6, 24 and 48 h. Hypoxia exposure increased the content of MDA, lactate, glucose and the level of T -AOC, enhanced the activity of T -SOD, CAT, GPx and LDH, and decreased SDH activity. Histological examination revealed that liver tissue was damaged after exposure, and the degree of damage increased with the prolongation of hypoxia time. In the liver transcriptome, 1527 DEGs were identified, of which 240 were co-identified in the three comparison groups. These DEGs showed significant enrichment in pathways associated with oxidation-reduction and metabolic processes. RT- qPCR confirmed transcriptional induction of metabolismrelated genes in response to hypoxia. Metabolomic results showed that the SDMs were related to amino acid, carbohydrate, and lipid metabolism. Moreover, a conjoint analysis demonstrated an increase in the activity of several key enzymes, such as phosphoenolpyruvate carboxykinase, phospholipid phosphatase 1a, and betainehomocysteine methyltransferase. An integrated regulatory network was observed to be involved in glycine, serine, and threonine metabolism; glycolysis/gluconeogenesis; tricarboxylic acid cycle; protein digestion and absorption; and lipid metabolism in response to hypoxia exposure. Overall, the findings indicated that hypoxia can cause oxidative stress in yellow catfish and that their bodies can resist stress by activating the antioxidant defense system and mobilizing multiple metabolic pathways to meet the energy demand. These results will aid in the development of strategies aimed at mitigating the adverse effects of hypoxic exposure on fish.

Abstract:The regulation mechanism of the hybrid yellow catfish "Huangyou-1" was assessed under conditions of hypoxia and reoxygenation by examination of oxygen sensors and by monitoring respiratory metabolism, oxidative stress, and apoptosis. The expressions of genes related to oxygen sensors (HIF-1α, HIF-2α, VHL, HIF-1β, PHD2, and FIH-1) were upregulated in the brain and liver during hypoxia, and recovered compared with control upon reoxygenation. The expressions of genes related to glycolysis (HK1, PGK1, PGAM2, PFK, and LDH) were increased during hypoxia and then recovered compared with control upon reoxygenation. The mRNA levels of CS did not change during hypoxia in the brain and liver, but increased during reoxygenation. The mRNA levels of SDH decreased significantly only in the liver during hypoxia, but later increased compared with control upon reoxygenation in both tissues. Under hypoxic conditions, the expressions of genes related to oxidative stress (SOD1, SOD2, GSH-Px, and CAT) and the activity of antioxidant enzymes (SOD, CAT, and GSH-Px) and MDA were upregulated compared with control. The expressions of genes related to apoptosis (Apaf-1, Bax, Caspase 3, Caspase 9, and p53) were higher than those in control during hypoxic exposure, while the expressions of Bcl-2 and Cyt C were decreased. The findings of the transcriptional analyses will provide insights into the molecular mechanisms of hybrid yellow catfish "Huangyou-1" under conditions of hypoxia and reoxygenation. Overall, these findings showed that oxygen sensors of "Huangyou-1" are potentially useful biomarkers of environmental hypoxic exposure. Together with genes related to respiratory metabolism, oxidative stress and apoptosis occupy a quite high position in enhancing hypoxia tolerance. Our findings provided new insights into the molecular regulatory mechanism of hypoxia in "Huangyou-1."

Abstract:Dissolved oxygen concentrations both in the open ocean and coast have been declining due to the interaction of global climate change and human activity. Fish have evolved different adaptative strategies to cope with possibly damage induced by hypoxic environments. Black rockfish as important economic fish widely reared in the offshore sea cage, whereas related physiological response subject to hypoxia stress remained unclear. In this study, hepatic anti-oxidant enzymes (superoxide dismutase [SOD], catalase [CAT], glutathione peroxidase [GSH-Px]), aminotransferase (AST) and alanine aminotransferase (ALT) activities, lipid peroxidation (LPO), malondialdehyde (MDA) and glutathione (GSH) content, immunological parameters and the expression of apoptosis (bax, bcl2, p53, caspase3, xiap) and immune-related genes (c3, il-1β, ccl25, saa, hap, isg15) of black rockfish were determined during hypoxia and reoxygenation to illustrate the underlying defense response mechanisms. Results showed that hypoxia stress remarkably increased hepatic LPO and MDA content, AST and ALT activity and proportion of pyknotic nucleus. Hepatic SOD, CAT and GSH-Px activity manifested similar results, whereas GSH levels significantly decreased under hypoxia stress. The apoptosis rate of hepatocyte increased during hypoxia stress and reoxygenation. Meanwhile, p53, caspase3, bax and xiap mRNAs and bax/bcl2 rations were significantly up-regulated under hypoxia stress. However, bcl2 mRNA was significantly down-regulated. Interestingly, hypoxia stress significantly increased NBT-positive cell percent, phagocytic index, respiratory burst and ACH50 activity, and lysozyme activity. The mRNA levels of c3, ilβ, ccl25, saa, hap and isg15 were significantly up-regulated in the liver, spleen and head-kidney under hypoxia stress. The above parameters recovered to normal status after reoxygenation for 24 h Thus, hypoxia stress impairs hepatic antioxidant capacity, induces oxidative damage and apoptosis via the xiap-p53-bax-bcl2 and the caspase-dependent pathways, but enhances host immunity by regulating nonspecific immune indices and related genes expression to maintain homeostasis in black rockfish. These findings will help fully understand the hypoxia tolerance mechanisms of black rockfish and provide more data for offshore open ocean farming.
Effects of β-glucan on ROS production and energy metabolism in yellow croaker (Pseudosciaena crocea) under acute hypoxic stress

