Abstract:The fundamental properties and pH-sensitivity of chitosan/gelating hydrogels were investigated using spectroscopic and microelectro mechanical (MEMS) measurement approaches. Turbidimetric titration revealed that there were electrostatic attractive interactions between tripolyphosphate (TPP), chitosan, and gelatin in the acidic pH range, depending on their degree of ionization. The pH-sensitive swelling behavior of the hydrogels was investigated by monitoring the deflection of hydrogel-coated microcantilevers, which exhibited a sensitive and repeatable response to solution pH. The deflection of the microcantilever increased as the pH decreased, and the response speed of the system exhibited a nearly linear relationship with pH. The effects of the pH and concentration of TPP solution, as well as the ratio of chitosan to gelatin in gel precursor solutions, on the pH sensitivity of the hydrogels were also investigated. It was found that the swelling of the hydrogel is mainly a result of chain relaxation of chitosan–TPP complexes caused by protonation of free amino groups in chitosan, which depends on the crosslinking density set during the formation of the network. An increase in initial crosslink density induced a decrease in swelling and pH sensitivity. It can be concluded from this study that pH-sensitive chitosan gel properties can be tuned by preparatory conditions and inclusion of gelatin. Furthermore, microcantilevers can be used as a platform for gaining increased understanding of environmentally sensitive polymers. © 2006 Wiley Periodicals, Inc.

Abstract:The relative molecular weight of chitosan was determined by the relationships of three methods,HPLC,intrinsic viscosity,and dynamics viscosity.The intrinsic viscosity of chitosan solution was measured with one-point Ubbelohde viscometer,the relative molecular weight of chitosan was determined with HPLC,and the MHS equation was obtained as [?] = 3.72 × 10-5Mw 1.37.This paper also sets up the relationship between dynamics viscosity and the molecular weight of chitosan during 10 mg/mL solution degradation by chitosanase.

Abstract:Functionalized biopolymers, especially at the nanoscale, boast a broad spectrum of applications. In this context, our laboratory synthesized chitosan, an example of such biopolymers, through the deacetylation of chitin derived from shrimp. The molecular structure of the prepared chitosan was examined using FTIR, which confirmed its similarity to commercially available chitosan. A significant aspect of chitosan’s functionalization largely hinges on its solubility in major solvents, which in turn affects its potential applications. In this study, the DFT:B3LYP/3-21g* model was employed to investigate the influence of various solvents on the structural and physical properties of chitosan. Key parameters such as the total dipole moment (TDM), HOMO/LUMO energy gap, and molecular electrostatic potential (MESP) were calculated to ascertain the effect of solvation on nanoscale chitosan. The findings suggest that solvation enhances chitosan’s reactivity in terms of the TDM, HOMO/LUMO energy gap, and MESP. Additionally, solvation led to minor alterations in chitosan’s structural parameters. To experimentally validate the theoretical outcomes, chitosan was dissolved in several solvents. The FTIR spectrum of each solution was then recorded and juxtaposed against raw (undissolved) chitosan. The analyses showed no modifications in the molecular structure due to solvation.

Preparation and Characterization of N-succinyl-chitosan

Abstract:Poisonous solvents were widely used in the preparation of N-succinyl-chitosan,which were easy to cause environmental pollution and difficult to remove completely from the products.These disadvantages limited their uses in biomedical materials.In the present work,a novel method was proposed to successfully synthesize N-succinyl-chitosan,which was safe and environmentally friendly.A series of N-succinyl-chitosan with different degrees of substitution were prepared in absolute ethanol by controlling the reaction time.The different degrees of substitution,molecular weight and polydispersity index of the products were measured.The products were characterized by Fourier transform infrared(FTIR) spectroscopy,X-ray diffractometry(XRD) and thermogravimetry(TG).Furthermore,the water-solubility and biocompatibility were studied.When the reaction time is 15 h,the degree of substitution and molecular weight of the product are highest.Moreover,polydispersity index of the product is smallest among all the products(10 h,12 h,15 h,18 h,20 h).In addition,the results indicate N-succinyl-chitosan has been synthesized with decreased degree of crystallization and thermostability.Water-solubility of modified chitosan is improved due to the decrease of crystallization.N-succinyl-chitosan prepared in absolute ethanol has good biocompatibility,which has great potential in biomedical fields.

