Abstract:Traditional wound dressings have a limited capacity to absorb exudates, are permeable to microbes, and may adhere to wounds, which leads to secondary injuries. Hydrogels are promising alternative dressings to overcome the above challenges. In this study, we developed sodium alginate-based hydrogel films loaded with Betula utilis bark extract. These films were prepared via solvent-casting crosslinking method and evaluated for wound healing activity. Prepared films were 0.05–0.083 mm thick, flexible with folding endurance ranging from 197–203 folds, which indicates good physical properties. Optimized formulations exhibited successful loading of extract in the film matrix without any interaction as confirmed by FTIR. Maximum zone of inhibition against Gram-positive and Gram-negative bacteria was achieved by optimum formulation (B6), i.e., 19 mm and 9 mm, respectively, with > 90% scavenging activity. Furthermore, this optimum formulation (B6) was able to achieve 93% wound contraction in rats. Histograms of the optimized formulation treated group also revealed complete reepithelization of wounds. Conclusively, our extract-loaded hydrogel dressing successfully demonstrated its potential for cutaneous wound healing.

Abstract:Bacterial cellulose is a bacterially derived polymer with great potential for application in wound healing due to its innate properties such as high biocompatibility and biodegradability. In addition to this, it is naturally biosynthesized by bacteria as a hydrogel, which makes it an optimal substrate for the treatment of dry wounds, where additional moisture is required to facilitate the healing process. However, this polymer lacks antibacterial properties. As bacterial infections are becoming increasingly common and difficult to treat due to antimicrobial resistance, it is of crucial importance to develop strategies for the modification of cellulose to ensure protection against bacterial contamination. In this study, a green-chemistry approach was proposed for the functionalization of cellulose to introduce antibacterial functional groups. Two different active agents, namely glycidyl trimethylammonium chloride and glycidyl hexadecyl ether, were used for the covalent derivatization of the hydroxyl groups of glucose through a heterogeneous reaction in basic aqueous conditions. The modified material was chemically and mechanically characterized by solid-state techniques and rheological measurements. A biological assessment was then carried out both using bacterial cells and human keratinocytes. It was observed that the functionalization performed induced a reduction of approximately half of the bacterial population within 24 h of direct contact withStaphylococcus aureussubsp. aureus Rosenbach 6538P(TM)andEscherichia coli(Migula) Castellani and Chalmers ATCC(R)8739(TM)(respectively, a reduction of 53% and 43% in the cell number was registered for the two strains). In parallel, cytotoxicity studies performed on keratinocytes (HaCaT cell line) showed cell viability in the range of 90 to 100% for up to 6 days of direct contact with both unmodified and modified samples. The morphology of the cells was also visually evaluated, and no significant difference was noted as compared to the control. Finally, thein vitroscratch assay evidenced good wound closure rates in the presence of the samples, with complete coverage of the scratched area after 5 days for both the modified cellulose and the positive control (i.e., keratinocytes growth medium). Overall, the modified hydrogel showed promising features, confirming its potential as an alternative substrate to develop a sustainable, antibacterial and biocompatible wound dressing.
Capparis sepiaria-Loaded Sodium Alginate Single- and Double-Layer Membrane Composites for Wound Healing

Abstract:Background: Effective wound dressing is the key solution to combating the increased death rate and prolonged hospital stay common to patients with wounds. Methods: Sodium alginate-based single- and double-layer membranes incorporated with Capparis sepiaria root extract were designed using the solvent-casting method from a combination of polyvinyl alcohol (PVA), Pluronic F127 (PF127), and gum acacia. Results: The successful preparation of the membranes and loading of the extract were confirmed using Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD). The prepared membranes were biodegradable and non-toxic to human skin cells (HaCaT), with high biocompatibility of 92 to 112% cell viability and good hemocompatibility with absorbance ranging from 0.17 to 0.30. The membrane's highest water vapor transmission rate was 1654.7333 ± 0.736 g/m2/day and the highest % porosity was 76%. The membranes supported cellular adhesion and migration, with the highest closure being 68% after 4 days compared with the commercial wound dressings. This membrane exhibited enhanced antimicrobial activity against the pathogens responsible for wound infections. Conclusions: The distinct features of the membranes make them promising wound dressings for treating infected wounds.

