Abstract:Modern micro/nanorobots can perform multiple tasks for biomedical and environmental applications. Particularly, magnetic microrobots can be completely controlled by a rotating magnetic field and their motion powered and controlled without the use of toxic fuels, which makes them most promising for biomedical application. Moreover, they are able to form swarms, allowing them to perform specific tasks at a larger scale than a single microrobot. In this work, we developed magnetic microrobots composed of halloysite nanotubes as backbone and Fe3O4 nanoparticles as magnetic material allowing magnetic propulsion and covered these with polyethylenimine to load ampicillin and prevent the microrobots from disassembling. These microrobots exhibit multimodal motion as single robots as well as in swarms. In addition, they can transform from tumbling to spinning motion and vice‐versa, and when in swarm mode they can change their motion from vortex to ribbon and back again. Finally, the vortex motion mode is used to penetrate and disrupt the extracellular matrix of Staphylococcus aureus biofilm colonized on titanium mesh used for bone restoration, which improves the effect of the antibiotic's activity. Such magnetic microrobots for biofilm removal from medical implants could reduce implant rejection and improve patients' well‐being. This article is protected by copyright. All rights reserved

Abstract:Targeted drug delivery using a microrobot is a promising technique capable of overcoming the limitations of conventional chemotherapy that relies on body circulation. However, most studies of microrobots used for drug delivery have only demonstrated simple mobility rather than precise targeting methods and prove the possibility of biodegradation of implanted microrobots after drug delivery. In this study, magnetically guided self‐rolled microrobot that enables autonomous navigation‐based targeted drug delivery, real‐time X‐ray imaging, and microrobot retrieval is proposed. The microrobot, composed of a self‐rolled body that is printed using focused light and a surface with magnetic nanoparticles attached, demonstrates the loading of doxorubicin and an X‐ray contrast agent for cancer therapy and X‐ray imaging. The microrobot is precisely mobilized to the lesion site through automated targeting using magnetic field control of an electromagnetic actuation system under real‐time X‐ray imaging. The photothermal effect using near‐infrared light reveals rapid drug release of the microrobot located at the lesion site. After drug delivery, the microrobot is recovered without potential toxicity by implantation or degradation using a magnetic‐field‐switchable coiled catheter. This microrobotic approach using automated control method of the therapeutic agents‐loaded microrobot has potential use in precise localized drug delivery systems.

Abstract:During recent years, microrobots have drawn extensive attention owing to their good controllability and great potential in biomedicine. Powered by external physical fields or chemical reactions, these untethered microdevices are promising candidates for in vivo complex tasks, such as targeted delivery, imaging and sensing, tissue engineering, hyperthermia, and assisted fertilization, among others. However, in clinical use, the biodegradability of microrobots is significant for avoiding toxic residue in the human body. The selection of biodegradable materials and the corresponding in vivo environment needed for degradation are increasingly receiving attention in this regard. This review aims at analyzing different types of biodegradable microrobots by critically discussing their advantages and limitations. The chemical degradation mechanisms behind biodegradable microrobots and their typical applications are also thoroughly investigated. Furthermore, we examine their feasibility and deal with the in vivo suitability of different biodegradable microrobots in terms of their degradation mechanisms; pathological environments; and corresponding biomedical applications, especially targeted delivery. Ultimately, we highlight the prevailing obstacles and perspective solutions, ranging from their manufacturing methods, control of movement, and degradation rate to insufficient and limited in vivo tests, that could be of benefit to forthcoming clinical applications.

Monte Carlo study of an unconventional superconducting phase in iridium oxide J(eff)=1/2 Mott insulators induced by carrier doping.

