Abstract:In recent years, climate change has altered many ecosystems due to a combination of frequent droughts, irregular precipitation, increasingly salinized areas and high temperatures. These environmental changes have also caused a decline in crop yield worldwide. Therefore, there is an urgent need to fully understand the plant responses to abiotic stress and to apply the acquired knowledge to improve stress tolerance in crop plants. The accumulation of polyamines (PAs) in response to many abiotic stresses is one of the most remarkable plant metabolic responses. In this review, we provide an update about the most significant achievements improving plant tolerance to drought, salinity, low and high temperature stresses by exogenous application of PAs or genetic manipulation of endogenous PA levels. We also provide some clues about possible mechanisms underlying PA functions, as well as known cross-talks with other stress signaling pathways. Finally, we discuss about the possible use of PAs for seed priming to induce abiotic stress tolerance in agricultural valuable crop plants.

Abstract:In recent years, extreme environmental cues such as abiotic stresses, including frequent droughts with irregular precipitation, salinity, metal contamination, and temperature fluctuations, have been escalating the damage to plants' optimal productivity worldwide. Therefore, yield maintenance under extreme events needs improvement in multiple mechanisms that can minimize the influence of abiotic stresses. Polyamines (PAs) are pivotally necessary for a defensive purpose under adverse abiotic conditions, but their molecular interplay in this remains speculative. The PAs' accretion is one of the most notable metabolic responses of plants under stress challenges. Recent studies reported the beneficial roles of PAs in plant development, including metabolic and physiological processes, unveiling their potential for inducing tolerance against adverse conditions. This review presents an overview of research about the most illustrious and remarkable achievements in strengthening plant tolerance to drought, salt, and temperature stresses by the exogenous application of PAs. The knowledge of underlying processes associated with stress tolerance and PA signaling pathways was also summarized, focusing on up-to-date evidence regarding the metabolic and physiological role of PAs with exogenous applications that protect plants under unfavorable climatic conditions. Conclusively, the literature proposes that PAs impart an imperative role in abiotic stress tolerance in plants. This implies potentially important feedback on PAs and plants' stress tolerance under unfavorable cues.

Abstract:Polyamines (PAs) metabolism and functions have started to raise attention from plant scientists in the last years. PAs have been investigated for their involvement in plant cell signaling and protection, proving that alterations in their endogenous levels can affect plant growth, development, and survival. The recognized roles of PAs in metal‐stressed plants are presented and discussed on a "case‐study" basis, for each metal. Bearing in mind that the contamination of soils by heavy metals (HMs) is a growing problem worldwide, it is important to find efficient mechanisms through which agricultural productivity and food quality are safeguarded in a scenario of increased pollution. Making cultivars more tolerant to HM‐stress, capable of detoxifying or accumulating them safely, is a goal that will most certainly benefit from researching the functions and applicability of PAs. To date, plant PAs have been recognized for their roles as membrane‐, protein‐ and nucleic acid‐stabilizers, as protectors of cellular integrity and photosynthetic machinery, as direct and indirect signaling agents, and as emerging members of the non‐enzymatic antioxidant (AOX) system. Moreover, PAs are not only important in normal plant developmental processes but have also been suggested to induce stress priming, to act as epigenetic regulators of gene expression, and to enhance the detoxification and vacuolar compartmentalization of HMs. Although the stress‐ameliorating effects of PAs have been widely studied for several abiotic stresses, not much is known regarding their effects on metal induced‐stress, except for Cd. Abstract:Purpose of ReviewEnvironmental stress is increasingly wearing down crop productivity. As a consequence, a major priority of plant research is to get deeper insights on tolerance mechanisms, providing solutions with the generation of stress-tolerant cultivars. To this goal, a common target of genetic modification is the polyamine signaling pathway. Its association with a protective role against stressful stimuli is widely acknowledged; however, the nature of this function is highly complex. In consequence, this review aims to present up-to-date evidence in regard with metabolic and physiological role of polyamines, protecting plants during severe stressful events (such as drought and soil salinization).Recent FindingsThe most recent evidence from stress physiology research highlight polyamines as key players in signaling responses involved in central metabolism, sugar and lipid homeostasis, maintenance, and induction of antioxidant capacity as well as osmotic regulation. Nevertheless, a number of questions remain open, such as the extent of their roles and whether they represent hub metabolic molecules. Recent advances on polyamine metabolism are therefore summarized in relation to salt and drought stress tolerance and its possible implication on the generation of tolerant crops.SummaryThis review highlights recent findings related to polyamine protective role during drought and salt stress. A clear synergy is established between these amine compounds, ABA, and reactive oxygen and nitrogen species. However, the involvement of these amine compounds on stress physiology goes far beyond the modulation of nitro-oxidative homeostasis and ABA signaling, with an increasing body of evidence demonstrating that polyamines are emerging metabolic hubs of plant stress signaling.

