Abstract:Cadmium (Cd) is a non-essential heavy metal with high toxicity to plants. Plants have acquired specialized mechanisms to sense, transport, and detoxify Cd. Recent studies have identified many transporters involved in Cd uptake, transport, and detoxification. However, the complex transcriptional regulatory networks involved in Cd response remain to be elucidated. Here, we provide an overview of current knowledge regarding transcriptional regulatory networks and post-translational regulation of the transcription factors involved in Cd response. An increasing number of reports indicate that epigenetic regulation and long non-coding and small RNAs are important in Cd-induced transcriptional responses. Several kinases play important roles in Cd signaling that activate transcriptional cascades. We also discuss the perspectives to reduce grain Cd content and improve crop tolerance to Cd stress, which provides a theoretical reference for food safety and the future research of plant varieties with low Cd accumulation.
Transcriptomic Sequencing Analysis on Key Genes and Pathways Regulating Cadmium (Cd) in Ryegrass (Lolium perenne L.) under Different Cadmium Concentrations

Abstract:Perennial ryegrass (Lolium perenne L.) is an important forage grass and has the potential to be used in phytoremediation, while little information is available regarding the transcriptome profiling of ryegrass leaves in response to high levels of Cd. To investigate and uncover the physiological responses and gene expression characteristics of perennial ryegrass under Cd stress, a pot experiment was performed to study the transcriptomic profiles of ryegrass with Cd-spiked soils. Transcriptome sequencing and comparative analysis were performed on the Illumina RNA-Seq platform at different concentrations of Cd-treated (0, 50 and 500 mg·kg−1 soil) ryegrass leaves and differentially expressed genes (DEGs) were verified by RT-qPCR. The results show that high concentrations of Cd significantly inhibited the growth of ryegrass, while the lower concentrations (5 and 25 mg·kg−1) showed minor effects. The activity levels of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and malondialdehyde (MDA) increased in Cd-treated ryegrass leaves. We identified 1103 differentially expressed genes (DEGs) and profiled the molecular regulatory pathways of ryegrass leaves with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis in response to Cd stress. Cd stress significantly increased the membrane part, the metabolic process, the cellular process and catalytic activity. The numbers of unigenes related to signal transduction mechanisms, post-translational modification, replication, recombination and repair significantly increased. KEGG function annotation and enrichment analysis were performed based on DEGs with different treatments, indicating that the MAPK signaling pathway, the mRNA surveillance pathway and RNA transport were regulated significantly. Taken together, this study explores the effect of Cd stress on the growth physiology and gene level of ryegrass, thus highlighting significance of preventing and controlling heavy metal pollution in the future.

Co-expression analyses reveal key Cd stress response-related metabolites and transcriptional regulators in Kentucky bluegrass

Abstract:Kentucky bluegrass (Poa pratensis) is known for its high cadmium (Cd) tolerance and accumulation, and it is therefore considered to have the potential for phytoremediation of Cd-contaminated soil. However, the mechanisms underlying the accumulation and tolerance of Cd in Kentucky bluegrass are largely unknown. In this study, we examined variances in the transcriptome and metabolome of a Cd-tolerant variety (Midnight, M) and a Cd-sensitive variety (Rugby II, R) to pinpoint crucial regulatory genes and metabolites associated with Cd response. We also validated the role of the key metabolite, l-phenylalanine, in Cd transport and alleviation of Cd stress by applying it to the Cd-tolerant variety M. Metabolites of the M and R varieties under Cd stress were subjected to co-expression analysis. The results showed that shikimate-phenylpropanoid pathway metabolites (phenolic acids, phenylpropanoids, and polyketides) were highly induced by Cd treatment and were more abundant in the Cd-tolerant variety. Gene co-expression network analysis was employed to further identify genes closely associated with key metabolites. The calcium regulatory genes, zinc finger proteins (ZAT6 and PMA), MYB transcription factors (MYB78, MYB62, and MYB33), ONAC077, receptor-like protein kinase 4, CBL-interacting protein kinase 1, and protein phosphatase 2A were highly correlated with the metabolism of phenolic acids, phenylpropanoids, and polyketides. Exogenous l-phenylalanine can significantly increase the Cd concentration in the leaves (22.27%-55.00%) and roots (7.69%-35.16%) of Kentucky bluegrass. The use of 1 mg/L of l-phenylalanine has been demonstrated to lower malondialdehyde levels and higher total phenols, flavonoids, and anthocyanins levels, while also significantly enhancing the uptake of Cd and its translocation from roots to shoots. Our results provide insights into the response mechanisms to Cd stress and offer a novel l-phenylalanine-based phytoremediation strategy for Cd-containing soil.
Transcriptome-wide m6A methylation profile reveals tissue specific regulatory networks in switchgrass (Panicum virgatum L.) under cadmium stress

