Abstract:In order to achieve efficient bioconversion of L-glutamic acid to alpha ketoglutaric acid (α-KG),the L-glutamate oxidase (LGOX) gene from Kitasatospora setae KM-6054 was expressed in Escherichia coli BL21.According to the known LGOX sequence from K.setae and the codon bias of E.coli,the optimized LGOX gene sequence was synthesized and cloned into the pET28a (+) vector,which was then transferred into E.coli BL21(DE3) to obtain a recombinant LGOX.And enzymatic properties of the recombinant LGOX were also investigated.Results showed that the recombinant LGOX activity could reach 49.10 U/mL after induction at an IPTG concentration of 0.1 mmol/L at 20 ℃ for 18 h.Subsequently,the recombinant LGOX was purified by affinity column chromatography to a specific activity of 45.98 U/mg and its molecular weight was about 70 kDa as determined by SDS-PAGE analysis.The enzymatic properties showed that the optimal reaction temperature and pH value were 40 ℃ and 6.0,respectively.The Michaelis-Menten constant Km was 1.23 mmol/L,and the maximum reaction rate Vmax was 76.24 μmol/(min·mg).L-Glutamic acid was the optimal substrate for the LGOX.The heterologous expression and characterization of recombinant L-glutamate oxidase in this study can provide a new way for biosynthesis of α-KG.
Global Landscape of Lysine Acylomes in Bacillus Subtilis

Abstract:Lysine acetylation is a common posttranslational modification that regulates numerous biochemical functions in both eukaryotic and prokaryotic species. In addition, several new non-acetyl acylations are structurally different from lysine acetylation and participate in diverse physiological functions. Here, a comprehensive analysis of several lysine acylomes was performed by combining the high-affinity antibody enrichment with high-resolution LC-MS/MS. In total, we identified 2536 lysine acetylated sites, 4723 propionylated sites, 2150 succinylated sites and 3001 malonylated sites in Bacillus subtilis, respectively. These acylated proteins account for 35.8% of total protein in this bacterium. The four lysine acylomes showed a motif preference for glutamate surrounding the modified lysine residues, and a functional preference for several metabolic pathways, such as carbon metabolism, fatty acid metabolism, and ribosome. In addition, more protein-protein interaction clusters were identified in the propionylated substrates than other three lysine acylomes. In summary, our study presents a global landscape of acylation in the Gram-positive model organism Bacillus and their potential functions in metabolism and physiology.
Ac-LysargiNase Complements Trypsin for the Identification of Ubiquitinated Sites

Abstract:Mass spectrometry (MS)-based identification of ubiquitinated sites requires trypsin digestion prior to MS analysis, and a signature peptide was produced with a diglycine residue attached to the ubiquitinated lysine (K-ε-GG peptide). However, the missed cleavage of modified lysines by trypsin results in modified peptides with increased length and charge, whose detection by MS analysis is suppressed by the vast majority of internally unmodified peptides. LysargiNase, the mirrored trypsin, is reported to cleave before lysine and arginine residues and to be favorable for the identification of methylation and phosphorylation, but its digestive characteristics related to ubiquitination are unclear. Herein, we tested the capacity of the in-house developed acetylated LysargiNase (Ac-LysargiNase) with high activity and stability, for cleaving ubiquitinated sites in both the seven types of ubiquitin chains and their corresponding K-ε-GG peptides. Interestingly, Ac-LysargiNase could efficiently cleave the K63-linked chain but had little effect on the other types of chains. Additionally, Ac-LysargiNase had higher exopeptidase activity than trypsin. Utilizing these features of the paired mirror proteases, a workflow of trypsin and Ac-LysargiNase tandem digestion was developed for the identification of ubiquitinated proteins. Through this method, the charge states and ionization capacity of the unmodified peptides were efficiently reduced, and the identification of modified sites was consequently increased by 30% to 50%. Strikingly, approximately 15% of the modified sites were cleaved by Ac-LysargiNase, resulting in shorter K-ε-GG peptides for better identification. The enzyme Ac-LysargiNase is expected to serve as an option for increasing the efficiency of modified site identification in ubiquitome research.
Overexpression of Rhizomucor miehei Lipase and Activity Assay

