Abstract:Molecular imprinting polymers(MIPs) were synthesized by bulk polymerization,using capsaicin as the template molecule and 2-vinylpyridine(2-VP) and Acrylamide(AM) as the function monomers.The specific adsorption ability of the molecularly imprinted polymers(MIPs) to capsaicin was evaluated by equilibrium binding experiments,saturated adsorption time was assessed by saturated adsorption experiments and selective adsorption ability to capsaicin analogs was appraised by selective adsorption experiments.The results showed that MIPs exerted the best adsorption efficiency to capsaicin when the ratio of capsaicin:2-VP:AM was 1:2:2,with a adsorption capacity of 20.46 mg/g and a recognition factor of 3.05.Morevoer saturated adsorption time was 6 h and better selective recognition to capsaicin was found.In this study,the elution efficiencies of eluents of different polarity for specific adsorption and non-specific adsorption of capsaicin in solid phace extraction column of MIPs were investigated.The specific adsorption was kept while the non-specific adsorption was discharged to the greatest extent when using chloroform as the eluent.

Abstract:A pre-treatment methodology for clenbuterol hydrochloride (CLEN) isolation and enrichment in a complex matrix environment was developed through exploiting molecularly imprinted polymers (MIPs). CLEN-imprinted polymers were synthesized by the combined use of ally-β-cyclodextrin (ally-β-CD) and methacrylic acid (MAA), allyl-β-CD and acrylonitrile (AN), and allyl-β-CD and methyl methacrylate (MMA) as the binary functional monomers. MAA-linked allyl-β-CD MIPs (M-MAA) were characterized by Fourier transform-infrared (FT-IR) spectroscopy and a scanning electron microscope (SEM). Based upon the results, M-MAA polymers generally proved to be an excellent selective extraction compared to its references: AN-linked allyl-β-CD MIPs (M-AN) and MMA-linked allyl-β-CD MIPs (M-MMA). M-MAA polymers were eventually chosen to run through a molecularly imprinted solid-phase extraction (MISPE) micro-column to enrich CLEN residues spiked in pig livers. A high recovery was achieved, ranging from 91.03% to 96.76% with relative standard deviation (RSD) ≤4.45%.

Abstract:SUMMARY In order to develop a test system for predicting cytotoxic response to chemotherapeutic agents, leukocytes from 36 adult patients and 2 teenagers with acute leukemia were cul tured in vitro for 24 hr and the effects of drugs on the incor poration of uridine-3H in RNA and of thymidine-'4 C in DNA were measured. Cortisol, vincristine, and cytosine arabinoside produced inhibition of uridine-3H incorporation in sensitive leukocytes. The effects of cortisol and vincristine on the incor poration of uridine-3H in RNA and of thymidine-'4 C were, in general, similar. In two studies, cytosine arabinoside produced a more profound depression of incorporation of radioactive thymidine than of radioactive uridine. From 45 in vitro and clinical studies of leukocyte response to glucorticoids and vincristine, it was concluded that the in vitro test system can predict the in vivo cytotoxicity of these compounds in acute leukemia in adults. From 8 in vitro and clinical studies of cytosine arabinoside, it appeared likely that the in vitro test system may have predictive value for the cytotoxic effect of this drug in vivo. Whereas the test system may have use in predicting in vivo cytotoxic effects, it cannot be used for predicting the likelihood of inducing a hematologic remission.
HPLC Determination of Midazolam in Rat Plasma and Applied to Investigation the Drug Interaction

