Abstract:Stroke results in the formation of a cavity in the infarcted brain tissue. Angiogenesis and neurogenesis are poor in the cavity, preventing brain-tissue regeneration for stroke therapy. To regenerate brain tissue in the cavity, filamentous phages, the human-safe nanofiber-like bacteria-specific viruses, are genetically engineered to display many copies of RGD peptide on the sidewalls. The viral nanofibers, electrostatically coated on biocompatible injectable silk protein microparticles, not only promote adhesion, proliferation, and infiltration of neural stem cells (NSCs), but also induce NSCs to differentiate preferentially into neurons in basal medium within 3 d. After the NSC-loaded microparticles are injected into the stroke cavity of rat models, the phage nanofibers on the microparticles stimulate angiogenesis and neurogenesis in the stroke sites within two weeks for brain regeneration, leading to functional recovery of limb motor control of rats within 12 weeks. The viral nanofibers also brought about the desired outcomes for stroke therapy, such as reducing inflammatory response, decreasing thickness of astrocytes scars, and increasing neuroblasts response in the subventricular zone. As virtually any functional peptide can be displayed on the phage by genetic means, the phage nanofibers hold promise as a unique and effective injectable biomaterial for stroke therapy.
Viral vector eluting lenses for single-step targeted expression of genetically-encoded activity sensors for in vivo microendoscopic calcium imaging

Abstract:Optical methods for studying the brain offer powerful approaches for understanding how neural activity underlies complex behavior. These methods typically rely on genetically encoded sensors and actuators to monitor and control neural activity. For microendoscopic calcium imaging, injection of a virus followed by implantation of a lens probe is required to express a calcium sensor and enable optical access to the target brain region. This two-step process poses several challenges, chief among them being the risks associated with mistargeting and/or misalignment between virus expression zone, lens probe and target brain region. Here, we engineer an adeno-associated virus (AAV)-eluting polymer coating for gradient refractive index (GRIN) lenses enabling expression of a genetically encoded calcium indicator (GCaMP) directly within the brain region of interest upon implantation of the lens. This approach requires only one surgical step and guarantees alignment between GCaMP expression and lens in the brain. Additionally, the slow virus release from these coatings increases the working time for surgical implantation, expanding the brain regions and species amenable to this approach. These enhanced capabilities should accelerate neuroscience research utilizing optical methods and advance our understanding of the neural circuit mechanisms underlying brain function and behavior in health and disease.

Multichannel Fiber-Based Simultaneous Neural Stimulation and Signal Readout in Vivo

Abstract:In order to overcome the depth limitation of light penetration in brain tissue, a promising approach in optogenetics is implanting multimode optical fibers for stimulation and fluorescence imaging. However, traditional fiber photometry can only realize stimulation or recording in a single brain region, so optogenetic stimulation and simultaneous recording at the implantation site cannot be achieved. And there is currently no scheme for synchronization of multiple brain regions, therefore the connection between brain regions cannot be strictly proved. To overcome these issues, we developed a multichannel fiber-based in vivo simultaneous neural stimulation and signal readout system. Different from traditional fiber photometry, our system uses the galvanometer scanning technology and a specially-made optical fiber bundle with 200-μm diameter to realize high-precision multichannel stimulation with up to 1-kHz frequency and simultaneous recording. Combined with the specific light-sensitive proteins such as ChrimsonR and genetically encoded calcium indicator GCamP6s, we realize simultaneous optogenetic stimulation and recording of fluorescence signals representing neural activity in up to 7 brain regions of a mouse in vivo. At the same time, we performed optogenetic stimulation in the SNc/VTA brain region of 3 mice expressing the ChrimsonR light-sensitive protein, and recorded the fluorescence signal of the corresponding site simultaneously. In addition, our scheme is also suitable for stimulation and simultaneous recording of multiple brain regions in a single mouse, as well as signal recording and regulation in social and other behaviors in multiple mice. Our system and experimental method allow acquisition and analysis of functional information about relevant brain regions in specific behaviors, thus providing important data and theoretical support for further understanding and treatment of related diseases.

