Abstract:A series of well-designed highly branched PEG-catechol based thermo-responsive copolymers were synthesized via a one-pot RAFT polymerization. A varying degree of photocrosslinkable (meth)acrylate moieties were incorporated within the 3D structure to allow on-demand photocuring (strong cohesion, unlike conventional PEG adhesives). At the same time, multitudes of free catechol groups inspired from adhesive proteins of marine mussels were also introduced in the hyperbranched structure, giving rise to adherence to skin and cardiac tissue. The resulting ambivalent PEG-catechol based copolymers were systematically studied to investigate the effects of polymer composition on tissue bioadhesive and swelling properties, comparing acrylates to methacrylates and PEG to 2-hydroxyethyl acrylamide (HEAA). It was proved that DOPA played a major role in the adhesion performance as it significantly enhanced the adhesion performances on varied substrates. The highly branched PEG-catechol copolymers demonstrate the great potential in the design of novel surgical glues, sealants or drug delivery vectors.

Abstract:Current medical adhesives based on cyanoacrylates typically exhibit cellular toxicity. In contrast, fibrin adhesives are non-toxic but have poor adhesive properties. To overcome these drawbacks we designed a simple and scalable adhesive precursor inspired by marine mussel adhesion that functioned with strong adhesion in wet conditions and with low cytotoxicity. Dopamine, an-amine derivative of an amino acid abundantly present in mussel adhesive proteins, was co-polymerised with a tri-functional vinyl monomer, to form a hyperbranched poly(β-amino ester) polymer termed poly(dopamine-co-acrylate) (PDA). A variety of molecular weights and crosslinking methods were analysed using an ex vivo porcine skin model and an almost 4 fold increase in wet adhesion strength was observed compared to TISSEEL® fibrin sealant. With a fast curing time, degradable properties and low cytotoxicity, PDA is highly attractive for medical purposes and could have a broad impact on surgeries where surgical tissue adhesives, sealants, and haemostatic agents are used.

Abstract:Tissue adhesives are a versatile and valuable alternative for wound closure. With fast application, the prevention of body fluid leakage and additional trauma to the wound, adhesives are desirable, especially in friable tissues. Although several options are already in the market, these present some drawbacks, namely poor adhesion in wet substrates and toxicity. Here, the main adhesives both synthetic and biomimetic, commercially available and those still in research, are analyzed with a focus on their adhesion mechanisms. At present, the strongest adhesive able to replace sutures in skin wounds is cyanoacrylate based. However it is toxic and therefore cannot be used internally. More recently alternatives have emerged, with PEG-based adhesives being biocompatible but mainly used as tissue sealants due to their low strength. The most recent approaches under development try to mimic the adhesion strategies of several organisms, such as the extensively studied blue mussels, sandcastle worms, barnacles and geckos. Although no ideal results have been achieved so far, large improvements have been accomplished in the last decade and research is reaching to a new level where the results are starting to meet the needs for medical applications.

Abstract:The development of new tissue adhesives is still in great demand as the current available tissue adhesives cannot meet the requirements of sophisticated surgeries. One major limitation of current adhesives is difficult to balance the adhesive strength, the mechanical property and the responsive degradation rate. Herein, we report a new injectable and self-healable hydrogel, which can function as a multi-responsive tissue adhesive with on-demand degradation profile under different external stimuli. The adhesive hydrogel is composed of aldehyde functionalized hyaluronic acid (HA-CHO) and a disulfide containing crosslinker - 3,3'-dithiobis(propionic hydrazide) (DTPH). With the optimized formulation, the gelation can be formed within 1 min in situ. Multi responsive properties were introduced into this adhesive hydrogel, namely (1) Strain-responsive; (2) Reduction-responsive; (3) Oxidation-responsive; (4) Enzyme-responsive; and (5) UV-responsive properties. Bovine serum albumin (BSA)/Glutaraldehyde hydrogel - a commercial tissue adhesive BioGlue (R), was selected as the control. Remarkably, this new hydrogel showed much high lap shear adhesive strength (up to 120 kPa) to the porcine skin, which surpasses 65.8% to BioGlue (R). (C) 2021 Elsevier Ltd. All rights reserved.

