Abstract:Objective: This study aimed to evaluate the effect of simvastatin-hydroxyapatite scaffolds in the restoration of mandibular defects.Method: 24 New Zealand white rabbits were used in the study.A 10 mm × 4 mm × 3 mm defect was manufactured in the mandible body of each rabbit.The animals were randomly divided into two groups: group A(n=12) the defect was restored by a composite of Simvastatin+BMSCs+nano HA scaffolds;group B(n=12),defects were restored by BMSCs-nano HA scaffolds.3 animals in each group were sacrificed at 2,4,8,and 12 weeks after restoration.The specimen were underwent line imaging analysis,histological observation,and scanning electron microscopy(SEM) examination. Result: Imaging examination and histological staining results showed that group A healing degree,bone and its quality is superior than group B.SEM showed good histocompatibility and better absorption of the compound material in group A. Through the statistical analysis,the bone mineral density was significantly higher in group A than group B(P 0.05). Conclusion: The results suggest that Simvastatin + BMSCs + nano HA scaffold material have a higher osteogenic potential.

Abstract:Objective. Recent studies demonstrate that simvastatin stimulates bone formation, suggesting the potential application in dental implantology. In this study, our lab developed a simvastatin-loaded titanium porous surface. The aim was to investigate the effect of simvastatin-loaded titanium surfaces on the promotion of osteogenesis in preosteoblasts (MC3T3-E1) in vitro.Study design. The control group consisted of cells cultured on titanium disks without any intervention for different time intervals (4, 7, and 14 days), and the experimental groups (simvastatin-loaded groups) consisted of cells cultured on titanium disks that were preincubated in varying concentration (10(-7) mol/L, 10(-6) mol/L, 10(-5) mol/L, and 10(-4) mol/L) of simvastatin for the same time intervals of the control group. Alkaline phosphatase (ALP) activity, type I collagen synthesis, and osteocalcin release were used to measure the cellular osteoblastic activities.Results. All simvastatin-loaded groups showed increased ALP activity compared with the control group at every time point, especially the 10(-7) mol/L group, which significantly increased the activity almost fourfold at 4 days (P < .05). In the type I collagen synthesis assay, all simvastatin-loaded groups showed an increase, and the effect was inverse dose dependent (maximal at 10(-7) mol/L). This stimulatory effect of simvastatin was also observed in the osteocalcin release assay (P < .05; at 10(-7) mol/L, 10(-6) mol/L, maximal at 10(-7) mol/L).Conclusion. These results indicate that simvastatin-loaded porous implant surfaces promote accelerated osteogenic differentiation of preosteoblasts, which have the potential to improve the nature of osseointegration. Effects of Simvastatin-Loaded Nanomicelles on the Early Preservation of Tooth Extraction Sites

Abstract:Object: The present study intended to evaluate the effect of simvastatin-loaded nanomicelles (SVNs) on promoting new bone formation and reducing alveolar ridge resorption at the tooth extraction sites at the early healing of the extraction sockets. Methods: SVNs were synthesized using a dialysis method. The rabbit tooth extraction model was established, SVNs and simvastatin (SV) were loaded on gelatin sponge and inserted into the extraction socket. CBCT scans were performed at 0, 2, and 4 weeks postoperatively to evaluate bone formation and alveolar ridge absorption in the extraction sockets. And all the animals were sacrificed and the mandibles were harvested. And HE staining and Masson staining were used for histological evaluation of the bone formation in the extraction sockets. Results: Radiographic evaluation showed that compared with the blank control group, at 2 and 4 weeks after extraction, SVNs increased the new bone density in the extraction sockets by 75.7% and 96.5%, and reduced the absorption rate of alveolar ridge length at the extraction sites by 60.8% and 49.1%, respectively. Histological evaluation showed that SVNs significantly improved the maturation of new bone tissue in the extraction sockets. Conclusion: SVNs can significantly accelerate healing and effectively reduce the absorption of alveolar ridge at the extraction sites in the early stage of tooth extraction socket healing.
Bone Responses to Simvastatin-Loaded Porous Implant Surfaces in an Ovariectomized Model.