Abstract:Abstract Optogenetics combined with electrical recording has emerged as a powerful tool for investigating causal relationships between neural circuit activity and function. However, the size of optogenetically manipulated tissue is typically 1-2 orders of magnitude larger than that can be electrically recorded, rendering difficulty for assigning functional roles of recorded neurons. Here we report a viral vector-delivery optrode (VVD-optrode) system for precise integration of optogenetics and electrophysiology in the brain. Our system consists of flexible microelectrode filaments and fiber optics that are simultaneously self-assembled in a nanoliter-scale, viral vector-delivery polymer carrier. The highly localized delivery and neuronal expression of opsin genes at microelectrode-tissue interfaces ensure high spatial congruence between optogenetically manipulated and electrically recorded neuronal populations. We demonstrate that this multifunctional system is capable of optogenetic manipulation and electrical recording of spatially defined neuronal populations for three months, allowing precise and long-term studies of neural circuit functions.

A Precise and Minimally Invasive Approach to Optogenetics in the Awake Primate

Abstract:Optogenetics has proven to be a powerful tool for understanding the function of specific cell types and circuits within the central nervous system and establishing a causal link between their activity and behavior. Its application in non-human primates has been slow to develop. One challenge has been the damage caused by transdural delivery of viruses and light to the brain. Here, we report optogenetic activation of neuronal responses in the alert and behaving monkey after replacement of the native dura with a transparent artificial dura. This approach enables the use of fine glass micropipettes to inject virus with minimal damage and transdural illumination, obviating the damage that would otherwise occur as a result of lowering optical fibers into the brain. It also permits visualization of the underlying cortical micro-vasculature, which has proven to be helpful in targeting electrodes and laser illumination to the virus location. This approach promises to greatly assist in the dissection of cortical circuits underlying visual perception and behavior.
Intracerebroventricular Administration of AAV9-PHP.B SYN1-EmGFP Induces Widespread Transgene Expression in the Mouse and Monkey Central Nervous System

Abstract:Viral vectors made from adeno-associated virus (AAV) have emerged as preferred tools in basic and translational neuroscience research to introduce or modify genetic material in cells of interest. The use of viral vectors is particularly attractive in nontransgenic species, such as nonhuman primates. Injection of AAV solutions into the cerebrospinal fluid is an effective method to achieve a broad distribution of a transgene in the central nervous system. In this study, we conducted injections of AAV9-PHP.B, a recently described AAV capsid mutant, in the lateral ventricle of mice and rhesus macaques. To enhance the expression of the transgene (the tag protein emerald green fluorescent protein [EmGFP]), we used a gene promoter that confers high neuron-specific expression of the transgene, the human synapsin 1 (SYN1) promoter. The efficacy of the viral vector was first tested in mice. Our results show that intracerebroventricular injections of AAV9-PHP.B SYN1-EmGFP-woodchuck hepatitis virus posttranscriptional regulatory element resulted in neuronal EmGFP expression throughout the mice and monkey brains. We have provided a thorough characterization of the brain regions expressing EmGFP in both species. EmGFP was observed in neuronal cell bodies over the whole cerebral cortex and in the cerebellum, as well as in some subcortical regions, including the striatum and hippocampus. We also observed densely labeled neuropil in areas known to receive projections from these regions. Double fluorescence studies demonstrated that EmGFP was expressed by several types of neurons throughout the mouse and monkey brain. Our results demonstrate that a single injection in the lateral ventricle is an efficient method to obtain transgene expression in many cortical and subcortical regions, obviating the need of multiple intraparenchymal injections to cover large brain areas. The use of intraventricular injections of AAV9-PHP.B SYN1-EmGFP could provide a powerful approach to transduce widespread areas of the brain and may contribute to further development of methods to genetically target-specific populations of neurons.

