Abstract:Pediatric porcines have widely been used as substitute for children in biomechanical research. Previous studies have used entire piglet cranium when testing their properties. Here, the piglet craniums from the frontal and parietal locations were carefully dissected into spongy and cortical part, and tensile tests at different strain rates were then conducted on these two bone types. It is found that the elastic modulus, yield stress, and ultimate stress of the cortical bone were all significantly higher than those of the spongy bone. The ultimate strains of the cortical and spongy bone were similar. Overall, the effect of the position on the mechanical properties did not reach significance. Cortical bone strength from the frontal location was slightly higher than that obtained from the parietal location; however, spongy bone did not show this location difference. The mechanical properties of both the cortical and spongy bone are significantly strain-rate dependent. Specifically, the elastic modulus, yield stress, and the ultimate stress of the cortical bone increased by approximately 123%, 63%, and 50%, respectively, with strain rates ranging from 0.001 to 10/s. For spongy bone, increases were approximately 128%, 73%, and 77%, respectively. Ultimate strain decreased by approximately 37% and 7% for cortical and spongy bone, respectively. An elastic-plastic constitutive model incorporating with strain rate based on a combined exponential and logarithmic function was proposed and implemented into LS-DYNA through user-defined material. The developed model and the subroutine code successfully simulated the strain-rate characteristics and the fracture process of the bone samples.

Abstract:A finite element (FE) validation and sensitivity study was undertaken on a modern domestic pig cranium. Bone strain data were collected ex vivo from strain gauges, and compared with results from specimen-specific FE models. An isotropic, homogeneous model was created, then input parameters were altered to investigate model sensitivity. Heterogeneous, isotropic models investigated the effects of a constant-thickness, stiffer outer layer (representing cortical bone) atop a more compliant interior (representing cancellous bone). Loading direction and placement of strain gauges were also varied, and the use of 2D membrane elements at strain gauge locations as a method of projecting 3D model strains into the plane of the gauge was investigated. The models correctly estimate the loading conditions of the experiment, yet at some locations fail to reproduce correct principal strain magnitudes, and hence strain ratios. Principal strain orientations are predicted well. The initial model was too stiff by approximately an order of magnitude. Introducing a compliant interior reported strain magnitudes more similar to the ex vivo results without notably affecting strain orientations, ratios or contour patterns, suggesting that this simple heterogeneity was the equivalent of reducing the overall stiffness of the model. Models were generally insensitive to moderate changes in loading direction or strain gauge placement, except in the squamosal portion of the zygomatic arch. The use of membrane elements made negligible differences to the reported strains. The models therefore seem most sensitive to changes in material properties, and suggest that failure to model local heterogeneity in material properties and structure of the bone may be responsible for discrepancies between the experimental and model results. This is partially attributable to a lack of resolution in the CT scans from which the model was built, and partially due to an absence of detailed material properties data for pig cranial bone. Thus, caution is advised when using FE models to estimate absolute numerical values of breaking stress and bite force unless detailed input parameters are available. However, if the objective is to compare relative differences between models, the fact that the strain environment is replicated well means that such investigations can be robust.

Abstract:<h2>Abstract</h2><p>The Göttingen minipig has been used as a surrogate in impact experiments designed to better understand the mechanisms by which mechanical loading induces traumatic brain injury (TBI). However, the relationship between mechanical response and structural morphology of the minipig cranium must be understood relative to the human skull in order to accurately scale any quantitative results, such as injury thresholds, from non-human TBI experiments to the human anatomy. In this study, bone specimens were dissected from the crania of adolescent Göttingen minipigs. These specimens were small cubes that contained the entire thickness of the skull. The microstructure of these skull specimens was quantified at the micron-length scale using micro-computed tomography (micro-CT). The skull was found to be highly porous near the skin-side surface and became less porous nearer the brain-side surface. The skull specimens were then loaded in quasi-static compression to obtain their mechanical response. The surface strain distribution on the specimen face was measured during loading using digital image correlation (DIC). The 2-D strain field formed a gradient of iso-strain bands along the thickness (depth) dimension from the skin-most to brain-most sides of the skull. The variation of the minipig microstructure along the thickness differed significantly from that of the adult human skull; thus the mechanical load transmission through the minipig skull is expected to be quite different from that of the human skull. The objective was to develop the methodology of relating the microstructure, as quantified by the bone volume fraction (BVF), to the mechanical response. The specimen was modeled by discretizing the depth dimension into a series of layers, which enabled the calibration of a power law relating the depth-dependent BVF to the depth-varying modulus. The relationship was used to predict moduli values for the adolescent minipig skull to provide updated, biofidelic parameters for finite element simulations at varying levels of complexity. Moreover, the methodology outlined in this paper can be applied to other skulls with different structural variations, such as the human.</p>