The HPEGDA polymers have been synthesized from controlled homopolymerization of poly(ethylene glycol) diacrylate (PEGDA 700) via in situ deactivation enhanced atom transfer radical polymerization (DE-ATRP). By introducing a high initiator-to-monomer ratio, the obtained HPEGDA polymer is composed of extremely short carbon-carbon backbones interconnected together by the long PEG chains as well as pendent photocrosslinkable acrylate moieties. Due to the extremely short C-C backbone, the long PEG chains can therefore be seen as the main chain, thus, HPEGDA polymers behave more like polyester which is a category of polymers that contain the ester functional group in their main chain. Photo-cured HPEGDA can be readily adhered to tissue forming a patch with robust mechanical and adhesive strengths. The degradation profile by hydrolysis of polyester blocks as well as a significantly low swelling ratio of HPEGDA gels in an aqueous environment allow them to have great potential for sealing and repair of internal tissue. Furthermore, HPEGDA gels appear to have minor significant cytotoxicity in vitro. These unique properties indicate that the reported HPEGDA polymers are well poised for the development of adhesive tissue engineering matrixes, wound dressings, and sealants.

Abstract:<p>Closure of wounds with tissue adhesives has many advantages over sutures, but existing synthetic adhesives are toxic and have poor workability. Blood-derived adhesives display complete resorption but have adhesion too weak for reliable wound dressings. We propose a semi-synthetic design that combines the positive attributes of synthetic and blood-derived tissue adhesives. PAMAM-g-diazirine (PDz) is a rapidly gelling bioadhesive miscible in both aqueous and organic solvents. PDz blended with platelet-rich plasma (PRP) forms PDz/PRP composite, a semi-synthetic formulation that combines PDz's wet tissue adhesion with PRP's potent wound healing properties. Light-activated PDz/PRP bioadhesive composite has similar elasticity to soft tissues and behaves as an induced hemostat—an unmet clinical need for rapid wound dressings. PDz/PRP composite applied to in vivo full-thickness wounds observed a 25% reduction in inflammation, as assessed by the host-cell response.</p>

