Abstract:Changes in the structural properties of the skin due to collagen alterations are an important factor in diabetic skin complications. Using a combination of photonic methods as an optic diagnostic tool, we investigated the structural alteration in rat dermal collagen I in diabetes, and after short-term l-arginine treatment. The multiplex approach shows that in the early phase of diabetes, collagen fibers are partially damaged, resulting in the heterogeneity of fibers, e.g., “patchy patterns” of highly ordered/disordered fibers, while l-arginine treatment counteracts to some extent the conformational changes in collagen-induced by diabetes and mitigates the damage. Raman spectroscopy shows intense collagen conformational changes via amides I and II in diabetes, suggesting that diabetes-induced structural changes in collagen originate predominantly from individual collagen molecules rather than supramolecular structures. There is a clear increase in the amounts of newly synthesized proline and hydroxyproline after treatment with l-arginine, reflecting the changed collagen content. This suggests that it might be useful for treating and stopping collagen damage early on in diabetic skin. Our results demonstrate that l-arginine attenuates the early collagen I alteration caused by diabetes and that it could be used to treat and prevent collagen damage in diabetic skin at a very early stage.
Marine Collagen Peptides Reduce Endothelial Cell Injury in Diabetic Rats by Inhibiting Apoptosis and the Expression of Coupling Factor 6 and Microparticles

Abstract:The present study aimed to elucidate the role of marine collagen peptides (MCPs) in protection of carotid artery vascular endothelial cells (CAVECs) in type 2 diabetes mellitus (T2DM), and the mechanism underlying this process. In an in vivo experiment, diabetic Wistar rats were divided randomly into four groups (n=10/group): Diabetes control, and three diabetes groups administered low, medium and high doses of MCPs (2.25, 4.5 and 9.0 g/kg body weight/day, respectively). Another 10 healthy rats served as the control. In an in vitro experiment, human umbilical-vein endothelial cells (HUVECs) were incubated in normal and high concentrations of glucose with or without MCPs (3.0, 15.0 and 30.0 mg/ml, respectively) for 24, 48 or 72 h. Blood vessel/endothelial construction, inflammatory exudation and associated molecular biomarkers in CAVECs were detected and analyzed. The results of the present study demonstrated that in rats, MCP treatment for 4 weeks significantly lowered blood glucose and attenuated endothelial thinning and inflammatory exudation in carotid-artery vascular endothelial cells. In vitro, the high-glucose intervention significantly increased cell apoptosis in HUVECs, and medium and high doses of MCPs (4.5 and 9.0 g/kg body weight/day, respectively) partially ameliorated this high glucose-mediated apoptosis and decreased levels of apoptosis biomarkers. In conclusion, a moderate oral MCP dose (≥4.5 g/kg body weight/day) may be a novel therapeutic tool to protect against early cardiovascular complications associated with T2DM by inhibiting apoptosis and reducing the expression of coupling factor 6 and microparticles.

Abstract:Oxidative stress and abnormal glucose metabolism are the important physiological mechanisms in the occurrence and development of diabetes. Antioxidant peptides have been reported to attenuate diabetes complications by regulating levels of oxidative stress, but few studies have focused on peptides from marine bone collagen. In this study, we prepared the peptides with a molecular weight of less than 1 kD (HNCP) by enzymolysis and ultrafiltration derived from Harpadon nehereus bone collagen. Furthermore, the effects of HNCP on blood glucose, blood lipid, liver structure and function, oxidative stress, and glucose metabolism were studied using HE staining, kit detection, and Western blotting experiment in streptozocin-induced type 1 diabetes mice. After the 240 mg/kg HNCP treatment, the levels of blood glucose, triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) in streptozotocin-induced diabetes mice decreased by 32.8%, 42.2%, and 43.2%, respectively, while the levels of serum insulin and hepatic glycogen increased by 142.0% and 96.4%, respectively. The antioxidant enzymes levels and liver function in the diabetic mice were markedly improved after HNCP intervention. In addition, the levels of nuclear factor E2-related factor 2 (Nrf2), glucokinase (GK), and phosphorylation of glycogen synthase kinase-3 (p-GSK3β) in the liver were markedly up-regulated after HNCP treatment, but the glucose-6-phosphatase (G6Pase) and phosphoenolpyruvate carboxykinase1 (PEPCK1) were down-regulated. In conclusion, HNCP could attenuate oxidative stress, reduce blood glucose, and improve glycolipid metabolism in streptozocin-induced type 1 diabetes mice.