Abstract:In order to overexpress the Rhizomucor miehei Lipase(RML) gene and obtain a large number of bioactive proteins in Escherichia coli.Firstly,the research improved the amount of the soluble protein through three methods:1) RML was introduced at 16 ℃.2) RML was fused with a new chaperone(Skp) which was located at the N terminal of RML;3) RML was fused with the Trx.tag in pET32a and the tag was located at the N terminal of RML.Secondly,the specific activity of purified RML derived from different methods was detected as(226±10~247±10) U/mg.The expression result showed that the low-temperature inducing was best method and the concentration of RML was 0.86 mg/mL.It was proposed that the temperature was the key factor of controling the amount of soluble protein.The activity test showed that fusion of Skp/Trx·tag and the N terminal of RML did not change the activity center(C terminal) and the catalytic ability of RML.Therefore,the strategy for overexpression of RML in E.coli kept the biologycal activity and was valuble in the practice.
LysargiNase and Chemical Derivatization Based Strategy for Facilitating In-Depth Profiling of C-Terminome.

Abstract:Global identification of protein C-termini is highly challenging due to their low abundance in conventional shotgun proteomics. Several enrichment strategies have been developed to facilitate the detection of C-terminal peptides. One major issue of previous approaches is the limited C-terminome coverage. Herein, we integrated LysargiNase digestion, chemical acetylation on neo-N-terminus, and a-ion-aided peptide matching into poly(allylamine)-based C-terminomics (termed as LAACTer). In this strategy, we leveraged LysargiNase, a protease with cleavage specificity N-terminal to Lys and Arg residues, to cover previously unidentifiable C-terminome and employed chemical acetylation and a-ion-aided peptide matching to efficiently boost peptide identifications. Triplicates of LAACTer identified a total of 834 C-termini from proteome of 293T cell, which expanded the coverage by 164% (643 more unique C-termini) compared with the parallel experiments using the original workflow. Compared with the largest human C-terminome data sets (containing 800-900 C-termini), LAACTer not only achieved comparable profiling depth but also yielded 465 previously unidentified C-termini. In a SILAC (stable isotope labeling with amino acids in cell culture)-based quantitative study for identification of GluC-cleaved products, LAACTer quantified 300% more C-terminal peptides than the original workflow. Using LAACTer and the original workflow, we performed global analysis for the C-terminal sequences of 293T cell. The original and processed C-termini displayed distinct sequence patterns, implying the "C-end rules" that regulates protein stability could be more complex than just amino acid motifs. In conclusion, we reason LAACTer could be a powerful proteomic tool for in-depth C-terminomics and would benefit better functional understanding of protein C-termini.
Functional expression and purification of bovine enterokinase light chain in recombinant Escherichia coli.

Abstract:Enterokinase (EC 3.4.21.9) is a serine proteinase of the intestinal brush border that exhibits specificity for the sequence (ASP)4-Lys and converts trypsinogen into its active form, trypsin. A codon optimized sequence coding light chain (catalytic subunit) of bovine enterokinase gene (sBEKLC) was synthesized, and it was fused with DsbA to construct the expression vector (pET39-sBEKLC). Then, the plasmid was transformed into E. coli BL21 (DE3) for expression. Under optimal conditions, the volumetric productivity of fusion protein reached 151.2mg L-1, i.e., 80.6 mg sBEKLC L-1. The cold osmotic shock technique was successfully used to extract sBEKLC from periplasmic space, and nickel affinity chromatography was employed to obtain mature sBEKLC. Finally, about 6.8 mg of bioactive sBEKLC was purified from 1 liter fermentation broth and could be used to cleave one tested fusion protein with an inter-domain enteropeptidase recognition site. This work will be helpful for large-scale production of this increasingly demanded enterokinase.
[Study on the Properties of a Novel Glycine Amino Peptidase from Actinomucor Elegans].

