Abstract:The aim of this study was to design a pH- and redox-dual responsive Bletilla striata polysaccharide (BSP)-based copolymer to enhance anti-tumor drugs release at tumor sites and improve the therapeutic effect. The copolymer was synthesized using stearic acid (SA) and cystamine via a disulfide linkage and characterized using 1H-Nuclear Magnetic Resonance spectroscopy and Fourier Transform Infrared spectroscopy. The BSP-ss-SA copolymer could self-assemble into micelle in an aqueous environment and could encapsulate docetaxel therein. Its inhibitory effects on HepG2 cells and 4 T1 cells were determined. Besides, the anti-cancer effects in vivo and histopathological study of 4 T1-bearing tumor mice were also evaluated. Docetaxel-loaded BSP-ss-SA micelles showed significant pH-sensitive release behavior, supplying a greater drug release percentage in pH 5.0 media compared to pH 7.4 media. BSP-ss-SA micelles exhibited a clear redox-responsive release property in pH 7.4 media whereas the similar cumulative release percentage of docetaxel from BSP-ss-SA micelles in pH 5.0 media in the presence and absence of DL-dithiothreitol. The Docetaxel-loaded BSP-ss-SA micelles clearly inhibited the proliferation of HepG2 and 4 T1 cells compared with docetaxel solution. The results of MTT and histopathological study indicated that BSP-ss-SA copolymer exhibited good blood compatibility. The BSP-ss-SA copolymer may be used as carriers to deliver anti-tumor drugs to special tumor tissues.

Abstract:A novel redox-sensitvie core cross-linked micelles based on disulfide-linked PEG-polypeptide hybrid polymer were prepared and demonstrated for anticancer drug delivery, where N,N-bis(acrylate) cystamine (BAC) served as cross-linker, allyl-terminated poly(gamma-benzyl-L-glutamate) (PBLG) and polyethylene glycol (PEG) methyl ether methacrylate acted as comonomers. The molecular structure and characteristics of the cross-linked micelle and the precursor were confirmed by H-1 NMR and FT-IR. The cross-linked micelles could be easily degraded into individual linear short chains (M-n = 1800) in the presence of 20 mM glutathione (GSH) by the cleavage of the disulfide linkages from the cross-linker BAC. Doxorubicin (DOX) was selected as a model anticancer drug and encapsulated into the micelles with a decent drug loading content of 21.6 wt%. Compared to the burst release of free DOX in first 6 hours, the in vitro release studies revealed that the micelles exhibited a sustained and high cumulative drug release in GSH (up to 86%) within 24 h, rather than the relatively low release rate of 62% in PBS (pH 7.4). Cell cytoxicity experiments showed that the obtained micelles exhibited nontoxic, and the drug-loaded micelles exhibited high anti-cancer efficacy. All the results showed that the designed cross-linked PEG-polypeptide hybrid micelles may be a promising vehicle for anticancer drug delivery with stimuli-triggered drug release behavior in reducing environment.

Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.

Abstract:The amphiphilic diblock copolymer composed of methoxy poly(ethylene glycol) and racemic oligoleucine was synthesized which formed into micelle with uniform size in aqueous environment. Doxorubicin (DOX) was loaded into micelle aided by noncovalent interactions with high drug loading efficiency. The DOX-loaded micelle (referred as M-DOX) demonstrated the sustained drug release in vitro and excellent antiproliferative capability toward both MG63 and Saos-2 cells. Furthermore, for both MG63 and Saos-2-xenografted BALB/c nude mouse models, M-DOX exhibited enhanced intratumoral distribution, improved antitumor efficacy, and reduced side effects compared with free DOX. Therefore, the polypeptide micelle showed a bright prospect for controlled delivery of antitumor drugs in vivo.