Abstract:Diabetic wounds require a multifactorial approach because several factors are involved in its occurrence. Herein we investigated whether transplantation of hyaluronic acid (HA) in combination with menstrual blood derived stem cells (MenSCs) could promote healing in diabetic rats. Thirty days after induction of diabetes, sixty animals were randomly planned into four equal groups: the untreated group, HA group, MenSC group, and HA+MenSC group. Sampling was done for histological, molecular, and tensiometrical assessments. Our results indicated that the wound contraction rate, volumes of new epidermis and dermis, collagen density, as well as tensiometrical parameter were considerably increased in the treatment groups compared to the untreated group and these changes were more obvious in the HA+MenSC ones. In addition, the expression levels of TGF-β and VEGF genes were significantly upregulated in treatment groups in comparison with the untreated group and were greater in the HA+MenSC group. This is while expression levels of TNF-α and IL-1β genes were more considerably downregulated in the HA+MenSC group than the other groups. We concluded that the combined use of HA and MenSCs has more effects on diabetic wound healing.

Abstract:Chronic diabetic wounds have become a major healthcare challenge worldwide. Improper treatment may lead to serious complications. Current treatment methods including biological and physical methods and skin grafting have limitations and disadvantages, such as poor efficacy, inconvenience of use, and high cost. Therefore, developing a more effective and feasible treatment is of great significance for the repair of chronic diabetic wounds. Hydrogels can be designed to serve multiple functions to promote the repair of chronic diabetic wounds. Furthermore, 3D bioprinting enables hydrogel customization to fit chronic diabetic wounds, thus facilitating the healing process. This paper reports a study of 3D printing of a collagen-hyaluronic acid composite hydrogels with application for chronic diabetic wound repair. In situ printed hydrogels were developed by a macromolecular crosslinking network using methacrylated recombinant human collagen (RHCMA) and methacrylated hyaluronic acid (HAMA), both of which can respond to ultraviolet (UV) irradiation. The hydrogels were also loaded with silver nanoclusters (AgNCs) with ultra-small-size nanoparticles, which have the advantages of deep penetration ability and broad-spectrum high-efficiency antibacterial properties. The results of this study show that the developed RHCMA, HAMA, and AgNCs (RHAg) composite hydrogels present good UV responsiveness, porosity, mechanical properties, printability, and biocompatibility, all of which are beneficial to wound healing. The results of this study further show that the developed RHAg hydrogels not only effectively inhibited Staphylococcus aureus and Pseudomonas aeruginosa but also promoted the proliferation and migration of fibroblasts in vitro and tissue regeneration and collagen deposition in vivo, thus producing a desirable wound repair effect and can be used as an effective functional biomaterial to promote chronic diabetic wound repair.
Biocompatible zinc oxide nanocrystals stabilized via hydroxyethyl cellulose for mitigation of diabetic complications

Abstract:The vascular complications of diabetes are the most serious manifestations of the disease. The hyperglycemia can directly promote an inflammatory state where the increase C-reactive (CRP) and cytokines, such as interleukins (IL-1 and IL-6), which contribute to the development of cardiovascular diseases. The current study was aimed to evaluate the role of environmentally-synthesized zinc oxide nanocrystals (ZnO-NPs) in augmentation of hyperglycemia and its complications, as well as the preservation of asymmetrical dimethylarginine (ADMA) level as a specific marker for endothelial dysfunction in streptozotocin (STZ)-induced diabetic rats. ZnO-NPs was chemically-synthesized using environmental benign biodegradable hydroxyl ethyl cellulose (HES) as both a stabilizing and directing agent in the presence of potassium hydroxide. HES is a biomaterial compound used in many biomedical applications due to its biodegradability and biocompatibility in nature. Particle size, morphological structure, purity, and crystallinity of the as-prepared ZnO-NPs were evaluated through different techniques, such as transmission electron microscopy (TEM), X-ray diffraction (XRD), and scanning electron microscopy connected to energy-dispersive X-ray spectra (SEM-EDS). Sixty male albino rats were used in this study and divided into four groups: control, ZnO-NPs, diabetic and treated groups; after the experimental period, CRP and interleukin-1 (IL-1α) were determined by ELISA. ADMA was estimated by RP-HPLC using a fluorescence detector. The results obtained indicate that CRP, IL-1α, and ADMA levels increased significantly concomitant with a reduction in NO level in the diabetic group, whereas ZnO-NPs supplementation significantly attenuated these parameters. Based on these encouraging results, the reported approach of environmental synthesis and application has the potential of leading to a new generation of nanometerials for treatment of diabetic complications with considerably enhanced selectively towards atherosclerosis.
Effect of marine collagen peptide on the expression of overoxidation stress markers in rats with type 2 diabetes