Abstract:Objective:To develop an RP-HPLC method for the determination of midazolam in rats plasma.And the method was applied to investigate the induction effect of zolmitriptan on rat hepatic CYP3A activity in vivo. Method:HPLC for midazolam:The analytical column was Diamonsil~(TM)C_(18)(200 mm×4.6 mm,5 μm),the mobile phase consisted of acetonitrile-0.01 mol·L~(-1)potassium dihydrogen phosphate buffer solution(pH 5.0)(58: 42),the flow rate was 1.0 mL·min~(-1),and detected at 220 nm.Male and female SD-rats treated with zolmitriptan, dexamethasone and placebo(blank control)separately.And then animals received a single injection of midazolam (10 mg·kg~(-1))in the tail vein after the last dose of zolmitriptan,dexamethasone or placebo.The midazolam phar- macokinetics of zolmitriptan group,dexamethasone group and blank control were compared.Results:The standard curves for midazolam were linear in the range of 0.3-375 μmol·L~(-1),r=0.9997,and the detection concentration of midazolam was 0.0018 μmol·L~(-1).The average recovery of midazolam was 97.9%.The area under the curve (AUC_(0-t)and the elimination half-life of midazolam was significantly reduced in male rats by the zolmitriptan and dexamethasone pretreatment.While in female rats there were no significantly changes.Conclusion:The method is stable,accurate and simple,and can be used for investigation the effect of zolmitriptan on CYP3A activity in rats. Zolmitriptan has the gender-different effect of induction on male rat hepatic CYP3A activity.
Computer aided-molecular design and synthesis of a high selective molecularly imprinted polymer for solid-phase extraction of furosemide from human plasma

Abstract:Highly selective molecularly imprinted polymers (MIPs) for solid-phase extraction and determination of furosemide in human plasma have been designed and prepared. In order to study the intermolecular interactions in the pre-polymerization mixture and to find a suitable functional monomer in MIP preparation, a computational approach was developed. It was based on the comparison of the binding energy of the complexes between the template and functional monomers. Having confirmed the results of computational method, three MIPs were synthesized with different functional monomers, i.e. acrylamide (AAM), 4-vinylpiridine (4-VP) and acrylonitrile (ACN), and then evaluated using Langmuir-Freundlich (LF) isotherm. Using the MIP prepared by AAM as functional monomer, a molecularly imprinted solid-phase extraction procedure followed by high performance liquid chromatography with ultraviolet detector (MISPE-HPLC-UV) was developed for selective extraction and determination of furosemide in human plasma. For the proposed MISPE-HPLC-UV method, the linearity between responses (peak area) and concentration was found over the range of 75-3500 ng mL(-1) with a linear regression coefficient (R(2)) of 0.997. The limit of detection (LOD) and quantification (LOQ) in plasma were 12.9 and 43.3 ng mL(-1), respectively.

Abstract:Immunohistochemical analysis was carried out to examine the characteristics of nasopharyngeal carcinoma (NPC) using 38 biopsy cases obtained from southern China. These cases were divided into 3 groups according to their predominant pattern associated with cell and tissue differentiation which is based on World Health Organization (WHO) classification as follows: 6 cases of squamous cell carcinoma (16%), 25 cases of differentiated non-keratinizing carcinoma (66%), 7 cases of undifferentiated carcinoma (18%). All tumor tissues reacted with MB-1, but they did not react with L26 (CD20), 4KB5 (CD45R), MT-1, and leukocyte common antigen (LCA). Keratin and epithelial membrane antigen (EMA) as epithelial markers focally stained NPC tissues in all cases. Carcinoembryonic antigen (CEA)-positive staining was detected in 7 (28%) of the 25 cases of differentiated non-keratinizing carcinoma and in 3 (43%) of the 7 cases of undifferentiated carcinoma; thus, of 38 cases, 10 (26%) were CEA-positive. On the other hand, squamous cell carcinoma cases did not react with CEA. These NPC tissues did not react with S-100 protein, alpha-1-antichymotrypsin (ACT), lysozyme, vimentin, and desmin. Therefore, it is concluded that some cases of NPC are difficult to distinguish from malignant lymphoma. In certain cases, NPC may be distinguished from malignant lymphoma, using immunohistochemical methods for the detection of MB-1, keratin, EMA, and LCA. Specifically, this evidence suggests that MB-1 may be useful as a tumor marker of NPC. Moreover, the CEA reaction to NPC may be related to the cell differentiation.