Abstract:In gastropods that have been studied, there is some indication of the presence of neurons with specialized staining properties that are possibly neurosecretory. However, there are no stains specific for neurosecretion, and certain cellular inclusions can simulate secretory products. The presence of elementary neurosecretory granules does not as such determine neurosecretory status since such granules may represent neurotransmitters. Evidence of cyclical secretory activity of so-called neurosecretory neurons is most frequently related to reproductive phenomena, but there are also instances of control of water balance and hibernation. The existence of neurohemal organs within nerves and commissures has been described in several species. So-called neuroglandular complexes have been reported in association with the nervous system. Vesicles incorporated in the central nervous system may be sensory rather than glandular. Some caution is needed in estimating secretory activity of these complexes. Possible non-neural endocrine glands adjacent to the central ganglia have been described. In pulmonates granule-laden fibers are associated with the mediodorsal bodies; however, the nature of the neurosecretomotor or neurosecretory-neuropil control is still unclear, both morphologically and physiologically.

Abstract:Optogenetics combines optics and genetics to control neuronal activity with cell-type specificity and millisecond temporal precision. Its use in model organisms such as rodents, Drosophila, and Caenorhabditis elegans is now well-established. However, application of this technology in nonhuman primates (NHPs) has been slow to develop. One key challenge has been the delivery of viruses and light to the brain through the thick dura mater of NHPs, which can only be penetrated with large-diameter devices that damage the brain. The opacity of the NHP dura prevents visualization of the underlying cortex, limiting the spatial precision of virus injections, electrophysiological recordings, and photostimulation. Here, we describe a new optogenetics approach in which the native dura is replaced with an optically transparent artificial dura. This artificial dura can be penetrated with fine glass micropipettes, enabling precisely targeted injections of virus into brain tissue with minimal damage to cortex. The expression of optogenetic agents can be monitored visually over time. Most critically, this optical window permits targeted, noninvasive photostimulation and concomitant measurements of neuronal activity via intrinsic signal imaging and electrophysiological recordings. We present results from both anesthetized-paralyzed (optical imaging) and awake-behaving NHPs (electrophysiology). The improvements over current methods made possible by the artificial dura should enable the widespread use of optogenetic tools in NHP research, a key step toward the development of therapies for neuropsychiatric and neurological diseases in humans.
Untying a Nanoscale Knotted Polymer Structure to Linear Chains for Efficient Gene Delivery in Vitro and to the Brain

Abstract:The purpose of this study was to develop a platform transfection technology, for applications in the brain, which could transfect astrocytes without requiring cell specific functionalization and without the common cause of toxicity through high charge density. Here we show that a simple and scalable preparation technique can be used to produce a "knot" structured cationic polymer, where single growing chains can crosslink together via disulphide intramolecular crosslinks (internal cyclizations). This well-defined knot structure can thus "untie" under reducing conditions, showing a more favorable transfection profile for astrocytes compared to 25 kDa-PEI (48-fold), SuperFect® (39-fold) and Lipofectamine®2000 (18-fold) whilst maintaining neural cell viability at over 80% after four days of culture. The high transfection/lack of toxicity of this knot structured polymer in vitro, combined with its ability to mediate luciferase transgene expression in the adult rat brain, demonstrates its use as a platform transfection technology which should be investigated further for neurodegenerative disease therapies.

