Abstract:Machine learning methodology with neural network models was developed using the datasets based on the overlay sampling uniform design (OSUD) for optimization of phenyllactic acid biosynthesis and separation processes by cryogels. Compared with the multiple regression, the machine learning models exhibited a significant improvement of predictive accuracy of phenyllactic acid biosynthesis, in which the radial basis function neural network (RBFNN) model had the best predictive performance with the accuracy increased by 65.2%. The combination of RBFNN and OSUD was further employed to optimize the chromatographic separation of phenyllactic acid from crude fermentation broth using two poly(hydroxyethyl methacrylate) based anion-exchange cryogel packed-beds (grafted with (vinylbenzyl)trimethylammonium chloride and N,N-dimethylaminoethyl methacrylate). After optimizing the three critical separation parameters: sample volume (5.3-31.8mL), flow velocity (1.0-6.0cm/min), and elution salt concentration (0.05-0.3mol/L), it was found that the models provided excellent predictions. The optimized recovery rates for the two packed-beds were determined to be 76.5% and 83.0%, and the optimal adsorption capacities were 0.26mg/mL and 0.39mg/mL from the fermentation broth, respectively. This study provides a reliable integrated approach for optimizing the synthesis and separation processes of high-value bioproducts like phenyllactic acid from crude feedstocks.

Abstract:The production of 2-keto-L-gulonic acid (2-KGA) during the conversion from L-sorbose to 2-KGA in the two-step fermentation of vitamin C can be improved by using an efficient companion strain Bacillus subtilis A9 to facilitate the growth of Ketogulonicigenium vulgare and the production of 2-KGA. Two optimization models, namely response surface methodology (RSM) and artificial neural network (ANN), were built to optimize the medium components for mixed-culture fermentation of 2-KGA. The root mean square error, R2 and the standard error of prediction given by the ANN model were 0.13%, 0.99% and 0.21%, respectively, while the RSM model gave 1.89%, 0.84% and 2.9%, respectively. This indicated that the fitness and the prediction accuracy of the ANN model were higher than those of the RSM model. Furthermore, using genetic algorithm (GA), the input space of the ANN model was optimized, predicting that the maximum 2-KGA production of 72.54 g·L−1 would be obtained at the GA-optimized concentrations of the medium components (L-sorbose, 92.5 g·L−1; urea, 10.2 g·L−1; corn steep liquor, 16 g·L−1; CaCO3, 3.96 g·L−1; MgSO4, 0.28 g·L−1). The 2-KGA production experimentally obtained using the ANN–GA-designed medium was 71.21 ± 1.53 g·L−1, which was in good agreement with the predicted value. The same optimization process may be used to improve the production during bacterial mixed-cultures fermentation by changing the fermentation parameters.

Abstract:A five-level-three factor central composite rotary design was employed to find out the interactive effects of three variables, i.e. static magnetic field intensity, concentration of NH3–N and initial pH on polyhydroxyalkanoates (PHA) production by activated sludge under the aerobic dynamic feeding (ADF) technique. Response surface methodology (RSM) was utilized for process optimization and a second-order polynomial equation was obtained by multiple regression analysis. A yield of 49.5% of dry cell weight (dcw) was achieved at optimized conditions, i.e. magnetic field 11mT, NH3–N 4.8mgl−1 (C:N=60:1)and initial pH 9.0.

Abstract:The optimization of the batch size experiment was run for a hydraulic retention time of 45 days using proteolytic enzyme pretreatment. The highest amounts of biogas were produced in comparison to conventional BDS (25:75), which is not processed with enzymes, and there was an increase in the biogas generation of 13.9 and 18.57%. The kinetic models show the goodness of fit between 0.993 and 0.998 and the correlation coefficient's value domain was [-1, 1] from a statistical perspective. The Box-Behnken design was carried out using the response surface methodology at different levels of independent parameters to optimize the process. Different instruments were evaluated to determine the chemical structure change and the contamination of the different treatments and the raw sample of tannery fleshings was determined. Thermogravimetric analysis was conducted to determine the loss of weight on thermal degradation. The Fourier transform infrared spectrometry was carried out to determine the different functional groups, such as -OH, -CH, -NH, and C-O, present in the samples of tannery fleshings. Scanning electron microscopy and energy dispersive X-ray analysis were carried out to determine the morphological alterations in the substrate, digestate, enzyme-pretreated fleshings, and the chemical composition of samples.
Electromechanics of fluidized beds of nanoparticles.