Abstract:The glycine amino peptidase of Actinomucor elegans was studied in this work. For the enzyme production Actinomucor elegans was cultured with an enzyme producing medium. Then the cells were collected and subjected to enzyme purification. The glycine aminopeptidase was purified 592 times by a DEAE-Toyopearl column, a Toyopearl HW 65-C column and a Superdex 200 column subsequently and the purified enzyme had a specific activity of 14.2 u/mg. The enzyme was estimated to have molecular mass of 320kD by gel filtration and a subunit size of 56.5kD by SDS-PAGE. It hydrolyzes glycine residue containing substrates such as glycine-betanaphthylamine more efficiently than those containing other amino acid residue. Addition to Gly-betaNA, the enzyme could also hydrolyze Ala-betaNA, Met-betaNA, Leu-betaNA, Arg-betaNA and Ser-betaNA but it had no activity on the substrates such as Trp-betaNA, Pyr-betaNA, Pro-betaNA, Asp-betaNA, Lys-betaNA, Val-betaNA. It was also observed when the glycine-betanaphthylamine concentration was higher than 2mmol/L the enzyme showed a substrate inhibition, and at the 20 mmol/L the enzyme only showed about 55% activity as it showed at the 2mmol/L. Whereas no such phenomenon was observed on the other substrate such as alanine-betanaphthylamine. The optimal temperature and pH for the reaction of this enzyme is 30 degrees C and pH 8.0, respectively. The Km and Kcat of the enzyme for glycine-betanaphthylamine is 0.24 mmol/L and 100.8 s(-1), respectively. Zn2+, Cu2+ and Cd2+ suppress almost all activities of the enzyme at the concentration of 1.0 mmol/L. Based on the study of chelating reagents, GAP belongs to the metalloenzyme. When a gelatin solution was hydrolyzed with 0.5% of alkaline proteinase together with glycine aminopeptidase at 50 degrees C for 18 hours, the glycine aminopeptidase could improve the hydrolysis degree of the protease. The total free amino acid was improved about 13% and although the enzyme mainly had the activity to hydrolyze the glycine residue, individual amino acids analysis with an amino acid analyzer showed that the contents of glycine, proline, alanine, arginine and glutamate were considerably increased. The results of this study showed that the glycine aminopeptidase would be useful in the food industry.

Abstract:The gene coding plectasin mature peptide isolated from a saprophytic fungus was optimized according to the E.coli codon preference,and inserted into a pET-32a(+)-TEV carrier.Then a recombinant plasmid pYG330 for high-level expression of recombinant plectasin fusion protein was constructed.Fusion protein were expressed in the soluble fraction.The fusion protein contained both His-tag label for affinity chromatography and specific enzymes recognition sites of TEV enzyme for fusion protein efficient cutting.High purity plectasin was achieved through three key purification processes,affinity chromatography after desalting,enrichment by freeze-dry,and another step of HPLC.Plectasin obtained in this study showed marked antibacterial activity against clinical drug-resistance bacteria of Staphylococcus aureus M-2 and Bacillus subtilis.
One-step purification and immobilization of his-tagged GL-7-ACA acylase

Abstract:GL-7-ACA acylase (GLA) is an enzyme that converts GL-7-ACA to 7-ACA, a starting material for semi-synthetic cephalosporin antibiotics. Four his-tagged GLAs were constructed in this work different in their N-terminal amino acid residues. Among them the highest activity of GLA reached 3800U/l in 19h for the recombinant strain BL21 (DE3)/pET28GA01. The immobilized GLA on TALON affinity support, agar-EPI-IDA-Co2+ and FP-IDA-Ni2+ was evaluated by one-step purification and immobilization, and the highest immobilized activity was 163U/g for TALON affinity support. But more robust matrix made FP-IDA-Ni2+ suitable for the industrial application. The activities of four immobilized GLAs on FP-IDA-Ni2+ ranged from 66.7 to 80U/g. Immobilized GLA showed good stability at pH value below 11.0 and at temperature up to 30°C. To the consecutive conversion of GL-7-ACA, immobilized GLA was recycled 21 times without significant loss of activity, and the average yield rate of 7-ACA reached 90%. These results indicated that intracellular production of his-tagged GLA in E. coli, followed by one-step affinity purification and immobilization on FP-IDA-Ni2+ resins, might serve as an effective process for the industrial application.
[Studies on the expression, purification and renaturation of recombinant N-acety-L-ornithine deacetylase]