Abstract:Based on a microscopic theoretical study, we show that novel superconductivity is induced by carrier doping in layered perovskite Ir oxides where a strong spin-orbit coupling causes an effective total angular momentum J(eff)=1/2 Mott insulator. Using a variational Monte Carlo method, we find an unconventional superconducting state in the ground state phase diagram of a t(2g) three-orbital Hubbard model on the square lattice. This superconducting state is characterized by a d(x(2)-y(2))-wave "pseudospin singlet" formed by the J(eff)=1/2 Kramers doublet, which thus contains interorbital as well as both singlet and triplet components of t(2g) electrons. The superconducting state is found stable only by electron doping, but not by hole doping, for the case of carrier doped Sr2IrO4. We also study an effective single-orbital Hubbard model to discuss the similarities to high-T(c) cuprate superconductors and the multiorbital effects.
Programming self-assembling magnetic microrobots with multiple physical and chemical intelligence

Abstract:Medical microrobots represent the cutting-edge of biomedical research, showcasing their potential as versatile tools. They exhibit promise in acting as carriers for cancer cell therapy, effectively delivering drugs, and as manipulators equipped for biosensing, offering mobility and adaptability. Despite these advancements, the intricate challenge of creating a microrobot that seamlessly integrates various physical and chemical functionalities persists. This includes the fusion of selective sensing, manipulation capabilities, carrier functionality, precise time-based actuators for motion control, and adaptive shaping. Addressing these complexities remains an ongoing endeavor. In this context, our work introduces a pioneering magnetic microrobot founded on CaCO 3 microparticles (MPs) synthesized alongside polyethylenimine (CaCO 3 -PEI), forming the core body. This is combined with Fe 3 O 4 nanoparticles (NPs) enveloped in glutaraldehyde (Fe 3 O 4 -Glu), constituting the propulsive engine. The synergy of these elements enables the microrobot to execute multimodal motions, orchestrating its movement with finesse. This dynamic capability follows a "deliver-and-return" pattern for precise targeting applications with real-world relevance. Furthermore, the Fe 3 O 4 -Glu/CaCO 3 -PEI microrobots demonstrated remarkable proficiency in the targeted identification, manipulation, and transportation of cancer cells through the strategic integration of specific antibodies onto their structure. Within the realm of selective cancer cell detection, these microrobots adeptly function as dynamic mobile immunosensors. The versatile utility of the Fe 3 O 4 -Glu/CaCO 3 -PEI microrobots extends to their role as carriers for drugs and imaging agents, facilitated by the mediation of extracellular pH modulation in cancer cells orchestrated by CaCO 3 . This innovative work introduces a novel "on-the-fly" concept, revolutionizing the landscape of robotics programmed with multifaceted chemical and physical intelligences.

Abstract:Microrobots have attracted considerable attention due to their extensive applications in microobject manipulation and targeted drug delivery. To realize more complex micro-/nanocargo manipulation (e.g., encapsulation and release) in biological applications, it is highly desirable to endow microrobots with a shape-morphing adaptation to dynamic environments. Here, environmentally adaptive shape-morphing microrobots (SMMRs) have been developed by programmatically encoding different expansion rates in a pH-responsive hydrogel. Due to a combination with magnetic propulsion, a shape-morphing microcrab (SMMC) is able to perform targeted microparticle delivery, including gripping, transporting, and releasing by "opening-closing" of a claw. As a proof-of-concept demonstration, a shape-morphing microfish (SMMF) is designed to encapsulate a drug (doxorubicin (DOX)) by closing its mouth in phosphate-buffered saline (PBS, pH ∼ 7.4) and release the drug by opening its mouth in a slightly acidic solution (pH < 7). Furthermore, localized HeLa cell treatment in an artificial vascular network is realized by "opening-closing" of the SMMF mouth. With the continuous optimization of size, motion control, and imaging technology, these magnetic SMMRs will provide ideal platforms for complex microcargo operations and on-demand drug release.

Prediction of visual content across eye movements and their modulation by inferred information in the blind spot.

Abstract:The probabilistic view of brain function postulates that the brain operates by testing and refining predictions about the world. Here, we tested the existence of sensory predictive signals that could occur during eye movements. The local changes in acuity and global visual-field shifts of saccades could in principle be, at least partially, predicted. We analyzed whether these predictions are implemented differently for direct sensory information and for information that is exclusively inferred, as in the monocular blind spot area. This is performed by comparing the electroencephalogram (EEG) of responses after an eye movement to a peripheral visual stimulus, presented monocularly either inside or outside the blind spot. In some trials the stimulus remained the same pre and post eye-movement but in others it was exchanged. The EEG was analyzed using univariate general linear models, corrected for multiple comparison using threshold-free cluster based methods. We observe two main results: (1) early (~100 ms) and intermediate (~200 ms) lateralized EEG responses to saccade-contingent stimulus changes that suggest the occurrence of error signals to low and middle-level predictions within the visual modality. And (2), a late effect which was compatible in timing and topography with a P3 response, suggestive of violations of global constancy predictions. Remarkably, this late, change-related response was diminished when the change occurred in relation to a filled-in percept inside the subjects' blind spots. The results indicate that both sensory and associative predictive signals exist for transformations of visual input secondary to subjects' eye-movements. We show that these predictions occur across multiple levels of visual processing and are based on internal models that can differentiate between bottom-up input originating from the outside world or inferred from surrounding information. Physiological Mg2+ Conditions Significantly Alter the Inhibition of HIV-1 and HIV-2 Reverse Transcriptases by Nucleoside and Non-Nucleoside Inhibitors in Vitro.