Abstract:The electromechanical behavior of a gas-fluidized bed of insulating silica nanoparticles is investigated. When fluidized by gas, these nanoparticles form highly porous agglomerates of size of the order of hundreds of microns, which gives rise to a nonbubbling fluidization regime. Bed expansion is enhanced by an imposed alternating electric field for oscillation frequencies in the range between tens and hundreds of hertzs and field strengths of about 1 kV/cm . Nanoparticle agglomerates are naturally charged and experience forced oscillations that cause an increase of the gas flow shear on their surface. As a consequence, the agglomerate size is expected to decrease, which can explain the observed behavior. A model based on the balance between attractive and flow shear forces is presented that accounts for agglomerate size reduction as the strength of the field is increased.

Abstract:The biotransformation of L-sodium glutamate (L-MSG) to gamma-aminobutyric acid (GABA) catalyzed by the cells of Lactobacillus brevis with higher glutamate decarboxylase activity was investigated. The results showed that pH, temperature, and FeSO4 (.) 7H(2)O concentration had significantly positive effect on GABA yield. The individual and interactive effects of pH, temperature, and FeSO4 (.) 7H(2)O concentration were further optimized in terms of GABA yield. In the present work; an artificial neural network (ANN) and response surface methodology (RSM) models were developed, which incorporated pH, temperature, and FeSO4 (.) 7H(2)O concentration as input variables, and GAGA yield as output variable. The optimized ANN topology included four neurons in the hidden layer and the best network architecture was 3-4-1. The trained ANN gave total root-mean square error (sigma) equal to 1.84 for GABA yield while the RSM gave a equal to 2.63. The results demonstrated a slightly higher prediction accuracy of ANN compared to RSM. The modeled maximum GABA yield was identified by applying particle swarm optimization algorithm to the ANN model developed. The modeled maximum GABA yield reached 91 mM under the following optimal conditions: 25 ml, Na2HPO4-citric acid buffer (100 mM, pH 4.23), 120 MM L-MSG, 0.83 g/L FeSO4 (.) 7H(2)O, 10 mu M PLP, the resting cells obtained from a 60-h culture broth, 2.68 g dry cell weight (DCW)/L, and without agitation at 40 degrees C for 5 h. The previous high value of GABA yield that was observed was 81.8 mM. The optimized conditions allowed GABA yield to be increased from 81.8 to 90.57 mM after verification experiments test.

Abstract:In this research, an industrially important enzyme tannase and product gallic acid was produced with an inexpensive novel substrate Swietenia macrophylla . Fermentation of S. macrophylla was optimized using a two-step approach: First, the traditional One variable at-a-time technique, and second, the statistical Box-Behnken design for co-production of tannase enzyme and gallic acid. This two-step method of optimization showed the highest tannase activity and gallic acid yield of 0.0497 U/mL and 225 μg/mL respectively which is a 29.5 and 49-fold increase when compared to unoptimized conditions. Further, the partially purified tannase enzyme was characterized and showed optimal tannase activity at pH 4.0 and 30 °C, and was stable between pH 3.0–6.0 and 4−40 °C for 24 h and 10 h, respectively. Also, metal ions such as Ca 2+ , Na + , and K + at 1 mM concentration; and organic solvents methanol, and isoamyl alcohol at 20% v/v exhibited the highest activity at optimized reaction conditions. Whereas, Mn 2+ , Zn 2+ , Mg 2+ , Fe 2+ and Fe 3+ , EDTA, TritonX 100, toluene, and hexane caused the tannase inhibition at higher concentrations. In the end, the fermentative production of gallic acid was verified qualitatively through thin-layer chromatography and Fourier transform infrared spectroscopy.