Abstract:The extended optical theorem is generalized for scalar acoustical beams of arbitrary character with any angle of incidence interacting with an object of arbitrary geometric shape and size, and placed randomly in the beam's path with any scattering angle. Analytical expressions for the extinction, absorption, and scattering cross sections are derived, and the connections with the axial (i.e., along the direction of wave propagation) torque and radiation force calculations are discussed. As examples to illustrate the analysis for a viscoelastic object, the extinction, absorption, and scattering cross sections are provided for an infinite plane progressive wave, infinite nondiffracting Bessel beams, a zero-order spherical quasi-Gaussian beam, and a Bessel-Gauss vortex beam emanating from a finite circular aperture, which reduces to a finite high-order Bessel beam, a finite zero-order Bessel beam, and a finite piston radiator vibrating uniformly with appropriate selection of beam parameters. The similarity with the asymptotic quantum inelastic cross sections is also mentioned.

Abstract:A new scheme was proposed for synthesizing poly(ethylene glycol)-g-chitosan (PEG-g-CS), where methoxy poly(ethylene glycol) iodide (MPEG-I) (Mn 2000) was used for N-substitution of triphenylmethyl chitosan (TPM-CS) in organic medium. The graft copolymers were obtained by subsequent removal of protecting groups with dichloroacetic acid. By varying PEG-I/TPM-CS feed ratio, the grafting levels (GL) of PEG can be adjusted. The chitosan derivatives were characterized by FTIR, 1H NMR, 13C NMR and DSC. All the copolymers were soluble in water over wide pH range. Furthermore, organosolubility of the hybrids in DMF and DMSO was also achieved when the DS value more than 24%. The lysozyme degradation rate of the copolymers in aqueous neutral medium decreased with the increase of GL value.

Abstract:In this study, we analyzed gallium nitride nanotubes which were formed highly oriented on a c-axis perpendicular to substrate surface by using nanoindentation technique. We were focusing experimental observations on the nanomechanical properties. The behavior of the gallium nitride nanotube breakage accompanying with compression test was examined using a nanoindentation system enabling the application of compressive force to the nanotubes homogeneously. The buckling energy of compression for a single gallium nitride nanotube had been measured. The corresponding value of elastic modulus of gallium nitride nanotube was also characterized. Abstract:The ability to modulate the degradation properties of biomaterials such as thermally responsive hydrogels is desirable when exploring new therapeutic strategies that rely on the temporary presence of a placed scaffold or gel. Here we report a method of manipulating the absorption rate of a poly(N-isopropylacrylamide) ((poly(NIPAAm)) based hydrogel across a wide range (from 1 d to 5 mo) by small alterations in the composition. Relying upon the autocatalytic effect, the degradation of poly(NIPAAm-co-HEMA-co-MAPLA), (HEMA = 2-hydroxyethyl methacrylate; MAPLA = methacrylate-polylactide) was greatly accelerated by adding a fourth monomer methacrylic acid (MAA) at no more than 2 mol% to obtain poly(NIPAAm-co-HEMA-co-MAPLA-co-MAA) (pNHMMj) where j reflects the MAA molar % in the reactant mixture. MAA residue introduction decreased the pH inside the hydrogels and in surrounding buffered solutions. Accelerated degradation positively correlated with MAA content in pNHMMj polymers, putatively by the accelerated cleavage of MAPLA residues to raise the transition temperature of the polymer above body temperature. Physical properties including thermal transition behavior and initial mechanical strength did not vary significantly with MAA content. A rat hindlimb injection model generally reflected the in vitro observation that higher MAA content resulted in more rapid degradation and cellular infiltration. The strategy of tuning the degradation of thermally responsive hydrogels where degradation or solubilization is determined by their polyester components might be applied to other tissue engineering and regenerative medicine applications where designed biomaterial degradation behavior is needed.

Abstract:Carboxymethyl chitosans (CM-chitosan) of various degrees of deacetylation (DD 28-95%) and substitution (DS 0.15-1.21) were successfully prepared from N-acetylchitosans in NaOH of varying concentrations. Infrared spectroscopy (IR), elemental analysis, potentiometric titration, 13C NMR, X-ray diffraction and gel-permeation chromatographic (GPC) techniques were used to characterize their molecular structures. The moisture-absorption (R(a)) and -retention (R(h)) abilities of CM-chitosan are closely related to the DD and DS values. Under conditions of high relative humidity, the maximum R(a) and R(h) were obtained at DD values of about 50%, and when the DD value deviated from 50%, R(a) and R(h) decreased. Under dry conditions, when the DD value was 50%, the R(h) was the lowest. With the DS value increasing, R(a) and R(h) increased. However, further increase of the DS value above 1.0 reduced the increasing tendency of R(a) and R(h), and even some decreases in R(a) and R(h) were observed. Intermolecular hydrogen bonds play a very important role in moisture-absorption and retention ability of CM-chitosan.