Abstract:Delivering genes to and across the brain vasculature efficiently and specifically across species remains a critical challenge for addressing neurological diseases. We have evolved adeno-associated virus (AAV9) capsids into vectors that transduce brain endothelial cells specifically and efficiently following systemic administration in wild-type mice with diverse genetic backgrounds, and in rats. These AAVs also exhibit superior transduction of the CNS across non-human primates (marmosets and rhesus macaques), and in ex vivo human brain slices, although the endothelial tropism is not conserved across species. The capsid modifications translate from AAV9 to other serotypes such as AAV1 and AAV-DJ, enabling serotype switching for sequential AAV administration in mice. We demonstrate that the endothelial-specific mouse capsids can be used to genetically engineer the blood-brain barrier by transforming the mouse brain vasculature into a functional biofactory. We apply this approach to Hevin knockout mice, where AAV-X1-mediated ectopic expression of the synaptogenic protein Sparcl1/Hevin in brain endothelial cells rescued synaptic deficits.
Flexible and stretchable polymer optical fibers for chronic brain and vagus nerve optogenetic stimulations in free-behaving animals

Abstract:Abstract Background Although electrical stimulation of the peripheral and central nervous systems has attracted much attention owing to its potential therapeutic effects on neuropsychiatric diseases, its non-cell-type-specific activation characteristics may hinder its wide clinical application. Unlike electrical methodologies, optogenetics has more recently been applied as a cell-specific approach for precise modulation of neural functions in vivo, for instance on the vagus nerve. The commonly used implantable optical waveguides are silica optical fibers, which for brain optogenetic stimulation (BOS) are usually fixed on the skull bone. However, due to the huge mismatch of mechanical properties between the stiff optical implants and deformable vagal tissues, vagus nerve optogenetic stimulation (VNOS) in free-behaving animals continues to be a great challenge. Results To resolve this issue, we developed a simplified method for the fabrication of flexible and stretchable polymer optical fibers (POFs), which show significantly improved characteristics for in vivo optogenetic applications, specifically a low Young’s modulus, high stretchability, improved biocompatibility, and long-term stability. We implanted the POFs into the primary motor cortex of C57 mice after the expression of CaMKIIα-ChR2-mCherry detected frequency-dependent neuronal activity and the behavioral changes during light delivery. The viability of POFs as implantable waveguides for VNOS was verified by the increased firing rate of the fast-spiking GABAergic interneurons recorded in the left vagus nerve of VGAT-ChR2 transgenic mice. Furthermore, VNOS was carried out in free-moving rodents via chronically implanted POFs, and an inhibitory influence on the cardiac system and an anxiolytic effect on behaviors was shown. Conclusion Our results demonstrate the feasibility and advantages of the use of POFs in chronic optogenetic modulations in both of the central and peripheral nervous systems, providing new information for the development of novel therapeutic strategies for the treatment of neuropsychiatric disorders.

ARViS: A bleed-free multi-site automated injection robot for accurate, fast, and dense delivery of virus to mouse and marmoset brains

Abstract:Genetically encoded fluorescent sensors continue to be developed and improved. If they could be expressed across multiple cortical areas in non-human primates, it would be possible to measure a variety of spatiotemporal dynamics of primate-specific cortical activity. Here, we develop an Automated Robotic Virus injection System (ARViS) for broad expression of a biosensor. ARViS consists of two technologies: image recognition of vasculature structures on the cortical surface to determine multiple injection sites without hitting them, and robotic control of micropipette insertion perpendicular to the cortical surface with 50-μm precision. In mouse cortex, ARViS sequentially injected virus solution into 100 sites over a duration of 100-minutes with a bleeding probability of only 0.1% per site. Furthermore, ARViS successfully achieved 266-site injections over the frontoparietal cortex of a common marmoset. We demonstrate one-photon and two-photon calcium imaging in the marmoset frontoparietal cortex, illustrating the effective expression of biosensors delivered by ARViS.

Abstract:Optogenetics has the potential to transform the study of the peripheral nervous system (PNS), but the complex anatomy of the PNS poses unique challenges for the focused delivery of light to specific tissues. This protocol describes the fabrication of a wireless telemetry system for studying peripheral sensory pathways. Unlike existing wireless approaches, the low-power wireless telemetry offers organ specificity via a sandwiched pre-curved tether, and enables high-throughput analysis of behavioral experiments with a channel isolation strategy. We describe the technical procedures for the construction of these devices, the wireless power transmission (TX) system with antenna coils, and their implementation for in vivo experimental applications. In total, the timeline of the procedure, including device fabrication, implantation, and preparation to begin in vivo experimentation can be completed in ~2-4 weeks. Implementation of these devices allows for chronic (>1 month) wireless optogenetic manipulation of peripheral neural pathways in freely behaving animals navigating homecage environments (up to 8).