Abstract:The need for wound closure or surgical procedures has been commonly met by application of sutures. Unfortunately, these are often invasive or subject to contamination (and consequent infection and negative impact on patients). Alternative solutions are offered by surgical adhesives that can be applied and set without major disruption; a new class of supramolecular‐based adhesives provide potential solutions to some of these challenges. In this study a series of polymers utilising dopamine as a self‐assembling unit were synthesized. It was found that these motifs act as extremely effective adhesives, with control over the mechanical strength of the adhesion and materials' tensile properties enabled by changing monomer feed ratios and levels of crosslinking. These materials significantly outperform commercially available bio‐adhesives, showing yield strengths after adhesion at least 2 times higher than that of BioGlue and Tisseel, as well as the ability to re‐adhere with significant recovery of adhesion strength. Promisingly, the materials were shown to be non‐cytotoxic, with cell viability > 90%, and able to perform in aqueous environments without significant loss in strength. Finally, the removal of the materials, after use as an adhesive, was possible using benign organic solvents such as ethanol, ensuring recyclability. These properties all demonstrate the effectiveness of the materials as potential bio‐adhesives, with potential advantages for use in surgery. This article is protected by copyright. Abstract:Although tissue adhesives have potential advantages over traditional sutures, existing ones suffer from several limitations: slow adhesion kinetic, low mechanical strength, and poor interfacial bonding with wet biological tissues. Herein, a cooperative mussel/slug double‐bioinspired hydrogel adhesive (DBHA) composed of a robust adhesive interface and a stretchable dissipative matrix is developed. The DBHA is formed by a cationic polysaccharide (chitosan), an anionic polysaccharide (carboxymethyl cellulose), and a barbell‐like dendritic lysine grafted with catechol groups. Compared to various commercial bio‐glues and traditional adhesives, the DBHA has significantly stronger tissue adhesion and enhanced toughness both ex vivo and in vivo. Meanwhile, the DBHA exhibits fast, strong, tough and durable adhesion to diverse ex vivo tissue surfaces with blood. The adhesion energy between the adhesive and porcine skin can reach 200–900 J m−2. Additionally, in vivo studies prove that DBHA has good hemostasis of rabbit artery trauma and achieves better wound healing of tissue incision than commercial bio‐glues. This study provides a novel strategy for fabricating fast and strong wet adhesives, which can be used in many applications, such as soft robots, tissue adhesives, and hemostats. Abstract:Glue-type bio-adhesives are in high demand for many applications, including hemostasis, wound closure, and integration of bioelectronic devices, due to their injectable ability and in situ adhesion. However, most glue-type bio-adhesives cannot be used for short-term tissue adhesion due to their weak instant cohesion. Here, we show a novel glue-type bio-adhesive based on the phase separation of proteins and polysaccharides by functionalizing polysaccharides with dopa. The bio-adhesive exhibits increased adhesion performance and enhanced phase separation behaviors. Because of the cohesion from phase separation and adhesion from dopa, the bio-adhesive shows excellent instant and long-term adhesion performance for both organic and inorganic substrates. The long-term adhesion strength of the bio-glue on wet tissues reached 1.48 MPa (shear strength), while the interfacial toughness reached ~880 J m−2. Due to the unique phase separation behaviors, the bio-glue can even work normally in aqueous environments. At last, the feasibility of this glue-type bio-adhesive in the adhesion of various visceral tissues in vitro was demonstrated to have excellent biocompatibility. Given the convenience of application, biocompatibility, and robust bio-adhesion, we anticipate the bio-glue may find broad biomedical and clinical applications.

Abstract:In this paper, a novel biocompatible and biodegradable tissue adhesive composed of poly(ethylene glycol)-methacrylate (PEGDMA) and thiolated chitosan (CSS) was prepared. PEGDMA and CSS cross-linked rapidly under physiological conditions through the Michael addition reaction via UV lamp irradiation. The chemical structures of PEGDMA and CSS were confirmed via FTIR and 1H NMR. The equilibrium swelling ratio and biodegradation of the hydrogels were tunable by varying the component ratios of the hydrogels. The compression strength and adhesive strength of the resulting hydrogels were measured with a tensile tester, and the adhesion strength of the hydrogel was higher than the fibrin glues. Moreover, the cytotoxicity of the PEGDMA/CSS hydrogels for L929 cells was evaluated by the MTT assay, and the results indicate that the photocured hydrogels are biocompatible and less cytotoxic towards the growth of L929 cells. These findings imply that the obtained hydrogel adhesives are a potential bioadhesive for clinical application in the future.

Abstract:To date, the robust and durable adhesion capability of hydrogel adhesives in wet environments remains a huge challenge. Herein, a physicochemically double-network crosslinked hydrogel matrix was prepared by mixing acrylic acid (AAc), chitosan (CS) and tannic acid (TA) as the main components and the subsequent in situ polymerization of AAc. The abundant reactive sites on the surface of the hydrogel matrix facilitate rapid, strong and repeatable adhesion to different surfaces of engineering solids and biological tissues in an aquatic environment. The formation of amide covalent bonds resulting from the addition of the bridging agent further expands the long-term application of the hydrogel in tissue repair, and the constructed hydrogel-tissue adhesive interface still has robust adhesion energy after soaking in a physiological environment for up to one month. Moreover, the hydrogel showed fantastic hemostatic performance due to its characteristics of platelet adhesion and high burst pressure. Overall, the persistent adhesion and excellent cytocompatibility of the hydrogel adhesive make it potentially applicable in medical adhesives.