Abstract:Background: Marine collagen peptide is a kind of oligopeptide powder at the relative molecular mass of less than 1 000, comprising 2-6 amino acids. Its antioxidation property has been proved in vitro. Objective: To study the effect of marine collagen peptide on the expression of overoxidation stress markers in the blood serum of rats with type 2 diabetes. Design, time and setting: A randomized control animal study was carried out from October 2006 to June 2007 at the Laboratory of Animal Experiment in Shenzhen Center for Disease Control and Prevention (Shenzhen, Guangdong, China). Materials: Marine collagen peptide was offered by the China National Research Institute of Food and Fermentation Industry. Thirty-six male SD rats were assigned into 6 controls and 30 models. Normal control group fed with basal diet, while 26 of other 30 rats were successfully induced the model of type 2 diabetes by intraperitoneal injection of streptozotocin. Methods: Twenty-four rats with type 2 diabetes were randomly divided into four groups, namely marine collagen peptide low dose group (0.75 g/ kg ·d), medium dose group (1.5 g/kg ·d), high dose group (3.0 g/kg ·d) and diabetes control group. They were all processed into intragastric administration of tap water for 4 weeks. Main outcome measures: Blood glucose of rats before intervention and at four weeks after intervention. Blood level of superoxide dismutase (SOD), malondialdehyde (MDA), reduced glutathione hormone (GSH) and NO of rats at four weeks after intervention. Results: There were no significant differences among three marine collagen peptide groups on the blood glucose (P > 0.05). Compared to normal control group, the levels of SOD and GSH of all groups were significantly lowered, while the levels of MDA and NO were significantly enhanced (P < 0.01, 0.05); Compared to the diabetes control group, the SOD and GSH levels of marine collagen peptide middle and high dose groups were significantly increased, while the MDA and NO levels were significantly lowered (P < 0.01, 0.05). Conclusion: Higher dosage of marine collagen peptide (≥ 1.5 g/kg ·d) may have a regulating effect on the expression of overoxidation markers in rats with type 2 diabetes. The exact effect of marine collagen peptide on fasting blood glucose deserves further study of large scale samples and intervention time.
Homocysteine and Asymmetrical Dimethylarginine in Diabetic Rats Treated with Docosahexaenoic Acid–Loaded Zinc Oxide Nanoparticles

Abstract:Hyperglycemia, the hallmark of diabetes mellitus, is considered one of the endothelial dysfunction risk factors, the main reason of vascular complication. In this study, we aimed to evaluate homocysteine (Hcy) and asymmetrical dimethylarginine (ADMA) levels in diabetic rats and the possibility to attenuate the elevation of these two parameters by supplementation of docosahexaenoic acid (DHA) alone or loaded zinc oxide nanoparticles (ZnONPs) to improve endothelial dysfunction in streptozotocin (STZ)-induced diabetic rats. Forty male albino rats weighing 180–200 g were classified as control, diabetic, diabetic treated with DHA, and diabetic treated with DHA-loaded zinc oxide nanoparticles (DHA/ZnONPs) groups. Fasting blood glucose, insulin, ADMA, Hcy, and nitric oxide (NO) were estimated. Fatty acids (linoleic acid (LA), arachidonic acid (AA), DHA, α-linolenic acid (ALA), and oleic acid (OA)) were also evaluated by reversed phase HPLC using a UV detector. The results showed that fasting blood sugar, insulin resistance, LA, AA, OA, ADMA, and Hcy increased significantly in diabetic rats compared with control while fasting insulin, DHA, ALA, and NO decreased significantly in diabetic rats. In both treated groups, fasting blood sugar, insulin resistance, LA, AA, OA, ADMA, and Hcy significantly decreased as compared with the diabetic group while fasting insulin, DHA, ALA, and NO were significantly increased. In conclusion, DHA and DHA/ZnONP supplementation protect against diabetic complications and improve endothelial dysfunction as well as hyperhomocysteinemia in diabetes. DHA/ZnONP-treated group appeared more efficient than DHA alone.