Abstract:Spoofing is a great threat to GNSS (Global Navigation Satellite System) based applications. Many effective spoofing detection methods have been proposed, while few effort is paid to spoofing localization, which is of equally importance. GNSS interference localization is mainly based on TDOA (Time Difference of Arrival) estimations and one widely adopted method to measure the TDOA is based on signal cross-correlation. This paper proposes a differential code phase (DCP) based technique to estimate the spoofing TDOA. The TDOA is estimated as the ratio between DCP and spoofing code frequency. The DCP based TDOA model and its estimation error model are mathematically formulated. Also, the performance comparison between the DCP based and the widely used cross-correlation based TDOA techniques is conducted based on simulation results. Results show the DCP based method can be more accurate in TDOA estimation.
The mechanical and microstructural properties of the pediatric skull

Abstract:The pediatric skull differs drastically from the adult skull in terms of composition, rigidity, and structure. However, there is limited data which quantifies the mechanical properties of the pediatric skull. The lack of mechanical data may inhibit desired pediatric craniofacial surgical outcomes as current methodologies and materials employed for the pediatric population are adapted from those used for adults. In this study, normally discarded parietal bone tissue from eight pediatric craniosynostosis surgery patients (aged 4 to 10 months) was collected during reconstructive surgery and prepared for microstructural analysis and mechanical testing. Up to 12 individual coupon samples of fresh, never frozen tissue were harvested from each specimen and prepared for four-point bending testing to failure. The microstructure of each sample was analyzed using micro-computed tomography before and after each mechanical test. From this analysis, effective geometric and mechanical properties were determined for each sample (n = 68). Test results demonstrated that the pediatric parietal skull was 2.0 mm (±0.4) thick, with a porosity of 36% (±14). The effective modulus of the tissue samples, determined from the initial slope of the sample stress-strain response using Euler beam theory and a nonlinear Ramberg-Osgood stress-strain relationship, was 4.2 GPa (±2.1), which was approximately three times less stiff than adult skull tissue reported in the literature. Furthermore, the pediatric skull was able to bend up to flexural failure strains of 6.7% (±2.0), which was approximately five times larger than failure strains measured in adult skull. The disparity between the measured mechanical properties of pediatric skull tissue and adult skull tissue points towards the need to reevaluate current surgical technologies, such as pediatric cranial surgical hardware, so that they are more compatible with pediatric tissue.
Age-dependent changes in material properties of the brain and braincase of the rat

Abstract:Clinical and biomechanical evidence indicates that mechanisms and pathology of head injury in infants and young children may be different from those in adults. Biomechanical computer-based modeling, which can be used to provide insight into the thresholds for traumatic tissue injury, requires data on material properties of the brain, skull, and sutures that are specific for the pediatric population. In this study, brain material properties were determined for rats at postnatal days (PND) 13, 17, 43, and 90, and skull/suture composite (braincase) properties were determined at PND 13, 17, and 43. Controlled 1 mm indentation of a force probe into the brain was used to measure naive, non-preconditioned (NPC) and preconditioned (PC) instantaneous (G(i)) and long-term (G( infinity )) shear moduli of brain tissue both in situ and in vitro. Brains at 13 and 17 PND exhibited statistically indistinguishable shear moduli, as did brains at 43 and 90 PND. However, the immature (average of 13 and 17 PND) rat brain (G(i) = 3336 Pa NPC, 1754 Pa PC; G( infinity )= 786 Pa NPC, 626 Pa PC) was significantly stiffer (p < 0.05) than the mature (average of 43 and 90 PND) brains (G(i) = 1721 Pa NPC, 1232 Pa PC; G( infinity ) = 508 Pa NPC, 398 Pa PC). A "reverse engineering" finite element model approach, which simulated the indentation of the force probe into the intact braincase, was used to estimate the effective elastic moduli of the braincase. Although the skull of older rats was significantly thicker than that of the younger rats, there was no significant age-dependent change in the effective elastic modulus of the braincase (average value = 6.3 MPa). Thus, the increase in structural rigidity of the braincase with age (up to 43 PND) was due to an increase in skull thickness rather than stiffening of the tissue. These observations of a stiffer brain and more compliant braincase in the immature rat compared with the adult rat will aid in the development of age-specific experimental models and in computational head injury simulations. Specifically, these results will assist in the selection of forces to induce comparable mechanical stresses, strains and consequent injury profiles in brain tissues of immature and adult animals.
Rate- and Region-Dependent Mechanical Properties of Göttingen Minipig Brain Tissue in Simple Shear and Unconfined Compression