Polysaccharide Fabrication Platforms and Biocompatibility Assessment as Candidate Wound Dressing Materials

Abstract:Wound dressings are critical for wound care because they provide a physical barrier between the injury site and outside environment, preventing further damage or infection. Wound dressings also manage and even encourage the wound healing process for proper recovery. Polysaccharide biopolymers are slowly becoming popular as modern wound dressings materials because they are naturally derived, highly abundant, inexpensive, absorbent, non-toxic and non-immunogenic. Polysaccharide biopolymers have also been processed into biomimetic platforms that offer a bioactive component in wound dressings that aid the healing process. This review primarily focuses on the fabrication and biocompatibility assessment of polysaccharide materials. Specifically, fabrication platforms such as electrospun fibers and hydrogels, their fabrication considerations and popular polysaccharides such as chitosan, alginate, and hyaluronic acid among emerging options such as arabinoxylan are discussed. A survey of biocompatibility and bioactive molecule release studies, leveraging polysaccharide's naturally derived properties, is highlighted in the text, while challenges and future directions for wound dressing development using emerging fabrication techniques such as 3D bioprinting are outlined in the conclusion. This paper aims to encourage further investigation and open up new, disruptive avenues for polysaccharides in wound dressing material development.
Fabrication and Evaluation of a Soy Protein Isolate/Collagen/Sodium Alginate Multifunctional Bilayered Wound Dressing: Release of Cinnamaldehyde, Artemisia absinthium, and Oxygen

Abstract:Chronic wounds, such as diabetic ulcers and pressure sores, pose significant challenges in modern healthcare due to their prolonged healing times and susceptibility to infections. This study aims to engineer a bilayered wound dressing (BLWD) composed of soy protein isolate/collagen with the ability to release Cinnamaldehyde, Artemisia absinthium (AA), and oxygen. Cinnamaldehyde, magnesium peroxide (MgO2), and AA extract were encapsulated. Nanoparticles were evaluated using scanning electron microscopy (SEM), dynamic light scattering, and ZETA potential tests. Swelling, degradation, water vapor penetration, tensile, MTT, SEM, oxygen release, AA extract release, and antibacterial properties were performed. An in vivo study was carried out to assess the final wound dressing under Hematoxiline&Eosin and Masson trichrome staining analysis and compared to a commercial product. According to the results, the synthesized nanoparticles had an average diameter of about 20 nm with a zeta potential in the range of -20 to -30 mV. The layers had uniform and dense surfaces. The maximum swelling and degradation of the dressing was about 130 and 13% respectively. Generally, better mechanical properties were observed in BLWD than in the single-layer case. More than 90% biocompatibility for the wound dressing was reported. The BLWD could inhibit the growth of Gram-positive and Gram-negative microorganisms. Histopathological analysis showed an acceptable wound-healing property. To sum up, the engineered wound dressing can be a good candidate for more clinical trials.

Abstract:Keeping in view the progressive future prospectus of carbohydrate research, herein, the potential of bioactive polysaccharides aloe vera (AV) and sterculia gum (SG) was explored to develop network hydrogel wound dressings. Both polysaccharides, acemannan of AV gel and glucuronic-galacturonic acid of SG accelerate wound healing process. The antibiotic drug cefuroxime encapsulated dressings were designed by graft-copolymerization with carbopol. The copolymers were characterized by SEM, AFM, FTIR, C13 NMR, XRD, TGA-DTG, DSC and swelling studies. The slow diffusion of cefuroxime drug exhibited non-Fickian mechanism and release of drug was occurred with a first order kinetic model. Polymer-bio membrane interactions indicated mucoadhesion nature. The clot formation was reduced during polymer-blood interactions indicated bio-compatible nature of the dressings moreover dressings were found antioxidant in nature. The porous dressings were O2 permeable and microbial impermeable. The hydrogel dressings revealed antimicrobial activity against P. aeruginosa. Degree of antimicrobial activity enhanced in case of hydrogel wound dressings loaded with cefuroxime. Overall, the results of these biochemical assays, sustained release of antibiotic drug and inherent wound healing potential of AV-SG indicated the use of hydrogels as dressing materials for better wound healing.