Effects of Marine Collagen Peptides on Blood Glucose and Lipid Metabolism in Hyperinsulinemic Rats

Abstract:OBJECTIVE:To observe the effects of marine collagen peptides (MCPs) on blood glucose and lipid metabolism in hyperinsulinemic rats.METHODS:Male SD rats fed with high fat diet for 4 months were used to establish a hyperinsulinemic model. Thirty two hyperinsulinemic rats were divided into four groups according to body weight and serum insulin level, namely hyperinsulinemic control (HIC) group and 3 Marine Collagen Peptides treated groups with 8 rats in each group. Another 8 healthy male SD rats fed with basic diet were used as negative control (NC). Rats in MCPs groups were treated by intragastric way with MCPs at the dose of 0.225 g/kg bw, 0.45 g/kg bw and 1.35 g/kg bw respectively for 8 weeks, while rats in HIC and NC groups were treated with tap water in the same manner. During the test period, rats in HIC and MCPs groups were fed with high fat diet sequentially, while rats in NC group were fed with basic diet. Fasting serum glucose (GLU), insulin (INS) and lipids were assayed respectively before and after MCPs treatment. Oral glucose tolerance test, antioxidase activity, serum malondialdehyde (MDA) levels and the ultrastructure of islet beta cell were carried out at the end of the experiment.RESULTS:After 8 weeks of treatment, compared with HIC group, fasting INS, TC and TG of all three MCPs-treated groups decreased obviously, while fasting GLU level lowered significantly in 0.225 g/kg bw and 1.35 g/kg bw MCPs groups. Moreover, GLU level at 0.5 h, 2 h and the area under blood sugar curve in OGTT were evidently lower in 0.45 g/kg bw and 1.35 g/kg bw MCPs groups. Besides, serum SOD activity increased in all MCPs groups, and serum GSH-Px activity elevated obviously in 1.35 g/kg bw MCPs group, while serum MDA reduced. Finally, the ultrastructural impairment of islet resulted from high fat diet was improved evidently by 1.35 g/kg bw MCPs.CONCLUSION:MCPs was effective in improving glucose and lipid metabolism in hyperinsulinemic rats induced by high fat diet.

Abstract:The management of chronic wounds in diabetes remains challenging due to the complexity of impaired wound healing, delayed healing, susceptibility to infection, and elevated risk of reopening, highlighting the need for effective chronic wound management with innovative approaches such as multifunctional hydrogels. Here, we have produced HA-DA@rhCol hydrogels consisting of dopamine-modified hyaluronic acid and recombinant human collagen type-III (rhCol) by oxidative coupling of the catechol group using the H<sub>2</sub>O<sub>2</sub>/HRP catalytic system. The post-reactive hydrogel has a good porous structure, swelling rate, reasonable degradation, rheological and mechanical properties, and the catechol group and dopamine impart to the hydrogel tissue adhesiveness, antioxidant capacity, and excellent photothermal effects leading to superior in vitro antimicrobial activity. In addition, the ability of rhCol to confer hydrogels to promote angiogenesis and wound repair has also been investigated. Cytotoxicity and hemolysis tests demonstrated the good biocompatibility of the hydrogel. Wound closure, collagen deposition and immunohistochemical examination confirmed the ability of the hydrogel to promote diabetic wound healing. In summary, the adhesive hemostatic antioxidative hydrogel with rhCol to promote wound healing in diabetic rat is an excellent chronic wound dressing.