Abstract:In this study, the ideal polymer materials were selected by molecular dynamics (MD) and dissipative particle dynamics (DPD) simulation methods, and pH and redox-sensitive micelles were prepared according to the simulation results. Through experiments, we successfully prepared hydrophilic block polyethylene glycol monomethyl ether (mPEG) connected hydrophobic block polylactic acid-glycolic acid (PLGA) amphiphilic polymer, and connected with redox fragment containing disulfide bond, and then connected with amide bond doxorubicin (DOX) to form mPEG-PLGA-SS-DOX prodrug, and further physical embedded free DOX. Finally, we obtained a particle size of 47.04 ± 0.103 nm, the potential of −23.8 ± 0.189 mV, Polymer dispersity index (PDI) of 0.231 ± 0.001, transmission electron microscopy (TEM) showed that it was spherical mPEG-PLGA-SS-DOX/DOX nanoparticles, its entrapment efficiency (EE) was 68.2%, loading content was 16.72%. The cumulative release of mPEG-PLGA-SS-DOX/DOX was 87.39% in a pH 5.0 and 10 mM Glutathione (GSH) tumor simulation environment. It was less toxic to normal cell HEK293 and more cytotoxic to HepG2 tumor cells. It showed that it was an excellent anti-tumor nano-drug. In addition, the application of computer simulation in the design of nanomedicine also provided a new research idea for the research of nanomedicine.

Beta-Cyclodextrin-Centered Star-Shaped Amphiphilic Polymers for Doxorubicin Delivery

Abstract:Aim: Delivery of doxorubicin could be achieved by a novel micellar system based on beta-cyclodextrin-centered star-shaped amphiphilic polymers (sPEL/CD). This study specifically explored the effect of polylactide segments in sPEL/CD on various micelle properties, such as the critical micelle concentration, size, drug loading, cytotoxicity and drug resistance reversing effect. Method: The sPEL/CD was synthesized by the arm-first method. The critical micelle concentrations of polymeric micelles were determined by fluorescence spectrophotometry using pyrene as a probe. The oil/water method was applied to prepare doxorubicin-loaded micelles. 3-(4,5-dimethylthi-azol-2-yl)-2,5-diphenyltetrazolium bromide, confocal laser-scanning microscopy and flow cytometry were used to examine cell cytotoxicity and cellular uptake of the doxorubicin-loaded micelles. Finally, rhodamine-123 cellular uptake was determined to evaluate the polymer action on MCF-7 and MCF-7/ADR cells. Results: All polymers exhibited low cytotoxicity and their micelles had a desirable re lease-acceleration pH (pH 5.0) for cytoplasmic drug delivery. With the introduction of polylactide into the polymer, the micelle critical micelle concentration can be effectively decreased and the drug-loading content was enhanced. Most importantly, the drug resistance of MCF-7/ADR cells was significantly reversed via the interaction between polymer and Pgp. Therefore, this type of polymer has potential superiority for cancer therapy.
Fabrication of Core Crosslinked Polymeric Micelles as Nanocarriers for Doxorubicin Delivery: Self-Assembly, In Situ Diselenide Metathesis and Redox-Responsive Drug Release

Abstract:Polymeric micelles (PMs) have been used to improve the poor aqueous solubility, slow absorption and non-selective biodistribution of chemotherapeutic agents (CAs), albeit, they suffer from disassembly and premature release of payloads in the bloodstream. To alleviate the thermodynamic instability of PMs, different core crosslinking approaches were employed. Herein, we synthesized the poly(ethylene oxide)-b-poly((2-aminoethyl)diselanyl)ethyl l-aspartamide)-b-polycaprolactone (mPEG-P(LA-DSeDEA)-PCL) copolymer which self-assembled into monodispersed nanoscale, 156.57 ± 4.42 nm, core crosslinked micelles (CCMs) through visible light-induced diselenide metathesis reaction between the pendant selenocystamine moieties. The CCMs demonstrated desirable doxorubicin (DOX)-loading content (7.31%) and encapsulation efficiency (42.73%). Both blank and DOX-loaded CCMs (DOX@CCMs) established appreciable colloidal stability in the presence of bovine serum albumin (BSA). The DOX@CCMs showed redox-responsive drug releasing behavior when treated with 5 and 10 mM reduced glutathione (GSH) and 0.1% H2O2. Unlike the DOX-loaded non-crosslinked micelles (DOX@NCMs) which exhibited initial burst release, DOX@CCMs demonstrated a sustained release profile in vitro where 71.7% of the encapsulated DOX was released within 72 h. In addition, the in vitro fluorescent microscope images and flow cytometry analysis confirmed the efficient cellular internalization of DOX@CCMs. The in vitro cytotoxicity test on HaCaT, MDCK, and HeLa cell lines reiterated the cytocompatibility (≥82% cell viability) of the mPEG-P(LA-DSeDEA)-PCL copolymer and DOX@CCMs selectively inhibit the viabilities of 48.85% of HeLa cells as compared to 15.75% of HaCaT and 7.85% of MDCK cells at a maximum dose of 10 μg/mL. Overall, all these appealing attributes make CCMs desirable as nanocarriers for the delivery and controlled release of DOX in tumor cells.

