Abstract:Hierarchically assembled nanomaterials can find a variety of applications in medicine, energy, and electronics. Here, an automatically controlled dip-pulling method is developed and optimized to generate an unprecedented ordered nano-to-micro hierarchical nanoridge-in-microridge (NiM) structure from a bacteria-specific human-safe virus, the filamentous phage with or without genetically displaying a foreign peptide. The NiM structure is pictured as a window blind with each lath (the microridge) made of parallel phage bundles (the nanoridges). It is independent of the substrate materials supporting it. Surprisingly, it can induce the bidirectional differentiation of stem cells into neurons and astrocytes within a short timeframe (only 8 d) not seen before, which is highly desired because both neurons and astrocytes are needed simultaneously in treating neurodegenerative diseases. Since phages can direct tissue regeneration, template materials formation, sense molecules, and build electrodes, the NiM structures displaying different peptides and on varying materials hold promise in many technologically important fields.

Abstract:We report an unprecedented assembling performance of amide-containing, homo-structured poly(N-alkylacrylamide)s, through which they can assemble into discrete spherical particles with size ranging from tens of nanometres to several micrometres. For a specific polymer, a facile control over topology and scale of assembled structures can be accomplished via solvent conditions. Assembling using appropriate solvent gives rise to capsule-like assemblies. The molecular structure of polymers, such as the side chain chemistry and topology display striking impact on the size and morphology of resulting assemblies. These polymers might function as novel model systems for investigating complex functions performed by amide moieties in proteins, while this assembling offers a simple, facile but powerful and cost-effective manufacturing route for nano- or microstructured materials that may find great potential in diverse applications varying from cosmetics, drug delivery, nanomedicine, to biotechnology.

Abstract:Today various nano-moieties including nanoparticles, nanocubes, nanosticks, nanoflowers and nanoprimes have been synthesised. It is an interesting challenge to build nanostructures using those nano-moieties as blocks owing to their distinct properties and numerous applications. Herein, we demonstrate a smart way, self-assembling, to arrange nanoparticles and nanocubes into a line, sphere, network, bowl-bottom etc. The obtained anisotropic nanostructures have been employed to enhance the sensor sensitivity and to identify the hot-spot position for surface-enhanced Raman scattering (SERS).

Abstract:Controllable self-assembly of nanoscale building blocks into larger specific structures provides an effective route for the fabrication of new materials with unique optical, electronic, and magnetic properties. The ability of nanoparticles (NPs) to self-assemble like molecules is opening new research frontiers in nanoscience and nanotechnology. We present a new class of amphiphilic "colloidal molecules" (ACMs) composed of inorganic NPs tethered with amphiphilic linear block copolymers (BCPs). Driven by the conformational changes of tethered BCP chains, such ACMs can self-assemble into well-defined vesicular and tubular nanostructures comprising a monolayer shell of hexagonally packed NPs in selective solvents. The morphologies and geometries of these assemblies can be controlled by the size of NPs and molecular weight of BCPs. Our approach also allows us to control the interparticle distance, thus fine-tuning the plasmonic properties of the assemblies of metal NPs. This strategy provides a general means to design new building blocks for assembling novel functional materials and devices.

Abstract:The science of self-assembly has undergone a radical shift from asking questions about why individual components self-organize into ordered structures, to manipulating the resultant order. However, the quest for far-reaching nanomanufacturing requires addressing an even more challenging question: how to form nanoparticle (NP) structures with designed architectures without explicitly prescribing particle positions. Here we report an assembly concept in which building instructions are embedded into NPs via DNA frames. The integration of NPs and DNA origami frames enables the fabrication of NPs with designed anisotropic and selective interactions. Using a pre-defined set of different DNA-framed NPs, we show it is possible to design diverse planar architectures, which include periodic structures and shaped meso-objects that spontaneously emerge on mixing of the different topological types of NP. Even objects of non-trivial shapes, such as a nanoscale model of Leonardo da Vinci's Vitruvian Man , can be self-assembled successfully.