Abstract:Purpose: The purpose of this study was to investigate bone responses to simvastatin-loaded porous implant surfaces in an ovariectomized model. Materials and Methods: Roughened implants were divided into a control group (n = 32), test group 1 (n = 32), and test group 2 (n = 32). Test implants were immersed into 10-7 mol/L (test group I) or 10-6 mol/L (test group 2) simvastatin solutions for drug adsorption onto implant surfaces. Forty-eight ovariectomized rats randomly received an implant in each tibia. One, 2, 4, and 12 weeks later, the tibiae were retrieved and prepared for histomorphometric evaluation. Bone-to-implant contact and bone area around the implant were determined, and histologic observations were made. Results: New bone formation on test implant surfaces was seen after 1 week, while it was seen on the control implant surface after 2 weeks. There was more bone tissue and bone-to-implant contact along the test implant surfaces than along the control implant surface. The test group 1 and test group 2 implants showed significantly greater bone area and bone-to-implant contact compared to the control implant at all observed time points (P<.05). No differences were found between the two types of test group implants after 1, 2, 4, and 12 weeks (P>.05). Osteogenic effect in rat osteoporosis model and femur defect model by intraosseous administration of simvastatin microcrystals

Abstract:Background: Currently, drugs for local bone formation are very limited in clinic. Simvastatin is one of most potential translational drugs by repositioning for its significant osteogenic effect and its history as a safe lipid-lowering medicine. In this study, we prepared the pre-clinical simvastatin microcrystals which could be potentially translated and industrialized. Methods: Simvastatin microcrystals were made by wet media milling method. Characterization of microcrystal particles were evaluated by laser particle size analyzer and high-performance liquid chromatography (HPLC). Furthermore, we verified the osteogenic effect in rat ovariectomy(OVX)-induced osteoporosis model and femur defect model. For osteoporosis model, we delivered simvastatin microcrystals to tibia with poloxamer hydrogel by intraosseous injection. Bone mineral density (BMD) and ultimate force were assessed after treatment. For femur defect model, simvastatin microcrystal was incorporated in clinically used calcium phosphate cements (CPCs) as an implant. Histology and μCT were used for evaluation of the healing. Results: The D 10 , D 50, and D 90 showed size of the most particles ranged from 0.226-3.425 μm. The mean particle size of the microcrystals is determined as 1.365 μm which demonstrated a successful preparation. For osteoporosis model, BMD and the ultimate force of the treated tibia were significantly improved in osteoporosis rat after injection of 0.5 mg and 1 mg simvastatin microcrystals compared with OVX or 0 mg groups. There were no differences observed in BMD and ultimate force between 0.5 mg, 1 mg simvastatin microcrystals group and bone morphogenetic protein (BMP) 5 ug group. For femur defect model, quantitative analysis of bone regeneration by μCT showed bone volume/tissue volume (BV/TV), trabecular thickness (Tb.Th), trabecular number (Tb.N) were significantly increased in simvastatin microcrystals 50ug (SIM-C 50 ug), SIM-C 100 ug and BMP 5 ug group compared with CPC vehicle group. Trabecular separation (Tb.Sp), and cement volume/tissue volume (CV/TV) were decreased in SIM-C 50 ug, SIM-C 100 ug and BMP 5 ug groups compared with CPC vehicle group. Semiquantitative scale for histology assessment further demonstrated a higher bone regeneration score in drug loaded groups compared with CPC group.Conclusions: Our study shows that simvastatin microcrystals were successfully prepared by wet media milling method and the microcrystals can promote bone formation by local delivery by poloxamer hydrogel or CPC, which is of great translational potential.
In Vitro And In Vivo Evaluation Of The Osteogenic Ability Of Implant Surfaces With A Local Delivery Of Simvastatin