Abstract:The heavy metal cadmium (Cd), known for its high toxicity, poses a grave threat to human health through the food chain. N6-methyladenosine (m6A), the most abundant internal modification, regulates plant adaptation to various adversities, yet the panorama of m6A modifications in switchgrass under cadmium stress remains elusive. This study examines the physiological responses of switchgrass roots and shoots exposed to 50 μM CdCl2, alongside an overview of transcriptome-wide m6A methylation patterns. After cadmium treatment, methylation modifications are primarily enriched near stop codons and the 3'UTR region, with a negative correlation between m6A modification and gene expression levels. In shoots, approximately 58 % of DEGs with m6A modifications show upregulation in expression and decrease in m6A peaks, including zinc transporter 4-like (ZIP4). In roots, about 43 % of DEGs with m6A modifications exhibit downregulation in expression and increase in m6A peaks, such as the ABC transporter family member (ABCG25). We further validate the m6A enrichment, gene expression and mRNA stability of ZIP4 in response to Cd treatment. The results suggest that the negative correlation of m6A enrichment and gene expression is due to altered mRNA stability. Our study establishes an m6A regulatory network governing cadmium transport in switchgrass roots and shoots, offering new avenues for candidate gene manipulation in phytoremediation applications of heavy metal pollution.

Abstract:Kentucky bluegrass ( Poa pratensis L., KB) demonstrates superior performance in both cadmium (Cd) accumulation and tolerance; however, the regulatory mechanisms and detoxification pathways in this species remain unclear. Therefore, phenotype, root ultrastructure, cell wall components, proteomics, transcriptomics, and metabolomics were analyzed under the hydroponic system to investigate the Cd tolerance and accumulation mechanisms in the Cd-tolerant KB variety 'Midnight (M)' and the Cd-sensitive variety 'Rugby II (R)' under Cd stress. The M variety exhibited higher levels of hydroxyl and carboxyl groups as revealed by Fourier transform infrared spectroscopy spectral analysis. Additionally, a reduced abundance of polysaccharide degradation proteins was observed in the M variety. The higher abundance of glutathione S-transferase and content of L-cysteine-glutathione disulfide and oxidized glutathione in the M variety may contribute to better performance of the M variety under Cd stress. Additionally, the R variety had an enhanced content of carboxylic acids and derivatives, increasing the Cd translocation capacity. Collectively, the down-regulation of cell wall polysaccharide degradation genes coupled with the up-regulation of glutathione metabolism genes enhances the tolerance to Cd stress in KB. Additionally, lignification of the endodermis and the increase in carboxylic acids and derivatives play crucial roles in the redistribution of Cd in KB.

Comparison of cadmium uptake and transcriptional responses in roots reveal key transcripts from high and low-cadmium tolerance ryegrass cultivars

Abstract:Cadmium (Cd), which seriously affects plant growth and crop production, is harmful to humans. Previous studies revealed ryegrass (Lolium multiflorum Lam.) exhibits Cd tolerance, and may be useful as a potential hyperaccumulator because of its wide distribution. In this study, the physiological and transcriptional responses of two ryegrass cultivars [i.e., high (LmHC) and low (LmLC) Cd tolerance] to Cd stress were investigated and compared. The Cd tolerance of LmHC was greater than that of LmLC at various Cd concentrations. The uptake of Evans blue dye revealed that Cd-induced root cell mortality was higher in LmLC than in LmHC after a 12-h Cd treatment. Furthermore, the content and influx rate of Cd in LmLC roots were greater than in LmHC roots under Cd stress conditions. The RNA sequencing and quantitative real-time PCR data indicated that the Cd transport regulatory genes (ABCG37, ABCB4, NRAMP4, and HMA5) were differentially expressed between the LmLC and LmHC roots. This expression-level diversity may contribute to the differences in the Cd accumulation and translocation between LmLC and LmHC. These findings may help clarify the physiological and molecular mechanisms underlying ryegrass responses to Cd toxicity. Additionally, ryegrass may be able to hyperaccumulate toxic heavy metals during the phytoremediation of contaminated soil.
Cadmium phytoextraction potential of king grass (Pennisetum sinese Roxb.) and responses of rhizosphere bacterial communities to a cadmium pollution gradient