Abstract:The argE gene from Escherichia coli coding for N-acety-L-ornithine deacetylase(NAOase), the key enzyme involved in the L-arginine biosynthesis, had been cloned in pET22b and transformed into BL21 (DE3). With 32.5% expression level in the optimal fermentation medium at 37 degrees C, most NAOase was expressed as inclusion bodies. The soluble and active proportion could be slightly increased when expressed at low temperature. The specific activity of soluble NAOase purified by Ni-NTA resin chromatography was 1193.2u/mg. The species and proportions of whole cell proteins varied with induction conditions. The inclusion bodies expressed at 37 degrees C was more pure than 22 degrees C after gradient wash with urea. Inclusion bodies could be partly refolding and reactivated by dilution and dialysis. Low protein concentration and suitable rate of oxidant/reducing agents were important to renaturation. In the optimal conditions 17.78% of Urea-denatured NAOase could be refolding and reactivated by dilution. The purified fusion protein was obtained after wash, solubilization and Ni-NTA resin affinity chromatography purification of inclusion bodies.

Abstract:Atrial natriuretic peptide (ANP), a small peptide consisting of 28 amino acids, has been applied in clinical treatment for heart failure, but it can encounter proteolytic degradation during its expression in host cells. Therefore, it is usually reported that ANP was expressed as a part of fusion protein. The aim of our study was to use an overexpression system to express the fusion protein REF-ANP and to optimize a purification method. First, Escherichia coli DH5alpha was transformed with constructed expression vector containing two tandem copies of ref-anp gene and the fusion protein REF-ANP was overexpressed in shaking flask culture. Subsequently, the inclusion bodies were purified with reverse phase chromatography and pooled fractions were lyophilized. After this step, REF-ANP can be solubilized under native conditions without urea. After cleavage reaction, the sample was subjected to size exclusion chromatography and then rANP was polished with reverse phase chromatography. The final purity of rANP was more than 98% and the recovery of rANP per liter of shaking flask culture was more than 3mg. Such methods as mass spectrometry, capillary isoelectrofocusing analysis, and N-terminal amino acid sequence were used to identify rANP. The capillary isoelectrofocusing analysis showed that the pI of ANP was about pH 9.7. In this study, an efficient refolding and purification process should make scaling-up procedures easier and more successful than earlier reports. Moreover, it is possible that the refolding and purification method along with the overexpression system described in this article may offer new ideas on optimizing expression and purification of other kinds of short peptides.
Heterologous expression and purification of the phage lysin-like bacteriocin LysL from Lactococcus lactis LAC460

Abstract:The wild-type Lactococcus lactis strain LAC460 produces two bacteriocin-like phage lysins, LysL and LysP. This study aimed to produce and secrete LysL in various heterologous hosts and an in vitro cell-free expression system for further functional studies. Initially, the lysL gene from L. lactis LAC460 was cloned into Lactococcus cremoris NZ9000 and L. lactis N8 strains, with and without the usp45 signal sequence (SSusp45), under a nisin-inducible promoter. Active LysL was primarily produced intracellularly in recombinant L. lactis N8, with some secretion into the supernatant. Recombinant L. cremoris NZ9000 lysed upon nisin induction, indicating successful lysL expression. However, fusion with Usp45 signal peptide (SPUsp45-LysL) weakened LysL activity, likely due to incomplete signal peptide cleavage during secretion. Active LysL was also produced in vitro, and analysed in SDS-PAGE, giving a 42-kDa band. However, the yield of LysL protein was still low when produced from recombinant lactococci or by in vitro expression system. Therefore, His-tagged LysL was produced in Escherichia coli BL21(DE3). Western blot confirmed the intracellular production of about 44-kDa His-tagged LysL in E. coli. His-tagged active LysL was then purified by Ni-NTA affinity chromatography yielding sufficient 4.34 mg of protein to be used in future functional studies.
High-Level Extracellular Expression of a New β-N-Acetylglucosaminidase in Escherichia coli for Producing GlcNAc