Abstract:Recording and interrogating brain activities using optical methods have become emerging technologies in neuroscience. Traditional tools for optogenetic stimulation in the deep brain are mostly based on implantable fibers, imposing constraints on the animal movement. Recently developed microscale light-emitting diodes (micro-LEDs), which can be wirelessly operated, serve as injectable light sources that directly interact with neural systems. Here, we exploit a wirelessly controlled, implantable system for optogenetic studies in behaving animals. Thin-film indium gallium nitride (InGaN)-based blue micro-LEDs transferred onto flexible probes are injected into the animal brain and optically activate channelrhodopsin-2 expressing neurons. A customized circuit module with a battery is employed to modulate the micro-LED, which is remotely controlled at a distance up to 50mvia 2.4-GHz radio frequency communications. The systems are implemented on freely moving mice, and demonstrate optogenetic modulation of locomotive behaviors in vivo. Moreover, independent and synchronous control of multiple animals is accomplished with the communication unit in the design circuit. The proposed system provides the potential for advanced optical neural interfaces and offers solutions to study complicated animal behaviors in neuroscience research.

Abstract:cerebellar syndrome. These patients usually have cerebellar cortical atrophy.2 Autosomal dominant cerebellar ataxia resembles idiopathic cerebellar ataxia in many respects. Like idiopathic cerebellar ataxia, autosomal dominant cerebellar ataxia is a late onset disorder that may present as a pure cerebellar disorder or in combination with non-cerebellar symptoms. Neuropathological findings in idiopathic cerebellar ataxia and autosomal dominant cerebellar ataxia are often indistinguishable. These phenomenological similarities have prompted the hypothesis that at least some cases of idiopathic cerebellar ataxia arise from spontaneous mutation of a gene responsible for autosomal dominant cerebellar ataxia, from non-paternity, or falsely appear sporadic due to incomplete pedigree data.14 The recent discovery of an expanded trinucleotide repeat mutation in the SCAl region in American families with autosomal dominant cerebellar ataxia offers the opportunity to search for this mutation in isolated patients with idiopathic cerebellar ataxia by the polymerase chain reaction.5 We have analysed DNA from 61 patients with idiopathic cerebellar ataxia with a mean age of 53-6 (SD 12-8) years and a mean disease duration of 9-2 (SD 47) years. All patients satisfied the following diagnostic criteria: (a) progressive, otherwise unexplained ataxia, and (b) family history without evidence of heredity or consanguinity of parents. All diagnoses were made after exclusion of possible symptomatic causes (alcoholism, other toxic causes, malignancy, hypothyroidism, vitamin deficiency, inflammatory, metabolic, or vascular causes). Twenty five patients had a pure cerebellar syndrome (idiopathic cerebellar ataxia-C), whereas 36 had additional non-cerebellar symptoms (idiopathic cerebellar ataxia-P). Of these, 17 fulfilled the clinical criteria for probable multiple system atrophy.3 Polymerase chain reaction analysis was performed, as described by Orr et al.5 For comparison, DNA from 144 patients with autosomal dominant cerebellar ataxia was analysed. As a positive control, DNA from families with autosomal dominant cerebellar ataxia from Yakut tribes from Siberia was included, in which the SCAI expansion has been demonstrated and sequenced. A diagnosis of SCAl heterozygocity was made in five out of 19 German families with autosomal dominant cerebellar ataxia for which the family history was medically documented. This corresponds to a prevalence of SCAl in German families with autosomal dominant cerebellar ataxia of about 25%. By contrast, the SCAl expansion was found in none of the 61 cases of idiopathic cerebellar ataxia. The present results argue against the hypothesis that a relevant number of cases of idiopathic cerebellar ataxia are due to a trinucleotide expansion at the SCAl locus. The importance of our failure to show a mutation of the SCAl gene in patients with idiopathic cerebellar ataxia is underlined by the finding that the SCAl mutation is present in a considerable proportion of German families with autosomal dominant cerebellar ataxia. Our data do not exclude the possibility that some cases of idiopathic cerebellar ataxia are due to spontaneous mutations at other