Abstract:Screening for tolerant and high biomass producing plants is important for phytoextraction efforts in remediating agricultural soils contaminated by heavy metals. We carried out a greenhouse experiment involving a soil cadmium (Cd) concentration gradient (0.1, 0.5, 1, 2, 4, and 8 mg kg−1) to assess growth and phytoextraction capacity of king grass (Pennisetum sinese Roxb.) in soils contaminated by Cd and to explore changes in diversity and structure of rhizosphere soil bacterial communities in response to long-term Cd pollution. A significant positive relationship was observed between Cd concentrations in P. sinese stems, leaves, and roots and soil Cd concentration. The highest Cd concentrations in shoots and roots were 28.87 and 34.01 mg kg−1, respectively, at 8 mg kg−1of soil Cd supply. Total extraction amounts of Cd in P. sinese were 0.22–1.86 mg plant−1 corresponding to treatment with 0.5–8 mg kg−1 Cd. Most of the Cd was stored in shoots, and the largest accumulation was 1.56 mg plant−1 with 54.02 g dry shoot weight. After phytoextraction, changes in rhizobacterial community composition were found with different levels of Cd application, whereas there were no clear trends in diversity and richness. Results of this study show the feasibility of P. sinese in accumulating Cd and provide support for its application in remediation of soil moderately contaminated by Cd.

Abstract:Cadmium (Cd) contamination poses a substantial threat the environment, necessitating effective remediation strategies. Phytoremediation emerges as a cost-efficient and eco-friendly approach for reducing Cd levels in the soil. In this study, the suitability of A. venetum for ameliorating Cd-contaminated soils was evaluated. Mild Cd stress promoted seedling and root growth, with the root being identified as the primary tissue for Cd accumulation. The Cd content of roots ranged from 0.35 to 0.55 mg/g under treatment with 10-50 µM CdCl2·2.5 H2O, and the bioaccumulation factor ranged from 28.78 to 84.43. Transcriptome sequencing revealed 20,292 unigenes, and 7507 nonredundant differentially expressed genes (DEGs) were identified across five comparison groups. DEGs belonging to the "MAPK signaling pathway-plant," "monoterpenoid biosynthesis," and "flavonoid biosynthesis pathway" exhibited higher expression levels in roots compared to stems and leaves. In addition, cytokinin-related DEGs, ROS scavenger genes, such as P450, glutathione-S-transferase (GST), and superoxide dismutase (SOD), and the cell wall biosynthesis-related genes, CSLG and D-GRL, were also upregulated in the root tissue, suggesting that Cd promotes root development. Conversely, certain ABC transporter genes, (e.g, NRAMP5), and some vacuolar iron transporters, predominantly expressed in the roots, displayed a strong correlation with Cd content, revealing the mechanism underlying the compartmentalized storage of Cd in the roots. KEGG enrichment analysis of DEGs showed that the pathways associated with the biosynthesis of flavonoids, lignin, and some terpenoids were significantly enriched in the roots under Cd stress, underscoring the pivotal role of these pathways in Cd detoxification. Our study suggests A. venetum as a potential Cd-contaminated phytoremediation plant and provides insights into the molecular-level mechanisms of root development promotion and accumulation mechanism in response to Cd stress.