Abstract:A consecutive optimization based on statistical approach was applied for a-glucosidase production by both wild and mutant T. dupontii. Plackett Burman design (PBD) with two levels was employed in order to screen the significant effect of different nutritional and physical parameters through submerged fermentation. Among all nine variables tested in PBD, incubation time, inoculum size and ammonium sulphate concentration were selected. The Box-Behnken approach was further applied for process optimization. The a-glucosidase production for both wild and mutant T.dupontii was obtained at 72 h of incubation, 1.25 mL inoculum size and 0.25% ammonium sulphate concentration with relatively 95% correlation between the experimentally predicted and observed values. The duration of maximum enzyme production in RSM was cost-saving and fast. The quadratic model was in satisfactory adjustment with the experimental data with high R2 value which describes 98.90% of response variability of the model. Moreover, the novel approach of this present work is that, consecutive optimization were applied for maximum a-glucosidase production using response surface methodology by both wild and mutant thermophillic T. dupontii. Results revealed that thermophillic mutant T. dupontii could be potential candidate for industrial applications.
Optimized phenol degradation and lipid production by Rhodosporidium toruloides using response surface methodology and genetic algorithm-optimized artificial neural network

Abstract:Oleaginous yeast can produce lipids while degrading phenol in wastewater treatment. In this study, a Plackett-Burman Design (PBD) was adopted to identify key factors of phenol degradation and lipid production using R toruloides 9564T. While temperature, inoculum size, and agitation were significant for both the processes (p < 0.05), pH and incubation were significant for lipid production, and phenol removal, respectively. Results from four factors (pH, temperature, inoculum size, and incubation period) central composite design (CCD) experiment were used to formulate quadratic and genetic algorithm-optimized ANN models. The reduced quadratic model for phenol degradation (R2: 0.993) and lipid production (R2: 0.958) were marginally inferior to ANN models (R2: 0.999, 0.982, respectively) on training sets. Multi-objective optimization with equal importance suggests phenol degradation between 106.4 and 108.76%, and lipid production of 0.864-0.903 g/L, by polynomial and ANN models. Complete phenol degradation (100%) and 3.35-fold increment (0.918 g/L) in lipid production were obtained at pH 6.07, inoculum size 14.68% v/v, at 29.5 °C in 92.17 h experimentally.

Abstract:In this study, a thermo-tolerant isolated Bacillus licheniformis WF67 has produced considerable levels from the extracellular α-amylase under submerged fermentation conditions. To maximize the α-amylase production, both traditional "one factor per time" and statistical optimization were performed. The "one factor per time" optimization technique declared the most effective parameters in the process of α-amylase production. Those parameters were cultivation medium pH value, incubation time, soluble starch and beef extract as carbon and nitrogen sources. Response surface methodology (RSM) was employed to get the effective values of those most efficient parameters via different interaction ratios between all of them. Central composite design (CCD) was based on the following values: medium pH (pH 3.0-9.0), incubation time (15-75 h), soluble starch (1.5-19.5 g/L), and beef extract (1.0-13 g/L). Results obtained from CCD approach revealed that the incubation of B. licheniformis WF67 for 50h, at pH value of 6.33, using the specified medium containing 12.55 g/L soluble starch, and 8.31 g/L beef extract were the optimum values for α-amylase production. Under the previously mentioned conditions α-amylase productivity was improved by about 90% (6.633 U/mL) in comparison to the traditional "one factor per time" optimization results (3.486 U/mL).

Expression of biologically active bovine luteinizing hormone in Chinese hamster ovary cells.