Abstract:Degradability is often a critical property of materials utilized in tissue engineering. Although chitosan, a naturally derived polysaccharide, is an attractive material due to its biocompatibility and ability to form scaffolds, its slow and uncontrollable rate of degradation can be an undesirable feature. In this study, we characterize chitosan derivatives formed using a combination of carboxymethylation and a bimodal molecular weight distribution. Specifically, chitosan is carboxymethylated to a theoretical extent of similar to 30% as described in our previous work, in which carboxyl groups possessing negative charges are created at a physiological pH. Carboxymethyl chitosan is used to form films and constructs by varying the ratio of high to low molecular weight (MW) while maintaining the mechanical properties of the polymer. The rate of degradation is found to be dependent upon both the carboxymethylation and the ratio of high to low MW polymer, as determined by dry weight loss in lysozyme solution in PBS. Subsequently, biocompatibility is examined to determine the effects of these modifications upon Neuro-2a cells cultured on these films. Neuro-2a cells adher and proliferate on the modified films at a comparable rate to those cultured on unmodified films. This data indicates that these chitosan derivatives exhibit tunable degradation rates and result in a promising material system for neural tissue engineering.

Abstract:Shrimp waste is a valuable source for chitin extraction and consequently for chitosan preparation. In the process of obtaining chitosan, a determining step is the chitin deacetylation. The main characteristic of chitosan is the degree of deacetylation, which must be as high as possible. The molar mass is another important parameter that defines its utilizations, and according to these, high or low molar masses are required. The present study is an attempt to optimize the deacetylation step to obtain chitosan with a high degree of deacetylation and high or low molar mass. The study was carried out based on experimental data obtained in the frame of a central composite design where three working parameters were considered: NaOH concentration, liquid:solid ratio, and process duration. The regression models defined for the degree of deacetylation (DD) and for the mean molar mass (MM) of chitosan powders were used in the formulation of optimization problems. The objectives considered were simultaneous maximum DD and maximum/minimum MM for the final chitosan samples. For these purposes, multiobjective optimization problems were formulated and solved using genetic algorithms implemented in Matlab®. The multiple optimal solutions represented by trade-offs between the two objectives are presented for each case.

Abstract:Hydrophobic derivatives of polysaccharides possess an amphiphilic behavior and are widely used as rheological modifiers, selective sorbents, and stabilizers for compositions intended for various applications. In this work, we studied the mechanochemical reactions of chitosan alkylation when interacting with docosylglycidyl and hexadecylglycidyl ethers in the absence of solvents at shear deformation in a pilot twin-screw extruder. The chemical structure and physical properties of the obtained derivatives were characterized by elemental analysis, FT-IR spectroscopy, dynamic light scattering, scanning electron microscopy, and mechanical tests. According to calculations for products soluble in aqueous media, it was possible to introduce about 5–12 hydrophobic fragments per chitosan macromolecule with a degree of polymerization of 500–2000. The length of the carbon chain of the alkyl substituent significantly affects its reactivity under the chosen conditions of mechanochemical synthesis. It was shown that modification disturbs the packing ability of the macromolecules, resulting in an increase of plasticity and drop in the elastic modulus of the film made from the hydrophobically modified chitosan samples.

Abstract:Conformational changes of the thermal transitions of chitosan-graft-poly(N-vinylcaprolactam) copolymers in aqueous solution were studied by varying of the length of the grafted poly(N-vinylcaprolactam) (PVCL) chains, as well as the ionic strength and the pH of the solution. The conformational properties of the copolymer were monitored by means of dynamic light scattering and ζ-potential measurements. A series of copolymers with defined molecular architecture were synthesized. Obtained results point out that hydrophobic hydration plays a crucial role on the solubility of this copolymer at neutral and slightly alkaline solutions. The evolution of the size of macromolecular aggregates indicates that, in the coil state, there is a monomodal distribution, passing through a bimodal distribution in the pre-transition region, just before the phase separation. The role of the charge of the copolymers on the cooperative transition is also analyzed. The phase transition of these amphiphilic copolymers shows a strong dependence on the ionic strength of the solution.

Abstract:In this work, the transformation of chitosan-grafted-polyacrylamide (GPAM) aggregates in aqueous solution upon heating was explored by cryo-electron microscope (cryo-TEM) and dynamic light scattering (DLS), and larger aggregates were formed in GPAM aqueous solution upon heating, which were responsible for the thermo-thickening behavior of GPAM aqueous solution during the heating process. The heating initiates a transformation from H-bonding aggregates to a large-sized cluster formed by self-assembled hydrophobic chitosan backbones. The acetic acid (HAc) concentration has a significant effect on the thermo-thickening behavior of GPAM aqueous solution; there is a critical value of the concentration (>0.005 M) for the thermo-thickening of 10 mg/mL GPAM solution. The concentration of HAc will affect the protonation degree of GPAM, and affect the strength of the electrostatic repulsion between GPAM molecular segments, which will have a significant effect on the state of the aggregates in solution. Other factors that have an influence on the thermo-thickening behavior of GPAM aqueous solution upon heating were investigated and discussed in detail, including the heating rate and shear rate.