Abstract:Nanomaterial-based wound dressings have been extensively studied for the treatment of both minor and life-threatening tissue injuries. These wound dressings must possess several crucial characteristics, such as tissue compatibility, non-toxicity, appropriate biodegradability to facilitate wound healing, effective antibacterial activity to prevent infection, and adequate physical and mechanical strength to withstand repetitive dynamic forces that could potentially disrupt the healing process. Nevertheless, the development of nanostructured wound dressings that incorporate various functional micro- and nanomaterials in distinct architectures, each serving specific purposes, presents significant challenges. In this study, we successfully developed a novel multifunctional wound dressing based on poly(lactic acid) (PLA) fibrous membranes produced by solution-blow spinning (SBS) and electrospinning. The PLA-based membranes underwent surface modifications aimed at tailoring their properties for utilization as effective wound dressing platforms. Initially, beta-chitin whiskers were deposited onto the membrane surface through filtration, imparting hydrophilic character. Afterward, silver nanoparticles (AgNPs) were incorporated onto the beta-chitin layer using a spray deposition method, resulting in platforms with antimicrobial properties against both Staphylococcus aureus and Escherichia coli. Cytotoxicity studies demonstrated the biocompatibility of the membranes with the neonatal human dermal fibroblast (HDFn) cell line. Moreover, bilayer membranes exhibited a high surface area and porosity (> 80%), remarkable stability in aqueous media, and favorable mechanical properties, making them promising candidates for application as multifunctional wound dressings.

Abstract:Skin wounds, due to their high vulnerability to infections, represent a significant public health issue. These wounds are not only disabling but also entail costly treatments and slow recovery. Consequently, it is crucial to implement new treatments based on bioactive and natural antimicrobial compounds utilizing fibers, polymers, hydrocolloids, and hydrogels to control potential infections and promote wound healing. This study aimed to develop a biocomposite with antimicrobial activity for the treatment of skin wounds, using sodium alginate, bamboo fiber, and a natural antimicrobial as ingredients. The physico-mechanical properties (Young's modulus, tensile strength, elongation at break, moisture absorption, and water vapor permeability) and antimicrobial activity against Escherichia coli, Staphylococcus aureus, and Staphylococcus hominis were determined. The results demonstrated that the designed biocomposite possesses adequate physico-mechanical properties, such as flexibility, strength, and water absorption capacity, in addition to exhibiting antibacterial activity, making it suitable to be used as a dressing in wound treatment.

Abstract:Bacterial cellulose (BC) is an ideal candidate for wound dressings due to its natural origin, exceptional water-holding capacity, pliability, biocompatibility, and high absorption capability. However, the lack of essential antimicrobial activity limits its biomedical applications. This study reported BC-based wound dressings containing silk fibroin protein (SF), offering the potential for biomimetic properties, and (-)-epigallocatechin-3-gallate (EGCG) for polyphenol-assisted surface modification to promote infectious wound healing. Glycerol was used as the carbon source to promote the formation of an adhesive layer by facilitating the β-sheet folding of SF, and different concentrations of EGCG were employed to interact with SF through strong hydrogen bonding facilitated by the polyphenolic groups. The functionalized membrane exhibited outstanding water-holding capacity, swelling ratio, and degradation properties, along with enhanced hydrophilicity, adhesiveness, and a remarkable free radical scavenging ability. Both in vitro and in vivo experiments confirmed its potent bacteriostatic activity. The composite membrane displayed excellent biocompatibility, including cellular and hemocompatibility. Importantly, it effectively promoted wound healing in murine back infections. These findings suggest the significant feasibility of the innovative modification approach, and that functionalized membranes have great potential as wound-dressing materials for infection management in future clinical applications.
Gallic Acid-Loaded Sodium Alginate-Based (Polyvinyl Alcohol-Co-Acrylic Acid) Hydrogel Membranes for Cutaneous Wound Healing: Synthesis and Characterization