Abstract:Biologically active bovine luteinizing hormone (LH) has been obtained through expression of the alpha- and LH beta-subunit genes in stably transformed clones of DUXB11, a Chinese hamster ovary cell line deficient in dihydrofolate reductase (DHFR). Expression of alpha-and LH beta-subunit mRNAs of the expected sizes (approximately 910 and 770 nucleotides, respectively) were revealed by blot analysis after electrophoresis of total cellular RNA. Furthermore, presence or absence of the gonadotropin mRNAs in several clonal lines was directly correlated with the appearance of one or both bovine LH subunits in the culture medium. Media from three clones secreting significant immunoreactive levels of both subunits also stimulated the release of progesterone in ovine luteal cells, suggesting that the secreted LH was assembled into a biologically active and glycosylated dimer. Immunoprecipitation and NaDodSO4/PAGE of [35S]methionine-labeled proteins secreted from one of the clones, CHODLH20, further confirmed the presence of an alpha/beta dimer with apparent subunit molecular weights of 20,500 and 16,000, only slightly higher than those of pituitary alpha and LH beta subunits.

Abstract:The extraction of phytocompounds from Achocha ( Cyclanthera pedata ) vegetable juice using traditional methods often results in suboptimal yields and efficiency. This study aimed to enhance the extraction process through the application of thermosonication (TS). To achieve this, an artificial neural network (ANN) and a genetic algorithm (GA) were utilized to simulate and optimize the process parameters. The study investigated the influence of ultrasonic amplitude (30%–50%), temperature (30 °C–50 °C), and sonication duration (15–60 min) on total polyphenolic content (TPC), total flavonoid content (TFC), antioxidant activity (AOA), and ascorbic acid content (AA). Remarkably, the ANN-GA optimization resulted in optimal TS conditions: an ultrasonic amplitude of 40%, a temperature of 40 °C, and a sonication duration of 30 min. Subsequent analysis of extraction kinetics and thermodynamics across various temperatures (30 °C–50 °C) and extraction times (0–30 min) demonstrated R 2 (0.98821) and χ 2 (1.74773) for TPC with activation energy (E a ) 26.0456, R 2 (0.99906) and χ 2 (0.07215) for TFC with E a 26.2336, R 2 (0.99867) and χ 2 (0.03003) for AOA with E a 26.0987, R 2 (0.99731) and χ 2 (0.13719) for AA with E a 26.0984, validating the pseudo second-order kinetic model. These findings indicate that increased temperature enhances the saturation concentration and rate constant of phytochemical extraction.

Loki, Io: New ground‐based observations and a model describing the change from periodic overturn

Abstract:Loki Patera is the most powerful volcano in the solar system. We have obtained measurements of Loki's 3.5 micron brightness from NASA's Infrared Telescope Facility (IRTF) and have witnessed a major change in eruptive behavior. While Loki brightened by a factor of several every 540 days prior to 2001, from 2001 through 2004 Loki remained at a constant, intermediate brightness. We have constructed a quantitative model of Loki as a basaltic lava lake whose solidified crust overturns when it becomes buoyantly unstable. By altering the speed at which the overturn propagates across the patera, we can match our ground‐based brightness data. In addition, we can match other data taken at other times and wavelengths. By slowing the propagation speed dramatically, we can match the observations from 2001–2004. Such slowing could be due to a small change in volatile content in the lava.
Modeling and optimization of spray drying process parameters using artificial neural network and genetic algorithm for the production of probiotic (Lactiplantibacillus plantarum) finger millet milk powder