Abstract:A better understanding of the physicochemical phenomena underlying the direct solubilization of solid chitosan in an aqueous acetic acid medium without the addition of salt is needed. These phenomena have been studied using an inverse approach in which the chitosan is treated as a poly-acid and not as poly-base, and the macrophenomenon observed represents the aggregation of the macromolecules of chitosan instead of their solubilization. The purpose of this study is to present an overview of the physicochemical phenomena underlying this direct solubilization in an aqueous acetic acid medium without the addition of salt through a mathematical model based on equilibrium equations, mass balances, and correlations between experimental data. This model allowed us to model dilute chitosan/acetic acid/water systems without the addition of salt with an adequate fit (R2 > 0.9951) to the trend of the experimental data, enabling the calculation of the pH, degree of ionization, apparent dissociation constant, and the concentrations of the smaller molecules \(\left( {{\text{CH}}_{3} {\text{COOH}}, {\text{CH}}_{3} {\text{COO}}^{ - } ,{\text{H}}_{3} {\text{O}}^{ + } } \right)\), with only a priori knowledge of the initial concentration of acetic acid, initial concentration of amine groups, and the dissociation constant of acetic acid. The main finding of this study was that the diluted systems lacking salt studied here showed a tendency toward an apparent dissociation constant of \(pK_{1/2} \to 6.44\) as the polymer concentration increased. This value is similar to the values reported in the literature for systems containing salt and a degree of deacetylation \(\left( {f_{\text{D}} \sim 0.7} \right)\) similar to the chitosan evaluated here.

Abstract:Abstract Correctness and reliability of molar mass data by viscometry in organometallic solvents (cuen, cuoxam, cadoxen) are compromised by the alkalinity of these solvents which causes immediate depolymerization especially in the case of pulps with higher carbonyl content (oxidative damage). The viscosity values thus correspond to the molar mass after the beta-elimination reactions that underly these degradative processes, which is sometimes significantly smaller than the molar mass determined by gel permeation chromatography (GPC) in the non-degrading solvent system DMAc/LiCl. Despite this well-known drawback, viscosity measurements have become a standard approach for molar mass measurements due to their ease and fastness, especially in the pulp and paper industries. A potential way to reduce the inherent error of these molar mass determinations via viscosity measurements is a reductive treatment prior to dissolution of the pulp in the organometallic solvents, which converts the labile, alkali-sensitive carbonyl structures back to the respective alcohols. Using sodium borohydride (NaBH 4 ) on different types of cellulosic pulps, we demonstrate the beneficial effects of such a reduction step on the determined degree of polymerization (DP) for all three common solvents: cuen, cuoxam and cadoxen. Molar mass distributions and profiles of carbonyl groups were determined by GPC and by carbonyl selective fluorescence labeling (“CCOA method”). Such a reductive treatment was especially valuable for hemicellulose-containing pulps. While the decreased measurement error according to the new protocol is beyond doubt, an immediate acceptance in the pulp and paper industries is at least questionable, because the new, more correct data would not agree with the old – wrong, but consistent – numbers accumulated over years and decades. In the long run, however, the new, improved protocol will prevail here as well due to its lower error rate.

[Spectra analyses of chitosans degraded by hydrogen peroxide under optimal conditions]