Abstract:Traditional wound dressings often cannot treat wounds caused by bacterial infections or other wound types that are insensitive to these wound treatments. Therefore, a biodegradable, bioactive hydrogel wound dressing could be an effective alternative option. The purpose of this study was to develop a hydrogel membrane comprised of sodium alginate, polyvinyl alcohol, acrylic acid, and gallic acid for treating skin wounds. The newly developed membranes were analyzed using Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM), X-ray diffraction analysis (XRD), sol-gel fraction, porosity, mechanical strength, swelling, drug release and data modelling, polymeric network parameters, biodegradation, and antioxidation (DPPH and ABTS) and antimicrobial activity against Gram-positive and negative bacteria. The results revealed that hydrogel membranes were crosslinked successfully and had excellent thermal stability, high drug loading, greater mechanical strength, and exhibited excellent biodegradation. Additionally, the swelling ability and the porosity of the surface facilitated a controlled release of the encapsulated drug (gallic acid), with 70.34% release observed at pH 1.2, 70.10% at pH 5.5 (normal skin pH), and 86.24% at pH 7.4 (wounds pH) in 48 h. The gallic acid-loaded hydrogel membranes showed a greater area of inhibition against Pseudomonas aeruginosa, Staphylococcus aureus, and Escherichia coli bacteria as well as demonstrated excellent antioxidant properties. Based on Franz cell analyses, the permeation flux of the drug from optimized formulations through mice skin was 92 (pH 5.5) and 110 (pH 7.4) μg/cm2·h−1. Moreover, hydrogel membranes retained significant amounts of drug in the skin for 24 h, such as 2371 (pH 5.5) and 3300 μg/cm2 (pH 7.4). Acute dermal irritation tests in rats showed that hydrogel membranes were nonirritating. Hydrogel membranes containing gallic acid could be an effective option for improving wound healing and could result in faster wound healing.

Sodium alginate-based multifunctional sandwich-like system for treating wound infections

Abstract:Microbial colonization and development of infections in wounds is a sign of chronicity. The prevailing approach to manage and treat these wounds involves dressings. However, these often fail in effectively addressing infections, as they struggle to both absorb exudates and maintain optimal local moisture. The system here presented was conceptualized with a three-layer design: the outer layer made of a fibrous polycaprolactone (PCL) film, to act as a barrier for preventing microorganisms and impurities from reaching the wound; the intermediate layer formed of a sodium alginate (SA) hydrogel loaded with ampicillin (Amp) for fighting infections; and the inner layer comprised of a fibrous film of PCL and polyethylene glycol (PEG) for facilitating cell recognition and preventing wound adhesion. Thermal evaluations, degradation, wettability and release behavior testing confirmed the system resistance overtime. The sandwich demonstrated the capability for absorbing exudates (≈70 %) and exhibited a controlled release of Amp for up to 24 h. Antimicrobial testing was performed against Staphylococcus aureus and Escherichia coli, as representatives of Gram-positive and Gram-negative bacteria: >99 % elimination of bacteria. Cell cytotoxicity assessments showed high cytocompatibility levels, confirming the safety of the proposed sandwich system. Adhesion assays confirmed the system ease of detaching without mechanical effort (0.37 N). Data established the efficiency of the sandwich-like system, suggesting promising applications in infected wound care.
Synthesis and characterization of a novel hydrogel based on carboxymethyl chitosan/sodium alginate with the ability to release simvastatin for chronic wound healing