Abstract:The contamination of soils with cadmium (Cd) has become a serious environmental issue that needs to be addressed. Elucidating the mechanisms underlying Cd accumulation may facilitate the development of plants that accumulate both high and low amounts of Cd. In this study, a combination of phenotypic, physiological, and comparative transcriptomic analyses was performed to investigate the effects of different Cd concentrations (0, 5, 10, 30, 50 mg/kg) on Brassica juncea L. Our results suggest that B. juncea L. seedlings had a degree of tolerance to the 5 mg/kg Cd treatment, whereas higher Cd stress (10–50 mg/kg) could suppress the growth of B. juncea L. seedlings. The contents of soluble protein, as well as MDA (malondialdehyde), were increased, but the activities of CAT (catalase) enzymes and the contents of soluble sugar and chlorophyll were decreased, when B. juncea L. was under 30 and 50 mg/kg Cd treatment. Comparative transcriptomic analysis indicated that XTH18 (xyloglucan endotransglucosylase/hydrolase enzymes), XTH22 , and XTH23 were down-regulated, but PME17 (pectin methylesterases) and PME14 were up-regulated, which might contribute to cell wall integrity maintenance. Moreover, the down-regulation of HMA3 (heavy metal ATPase 3) and up-regulation of Nramp3 (natural resistance associated macrophage proteins 3) , HMA2 (heavy metal ATPase 2) , and Nramp1 (natural resistance associated macrophage proteins 1) might also play roles in reducing Cd toxicity in roots. Taken together, the results of our study may help to elucidate the mechanisms underlying the response of B. juncea L. to various concentrations of Cd.

Comparison and Characterization of Oxidation Resistance and Carbohydrate Content in Cd-Tolerant and -Sensitive Kentucky Bluegrass under Cd Stress

Abstract:Kentucky bluegrass (Poa pratensis L.), a turf grass species that is hypertolerant of cadmium (Cd), is a potential phytoremediation material for soil polluted with Cd. However, the mechanism of Cd phytotoxicity in Kentucky bluegrass is unclear. Here, we compared the phenotype, induction of oxidative stress, and structural and non-structural carbohydrate contents between a Cd-tolerant genotype (‘Midnight’, M) and Cd-sensitive genotype (‘Rugby’, R). The results showed that both genotypes accumulated more Cd in the roots, whereas the R genotype distributed more Cd into the leaves compared with the M genotype. In both genotypes, Cd inhibited the length and fresh weight of the leaves and roots; increased the peroxidase (POD) activity but inhibited ascorbate peroxidase (APX) and catalase (CAT) activity; and increased the superoxide radical (O2−), hydrogen peroxide (H2O2), and malondialdehyde (MDA) contents. However, the M genotype exhibited lower root length inhibition, and the H2O2 and MDA contents confirmed that the M genotype had increased Cd accumulation and resistance, while the R genotype exhibited a better distribution of Cd. Moreover, Cd stress significantly increased the soluble sugar, trehalose, and sucrose contents of both genotypes. Pectin, lignin, and cellulose were significantly increased to prevent the entry of Cd into the roots. The Cd-induced growth inhibition and physiological responses in Kentucky bluegrass were preliminarily explored herein, with the chelation of pectin, lignification, and antioxidant response being possible contributors to Cd detoxification in Kentucky bluegrass. In addition, the Cd-induced increase in trehalose, sucrose, and soluble sugar contents might play a pivotal role in the defense against Cd stress in Kentucky bluegrass.

Abstract:Cadmium (Cd) and dichlorodiphenyltrichloroethane (DDT) are frequently detected in agricultural soils, which poses a threat to public health. This study investigated the effects of inoculation of king grass with Piriformospora indica on the remediation of soils co-contaminated with Cd and DDTs. After treatment for 90 days, the dry shoot and root biomass of king grass inoculated with P. indica markedly increased by 13.0–15.8% and 24.1–46.4%, respectively, compared with those of uninoculated plants. Inoculation with P. indica also increased the uptake of Cd and DDTs by shoots and roots of king grass. The removal efficiency of Cd and DDTs from soils reached 4.88–17.4% and 48.4–51.0%, respectively, in the presence of king grass inoculated with P. indica. Under three Cd–DDTs contamination conditions, root secretion of organic acids, alcohol, and polyamines was distinctively stimulated by P. indica inoculation of king grass compared with planting king grass alone. After phytoremediation, changes in soil bacterial and fungal community composition occurred at different contamination levels. Overall, the results showed that king grass associated with P. indica can be adopted for phytoextraction of Cd and DDTs from moderately contaminated soils by regulating root excretion and reshaping rhizosphere microbial community structure.