Abstract:This article demonstrates the ability of viral fusion peptides to form spiral‐like nanostructures using uniaxial compression in a Langmuir–Blodgett trough. By controlling compression forces, nanofibrils with specific curvatures and inter‐fiber distances are achieved, transitioning from fluid to gel states. These peptide spirals also serve as templates for gold replicas, offering new possibilities in engineering functional nanomaterials. Self‐assembling peptides (SAPs) are versatile building blocks in the creation of hierarchical nanostructures. Although SAPs promise precise control over assembled morphology and dimensions, experimental validation is still crucial. In this sense, recent efforts have focused on nanospiral structures, which mimic natural architectures and hold potential for biomedical and nanotechnological applications. Here, it is demonstrated that SARS‐CoV‐2 fusion peptides are able to form specific spiral‐like structures using interfacial assembly as a fabrication methodology, along with fine‐tuning of the spiral ensembles through an imposed interfacial flow due to the uniaxial compression in a Langmuir–Blodgett trough. Molecular characterization by atomic force microscopy, neutron reflectometry, interfacial shear rheology, and infrared spectroscopy, highlighted how variations in fusion peptide sequences influence assembly, highlighting the importance of molecular design. These spiral structures are subsequently modified to serve as templates for metallic replicas, expanding the potential of peptide‐guided self‐assembly in fabricating tailored nanostructured surfaces with sub‐10 nm dimensions over cm2 areas.

Abstract:Biomolecule-mediated assembly of novel nanoconjugates has been subjected to numerous investigations nowadays, which provides a new insight into the material science and engineering. Via the molecular biology technology, the genetically engineered polypeptide for inorganics (GEPI) can be designed as a molecular binder into the bio-scaffold to assemble hybrid functional nanoarchitectures. In the present work, we constructed a multi-functional virus-like particle (VLP) scaffold based on the Tobacco mosaic virus (TMV) (wild-type strain U1) in a facile way. The S123C site-mutated coat protein (CP) of TMV recombinated with a Ti–GEPI and 6-histidine tag (His tag) was successfully cloned, expressed and purified. The as-produced r-CP products retained both binding affinity for inorganic nanoparticles and self-assembly capability. Analysis of r-CP self-assembly during condensation polymerization was conducted. The r-CP-assembled aggregates including disc-like structures and rod-like VLPs were recovered by preparative size-exclusion chromatography (SEC) and then examined by TEM analysis. Notably, r-CP protein displayed a thermo-triggered reversibly switchable transition between bistable states of transparency and opaqueness. Western blot analysis, Immunogold labelling and TEM analysis were employed to test the existence and binding function of the His tag group in r-CP VLPs. Furthermore, dispersion of TiO 2 NPs in r-CP solution and recyclable application in bio-benefication was introduced. Here we demonstrated the possibilities of combining peptide-mediated immobilization with VLP-based biotemplate bearing multi-binding moieties for prospectful functional applications.

Abstract:The co-self-assembly of proteins and nucleic acids (NAs) produces complex biomolecular machines (e.g., ribosomes and telomerases) that represent some of the most daunting targets for biomolecular design. Despite significant advances in protein and DNA or RNA nanotechnology, the construction of artificial nucleoprotein complexes has largely been limited to cases that rely on the NA-mediated spatial organization of protein units, rather than a cooperative interplay between protein- and NA-mediated interactions that typify natural nucleoprotein assemblies. We report here a structurally well-defined synthetic nucleoprotein assembly that forms through the synergy of three types of intermolecular interactions: Watson-Crick base pairing, NA-protein interactions, and protein-metal coordination. The fine thermodynamic balance between these interactions enables the formation of a crystalline architecture under highly specific conditions.

Abstract:When viewed through the lens of materials science, nature provides a vast library of hierarchically organized structures that serve as inspiration and raw materials for new synthetic materials. The structural organization of complex bioarchitectures with advanced functions arises from the association of building blocks and is strongly supported by ubiquitous mechanisms of self-assembly, where interactions among components result in spontaneous assembly into defined structures. Viruses are exemplary, where a capsid structure, often formed from the self-assembly of many individual protein subunits, serves as a vehicle for the transport and protection of the viral genome. Higher-order assemblies of viral particles are also found in nature with unexpected collective behaviors. When the infectious aspect of viruses is removed, the self-assembly of viral particles and their potential for hierarchical assembly become an inspiration for the design and construction of a new class of functional materials at a range of different length scales.<i>Salmonella typhimurium</i> bacteriophage P22 is a well-studied model for understanding viral self-assembly and the construction of virus-like particle (VLP)-based materials. The formation of cage-like P22 VLP structures results from scaffold protein (SP)-directed self-assembly of coat protein (CP) subunits into icosahedral capsids with encapsulation of SP inside the capsid. Employing the CP-SP interaction during self-assembly, the encapsulation of guest protein cargos inside P22 VLPs can be achieved with control over the composition and the number of guest cargos. The morphology of cargo-loaded VLPs can be altered, along with changes in both the physical properties of the capsid and the cargo-capsid interactions, by mimicking aspects of the infectious P22 viral maturation. The structure of the capsid differentiates the inside cavity from the outside environment and serves as a protecting layer for the encapsulated cargos. Pores in the capsid shell regulate molecular exchange between inside and outside, where small molecules can traverse the capsid freely while the diffusion of larger molecules is limited by the pores. The interior cavity of the P22 capsid can be packed with hundreds of copies of cargo proteins (especially enzymes), enforcing intermolecular proximity, making this an ideal model system in which to study enzymatic catalysis in crowded and confined environments. These aspects highlight the development of functional nanomaterials from individual P22 VLPs, through biomimetic design and self-assembly, resulting in fabrication of nanoreactors with controlled catalytic behaviors.Individual P22 VLPs have been used as building blocks for the self-assembly of higher-order structures. This relies on a balance between the intrinsic interparticle repulsion and a tunable interparticle attraction. The ordering of VLPs within three-dimensional assemblies is dependent on the balance between repulsive and attractive interactions: too strong an attraction results in kinetically trapped disordered structures, while decreasing the attraction can lead to more ordered arrays. These higher-order assemblies display collective behavior of high charge density beyond those of the individual VLPs.The development of synthetic nanomaterials based on P22 VLPs demonstrates how the potential for hierarchical self-assembly can be applied to other self-assembling capsid structures across multiple length scales toward future bioinspired functional materials.