Abstract:Redox-and pH-sensitive branched star polymers (BSPs), BP(DMAEMA-co-MAEBA- co-DTDMA)(PMAIGP) ns, have been successively prepared by two steps of reversible addition-fragmentation chain transfer (RAFT) polymerization. The first step is RAFT polymerization of 2-(N, N-dimethylaminoethyl) methacrylate (DMAEMA) and p-(methacryloxyethoxy) benzaldehyde (MAEBA) in the presence of divinyl monomer, 2,2'-dithiodiethoxyl dimethacrylate (DTDMA). The resultant branched polymers were used as a macro-RAFT agent in the subsequent RAFT polymerization. After hydrolysis of the BSPs to form BP(DMAEMA-co- MAEBA-co-DTDMA)(PMAGP)(n)s (BSP-H), the anticancer drug doxorubicin (DOX) was covalently linked to branched polymer chains by reaction of primary amine of DOX and aldehyde groups in the polymer chains. Their compositions, structures, molecular weights, and molecular weight distributions were respectively characterized by nuclear magnetic resonance spectra and gel permeation chromatography measurements. The DOX-loaded micelles were fabricated by self-assembly of DOX-containing BSPs in water, which were characterized by transmission electron microscopy and dynamic light scattering. Aromatic imine linkage is stable in neutral water, but is acid-labile; controlled release of DOX from the BSP-H-DOX micelles was realized at pH values of 5 and 6, and at higher acidic solution, fast release of DOX was observed. In vitro cytotoxicity experiment results revealed low cytotoxicity of the BSPs and release of DOX from micelles in HepG2 and HeLa cells. Confocal laser fluorescence microscopy observations showed that DOX-loaded micelles have specific interaction with HepG2 cells. Thus, this type of BSP micelle is an efficient drug delivery system.
Charge-Convertible and Reduction-Sensitive Cholesterol-Containing Amphiphilic Copolymers for Improved Doxorubicin Delivery

Abstract:For achieving successful chemotherapy against cancer, designing biocompatible drug delivery systems (DDSs) with long circulation times, high cellular endocytosis efficiency, and targeted drug release is of upmost importance. Herein, a well-defined PEG-b-P(MASSChol-co-MANBoc) block copolymer bearing redox-sensitive cholesteryl-side group was prepared via reversible addition-fragmentation chain transfer (RAFT) polymerization (with non-redox PEG-b-P(MACCChol-co-MAN-DCA) as the reference), and 1,2-dicarboxylic-cyclohexene acid (DCA) was then grafted onto the hydrophobic block to endow it with charge-convertible characteristics under a tumor microenvironment. The amphiphilic copolymer could be assembled into polymeric spherical micelles (SSMCs) with polyethylene glycol (PEG) as the corona/shell, and anti-cancer drug doxorubicin (DOX) was successfully encapsulated into the micellar core via strong hydrophobic and electrostatic interactions. This nanocarrier showed high stability in the physiological environment and demonstrated "smart" surface charge conversion from negative to positive in the slightly acidic environment of tumor tissues (pH 6.5~6.8), as determined by dynamic light scattering (DLS). Moreover, the cleavage of a disulfide bond linking the cholesterol grafts under an intracellular redox environment (10 mM GSH) resulted in micellar dissociation and accelerated drug release, with the non-redox-responsive micelles (CCMCs) as the control. Additionally, a cellular endocytosis and tumor proliferation inhibition study against MCF-7 tumor cells demonstrated the enhanced endocytosis and tumor cell inhibitory efficiency of dual-responsive SSMCs/DOX nanomedicines, revealing potentials as multifunctional nanoplatforms for effective oncology treatment.