Abstract:Reverse transcriptases (RTs) are typically assayed in vitro with 5-10 mM Mg2+, whereas the free Mg2+ concentration in cells is much lower. Artificially high Mg2+ concentrations used in vitro can misrepresent different properties of human immunodeficiency virus (HIV) RT, including fidelity, catalysis, pausing, and RNase H activity. Here, we analyzed nucleoside (NRTIs) and non-nucleoside RT inhibitors (NNRTIs) in primer extension assays at different concentrations of free Mg2+. At low concentrations of Mg2+, NRTIs and dideoxynucleotides (AZTTP, ddCTP, ddGTP, and 3TCTP) inhibited HIV-1 and HIV-2 RT synthesis less efficiently than they did with large amounts of Mg2+, whereas inhibition by the "translocation-defective RT inhibitor" EFdA (4'-ethynyl-2-fluoro-2'-deoxyadenosine) was unaffected by Mg2+ concentrations. Steady-state kinetic analyses revealed that the reduced level of inhibition at low Mg2+ concentrations resulted from a 3-9-fold (depending on the particular nucleotide and inhibitor) less efficient incorporation (based on kcat/Km) of these NRTIs under this condition compared to incorporation of natural dNTPs. In contrast, EFdATP was incorporated with an efficiency similar to that of its analogue dATP at low Mg2+ concentrations. Unlike NRTIs, NNRTIs (nevirapine, efavirenz, and rilviripine), were approximately 4-fold (based on IC50 values) more effective at low than at high Mg2+ concentrations. Drug-resistant HIV-1 RT mutants also displayed the Mg2+-dependent difference in susceptibility to NRTIs and NNRTIs. In summary, analyzing the efficiency of inhibitors under more physiologically relevant low-Mg2+ conditions yielded results dramatically different from those from measurements using commonly employed high-Mg2+ in vitro conditions. These results also emphasize differences in Mg2+ sensitivity between the translocation inhibitor EFdATP and other NRTIs.

Abstract:Microrobots and metal–organic frameworks (MOFs) have been identified as promising carriers for drug delivery applications. While clinical applications of microrobots are limited by their low drug loading efficiencies and the poor degradability of the materials used for their fabrication, MOFs lack motility and targeted drug delivery capabilities. The combination of these two fields marks the beginning of a new era; MOF‐based small‐scale robots (MOFBOTs) for biomedical applications. Yet, biodegradability is a major hurdle in the field of micro‐ and nanoswimmers including small‐scale robots. Here, a highly integrated MOFBOT that is able to realize magnetic locomotion, drug delivery, and selective degradation in cell cultures is reported for the first time. The MOF used in the investigations does not only allow a superior loading of chemotherapeutic drugs and their controlled release via a pH‐responsive degradation but it also enables the controlled locomotion of enzymatically biodegradable gelatin‐based helical microrobots under magnetic fields. The degradation of the integrated MOFBOT is observed after two weeks, when all its components fully degrade. Additionally, drug delivery studies performed in cancer cell cultures show reduced viability upon delivery of Doxorubicin within short time frames. This MOFBOT system opens new avenues for highly integrated fully biodegradable small‐scale robots.