Abstract:Modeling and optimization of spray drying process and quality parameters using ANN‐GA and RSM‐DF methods for production of spray‐dried probiotic finger millet milk powder. Consumers have been more inclined towards functional food due to various health issues. Non‐dairy‐based probiotic milk could be the potential alternative to fulfill consumer demand. Spray drying of functional health drinks could enhance the shelf life and reduce the transportation cost of developed food powder. The current study was focused on the modeling and optimization of spray‐drying process parameters using an artificial neural network (ANN) coupled with a genetic algorithm (GA). The data so obtained using ANN‐GA was also compared with the data obtained using response surface methodology (RSM) coupled with desirability function (DF). The results indicated that the ANN model was better at predicting the response parameters compared to the RSM model with a higher correlation coefficient (R) of .9997, .9994, .9964, and .9992 for training, testing, validation, and all datasets, respectively. The optimum conditions obtained using RSM‐DF were 160.41°C of inlet air temperature, 33.77% of maltodextrin content, and 138.79 mL/h of feed rate while that for ANN‐GA were 160.87°C, 20%, and 200 mL/h. The RSM‐DF method proved to be better for the optimization of response parameters. Therefore method selection for modeling and optimization of process and response parameters must be based on fulfilling the specific criteria. Practical Applications Non‐dairy milk production has gained popularity in the area of research and product development. Various process protocols have been reported to produce high‐quality non‐dairy milk to fulfill the demand for a vegan diet. However, nutrient bioavailability, longer shelf life, product stability, and consumer acceptability are hard to obtain from cereal‐based non‐dairy milk. The current study contributes to produce spray‐dried milk powder followed by fermentation at optimum drying conditions with good quality and stability. The process has been modeled and optimized in the study using advanced statistical tools such as ANN‐GA and RSM‐DF. They can effectively predict the quality parameters and determine the optimal conditions for new experimental data.

Abstract:An integrated approach involving response surface methodology (RSM) and artificial neural network-ant-colony hybrid optimization (ANN-ACO) was adopted to develop a bioprocess medium to increase the yield of Bacillus cereus neutral protease under submerged fermentation conditions. The ANN-ACO model was comparatively superior (predicted r 2 = 98.5%, mean squared error [MSE] = 0.0353) to RSM model (predicted r 2 = 86.4%, MSE = 23.85) in predictive capability arising from its low performance error. The hybrid model recommended a medium containing (gL −1 ) molasses 45.00, urea 9.81, casein 25.45, Ca 2+ 1.23, Zn 2+ 0.021, Mn 2+ 0.020, and 4.45% (vv −1 ) inoculum, for a 6.75-fold increase in protease activity from a baseline of 76.63 UmL −1 . Yield was further increased in a 5-L bioreactor to a final volumetric productivity of 3.472 mg(Lh) −1 . The 10.0-fold purified 46.6-kDa-enzyme had maximum activity at pH 6.5, 45–55 °C, with K m of 6.92 mM, V max of 769.23 μmolmL −1 min −1 , k cat of 28.49 s −1 , and k cat /K m of 4.117 × 103 M −1 s −1 , at 45 °C, pH 6.5. The enzyme was stabilized by Ca 2+ , activated by Zn 2+ but inhibited by EDTA suggesting that it was a metallo-protease. The biomolecule significantly clarified orange and pineapple juices indicating its food industry application.

Abstract:Bael fruit pulp was treated with pectinase enzyme for the easy extraction and high recovery of juice with nutritive properties. The response surface methodology (RSM) and artificial neural network (ANN) modeling were carried out on runs of Box-Behnken design of three variables for the optimization process and identification of the best modeling method. Pectinase concentration of 0.22 g/100 g of pulp, the temperature of 46.20°C, and time of 6.35 hours was found out to be RSM optimized value of variables whereas pectinase concentration of 0.18 g/100 g of pulp, the temperature of 46.81°C and time of 6.09 hours was obtained to be ANN-GA optimized value of variables. From the values of the coefficient of determination (R2), root mean squared error (RMSE), and mean absolute error (MAE), ANN was found out to be better than RSM. This research would help in the commercial production of bael fruit juice on an industrial scale.