Abstract:Since wound dressing has been considered a promising strategy to improve wound healing, recent attention has been focused on the development of modern wound dressings based on synthetic and bioactive polymers. In this study, we prepared a multifunctional wound dressing based on carboxymethyl chitosan/sodium alginate hydrogel containing a nanostructured lipid carrier in which simvastatin has been encapsulated. This dressing aimed to act as a barrier against pathogens, eliminate excess exudates, and accelerate wound healing by increasing the production of vascular endothelial growth factor (VEGF). Among various fabricated composites of dressing, the hydrogel composite with a carboxymethyl chitosan/sodium alginate ratio of 1:2 had an average pore size of about 98.44 ± 26.9 μm and showed 707 ± 31.9 % swelling and a 2116 ± 79.2 g/m2.per day water vapor transfer rate (WVTR), demonstrating appropriate properties for absorbing exudates and maintaining wound moisture. The nanostructured lipid carrier with optimum composition and properties had a spherical shape and uniform particle size distribution (74.46 ±7.9 nm). The prepared nanocomposite hydrogel displayed excellent antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria as well as high biocompatibility on L929 mouse fibroblast cells. It can release the loaded simvastatin drug slowly and over a prolonged period of time. The highest drug release occurred (80%) within 14 days. The results showed that this novel nanocomposite could be a promising candidate as a wound dressing for treating various chronic wounds in skin tissues.
Fabrication of Bacterial Cellulose-Based Dressings for Promoting Infected Wound Healing

Abstract:Bacterial cellulose (BC) holds several unique properties such as high water retention capability, flexibility, biocompatibility, and high absorption capacity. All these features make it a potential material for wound healing applications. However, it lacks antibacterial properties, which hampers its applications for infectious wound healings. This study reported BC-based dressings containing ε-polylysine (ε-PL), cross-linked by a biocompatible and mussel-inspired polydopamine (PDA) for promoting infectious wound healing. BC membranes were coated with PDA by a simple self-polymerization process, followed by treating with different contents of ε-PL. The resulted membranes showed strong antibacterial properties against tested bacteria by both in vitro and in vivo evaluations. The membranes also exhibited hemocompatibility and cytocompatibility by in vitro investigations. Moreover, the functionalized membranes promoted infected wound healing using Sprague–Dawley rats as a model animal. A complete wound healing was observed in the group treated with functionalized membranes, while wounds were still open for control and pure BC groups in the same duration. Histological investigations indicated that the thickness of newborn skin was greater and smoother in the groups treated with modified membranes in comparison to neat BC or control groups. Abstract:Abstract Chronic nonhealing wounds, such as pressure sores, venous leg ulcers and diabetic foot ulcers are a silent pandemic, and represent a significant economic and health burden, with billions exhausted per annum in the UK and USA alone. Chronic wounds are at high risk of infection with biofilms that are difficult to treat with broad-spectrum antibiotics. Another option is to develop a nontoxic dressing that can physically remove the live bacteria, thus reducing the risk of antimicrobial resistance (AMR). Thousands of wound dressings are commercially available to combat various degrees of wounding. Polydimethylsiloxane (PDMS), a biocompatible and inexpensive polymer to fabricate, can undergo various modifications. The ability of fabricated, patterned polymers to attract the opportunist Staphylococcus aureus and Pseudomonas aeruginosa, either from the planktonic state or while sessile in biofilms, on ex vivo human skin was investigated. These are the most common species isolated from chronic wounds, contributing to prolific biofilm formation, and decreasing antimicrobial efficiency. Different combinations of PDMS polymers patterned (PT) or flat (FL) ± triclosan (1% or 10%) were assessed with regard to their ability for selective bacterial attachment. Patterned PDMS + 10% triclosan (PT 10%) attracted significantly more live S. aureus and P. aeruginosa, as determined via colony-forming units (CFU) mL–1 (P < 0.01), scanning electron microscopy (P < 0.01) and confocal scanning laser microscopy (P < 0.01). The released triclosan was not cytotoxic against either type of bacteria (P < 0.05) or primary cultures of human dermal fibroblasts (P < 0.05). High performance liquid chromatography analysis of the culture medium confirmed a low level of phase release. Dermal fibroblast viability increased in the presence of PDMS, when subject to infection in co-culture via the Boyden chamber assay (P < 0.05). Compared to all the other polymer combinations, PT 10% demonstrated superior removal of biofilms that had been established on human ex vivo epidermis (P < 0.5). Histological examination of ex vivo full-thickness human skin confirmed the superior attraction qualities of PT 10% on the removal of the bacterial biofilm. In summary, the unique topography of PDMS combined with triclosan attracted a substantial number of live bacteria, in their planktonic state, and, significantly, when in an established biofilm. A coinfection model demonstrated the protective nature of the polymer on cultured dermal fibroblast viability by drawing the planktonic bacteria away from them in a co-culture model. Successful establishment of biofilms on epidermal sheets from full-thickness skin allowed the demonstration of the favourable polymer topography for biofilm removal from human skin. Defined patterned polymers also presented with higher attachment rates than nonpatterned polymers, highlighting the potential for engineering specific patterned polymers to transfer biofilms physically from wounds. Significantly, these polymers lack bactericidal properties, which is vital in the quest to combat antimicrobial resistance.