Abstract:Microrobots hold broad application prospects in the field of precision medicine, such as intravenous drug injection, tumor resection, opening blood vessel and imaging during abdominal surgery. However, the rapid and controllable preparation of biocompatible hydrogel microparticles still poses challenges. This study proposes the one-step direct acquisition of biocompatible sodium alginate and gelatin methacrylate (GelMA) hydrogel microparticles using a oil-free aqueous solution, ensuring production with controllable generation frequency. An adaptive interface shearing platform is established to fabricate Alginate/GelMA microparticles using .a mixture of hydrogel, photoinitiator, and Fe3O4 nanoparticles (NPs). By adjusting the static magnetic field intensity (Bs), vibration frequency, and flow rate (Q) of dispersed phase, the size and morphology of the hydrogel microparticles can be controlled. These hydrogel microparticle robots exhibit magnetic responsiveness, demonstrating precise rotating and rolling movements under the influence of an externally rotating magnetic field (RMF). Moreover, the hydrogel microparticle robots with specific critical frequency (Cf) can be customized by adjusting the Bs and the concentration of Fe3O4 NPs. The directional in-situ untethered motion of the hydrogel microparticle robots can be successfully realized and accurately controlled in the climbing over obstacles and in vitro experiments of animals, respectively. This versatile and fully biodegradable microrobot has the potential to precisely control movement to bone tissue and the natural cavity of human body, as well as drug delivery.

Abstract:A multidrug delivery microrobot inspired by the fish structure with three components: skeleton, head, and body, is fabricated. The skeleton is made by poly(ethylene glycol) diacrylate (PEGDA) photoresist embedding with Fe3O4 nanoparticles and can respond to magnetic fields for microrobot actuation and drug‐targeted delivery. The drug storage structures, head, and body, made by biodegradable GelMA exhibit enzyme‐responsive drug release. Multidrug combination therapy provides an effective strategy for malignant tumor treatment. This paper presents the development of a biodegradable microrobot for on‐demand multidrug delivery. By combining magnetic targeting transportation with tumor therapy, it is hypothesized that loading multiple drugs on different regions of a single magnetic microrobot can enhance a synergistic effect for cancer treatment. The synergistic effect of using two drugs together is greater than that of using each drug separately. Here, a 3D‐printed microrobot inspired by the fish structure with three hydrogel components: skeleton, head, and body structures is demonstrated. Made of iron oxide (Fe3O4) nanoparticles embedded in poly(ethylene glycol) diacrylate (PEGDA), the skeleton can respond to magnetic fields for microrobot actuation and drug‐targeted delivery. The drug storage structures, head, and body, made by biodegradable gelatin methacryloyl (GelMA) exhibit enzyme‐responsive cargo release. The multidrug delivery microrobots carrying acetylsalicylic acid (ASA) and doxorubicin (DOX) in drug storage structures, respectively, exhibit the excellent synergistic effects of ASA and DOX by accelerating HeLa cell apoptosis and inhibiting HeLa cell metastasis. In vivo studies indicate that the microrobots improve the efficiency of tumor inhibition and induce a response to anti‐angiogenesis. The versatile multidrug delivery microrobot conceptualized here provides a way for developing effective combination therapy for cancer.

Abstract:Metal-organic frameworks (MOFs) hold significant potential as vehicles for drug delivery due to their expansive specific surface area, biocompatibility, and versatile attributes. Concurrently, magnetically actuated micro/nano-robots (MNRs) offer distinct advantages, such as untethered and precise manipulation. The fusion of these technologies presents a promising avenue for achieving non-invasive targeted drug delivery. Here, we report a MOF-based magnetic microrobot swarm (MMRS) for targeted therapy. Our approach overcomes limitations associated with a single MNR, including limited drug loading and the risk of loss during manipulation. We select Zeolitic Imidazolate Framework-8 (ZIF-8) as the drug vehicle for its superior loading potential and pH-sensitive decomposition. Our design incorporates magnetic responsive components into the one-pot synthesis of Fe@ZIF-8, enabling collective behaviors under actuation. Tuning the yaw angle of alternating magnetic fields and nanoparticles' amount, the MMRSs with controllable size achieve instantaneous transformation among different configurations, including vortex-like swarms, chain-like swarms, and elliptical swarms, facilitating adaptation to environmental variations. Transported to the subcutaneous T24 tumor site, the MMRSs with encapsulated doxorubicin (DOX) automatically degrade and release the drug, leading to a dramatic reduction of the tumor in vivo. Our investigation signifies a significant advancement in the integration of biodegradable MOFs into microrobot swarms, ushering in new avenues for accurate and non-invasive targeted drug delivery.