Abstract:Purpose: This study established local delivery with a calcium phosphate (CaP) coating and investigated effects of delivery on implant osseointegration by in vitro and in vivo experiments. Material and Methods: Simvastatin was prepared onto titanium surfaces with varying concentration (10(-7), 10(-6), 10(-6), and 10(-4) moVL). Surface characteristics were performed by field-emission scanning electron microscope (FSEM), x-ray diffractometer (XRD), and fourier transform infrared spectroscopy (FTIR). Alkaline phosphatase activity (ALP) and osteocalcin release were used to measure osteoblastic activities. Ovariectomized rats randomly received control and test implants in both tibiae. After 4 and 12 weeks of implantation, the tibiae were retrieved and prepared for histomorphometric evaluation. Results: FSEM observation showed that the size of flakes decreased with an increase of simvastatin concentrations. XRD and FUR examinations demonstrated that all coatings were composed of octacalcium phosphate (OCP). Simvastatin-loaded titanium surface had an increased effect on ALP activities at different concentrations on day 4 and day 7, and only the 10(-6) moVL group showed significant differences on day 14 (P < .05). The 10(-6) moVL group showed significant expression of osteocalcin (P < .05). Test implants (10-6 moVL) showed a significantly greater bone area and boneimplant contact compared to control implants during the observation periods (P < .05). Conclusions: It was concluded that the local delivery of simvastatin was established onto implant surfaces using the biomimetic CaP coating and could improve osteoblast function and implant osseointegration in ovariectomized rats.

Abstract:Gellan gum is obtained from the bacterium Sphingomonas elodea and is a polysaccharide with carboxylic acid functional groups. The goal of this project was to investigate the osteoinductive effect of local administration of calcitonin through an injectable scaffold of gellan gum containing salmon calcitonin loaded in silsesquioxane nanoparticles, hydroxyapatite, and platelets rich plasma. The femur of rats was defected by creating a 2 × 5 mm 2 hole using an electric drill. The defect was filled with an injectable hydrogel scaffold composed of gellan gum enriched with salmon calcitonin loaded in silsesquioxane nanoparticles, hydroxyapatite, platelets rich plasma, and then the radiologic images were taken. Bone densitometry and the histologic studies were carried out by Hematoxylin & Eosin test. Biochemical analysis was done to measure the serum alkaline phosphatase (ALP), calcium, and calcitonin concentration. Healing of the bone defects and bone densitometry in the treated group by calcitonin-loaded scaffold was significantly higher ( p < 0.05) and bone formation occupied 75% of the defect was greater than in other groups. Serum ALP and calcium levels in the scaffold-loaded calcitonin group were more than in the other groups ( p < 0.05). The osteogenic marker genes also increased significantly ( p < 0.05) with free calcitonin and the scaffold. Gellan gum-based scaffold loaded with calcitonin may be considered a promising local treatment to progress bone formation in repairing skeletal injuries.

Controlled release of simvastatin-loaded thermo-sensitive PLGA-PEG-PLGA hydrogel for bone tissue regeneration: in vitro and in vivo characteristics

Abstract:Reports on the local delivery of drug loaded injectable hydrogels for bone regeneration are currently limited. This study assessed the effect of controlled simvastatin (SIM) release from a thermo-sensitive hydrogel in vitro and in vivo. We successfully manufactured and evaluated thermo-sensitive poly(d,l-lactide-co-glycolide)-poly(ethylene glycol)-poly(d,l-lactide-co-glycolide) triblock copolymers (PLGA-PEG-PLGA) loaded with SIM. The osteogenic effect of this hydrogel was tested in vitro and in vivo. MC-3T3 E1 cells proliferation and osteoblastic differentiation was analyzed after cultivation with the hydrogel extracts. Cells co-cultured with SIM/PLGA-PEG-PLGA extracts showed an increase in mineralization and osteogenic gene expression compared to the other two groups. Additionally, the characteristics of this composite in vivo were demonstrated using a rat bone defect model. The bone defects injected with SIM/PLGA-PEG-PLGA hydrogel showed increased new bone formation compared to samples treated with PLGA-PEG-PLGA and control samples. The results of this study suggest that SIM/PLGA-PEG-PLGA might provide potential therapeutic value for bone healing.
Osteogenic effects in a rat osteoporosis model and femur defect model by simvastatin microcrystals

