Occurrence of Stem Rot on Canola Caused by Sclerotinia sclerotiorum in Argentina

Abstract:Canola (Brassica napus) was introduced as an alternative crop for wheat in Argentina. During 2003, typical symptoms of stem rot disease were observed on canola plants in two commercial fields located at Bragado, in northern Buenos Aires Province in Argentina. Average disease incidence across four canola cultivars was 21% (range = 13 to 29%). Symptoms included chlorosis and wilting of foliage and necrosis of basal stems. The disease appeared singly or in patches consisting of 4- to 5-month-old plants. The first visible symptom noticed was chlorosis and wilting of the foliage beginning from the basal leaves. Infection of the main stem at ground level typically was followed by a grayish white discoloration that progressed above the soil line to the apex. In advanced stages of the disease, stems and branches became bleached and eventually died. Black and irregularly shaped sclerotia (average size 5.5 × 2.8 mm) inside necrotic stem tissue were the typical signs of the pathogen. From September to October 2003, four samples consisting of six affected plants per sample were arbitrarily collected from two commercial fields located at Bragado. Sclerotia were taken from diseased stems, dipped in 70% ethanol, surface sterilized with 1% sodium hypochlorite for 1 min, and rinsed in sterile water. Each sclerotium was blotted dry on sterile Whatman's filter paper and placed on potato dextrose agar. Plates were incubated in the dark at 25°C for 2 to 3 days, followed by incubation under 12-h NUV light/12-h dark for 6 to 8 days. Six resulting colonies were identified as Sclerotinia sclerotiorum (Lib.) de Bary on the basis of taxonomic characteristics of the plant pathogenic species of Sclerotinia (3). Koch's postulates for three fungal isolates from infected plants were carried out on 6-week-old canola plants (cvs. Eclipse, Impulse, Master, and Mistral) by placing a colonized agar disk into wounds made in the basal stem region with a sterile scalpel. Pathogenicity tests, which included five inoculated and three control plants potted in a sterilized soil mix (soil/sand, 3:1), were conducted in a greenhouse at 23 to 26°C and 75% relative humidity with no supplemental light. Characteristic symptoms identical to the original observations developed within 12 days after inoculation on 100% of the inoculated plants for three isolates. Symptoms included wilted foliage, collapsed plants, and plant death. White mycelium and sclerotia developed on infected tissues, and the pathogen was successfully reisolated from symptomatic plants in all instances. Control plants, which were treated similarly except that the agar disk did not contain fungal growth, remained healthy. The experiment was repeated, and the results were identical to the first inoculations. Canola stem rot disease incited by S. sclerotiorum was first reported in Argentina during 1995 at experimental field plots in Buenos Aires. S. sclerotiorum, which has been reported to cause disease in canola in Canada (2) and the United States (1,4), currently represents a serious problem to the main canola cultivars grown in Argentina. To our knowledge, this is the first report of the occurrence of S. sclerotiorum causing a high incidence of stem rot in commercial crops of canola in Argentina. References: (1) D. F. Farr et al. Evaluation of L-asparaginase: polyethylene glycol conjugate versus native L-asparaginase combined with chemotherapy. A randomized double-blind study in canine lymphoma.

Abstract:L-asparaginase is an enzyme that inhibits protein synthesis by the depletion of sources of L-asparagine, which is necessary for transformed lymphoid cells to proliferate. L-asparaginase is used in the treatment of childhood acute lymphoblastic leukemia. A problem with L-asparaginase therapy is the immunogenicity of the enzyme and the development of anaphylactic reactions. Canine lymphoma is a predominantly B-cell tumor with widespread disease; without treatment, dogs with lymphoma usually survive 1-2 months. Canine lymphoma will respond to L-asparaginase therapy. A randomized double-blind study evaluated a polyethylene glycol (PEG) conjugate L-asparaginase combined with chemotherapy (vincristine, cyclophosphamide, doxorubicin, and prednisone). Thirty-five dogs were randomized to the PEG L-asparaginase group, and 34 dogs were randomized to the native L-asparaginase group. Thirty dogs (85.7%) achieved a complete remission (CR) with a median time to relapse of 217 days, and 32 (94.1%) dogs in the native L-asparaginase group achieved a CR with a median time to relapse of 214 days (P greater than 0.05). The asparaginase was well tolerated in both groups. Two dogs in the native L-asparaginase group had severe allergic reactions, and one dog in the PEG asparaginase group had a generalized urticarial reaction after repeated injections. This study indicates that PEG L-asparaginase has equal therapeutic efficacy to native L-asparaginase.
Diversity and Pathogenicity of Fusarium Root Rot Fungi from Canola (Brassica napus) in Alberta, Canada