Strong poly(ethylene oxide) based gel adhesives via oxime cross-linking

Abstract:There is a demand for materials to replace or augment the use of sutures and staples in surgical procedures. Currently available commercial surgical adhesives provide either high bond strength with biological toxicity or polymer and protein-based products that are biologically acceptable (though with potential sensitizing potential) but have much reduced bond strength. It is desirable to provide novel biocompatible and biodegradable surgical adhesives/sealants capable of high strength with minimal immune or inflammatory response. In this work, we report the end group derivatization of 8-arm star PEOs with aldehyde and amine end groups. Gels were prepared employing the Schiff-base chemistry between the aldehydes and the amines. Gel setting times, swelling behavior and rheological characterization were carried out for these gels. The mechanical-viscoelastic properties were found to be directly proportional to the crosslinking density of the gels, the 10K PEO gel was stiffer in comparison to the 20K PEO gel. The adhesive properties of these gels were tested using porcine skin and showed excellent adhesion properties. Cytotoxicity studies were carried out for the individual gel components using two different methods: (a) Crystal Violet Staining assay (CVS assay) and (b) impedance and cell index measurement by the xCELLigence system at concentrations >5%. Gels prepared by mixing 20% w/w solutions were also tested for cytotoxicity. The results revealed that the individual gel components as well as the prepared gels and their leachables were non-cytotoxic at these concentrations. Statement of significance: This work presents a new type of glue that is aimed at surgery applications using a water soluble star shaped polymer. It show excellent adhesion to skin and is tough and easy to use. We show that it is very biocompatible based on tests on live human cells, and could therefore in principle be used for internal surgery. Comparison with other reported and commercial glues shows that it is stronger than most, and does not swell in water to the same degree as many other water based bioadhesives.

Abstract:Engineered protein bioadhesives with instantaneous and strong adhesion performances are developed for clinical dynamic wound repair and noncompressible hemorrhage. They can be massively produced and showed outstanding preponderances in terms of adhesion performance, production cost, curing rate, and flexibility, overwhelming existing clinical bioglues. This work offers promising alternatives to current commercial bioadhesives for clinical surgery. Surgical adhesives are playing an important role in wound repair and emergency hemostasis in clinical treatment. However, the development of strong bioglue with rapid in situ adhesion, durable adhesiveness, and flexibility in dynamic and moist physiological environments is still challenging. Herein, a new type of biosynthetic protein bioadhesives with superior adhesion performance is reported by developing a protein aldimine condensation strategy. Lysine‐rich recombinant proteins are designed and massively biosynthesized to instantaneously react with aldehyde cross‐linkers to realize in situ strong adhesion. The obtained bioadhesives show an ultra‐high adhesion strength of ≈101.6 kPa on porcine skin, outperforming extant clinical bioglues. In addition, they possess super biocompatibility, flexibility, biodegradability, and compliance with the tissues. Owing to the strong and instantaneous adhesion properties, the bioadhesives are qualified for dynamic wound closure, facilitating wound repair, and noncompressible hemorrhage. Importantly, they can be industrially encapsulated into custom‐made cartridge delivery tubes at low cost for clinical use. Therefore, biosynthetic bioadhesives have great potential for biological applications and are capable of scaling up to the industrial level for clinical transformation, which will be a successful paradigm for reforming existing clinical products.

