Abstract:The solvation and solubilization of selected anesthetic active pharmaceutical ingredients (bupivacaine, prilocaine, and procaine) in arginine-based deep eutectic solvents are studied using a theoretical approach considering quantum chemistry and classical molecular dynamics. The intermolecular forces between the anesthetics and the solvent are characterized, with particular attention to hydrogen bonding, in terms of strength, topological properties, interaction mechanism, structuring, and dynamic properties of solvation shells. The reported results show the nanoscopic properties that confirm these solvents as suitable materials for anesthetics drug delivery in the liquid phase.
Towards a greater understanding of the deep eutectic phenomenon through examination of the lidocaine-NSAID therapeutic deep eutectic systems

Abstract:Therapeutic deep eutectic solvents (THEDES) have been attracting increasing attention in the pharmaceutical literature as a promising enabling technology capable of improving physicochemical and biopharmaceutical properties for difficult-to-deliver drug compounds. The current literature has explored amide local anaesthetics and carboxylic acid nonsteroidal anti-inflammatories (NSAIDs) as commonly used THEDES formers for their active hydrogen-bonding functionality. However, little is known about what happens within the "deep eutectic" region where a range of binary compositions present simply as a liquid with no melting events detectable across experimentally achievable conditions. There is also very limited understanding of how parent compounds' physicochemical properties could impact upon the formation, interaction mechanism, and stability of the formed liquid systems, despite the significance of these information in dose adjustment, industrial handling, and scaling-up of these liquids. In the current work, we probed the "deep eutectic" phenomenon by investigating the formation and physicochemical behaviours of some chosen lidocaine-NSAID systems across a wide range of composition ratios. Our data revealed that successfully formed THEDES exhibited composition dependent Tg variations with strong positive deviations from predicted Tg values using the Gordon-Taylor theory, suggesting substantial interactions within the formed supramolecular structure. Interestingly, it was found that the parent compound's glass forming ability had a noticeable impact upon such profound interaction and hence could dictate the success of THEDES formation. It has also been confirmed that all successful systems were formed based on charge-assisted hydrogen bonding within their THEDES network, affirming the significant role of partial protonisation on achieving a profound melting point depression. More importantly, the work found that within the "deep eutectic" region there was still an ideal, or thermodynamically preferrable "THEDES point", which would exhibit excellent stability upon exposure to stress storage conditions. The discoveries of this study bring the literature one step closer to fully understanding the "therapeutic deep eutectic" phenomenon. Through correlation between parent reagents' physicochemical properties and the synthesised products' characteristics, we establish a more educated process for the prediction and engineering of THEDES.

Abstract:Hydrophobic deep eutectic solvents (DESs) emerge as candidates to extract organic substrates from aqueous solutions. The DES-aqueous liquid-liquid interface plays a vital role in the extraction ability of hydrophobic DES because the non-bulk structure of molecules at the interface could cause thermodynamic and kinetic barriers. One question is how the DES compositions affect the structural features of the DES-aqueous liquid-liquid interface. We investigate the density profile, dipole moment and hydrogen bonds of eight hydrophobic DES-aqueous liquid-liquid interfaces using molecular dynamics simulations. The eight DESs are composed of four organic compounds: decanoic acid, menthol, thymol, and lidocaine. The simulation results show the variations of dipole moment and hydrogen bond structure and dynamics at the liquid-liquid interfaces. Such variations could influence the extraction ability of DES through adjusting the partition and kinetics of organic substrates in the DES-aqueous biphasic systems.

Abstract:Hydrophobic deep eutectic solvents (DESs) have emerged as excellent extractants. Their performance depends on the heterogeneous hydrogen bond environment formed by multiple hydrogen bond donors and acceptors. An understanding of this heterogeneous hydrogen bond environment can be used to develop principles for designing high-performance DESs for extraction and other separation applications. We investigate the structure and dynamics of hydrogen bonds in eight hydrophobic DESs formed by decanoic acid, menthol, thymol, and Lidocaine using molecular dynamics simulations. The results show the diversity of hydrogen bonds in the eight DESs and their impact on diffusivity and molecular association. Each DES possesses four-six types of hydrogen bonds and one or two of them overwhelm the others in quantity and lifetime. The dominating hydrogen bonds determine whether the DESs are governed by intra- or inter-component associations. The component diffusivity presents an inverse relationship with the hydrogen bond strength.