Abstract:<p>Simvastatin is a translational drug that may be used to induce local bone formation. In this study, simvastatin microcrystals were made by a wet media milling method, and then we verified the osteogenic effect of the microcrystals in rat ovariectomy (OVX)–induced osteoporosis and femur defect models. For the osteoporosis model, we delivered simvastatin microcrystals to the tibia with poloxamer hydrogels via an intraosseous injection. Bone mineral density and the ultimate force of the treated tibia were significantly improved after injection of simvastatin microcrystals at 0.5 and 1 mg compared with the OVX or 0‐mg control groups. For the femur defect model, simvastatin microcrystals were incorporated in clinically used calcium phosphate cements (CPCs) as an implant. Quantitative analysis of bone regeneration by microcomputed tomography (μCT) showed improved bone morphology with simvastatin microcrystals at 50 and 100 μg, compared with the CPC vehicle. A semiquantitative scale for histology assessment further demonstrated a higher bone regeneration score in the drug‐loaded groups. Our study shows that simvastatin microcrystals can promote bone formation by local delivery using a poloxamer hydrogel or CPC, which may be translationally useful.</p>

Abstract:Nonunion of fractured bones is a common clinical problem for orthopedic surgeons. This study aimed to investigate the effects of simvastatin locally applied from calcium sulfate (CS) combined with a mesenchymal stem cell (MSC) sheet on fracture healing. In vitro, the proliferation and differentiation of rat bone marrow-derived MSCs stimulated by simvastatin were investigated. In vivo, an osteotomy model was made in rat tibia, and fractured tibias were treated with CS, CS/simvastatin, CS/MSC sheet or simvastatin-loaded CS with MSC or untreated (control). Tibias were harvested at 2 or 8 weeks and underwent real-time quantitative polymerase chain reaction, x-ray, micro-CT and histological analysis. The expression levels of bone morphogenetic protein 2, alkaline phosphatase, osteocalcin, osteoprotegerin and vascular endothelial growth factor of simvastatin-induced MSCs increased with the concentrations of the simvastatin, significantly higher than those in the MSCs group. At 2 weeks, the CS/simvastatin/MSC sheet group showed significantly higher expressions of bone morphogenetic protein 2, alkaline phosphatase, osteocalcin, osteoprotegerin and vascular endothelial growth factor, with more callus formation around the fracture site compared with the other four groups. At 8 weeks, complete bone union was obtained in the CS/simvastatin/MSC sheet group. By contrast, newly regenerated bone tissue partially bridged the gap in the CS/simvastatin group and the CS/MSC sheet group; the control and CS group showed nonunion of the tibia. These results show that both simvastatin and the MSC sheet contributed to the formation of new bone and that the tibia fracture was completely healed by transplantation of the MSC sheet with locally applied simvastatin. Such MSC sheet with locally applied simvastatin might contribute to the treatment of fractures, bone delayed unions or nonunions in clinical practice.
Combination of Simvastatin, Calcium Silicate/gypsum, and Gelatin and Bone Regeneration in Rabbit Calvarial Defects