Abstract:Numerous environmental conditions negatively affect plant production. Abiotic stresses, such as salinity, drought, temperature, and heavy metals, cause damage at the physiological, biochemical, and molecular level, and limit plant growth, development, and survival. Studies have indicated that small amine compounds, polyamines (PAs), play a key role in plant tolerance to various abiotic stresses. Pharmacological and molecular studies, as well as research using genetic and transgenic approaches, have revealed the favorable effects of PAs on growth, ion homeostasis, water maintenance, photosynthesis, reactive oxygen species (ROS) accumulation, and antioxidant systems in many plant species under abiotic stress. PAs display a multitrack action: regulating the expression of stress response genes and the activity of ion channels; improving the stability of membranes, DNA, and other biomolecules; and interacting with signaling molecules and plant hormones. In recent years the number of reports indicating crosstalk between PAs and phytohormones in plant response to abiotic stresses has increased. Interestingly, some plant hormones, previously known as plant growth regulators, can also participate in plant response to abiotic stresses. Therefore, the main goal of this review is to summarize the most significant results that represent the interactions between PAs and plant hormones, such as abscisic acid, brassinosteroids, ethylene, jasmonates, and gibberellins, in plants under abiotic stress. The future perspectives for research focusing on the crosstalk between PAs and plant hormones were also discussed.

Abstract:Polyamines (PAs) are found in all living organisms and serve many vital physiological processes. In plants, PAs are ubiquitous in plant growth, physiology, reproduction, and yield. In the last decades, PAs have been studied widely for exploring their function in conferring abiotic stresses (salt, drought, and metal/metalloid toxicity) tolerance. The role of PAs in enhancing antioxidant defense mechanism and subsequent oxidative stress tolerance in plants is well-evident. However, the enzymatic regulation in PAs biosynthesis and metabolism is still under research and widely variable under various stresses and plant types. Recently, exogenous use of PAs, such as putrescine, spermidine, and spermine, was found to play a vital role in enhancing stress tolerance traits in plants. Polyamines also interact with other molecules like phytohormones, nitric oxides, trace elements, and other signaling molecules to providing coordinating actions towards stress tolerance. Due to the rapid industrialization metal/metalloid(s) contamination in the soil and subsequent uptake and toxicity in plants causes the most significant yield loss in cultivated plants, which also hamper food security. Finding the ways in enhancing tolerance and remediation mechanism is one of the critical tasks for plant biologists. In this review, we will focus the recent update on the roles of PAs in conferring metal/metalloid(s) tolerance in plants.
Role of Polyamines in the Response to Salt Stress of Tomato