Abstract:Traumatic brain injury (TBI), particularly from explosive blasts, is a major cause of casualties in modern military conflicts. Computational models are an important tool in understanding the underlying biomechanics of TBI but are highly dependent on the mechanical properties of soft tissue to produce accurate results. Reported material properties of brain tissue can vary by several orders of magnitude between studies, and no published set of material parameters exists for porcine brain tissue at strain rates relevant to blast. In this work, brain tissue from the brainstem, cerebellum, and cerebrum of freshly euthanized adolescent male Göttingen minipigs was tested in simple shear and unconfined compression at strain rates ranging from quasi-static (QS) to 300 s-1. Brain tissue showed significant strain rate stiffening in both shear and compression. Minimal differences were seen between different regions of the brain. Both hyperelastic and hyper-viscoelastic constitutive models were fit to experimental stress, considering data from either a single loading mode (unidirectional) or two loading modes together (bidirectional). The unidirectional hyper-viscoelastic models with an Ogden hyperelastic representation and a one-term Prony series best captured the response of brain tissue in all regions and rates. The bidirectional models were generally able to capture the response of the tissue in high-rate shear and all compression modes, but not the QS shear. Our constitutive models describe the first set of material parameters for porcine brain tissue relevant to loading modes and rates seen in blast injury.

Abstract:In the early 1960's, the University of Waterloo recognized a growing need in society both for computer specialists and for non-specialists having some familiarity with computers. Programmes initiated at that time have grown so rapidly that computer science courses are now attended by approximately 4000 Waterloo undergraduates, about 1000 of whom may be regarded as computer science majors. Courses have been designed upon the premise that hands-on experience is one of the most effective vehicles for teaching the practice of computer science. Many of the courses require that a student write programmes and run between 100 and 500 jobs through the computer. The objective of providing prompt service to student programmers entailed major operational difficulties for the University Computing Centre as the volume of work approached a level of 1 million student jobs per year.
Development and global validation of a 1-week-old piglet head finite element model for impact simulations

Abstract:Purpose: Child head injury under impact scenarios (e.g. falls, vehicle crashes, etc.) is an important topic in the field of injury biomechanics. The head of piglet was commonly used as the surrogate to investigate the biomechanical response and mechanisms of pediatric head injuries because of the similar cellular structures and material properties. However, up to date, piglet head models with accurate geometry and material properties, which have been validated by impact experiments, are seldom. We aim to develop such a model for future research. Methods: In this study, first, the detailed anatomical structures of the piglet head, including the skull, suture, brain, pia mater, dura mater, cerebrospinal fluid, scalp and soft tissue, were constructed based on CT scans. Then, a structured butterfly method was adopted to mesh the complex geometries of the piglet head to generate high-quality elements and each component was assigned corresponding constitutive material models. Finally, the guided drop tower tests were conducted and the force-time histories were ectracted to validate the piglet head finite element model. Results: Simulations were conducted on the developed finite element model under impact conditions and the simulation results were compared with the experimental data from the guided drop tower tests and the published literature. The average peak force and duration of the guide drop tower test were similar to that of the simulation, with an error below 10%. The inaccuracy was below 20%. The average peak force and duration reported in the literature were comparable to those of the simulation, with the exception of the duration for an impact energy of 11 J. The results showed that the model was capable to capture the response of the pig head. Conclusion: This study can provide an effective tool for investigating child head injury mechanisms and protection strategies under impact loading conditions.
Mechanical characterization of brain tissue in simple shear at dynamic strain rates