Abstract:The aim of the present study was to evaluate the effect of β-glucan on acute hypoxia-induced oxidative stress and the changes in energy metabolism by determining ROS production, activities and mRNA levels of energy metabolism enzyme (PK, F-ATPase, SDH and MDH), and in gene expression of HIF-1α in the liver of large yellow croaker. Fish were injected with β-glucan at a dose of 0 or 5 mg kg−1 body weight on 6, 4 and 2 days before exposed to 1.5 and 7.0 mg DO L−1 for 48 h. The results showed that β-glucan enhanced survival rate and reduced ROS during the lethal hypoxic stress, indicating that β-glucan could ameliorate hypoxia-induced oxidative stress. Obtained results also showed that β-glucan could up-regulate activities and mRNA levels of PK, demonstrating that β-glucan increased anaerobic glycolysis capacity. Furthermore, a coordinated transcriptional regulation of energy metabolism enzyme genes was observed, suggesting that HIF-1α is required for regulating these genes. In conclusion, β-glucan could alleviate cute hypoxia-induced oxidative stress in large yellow croker by enhancing anaerobic glycolysis capacity, emphasizing a central role of transcription factor HIF-1α in the process.

Different cold tolerances among three strains of large yellow croaker: related to antioxidant defense and energy metabolism

Abstract:The aim of this study was to compare low-temperature tolerances in different strains of large yellow croaker. Dai Qu (DQ), Min-Yue Dong (MY), and Quan Zhou (NZ) strains of large yellow croaker were subjected to cold stress (8.6 °C) for 12 h, 24 h, 48 h, and 96 h. Survival rate, histological observation, and antioxidant and energy metabolism indicators were determined. The results showed that compared with the DQ group and MY group, NZ group aggravated hepatic structure, enhanced ROS, lactate, and anaerobic metabolism (PK gene expression and activity), while inhibited ATP, GSH, antioxidant enzymes (mRNA levels and activities of SOD, GPx, and CAT), and aerobic metabolism enzymes (mRNA levels and activities of F-ATPase, SDH, and MDH), indicating the reduction of cold tolerance in the NZ group was closely correlated with the decrement of antioxidative capacity and energy metabolism efficiency. Nrf2 and AMPK gene expressions were correlated with antioxidant and energy metabolism mRNA levels, respectively, suggesting Nrf2 and AMPK might participate in the modulation of target genes during the cold-stress adaptation. In conclusion, low temperature tolerance of fish depended on the antioxidant defense and energy metabolism efficiency, which contributes to understanding the underlying mechanisms of cold adaptation in large yellow croaker.
Chronic exposure of hydrogen peroxide alters redox state, apoptosis and endoplasmic reticulum stress in common carp (Cyprinus carpio)