Abstract:The development of a drug delivery liquid formulation of ester – based tetracaine local anaesthetic is synthesised in silico via the formation of a therapeutic deep eutectic solvent in combination with citral, a natural monoterpenoid. The properties of the obtained liquid system, studied at the nanoscopic level, are derived from a combination of quantum chemistry and classical molecular dynamics simulations. The intermolecular forces (hydrogen bonding) were analysed and their relationship with liquid phase properties were established. Water effect on the liquid phase properties were also considered in terms of competing for hydrogen bonding sites in the components of the developed fluid. The systems proposed in this work were studied using CIT (HBA) 1:1 TET (HBD) molar ratio at temperatures ranging from 288 K to 318 K and 1 bar. The hydrogen bond network developed between the oxygen atom in citral molecules and the –NH group in tetracaine molecules under those conditions, confirmed via minimal cluster analysis, is maintained during the addition of water up to 1.3 wt% as the water molecules do not disrupt the intermolecular hydrogen bond but develop new interactions (hydrogen bonds) with citral and tetracaine monomers presented in the fluid. The reported results probed the suitability of the considered system as an alternative drug delivery method as well as the possibility of developing deep eutectic-based formulation of anaesthetics using natural compounds due to the formation of effective and strong intermolecular hydrogen bonds.

Abstract:Objective Shortening the pulse width to 0.3 millisecond holds neurophysiological and clinical promise of making electroconvulsive therapy (ECT) safer by reducing cognitive adverse effects. The exclusive effects of pulse width on autobiographical and subjective memory are largely unstudied. The aim was to principally investigate during the acute ECT course and at 3 months after ECT autobiographical and subjective memory effects of 0.3-millisecond ultrabrief and 1-millisecond brief-pulse ECT. Methods Patient with severe depression were randomized to 1-millisecond brief-pulse (n = 20) or 0.3-millisecond ultrabrief-pulse (n = 20) right unilateral ECT, both at 6 times the threshold stimulus dose given thrice weekly. Autobiographical Memory Interview (Kopelman) and Squire Subjective Memory Questionnaire were administered at baseline, after 8 sessions, end of the acute course, and 3 months later. Results Early adult semantic memory worsened significantly over time with brief-pulse compared with ultrabrief ECT: F3,88 = 2.60, P = 0.05. There was a significant decline in brief-pulse treatment vis-à-vis a significant improvement with ultrabrief-pulse treatment with childhood semantic memory (P = 0.022), early adult semantic (P = 0.03), and recent semantic memory (P = 0.018) only at 24 hours after the eighth ECT treatment. Conclusions Clinically meaningful and significant improvement in semantic autobiographical memory occurred in ultrabrief treatment vis-à-vis brief-pulse ECT after 8 treatments. Ultrabrief treatment offered a small but significant advantage over 1-millisecond brief-pulse high-dose right unilateral ECT with early adult semantic autobiographical memory, which persisted up to 3 months.

Abstract:The reduced solubility of some active pharmaceutical ingredients (APIs) in water is a limiting factor for the development of new formulations. Therefore, the search of new compatible solvents would be convenient to overcome such limitation. Among this group, eutectic solvents stand out as good candidates. In this work, the systematic application of COSMO-RS as a predictive method to anticipate APIs solubility in systems involving natural eutectic solvents (NAESs) is presented. The methodology has been validated by using a collection of 50 APIs – NAESs systems from the literature. This predictive tool allowed the selection of four NAESs (reported in literature as natural eutectic mixtures) as good candidates to improve lidocaine and caffeine solubility in water and PEG 300 (benchmark solvents). The four NAESs candidates were prepared and experimentally characterized by means of density and viscosity measurements. Density values ranging from 1.0 to 1.3 g/mL and viscosities from 200 to 2,200 cP were found, with glycerol based NAESs as the most viscous systems. Solubility experiments were also carried out to analyze COSMO-RS predictions validity. Betaine: lactic acid and betaine: 1,2-propanediol are good candidates for lidocaine solutions, achieving 2,300 and 572 mg/mL values respectively, much higher than polyethylene glycol, PEG 300, (173 mg/mL), typically used as benchmark solvent. In the case of caffeine, proline:lactic acid was found as the best candidate with 30 mg/mL solubility (higher than water: 23 mg/mL and PEG 300: 18 mg/L). This work permits a systematic analysis of APIs solubility in NAESs (more than 80 systems reported) by using a total predictive tool (COSMO-RS predictions) to anticipate that promising solvents, with high solubilities for lidocaine and caffeine.