Abstract:During severe impact conditions, brain tissue experiences a rapid and complex deformation, which can be seen as a mixture of compression, tension and shear. Moreover, diffuse axonal injury (DAI) occurs in animals and humans when both the strains and strain rates exceed 10% and 10/s, respectively. Knowing the mechanical properties of brain tissue in shear at these strains and strain rates is thus of particular importance, as they can be used in finite element simulations to predict the occurrence of brain injuries under different impact conditions. In this research, an experimental setup was developed to perform simple shear tests on porcine brain tissue at strain rates < 120/s. The maximum measured shear stress at strain rates of 30, 60, 90 and 120/s was 1.15 +/- 0.25 kPa, 1.34 +/- 0.19 kPa, 2.19 +/- 0.225 kPa and 2.52 +/- 0.27 kPa, (mean +/- SD), respectively, at the maximum amount of shear, K = 1. Good agreement of experimental, theoretical (Ogden and MooneyRivlin models) and numerical shear stresses was achieved (p = 0.7866 - 0.9935). Specimen thickness effects (2.0 - 10.0 mm thick specimens) were also analyzed numerically and we found that there is no significant difference (p = 0.9954) in the shear stress magnitudes, indicating a homogeneous deformation of the specimens during simple shear tests. Stress relaxation tests in simple shear were also conducted at different strain magnitudes (10% - 60% strain) with the average rise time of 14 ms. This allowed us to estimate elastic and viscoelastic parameters (mu = 4942.0 Pa and Prony parameters: g1 = 0.520, g2 = 0.3057, tau1 = 0.0264 s, tau2 = 0.011 s) that can be used in FE software to analyze the hyperviscoelastic behavior of brain tissue.
Material Characterization of in Vivo and in Vitro Porcine Brain Using Shear Wave Elasticity

Abstract:Realistic computer simulation of closed head trauma requires accurate mechanical properties of brain tissue, ideally in vivo. A substantive deficiency of most existing experimental brain data is that properties were identified through in vitro mechanical testing. This study develops a novel application of shear wave elasticity imaging to assess porcine brain tissue shear modulus in vivo. Shear wave elasticity imaging is a quantitative ultrasound technique that has been used here to examine changes in brain tissue shear modulus as a function of several experimental and physiologic parameters. Animal studies were performed using two different ultrasound transducers to explore the differences in physical response between closed skull and open skull arrangements. In vivo intracranial pressure in four animals was varied over a relevant physiologic range (2-40 mmHg) and was correlated with shear wave speed and stiffness estimates in brain tissue. We found that stiffness does not vary with modulation of intracranial pressure. Additional in vitro porcine specimens (n = 14) were used to investigate variation in brain tissue stiffness with temperature, confinement, spatial location and transducer orientation. We observed a statistically significant decrease in stiffness with increased temperature (23%) and an increase in stiffness with decreasing external confinement (22-37%). This study determined the feasibility of using shear wave elasticity imaging to characterize porcine brain tissue both in vitro and in vivo. Our results underline the importance of temperature- and skull-derived boundary conditions to brain stiffness and suggest that physiologic ranges of intracranial pressure do not significantly affect in situ brain tissue properties. Shear wave elasticity imaging allowed for brain material properties to be experimentally characterized in a physiologic setting and provides a stronger basis for assessing brain injury in computational models.

Relationships between biochemical bone metabolism indices and morphometric, densitometric and mechanical properties of mandible in 6-month-old pigs