Abstract:Demicellization of the self-assembled multimolecular micelles upon dilution restricts their application as drug delivery systems (DDSs) for tumor treatment. Here, a redox-responsive hyperbranched polymer prodrug (HBPP) was designed with a high drug content of 62.0% as a unimolecular micelle for the tumor-selective drug delivery, via the facile self-condensing vinyl polymerization (SCVP) of redox-responsive doxorubicin-based prodrug monomer MA-SS-DOX and poly(ethylene glycol) methacrylate (PEGMA) with p-chloromethylstyrene (CMS) as an inimer. The unimolecular micelle could be easily obtained with a hydrodynamic diameter of 122 nm, showing excellent GSH-triggered drug release performance with a cumulative release of 60.9% within 85 h but a low premature drug leakage of 3.2%. Abstract:Reduction-responsive biodegradable polymeric micelles based on functional 2-methylene-1,3-dioxepane (MDO) copolymers are developed and investigated for triggered doxorubicin (DOX) release. The MDO-based copolymers P(MDO-co-PEGMA-co-PDSMA) are synthesized via the simple one-step radical ring-opening copolymerization of MDO, poly(ethylene glycol) methyl ether methacrylate (PEGMA), and pyridyldisulfide ethylmethacrylate (PDSMA). The copolymers can self-assemble to form micelles in aqueous solution. DOX, a model anticancer drug, is loaded into the micelles with the drug loading content (DLC) of 11.3%. The micelles can be disassembled under a reductive environment (10 x 10(-3)m glutathione), which results in a triggered drug release behavior. The glutathione-mediated intracellular drug release of DOX-loaded micelles is investigated against A549 cells. Confocal laser scanning microscopy (CLSM) results demonstrated that DOX-loaded micelles exhibits faster drug release in glutathione monoester (GSH-OEt)-pretreated A549 cells, compared with untreated and buthionine sulfoximine (BSO)-pretreated A549 cells. Based on the facile synthetic strategy, the reduction-sensitive biodegradable micelles with triggered intracellular drug release are promising for anticancer drug delivery.

Abstract:Here, a biodegradable polymer-drug conjugate of doxorubicin (DOX) conjugated with a stearic acid-grafted chitosan oligosaccharide (CSO-SA) was synthesized via disulfide linkers. The obtained polymer-drug conjugate DOX-SS-CSO-SA could self-assemble into nanosized micelles in aqueous medium with a low critical micelle concentration. The size of the micelles was 62.8 nm with a narrow size distribution. In reducing environments, the DOX-SS-CSO-SA could rapidly disassemble result from the cleavage of the disulfide linkers and release the DOX. DOX-SS-CSO-SA had high efficiency for cellular uptake and rapidly released DOX in reductive intracellular environments. In vitro antitumor activity tests showed that the DOX-SS-CSO-SA had higher cytotoxicity against DOX-resistant cells than free DOX, with reversal ability up to 34.8-fold. DOX-SS-CSO-SA altered the drug distribution in vivo, which showed selectively accumulation in tumor and reduced nonspecific accumulation in hearts. In vivo antitumor studies demonstrated that DOX-SS-CSO-SA showed efficient suppression on tumor growth and relieved the DOX-induced cardiac injury. Therefore, DOX-SS-CSO-SA is a potential drug delivery system for safe and effective cancer therapy.
Esterase-Responsive Polymeric Micelles Containing Tetraphenylethene and Poly(ethylene glycol) Moieties for Efficient Doxorubicin Delivery and Tumor Therapy

Abstract:Enzyme-responsive drug delivery systems have drawn much attention in the field of cancer theranostics due to their high sensitivity and substrate specificity under mild conditions. In this study, an amphiphilic polymer T1 is reported, which contains a tetraphenylethene unit and a poly(ethylene glycol) chain linked by an esterase-responsive phenolic ester bond. In aqueous solution, T1 formed stable micelles via self-assembly, which showed an aggregation-induced emission enhancement of 32-fold at 532 nm and a critical micelle concentration of 0.53 μM as well as esterase-responsive activity. The hydrophobic drug doxorubicin (DOX) was efficiently encapsulated into the micelles with a drug loading of 21%. In the presence of the esterase, the selective decomposition of drug-loaded T1 micelles was observed, and DOX was subsequently released with a half-life of 5 h. In vitro antitumor studies showed that T1@DOX micelles exhibited good therapeutic effects on HeLa cells, while normal cells remained mostly intact. In vivo anticancer experiments revealed that T1@DOX micelles indeed suppressed tumor growth and had reduced side effects compared to DOX·HCl. The present work showed the potential clinical application of esterase-responsive drug delivery in cancer therapy.