Abstract:By selecting L-arginine as the hydrogen bond acceptor (HBA) and 2-hydroxypropyl-β-cyclodextrin (2HPβCD) as the hydrogen bond donor (HBD), deep eutectic solvents (DESs) with various water content were prepared at the 4:1 mass ratio of L-arginine to 2HPβCD with 40 to 60% of water, and were studied for its application in transdermal drug delivery system (TDDS). The hydrogen bond networks and internal chemistry structures of the DESs were measured by attenuated total reflection Fourier transform infrared (ATR-FTIR) and 1 H-nuclear magnetic resonance spectroscopy ( 1 H-NMR), which demonstrated the successful synthesis of DESs. The viscosity of DES was decreased from 10,324.9 to 3219.6 mPa s, while glass transition temperature (Tg) of the DESs was increased from − 60.8 to − 51.4 °C, as the added water was increased from 45 to 60%. The solubility of ibuprofen, norfloxacin, and nateglinide in DES with 45% of water were increased by 79.3, 44.1, and 3.2 times higher than that in water, respectively. The vitro study of transdermal absorption of lidocaine in DESs showed that the cumulative amounts of lidocaine reached 252.4 μg/cm 2 , 226.1 μg/cm 2 , and 286.1 μg/cm 2 at 8 h for DESs with 45%, 50%, and 60% of water, respectively. The permeation mechanism of DES with lower content of water (45%) was mainly by changing the fluidization of lipids, while changing the secondary structure of keratin in stratum corneum (SC) at higher water content (50% and 60%). These nonirritant and viscous fluid like DESs with good drug solubility and permeation enhancing effects have broad application prospect in the field of drug solubilization and transdermal drug delivery system. Abstract:A theoretical and experimental study was performed on natural deep eutectic solvents (NADES) formed by lactic acid-glucose (LGH), citric acid-fructose (CFH) and citric acid-glucose (CGH). The presence of nuclear Overhauser effect (NOE) in the proton nuclear magnetic resonance (1H NMR) spectra of the NADES was studied. The spatial proximity between the NADES components was experimentally confirmed by the detection of multiple NOE effects in the dilutions analyzed. LGH showed the best outcome by partially maintaining its supramolecular structure throughout the dilutions. In order to rationalize the intermolecular interactions generated among the components, a theoretical study was performed using a density functional theory (DFT) computational method. A simplified dimeric model of the NADES was selected in order to achieve a rapid screening of the system searching for interactions between their constituents. In agreement with the experimental evidence, the calculations allowed to confirm the spatial proximity, by finding at least two hydrogen bonds between the components of every NADES.

Abstract:In this work, we report the preparation of novel therapeutic deep eutectic solvents (THEDES) based on choline chloride (CC) and ascorbic acid (AA), in which dexamethasone (DEX) was successfully solubilized. THEDES were synthesized and characterized for thermal behavior and polarized optic microscopy to confirm the formation of a eutectic mixture. The solubility profile of DEX solubilized in THEDES was greatly improved when compared with DEX in powder form. Additionally, the diffusion and permeability of DEX were also slightly increased. The antioxidant properties of AA were maintained up to 6 months when the compound is in THEDES form. Biological tests were also performed, and the results indicate that cell viability varies in a dose-dependent manner. After the half maximal effective concentration (EC50) was determined, a controlled drug delivery system was successfully developed by supercritical fluid sintering of a starch:poly-ε-caprolactone blend doped with THEDES containing solubilized DEX. The present study highlights the potential applicability of THEDES based on CC and AA in the pharmaceutical industry and/or tissue engineering (TE) applications.

Abstract:To overcome the toxic nature of organic solvents, scientific interest in the use of green solvents, particularly natural deep eutectic solvents (NADES), has increased over the past decade, leading to new applications in the food, nutraceutical, pharmaceutical, and cosmetic industries. Understanding the physicochemical properties and molecular interactions of NADES is essential for uncovering new potential applications in these fields. In this study, several lactic and citric acid–based NADES, as well as chloride choline- and urea-based NADES, were evaluated for their physicochemical properties, including density, pH, viscosity, conductivity, and refractive index. Additionally, nuclear magnetic resonance (NMR), and in particular nuclear Overhauser enhancement spectroscopy (NOESY), was employed to investigate the intermolecular interactions between the NADES components to confirm the formation of the eutectic mixture. The extraction efficiency of the confirmed NADES was tested for extracting polyphenols as a proof of concept to highlight their relationship with the measured properties. Lactic and choline chloride–based NADES provided the highest extraction yields. These results were also compared with the predicted extraction capabilities of each NADES provided by the COSMO-RS software.