Abstract:Plants irrigated with saline solutions undergo osmotic and oxidative stresses, which affect their growth, photosynthetic activity and yield. Therefore, the use of saline water for irrigation, in addition to the increasing soil salinity, is one of the major threats to crop productivity worldwide. Plant tolerance to stressful conditions can be improved using different strategies, i.e., seed priming and acclimation, which elicit morphological and biochemical responses to overcome stress. In this work, we evaluated the combined effect of priming and acclimation on salt stress response of a tomato cultivar (Solanum lycopersicum L.), very sensitive to salinity. Chemical priming of seeds was performed by treating seeds with polyamines (PAs): 2.5 mM putrescine (PUT), 2.5 mM spermine (SPM) and 2.5 mM spermidine (SPD). Germinated seeds of primed and non-primed (controls) were sown in non-saline soil. The acclimation consisted of irrigating the seedlings for 2 weeks with tap water, followed by irrigation with saline and non-saline water for 4 weeks. At the end of the growth period, morphological, physiological and biochemical parameters were determined. The positive effects of combined treatments were evident, when primed plants were compared to non-primed, grown under the same conditions. Priming with PAs improved tolerance to salt stress, reduced the negative effects of salinity on growth, improved membrane integrity, and increased photosynthetic pigments, proline and enzymatic and non-enzymatic antioxidant responses in all salt-exposed plants. These results may open new perspectives and strategies to increase tolerance to salt stress in sensitive species, such as tomato.

Metabolomics and lipidomics insight into the effect of different polyamines on tomato plants under non-stress and salinity conditions

Abstract:Polyamines (PAs) are key signaling molecules involved in plant growth and stress acclimation processes. This work investigated the effect of spermidine, spermine, and putrescine (alone and in a mixture) in tomato plants using a combined metabolomics and lipidomics approach. The experiments were carried out under non-stress and 100 mM NaCl salinity conditions. Shoot and root biomass, as well as SPAD values, were increased by the application of exogenous PAs but with differences across treatments. Similarly, root length density (F: 34, p < 0.001), average root diameter (F: 14, p < 0.001), and very fine roots (0.0-0.5 mm) increased in PA-treated plants, compared to control. Metabolomics and lipidomics indicated that, despite being salinity the hierarchically prevalent factor, the different PA treatments imposed distinct remodeling at the molecular level. Plants treated with putrescine showed the broader modulation of metabolite profile, whereas spermidine and spermine induced a comparatively milder effect. The pathway analysis from differential metabolites indicated a broad and multi-level intricate modulation of several signaling molecules together with stress-related compounds like flavonoids and alkaloids. Concerning signaling processes, the complex crosstalk between phytohormones (mainly abscisic acid, cytokinins, the ethylene precursor, and jasmonates), and the membrane lipids signaling cascade (in particular, sphingolipids as well as ceramides and other glycerophospholipids), was involved in such complex response of tomato to PAs. Interestingly, PA-specific processes could be observed, with peculiar responses under either control or salinity conditions.

Abstract:AbstractSalinity stress is a chief abiotic hindrance affecting crop productivity and yield particularly in arid and semi-arid regions across the globe. Plants have evolved a network of physiological and molecular mechanisms to cope with salinity stress. The complex and multi-faceted nature of salinity tolerance involve several mechanisms and a comprehensive approach on how plants employ tolerance to salinity at distinct levels and combining physiological measurements with new molecular technologies is an emerging area of crop research. Recent evidences from salinity tolerance mechanisms highlight osmolytes that play a key role in quenching free radicals, induce antioxidant machinery, and osmotic regulation. Polyamines (PAs) also exhibit a promising role in salt tolerance mechanism and adaptation strategies mainly by stabilization of membranes, neutralization of acids, and suppressing ROS synthesis. Interconnection of PAs and nitric oxide biosynthesis also raise hopes for better tolerance to plants under stressful environment. Based on recent findings, the present review highlights the protective role of ion pumps, osmolytes, and polyamines during salt stress. All these strategies have emerged as potential targets for researchers to confer salt stress tolerance in plants.

