Abstract:Cisplatin is a platinum-based chemotherapy drug widely used to treat various solid tumours. Although it is effective in anti-cancer therapy, many patients develop peripheral neuropathy during and after cisplatin treatment. Peripheral neuropathy results from lesions or diseases in the peripheral somatosensory nervous system and is a significant cause of debilitation and suffering in patients. In recent years, preclinical studies have been conducted to elucidate the mechanisms involved in chemotherapy-induced peripheral neuropathic pain, as well as to promote new therapeutic targets since current treatments are ineffective and are associated with adverse effects. G-protein coupled receptors and ion channels play a significant role in pain processing and may represent promising targets for improving the management of cisplatin-induced neuropathic pain. This review describes the role of G protein-coupled receptors and ion channels in cisplatin-induced pain, analysing preclinical experimental studies that investigated the role of each receptor subtype in the modulation of cisplatin-induced pain.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is the mechanism of peripheral neuropathy (PN) caused by cisplatin and its treatment strategies. Specifically, the paper focuses on exploring the roles of G protein - coupled receptors (GPCRs) and ion channels in cisplatin - induced peripheral neuropathy, and aims to provide new pharmacological intervention methods for the management of this serious side effect by analyzing data from pre - clinical studies. ### Background of the Paper Cisplatin is a chemotherapeutic drug widely used in the treatment of various solid tumors. Although it is very effective in anti - cancer treatment, many patients will develop peripheral neuropathy during or after cisplatin treatment. This neuropathy will cause patients to experience symptoms such as pain, numbness, and tingling, which seriously affect the patient's quality of life. Currently, effective treatment methods for cisplatin - induced peripheral neuropathy are still limited, and existing treatment methods are often accompanied by adverse reactions. ### Research Objectives 1. **Clarify the Mechanism**: Explore the specific mechanism of action of GPCRs and ion channels in cisplatin - induced peripheral neuropathy through the analysis of pre - clinical studies. 2. **Find New Targets**: Identify possible new pharmacological targets to develop more effective treatment strategies, improve patients' symptoms without affecting the effectiveness of anti - cancer treatment. ### Main Contents - **Toxicity and Mechanism of Cisplatin**: The paper describes in detail how cisplatin forms adducts by binding to DNA, leading to apoptosis and neuronal damage, especially the impact on dorsal root ganglia (DRG). - **Role of GPCRs**: Discusses the roles of different types of GPCRs, such as kinin receptors, adenosine receptors, and cannabinoid receptors, in cisplatin - induced peripheral neuropathy and their signal transduction pathways. - **Role of Ion Channels**: Analyzes the roles of voltage - gated channels and transient receptor potential (TRP) channels in pain perception and how they are activated or regulated by cisplatin. - **Potential Treatment Strategies**: Proposes potential treatment strategies for these receptors and channels, such as the use of kinin receptor antagonists, adenosine receptor agonists, and cannabinoid receptor agonists. ### Conclusion Through in - depth research on the mechanism of cisplatin - induced peripheral neuropathy, the paper provides a theoretical basis for the development of new treatment strategies, which is expected to improve patients' symptoms, improve the quality of life, and at the same time not affect the effectiveness of anti - cancer treatment. ### Keywords - Chemotherapy - Cancer - Neuropathic Pain - Kinin - Cannabinoid - Voltage - Gated Channel - Transient Receptor Potential