Immune Cell‐Based Microrobots for Remote Magnetic Actuation, Antitumor Activity, and Medical Imaging

Abstract:Translating medical microrobots into clinics requires tracking, localization, and performing assigned medical tasks at target locations, which can only happen when appropriate design, actuation mechanisms, and medical imaging systems are integrated into a single microrobot. Despite this, these parameters have not been fully considered when designing macrophage‐based microrobots. This study presents living macrophage‐based microrobots that combine macrophages with magnetic Janus particles coated with FePt nanofilm for magnetic steering and medical imaging and bacterial lipopolysaccharides for stimulating macrophages in a tumor‐killing state. The macrophage‐based microrobots combine wireless magnetic actuation, tracking with medical imaging techniques, and antitumor abilities. These microrobots are imaged under magnetic resonance imaging and optoacoustic imaging in soft‐tissue mimicking phantoms and ex vivo conditions. Magnetic actuation and real‐time imaging of microrobots are demonstrated under static and physiologically relevant flow conditions using optoacoustic imaging. Further, macrophage‐based microrobots are magnetically steered toward urinary bladder tumor spheroids and imaged with a handheld optoacoustic device, where the microrobots significantly reduced the viability of tumor spheroids. The proposed approach demonstrates the proof‐of‐concept feasibility of integrating macrophage‐based microrobots into clinic imaging modalities for cancer targeting and intervention, and can also be implemented for various other medical applications. High-performance Magnetic Metal Microrobot Prepared by a Two-Photon Polymerization and Sintering Method.

Abstract:Magnetically-actuated microrobots (MARs) exhibit great potential in biomedicine owing to their precise navigation, wireless actuation and remote operation in confined space. However, most previously explored MARs unfold the drawback of hypodynamic magnetic torque due to low magnetic content, leading to their limited applications in controlled locomotion in fast-flowing fluid and massive cargo carrying to the target position. Here, we report a high-performance pure-nickel magnetically-actuated microrobot (Ni-MAR), prepared by a femtosecond laser polymerization followed by sintering method. Our Ni-MAR possesses a high magnetic content (∼90 wt%), thus resulting in enhanced magnetic torque under low-strength rotating magnetic fields, which enables the microrobot to exhibit high-speed swimming and superior cargo carrying. The maximum velocity of our Ni-MAR, 12.5 body lengths per second, outperforms the velocity of traditional helical MARs. The high-speed Ni-MAR is capable of maintaining controlled locomotion in fast-flowing fluid. Moreover, the Ni-MAR with massive cargo carrying capability can push a 200-times heavier microcube with translation and rotation motion. A single cell and multiple cells can be transported facilely by a single Ni-MAR to the target position. This work provides a scheme for fabricating high-performance magnetic microrobots, which holds great promise for targeted therapy and drug delivery in vivo.
Autonomous Bionanorobots via a Cage-Shaped Silsesquioxane Vehicle for In Vivo Heavy Metal Detoxification

Abstract:Nanorobots hold great promise for integrated drug delivery systems that are responsive to molecular triggers. Herein, we successfully developed an automatic smart bionanorobot that has transport capability and recognizes and removes zinc ions from poisoned cells based on nanoscale polyhedral oligomeric silsesquioxane molecules. This intelligent bionanorobot can easily move inside and outside the cell and find zinc ions owing to its highly selective recognition to zinc ions and high cell permeability, especially the well-combined high penetration and strong binding energy. More importantly, it was also found that this intelligent bionanorobot can restore round HeLa cells to a normal fusiform cell morphology following high-concentration zinc treatment and does not interfere with cell proliferation and division. It was also shown by in vivo experiments that the bionanorobot can inhibit persistent enlargement of the liver caused by zinc ion poisoning.