Abstract:An increasing number of studies have demonstrated that inflammation has a close correlation with multiple stages of tumor development, including tumor initiation, promotion, and migration. Cyclooxygenase-2 (COX-2), which is overexpressed in inflammation, has also been found to be overexpressed in several types of cancer. Some anti-inflammatory drugs have been shown to have potential effects on the inhibition of tumor pathogenesis and development. Among them, non-steroidal anti-inflammatory drugs (NSAIDs) play a role by inhibiting the overexpression of COX-2 with low price and few side effects. In this work, ibuprofen was linked to an amphiphilic polymer mPEG-PLA via disulfide bond to form a polymeric prodrug molecule with reductive sensitivity. The amphiphilic molecule can self-assemble into nanoparticles in an aqueous solution and encapsulate hydrophobic drug, doxorubicin, into the core. Electrostatic potential analysis and weak interaction analysis showed different weak interactions, including hydrogen bonds and π-π stackings, are the driving force of the formation of the complex system between doxorubicin and polymeric prodrug molecules. These quantitative calculations were helpful to understand the formation mechanism of prodrug polymeric carriers. Characterization methods such as TEM and DLS proved that mPEG-PLA- s -IHE/DOX (PDC/DOX) micelles had an excellent uniform nanostructure, and in vitro drug release experiments showed superior sustained-release and reductive environment-dependent release of this drug. More importantly, in vitro cytotoxicity and cell uptake studies showed that the PDC/DOX micelles induced apoptosis of A549 lung cancer cell lines. This study demonstrates the advantage of combining NSAIDs with chemotherapy agents in the treatment of lung cancer, which provides a feasible and novel polymer delivery system, and a reasonable explanation of formation mechanism.

Synthesis and physicochemical properties of doxorubicin-loaded PEGA containing amphiphilic block polymeric micelles

Abstract:In this study, we aim to synthesize self-assembled amphiphilic diblock poly(PEGA- b -HEA-PCL) copolymers through RAFT living polymerization, targeting the delivery of hydrophobic anticancer drugs. The synthesized self-assembled diblock copolymers polymeric micelles (PMs) comprising poly(ethylene glycol) methyl ether acrylate (PEGA), as a hydrophilic segment and 2-hydroxyethyl acrylate-polyhexanoate monomer (HEA-PCL) with different block lengths, as a hydrophobic segment. The chemical structures, compositions, and self-assembled behavior were identified through 1 H NMR spectroscopy. The thermal stability was assessed through TGA and DSC. Furthermore, DOX was encapsulated into all PMs. The drug-loaded PMs exhibited enhanced drug release profiles in acidic medium. Particle diameter was measured through DLS and TEM techniques. The cell viability of diblock polymers and selected DOX-loaded PMs were evaluated against non-cancerous (L929) and cancerous cells (SK-N-AS), respectively, through well-known MTT assay. Micellar aggregates with mean diameters of approximately 127.2–145.3 nm formed in aqueous solution. The diameters of PMs increased to 141.5–173.1 nm upon the incorporation of DOX. The drug loading content and encapsulation efficiency of PMs were approximately 8.09–18.84% and 30.43–54.07%, respectively. The MTT assay results indicated that all synthesized materials had minimal effects on the viability of L929 cells, while DOX-loaded materials inhibited the viability of neuroblastoma cells by 68.7%. The highest drug release was 89.20% at pH 7.4, while 83.45% at pH 5.0 for 40 h. These findings suggest that the synthesized amphiphilic PMs are promising candidates for drug delivery systems.

Abstract:An amphiphilic copolymer, PEG-b-PLA-b-(PAA-co-PNIPAM), was synthesized via atom transfer radical polymerization of tert-butyl acrylate and N-isopropyl acryannide using PEG-b-PLA-Br as a macroinitiator and CuBr/Me6TREN as a catalytic system, followed by selectively hydrolyzing tert-butyl groups to carboxyl groups. Then, doxorubicin (DOX) was loaded to the hydrophobic core, and the PAA segments were cross-linked by cystamine to get disulfide cross-linked and DOX loaded micelles. The transmission electron microscopy images showed that the cross-linked DOX loaded micelles were spherical nanoparticles, with a mean diameter of 100 nm. The drug release behavior of the cross-linked DOX loaded micelles was redox responsive. The cumulative release amount of DOX was 33.1% with the presence of 10 mM of glutathione (GSH) at 37 degrees C, much higher than that without the presence of GSH, which was only 4.3%. Furthermore, the cumulative release amount of DOX was 50.9% at 25 degrees C, higher than that at 37 degrees C, indicating that the cross-linked DOX loaded micelles were also thermo-sensitive.