Abstract:The present study was performed to determine whether simvastatin improves bone regeneration when combined with calcium silicate/gypsum and gelatin (CS-GEL). The surface morphology was determined using field-emission scanning electron microscopy (FSEM). Degradation in vitro was evaluated by monitoring the weight change of the composites soaked in phosphate buffered saline (PBS). Drug release was evaluated using high-performance liquid chromatography (HPLC). Cytotoxicity testing was performed to assess the biocompatibility of composites. Four 5 mm-diameter bone defects were created in rabbit calvaria. Three sites were filled with CS-GEL, 0.5 mg simvastatin-loaded CS-GEL (SIM-0.5) and 1.0 mg simvastatin-loaded CS-GEL (SIM-1.0), respectively, and the fourth was left empty as the control group. Micro-computed tomography (micro-CT) and histological analysis were carried out at 4 and 12 weeks postoperatively. The composites all exhibited three-dimensional structures and showed the residue with nearly 80% after 4 weeks of immersion. Drug release was explosive on the first day and then the release rate remained stable. The composites did not induce any cytotoxicity. The results in vivo demonstrated that the new bone formation and the expressions of BMP-2, OC and type I collagen were improved in the simvastatin-loaded CS-GEL group. It was concluded that the simvastatin-loaded CS-GEL may improve bone regeneration.
Influence of simvastatin-loaded implants on osseointegration in an ovariectomized animal model.

Abstract:The success of bone implants in the presence of osteoporosis is limited by lack of osseointegration between the implant and the natural bone. This study applied an electrochemical process to deposit simvastatin-nanohydroxyapatite (HA) coatings on porous implant surfaces and investigated the effects of these simvastatin-HA coatings on implant surfaces in an animal model of osteoporosis. In this study, simvastatin-HA coated implants were inserted into the tibia of osteoporotic rats. After 2, 4, and 12 weeks, tissue was retrieved for histomorphometric evaluation. The results indicated that the simvastatin-HA coatings increased bone-implant contact and new bone formation around implant surfaces. In conclusion, implants loaded with simvastatin by an electrochemical process improved implant osseointegration in osteoporotic rats. Furthermore, the increased concentration of simvastatin could affect the osseointegration, but the dose-effects also need further investigation.
Single-dose Local Simvastatin Injection Improves Implant Fixation Via Increased Angiogenesis and Bone Formation in an Ovariectomized Rat Model.

Abstract:BACKGROUND:Statins have been reported to promote bone formation. However, taken orally, their bioavailability is low to the bones. Implant therapies require a local repair response, topical application of osteoinductive agents, or biomaterials that promote implant fixation.MATERIAL/METHODS:The present study evaluated the effect of a single local injection of simvastatin on screw fixation in an ovariectomized rat model of osteoporosis.RESULTS:Dual-energy X-ray absorptiometry, micro-computed tomography, histology, and biomechanical tests revealed that 5 and 10 mg simvastatin significantly improved bone mineral density by 18.2% and 22.4%, respectively (P<0.05); increased bone volume fraction by 51.0% and 57.9%, trabecular thickness by 16.4% and 18.9%, trabeculae number by 112.0% and 107.1%, and percentage of osseointegration by 115.7% and 126.3%; and decreased trabeculae separation by 34.1% and 36.6%, respectively (all P<0.01). Bone mineral apposition rate was significantly increased (P<0.01). Furthermore, implant fixation was significantly increased (P<0.05), and bone morphogenetic protein 2 (BMP2) expression was markedly increased. Local injection of a single dose of simvastatin also promoted angiogenesis. Vessel number, volume, thickness, surface area, and vascular volume per tissue volume were significantly increased (all P<0.01). Vascular endothelial growth factor (VEGF), VEGF receptor-2, von Willebrand factor, and platelet endothelial cell adhesion molecule-1 expression were enhanced.CONCLUSIONS:A single local injection of simvastatin significantly increased bone formation, promoted osseointegration, and enhanced implant fixation in ovariectomized rats. The underlying mechanism appears to involve enhanced BMP2 expression and angiogenesis in the target bone.