Abstract:Molecularly imprinted polymers (MIPs) are being increasingly used as selective adsorbents in different analytical applications. To satisfy the different application purposes, MIPs with well controlled physical forms in different size ranges are highly desirable. For examples, MIP nanoparticles are very suitable to be used to develop binding assays and for microfluidic separations, whereas MIP beads with diameter of 1.5-3 microm can be more appropriate to use in new analytical liquid chromatography systems. Previous studies have demonstrated that imprinted microspheres and nanoparticles can be synthesized using a simple precipitation polymerization method. Despite that the synthetic method is straightforward, the final particle size obtained has been difficult to adjust for a given template. In this work, we initiated to study new synthetic conditions to obtain MIP beads with controllable size in the nano- to micro-meter range, using racemic propranolol as a model template. Varying the composition of the cross-linking monomer allowed the particle size of the MIP beads to be altered in the range of 130 nm to 2.4 microm, whereas the favorable binding property of the imprinted beads remained intact. The chiral recognition sites were further characterized with equilibrium binding analysis using tritium-labeled (S)-propranolol as a tracer. In general, the imprinted sites displayed a high chiral selectivity: the apparent affinity of the (S)-imprinted sites for (S)-propranolol was 20 times that of for (R)-propranolol. Compared to previously reported irregular particles, the chiral selectivity of competitive radioligand binding assays developed from the present imprinted beads has been increased by six to seven folds in an optimized aqueous solvent.

Abstract:Uniformly imprinted polymer microspheres were prepared by precipitation polymerization in chloroform.MCP(monocrotophos),MAA(methacrylic acid),EGDMA(ethylene glycol dimethacrylate) and AIBN(azobisisobutyronitrile) were used as template,functional monomer,cross_linker and initiator,respectively.UV and IR spectra were used to demonstrate the mechanism of the interaction between MCP and MAA.It was found that one MCP was entrapped by three MAA molecules in chloroform.Langmuir model was used to fit the adsorption data,the results showed that the MCP imprinted polymer microspheres showed specific binding sites to the MCP.The data obtained in equilibrium adsorption experiments were processed by Scatchard analysis,the dissociation contents(K_(D1)) and the apparent maximum binding capacity (B_(max1)) were K_(D1)=0.062?mmol/L,B_(max1)=3.26?μmol/g for high affinity binding sites and K_(D2)=3.03?mmol/L,B_(max2)=65.70?μmol/g for low affinity binding sites,respectively.The MCP imprinted polymers have showed higher affinity to MCP than to the other organophosphorus pesticide used.So the imprinted polymers microspheres can be used for trace pesticide residue separation,enrichment and analysis.
Selective Extraction of Diazepam and Its Metabolites from Urine Samples by a Molecularly Imprinted Solid-Phase Extraction (MISPE) Method

Abstract:In this research, a molecularly imprinted polymer (MIP) was synthesized by precipitation polymerization using oxazepam (OZ) as a template molecule and was subsequently applied as a selective sorbent for the extraction of diazepam (DZP) and its metabolites in urine samples using an SPE cartridge. OZ, temazepam (TZ), nordiazepam (NZ) and DZP were analyzed in the final extracts by high-performance liquid chromatography with diode array detection (HPLC-DAD). The SPE extraction steps were optimized, and the evaluation of an imprinting factor was carried out. The selectivity of the method for OZ versus structurally related benzodiazepines (BZDs), such as bromazepam (BRZ), tetrazepam (TTZ) and halazepam (HZ), was investigated. Under the optimum conditions, the proposed methodology provided good linearity in the range of 10–1500 ng/mL, with limit of detection values between 13.5 and 21.1 ng/mL and recovery levels for DZP and its metabolites from 89.0 to 93.9% (RSD ≤ 8%) at a concentration level of 1000 ng/mL. The proposed method exhibited good selectivity, precision and accuracy and was applied to the analysis of urine samples from a real case of DZP intake.