Abstract:Chitosans with different average molecular weight (1,500-156 kDa) were prepared under optimal conditions based on our previous research by the depolymerization of initial chitosan using hydrogen peroxide, with maintaining the native structure of natural chitosan as principal consideration. Nitrogen and Carbon components of initial chitosan and degraded products were analyzed by BUCHI fully automatic nitrogen determination system and Liquid total organic carbon (TOC) analyzer respectively. Fourier transform infrared (FTIR), 13C nuclear magnetic resonance (13C NMR), X-ray diffraction and circular dichroism (CD) analyses were used to characterize the structure properties of samples. The results indicated that the maximum standard deviations of carbon content and nitrogen content and the degree of deacetylation (DD) of degraded products with the corresponding values of the initial chitosan are 2.4%, 2.3% and 6.9% respectively. Besides, the differentiations of the nitrogen content and DD value of the degraded product with the corresponding values of the initial chitosan are narrowed with the increase in the reaction time. FTIR spectra of resulting products are similar to that of initial chitosan in terms of peak number and position, indicating that there are no other functional groups formed during the degradation. 13C-NMR analyses of initial chitosan and degraded products revealed that the chemical structures of resulting chitosans are not changed to any noticeable extent. X-ray diffraction patterns of initial chitosan and degraded chitosans are alike and show characteristic peaks at 2theta = 10. 4 degrees and 19.8 degrees under the condition that the initial chitosan was disposed as degraded chitosans. Circular dichroism analyses showed that all the samples exhibit a broad negative band located at about 210 nm assigned to n --> pi* electronic transition of the --NH--CO-- chromophore on a glycosidic ring in acidic media, which demonstrated that degraded chitosans maintain their natural conformation in liquid state substantially. All these confirmed that the degraded chtiosans maintain their natural structure and conformation, and the breakage of beta-1,4-glucoside bonds in macromolecule is the basic process under optimal degradation conditions.

Abstract:Chitosan hydrogels are highly sought after in biomedical applications due to their biocompatibility, non-toxicity, and tunable mechanical properties. Nevertheless, achieving stable hydrogels with the required mechanical strength remains a notable obstacle. In this study, we prepared chitosan hydrogels with enhanced stability using minimal concentrations of glutaraldehyde to mitigate toxicity risks. The resulting hydrogel system is characterized by strong covalent bonds formed through imine and hydrogen bonding, facilitated by the aldehyde groups of glutaraldehyde. Rheological analyses reveal the viscoelastic properties of the hydrogels, showcasing stable gel-like structures with shear-thinning behavior, which are further enhanced with increasing chitosan concentration. Our observation demonstrates that the complex shear moduli of 0.5 - 1.5 w/v % chitosan hydrogel ranged between 3.08 kPa and 8.5 kPa, closely matching the shear moduli of the brain and connective tissues. The shear moduli obtained for the 2w/v% (10.2 kPa) and 2.5w/v% hydrogels (11.34 kPa) conform to the shear moduli of liver, fat, relaxed muscle, and breast gland tissue. Thixotropic studies reveal good structural recovery behaviour in the hydrogels. The swellability of 2w/v% and 2.5w/v% hydrogels (50-70%) suggests their suitability for bone tissue engineering, with moderate porosity required for bone growth (60-75%). Moreover, the yield stress values of 0.5 - 1.5 w/v % hydrogels satisfy the requirements for soft tissue engineering, while those of 2 and 2.5 w/v% match the requirements for bone tissue engineering. Our findings suggest that chitosan hydrogels of varying concentrations (0.5–2.5% w/v) hold promise for a range of biomedical applications, including tissue engineering for brain, nerve, soft, and bone tissues, and wound dressing. This research addresses the pressing need for stable chitosan hydrogels with tailored mechanical properties, potentially offering solutions to challenges in biomedical materials design.

[Activity of renin-angiotensin-aldosterone system (RAAS) and vasopressin level in patients with primary pulmonary hypertension].

Abstract:AIM Assessment of RAAS and vasopressin in patients with primary pulmonary hypertension (PPH). MATERIALS AND METHODS Activity of plasma renin (APR), angiotensin-converting enzyme (ACE), plasma levels of angiotensin II (AII) and vasopressin (VP), serum concentration of aldosteron (AS) were measured by radioimmunoassay and enzyme immunoassay in 21 PPH patients with circulatory failure (age 34.7 +/- 2.1 years), 11 patients with NYHA functional class II-III, 10 with class IV, and 10 control subjects (age 29.8 +/- 1.5 years). RESULTS Compared to controls, 21 PPH patients had elevated RAAS parameters: APR up to 3.52 ng/ml/h (p < 0.05), activity of ACE up to 43.13 units, AII level up to 33.93 ng/ml (p < 0.01), AS up to 468.86 ng/ml (p < 0.01), VP up to 5.26 ng/ml (p < 0.001). Circulatory failure progression resulted in activation of all the RAAS components. This and VP activation was the greatest in PPH patients with ACE > 5 ng/ml/h. PPH patients with mean pressure in the pulmonary artery higher than 60 mm Hg demonstrated higher ARP, AS, VP, AII, ACE than those who had this pressure under 60 mm Hg. CONCLUSION PPH patients display a noticeable activation of RAAS and VP. This activation seems to be secondary as the changes increase with elevation of the pressure in the pulmonary artery and aggravation of circulatory insufficiency. Plasma renin activity determines the degree of RAAS activation as a whole. The discovered activation of RAAS in PPH gives grounds for doubts in the validity of using ACE inhibitors in the treatment of PPH.