Abstract:Root rot disease poses a significant threat to canola (Brassica napus), underscoring the need for a comprehensive understanding of its causal agents for more effective disease mitigation. The composition and diversity of fungal pathogens associated with root rot of canola in Alberta, Canada, were evaluated from plant tissue samples collected in 2021 and 2022. The study revealed Fusarium spp. as the predominant pathogens found in almost all surveyed fields. Fusarium avenaceum, F. redolens, and F. solani were among the most frequently recovered species. Greenhouse trials confirmed their pathogenicity, with F. avenaceum and F. sporotrichioides found to be particularly aggressive. Additionally, F. sporotrichioides and F. commune were identified for the first time as canola root rot pathogens. Inoculation with isolates of most species resulted in significant reductions in seedling emergence, plant height, and shoot and root dry weights. Analysis of translation elongation factor 1-α (TEF-1α) and internal transcribed spacer (ITS) sequences confirmed the identity of the Fusarium spp., while concatenating the ITS and TEF-1α sequences enabled improved species differentiation. Geographic and year effects did not influence fungal diversity or aggressiveness, as determined by principal component analysis. This study emphasized the high diversity and impact of Fusarium spp. in causing canola root rot.

In 2017, soybean plants exhibiting symptoms of root rot characterized by brown lesions on the tap and lateral roots at the beginning pod (R3) growth stage were collected from 15 fields representing eight counties in Henan Province. The symptomatic root tissues were surface-sterilized with 70% ethanol for 50 s and rinsed in sterilized water three times. Small pieces of symptomatic tissue were then placed on potato dextrose agar (PDA) and incubated at 25°C for 5 days in the dark. A total of 132 Fusarium-like colonies were isolated, and hyphal tips were transferred to fresh PDA plates to obtain pure cultures. Single-conidia isolates were then produced by dilution plating conidia from pure cultures and then subculturing from colonies that developed from the single-conidia isolations. The average mycelial growth rate was 5.3 to 8.7 mm/day at 25°C on PDA, and the colonies produced aerial mycelium varying from rose to yellow white, and rose to burgundy pigment in agar. Macroconidia of the isolates were produced on carnation leaf agar (CLA) incubated under black light and observed to be abundant, relatively slender, almost straight, commonly 3- to 7-septate, averaged 38.6 × 4.1 μm, matching that reported for F. pseudograminearum (Aoki and O’Donnell 1999). No perithecia were observed. DNA of isolates YB17SN07 and YB17SN08 from Yuanyang County and isolate YB17SN09 from Yongcheng County was extracted using the CTAB method. The translation elongation factor 1 alpha (EF1-α) gene was amplified and sequenced (O’Donnell et al. 1998). Results of sequences were deposited in GenBank (accession nos. MG189598 to MG189600). BLASTn analysis of the EF1-α gene shared 99 to 100% similarity with those of F. pseudograminearum (HBZX1675 and CBS131261). The pathogen identity was further confirmed using species-specific PCR primers, Fp1-1 and Fp1-2 (Aoki and O’Donnell 1999). To test pathogenicity, inoculum was grown for 10 days at 25°C on 100 g of sterilized boiled wheat grain and sand (3:1 v/v) after infesting with a 1-ml spore suspension (108 conidia/ml) of either isolate YB17SN07, YB17SN08, or YB17SN09. Five grams of this inoculum was placed 3 cm below the surface of autoclaved soil in 15 × 20 cm pots prior to planting pregerminated soybean seeds (cv. Wandou 35). Six plants were grown per pot, five pots were used per isolate, and the experiment was repeated three times. Noninoculated plants were grown in autoclaved soil amended with sterile wheat/sand mix and served as controls. Plants were grown in the greenhouse with a 14:10 h day/night cycle. After 42 days, the root system of all inoculated soybean plants exhibited dark brown lesions over the entire taproot. Noninoculated plants did not exhibit symptoms. The fungus was reisolated and identified from infected plants for each isolate as described above. This is the first report of F. pseudograminearum causing root rot on soybean in China. First report of Fusarium luffae causing soybean wilt in the USA