Abstract:Deep eutectic solvents (DES) are environmentally friendly solvents with the potential to dissolve bioactive compounds without affecting their characteristics. DES has special qualities that can be customized to meet the unique characteristics of a biomolecule/active pharmaceutical ingredient (API) in accordance with various therapeutic needs, providing a reliable approach in opening the door for the creation of cutting-edge drug formulations by resolving solubility issues in pharmaceutics. This study outlines newly developing approaches to solve the problem of inefficient API extraction due to poor solubility. These emerging strategies also have the capacity to alter the chemical and physical stability of API, which triggers drug's shelf life and their possible health benefits. It is anticipated that the highlighted methods and processes will be developed to capitalize on the DES potential to improve drug solubility and delivery in the pharmaceutical sector.

Abstract:Modern strategies to deliver drugs efficiently via less toxic non-aqueous carriers like deep eutectic solvents (DES) has grown tremendously. Herein, we develop several synthetic and natural DES to improve solubility of poorly soluble drug, docetaxel (DTX). Menthol:Thymol-based natural DES showed 1500-folds higher solubility for DTX than that in water. FTIR-spectroscopy confirmed reduced crystallinity and stable encapsulation of DTX in DES through formation of hydrogen-bond. DTX-DES was developed further into self-emulsified formulation that upon dilution formed nano-sized globules (<200 nm) and released 20-folds higher DTX than pure drug thereby highlighting the potential of DES for designing formulations of poorly soluble drugs.

Abstract:Deep eutectic solvents (DESs) are commonly used in pharmaceutical applications as excellent solubilizers of active substances. This study investigated the tuning of ibuprofen and ketoprofen solubility utilizing DESs containing choline chloride or betaine as hydrogen bond acceptors and various polyols (ethylene glycol, diethylene glycol, triethylene glycol, glycerol, 1,2-propanediol, 1,3-butanediol) as hydrogen bond donors. Experimental solubility data were collected for all DES systems. A machine learning model was developed using COSMO-RS molecular descriptors to predict solubility. All studied DESs exhibited a cosolvency effect, increasing drug solubility at modest concentrations of water. The model accurately predicted solubility for ibuprofen, ketoprofen, and related analogs (flurbiprofen, felbinac, phenylacetic acid, diphenylacetic acid). A machine learning approach utilizing COSMO-RS descriptors enables the rational design and solubility prediction of DES formulations for improved pharmaceutical applications.

Abstract:Solubility is a major concern for many active pharmaceutical ingredients (APIs) and drugs. Owing to their better solubility, biodegradability and low toxicity of APIs and drugs, eutectic mixtures are emerging as the system to enhance their solubility with no loss of biological activity. In the present work, authors have taken camizestrant, deucravacitinib, and iclepertin as the active pharmaceutical ingredients (API) because they have poor solubility in water. Authors have designed eutectic mixtures (EMs) of the selected APIs with the amino acids (aspartic acid, lysine, phenylalanine, and proline) and studied for their physiochemical properties in default and water using density functional theory calculations, performed by Gaussian. The solubility of the APIs, amino acids and the designed eutectic mixtures of APIs have been determined and discussed using the dipole moment. The results showed that the solubility of the designed eutectic mixtures have better aqueous solubility than the APIs alone. Further, molecular dynamics (MD) simulations were done for APIs and EMs to study their stability and interactions.

Abstract:Deep eutectic solvents (DESs) have emerged as an alternative to both common organic solvents and ionic liquids (ILs). DESs share physicochemical properties with ILs such as low vapor pressure, high thermal stability, while offering advantages such as low toxicity, lower cost, and ease of preparation. Moreover, DESs are attractive candidates for electrochemical applications due to their large voltage windows and solubility properties. DESs as a solvent class share a general composition of a hydrogen bond donor (HBD), typically a polyol, amide, or acid, and a hydrogen bond acceptor (HBA), usually a quaternary ammonium or phosphonium salt. At a specific molar composition of a HBD and HBA, the DES forms a eutectic mixture resulting in a large melting point depression. Despite being widely studied, the microscopic structures of DESs have remained largely uncharacterized. Herein, we present a multitechnique NMR study of ethaline (ethylene glycol + choline chloride). Pulsed field gradient diffusion, nuclear Overhauser effect spectroscopy (NOESY), 13 C NMR relaxation experiments are deployed to sample a range of frequencies and modes of motion of the polyol and choline components of the DES. Generally, translational and rotational diffusion of polyols are more rapid than those of choline while short-range rotational motions observed from 13 C relaxation indicate slow local motion of ethylene glycol at low choline chloride (ChCl) content. Additionally, we have investigated the solubility properties of several lithium salts (LiTFSI, LiFSI, LiBF 4 , and LiOAc) in these DESs and utilized the same suite of NMR techniques to understand how they act as solvents. One interesting observation is that the Li + diffusivity exceeds that of the counterion for TFSI - , but not for FSI - . Finally, large changes in the structural organization of DESs that result from the presence of lithium salts may serve as a guide to the design of a new class of electrolytes for lithium-ion batteries.