Abstract:Cadmium (Cd) and arsenic (As) are two highly toxic heavy metals and metalloids that coexist in many situations posing severe threats to plants. Our investigation was conducted to explore the different regulatory mechanisms of ryegrass ( Lolium perenne L.) responding to individual and combined Cd and As stresses in hydroponics. Results showed that the ryegrass well-growth phenotype was not affected by Cd stress of 10 mg·L -1 . However, As of 10 mg·L -1 caused rapid water loss, proline surge, and chlorosis in shoots, suggesting that ryegrass was highly sensitive to As. Transcriptomic analysis revealed that the transcription factor LpIRO2 mediated the upregulation of ZIP1 and YSL6 that played an important role in Cd tolerance. We found that the presence of As caused the overexpression of LpSWT12 , a process potentially regulated by bHLH14, to mitigate hyperosmolarity. Indoleacetic acid (IAA) and abscisic acid (ABA) contents and expression of their signaling-related genes were significantly affected by As stress rather than Cd. We predict a regulatory network to illustrate the interaction between transporters, transcription factors, and signaling transduction, and explain the antagonism of Cd and As toxicity. This present work provides a research basis for plant protection from Cd and As pollution.

Abstract:Using king grass (Pennisetum purpureum Schumach. × Pennisetum glaucum (L.) R.Br.) for phytoextraction is a promising technology for producing large amounts of biomass fuel while remediating contaminated soil. To assess the practical phytoextraction capacity of king grass, we conducted a field experiment with three different soil types (loam, sandy loam, clay loam) and cadmium (Cd) concentrations (0, 0.25, 0.5, 1, 2, 4, 8, and 16 mg kg−1, aged stably for 6 years). King grass were harvested at two different periods (elongation and maturity) to identify the optimal harvest time for extraction efficiency. The results showed that all treatments had bioconcentration factor (BCF) > 1 and translocation factor (TF) < 1; Cd is mainly stored in the roots. However, due to a high shoot biomass, the highest quantity of Cd extracted from shoots was 2.75 mg plant−1, from the experimental group with 16 mg kg−1 Cd added in sandy loam. A significant positive relationship (P < 0.05) was observed between the amount of Cd extracted from king grass stems, leaves, and roots from soil with the diethylene triamine pentacetate acid (DTPA) extractable Cd concentration. The Cd concentration in shoots at the maturity stage is lower than at the elongation stage, mainly due to the effect of biological dilution. Meanwhile, there is significantly more biomass (P < 0.05) at the maturity stage than at the elongation stage. At the latter, the extraction efficiency of the three soils was loam > sandy loam > clay loam, while at maturity it was sandy loam > clay loam > loam. This change in extraction efficiency can be attributed mainly to differences in soil DTPA-extractable Cd concentration and growth rate caused by differences in soil physical and chemical properties. According to calculations from multiple harvests using three types of soil, remediating contaminated soil with 0–16 mg kg−1 Cd would take 13.9–224.5 and 19.5–250.6 years, extracting 7.21–265.23 and 4.96–330.52 g ha−1 Cd while producing 33.62–66.50 and 73.8–110.5 t ha−1 dry biomass at the elongation (90 days) and maturity (120 days) stages, respectively. In summary, king grass has major potential for remediating Cd-contaminated soil while producing large volumes of biofuel.

Abstract:Cadmium (Cd) pollution is a widespread environmental problem. In this study, we explored the transcriptome and biochemical responses of goldenrain tree (Koelreuteria paniculata Laxm.) leaves and roots to Cd stress. Leaf and root growth decreased substantially under Cd stress (50 mg/L CdCl2), but leaf and root antioxidant mechanisms were significantly activated. In RNA-seq analysis, roots treated with 25 mg/L CdCl2 featured enriched GO terms in cellular components related to intracellular ribonucleoprotein complex, ribonucleoprotein complex, and macromolecular complex. In leaves under Cd stress, most differentially expressed genes were enriched in the cellular component terms intrinsic component of membrane and membrane part. Weighted gene co-expression network analysis and analysis of module–trait relations revealed candidate genes associated with superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities and malondialdehyde (MDA). Ten transcription factors responded to Cd stress expression, including those in C2H2, MYB, WRKY, and bZIP families. Transcriptomic analysis of goldenrain tree revealed that Cd stress rapidly induced the intracellular ribonucleoprotein complex in the roots and the intrinsic component of membrane in the leaves. The results also indicate directions for further analyses of molecular mechanisms of Cd tolerance and accumulation in goldenrain tree.