Abstract:Hydrogen peroxide (H2O2) appears to be ubiquitous in natural water. Higher level of H2O2 can cause physiological stress, immunosuppression and even death in aquatic animals, but the physiological and molecular mechanisms of H2O2 toxicity are not well studied. Thus, the aim of the present study was to exposure potential toxic mechanisms of H2O2 via assessing the effects on redox state, apoptosis and endoplasmic reticulum (ER) stress in common carp. The fish were subjected to four concentrations of H2O2 (0, 0.25, 0.5 and 1 mM) for 14 days. And then, the tissues including blood, liver, muscle, gills, intestines, heart, kidney and spleen were collected to measure biochemical parameter and gene expression. The results showed that H2O2 exposure suppressed the majority antioxidative parameters in serum, liver, muscle and intestines, but enhanced T-SOD, CAT and T-AOC levels in gills. In all tested tissues, the MDA content was significantly promoted by H2O2 exposure. The oxidative stress-related genes including nrf2, gstα, sod, cat and/or gpx1 were upregulated in liver, gills, muscle, intestines, and/or kidney, but downregulated in heart after H2O2 exposure. Moreover, the ho-1 mRNA level was inhibited by H2O2 exposure in all tissues except intestines and spleen. After 14 days of exposure, H2O2 induced ER stress and initiated IRE1 and PERK pathways, which activated downstream genes, including chop, grp78 and/or xbp1s, to regulate UPR in liver, gills, muscle and/or heart. Meanwhile, H2O2 exposure activated MAPK pathway to regulate mitochondria-related genes including bcl-2, bax and cytc, which further triggered cas-8, cas-9 and cas-3, and accelerated apoptosis in liver, gills, muscle and heart. Importantly, in different tissues, the genes associated with oxidative stress, ER stress and apoptosis showed a different influence, and more significant influence was observed in the muscle, gills and liver. Overall results suggested that long-term H2O2 exposure induced oxidative stress, ER stress and apoptosis in the majority of tested tissues of common carp. The Nrf2, IRE1, PERK and MAPK pathways played important roles in H2O2-induced toxicity in fish. These data enriched the toxicity mechanism of H2O2 in fish, which might contribute to the risk assessment of H2O2 in aquatic environment.
Copper-induced oxidative stress, transcriptome changes, intestinal microbiota, and histopathology of common carp (Cyprinus carpio)

Abstract:Copper (Cu) is a common contaminant in aquatic environments, which could cause physiological dysfunction in aquatic organisms. However, few studies have comprehensively examined the impact of copper toxicity in freshwater fish over the past decade. In this research, the oxidative stress, liver transcriptome, intestinal microbiota, and histopathology of common carp (C. carpio) in response to Cu exposure were studied, by exposing juvenile carp to 0.2 mg/ml Cu2+ for 30 days. The results revealed that Cu2+ could induce significant changes in malondialdehyde (MDA) content and antioxidant enzyme (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx)) activity. The changes in antioxidant enzyme activities indicate that Cu can induce oxidative stress by generating reactive oxygen species (ROS) content. RNA-seq analysis of the liver identified 1069 differentially expressed genes (DEGs) after treatment with 2.0 mg/L Cu2+. Among the DEGs, 490 genes were upregulated and 579 genes were downregulated. GO functional enrichment analysis revealed that Cu could affect the fatty acid biosynthetic process, carnitine biosynthetic process, and activity of carboxylic acid transmembrane transporter. Meanwhile, the most significantly enriched KEGG pathway also included the lipid metabolism pathway. In addition, Cu2+ exposure increased bacterial richness and changed bacterial composition. At the phylum level, we found that the ratio of Bacteroidetes to Firmicutes was increased in the treatment carps, which can regulate intestinal epithelium function and reduce inflammation and immune responses. At the genus level, the abundances of 11 genera were significantly altered after exposure to Cu2+. The altered composition of the microbial community caused by Cu exposure may play a useful role in compensation of the intestinal lesions by Cu exposure. Furthermore, we found that Cu2+ exposure could cause histological alterations such as structural damage to the liver and intestines. The results of this research contribute to a better understanding of mechanisms related to Cu toxicity in fish.
Distributed flow sensing for closed-loop speed control of a flexible fish robot

Abstract:Flexibility plays an important role in fish behavior by enabling high maneuverability for predator avoidance and swimming in turbulent flow. This paper presents a novel flexible fish robot equipped with distributed pressure sensors for flow sensing. The body of the robot is molded from soft, hyperelastic material, which provides flexibility. Its Joukowski-foil shape is conducive to modeling the fluid analytically. A quasi-steady potential-flow model is adopted for real-time flow estimation, whereas a discrete-time vortex-shedding flow model is used for higher-fidelity simulation. The dynamics for the flexible fish robot yield a reduced model for one-dimensional swimming. A recursive Bayesian filter assimilates pressure measurements to estimate flow speed, angle of attack, and foil camber. The closed-loop speed-control strategy combines an inverse-mapping feedforward controller based on an average model derived for periodic actuation of angle-of-attack and a proportional-integral feedback controller utilizing the estimated flow information. Simulation and experimental results are presented to show the effectiveness of the estimation and control strategy. The paper provides a systematic approach to distributed flow sensing for closed-loop speed control of a flexible fish robot by regulating the flapping amplitude.
Effects of Dietary Cu and Zn on the Accumulation, Oxidative Stress and the Expressions of Immune-Related Genes in the Livers of Nile Tilapia (oreochromis Niloticus).