Abstract:Introduction and objective: Mandible is used as a bone model for monitoring bone tissue responses to various factors influencing skeletal homeostasis. Considering the lack of experimental data on interrelationships between bone metabolism indices and morphometric, densitometric and mechanical properties of mandible, the aim of this study was to perform such an evaluation in 6-month-old pigs. Materials and methods: Quantitative computed tomography was used to determine bone volume, mean volumetric bone mineral density, cortical bone density and cortical bone area. Using dual-energy X-ray absorptiometry, bone mineral density and bone mineral content were measured for ramus, body and whole jaw. In the three-point bending test, maximum elastic strength and ultimate strength of jaw was determined. Assessment of calcium (Ca), phosphorus (P), magnesium (Mg), parathormone (PTH), growth hormone (GH), insulin-like growth factor-1 (IGF-1), alkaline phosphatase (ALP), bone-specific alkaline phosphatase (BAP), osteocalcin (OC) and C-terminal telopeptide of collagen type-I (CTX) in blood was performed. Results: Statistically significant correlations in relation to the investigated traits of the jaw were found in the case of ALP, OC, CTX, GH and IGF-1. Significant correlations of ALP activity, OC and IGF-1 concentrations with final body weight were stated (p<0.05). Conclusions: This study shows the highest predictive value of ALP activity determination in relation to assessment of morphological, densitometric and biomechanical properties of mandible. Evaluation of Ca, P, Mg, BAP and PTH has not confirmed its significance for morphological, densitometric and biomechanical properties prediction in the jaw of pigs. ALP activity, OC and IGF-1 concentrations would be prognostic for body weight prediction.
Microstructural Analysis of Porcine Skull Bone Subjected to Impact Loading

Abstract:Skull fracture can be a complex process involving various types of bone microstructure. Finite element analysis of the microscopic architecture in the bone allows for a controlled evaluation of the stress wave interactions, micro-crack growth, propagation and eventual coalescence of trabecular fracture. In this paper, the microstructure and mechanics of small-volume sections of a 6-month-old Gottingen Minipig skull were analyzed. MicroCT scans were used to generate finite element models. Various computational methods were investigated for modeling the intricacies contained within the porous microstructure of the trabecular bone. Pores were explicitly meshed in one method, whereas in the second, a mesh was created from a microCT image-informed mapping algorithm that mapped the trabecular porosity from an image stack to a solid volume mesh of the model. From here, all models were subject to uniaxial compression simulations. The output of the simulations allowed for a detailed understanding of the failure mechanics of the skull structure and allowed for comparison between the methods. Fracture typically occurs in the weakest areas where the bone is highly porous and forms a fracture surface throughout the material, which causes the bone to collapse upon itself.
Influence of strain rate on indentation response of porcine brain

Abstract:Knowledge of brain tissue mechanical properties may be critical for formulating hypotheses about some specific diseases mechanisms and its accurate simulations such as traumatic brain injury (TBI) and tumor growth. Compared to traditional tests (e.g. tensile and compression), indentation shows superiority by virtue of its pinpoint and nondestructive/quasi-nondestructive. As a viscoelastic material, the properties of brain tissue depend on the strain rate by definition. However most efforts focus on the aspect of velocity in the field of brain indentation, rather than strain rate. The influence of strain rate on indentation response of brain tissue is taken little attention. Further, by comparing different results from literatures, it is also obvious that strain rate rather than velocity is more appropriate to characterize mechanical properties of brain. In this paper, to systematically characterize the influence of strain rate, a series of indentation-relaxation tests n = 210) are performed on the cortex of porcine brain using a custom-designed indentation device. The mechanical response that correlates with indenter diameters, depths of indentation and velocities, is revealed for the indentation portion, and elastic behavior of brain tissue is analyzed as the function of strain rate. Similarly, a linear viscoelastic model with a Prony series is employed for the indentation-relaxation portion, wherein the brain tissue shows more viscous and responds more quickly with increasing strain rate. Understanding the effect of strain rate on mechanical properties of brain indentation may be far-reaching for brain injury biomechanics and accurate simulations, but be important for bridging between indentation results of different literatures.

Biomechanics and Modeling in Mechanobiology

Abstract:Traumatic brain injury is a major cause of morbidity in civilian as well as military populations. Computational simulations of injurious events are an important tool to understanding the biomechanics of brain injury and evaluating injury criteria and safety measures. However, these computational models are highly dependent on the material parameters used to represent the brain tissue. Reported material properties of tissue from the cerebrum and cerebellum remain poorly defined at high rates and with respect to anisotropy. In this work, brain tissue from the cerebrum and cerebellum of male Göttingen minipigs was tested in one of three directions relative to axon fibers in oscillatory simple shear over a large range of strain rates from 0.025 to 250 s −1 . Brain tissue showed significant direction dependence in both regions, each with a single preferred loading direction. The tissue also showed strong rate dependence over the full range of rates considered. Transversely isotropic hyper-viscoelastic constitutive models were fit to experimental data using dynamic inverse finite element models to account for wave propagation observed at high strain rates. The fit constitutive models predicted the response in all directions well at rates below 100 s −1 , after which they adequately predicted the initial two loading cycles, with the exception of the 250 s −1 rate, where models performed poorly. These constitutive models can be readily implemented in finite element packages and are suitable for simulation of both conventional and blast injury in porcine, especially Göttingen minipig, models.