Abstract:Objective To investigate that locally applied simvastatin promotes bone healing by recruiting autogenous bone marrow stromal cells(BMSC)to the defect.Methods Simvastatin(5 mg)compounded with polylactic acid(PLa,20 mg)or only PLA(20 mg)was dissolved in acetone(200μl)to prepare locally applied materials(simvastatin-PLA scaffolds,PLA scaffolds).Green fluoresent protein(GFP)-BMSCs were transplanted into sixteen Sprague Dawley rats through the tail vein as tracer cells.After 48 hours,critical-sized calvarial defects(=5 mm)were surgically created on rats and implanted with simvastatin-PLA scaffolds(n=8)or PLA scaffolds(n=8)as control.Fourteen days later,GFP signals in the defect were detected using a fluorescence-based small animal imaging system and rat calvarial bones were sliced and observed under a fluorescence microscope(n=4).The expression of bone morphogenetic protein-2(BMP-2)in the defect area was detected by immunohistochemistry staining(n=4).Results Intense GFP signal was detected in the rat defect region 14 days after simvastatin implantation,while tenuous GFP signal was presented in the control group.Frozen sections histology of the defect region confirmed the existence of GFP-BMSCs in the simvastatin group under fluorescence microscopy.The expression of BMP-2 was up-regulated in the simvastatin group.Conclusions Locally applied simvastatin can recruit autogenous osteogenic stem cells to participate bone healing and BMP-2 may mediate the process of recruitment.
Calvarial Defect Healing by Recruitment of Autogenous Osteogenic Stem Cells Using Locally Applied Simvastatin.

Abstract:Local statins implant has been shown to promote bone healing, the underlying mechanisms are unclear. The purpose of this study was to test the effect of local simvastatin implant on bone defect healing; to evaluate the mobilization, migration, and homing of bone marrow-derived mesenchymal stem cells (BMSCs) and endothelial progenitor cells (EPCs) induced by simvastatin. We found that local simvastatin implant increased bone formation by 51.8% (week 6) and 64.8% (week 12) compared with polyglycolic acid controls (P < 0.01), as verified by X-ray, CT, and histology. Simvastatin increased migration capacity of BMSCs and EPCs in vitro (P < 0.05). Local simvastatin implant increased mobilization of EPCs to the peripheral blood by 127% revealed by FACS analysis (P < 0.01), and increased osteogenic BMSCs to the peripheral blood dramatically revealed by Alizarin Red-S staining for mineralized nodules formation. Pre-transplanted GFP-transfected BMSCs as a tracing cell and bioluminescence imaging revealed that local simvastatin implant recruited GFP-labeled BMSC. Also, local simvastatin implant induced the HIF-1α and BMP-2 expression. In conclusion, local simvastatin implantation promotes bone defect healing, where the underlying mechanism appears to involve the higher expression of HIF-1α and BMP-2, thus recruit autogenous osteogenic and angiogenetic stem cells to the bone defect area implanted with simvastatin.
Stimulatory Effects of Simvastatin on Bone Regeneration of the Expanded Suture in Rats.

Abstract:Simvastatin belongs to the family of statins and is found to have some osteopromotive properties in recent years. The aim of the present study was to investigate the potential effects of simvastatin on bone formation of the expanded mid-palatal suture of rats. Forty-five Wistar rats were randomly divided into three groups: control (C), expansion (EP), and expansion plus simvastatin (ES) groups. Rats in the ES group were administrated with simvastatin (20 mg/kg/d body weight). According to the schedule of sacrifice (days 3, 7 and 14), the suture width and bone volume changes of the region of interest (ROI) were detected by micro-computed tomography during RME. Besides, morphological changes and bone morphogenetic protein 2 (BMP-2) expression in the mid-palatal suture were observed by hematoxylin and eosin (HE) and immunohistochemical staining. Kruskal-Wallis one-way analysis of variance (ANOVA) and LSD method were applied to analyze the data at P<0.05 level. By the RME appliance, the suture was successfully widened. On days 7, 14, the bone volume of ROI in the ES group was more than that in the EP group (P<0.05). Besides, histological examinations also demonstrated that more bone regeneration and capillaries in the suture in the ES group were observed than that in the EP group. The BMP-2 expression in the ES group was more (P<0.05) than that in the EP and C groups on days 3, 7, 14. Consequently, those findings showed that simvastatin can induce a favorable effect on bone regeneration in the mid-palatal suture of rats during RME.
Injectable in Situ Shape-Forming Osteogenic Nanocomposite Hydrogel for Regenerating Irregular Bone Defects