High Glucose Concentration Stimulates NHE-1 Activity in Distal Nephron Cells: the Role of the Mek/Erk1/2/p90RSK and p38MAPK Signaling Pathways

Abstract:Aims: In models of diabetes, distal nephron cells contribute to glucose uptake and oxidation. How these cells contribute to the use of glucose for the regulation of H+ extrusion remains unknown. We used Madin-Darby Canine Kidney (MDCK) cells to investigate the effect of acute or chronic high glucose concentration on the abundance and activity of the Na+/H+ exchanger (NHE-1). Methods: Using RT-PCR, we also evaluated the mRNA expression for sodium glucose co-transporters SGLT1 and SGLT2. Protein abundance was analyzed using immunoblotting, and intracellular pH (pHi) recovery was evaluated using microscopy in conjunction with the fluorescent probe BCECF/AM. The Na+-dependent pHi recovery rate was monitored with HOE-694 (50 µM) and/or S3226 (10 µM), specific NHE-1 and NHE-3 inhibitors. Results: MDCK cells did not express the mRNA for SGLT1 or SGLT2 but did express the GLUT2, NHE-1 and NHE-3 proteins. Under control conditions, we observed a greater contribution of NHE-1 to pHi recovery relative to the other H+ transporters. Acute high glucose treatment increased the HOE-694-sensitive pHi recovery rate and p-Erk1/2 and p90RSK abundance. These parameters were reduced by PD-98059, a Mek inhibitor (1 µM). Chronic high glucose treatment also increased the HOE-694-sensitive pHi recovery rate and p-p38MAPK abundance. Both parameters were reduced by SB-203580, a p38MAPK inhibitor (10 µM). Conclusion: These results suggested that extracellular high glucose stimulated NHE-1 acutely and chronically through Mek/Erk1/2/p90RSK and p38MAPK pathways, respectively.
In Situ Hydrogel Formulation for Advanced Wound Dressing: Influence of Co-Solvents and Functional Excipient on Tailored Alginate–Pectin–Chitosan Blend Gelation Kinetics, Adhesiveness, and Performance