Abstract:The applicability of the statistical tools coupled with artificial intelligence techniques was tested to optimize the critical medium components for the production of extracellular cholesterol oxidase (COD; an enzyme of commercial interest) from Streptomyces rimosus MTCC 10792. The initial medium component screening was performed using Placket-Burman design with yeast extract, dextrose, starch and ammonium carbonate as significant factors. Response surface methodology (RSM) was attempted to develop a statistical model with a significant coefficient of determination (R2 = 0.89847), followed by model optimization using Genetic Algorithm (GA). RSM-GA based optimization approach predicted that the combination of yeast extract, dextrose, starch and ammonium carbonate at concentrations 0.99, 0.8, 0.1, and 0.05 g/100 ml respectively, has resulted in 3.6 folds increase in COD production (5.41 U/ml) in comparison with the un-optimized medium (1.5 U/ml). COD was purified 10.34 folds having specific activity of 12.37 U/mg with molecular mass of 54 kDa. The enzyme was stable at pH 7.0 and 40 °C temperature. The apparent Michaelis constant (Km) and Vmax values of COD were 0.043 mM and 2.21 μmol/min/mg, respectively. This is the first communication reporting RSM-GA based medium optimization, purification and characterization of COD by S. rimosus isolated from the forest soil of eastern India.

Optimization of tannase production by Aspergillus glaucus in solid-state fermentation of black tea waste

Abstract:Tannases are valuable industrial enzymes used in food, pharmaceutical, cosmetic, leather manufacture and in environmental biotechnology. In this study, 15 fungal isolates were obtained from Egyptian cultivated soil and marine samples. The isolated fungi were qualitatively and quantitatively screened for their abilities to produce tannase. The selected fungal isolate NRC8 giving highest tannase activity was identified by molecular technique (18S rRNA) as Aspergillus glaucus. Among different tannin-containing wastes tested, the black tea waste was the best substrate for tannase production by Aspergillus glaucus in solid-state fermentation (SSF). Optimization of the different process parameters required for maximum enzyme production was carried out to design a suitable SSF process. Maximal tannase production was achieved with moisture content of 75%, an inoculums size of 6 × 108 spore/ml and sodium nitrate 0.2% (pH of 5.0) at 30 °C after 5 days of incubation. Box-Behnken experiment was designed to get a quadratic model for further optimization studies. Four-factor response-surface method with 27 runs was prepared using independent parameters including (moisture content %, initial pH, substrate concentration (g) and sodium nitrate concentration (g) for tannase model. The F- and P-values of the model were 4.30 and 0.002, respectively, which implied that the model is significant. In addition, the lack-of-fit was 1040.37 which indicates the same significance relative to the pure error. A. glaucus tannase was evaluated by the efficiency of conversion of tannic acid to gallic acid. Moreover, production of gallic acid from SSF process of A. glaucus using black tea waste was found to be 38.27 mg/ml. The best bioconversion efficiency was achieved at 40 °C with tannic acid concentration up to 200 g/L.

Abstract:4′-N-demethyl-vicenistatin is a vicenistatin analogue that has better antitumor activity with promising applications in the pharmaceuticals industry. The harnessing of the complete potential of this compound necessitates a systematic optimization of the culture medium to enable the cost-effective production of 4′-N-demethyl-vicenistatin by Streptomyces parvus SCSIO Mla-L010/ΔvicG. Therefore, in this study, a sequential approach was employed to screen the significant medium compositions, as follows: one-factor-at-a-time (OFAT) and Plackett–Burman designs (PBD) were initially utilized. Cassava starch, glycerol, and seawater salt were identified as the pivotal components influencing 4′-N-demethyl-vicenistatin production. To further investigate the direct and interactive effects of these key components, a three-factor, five-level central composite design (CCD) was implemented. Finally, response surface methodology (RSM) and an artificial-neural-network-genetic-algorithm (ANN-GA) were employed for the modeling and optimization of the medium components to enhance efficient 4′-N-demethyl-vicenistatin production. The ANN-GA model showed superior reliability, achieving the most 4′-N-demethyl-vicenistatin, at 0.1921 g/L, which was 17% and 283% higher than the RSM-optimized and initial medium approaches, respectively. This study represents pioneering work on statistically guided optimization strategies for enhancing 4′-N-demethyl-vicenistatin production through medium optimization.