Abstract:Abstract Aims The aim of this study was to compare the efficiency of three defense mechanisms (ionic balance, osmotic adjustment and counteracting oxidative stress) under low, moderate and high soil salinity in two related species of contrasting tolerance to salinity: the halophyte Aster tripolium and the glycophyte Aster alpinus , and to elucidate their phytohormone-mediated regulation. Methods The phytohormonal profiling was performed to asses correlations between the pool of plant growth regulators and parameters depicting ionic homeostasis, osmotic adjustment and antioxidant system. Results Defense mechanisms in both species were based on accumulation/activity of distinct compounds (Na + and K + ions, antioxidants), but differences among species concerned particularly soluble carbohydrates and betaines. The halophyte accumulated mannitol, uronic acids and sucrose, while the glycophyte mostly glucose and proline-betaine. The halophyte responses also correlated with changes in the content of plant growth promoting PGRs, as well as jasmonates and benzoic acid. The glycophyte responses corresponded with changes in content of abscisic acid and ethylene precursor, as well as salicylic acid. Conclusions We provided evidence that benzoic acid rather than salicylic acid is involved in salt tolerance in the halophyte and elevated SA content may enhance vulnerability to salt excess. An important element of tolerance trait is also JA-GA network that influences the intensity of defense responses. This study uncovers new aspects of internal phytohormonal regulation of plant reaction to soil salinity and enables further insight into extremophyte biology.

Combined Transcriptome and Proteome Analysis to Elucidate Salt Tolerance Strategies of the Halophyte Panicum antidotale Retz

Abstract:Panicum antidotale , a C4 monocot, has the potential to reclaim saline and drylands and to be utilized as fodder and forage. Its adaptability to survive saline stress has been proven with eco-physiological and biochemical studies. However, little is known about its molecular mechanisms of salt tolerance. In this study, an integrated transcriptome and proteome analysis approach, based on RNA sequencing and liquid chromatography tandem mass spectrometry (LC-MS/MS), was used to identify the said mechanisms. Plants were treated with control (0 mM), low (100 mM), and high (300 mM) sodium chloride (NaCl) treatments to distinguish beneficial and toxic pathways influencing plant biomass. The results indicated differential expression of 3,179 (1,126 upregulated/2,053 downregulated) and 2,172 (898 upregulated/1,274 downregulated) genes (DEGs), and 514 (269 upregulated/245 downregulated) and 836 (494 upregulated/392 downregulated) proteins (DEPs) at 100 and 300 mM NaCl, respectively. Among these, most upregulated genes and proteins were involved in salt resistance strategies such as proline biosynthesis, the antioxidant defense system, ion homeostasis, and sugar accumulation at low salinity levels. On the other hand, the expression of several genes and proteins involved in the respiratory process were downregulated, indicating the inability of plants to meet their energy demands at high salinity levels. Moreover, the impairments in photosynthesis were also evident with the reduced expression of genes regulating the structure of photosystems and increased expression of abscisic acid (ABA) mediated pathways which limits stomatal gas exchange. Similarly, the disturbance in fatty acid metabolism and activation of essential ion transport blockers damaged the integrity of the cell membrane, which was also evident with enhanced malondialdehyde (MDA). Overall, the analysis of pathways revealed that the plant optimal performance at low salinity was related to enhanced metabolism, antioxidative defense, cell growth, and signaling pathways, whereas high salinity inhibited biomass accumulation by altered expression of numerous genes involved in carbon metabolism, signaling, transcription, and translation. The data provided the first global analysis of the mechanisms imparting salt stress tolerance of any halophyte at transcriptome and proteome levels.