Abstract:Chronic skin wounds affect more than 40 million patients worldwide, representing a huge problem for healthcare systems. This study elucidates the optimization of an in situ gelling polymer blend powder for biomedical applications through the use of co-solvents and functional excipients, underlining the possibility of tailoring microparticulate powder properties to generate, in situ, hydrogels with advanced properties that are able to improve wound management and patient well-being. The blend was composed of alginate, pectin, and chitosan (APC). Various co-solvents (ethanol, isopropanol, and acetone), and salt excipients (sodium bicarbonate and ammonium carbonate) were used to modulate the gelation kinetics, rheology, adhesiveness, and water vapor transmission rate of the gels. The use of co-solvents significantly influenced particle size (mean diameter ranging from 2.91 to 5.05 μm), depending on the solvent removal rate. Hydrogels obtained using ethanol were able to absorb over 15 times their weight in simulated wound fluid within just 5 min, whereas when sodium bicarbonate was used, complete gelation was achieved in less than 30 s. Such improvement was related to the internal microporous network typical of the particle matrix obtained with the use of co-solvents, whereas sodium bicarbonate was able to promote the formation of allowed particles. Specific formulations demonstrated an optimal water vapor transmission rate, enhanced viscoelastic properties, gel stiffness, and adhesiveness (7.7 to 9.9 kPa), facilitating an atraumatic removal post-use with minimized risk of unintended removal. Microscopic analysis unveiled that porous inner structures were influencing fluid uptake, gel formation, and transpiration. In summary, this study provided valuable insights for optimizing tailored APC hydrogels as advanced wound dressings for chronic wounds, including vascular ulcers, pressure ulcers, and partial and full-thickness wounds, characterized by a high production of exudate.

Abstract:Dual drug antibacterial wound dressings with biological materials possess crucial wound healing characteristics including biocompatibility, non-toxicity, degradability, mechanical strength and antibacterial properties. The study focusses on fabricating keratin (K)‐sodium alginate (A) based wound dressings by loading green synthesized zinc oxide nanoparticles (ZnO NPs) using C. roseus (leaf extract) and M. recutita (Chamomile flower part) herbal drug (CH) as a bioactive dual antibacterial wound dressing for the first time. The optimized ZnO NPs and CH exhibits strong physiochemical and electrostatic interactions (FT-IR, XRD and SEM) on the fabricated K-A-CH-ZnO biopolymeric mats. Moreover, the tiny porous network of the biopolymeric mat enhances thermal stability, hydrophilicity, mechanical strength and explores the water vapor transmission (2538.07 g/m 2 /day) and oxygen permeability (7.38 ± 0.31 g/m 2 ) to maintain moist environment and cell-material interactions. During enzymatic degradation studies, ZnO NPs and CH of biopolymeric mat not only retains structural integrity but also increases the characteristic of swelling with sustained drug release (57 %) in 144 h which accelerates wound healing process. Also, K-A-CH-ZnO mat exhibited excellent antibacterial effects against B. subtilis and E. coli . Furthermore, NIH 3T3 fibroblast cell behavior using MTT assay and in vivo evaluations of biopolymeric mat depicted enhanced biocompatibility with increased collagen deposition at the wound site as a prominent dual drug medicated antimicrobial wound dressing.

Abstract:Recently, managing the chronic skin wounds has become increasingly challenging for healthcare professionals due to the intricate orchestration of cellular and molecular processes involved that lead to the uncontrollable inflammatory reactions which hinder the healing process. Therefore, different types of wound dressings with immunomodulatory properties have been developed in recent years to effectively regulate the immune responses, enhance angiogenesis, promote re-epithelialization, and accelerate the wound healing process. This study aims to develop a new type of immunomodulatory wound dressing utilizing carboxymethyl cellulose (CMC)/sodium alginate (Alg)-simvastatin (SIM) to simultaneously enhance the inflammatory responses and the wound healing ratio. The CMC/Alg-SIM hydrogels exhibited appropriate swelling ratio, water vapor transmission rate, and desirable degradation rate, depending on the SIM content. The fabricated dressing showed sustained release of SIM (during 5 days) that improved the proliferation of skin cells. According to the in vitro findings, the CMC/Alg-SIM hydrogel exhibited controlled pro-inflammatory responses (decreased 2.5- and 1.6-times IL-6 and TNF-α, respectively) and improved secretion of anti-inflammatory cytokines (increased 1.5- and 1.3-times IL-10 and TGF-β, respectively) in comparison with CMC/Alg. Furthermore, the CMC/Alg-SIM hydrogel facilitated rapid wound healing in the rat model with a full-thickness skin defect. After 14 days post-surgery, the wound healing ratio in the CMC/Alg hydrogel group (∼93%) was significantly greater than the control group (∼58%). Therefore, the engineered CMC/Alg-SIM hydrogel with desired immunomodulatory properties possesses the potential to enhance and accelerate skin regeneration for the management of chronic wound healing.