Abstract:This article describes a novel supramolecular assembly-mediated strategy for the organization of Au nanoparticles (NPs) with different shapes (e.g., spheres, rods, and cubes) into large-area, free-standing 2D and 3D superlattices. This robust approach involves two major steps: (i) the organization of polymer-tethered NPs within the assemblies of supramolecular comblike block copolymers (CBCPs), and (ii) the disassembly of the assembled CBCP structures to produce free-standing NP superlattices. It is demonstrated that the crystal structures and lattice constants of the superlattices can be readily tailored by varying the molecular weight of tethered polymers, the volume fraction of NPs, and the matrix of CBCPs. This template-free approach may open a new avenue for the assembly of NPs into 2D and 3D structures with a wide range of potential applications.

Abstract:The ability to modulate nanoparticle (NP) assemblies with atomic precision is still lacking, which hinders us from creating hierarchical NP organizations with desired properties. In this work, a hierarchical fibrous (1D to 3D) assembly of Au NPs (21-gold atom, Au21) is realized and further modulated with atomic precision via site-specific tailoring of the surface hook (composed of four phenyl-containing ligands with a counteranion). Interestingly, tailoring of the associated counterion significantly changes the electrical transport properties of the NP-assembled solids by two orders of magnitude due to the altered configuration of the interacting π-π pairs of the surface hooks. Overall, our success in atomic-level modulation of the hierarchical NP assembly directly evidences how the NP ligands and associated counterions can function to guide the 1D, 2D, and 3D hierarchical self-assembly of NPs in a delicate manner. This work expands nanochemists' skills in rationally programming the hierarchical NP assemblies with controllable structures and properties.

This patterning is realized by use of a rigid and coordinative DNA frame as a molecular stamping apparatus (MOST App). The MOST App transfers multiple types of molecular "inks", DNA sequences, onto a nanoparticle surface and fixes these molecular inks into place to form a designed pattern. After a nanoparticle is released the from MOST App, it possesses single-molecule patches that can provide anisotropic bonds with distinctive affinities. We further use these stamped nanoparticles to assemble prescribed clusters, whose structure is determined by the locations of patches. Using electron microscopy and tomographic methods, we investigate the efficiency of cluster formation and the resulting spatial arrangements of nanoparticles. The presented approach provides a single-molecule and spatially determined control over nanoparticle functionalization for creating nanoparticles with designed placement of different molecules and for realizing a rational fabrication of nanomaterial architectures.

Abstract:In bottom–up self-assembly, DNA nanotechnology plays a vital role in the development of novel materials and promises to revolutionize nanoscale manufacturing technologies. DNA shapes exhibit many versatile characteristics, such as their addressability and programmability, which can be used for determining the organization of nanoparticles. Furthermore, the precise design of DNA tiles and origami provides a promising technique to synthesize various complex desired architectures. These nanoparticle-based structures with targeted organizations open the possibility to specific applications in sensing, optics, catalysis, among others. Here we review progress in the development and design of DNA shapes for the self-assembly of nanoparticles and discuss the broad range of applications for these architectures.