Abstract:In the context of an epidemiological study of autism in Nova Scotia, subjects were evaluated for minor physical anomalies and physical measurements. Normal control children, children with autism and their siblings, and children with developmental disabilities and their siblings were compared. Posterior rotation of the external ears was found to be a characteristic related to autism specifically, rather than to developmental disabilities in general. Small feet and normal-to-large hands also were observed in the autism group. Children with autism had a significant reduction in interpupillary distance, but not intercanthic distance or head circumference. In contrast, children with other developmental disabilities were notable for general small stature, which affected the hands, feet, eyes, and head size, as well as height. Abnormal ear configuration was the minor malformation most characteristic of the developmental disability group, and the subset of Down syndrome children had single transverse creases of the palm and epicanthic folds that resulted in significantly increased rates of these anomalies in the developmentally disabled controls. Siblings of the two disabled groups were not significantly different from normal controls on any of the measures that characterized children with autism or other developmental disabilities. The results agree with those of several previous studies, which have suggested that abnormalities of the ears are the general category of minor anomalies most associated with autism. Recent evidence regarding the embryological origin of autism suggests that the ear effects may be an important marker of the initiating events that lead to the disorder.

Abstract:Salinity is one of the main abiotic stresses, which adversely affects the growth, and development and ultimately yield of crop plants. In recent years, phytohormones have been involved in reducing the adverse effects of salt stress. For instance, phytohormone auxin is important for stabilizing the cellular pH environment and activating Na + /H + antiporters under salt stress. The application of gibberellins helps increase the seedlings' water status and maintain RNA and protein levels to maintain growth under salinity stress. Some other types of phytohormones and osmolytes such as abscisic acid, salicylic acid, and ethylene have also been reported to reduce the Na + /K + ratio and Na + and Cl − content, improve the antioxidant enzymes activities and regulate the expression of various genes, including NTHK1, AtMEKK1, AtRSH3, Cat1 , thereby enhancing the plant resistance to salt stress. The individual effects of phytohormones have been discussed in various studies, but comprehensive studies on the application of various phytohormones under salinity stress are lacking. Hence the emphasis of this review is on current and former studies that identify the molecular mechanisms by which phytohormones/osmolytes regulate plant tolerance to salinity stress in promoting growth and development. In this review, we summarized the role of various phytohormones, i.e., cytokinin, abscisic acid, gibberellins, jasmonic acid, auxins, brassinosteroids, and ethylene which directly or indirectly play a vital role in modulating the response of the crop to salinity stress. Furthermore, the role of various osmolytes, i.e., proline and glycine betaine, to enhance salinity tolerance was also highlighted. In addition, the interaction and crosstalk between phytohormones and osmolytes under salinity stress in the plant was adequately elaborated. In the end, future research needs about the application of phytohormones and osmolytes have also been discussed.

Abstract:Halophytes, salt-tolerant plants, are a source of valuable secondary metabolites with potential economic value. The steady-state pools of many stress-related metabolites are already enhanced in halophytes when compared with glycophytes, but growth under conditions away from the optimum can induce stress and consequently result in changes to secondary metabolites such as antioxidants. However, direct evidence for increasing the concentration of valuable secondary metabolites as a consequence of altering the salinity of the growing environment still remains equivocal. To address this, we analysed a range of metabolites with antioxidant capacity (including total phenols, flavonoids, ascorbate, reduced/oxidized glutathione and reactive oxygen species scavenging enzymes) in seedlings and plants from different families (Amaranthaceae, Brassicaceae, Plantaginaceae and Rhizophoraceae) and habitats grown under different salt concentrations. We show that it is possible to manipulate the antioxidant capacity of plants and seedlings by altering the saline growing environment, the length of time under saline cultivation and the developmental stage. Among the species studied, the halophytes Tripolium pannonicum, Plantago coronopus, Lepidium latifolium and Salicornia europaea demonstrated the most potential as functional foods or nutraceuticals.