Abstract:During the 2023 soybean growing season in South Dakota, we scouted a farmer's field and observed soybean (Glycine max (L.) Merr.) plants with wilting symptoms and blighted leaves. Symptomatic stems and leaves were collected from the field to identify associated pathogens. 0.5 cm2 size leaf and stem segments of the sample were surface sterilized by rinsing with 10% bleach for 5 minutes then dipping in 70% ethanol for one minute, and later placing in deionized sterile water for one minute. The sterilized segments were placed on wet filter paper and incubated under fluorescent light for three days. Fungal growth was observed, and the growing mycelia were transferred to potato dextrose agar plates amended with 50 µg/ml Ampicillin (PDAa). Pure culture of the isolate was obtained using single sporing and transferring on new PDAa plates. A dense aerial mycelial growth showing waxy yellow color with a pale orange tinge on the rear side covered the full plate after seven days of incubation at room temperature under fluorescent lights (Figure S1a and b). Developing macroconidia were falcate, curved, smooth to slightly rough, and hyaline with three-five septa (Figure S1c). For molecular identification, DNA of the recovered isolate was extracted and subjected to multiloci PCR (O'Donnell et al., 2010) to amplify and Sanger sequence the internal transcribed spacers region (ITS) (GenBank accession number PP393518), calmodulin (CAM-PP401978), RNA polymerase II second largest subunit (RPB2-PP401980), and translation elongation factor 1-α gene (TEF1-PP401979). The South Dakota isolate (SLSDF2) was identified as Fusairum luffae on NCBI and Fusarioid polyphasic identification databases with 99.40% similarity to Fusarium luffae strain NRRL31167. A phylogeny was inferred based on concatenated TEF1, RPB2, and CAM sequences to show species relatedness (Figure S3). The characterized isolate SDSLF2 was evaluated for soybean pathogenicity using spray inoculations on detached leaves and V2 stage soybean plants (Figure S2a and b). The conidial suspension was prepared by growing the pathogen on mung bean agar for seven days. 2 ml of conidial suspensions (2.6 × 104 conidia/ml) and mock control (sterilized water with 0.1% Tween-20) was sprayed on the detached leaves and whole plants. The experiment was repeated three times with four replicates in each. In the detached leaf assay, leaves were completely blighted (Figure S2a) within 96 hours. In whole plant assays, after two days of incubation, leaf blighting was visible and progressed with time. Four days post-inoculation, the infected plants showed extensive leaf symptoms, and ultimately defoliation occurred (Figure S2b). No symptoms were observed in mock controls of either of the experiments. The pathogen was reisolated from the infected tissues and its identity was confirmed as F. luffae by CAM sequencing fulfilling Koch's postulates. F. luffae has been reported to associated with soybeans in China (Zhao et al., 2022), however, to our knowledge, this is the first report of F. luffae pathogenic on soybeans in the USA, stressing the need to identify resistance sources to avoid any potential disease epidemic.
First Report of Wilt Caused by Fusarium nanum (FIESC 25) on Mungbean (Vigna radiata) in Pakistan