Abstract:The assembly of nanoparticles (NPs) into complex structures is a fundamental topic in nanochemistry. Although progress has been made in this respect, the classical treatment of NPs as hard building blocks that lack the ability to anisotropically “bond” to each other limits the construction of more complex assemblies. More importantly, extension of methods of assembly of robust NPs to the assembly of ultrasmall ones with size below 2 nm is still challenging. Here we report the controlled self-assembly of ∼2 nm gold NPs into one-dimensional (1D) nanochain, two-dimensional (2D) nanobelt and three-dimensional (3D) nanocomet architectures by kinetically controlling the diffusion of ultrasmall gold NPs with anisotropic surfaces in solution. This process is presumed to allow selective ligand exchange with linkers at different binding sites on the NP surface, and results in “multivalent” interactions between NPs. Different from the assembly of “hard building blocks”, our results demonstrate the significance of surface effects for ultrasmall NPs (or clusters) in determining the structure of complex self-assemblies, and suggest the possibility of assembling these “molecule-like” ultrasmall nanocrystals into novel complex materials on a nanoscale approaching that of real atoms or molecules.

Abstract:Self-assembly processes, while promising for enabling the fabrication of complexly organized nanomaterials from nanoparticles, are often limited in creating structures with multiscale order. These limitations are due to difficulties in practically realizing the assembly processes required to achieve such complex organizations. For a long time, a hierarchical assembly attracted interest as a potentially powerful approach. However, due to the experimental limitations, intermediate-level structures are often heterogeneous in composition and structure, which significantly impacts the formation of large-scale organizations. Here, we introduce a two-stage assembly strategy: DNA origami frames scaffold a coordination of nanoparticles into designed 3D nanoclusters, and then these clusters are assembled into ordered lattices whose types are determined by the clusters' valence. Through modulating the nanocluster architectures and intercluster bindings, we demonstrate the successful formation of complexly organized nanoparticle crystals. The presented two-stage assembly method provides a powerful fabrication strategy for creating nanoparticle superlattices with prescribed unit cells.

Abstract:The ability to organize nanoscale objects into well-defined three-dimensional (3D) arrays can translate advances in nanoscale synthesis into targeted material fabrication. Despite successes in nanoparticle assembly, most extant methods are system specific and not fully compatible with biomolecules. Here, we report a platform for creating distinct 3D ordered arrays from different nanomaterials using DNA-prescribed and valence-controlled material voxels. These material voxels consist of 3D DNA frames that integrate nano-objects within their scaffold, thus enabling the object’s valence and coordination to be determined by the frame’s vertices, which can bind to each other through hybridization. Such DNA material voxels define the lattice symmetry through the spatially prescribed valence decoupling the 3D assembly process from the nature of the nanocomponents, such as their intrinsic properties and shapes. We show this by assembling metallic and semiconductor nanoparticles and also protein superlattices. We support the technological potential of such an assembly approach by fabricating light-emitting 3D arrays with diffraction-limited spectral purity and 3D enzymatic arrays with increased activity.

Abstract:The self-assembly of nanoparticles is a challenging process for organizing precise structures with complicated and ingenious structures. In the past decades, a simple, high-efficiency, and reproducible self-assembly method from nanoscale to microscale has been pursued because of the promising and extensive application prospects in bioanalysis, catalysis, photonics, and energy storage. However, microscale self-assembly still faces big challenges including improving the stability and homogeneity as well as pursuing new assembly methods and templates for the uniform self-assembly. To address these obstacles, here, a novel silver-coated nanopore is developed which serves as a template for electrochemically generating microcyclic structures of gold nanoparticles at micrometers with highly homogenous size and remarkable reproducibility. Nanopore-induced microcyclic structures are further applied to visualize the diffusion profile of ionic flux. Based on this novel strategy, a nanopore could potentially facilitate the delivery of assembled structures for many practical applications including drug delivery, cellular detection, catalysis, and plasmonic sensing.

Abstract:Combination of top-down and bottom-up process is crucial for fabricating ordered complex micronanostructures. Here we report a simple, rapid, and versatile approach to demonstrate this useful concept, which involves the joint use of multibeam interference patterning and capillary force self-organization. Regular hydrophobic arrays of four-peddle nanoflowers consisting of bent needles with 300 nm tip diameters are readily produced. A "domino model" based on the balance of the capillary and support forces were proposed to interpret realization of large-area homogeneity of the array. The technology, promising for preparing more complex and functional structures, may find broad utilization in nano and biological researches. (C) 2009 American Institute of Physics. Abstract:The construction of variable structured multi-protein nano-assemblies is of great interest for the development of protein-based therapeutic systems. This study showcases the synthesis of polymer-protein assemblies with tunable structure like single- and multi-layer polymer-crosslinked protein vesicles, Janus protein vesicles and other hierarchical-structured assemblies by utilizing a dynamic template-assistant intermittent-assembly approach. The generalization of the methodology helps the protein assemblies to gain notable functional complexity. And we demonstrate compelling evidence highlighting the substantial impact of the topological morphology of protein nanoaggregates on their cellular uptake capacity.