Docosahexaenoic Acid Reduced Vascular Endothelial Cell Injury in Diabetic Rats Via the Modulation of Autophagy

Abstract:Purpose: Among varied ω-3 polyunsaturated fatty acid types, the therapeutic properties of docosahexaenoic acid (DHA) have been indicated under diabetic conditions in different cell lineages. Here, we investigated the anti-diabetic properties of DHA in rats with type 2 diabetes mellitus (D2M) focusing on autophagy-controlling factors. Methods: D2M was induced in male Wistar rats using a single dose of streptozocin (STZ) and a high-fat diet for 8 weeks. On week 2, diabetic rats received DHA 950 mg/kg/d until the end of the study. After that, rats were euthanized, and aortic and cardiac tissue samples were stained with H&E staining for histological assessment. The expression of adhesion molecules, ICAM-1 and VCAM-1, was measured in heart samples using real-time PCR analysis. Using western blotting, protein levels of BCLN1, LC3, and P62 were measured in D2M rats pre- and post-DHA treatment. Results: Data showed intracellular lipid vacuoles inside the vascular cells, and cardiomyocytes, after induction of D2M and DHA reduced intracellular lipid droplets and in situ inflammatory response. DHA can diminish increased levels of ICAM-1 in diabetic conditions (P Control vs. D2M rats=0.005) and reach near-to-control values (P Control vs. D2M rats=0.28; P D2M rats vs. D2M rats+DHA=0.033). Based on western blotting, D2M slightly increased the BCLN1 and LC3-II/I ratio without affecting P62. DHA promoted the LC3II/I ratio (P=0.303) and reduced P62 (P Control vs. D2M rats+DHA =0.0433; P D2M vs. D2M rats+DHA=0.096), leading to the completion of autophagy flux under diabetic conditions. Conclusion: DHA can reduce lipotoxicity of cardiovascular cells possibly via the activation of adaptive autophagy response in D2D rats.
A Three-Month Clinical Trial on the Efficacy of Hyaluronic Acid Adjunctive Non-Surgical Therapy for Periodontitis in Patients with Type 2 Diabetes Mellitus

Abstract:Background/Objectives: Conventional periodontal treatment for patients with diabetes has shown promising results, primarily focusing on glycated hemoglobin (HbA1c) levels as an endpoint measure. The properties of hyaluronic acid (HA) have been harnessed in various periodontal therapies, and it is a promising agent also in a non-surgical approach. The aim of this clinical trial was to assess the efficacy of hyaluronic acid in a local adjunctive non-surgical treatment for periodontitis in patients with type 2 diabetes. Methods: Eighty adult participants with well-controlled type 2 diabetes (HbA1c 7% (53 mmol/mol) or less) took part in the trial. The clinical parameters of periodontitis as well as the glycated hemoglobin (HbA1c) levels were evaluated, and an analysis of the potential differences between the control (placebo) and intervention (HA) groups was performed. Results/Conclusions: A decrease in all the clinical values of periodontitis after treatment was observed in the vast majority of patients in both groups. Differences in the clinical parameters were observed 12 weeks after the intervention between the patients in the placebo and HA therapy groups. Bleeding on probing (BoP) was reduced in the control group to 15–25% and was approximately 5.5% more in the intervention group (9.5–18.25%). The clinical attachment level (CAL) decreased 1 mm more in the HA therapy group (1–2 mm) than in the no adjunctive treatment group (2–3 mm). The probing depth (PD) was reduced similarly in both groups (3–3.75 mm). Due to the bilateral relationship between diabetes and periodontitis, healthcare professionals seek advancements in managing periodontal inflammation. The results of this study indicate that non-surgical periodontal treatment with HA as an adjunctive agent is worth considering in the therapy for patients with diabetes.