Abstract:Molecular imprinted polymers (MIPs) prepared using combination of acrylamide (ACM) and 4-vinylpyridine (4-Vpy) as co-functional monomers exhibited efficient recognition properties in both organic and aqueous media as HPLC stationary phase. The results indicate that amide and pyridine groups in functional monomers formed strong hydrogen-bonding interaction with the template molecule, and specific recognition sites were created within the polymer matrix during the imprinting process. When sulfamethoxazole (SMO) was used as template, a MIP prepared in a polar organic solvent (acetonitrile) using the combination of ACM and 4-Vpy showed better recognition of template than the polymer prepared in the same solvent using the combination of acidic monomer (methacrylic acid) and basic monomer 4-Vpy. On the contrary, when sulfamethazine (SMZ) was used as template, a MIP prepared using the combination of methacrylic acid (MAA) and 4-Vpy showed better recognition of template than the polymer prepared using the combination of ACM and 4-Vpy. Our results indicate that in organic media the degree of retention of the sample molecules on the imprinted polymers was controlled by the hydrogen-bonding interaction between the sample molecules and the polymer, while in aqueous media it was determined to a considerable extent by hydrophobic interactions. In both media the shape, size and the electronic structure of the template molecule were all-important factors in the recognition process.

Abstract:Molecular imprinting technology (MIT) offers an effective technique for efficient separation and enrichment of specific analytes from complicated matrices and has been used for illicit veterinary drug detectionin recent years due to its high selectivity, good chemical stability, and simple preparation. The development of in silico-based approaches has enabled the simulation of molecularly imprinted polymers (MIPs) to facilitate the selection of imprinting conditions such as template, functional monomer, and the best suitable solvent. In this work, using density functional theory (DFT), the molecularly imprinted polymers of clenbuterol and its metabolites were designed by computer-aided at B3LYP/6-31 + G (d, p) level. Screening molecular imprinting components such as functional monomers, cross-linkers, and solvents has been achieved in the computational simulation considerations. The simulation results showed that methacrylic acid (MAA) is the best functional monomer; the optimal imprinting ratio for both clenbuterol (CLB) and its dummy template molecule of phenylephrine (PE) to functional monomer is 1:3, while the optimal imprinting ratio for the two dummy template molecules of CLB’s metabolites is 1:5. Choosin gethyleneglycol dimethacrylate (EDGMA) as a crosslinker and aprotic solvents could increase the selectivity of the molecularly imprinted system. Atoms in Molecules (AIM) topology analysis was applied to investigate the template-monomer complexes bonding situation and helped to explain the nature of the reaction in the imprinting process. These theoretical predictions were also verified by the experimental results and found to be in good agreement with the computational results. The computer-simulated imprinting process compensates for the lack of clarity in the mechanism of the molecular imprinting process, and provides an important reference and direction for developing better recognition pattern towards CLB and its metabolite analytes in swine urine samples at the same time.

Abstract:The integration of molecular imprinting technique with chromatographic one has a great impact on the assay's selectivity and sensitivity. Herein, a molecularly imprinted solid-phase extraction associated with high performance liquid chromatography (MISPE-HPLC) was employed for simultaneous determination of the co-formulated drugs; tetracycline hydrochloride (TET) and metronidazole (MET), in plasma and in their anti-H-pylori drug for the first time. Two sorts of molecularly imprinted polymers (MIPs) were fabricated using TET and MET as the template molecules, while ethylene glycol dimethacrylate and methacrylic acid were used as a cross-linker and a monomer, respectively. The synthesized MIPs were identified using different techniques. The adsorption-desorption capability of each template was investigated towards its corresponding MIP. The extraction conditions of MISPE was optimized with respect to TET/MIP and MET/MIP sorbent. Bismuth subcitrate (BSC), the third co-formulated drug was analyzed in spiked human plasma using an atomic absorption spectrometric (AAS) method. The performance of the developed methods was assured as per ICH guidelines for analyzing the studied drugs in their pharmaceutical dosage form along with two of their official impurities. In addition, bioanalytical method validation was conducted where linearity was achieved at 2.0-40.0 μg mL-1, 2.0-40.0 μg mL-1 and 5.0-80.0 μg mL-1 for TET, MET and BSC, respectively.