Abstract:Mungbean, Vigna radia (L.) R. Wilczek, is ranked 2nd next to chickpea (Cicer arietinum) in total cultivation and production in Pakistan. In August of 2022 and 2023, mungbean plants (cv. PRI Mung-2018) were found wilting in a field at the Ayub Agricultural Research Institute, Faisalabad, Pakistan. Wilted leaves turned yellow, died, but remained attached to the stem. Vascular tissue at the base of the stem showed light to dark brown discoloration. Roots were stunted with purplish brown to black discoloration. Symptomatic mungbean plants were collected from fields at five different locations (20 samples/location). Disease incidence was similar among the five fields, ranging from 5 to 10% at each location depending upon type of germplasm and date of sowing. For fungal isolation and morphological identification, symptomatic stem and root tissues were cut into ~5 mm2 pieces with a sterilized blade. Tissues were surface-sterilized for one min in a 0.5% sodium hypochlorite solution, rinsed twice in sterilized water, air dried on sterilized filter paper, and aseptically placed on potato dextrose agar (PDA) containing 0.5 g/L-1 streptomycin sulphate. Plates were incubated for 3-4 days at 25 ± 2°C with a 12-h photoperiod. Single-spore cultures were used for morphological and molecular analyses. Isolates on PDA grew rapidly and produced abundant white aerial mycelium that turned off-white to beige with age. Macroconidia were hyaline, falcate, typically 3-to-6 septate with a pointed apical cell and a foot-shaped basal cell, measuring 24.5-49.5 x 2.7-4.7 μm (n = 40). Globose to obovate chlamydospores measuring 5.8 ± 0.5 μm (n = 40) were produced singly or in chains and were intercalary or terminal and possessed roughened walls. The morphological data indicated the isolates were members of the genus Fusarium (Leslie and Summerell 2006). To obtain a species-level identification, a portion of translation elongation factor 1-α (TEF1), the largest subunit of RNA polymerase (RPB1), and the second largest subunit of RNA polymerase (RPB2) region were PCR amplified and sequenced using EF1/EF2 (O'Donnell et al. 1998), Fa/G2R (Hofstetter et al. 2007), and 5f2/7cr (Liu et al. 1999) primers, respectively. DNA sequences of these genes were deposited in GenBank under accession numbers MW059021, MW059017 and MW059019, respectively. The partial TEF1, RPB1 and RPB2 sequences were queried against the Fusarium MLST database (https://fusarium.mycobank.org/page/Fusarium_identification), using the polyphasic identification tool. The BLASTn search revealed 99.9% identity of the isolate to F. nanum (Xia et al. 2019), formerly FIESC 25 of the F. incarnatum-equiseti species complex (MRC 2610, NRRL 54143; O'Donnell et al. 2018). To confirm pathogenicity, roots of 3-5 leaf stage mungbean seedlings were soaked in a 106 spores ml-1 conidial suspension of the fungus for 15 min and then planted in 10 cm pots containing sterilized soil. Mock-inoculated plants with sterile water served as a negative control. Twenty pots that were used for each inoculated and control treatment were maintained at 25 ± 2°C, 14:8 h photoperiod, and 80% relative humidity in a growth chamber. After 15 days, leaf yellowing, internal browning from the base of stems and root discoloration was observed in all the inoculated plants. The uninoculated negative control plants remained asymptomatic. Fusarium nanum was re-isolated from artificially inoculated plants and identified by colony growth, conidial characteristics on PDA and molecular analyses (TEF1). To our knowledge, this is the first report of wilt caused by F.nanum on mungbean in Pakistan. In Pakistan, mungbean cultivation in irrigated areas has increased in recent years. It has been introduced frequently in citrus orchards, crop rotation of maize and sesame, intercropping with sugarcane and as green manure. However, citrus, maize, sesame and sugarcane are also hosts of Fusarium spp. Therefore, this information warrants sustainable crop protection and may have an impact on further interaction of F. nanum with other wilt pathogens.
Fusarium and Fumonisin Occurrence in Argentinian Corn at Different Ear Maturity Stages

Abstract:The distribution of Fusarium species (Liseola section) and fumonisins B1, B2, and B3 (FB1, FB2, and FB3) in corn at the field stage to harvest time has been investigated. Corn samples were obtained from the 45th day after flowering to harvest stage, with 15 day intervals. A good correlation between fungal species and fumonisin contamination was observed. In particular, fumonisin contamination was higher in samples with fungal infection represented mainly by Fusarium moniliforme and Fusarium proliferatum, two well-known producers of fumonisins. Low levels of fumonisins were detected at the first and second samplings, when mainly Fusarium subglutinans, a fumonisin nonproducer species, was recorded. The three samplings after physiological maturity showed predominant F. moniliforme and F. proliferatum infection and a considerable contamination with fumonisins. In these samples the levels of FB1 averaged above 1 μg/g and FB2 and FB3 levels showed similar values but lower than FB1 values. Expression of CXCL16 in human rectal cancer.