Curcumin administration suppresses collagen synthesis in the hearts of rats with experimental diabetes

Abstract:Cardiac fibrosis is considered the initial change of diabetic cardiomyopathy (DCM). We have shown that curcumin alleviates collagen deposition in DCM, but the mechanism remains unknown. In this study we sought to investigate the effects of curcumin on cardiac fibrosis in vivo and in vitro and to elucidate the underlying mechanisms. Experimental diabetes was induced in rats by injection of low-dose streptozotocin (STZ) combined with high energy diet. The rats were orally treated with curcumin (300 mg·kg−1·d−1) for 16 weeks. Curcumin administration significantly suppressed the deposition of type I and type III collagens in the heart tissues of diabetic rats, accompanied by markedly reduced TGF-β1 production, suppressed TβR II levels and Smad2/3 phosphorylation, and increased Smad7 expression. Similar effects were observed in human cardiac fibroblasts exposed to high glucose (HG, 30 mmol/L) or exogenous TGF-β1 (5 ng/mL). Furthermore, TGF-β1 or HG treatment significantly increased the phosphorylation levels of AMPK and p38 MAPK in the fibroblasts. Application of curcumin (25 μmol/L) inhibited TGF-β1- or HG-induced AMPK/p38 MAPK activation and suppressed collagen synthesis in the fibroblasts. These effects were similar to those of the AMPK inhibitor compound C (10 μmol/L) but opposite to the effects of the AMPK activator metformin (2 mmol/L) in the fibroblasts. Our results demonstrate that curcumin suppresses diabetes-associated collagen synthesis in rat myocardium not only by inhibiting TGF-β1 production and canonical Smad signaling but also by blocking the non-canonical AMPK/p38 MAPK pathway.

Abstract:The multifactorial nature of diabetic wounds necessitates a mixed approach for successful treatment. Compensation of degenerated wound tissue extracellular matrix (ECM) and application of anti-inflammatory and antioxidant agents have been shown to be promising. Here, an attempt was made to fabricate a biocompatible wound dressing from curcumin-incorporated human amniotic membrane (HAM) ECM-derived scaffold to accelerate diabetic wound healing in rats. Therefore, after inducing diabetes, an excisional ischemic wound was created on rat skin, then treatments were administered for a period of 21 days. The main groups were the diabetic animals that received an engraftment of HAM scaffold (HAMS group) and the curcumin-incorporated HAMS (HAMS/β/C group). Evaluation at post-wounding days 7, 14, and 21 indicated that the parameters related to regeneration, including wound closure, volume of new epidermis and dermis, proliferating cells, fibroblasts, blood vessels, collagen deposition, and tensile strength, as well as transcripts of Vegf , bFgf , and Tgf-β genes of the healed wound in both HAMS and HAMS/β/C groups were considerably greater than those of the diabetic group. Conversely, the presence of inflammatory cells, i.e., neutrophils and macrophages, and the transcripts of Tnf-α and Il-1β showed a dramatic decrease in the treated groups relative to the diabetic group. Finally, compared to the HAMS group, considerable differences were found with the HAMS/β/C group in almost all evaluated parameters. Overall, these results suggest that using the complementary or synergistic effects of curcumin and HAMS could be a promising approach to improve diabetic wound healing.

Abstract:Aims Vascular endothelial growth factor (VEGF) is an important active protein for the induction of angiogenesis and plays an important role in the tissue regeneration of diabetic wounds. In this study, we used collagen-binding VEGF in a diabetic rat model to investigate the effects of this new method. Methods We produced a fusion protein (CBD-VEGF) consisting of VEGF and a collagen-binding domain (CBD), which allowed VEGF to bind to collagen. The diabetic rat models were made by injected streptozocin (STZ) peritoneally and removed full thickness skin on the back. All the rats were randomly divided into 3 groups: PBS group (n = 24), NAT-VEGF group (n = 24), and CBD-VEGF group (n = 24). After model establishment, the dissolved drugs were evenly given on the wounds using syringe. The healing rates were calculated and compared among the groups and the tissues of the wound were taken and evaluated for histological analysis. Results The CBD-VEGF group showed better result in wound healing rate, better vascularization and higher amount of VEGF in the wound granulation tissue compared with NAT-VEGF group and control. Conclusions Topical application of CBD-VEGF can promote diabetic wound healing in rat model, which could potentially provide a better therapeutic option for diabetic wounds.

Abstract:Continuous system identification of aircraft stability and control derivatives is proposed for failure identification. Prior information from flight mechanics is incorporated as constraints in linear regression, to improve the speed and accuracy of identification. A linearized fourth - order F-16 model with measurement (sensor) noise and damage to the horizontal tail is simulated. The simulated inflight failure problem demonstrates significant improvements in the short period parameter estimates.