Abstract:Alginate is a biocompatible polysaccharide matrix used for bioactive dressings with inherent healing properties. Most alginate dressings are produced as single-layer dressings. This study explores the potential of bilayer membranes to modulate drug release and enhance antimicrobial properties. We used alginate and chitosan loaded with simvastatin, an anti-inflammatory drug. One membrane comprised dense layers of both alginate and chitosan, while the other featured a dense alginate upper layer and a porous chitosan lower layer. The current study introduces a new approach in which a bilayer membrane is modeled instead of creating a polymeric blend between alginate and chitosan. The upper layer of the membrane contains only alginate loaded with simvastatin, while the bottom layer contains only chitosan. Another innovation is the study of the use of a porous lower layer of chitosan. Therefore, the association of these polymers in a bilayer and porous membrane gives advanced therapeutic dressings (with anti-inflammatory and antimicrobial properties intrinsic to the membrane) that are more efficient in the healing of complex wounds. Comprehensive characterization encompassed physicochemical, thermal, morphological, and mechanical properties. Microbiological tests were conducted using chitosan extract, and cytotoxicity evaluations were performed on fibroblast and keratinocyte cells. The results showed interlayer adhesion due to ionic interactions between alginate and chitosan surfaces. The drying process influenced the morphological and physicochemical features of the membranes. Simvastatin release profiles demonstrated sustained release over an extended period (approximately 60%–70% of the drug after 96 h). Storage assessments revealed that after six months, the membranes maintained around 98% of the initial simvastatin content. The antimicrobial activity test underscored the bacteriostatic efficacy of the chitosan porous layer, making it well-suited for infected wounds. Cell viability tests confirmed the non-cytotoxic nature of the films, highlighting their promising characteristics for treating diverse skin lesion types.

Abstract:The sustained release of antimicrobial therapeutics for wound dressing has become an attractive design strategy for prolonging the timespan of wound dressings and for reducing the risk of chronic wound infection. Recently, cellulose-based membrane has become a preferred option of wound dressings for the treatment of burn wounds and skin ulcers. In this work, novel cellulose membrane incorporated with mesoporous silica particles (SBA-15) was developed as an antimicrobial wound dressing with desirable sustained release functionality for targeting persistent bacterial pathogens. Attributed to a coated layer of calcium carbonate (CaCO3), SBA-15 particles were free from corrosion in alkaline condition during the preparation of cellulose-based composite membranes. SEM, TEM and BET results showed that the morphology, specific surface area, pore size and pore volume of pristine SBA-15 were preserved after the incorporation of CaCO3-coated SBA-15 into the cellulose matrix, while the mesoporous structure of SBA-15 was significantly disrupted without the use of CaCO3 coating. The resultant composite membranes containing 30 wt% SBA-15 (denoted as CM-Ca2-SBA(30%)) achieved 3.6 wt% of antimicrobial drug loading. Interestingly, CM-Ca2-SBA(30%) demonstrated the sustained release property of chloramphenicol for 270 h, driven by a two-stage drug release processes of SBA-15/cellulose. The water vapor permeability (WVTR) and swelling properties of composite membranes were shown to have complied with the primary requirements of wound dressing. Antibacterial assays revealed that strong antibacterial activities (144 h) of the composite membranes against Staphylococcus aureus and Eschericia coli were achieved. All results displayed that the strategy of coating silica with CaCO3 helps to obtain cellulose–silica composite membranes with desirable sustained release profiles and strong antibacterial activities. The antibacterial SBA-15/cellulose composite membranes show potential for the application of wound dressing.