Abstract:Introduction Cadmium (Cd) is a toxic heavy metal that severely threatens safe food production. Zhe-Maidong, a well-known Chinese traditional herbal medicine, is susceptible to Cd stress. However, the characteristics of Cd transformation and migration, as well as the regulatory system for genes conferring Cd accumulation of Zhe-Maidong, remains an essential issue to be addressed. Methods Zhe-Maidong seedling growth in Cd-contaminated and uncontaminated soil was conducted for 90 days. The Cd concentration was determined by inductively coupled plasma-mass spectrometry, and the Cd 2+ fluorescence probe detected Cd distributions. The root transcriptome of Zhe-Maidong was then evaluated using various Cd stress hydroponic treatments designated Cd-0, Cd-M, and Cd-H. Results and discussion The enrichment factor (EF) value in the root was four times that of the leaves, indicating that the root has a high ability to absorb and accumulate Cd. The Cd 2+ were mainly distributed in the root hair and the epidermis in both roots and leaves, revealing that the epidermal cells of roots may collect Cd 2+ and also have an outstanding role in Cd 2+ uptake. A total of 50 DEGs involved in Cd translocation and accumulation were identified. Among these DEGs, ANN, ABCC2/4, HMA1- 5, and CCX gene expression were positively correlated with EF-root, EF-leaf, EF-total, Cd-leaf, Cd-root, and Cd-plant, indicating their role in Cd transport and accumulation under Cd-stress. These data could be helpful in uncovering the Cd accumulation characteristics in Zhe-Maidong, as well as provide a bioinformatic foundation for investigations on finding gene functions and the screening of candidate genes related to Cd accumulation.

Abstract:To acquire a comprehensive understanding of the different plant organ molecular mechanisms in response to cadmium (Cd), transcriptome analyses of mung bean roots, stems, and leaves exposed to 100-μM Cd for 1, 5, and 9 days were performed. A total of 13,055, 8189, and 6705 differentially expressed genes (DEGs) were obtained in roots, stems, and leaves, respectively. The analysis of DEG enrichment and functional profiling showed that the upregulated DEGs in roots were significantly ( p < 0.05) enriched in signaling-, oxidation‒reduction-, and detoxification process-related GO terms, and the downregulated DEGs in roots were significantly ( p < 0.05) enriched in cell wall-, membrane-, and cell cytoskeleton-related GO terms. The upregulated DEGs in stems were significantly ( p < 0.05) enriched in signaling-, detoxification process-, and secondary metabolic process-related GO terms, and the downregulated DEGs in stems were significantly ( p < 0.05) enriched in cell wall- and membrane-related GO terms. The upregulated DEGs in leaves were significantly ( p < 0.05) enriched in regulation of transcription- and oxidation‒reduction process-related GO terms, and the downregulated DEGs in leaves were significantly ( p < 0.05) enriched in cell cytoskeleton- and photosynthesis-related GO terms. Key DEGs involved in the Cd response were obtained, including glutathione S-transferase-coding DEGs, pathogenesis-related protein-coding DEGs, isocitrate lyase-coding DEGs, malate synthase-coding DEGs, peroxidase-coding DEGs, photosynthesis-related DEGs, transporter-related DEGs, and secondary metabolism-related DEGs. Based on the results, it was proposed that the root was the principal organ involved in the response to Cd stress, the antioxidant system and secondary metabolic process were the main response pathways of the stem in response to Cd, and photosynthesis and the glyoxylate pathway were important biological processes in the leaf response to Cd.