Abstract:N-acetyl-β-D glucosamine (GlcNAc) is wildly used in cosmetics, nutraceuticals and pharmaceuticals. The traditional chemical process for GlcNAc production from chitin causes serious acidic pollution. Therefore, the enzymatic hydrolysis becomes a great promising and alternative strategy to produce GlcNAc. β-N-acetylglucosaminidase (NAGase) can hydrolyze chitin to produce GlcNAc. Here, a GH3 family NAGase encoding gene BlNagZ from Bacillus licheniformis was expressed extracellularly in Escherichia coli guided by signal peptide PelB. The recombinant BlNagZ presented the best activity at 60°C and pH 5.5 with a high specific activity of 13.05 U/mg. The BlNagZ activity in the fermentation supernatant can reach 13.62 U/mL after optimizing the culture conditions, which is 4.25 times higher than optimization before. Finally, combining BlNagZ with chitinase ChiA we identified before, chitin conversion efficiency to GlcNAc can reach 89.2% within 3.5 h. In all, this study provided not only a high active NAGase, and a secreted expression strategy to reduce the cost of production, which is conducive to the industrial application.

Development of a rapid high-efficiency scalable process for acetylated Sus scrofa cationic trypsin production from Escherichia coli inclusion bodies

Abstract:Trypsin is one of the most important enzymatic tools in proteomics and biopharmaceutical studies. Here, we describe the complete recombinant expression and purification from a trypsinogen expression vector construct. The Sus scrofa cationic trypsin gene with a propeptide sequence was optimized according to Escherichia coli codon-usage bias and chemically synthesized. The gene was inserted into pET-11c plasmid to yield an expression vector. Using high-density E. coli fed-batch fermentation, trypsinogen was expressed in inclusion bodies at 1.47 g/L. The inclusion body was refolded with a high yield of 36%. The purified trypsinogen was then activated to produce trypsin. To address stability problems, the trypsin thus produced was acetylated. The final product was generated upon gel filtration. The final yield of acetylated trypsin was 182 mg/L from a 5-L fermenter. Our acetylated trypsin product demonstrated higher BAEE activity (30,100 BAEE unit/mg) than a commercial product (9500 BAEE unit/mg, Promega). It also demonstrated resistance to autolysis. This is the first report of production of acetylated recombinant trypsin that is stable and suitable for scale-up.
Expression and Purification of Soluble Human Neutrophil Gelatinase Associated Lipocalin in E.coli

Abstract:To obtain soluble recombinant human neutrophil gelatinase associated lipocalin (NGAL) by prokaryotic expression,firstly,the encoding sequence of human NGAL was amplified with RT-PCR and cloned into plasmid pET28a,which was then transformed into DH5a,to establish the prokaryotic expressing system.E.coli BL21 was transformed by the recombinant plasmid pET28a-MBPNGAL.Expression of the recombinant protein was induced with IPTG and assayed by SDS-PAGE and immunoblot.For scaled production,we purified the MBP-NGAL fused protein by using immobilized metal affinity chromatography (Ni2+-NTA agarose column)and cation-affinity purification,and then assayed recombinant products by SDS-PAGE and immunoblot.From the results,the molecular weight of the Expressed fusion-protein was 25 k in SDS-PAGE as expected.After Ni2 +-NTA agarose column,we obtained purified recombinant protein at a single band by SDS-PAGE electrophoresis.And MBP-NGAL fused protein could react specifically with commercial NGAL polyclonal antibody (Clone NL-2).The recombinant NGAL fusion protein was highly soluble,and the fusion protein has been purified to greater than 90％ purity from E.coli expression vector.In conclusion,we provide a method to produce large amounts of soluble recombinant human NGAL in E.coli,which may benefit for preparation of specific antibodies and diagnosis for some related diseases.
A channel allocation algorithm for Citizens Broadband Radio Service/Spectrum Access System