gene loci associated with autosomal dominant cerebellar ataxia. Linkage studies provide evidence that there are at least two other gene loci (SCA2 and SCA3) for autosomal dominant cerebellar ataxia.67 The hypothesis that idiopathic cerebellar ataxia is due to a mutation at one of these loci cannot be tested at present because SCA2 and SCA3 genes are not identified. It is unlikely that a recessive gene defect accounts for many cases of idiopathic cerebellar ataxia because recessive inheritance of a late onset ataxic disorder has been documented only very rarely. It therefore seems likely that most cases of idiopathic cerebellar ataxia are non-inherited. This hypothesis is supported by a recent study reporting discordance for idiopathic cerebellar ataxia in monzygous triplets.4 In addition, detailed clinical, neuroradiological, and neuropathological investigations show subtle differences between idiopathic cerebellar ataxia and autosomal dominant cerebellar ataxia. Thus age of onset is more variable and tends to be lower in autosomal dominant cerebellar ataxia. Additional noncerebellar features of idiopathic cerebellar ataxia are often parkinsonism and autonomic failure, whereas ophthalmoplegia, pyramidal signs, dystonia, and amyotrophy are more common in autosomal dominant cerebellar ataxia. Abstract:In vivo pharmacology and optogenetics hold tremendous promise for dissection of neural circuits, cellular signaling, and manipulating neurophysiological systems in awake, behaving animals. Existing neural interface technologies, such as metal cannulas connected to external drug supplies for pharmacological infusions and tethered fiber optics for optogenetics, are not ideal for minimally invasive, untethered studies on freely behaving animals. Here, we introduce wireless optofluidic neural probes that combine ultrathin, soft microfluidic drug delivery with cellular-scale inorganic light-emitting diode (μ-ILED) arrays. These probes are orders of magnitude smaller than cannulas and allow wireless, programmed spatiotemporal control of fluid delivery and photostimulation. We demonstrate these devices in freely moving animals to modify gene expression, deliver peptide ligands, and provide concurrent photostimulation with antagonist drug delivery to manipulate mesoaccumbens reward-related behavior. The minimally invasive operation of these probes forecasts utility in other organ systems and species, with potential for broad application in biomedical science, engineering, and medicine.

Feeding infants directly at the breast during the postpartum hospital stay is associated with increased breastfeeding at 6 months postpartum: a prospective cohort study

Abstract:Objective To explore whether feeding only directly from the breast in the first 24–48 h of life increases the proportion of infants receiving any breast milk at 6 months. Design A prospective cohort study. Setting Three maternity hospitals in Melbourne, Australia. Participants 1003 postpartum English-speaking women with a healthy singleton term infant, who intended to breast feed, were recruited between 2009 and 2011. Women were excluded if they or their infant were seriously ill. 92% (n=924) were followed up at 6 months postpartum. Primary and secondary outcome measures Main exposure variable —type of infant feeding in hospital up to time of study recruitment (24–48 h postpartum), categorised as ‘fed directly at the breast only’ or ‘received at least some expressed breast milk (EBM) or infant formula’. Primary outcome—proportion of infants receiving any breast milk feeding at 6 months postpartum. Secondary outcomes—proportion of infants receiving only breast milk feeding at 6 months; breast milk feeding duration; and maternal characteristics associated with giving any breast milk at 6 months. Results Infants who had fed only at the breast prior to recruitment were more likely to be continuing to have any breast milk at 6 months than those who had received any EBM and/or infant formula (76% vs 59%; adjusted OR 1.76, 95% CI 1.24 to 2.48 (adjusted for parity, type of birth, breastfeeding intention, breastfeeding problems at recruitment, public/private status, epidural for labour or birth, maternal body mass index and education)). Conclusions Healthy term infants that fed only directly at the breast 24–48 h after birth were more likely to be continuing to breast feed at 6 months than those who received any EBM and/or formula in the early postpartum period. Support and encouragement to initiate breastfeeding directly at the breast is important.