Abstract:Excess Cu and Zn can cause many adverse effects in fish. However, few studies have addressed the effects of dietary Cu and Zn on antioxidant physiology and immunity and the underlying mechanisms in fishes. In this study, accumulation of Cu and Zn, effects on the antioxidant enzymes and the transcriptional expressions of immune-related genes were examined in the Oreochromis niloticus fed the Cu and/or Zn enriched duckweed. The results showed that the liver and intestine had the highest accumulation of Cu2+ and Zn2+ while the muscle had the lowest accumulation of these two metals. The activities of SOD, CAT, GPx and the contents of GSH, GSSG in the liver of all treatment groups were significantly decreased compared to the control group. MDA content was significantly elevated in all treatment groups after feeding for 21 days, implying lipid peroxidation in the liver. In the Cu + Zn group, the activities of SOD, GPx and the GSSG content in the liver were significantly decreased. Compared with the Zn group, the LZM activity in the Cu + Zn group was reversed after feeding for 42 days (P < 0.05). The transcriptional expressions of immune-related genes (TNF-α, INF-γ and IL-1β) in Cu, Zn, Cu + Zn groups were significantly inhibited compared with the control group after treatment for 21 days. Compared with the Cu + Zn group, the level of INF-γ transcripts was significantly reduced in the Cu and Zn group, while the TNF-α expression was elevated after treatment for 42 days. Cu and Zn had synergistic effects on the antioxidant system. Cu has greater effects than Zn on the immunity of O. niloticus. This study demonstrates that dietary Cu and Zn may pose a potential threat to the tilapia populations.
Effects of Acute Hypoxic Post-Feeding Condition on Respiratory Metabolism and Digestion in Yellow Catfish (pelteobagrus Fulvidraco)

Abstract:In this study, acute hypoxia was applied to two states of yellow catfish (Pelteobagrus fulvidraco) (non-feeding and feeding) for 48 h. We investigated the effects of acute hypoxia on the respiratory metabolism and digestive ability of yellow catfish (mean body length: 7.61 +/- 0.52 cm). The results indicated that the respiratory frequency of yellow catfish under acute hypoxic post-feeding condition was rapidly accelerated and the oxygen consumption rate was reduced. The level of succinate dehydrogenase in the gills first increased and then significantly decreased (p < 0.05) and the level of lactate dehydrogenase significantly increased (p < 0.05). When the expression level of hypoxia-inducing factor was significantly increased (p < 0.05), the downstream coding genes (hexokinase 1 (hk1), phosphofructokinase (pfkla), and pyruvate kinase M1/2a (pkma)), key molecules of glycolysis, and the activity of crucial enzymes (hexokinase, phosphofructokinase, and pyruvate kinase) increased. At the same time, the glucose level dropped and the lactic acid level increased, indicating that the metabolic mode in the gills of yellow catfish under acute hypoxic post-feeding condition gradually changed to anaerobic, and the anaerobic glycolysis process was activated to provide the required energy for the body. In addition, we found that the activity of digestive enzymes (trypsin, lipase, and amylase) and expression levels of digestive genes (trypsin, lipase, and amylase) were inhibited in the liver and intestine of yellow catfish under the acute hypoxic post-feeding condition, indicating that their digestive ability was reduced. This may adversely affect growth and development. This study analyzed the effects of acute hypoxic post-feeding exposure on the respiratory metabolism and digestive capacity of yellow catfish, providing new insights into fish physiology and sustainable fish culture.
The Combined Effect of Acute Hypoxic Stress and Feeding Status on the Metabolism of Yellow Catfish (pelteobagrus Fulvidraco)