Abstract:Recently, the Federal Communications Commission has proposed to create Citizens Broadband Radio Service (CBRS) with the three-tier spectrum sharing system to release more spectrum for the mobile broadband usage in the United States. At the moment, the Wireless Innovation Forum is involved in defining the operational and functional requirements for this framework based on [1], and also in standardizations. Evidently, the successful operation of this system depends on efficient channel allocation algorithms. In this paper, we consider the CBRS with the three-tier spectrum sharing system. A set of CBRS users, i.e., Priority Access License (PAL) and General Authorized Access (GAA) users are located in an area, which is divided into multiple census tracts. The PAL and GAA users request frequency channels from the spectrum access system (SAS). The role of the SAS is to allocate channels to these two types of users while providing interference protection to the incumbent users. For this setup, we consider the problem of channel allocation for PAL and GAA users. The objective is to allocate channels for these two types of users, while considering the channel allocation rules proposed in [1]. For this problem, we propose a channel allocation algorithm to be used by the SAS. The proposed algorithm allocates channels to the CBRS users in two steps: 1) allocate channels to the PAL users and 2) allocate channel to the GAA users. Numerically, we show that the proposed algorithm is able to allocate channels while satisfying the rules proposed in [1]. More importantly, this research provides new insights on investigating channel allocation algorithms in CBRS/SAS.
Generation of Acetyllysine Antibodies and Affinity Enrichment of Acetylated Peptides

Abstract:Lysine acetylation has emerged as one of the major post-translational modifications, as indicated by its roles in chromatin remodeling, activation of transcription factors and, most recently, regulation of metabolic enzymes. Identification of acetylation sites in a protein is the first essential step for functional characterization of acetylation in physiological regulation. However, the study of the acetylome is hindered by the lack of suitable physical and biochemical properties of the acetyl group and existence of high-abundance acetylated histones in the cell, and needs a robust method to overcome these problems. Here we present protocols for (i) using chemically acetylated ovalbumin and synthetic acetylated peptide to generate a pan-acetyllysine antibody and a site-specific antibody to Lys288-acetylated argininosuccinate lyase, respectively; (ii) using subcellular fractionation to reduce highly abundant acetylated histones; and (iii) using acetyllysine antibody affinity purification and mass spectrometry to characterize acetylome of human liver tissue. The entire characterization procedure takes ∼2–3 d to complete.
Recombinant production and characterization of a metal ion-independent Lysophospholipase from a hyperthermophilic archaeon Pyrococcus abyssi DSM25543

Abstract:Genome sequence of Pyrococcus abyssi DSM25543 contains a coding sequence (PAB_RS01410) for α/β hydrolase (WP_010867387.1). Structural analysis revealed the presence of a consensus motif GXSXG and a highly conserved catalytic triad in the amino acid sequence of α/β hydrolase that were characteristic features of lysophospholipases. A putative lysophospholipase from P. abyssi with its potential applications in oil degumming and starch processing was heterologously produced in E. coli Rosetta (DE3) pLysS in soluble form followed by its purification and characterization. The recombinant enzyme was found to be active at temperature of 40–90 °C and pH 5.5–7.0. However, the enzyme exhibited its optimum activity at 65 °C and pH 6.5. None of the metal ions (Mn 2+ , Mg 2+ , Ni 2+ , Cu 2+ , Fe 2+ , Co 2+ , Zn 2+ and Ca 2+ ) being tested had stimulatory effect on lysophospholipase activity. K m and V max for hydrolysis of 4-nitrophenyl butyrate were calculated to be 1 ± 0.089 mM and 1637 ± 24.434 U/mg, respectively It is the first report on the soluble production and characterization of recombinant lysophospholipase from P. abyssi which exhibits its lipolytic activity in the absence of divalent metal ions. Broad substrate specificity, activity and stability at elevated temperatures make recombinant lysophospholipase an ideal candidate for potential industrial applications.