Abstract:The development of bioadhesives is an important, yet challenging task as seemingly mutually exclusive properties need to be combined in one material, i.e., strong adhesion, water resistance, and high biocompatibility. Here we reported a biocompatible and biodegradable protein‐based bioadhesive patch (PBP) with high adhesion strength and low immunogenic response. PBP existed a strong adhesion for biological surfaces, which is higher than some conventional bioadhesives (i.e., polyethylene glycol and fibrin). Robust adhesion and strength are realized through the removal of interfacial water and fast formation of multiple supramolecular interactions induced by metal ions. We evaluated the PBP's high biocompatibility and neglected immunogenic response in vitro and in vivo. The strong adhesion on soft biological tissues qualifies the PBP as biomedical glue outperforming some commercial products for applications in hemostasis performance, accelerated wound healing, and sealing of defected organs, anticipating to be useful as a tissue adhesive and sealant. Abstract:Tissue adhesives have garnered extensive interest as alternatives and supplements to sutures, whereas major challenges still remain, including weak tissue adhesion, inadequate biocompatibility, and uncontrolled biodegradation. Here, injectable and biocompatible hydrogel adhesives are developed via catalyst-free o- phthalaldehyde/amine (hydrazide) cross-linking reaction. The hydrogels demonstrate rapid and firm adhesion to various tissues, and an o- phthalaldehyde–mediated tissue adhesion mechanism is established. The hydrogel adhesives show controlled degradation profiles of 6 to 22 weeks in vivo through the incorporation of disulfide bonds into hydrogel network. In liver and blood vessel injury, the hydrogels effectively seal the incisions and rapidly stop bleeding. In rat and rabbit models of full-thickness skin incision, the hydrogel adhesives quickly close the incisions and accelerate wound healing, which exhibit efficacies superior to those of commercially available fibrin glue and cyanoacrylate glue. Development and thermo-mechanical characterization of cyanoacrylate-based tissue adhesives

Abstract:Tissue adhesives are crucial in current medical practice, facilitating the healing process in traumatic lacerations and surgical incisions. This work aims to develop and characterize tissue adhesives based on cyanoacrylate-based n-butyl and octyl blends. The characterization of three different formulations was achieved through Fourier Transform Infrared Spectroscopy (FTIR), Thermogravimetric Analysis (TGA), Differential Scanning Calorimetry (DSC). FTIR spectra were obtained between 200 and 4000 cm −1 , and the results indicated the presence of specific functional groups, specifically for the base monomers of the formulas between 1000 and 1200 cm −1 . The samples started to degrade around 160 °C and were completely degraded between 200 and 250 °C. Viscosity analysis showed that the shear rate range was from 300 to 1 s −1 , while the temperature varied between 20 and 37 °C. Dynamic Mechanical Thermo-Analysis (DMTA) showed similar storage and loss modulus under a temperature range of 0–80 °C. In contrast, a commercial standard sample, utilized as a comparison standard, showed the lowest values, about 25% less than other samples. Oscillatory rheology performed under 37 °C and varying the deformation amplitude (0–100%) with an oscillation frequency of 5 rad/s allowed a comparison between the samples evaluated and the results from DMTA testing. Mechanical characterization was achieved through a comparison between these results and the correspondent results of mechanical testing as per ASTM standards applicable to tissue adhesives—F2258-05, F2255-05, F2256-05, and F2458-05. They suggest the utilization of a viscous n-butyl and 2-octyl cyanoacrylate-based blend, allying a better degree of biocompatibility with adequate mechanical performance.

Abstract:Medical adhesives are arising as an important clinical tool as adjuvants for sutures and staples in wound closure and healing, and in the achievement of hemostasis. However, clinical adhesives combining cytocompatibility, as well as strong and stable adhesion in physiological conditions are still in demand. Herein, a mussel‐inspired strategy is explored to produce adhesive coacervates using tannic acid (TA) and methacrylate pullulan (PUL‐MA). TA|PUL‐MA coacervates mainly comprise van der Waals forces and hydrophobic interactions. The methacrylic groups in PUL backbone increase the number of interactions in the adhesives matrix, resulting in enhanced cohesion and adhesion strength (72.7 Jm−2), comparing to the non‐methacrylated coacervate. The adhesive properties were kept in physiologic‐mimetic solutions (72.8 Jm−2) for 72 h. The photopolymerization of TA|PUL‐MA enabled the on‐demand detachment of the adhesive. The poor cytocompatibility associated with the use of phenolic groups was here circumvented by mixing reactive oxygen species (ROS)‐degrading enzyme in the adhesive coacervate. This addition did not hamper the adhesive character of the materials, neither their anti‐microbial nor hemostatic properties. This affordable and straightforward methodology, together with the tailorable adhesivity even in wet environments, high cytocompatibility and anti‐bacterial activity, enable foreseeing TA|PUL‐MA as a promising ready‐to‐use bioadhesive for biomedical applications.