Abstract:Metabolism is one of the most important physiological processes in fish that changes daily depending on the variability of disparate factors. Dissolved oxygen and feeding status are critical to the growth and metabolism of fish. In this study, the effects of dissolved oxygen and feeding status, as well as their interaction on the blood biochemistry and metabolism of yellow catfish (Pelteobagrus fulvidraco) were investigated using a two-factor test. At a dissolved oxygen content of 1.2 mg mL(-1), yellow catfish were exposed to hypoxia for 48 h and divided into four experimental groups: normoxia (6.5 mg L-1)-fasting group (C; Control group); normoxia-feeding group (F); hypoxia (1.2 mg L-1)-fasting group (H); and hypoxia-feeding group (FH). Analysis revealed that both hypoxia and feeding caused a short-term increase in the number of red blood cells and hemoglobin concentration in yellow catfish, thereby increasing the oxygen-carrying capacity of the blood. After feeding, the metabolic activity in yellow catfish showed a trend of rising and then falling. Following hypoxia, yellow catfish activated anaerobic glycolysis to provide the energy required by the body, and the mode of metabolism in yellow catfish gradually converted from aerobic to anaerobic. Moreover, the activity of respiratory metabolism enzyme succinate dehydrogenase (SDH) tended to decrease and then increase, while that of lactate dehydrogenase (LDH) tended to increase and then decrease, showing opposing trends. Furthermore, the liver glycogen was mobilized, blood glucose levels increased, and lactic acid accumulation occurred in the short term. Meanwhile, the hypoxia-inducible factor (hif-1 alpha) was activated, the expression of glucose metabolism-related genes (epo, hk1, pfkla, pkma, and gk) dramatically increased, and the activities of the three key enzymes involved in glycolysis (HK, PFK, and PK) were elevated for a short period of time. In conclusion, both hypoxia and feeding altered blood biochemical and metabolic responses in yellow catfish over time, and the metabolic responses were more intense in the post-feeding hypoxia group.
Acute exposure to waterborne cadmium induced oxidative stress and immunotoxicity in the brain, ovary and liver of zebrafish (Danio rerio)

Abstract:Cadmium (Cd) is an environmental contaminant that poses serious risks to aquatic organisms and their associated ecosystem. The mechanisms underlying Cd-induced oxidative stress and immunotoxicity in fish remain largely unknown. In this study, adult female zebrafish were exposed to 0 (control), 1mgL-1 Cd for 24h and 96h, and the oxidative stress and inflammatory responses induced by Cd were evaluated in the brain, liver and ovary. Reactive oxygen species (ROS), nitric oxide (NO), and malondialdehyde (MDA) increased in a time-dependent manner after treatment with Cd in the brain and liver. The increase may result from the disturbance of genes including copper and zinc superoxide dismutase (Cu/Zn-SOD), catalase (CAT), inducible nitric oxide synthase (iNOS), and ciclooxigenase-2 (COX-2) at mRNA, protein and activity levels. Although ROS, NO and MDA were not significantly affected by Cd in the ovary, the up-regulation of Cu/Zn-SOD, CAT, iNOS, and COX-2 was observed. Exposure to Cd induced a sharp increase in the protein levels of tumor necrosis factor alpha (TNF-α) in the brain, liver and ovary, possibly contributing to activate inflammatory responses. Furthermore, we also found a dramatic increase in mRNA levels of NF-E2-related factor 2 (Nrf2) and nuclear transcription factor κB (NF-κB) at 24h in the liver and ovary. The corresponding changes in the mRNA levels of Kelch-like-ECH-associated protein 1 (Keap1a and Keap1b) and the inhibitor of κBα (IκBαa and IκBαb) may contribute to regulate the transcriptional activity of Nrf2 and NF-κB, respectively. Contrarily, mRNA levels of Nrf2, NF-κB, Keap1, Keap1b, IκBαa and IκBαb remained stable at 24 and 96h in the brain. Taken together, we demonstrated Cd-induced oxidative stress and immunotoxicity in fish, possibly through transcriptional regulation of Nrf2 and NF-κB and gene modifications at transcriptional, translational, post-translational levels, which would greatly extend our understanding on the Cd toxicity.

Abstract:Aquaculture environments frequently experience hypoxia and subsequent reoxygenation conditions, which have significant effects on hypoxia-sensitive fish populations. In this study, hepatic biochemical activity indices in serum and the content of major neurotransmitters in the brain were altered markedly after acute hypoxia and reoxygenation exposure in silver carp (Hypophthalmichthys molitrix). Proteomics analysis of the liver showed that a number of immune-related and cytoskeletal organization-related proteins were downregulated, the ferroptosis pathway was activated, and several antioxidant molecules and detoxifying enzymes were upregulated. Proteomics analysis of the brain showed that somatostatin-1A (SST1A) was upregulated, dopamine-degrading enzyme catechol O methyltransferase (COMT) and ferritin, heavy subunit (FerH) were downregulated, and the levels of proteins involved in the nervous system were changed in different ways. In conclusion, these findings highlight that hypoxia–reoxygenation has potential adverse effects on growth, locomotion, immunity, and reproduction of silver carp, and represents a serious threat to liver and brain function, possibly via ferroptosis, oxidative stress, and cytoskeleton destruction in the liver, and abnormal expression of susceptibility genes for neurodegenerative disorders in the brain. Our present findings provide clues to the mechanisms of hypoxia and reoxygenation damage in the brain and liver of hypoxia-sensitive fish. They could also be used to develop methods to reduce hypoxia or reoxygenation injury to fish.