Abstract:This study investigates the use of aqueous eutectic mixtures to enhance the solubility of carvedilol, a drug with low solubility and bioavailability. It was found that mixtures of citric acid and choline chloride increased the maximum solubility. Additionally, the stability of carvedilol in these mixtures was demonstrated. These findings contribute to the sustainable development of liquid medications. Carvedilol is used to treat chronic hypertension, chronic stable angina pectoris and concomitant treatment heart failure and class II of the Biopharmaceutics Classification System (low solubility and poor bioavailability). This drug is only formulated as tablets, and liquid oral formulations are not available on the market. In this work, several aqueous eutectic mixtures formed by xylitol, citric acid, sorbitol and glucose (HBD) and choline chloride as HBA have been used to improve the solubility. Additionally, SwissADME was used to obtain physicochemical and pharmacokinetic parameters to further knowledge on the ADME (absorption, distribution, metabolism and excretion) parameters for carvedilol. The results reveal that the solubility of carvedilol increases as the content of water decreases, and the mixture formed by citric acid and choline chloride (CACh10) was the most soluble. The stability study shows that carvedilol in DES is stable during the test time, as the maximum and minimum concentration losses were 19.0 % and 0.2 %, respectively. These results help advance the green and sustainable development of new medicines in the liquid state.

Abstract:Solvents play key roles in designing drug delivery systems (DDSs). They are used as the reaction media in the preparation of DDSs and as vehicles for delivery of problematic drugs. The number of pharmaceutically acceptable solvents is limited and developing new green ones is of a great of interest. A deep eutectic solvent (DES) is a room temperature liquid composed of a mixture of hydrogen bond acceptor and hydrogen bond donor. DESs exhibit interesting physical and chemical characteristics such as low vapor pressure, non-inflammability, chemically tailorable, solvency power for a wide range of solutes, and water non-reactivity. Furthermore, they can be easily prepared from low toxicity, readily available, and inexpensive constituents. Due to these properties, DESs have attracted growing attention as green solvents in different areas of science from chemistry to material engineering and biology. In this review, after a brief introduction about DESs, we provide an overview about current advances made over the last decade in utilizing of DESs for solubilization of water insoluble drugs, transdermal drug delivery, inorganic nanoparticle synthesis, and designing polymeric and self-assembled drug carriers. This paper also discusses main challenges and limitations of DESs that should be considered before using of them.
Exploring Deep Eutectic Solvents as Pharmaceutical Excipients: Enhancing the Solubility of Ibuprofen and Mefenamic Acid

Abstract:Objectives: The study explores the potential of various deep eutectic solvents (DESs) to serve as drug delivery systems and pharmaceutical excipients. The research focuses on two primary objectives: evaluating the ability of the selected DES systems to enhance the solubility of two poorly water-soluble model drugs (IBU and MFA), and evaluating their physicochemical properties, including density, viscosity, flow behavior, surface tension, thermal stability, and water dilution effects, to determine their suitability for pharmaceutical applications. Methods: A range of DES systems containing pharmaceutically acceptable constituents was explored, encompassing organic acid-based, sugar- and sugar alcohol-based, and hydrophobic systems, as well as menthol (MNT)-based DES systems with common pharmaceutical excipients. MNT-based DESs exhibited the most significant solubility enhancements. Results: IBU solubility reached 379.69 mg/g in MNT: PEG 400 (1:1) and 356.3 mg/g in MNT:oleic acid (1:1), while MFA solubility peaked at 17.07 mg/g in MNT:Miglyol 812®N (1:1). In contrast, solubility in hydrophilic DES systems was significantly lower, with choline chloride: glycerol (1:2) and arginine: glycolic acid (1:8) showing the best results. While demonstrating lower solubility compared to the MNT-based systems, sugar-based DESs exhibited increased tunability via water and glycerol addition both in terms of solubility and physicochemical properties, such as viscosity and surface tension. Conclusions: Our study introduces novel DES systems, expanding the repertoire of pharmaceutically acceptable DES formulations and opening new avenues for the rational design of tailored solvent systems to overcome solubility challenges and enhance drug delivery.