Abstract:The in situ forming injectable hydrogels are appealing for irregular bone defects because of the ease of administration and the addition of ceramics, molecules, and proteins into the hydrogel. We have developed in situ shape-forming hydrogel using oxidized alginate and gelatin as the hydrogel matrix. Whitlockite bioceramic nanoparticles (WH NPs) were incorporated, as they provide enhanced osteogenic stimulation compared to hydroxyapatite via providing higher local ion concentration. The drug simvastatin was also incorporated into the hydrogel system, as it increases the expression of BMP-2 thereby provide environment for bone regeneration. The presence of both WH nanoparticles and simvastatin would enhance bone regeneration potential. The whitlockite nanoparticles (80 ± 8 nm) were synthesized by precipitation method and were characterized. The nanocomposite hydrogel system was characterized by SEM, FTIR and rheologically. The gelation time of the in situ nanocomposite hydrogel was determined by rheological analysis as 28 s, whereas hydrogel alone showed 132 s. Addition of WH NPs not only shortened the gelation time but also increased the gel strength. The in vitro release of simvastatin from the nanocomposite hydrogel showed a release over a period of 28 days. The alkaline phosphatase (ALP) level also showed a significant increase. RUNX2 and BMP2 expressions showed that nanocomposite hydrogel enhanced the osteogenic differentiation. In vivo bone regeneration studies in mice cranial defect studies showed nanocomposite hydrogel was effective in regenerating the bone compared to controls. Thus, the simvastatin-incorporated oxidized alginate-gelatin/WH NPs hydrogel system has the potential to be used as a repairing and regenerative system in cranial bone defects.

Abstract:The combination of bone tissue scaffolds with osteogenic induction factors is an effective strategy to facilitate bone healing processes. Here, chitosan (CS)/nano-hydroxyapatite (HA) scaffolds containing simvastatin (SIM)-loaded PLGA microspheres were fabricated by combining a freeze-drying technique with a modified water-oil-water emulsion method. The CS/HA weight ratio of 1:2 was selected by analyzing the effect of HA content on the micro-architecture, mechanical strength, and biocompatibility of the scaffold. Drug release kinetics showed that the SIM encapsulated in the scaffold was released in a sustained manner for up to 30 days. In vitro bioactivity study in rat bone marrow-derived mesenchymal stem cells showed that the SIM-loaded scaffolds had a strong ability in accelerating cell proliferation and inducing osteogenic differentiation. Moreover, an in vivo experiment using a rat calvarial defect model also documented that the SIM-loaded scaffolds had a remarkable effect on bone-promoting regeneration. The results of this study suggest that the SIM-loaded CS/HA scaffold is feasible and effective in bone repair and thus may provide a promising route for the treatment of critical-sized bone defects.

Abstract:BACKGROUND:In this study, simvastatin (SVT) in situ gels were successfully produced by our group.METHODS:The preparations were characterized in the following aspects: in vitro gelation, drug release, stability and pharmacodynamics.RESULTS:In this study, drug content of prepared gels was found to be in the range between 89 and 92%. The pH value was in the range between 6.5 and 7.0. The gelation temperature of the prepared thermogelling solutions was 37°C. In vitro release showed that the release of SVT from in situ gels was slow with burst effects at an early stage. Researches indicated that intraorally slow release SVT in situ gels could effectively promote bone regeneration repair of alveolar bone defect.CONCLUSION:This drug delivery system could prove to be a novel form able to prolong the residence time and to control the release of drug when administered into the oral cavity.
