Synthesis and characterization of La0.75Sr0.25MnO3/calcium phosphate composite bone cement with enhanced hyperthermia safety and radiopacity for bone tumor treatment

Abstract:In order to synthesize materials with safer hyperthermia ability for bone tumor treatment, different amount of La0.75Sr0.25MnO3 (LSM) has been incorporated to calcium phosphate cement to prepare self-setting La0.75Sr0.25MnO3/calcium phosphate composite bone cement (MCs) with low Curie temperature (Tc) and enhanced hyperthermia safety. The setting time, phase composition, chemical groups, magnetic property, hyperthermia ability, radiopacity, and cytocompatibility of the novel cement have been evaluated thoroughly. The results show that the particle size of the LSM powder is mainly distributed between 400 nm and 900 nm. The composite cements show a setting time from 15 min to 25 min. The crystalline phases of MCs are dominated by brushite, calcium phosphate, and La0.7Sr0.3MnO3. Moreover, the Tc of MCs is approximately 350 K (77 ℃). Under a magnetic field of 30000 Oe (2.39 ×106 A/m), the MCs show the highest magnetization approximately 1.03 emu/g (1.03 A·m2/Kg). The in vitro and ex vivo hyperthermia tests show that the magnetic cements exhibit good hyperthermia ability and can even realize a temperature rise of approximately 35 ℃ when exposed to an alternating magnetic field (AMF, 300 kHz). Moreover, the radiopacity of MCs is remarkedly enhanced as shown by the ex vivo X-ray transmission tests. Additionally, MTT assay reveals that the MCs display no cell cytotoxicity to MG-63 cells. The total protein qualification tests show that the addition of LSM does not affect the total protein content of cells. The cell/materials co-culturing results show that MG-63 cells can adhere and spread on the cements after co-culturing for 7 days. Therefore, this novel magnetic cement could be potentially used for safer hyperthermia treatment of bone cancer.