Abstract:The evaluation of molecularly imprinted polymers (MIPs) as selective sorbents for the solid-phase extraction of sildenafil and its principal metabolite, desmethylsildenafil, was investigated. Two MIPs were synthesised using structural analogues of sildenafil as templates, and a comparison of the performance of the two MIP sorbents in organic and aqueous media was performed. Additionally, the feasibility of applying molecularly imprinted solid-phase extraction (MISPE) to the clean-up of plasma samples containing sildenafil and desmethylsildenafil was investigated. A preliminary, quantitative MISPE for the determination of both compounds in plasma was also performed. The results showed that the MIPs used for the selective extraction of sildenafil gave better compound recovery when aqueous samples were used in comparison to organic-based samples. A preliminary, quantitative MISPE of sildenafil and desmethylsildenafil indicated that the imprinted materials could be used successfully as SPE sorbents for sample pre-treatment for the determination of sildenafil, and related compounds, in plasma.

Abstract:Sulfamethazine and sulfamethoxazole are anti-bacterial and anti-infective agents. Molecular imprinted polymers of these agents were prepared by the technique of non-covalent molecular imprinting polymerization. The functional monomers used include methacrylic acid, 4-vinylpyridine, acrylamide or a combination of methacrylic acid and 4-vinylpyridine. The polymers obtained were packed in a high-performance liquid chromatography (HPLC) column, and the molecular recognition mechanism regulating the binding behavior was studied using chromatography. The results showed that methacrylic acid was suitable for synthesizing sulfamethazine-selective polymer, whereas 4-vinylpyridine was suitable for sulfamethoxazole. Although these two compounds have similar structures, the interactions between them and the monomers are different. The selective polymers are expected to be of use for analyzing sulfonamide compounds in the food industry and clinic applications.
Selective Extraction of Functional Components Derived from Herb in Plasma by Using a Molecularly Imprinted Polymer Based on 2,2-Bis(hydroxymethyl)butanol Trimethacrylate.

Abstract:To make molecularly imprinted polymer (MIP) solid-phase extraction (SPE) capable of direct clean-up of aqueous biological samples, an anti-quercetin MIP with evident hydrophobic matrix was synthesized using acrylamide (AA) as the functional monomer and 2,2-bis(hydroxymethyl)butanol trimethacrylate (TRIM) as the crosslinker. The affinity and selectivity were evaluated by liquid chromatography, and the binding sites and the dissolution constants were measured by frontal chromatography. Compared with the AA-co-ethyleneglycol dimethacrylate (EDMA) MIP, the anti-quercetin AA-co-TRIM MIP exhibited stronger binding and possessed improved column efficacy. A linear plot of the peak area versus sample size (in the range of 0.4-2.2 microg) was obtained, which made it promising for the MIP columns to be directly used for analysis. Before MIP-SPE of the sample of plasma, several washing solvents were tested and it was shown that the careful choice of the right washing solvent is the key step to successful sample extraction. The anti-quercetin AA-co-TRIM polymer selectively extracted quercetin, the effective component in the plasma of rats fed the hydrolyzed extract of Gingko biloba L. The recovery (67%) for MIP-SPE was calculated using spiked plasma. The results of the present work showed that the properties of MIP could be improved by modifying the polymerization and that MIP-SPE could be used for direct clean-up of biological samples for the analysis of functional components in vivo originating from an extract of medicinal herbs.

Abstract:In this work, diflubenzuron (DBZ) molecularly imprinted polymers (MIPs) were synthesized by precipitation polymerization. The polymers were characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), dynamic light scattering (DLS). The influences of synthetic conditions including template molecules, functional monomers and crosslinker on the surface morphology, particle size and size distribution of polymers were investigated. Scatchard analysis indicated the presence of specified recognition sites in MIPs. The Sips model was fitted better to the equilibrium data of MIPs over whole concentrations. The pseudo-second-order kinetic model was well fitted to the experimental data of adsorption kinetic experiment, suggesting the chemisorption mechanism between MIPs and DBZ. Four structurally similar compounds were used for the selectivity test of MIPs. It was observed that MIPs can selectively rebind the template molecules (DBZ) under the interference of four other structurally similar compounds. The application of MIPs in solid-phase extraction (SPE) offers a method for extraction and determination of benzoylureas (BUs) pesticides in apple samples prior to high-performance liquid chromatography (HPLC) analysis.