Abstract:Local immunoregulation mediated by infiltration of inflammatory cells into colorectal adenocarcinomas is important for tumour progression. Tumour-associated macrophages and T cells are predominant components of chemokine-guided infiltrate of most colorectal tumours. CXCL16 is a newly discovered CXC chemokine expressed by antigen presenting cells attracting Th1, Tc and NK T cells. In this study, which is the first report on expression of the chemokine CXCL16 in human rectal cancer, CXCL16 gene and protein expression were analysed in cancer and normal adjacent tissue. Immunohistochemistry revealed CXCL16 expression in macrophages in normal tissue. The CXCL16 was found to a very limited extent in tumour-associated macrophages. Western blot analysis showed a suppression of CXCL16 protein in rectal cancer compared to non-cancer tissue in 83% of the patients (n=23, P=0.003). However, with real-time PCR mRNA was not down-regulated in the cancer compared to normal tissue, which may depend on regulated factor(s) at the level of translation and/or post-translation. The results may reflect one of the immunological mechanisms underlying carcinogenesis.

Abstract:Francisella tularensis, the causative agent of tularemia, can be spread by a variety of routes. Contaminated drinking water can serve as one of the means of transmission. The organism is of concern both as a cause of naturally occurring disease and as a potential agent of bioterrorism. This review article summarizes information on drinking water outbreaks, sources of contamination, and the detection and persistence of this bacterial pathogen in the aquatic environment. Information is provided on the adequacy of disinfection processes for controlling the organism in potable water supplies.
Phylogenetic, Phenotypic and Host Range Characterization of Five Fusarium Species Isolated from Chickpea in Alberta, Canada

Abstract:Chickpea (Cicer arietinum L.) is an important food legume worldwide, providing a source of high-quality protein. Fifteen chickpea samples showing leaf yellowing and root rot were obtained from southern Alberta, Canada, in 2018. Twenty single-spore Fusarium isolates were obtained from those samples. Based on the morphological characteristics, five species of Fusarium were identified, with F. redolens as the predominant species (11 isolates), followed by F. culmorum (3), F. sporotrichioides (3), F. oxysporum (2) and F. equiseti (1). The identification of each isolate was confirmed using partial sequences of the translation elongation factor 1-alpha (EF-1 alpha) gene. Each EF-1 alpha sequence displayed 99.54% to 100% similarity to sequences for each corresponding species in the GenBank database. The Fusarium isolates were tested for pathogenicity on chickpea 'CDC Frontier' in a greenhouse. Isolates recovered from inoculated plants were confirmed as the respective Fusarium species, fulfiling Koch's postulates. Selected isolates of each Fusarium species were able to infect common crops including barley, corn, canola, and lentil in Alberta. This is the first report of F. redolens on chickpea in Alberta, F. equiseti on chickpea in Canada, and F. culmorum and F. sporotrichioides on chickpea in the world.
First Report of Club Root Caused by Plasmodiophora brassicae on Canola in Australia