Simultaneous Expression of Glutaryl-7-aminocephalosporanic Acid Acylase Gene and Lysis Genes of Phage Λ in a Recombinant E. Coli

Abstract:Glutaryl-7-aminocephalosporanic acid acylase (GLA), recommended for use in the form of immobilized-enzyme, is one of the two key enzymes in the two-step synthesis of 7-aminocephalosporanic acid. For simplifying the process of cell disruption and immobilization, the lysis genes of phage λ (S−RRz) with the S amber mutation were designed to introduce into the over-expression system of GLA. A novel recombinant strain, E. coli TB1/pMKC-AS, simultaneously containing the maltose binding protein gene (malE), the lysis genes (S−RRz) and the target GLA gene (Acy) in a same operon, was successfully constructed. Under neutral pH conditions, cell growth and GLA activity of TB1/pMKC-AS was not affected by the presence of the lysis genes, however, autolysis phenomenon was observed under weak alkaline conditions. Through pH control and fed-batch culture, the GLA activity of TB1/pMKC-AS reached as high as 6810U/L with 24.8g/L dry cell density (OD600=67.9) in a 5L fermentor. In contrast to the cells of E. coli TB1/pMKC-Acy without the lysis genes, the mild EDTA/Tris buffer (pH 8.0) can cause the lysis of the cells of TB1/pMKC-AS containing the lysis genes. Correspondingly, a mild pH 9.0/42°C incubation method was developed for conveniently degrading the recombinant cells of TB1/pMKC-AS, based on the expression of the lysis genes. Further experiments showed that the cell lysate after the mild incubation disruption can be directly immobilized by 10% polyacrylamide to make the immobilized enzymes. In comparison with the immobilized GLA from TB1/pMKC-Acy, the immobilized cell lysate of TB1/pMKC-AS has the similar characteristics of catalysis stability, implying a great potential for industrial application of the lysis genes-assisted cell disruption.

Abstract:The expanding field of site-specific ligation of proteins and peptides has catalyzed the development of novel methods that enhance molecular modification. Among these methods, enzymatic strategies have emerged as dominant due to their specificity and efficiency in modifying proteins under mild conditions. Asparaginyl endopeptidase is a group of cyclotide-producing cysteine proteases from plants. These plant cysteine proteases, known for their specificity, effectively recognize the tripeptide motif (Asx-Xaa-Yaa) and cleave at the C-terminal side of Asx residues, forming acyl-enzyme intermediates that facilitate transpeptidation. Butelase 1 stands out as the most efficient AEP for protein engineering, yet challenges in its expression and purification limit its accessibility for widespread research and industrial use. To address these challenges, we engineered a new, catalytically efficient variant of Butelase 1, Butelase AY, by mutating the gatekeeping residues Val237Ala and Thr238Tyr within the LAD-1 region. These modifications significantly enhanced the stability and yield of Butelase AY, allowing for successful application in various peptide and protein engineering tasks. Butelase AY was tested on the peptide GLGKY, the globular protein GFP, and the intrinsically disordered protein α-synuclein, effectively labeling them with a fluorescent probe. Notably, Butelase AY maintained its efficiency with substrates containing unnatural amino acids, making it a promising candidate for biorthogonal applications. Importantly, the mutations did not compromise the enzyme's specificity, as it continued to process model peptides and native protein substrates with N-term NHV recognition motifs effectively. In conclusion, Butelase AY presents a novel recombinant tool for diverse protein labeling and modifications, particularly in biorthogonal strategies. This innovation has the potential to expand applications in biotechnology and therapeutic development, ultimately revolutionizing protein engineering and its utility in synthetic biology.