Abstract:Abstract Microrobots have attracted great attentions due to their wide applications in microobjects manipulation and targeted drug delivery. To realize more complex micro/nano cargos manipulation (e.g., encapsulation and release) in biological applications, endowing microrobots with shapes adaptability with the environment is highly desirable. Here, designable shape-morphing microrobots (SMMRs) have been developed by programmatically encoding different expansion rate in a pH-responsive hydrogel. Combined with magnetic propelling, the shape-morphing microcrab (SMMC) is capable of performing targeted microparticle delivery, including gripping, transporting, and releasing through claws morphing. As a proof-of-concept demonstration, the shape-morphing microfish (SMMF) is designed to encapsulate drug (doxorubicin (DOX)) by closing mouth in phosphate buffer saline (PBS, pH~7.4) and release them by opening mouth in slightly acid solution (pH<7), which realize localized Hela cells treatment in an artificial vascular network. These SMMRs with powerful shape morphing capabilities and remote motion controllability provide new platforms for complex microcargos operation and on-demand drug release.

Abstract:Abstract Self-propelling magnetic nanorobots capable of intrinsic-navigation in biological fluids with enhanced pharmacokinetics and deeper tissue penetration implicates promising strategy in targeted cancer therapy. Here, multi-component magnetic nanobot designed by chemically conjugating magnetic Fe 3 O 4 nanoparticles (NPs), anti-epithelial cell adhesion molecule antibody (anti-EpCAM mAb) to multi-walled carbon nanotubes (CNT) loaded with an anticancer drug, doxorubicin hydrochloride (DOX) is reported. Autonomous propulsion of the nanobots and their external magnetic guidance is enabled by enriching Fe 3 O 4 NPs with dual catalytic-magnetic functionality. The nanobots propel at high velocities even in complex biological fluids. In addition, the nanobots preferably release DOX in the intracellular lysosomal compartment of human colorectal carcinoma (HCT116) cells by the opening of Fe 3 O 4 NP gate. Further, nanobot reduce ex vivo HCT116 tumor spheroids more efficiently than free DOX. The multicomponent nanobot’s design represents a more pronounced method in targeting tumors with self-assisted anticancer drug delivery for ‘far-reaching’ sites in treating cancers.

Abstract:Remarkable evolution of drug-loaded microrobots has aroused growing interest in targeted therapy. However, existing microrobots encounter a number of challenges including maneuverability, biodegradability, and drug sustained release capabilities in medical applications. Designed double-layer drug-loaded microrobots (TDMs) were made of outer layer calcium alginate hydrogel and inner layer magnetic chitosan microspheres (mCSs). TDMs precisely deliver drug-loaded mCSs following the expected route towards the predefined destinations under a vision-guided magnetic driving system. In acidic conditions the outer layer of TDMs can protect mCSs, while in alkaline conditions calcium alginate hydrogel dissolves at an appropriate rate to render mCSs be continuously released. The mCSs fulfill drug sustained release tasks in vitro. The new designed low-cost microrobots will be competent for intestinal cancer targeted therapy after oral administration and bridge the gap between scientific research and market demand.

Abstract:The study explores the use of magnetic cyclodextrin (CD) functionalized microrobots for cleaning water contaminated with drug residues, specifically methamphetamine. These microrobots, with a hematite/magnetite core and CD surface, enhance pollutant removal due to their combined magnetic movement and hydrophobic cavities of CDs on microrobot surfaces. The design emphasizes autonomous movement, improved mass transfer, and targeted functionalization for effective remediation. The growing consumption of drugs of abuse together with the inefficiency of the current wastewater treatment plants toward their presence has resulted in an emergent class of pollutants. Thus, the development of alternative approaches to remediate this environmental threat is urgently needed. Microrobots, combining autonomous motion with great tunability for the development of specific tasks, have turned into promising candidates to take on the challenge. Here, hybrid urchin‐like hematite (α‐Fe2O3) microparticles carrying magnetite (Fe3O4) nanoparticles and surface functionalization with organic β‐cyclodextrin (CD) molecules are prepared with the aim of on‐the‐fly encapsulation of illicit drugs into the linked CD cavities of moving microrobots. The resulting mag‐CD microrobots are tested against methamphetamine (MA), proving their ability for the removal of this psychoactive substance. A dramatically enhanced capture of MA from water with active magnetically powered microrobots when compared with static passive CD‐modified particles is demonstrated. This work shows the advantages of enhanced mass transfer provided by the externally controlled magnetic navigation in microrobots that together with the versatility of their design is an efficient strategy to clean polluted waters.