Abstract:Genetically encoded fluorescent sensors continue to be developed and improved. If they could be expressed across multiple cortical areas in non-human primates, it would be possible to measure a variety of spatiotemporal dynamics of primate-specific cortical activity. Here, we develop an Automated Robotic Virus injection System (ARViS) for broad expression of a biosensor. ARViS consists of two technologies: image recognition of vasculature structures on the cortical surface to determine multiple injection sites without hitting them, and robotic control of micropipette insertion perpendicular to the cortical surface with 50 μm precision. In mouse cortex, ARViS sequentially injected virus solution into 100 sites over a duration of 100 min with a bleeding probability of only 0.1% per site. Furthermore, ARViS successfully achieved 266-site injections over the frontoparietal cortex of a female common marmoset. We demonstrate one-photon and two-photon calcium imaging in the marmoset frontoparietal cortex, illustrating the effective expression of biosensors delivered by ARViS.

Abstract:Gene therapy offers hope for currently untreatable diseases; the patient's own cellular machinery is recruited to create therapeutics. However, unpredictable responses that lead to neutralization by the host immune system and issues in constraining, controlling and sustaining delivery have presented clinical barriers to otherwise promising therapeutic developments. Here, we show that the protective environment provided by advanced biomaterials can function as injectable gene carriers to focus their therapeutic potential. Firstly, we investigated the potential of a tissue-specific molecular hydrogel to package recombinant adeno-associated viruses (rAAVs). Once a delivery pathway was confirmed, a set of rAAV variants were subsequently assessed for their ability to transduce various types of rodent and human neural cells in vitro and in vivo . Based on GFP expression, we identified a relatively new variant, rAAV-DJ, as showing desirable characteristics for constrained delivery and transduction efficiency. For the first time, we demonstrated precise control over the strength and type of interaction between biomaterials and rAAVs enabling the programmed release of viral payloads. This new approach enables specific infection of desired anatomical targets in a programmed fashion.

Abstract:Calcium ions (Ca 2+ ) play pivotal roles in regulating diverse brain functions, including cognition, emotion, locomotion, and learning and memory. These functions are intricately regulated by a variety of Ca 2+ -dependent cellular processes, encompassing synaptic plasticity, neuro/gliotransmitter release, and gene expression. In our previous work, we developed 'monster OptoSTIM1' (monSTIM1), an improved OptoSTIM1 that selectively activates Ca 2+ -release–activated Ca 2+ (CRAC) channels in the plasma membrane through blue light, allowing precise control over intracellular Ca 2+ signaling and specific brain functions. However, the large size of the coding sequence of monSTIM1 poses a limitation for its widespread use, as it exceeds the packaging capacity of adeno-associated virus (AAV). To address this constraint, we have introduced monSTIM1 variants with reduced coding sequence sizes and established AAV-based systems for expressing them in neurons and glial cells in the mouse brain. Upon expression by AAVs, these monSTIM1 variants significantly increased the expression levels of cFos in neurons and astrocytes in the hippocampal CA1 region following non-invasive light illumination. The use of monSTIM1 variants offers a promising avenue for investigating the spatiotemporal roles of Ca 2+ -mediated cellular activities in various brain functions. Furthermore, this toolkit holds potential as a therapeutic strategy for addressing brain disorders associated with aberrant Ca 2+ signaling.