Materials

Abstract:Diabetes mellitus continues to be one of the most common diseases often associated with diabetic ulcers. Chitosan is an attractive biopolymer for wound healing due to its biodegradability, biocompatibility, mucoadhesiveness, low toxicity, and hemostatic effect. A panel of hydrogels based on chitosan, collagen, and silver nanoparticels were produced to treat diabetic wounds. The antibacterial activity, cytotoxicity, swelling, rheological properties, and longitudinal sections of hydrogels were studied. The ability of the gels for wound healing was studied in CD1 mice with alloxan-induced diabetes. Application of the gels resulted in an increase in VEGF, TGF-b1, IL-1b, and TIMP1 gene expression and earlier wound closure in a comparison with control untreated wounds. All gels increased collagen deposition, hair follicle repair, and sebaceous glands formation. The results of these tests show that the obtained hydrogels have good mechanical properties and biological activity and have potential applications in the field of wound healing. However, clinical studies are required to compare the efficacy of the gels as animal models do not reproduce full diabetes pathology.

Abstract:Treating diabetic ulcers is a major challenge in clinical practice, persecuting millions of patients with diabetes and increasing the medical burden. Recombinant growth factor application can accelerate diabetic wound healing via angiogenesis. The local administration of recombinant growth factors has no robust clinical efficiency because of the degradation of append short duration of the molecules in the hostile inflammatoryenvironment.The present study focused on the pathophysiology of impaired neovascularization and growth factor short duration in the diabetic wound. We prepared a collagen-binding domain (CBD)-fused recombinant peptide (C-Histatin-1) that had both pro-angiogenesis capacity and collagen-affinity properties. Next, we created a biocompatible acellular dermal matrix (ADM) as a drug delivery carrier that featured collagen-richness, high porosity, and non-cytotoxicity. C-Histatin-1 was then tethered on ADM to obtain a sustained-release effect. Finally, a functional scaffold (C-Hst1/ADM) was developed. C-Hst1/ADM can sustain-release Histatin-1 to promote the adhesion, migration, and angiogenesisof vascular endothelial cells in vitro. Using a diabetic wound model, we showed that C-Hst1/ADM could significantly promote angiogenesis, reduce scar widths, and improve extracellular collagen accumulation. Therefore, the results of this study provide a foundation for the clinical application of C-Hst1/ADM covering scaffold in the treatment of diabetic wounds.

Adipose-derived stem cells from diabetic mice show impaired vascular stabilization in a murine model of diabetic retinopathy

Abstract:Diabetic retinopathy is characterized by progressive vascular dropout with subsequent vision loss. We have recently shown that an intravitreal injection of adipose-derived stem cells (ASCs) can stabilize the retinal microvasculature, enabling repair and regeneration of damaged capillary beds in vivo. Because an understanding of ASC status from healthy versus diseased donors will be important as autologous cellular therapies are developed for unmet clinical needs, we took advantage of the hyperglycemic Akimba mouse as a preclinical in vivo model of diabetic retinopathy in an effort aimed at evaluating therapeutic efficacy of adipose-derived stem cells (mASCs) derived either from healthy, nondiabetic or from diabetic mice. To these ends, Akimba mice received intravitreal injections of media conditioned by mASCs or mASCs themselves, subsequent to development of substantial retinal capillary dropout. mASCs from healthy mice were more effective than diabetic mASCs in protecting the diabetic retina from further vascular dropout. Engrafted ASCs were found to preferentially associate with the retinal vasculature. Conditioned medium was unable to recapitulate the vasoprotection seen with injected ASCs. In vitro diabetic ASCs showed decreased proliferation and increased apoptosis compared with healthy mASCs. Diabetic ASCs also secreted less vasoprotective factors than healthy mASCs, as determined by high-throughput enzyme-linked immunosorbent assay. Our findings suggest that diabetic ASCs are functionally impaired compared with healthy ASCs and support the utility of an allogeneic injection of ASCs versus autologous or conditioned media approaches in the treatment of diabetic retinopathy.