Polyamines (PAs) are nitrogenous and polycationic compounds containing more than two amine residues. Numerous investigations have demonstrated that cellular PA homeostasis plays a key role in various developmental and physiological processes. The PA balance, which may be affected by many environmental factors, is finely maintained by the pathways of PA biosynthesis and degradation (catabolism). In this review, the advances in PA transport and distribution and their roles in plants were summarized and discussed. In addition, the interplay between PAs and phytohormones, NO, and H2O2 were detailed during plant growth, senescence, fruit repining, as well as response to biotic and abiotic stresses. Moreover, it was elucidated how environmental signals such as light, temperature, and humidity modulate PA accumulation during plant development. Notably, PA has been shown to exert a potential role in shaping the domestication of rice. The present review comprehensively summarizes these latest advances, highlighting the importance of PAs as endogenous signaling molecules in plants, and as well proposes future perspectives on PA research.

Differential Salt Tolerance Strategies in Three Halophytes from the Same Ecological Habitat: Augmentation of Antioxidant Enzymes and Compounds

Abstract:Understanding the salt tolerance mechanism in obligate halophytes provides valuable information for conservation and re-habitation of saline areas. Here, we investigated the responses of three obligate halophytes namely Arthrocnemum macrostachyum, Sarcocornia fruticosa and Salicornia europaea to salt stress (0, 100, 200, 400 and 600 mM NaCl) during their vegetative growth with regard to biomass, ions contents (Na+, K+ and Ca2+), chlorophyll contents, carotenoids, phenolic compounds, flavonoids, and superoxide dismutase, peroxidase and esterase activities. S. europaea showed the lowest biomass, root K+ content, Chl a/b ratio, and carotenoids under salinity. This reduction of biomass is concomitant with the increase in proline contents and peroxidase activity. On the other hand, the promotion of growth under low salinity and maintenance under high salinity (200 and 400 Mm NaCl) in A. Macrostachyum and S. fruticosa are accompanied by an increase in Chl a/b ratio, carotenoids, phenolics contents, and esterase activity. Proline content was decreased under high salinity (400 and 600 mM NaCl) in both species compared to S. europaea, while peroxidase showed the lowest activity in both plants under all salt levels except under 600 mM NaCl in Arthrocnemum macrostachyum compared to S. europaea. These results suggest two differential strategies; (1) the salt tolerance is due to activation of antioxidant enzymes and biosynthesis of proline in S. europaea, (2) the salt tolerance in A. macrostachyum, S. fruticosa are due to rearrangement of chlorophyll ratio and biosynthesis of antioxidant compounds (carotenoids, phenolics and flavonoids) which their cost seem to need less energy than activation of antioxidant enzymes. The differential behavior in halophytes of the same habitat confirms that the tolerance mechanism in halophytes is species-specific which provides new insight about the restoration strategy of saline areas.

Abstract:Polyamines (PAs) are essential biomolecules that are known to be involved in the regulation of many plant developmental and growth processes as well as their response to different environmental stimuli. Maintaining the cellular pools of PAs or their metabolic precursors and by-products is critical to accomplish their normal functions. Therefore, the titre of PAs in the cells must be under tight regulation to enable cellular PA homeostasis. Polyamine homeostasis is hence achieved by the regulation of their input into the cellular PA pool, their conversion into secondary metabolites, their transport to other issues/organs, and their catabolism or turnover. The major contributors of input to the PA pools are their in vivo biosynthesis, interconversion between different PAs, and transport from other tissues/organs; while the output or turnover of PAs is facilitated by transport, conjugation and catabolism. Polyamine metabolic pathways including the biosynthesis, catabolism/turnover and conjugation with various organic molecules have been widely studied in all kingdoms. Discoveries on the molecular transporters facilitating the intracellular and intercellular translocation of PAs have also been reported. Numerous recent studies using transgenic approaches and mutagenesis have shown that plants can tolerate quite large concentrations of PAs in the cells; even though, at times, high cellular accumulation of PAs is quite detrimental, and so is high rate of catabolism. The mechanism by which plants tolerate such large quantities of PAs is still unclear. Interestingly, enhanced PA biosynthesis via manipulation of the PA metabolic networks has been suggested to contribute directly to increased growth and improvements in plant abiotic and biotic stress responses; hence greater biomass and productivity. Genetic manipulation of the PA metabolic networks has also been shown to improve plant nitrogen assimilation capacity, which may in turn lead to enhanced carbon assimilation. These potential benefits on top of the widely accepted role of PAs in improving plants' tolerance to biotic and abiotic stressors are invaluable tools for future plant improvement strategies.
Expression of insulinlike growth factor (IGF) and IGF-binding protein genes in human lung tumor cell lines.