Abstract:In 2009, a disease survey was conducted in 97 commercial canola (Brassica napus L.) fields in Western Australia by the Department of Agriculture and Food, Western Australia (DAFWA). In about 20% of the fields from the northern agricultural region of Western Australia, small patches were observed where canola plants showed symptoms of stunting and wilting. These plants were collected and roots of affected plants were washed thoroughly and examined for the presence of root disease. Small galls and clublike structures were observed on the secondary roots and sometimes on the main root of the affected plants. Examination of thin free hand sections from the root galls revealed that several cortical cells were enlarged and full of resting spores. The diameter of resting spores ranged between 2.5 and 3.0 μm. Plasmodia and zoosporangia were also observed in the root hairs. The identity of Plasmodiophora brassicae Woronin was confirmed by PCR using a modified method of Cao et al. 2007 (1). DNA from spores and slices of the galls of 14 different samples were extracted using DNeasy plant mini kit (QIAGEN Australia) as per manufacturer's instructions. Samples were disrupted by placing them into MPBIO tube A and placed in the Fast Prep machine at speed of 6 ms-1 for 40 s. This was repeated twice. The species-specific primers TC1F (5'-GTGGTCGAACTTCATTAAATTTGGGCTCTT-3')/TC1R (5'-TTCACCTACGGAACGTATATGTGCATGTGA-3') and TC2F (5'-AAACAACGAGTCAGCTTGAATGCTAGTGTG-3')/TC2R (5'-CTTTAGTTGTGTTTCGGCTAGGATGGTTCG-3') were used (1). The primers TC1F and TC1R failed to produce a PCR product of 548-bp size but using the primers TC2F and TC2R the PCR reaction resulted in a 519- bp fragment. Seven out of 14 samples gave positive results for P. brassicae with primers TC2F and TC2R. This indicates that the P. brassicae pathotype from Western Australia may be different than the one found in Alberta, Canada. However, pathotypes of P. brassicae from brassica vegetables from Australia have been found similar to the populations of P. brassicae present in the United States (2). Pathogenicity of P. brassicae was tested by dipping roots of five 10-day-old canola plants var. Cobbler in a spore suspension (1 × 106 resting spores/ml). Roots of five control plants were dipped in sterile water. Five weeks after inoculation, small galls were observed on the roots of three inoculated plants and the control plants remained symptomless. Resting spores were recovered from the galls developed on the roots of affected plants. Presence of P. brassicae in the affected roots was further confirmed by PCR using the method described above. To our knowledge, this is the first report of club root of canola in Australia. Club root is reported from vegetable brassicas and white mustard (Sinapis alba L.) in Australia. Club root has become a serious disease of canola in Canada since its detection in Alberta in 2006 (3). The resting spores of the fungus can survive for several years in soil, and therefore, this disease could pose a significant threat to canola production in Western Australia. Abstract:Fusarium spp. are commonly associated with the root rot complex of soybean (Glycine max). Previous surveys identified six common Fusarium species from Manitoba, including F. oxysporum, F. redolens, F. graminearum, F. solani, F. avenaceum, and F. acuminatum. This study aimed to determine their pathogenicity, assess host resistance, and evaluate the genetic diversity of Fusarium spp. isolated from Canada. The pathogenicity of these species was tested on two soybean cultivars, ‘Akras’ (moderately resistant) and ‘B150Y1′ (susceptible), under greenhouse conditions. The aggressiveness of the fungal isolates varied, with root rot severities ranging from 1.5 to 3.3 on a 0–4 scale. Subsequently, the six species were used to screen a panel of 20 Canadian soybean cultivars for resistance in a greenhouse. Cluster and principal component analyses were conducted based on the same traits used in the pathogenicity study. Two cultivars, ‘P15T46R2′ and ‘B150Y1′, were consistently found to be tolerant to F. oxysporum, F. redolens, F. graminearum, and F. solani. To investigate the incidence and prevalence of Fusarium spp. in Canada, fungi were isolated from 106 soybean fields surveyed across Manitoba, Saskatchewan, Ontario, and Quebec. Eighty-three Fusarium isolates were evaluated based on morphology and with multiple PCR primers, and phylogenetic analyses indicated their diversity across the major soybean production regions of Canada. Overall, this study contributes valuable insights into host resistance and the pathogenicity and genetic diversity of Fusarium spp. in Canadian soybean fields.

Abstract:Diseased canola plants which failed to produce pods and were shorter than healthy plants in the same field were submitted to the Alberta Plant Health Laboratory in September 2016. The predominant symptom on infected plants was root rot, resembling a pink root rot. The objective of this study was to confirm the occurrence of canola pink root rot in Alberta. Two of the fungal strains isolated from symptomatic root samples were subjected to DNA barcoding that targeted three genomic regions, namely the internal transcribed spacer (ITS) region, the translation elongation factor-1 alpha gene (EF1) and the beta-tubulin gene. By similarity search (BlastN) using the resultant sequences as queries against GenBank accession sequences, both isolates were identified as Setophoma terrestris. Experiments were conducted to fulfil Koch's postulates to demonstrate that this fungal species is a pathogen of canola. After inoculation, the two strains could cause pink root rot on canola. The fungal strains were re-isolated from the inoculated canola and their identity confirmed as S. terrestris. This is the first report of pink root rot on canola and on any Brassica host in the world.
The Host Range of Fusarium proliferatum in Western Canada

Abstract:Fusarium proliferatum is associated with the root rot of many plant species, but knowledge of its impact on western Canadian field crops is limited. This study assessed the host range of this fungus and its effect on plant emergence, plant height, and shoot and root dry weights in repeated greenhouse experiments with wheat, barley, faba beans, peas, lentils, canola, lupine, and soybeans. Infection was confirmed via PCR, and principal component analysis determined the utility of different parameters in assessing host responses. All crops were at least partly susceptible, developing mild to severe disease at the seedling and adult stages, and showing significant reductions in growth. In general, the barley and wheat demonstrated higher tolerances to infection, followed by the faba bean and the pea. The soybean, canola, lupine, and lentil were most susceptible. The canola and the soybean were particularly vulnerable to F. proliferatum at the pre-emergence stage, while infection greatly reduced the lentil's biomass. Reductions in the barley's emergence and other growth parameters, however, occurred only under a high inoculum concentration. Variability in root rot severity among cultivars of the same crop indicated some diversity in host reactions within species. Nonetheless, the absence of fully-resistant crops may pose challenges in managing F. proliferatum in western Canadian cropping systems.