Abstract:Kenaf is considered to have great potential for remediation of heavy metals in ecosystems. However, studies on molecular mechanisms of root Cd accumulation and tolerance are still inadequate. In this study, two differently tolerant kenaf cultivars were selected as materials and the physiological and transcriptomic effects were evaluated under Cd stress. This study showed that 200 µmol/L CdCl2 treatment triggered the reactive oxygen species (ROS) explosion and membrane lipid peroxidation. Compared with the Cd-sensitive cultivar 'Z', the Cd-tolerant cultivar 'F' was able to resist oxidative stress in cells by producing higher antioxidant enzyme activities and increasing the contents of ascorbic acid (AsA) and glutathione (GSH). The root cell wall of 'F' exhibited higher polysaccharide contents under Cd treatment, providing more Cd-binding sites. There were 3,439 differentially expressed genes (DEGs) that were co-regulated by Cd treatment in two cultivars. Phenylpropanoid biosynthesis and plant hormone signal transduction pathways were significantly enriched by functional annotation analysis. DEGs associated with pectin, cellulose, and hemi-cellulose metabolism were involved in Cd chelation of root cell wall; V-ATPases, ABCC3 and Narmp3 could participated in vacuolar compartmentalization of Cd; PDR1 was responsible for Cd efflux; the organic acid transporters contributed to the absorption of Cd in soil. These genes might have played key roles in kenaf Cd tolerance and Cd accumulation. Moreover, HcZIP2 was identified to be involved in Cd uptake and transport in kenaf. Our findings provide a deeper understanding of the molecular pathways underlying Cd accumulation and detoxification mechanisms in kenaf.

Abstract:To study the molecular mechanism of the hyperaccumulator plant Phytolacca americana against cadmium (Cd) stress, the leaves of P. americana treated with 400 μM Cd for 0, 2, 12, and 24 h were harvested for comparative transcriptome analysis. In total, 110.07 Gb of clean data were obtained, and 63,957 unigenes were acquired after being assembled. Due to the lack of P. americana genome information, only 24,517 unigenes were annotated by public databases. After Cd treatment, 5054 differentially expressed genes (DEGs) were identified. KEGG pathway enrichment analysis of DEGs showed that genes involved in the flavonoid biosynthesis and antenna proteins of photosynthesis were significantly down-regulated, while genes related to the lignin biosynthesis pathway were remarkably up-regulated, indicating that P. americana could synthesize more lignin to cope with Cd stress. Moreover, genes related to heavy metal accumulation, sulfur metabolism and glutathione metabolism were also significantly up-regulated. The gene expression pattern of several key genes related to distinct metabolic pathways was verified by qRT-PCR. The results indicated that the immobilization of lignin in cell wall, chelation, vacuolar compartmentalization, as well as the increase of thiol compounds content may be the important mechanisms of Cd detoxification in hyperaccumulator plant P. americana.

Abstract:A technique for automatically clustering linear envelopes of EMGs (electromyograms) during gait has been developed. It uses a temporal feature representation and a maximum peak matching scheme. This technique provides a viable way to define compact and meaningful EMG waveform features. The envelope matching is performed by dynamic programming, providing qualitatively the largest number of matched peaks and quantitatively a minimum distance measurement. The resulting averaged EMG profiles have low statistical variation and can serve as templates for EMG comparison and further classification.<<ETX>>

Abstract:Trifolium repens (T. repens) can accumulate significant amounts of heavy metal ions, and has strong adaptability to wide environmental conditions, and relatively large biomass, which is considered a potential plant for phytoremediation. However, the molecular mechanisms of T. repens involved in Cd tolerance have not yet been studied in detail. This study was conducted to examine the integrative responses of T. repens exposed to a high-level CdCl2 by investigating the physiological and transcriptomic analyses. The results suggested that T. repens seedlings had a high degree of tolerance to Cd treatment. The roots accumulated higher Cd concentration than leaves and were mainly distributed in the cell wall. The content of MDA, soluble protein, the relative electrolyte leakage, and three antioxidant enzymes (POD, SOD, and APX) was increased with the Cd treatment time increasing, but the CAT enzymes contents were decreased in roots. Furthermore, the transcriptome analysis demonstrated that the differentially expressed genes (DEGs) mainly enriched in the glutathione (GSH) metabolism pathway and the phenylpropanoid biosynthesis in the roots. Overexpressed genes in the lignin biosynthesis in the roots might improve Cd accumulation in cell walls. Moreover, the DEGs were also enriched in photosynthesis in the leaves, transferase activity, oxidoreductase activity, and ABA signal transduction, which might also play roles in reducing Cd toxicity in the plants. All the above, clearly suggest that T. repens employ several different mechanisms to protect itself against Cd stress, while the cell wall biosynthesis and GSH metabolism could be considered the most important specific mechanisms for Cd retention in the roots of T. repens.