Abstract:A chlorphenamine–imprinted polymer was prepared in this study. Chromatographic analysis showed that the retentivity and selectivity of the imprinted polymer were greatly strengthened through molecular imprinting. As a consequence, chlorphenamine could be easily separated from diphenhydramine. Ionic interaction was proved to be the main power for the imprinted polymer to bind chlorphenamine. The strong extraction ability of the imprinted polymer for chlorphenamine in aqueous solution was evaluated further. It was shown that chlorphenamine, as low as 0.02 μmol l−1, could be concentrated by 50 times with a recovery of more than 90% at pH 5. This study gave the potential of using the imprinted polymer for solid-phase extraction of practical samples.
The Selectivity of Polymers Imprinted with Amines

Abstract:One of the main reasons for making molecularly imprinted polymers (MIPs) has been that MIPs interact selectively with a specific target compound. This claim is investigated here with the example of a widely used type of noncovalent MIP, the MIP for the beta blocker propranolol. Adsorption isotherms of this MIP and of a nonimprinted control polymer (NIP), respectively, have been measured with a series of compounds in the porogen solvent acetonitrile. The results, visualized as "selectivity ladders", show that the MIP binds propranolol and many other amines better than the NIP does, but the selectivity of the MIP is actually inferior to that of the NIP. The selectivity of either polymer for propranolol is modest against many amines, but is remarkable with respect to other compounds. The contribution of imprinting towards selectivity can be better appreciated when three MIPs, made with different amine templates, are compared among themselves. Each MIP is seen to bind its own template slightly better than the other two MIPs do. In media different from the porogen, the selectivity patterns may change substantially. Propranolol seems to have properties that make it stand high on the selectivity scale in different solvents, albeit for different reasons.
Molecularly Imprinted Polymer Prepared by Precipitation Polymerization for Chloramphenicol

Abstract:OBJECTIVE To prepare molecularly imprinted polymer (MIP) for chloramphenicol by precipitation polymerization, and evaluate their adsorption properties. METHODS Using methacrylic acid as the functional monomer,ethylene glycol dimethacrylate as the crossing -linker, chloramphenicol as the template molecule. The optimal ratio among them was determined. The effect of initiator, solvent, temperature and water on the adsorption was investigated, and Scatchard analysis was performed. RESULTS The polymer of the ratio of CAP -functional monomer -crossing -linker(1 : 3 : 24) had the maximal adsorption, and the temperature and the amount of water influenced the binding performance greatly. CONCLUSION The chloramphenicol MIP prepared by precipitation polymeriza-tion with simple operation has a good adsorption property, and will have a better application prospect.

Abstract:Molecularly imprinted polymers (MIPs) have been used as useful sorbents in solid-phase extraction for a wide range of molecules and sample matrices. Their unique selectivity can be fine-tuned in the imprinting process and is crucial for the extraction of macromolecules from complex matrices such as serum. A relevant example of this is the application of MIPs to peptides in diagnostic assays. In this article the selectivity of MIPs, previously implemented in a quantitative mass-spectrometric assay for the biomarker pro-gastrin-releasing peptide, is investigated. Partial least squares regression was used to generate models for the evaluation and prediction of the retention mechanism of MIPs. A hypothesis on interactions of MIPs with the target peptide was verified by ad hoc experiments considering the relevant peptide physicochemical properties highlighted from the multivariate analysis. Novel insights into and knowledge of the driving forces responsible for the MIP selectivity have been obtained and can be directly used for further optimization of MIP imprinting strategies.Graphical AbstractApplied analytical strategy: the Solid Phase Extraction (SPE) of digested Bovin Serum Albumin (BSA), using Molecularly Imprinted Polymers (MIP), is followed by the liquid chromatography-mass spectrometry (LC-MS) analysis for the identification of the retained peptides.