Abstract:Understanding the viscoelastic behavior of pediatric brain tissue is critical to interpret how external mechanical forces affect head injury in children. However, knowledge of the viscoelastic properties of pediatric brain tissue is limited, and this reduces the biofidelity of developed numeric simulations of the pediatric head in analysis of brain injury. Thus, it is essential to characterize the viscoelastic behavior of pediatric brain tissue in various loading conditions and to identify constitutive models. In this study, the pediatric porcine brain tissue was investigated in compression with determine the viscoelasticity under small and large strain, respectively. A range of frequencies between 0.1 and 40 Hz was applied to determine frequency-dependent viscoelastic behavior via dynamic mechanical analysis, while brain samples were divided into three strain rate groups of 0.01/s, 1/s and 10/s for compression up to 0.3 strain level and stress relaxation to obtain time-dependent viscoelastic properties. At frequencies above 20 Hz, the storage modulus did not increase, while the loss modulus increased continuously. With strain rate increasing from 0.01/s to 10/s, the mean stress at 0.1, 0.2 and 0.3 strain increased to approximate 6.8, 5.6 and 4.4 times, respectively. The brain compressive response was sensitive to strain rate and frequency. The characterization of brain tissue will be valuable for development of head protection systems and prediction of brain injury.

Abstract:In this study, a statistical model of cranium geometry for 0- to 3-month-old children was developed by analyzing 11 CT scans using a combination of principal component analysis and multivariate regression analysis. Radial basis function was used to morph the geometry of a baseline child head finite element (FE) model into models with geometries representing a newborn, a 1.5-month-old, and a 3-month-old infant head. These three FE models were used in a parametric study of near-vertex impact conditions to quantify the sensitivity of different material parameters. Finally, model validation was conducted against peak head accelerations in cadaver tests under different impact conditions, and optimization techniques were used to determine the material properties. The results showed that the statistical model of cranium geometry produced realistic cranium size and shape, suture size, and skull/suture thickness, for 0- to 3-month-old children. The three pediatric head models generated by morphing had mesh quality comparable to the baseline model. The elastic modulus of skull had a greater effect on most head impact response measurements than other parameters. Head geometry was a significant factor affecting the maximal principal stress of the skull (p = 0.002) and maximal principal strain of the suture (p = 0.021) after controlling for the skull material. Compared with the newborn head, the 3-month-old head model produced 6.5% higher peak head acceleration, 64.8% higher maximal principal stress, and 66.3% higher strain in the suture. However, in the skull, the 3-month-old model produced 25.7% lower maximal principal stress and 11.5% lower strain than the newborn head. Material properties of the brain had little effects on head acceleration and strain/stress within the skull and suture. Elastic moduli of the skull, suture, dura, and scalp determined using optimization techniques were within reported literature ranges and produced impact response that closely matched those measured in previous cadaver tests. The method developed in this study made it possible to investigate the age effects from geometry changes on pediatric head impact responses. The parametric study demonstrated that it is important to consider the material properties and geometric variations together when estimating pediatric head responses and predicting head injury risks.

Abstract:Experimental tests using porcine brainstem samples were performed on a custom designed stress relaxation shear device. Tests were performed dynamically at strain rates >1 s−1, to three levels of peak strain (2.5%-7.5%). The directional dependence of the material properties was investigated by shearing both parallel and transverse to the predominant direction of the axonal fibers. Quasi-linear viscoelastic theory was used to describe the reduced relaxation response and the instantaneous elastic function. The time constants of the reduced relaxation function demonstrate no directional dependence; however, the relative magnitude of the exponential functions and the parameter representing the final limiting value are significantly different for each direction. The elastic function qualitatively demonstrates a dependence on direction. These results suggest that the brainstem is an anisotropic material. The data provides a starting point for analyzing the directionally dependent nature of brainstem material and can serve as a guide in developing new material models for finite element analysis.