Abstract:Currently, tumor-targeted nanocarriers self-assembled from amphiphilic polymer-drug conjugates are of great demand. The appeal of these carriers arises mainly through their excellent loading efficiency of homologous drug molecules with microenvironment-triggered drug release. Herein, doxorubicin (DOX) was constructed to a hyaluronic acid (HA) backbone through hydrazone and disulfide linkages to construct pH and reduction coresponsive prodrug conjugates (HA-ss-DOX). During formulation, the amphipathic HA-ss-DOX spontaneously assembled into distinct core/shell micelles in aqueous media and showed conspicuous physical DOX loading capabilities (29.1%, DOX/HA-ss-DOX) based on homologous compatibility. DOX/HA-ss-DOX micelles were shown to be stable in normal physiological environments, while accomplishing selective, rapid DOX release at acidic pH and/or highly reducing conditions. The efficacy of DOX/HA-ss-DOX micelles was tested on A549 human lung cancer cells, wherein flow cytometry and confocal microscopy analysis revealed their HA receptor-mediated endocytosis mechanism. In comparison, DOX-loaded redox-insensitive micelles (DOX/HA-DOX) still demonstrated pH-dependent drug release. However, a more rapid intracellular DOX release profile was achieved in DOX/HA-ss-DOX micelles because of their sensitivity to both acidic and reducing environments. Resultantly, DOX/HA-ss-DOX exhibited the strongest cytotoxicity and apoptosis-inducing ability among all tested groups when tested on an A549 cell line and xenograft model.

Abstract:The pH-triggered biodegradable micelles based on poly(beta-amino ester)-graft-12-acryloyloxy dodecyl phosphorylcholine (PAE-graft-ADPC) were explored as an efficient acid-triggered anticancer drug delivery system. A pH-responsive biodegradable polymer was synthesized by using hydrophilic 12-acryloyloxy dodecyl phosphorylcholine (ADPC) and poly(beta-amino ester) (PAE) via Michael addition reaction. Micelles with phosphorylcholine shells were prepared in aqueous solution and doxorubicin (DOX) was loaded into the core of the micelles with high efficiency (73%). Dynamic light scattering (DLS) and transmission electron microscopy (TEM) demonstrated that the size of the drug-loaded micelles was about 230 nm. DOX-loaded micelles were stable in the physiological environment, whereas they disassembled at pH 6.5 which is similar to the extracellular pH of most tumors. In vitro drug release showed that DOX release from micelles was faster at pH 6.5 than at 7.4. Intracellular uptake demonstrated that DOX was delivered into the cells effectively after the cells were incubated with DOX-loaded micelles at pH 6.5. Moreover, an in vitro cytotoxicity study indicated that the growth of HepG2 cells was inhibited remarkably when the cells were preincubated with DOX-loaded micelles at pH 6.5. The results indicated that the pH-triggered biodegradable micelles could serve as promising drug carriers for cancer therapy.
De novo synthesis of pH-responsive, self-assembled, and targeted polypeptide nano-micelles for enhanced delivery of doxorubicin

Abstract:Abstract Doxorubicin (DOX) is a commonly used anticancer drug, but it is inefficient as a therapeutic due to a lack of targeting. Peptide-tuned self-assembly of DOX offers a strategy to improve targeting for greater efficacy. In this work, we designed and prepared an amphiphilic tumor cell-targeting peptide, P14 (AAAAFFFHHHGRGD), able to encapsulate DOX by self-assembly to form tumor cell-targeting and pH-sensitive nano-micelles. The results showed a critical P14-micelle concentration of 1.758 mg l −1 and an average particle size of micelles of 121.64 nm, with entrapment and drug-loading efficiencies of 28.02% ± 1.35% and 12.06% ± 0.59%, respectively. The prepared micelles can release 73.52 ± 1.27% DOX within 24 h in pH 4.5 medium, and the drug cumulative release profile of micelles can be described by the first-order model. Compared with free DOX, the micelles exhibited an increased ability to inhibit tumor cell growth and cause tumor apoptosis in vitro , with IC 50 values of DOX and P14-DOX micelles against human breast cancer cells (MCF-7) of 0.91 ± 0.07 and 0.75 ± 0.06 μ g ml −1 , respectively, and cellular apoptotic rates of DOX and P14-DOX micelles of 70.3% and 42.4%, respectively. Cellular uptake experiments revealed high concentrations of micelles around and inside MCF-7 cells, demonstrating that micelles can target tumor cells. These results indicate the excellent potential for the application of this amphiphilic peptide as a carrier for small-molecule drugs and suggest a strategy for the design of effective anti-tumor drugs.