Abstract:BACKGROUND The presence of multiple, low-molecular-weight, insulinlike growth factor (IGF)-binding proteins in lung tumor cell-conditioned medium and lung cancer patient serum has been recently reported. PURPOSE To begin to elucidate the genetic basis for these observations, the present study examines the expression by lung tumor cell lines of three IGF-binding protein genes, namely, IGFBP-1, IGFBP-2, and IGFBP-3. Since IGF-binding proteins are thought to modulate the biologic action of the IGFs, the relationship between the expression of IGF-binding protein genes and the genes encoding IGF-I and IGF-II also has been investigated. METHODS Gene expression was studied in four small-cell lung cancer (SCLC) and three non-small-cell lung cancer (NSCLC) cell lines using Northern blot analysis and reverse transcriptase polymerase chain reaction (RT-PCR) for IGFBP-1. RESULTS IGFBP-1 gene expression was detected by Northern blot analysis in one NSCLC cell line only. However, RT-PCR revealed that the IGFBP-1 gene was expressed in all four SCLC cell lines and in two of the three NSCLC lines. Northern blot analysis of IGFBP-2 gene expression demonstrated that all lung tumor cell lines expressed this gene. A low level of IGFBP-3 gene expression was detected in one SCLC cell line and in all three NSCLC cell lines. All lung tumor cell lines expressed the IGF-II gene as determined by Northern blot analysis. In marked contrast, none of the lines showed evidence of IGF-I gene expression using this method. However, RT-PCR revealed a low level of IGF-I gene expression in one SCLC and one NSCLC cell line only. CONCLUSIONS These observations indicate 1) that IGF-binding proteins secreted by lung tumors are encoded by at least three different genes; 2) that there may be a close association between IGF-II and IGFBP-2 gene expression, such that, where there is production of IGF-II, IGFBP-2 is the principal BP; and 3) that the IGF-II gene is more widely expressed than the IGF-I gene in human lung tumor cell lines.

Abstract:Soil salinization, which is aggravated by climate change and inappropriate anthropogenic activities, has emerged as a serious environmental problem, threatening sustainable agriculture and future food security. Although there has been considerable progress in developing crop varieties by introducing salt tolerance-associated traits, most crop cultivars grown in saline soils still exhibit a decline in yield, necessitating the search for alternatives. Halophytes, with their intrinsic salt tolerance characteristics, are known to have great potential in rehabilitating salt-contaminated soils to support plant growth in saline soils by employing various strategies, including phytoremediation. In addition, the recent identification and characterization of salt tolerance-related genes encoding signaling components from halophytes, which are naturally grown under high salinity, have paved the way for the development of transgenic crops with improved salt tolerance. In this review, we aim to provide a comprehensive update on salinity-induced negative effects on soils and plants, including alterations of physicochemical properties in soils, and changes in physiological and biochemical processes and ion disparities in plants. We also review the physiological and biochemical adaptation strategies that help halophytes grow and survive in salinity-affected areas. Furthermore, we illustrate the halophyte-mediated phytoremediation process in salinity-affected areas, as well as their potential impacts on soil properties. Importantly, based on the recent findings on salt tolerance mechanisms in halophytes, we also comprehensively discuss the potential of improving salt tolerance in crop plants by introducing candidate genes related to antiporters, ion transporters, antioxidants, and defense proteins from halophytes for conserving sustainable agriculture in salinity-prone areas.