Abstract:Fusarium wilt is one of the most devastating diseases of cotton ( Gossypium spp.). It is caused by the soil-borne pathogen Fusarium oxysporum Schlechtend. f. sp. vasinfectum (Atk.) Snyd. & Hans (Atkinson 1892). To date, eight races of F. oxysporum f. sp. vasinfectum have been reported worldwide based on their diverse genetics and reactions to the host (Cianchetta et al. 2015). Fusarium wilt symptoms appear at all development stages. The fungus enters through the roots and colonizes the vascular system, leading to discoloration and wilting (Davis et al. 2006). During a survey in the July 2021 growing season, cotton plants showing typical wilting symptoms, stem discoloration, and root rot were observed in two cotton fields in Sumner County, Kansas. To confirm the causal agent, root tissues and lower parts of the stems were collected from 25 diseased cotton plants and cleaned as described by Larran et al. (2001). Small segments (1 to 2 cm) of roots and stems were incubated on potato dextrose agar (PDA) at room temperature. The isolates were purified with the single spore method and 23 of 25 isolates (92%) showed typical morphology of F. oxysporum , with a white-peach pigmentation as reported previously (Leslie and Summerell 2008). Two isolates, CK13B and CK18A, one from each field, were selected, and genomic DNA was extracted with E.Z.N.A Fungal DNA Mini kit (Omega Bio-tek, Norcross, GA). PCR assays were performed as reported previously (Ortiz et al. 2017) to amplify and sequence a portion of three nuclear genes: translation elongation factor ( EF-1α ), phosphate: H * symporter ( PHO ), and β-tubulin ( BT ). BLASTn analysis of CK13B sequences showed 93 to 98% identity with F . oxysporum f. sp. vasinfectum isolates from Australia, with 93.43% (1,706/1,826 bp) identity of EF-1α to isolate AuSeed14 (KT323873), 95.11% (1,849/1,944 bp) of PHO to isolate AuK24232 (KT323909), and 97.56% (1,840/1,886 bp) identity of BT to isolate AusSeed14 (KT323833). Sequences of CK13B for EF1-α , PHO , and BT genes were submitted to GenBank (accession no. ON754247, ON754248, and ON754249, respectively). The CK18A isolate showed high identity with F . oxysporum f. sp. vasinfectum races 1, 2, and 6 lineage isolates (Ortiz et al. 2017), with 99.89% (1,822/1,824 bp) identity of the EF1-α gene to isolate CDR238 (KT323838) from Arkansas, 98.36% (1,916/1,948 bp) identity of the PHO gene to isolate CDR1131 (KT323887) from Louisiana and 98.78% (1,864/1,887 bp) identity of the BT gene to isolate ATCC36198 (KT323799) from Brazil. Sequences of the CK18A isolate were submitted to GenBank (accession no. ON651444, ON725043, and ON725044, respectively). Pathogenicity of both isolates was tested on G . hirsutum as previously described (Kim et al. 2005), with three replicates per isolate and a control treatment in a growth chamber. Briefly, cotton seedlings were maintained with a 12 h light/dark cycle at 25 to 20°C. At the first true leaf stage, seedlings were uprooted and roots were cleaned by rinsing with sterile water and placed in a conidial suspension (1 × 10 6 conidia/ml) for 2 h while healthy control seedlings were dipped in water only. Plants were put into sterile soil, and the results were observed and recorded every 7 days for 30 days. The inoculated plants showed the same symptoms as those observed in the fields: wilting, leaf chlorosis, and necrosis. Both isolates were reisolated from the roots of the inoculated plants. DNA was extracted and used for PCR with the specific gene primers listed above. The sequences matched those of the original isolates, completing Koch's pustulates. To our knowledge, this is the first report of F. oxysporum f. sp . vasinfectum causing Fusarium wilt disease of cotton in Kansas. These results will help growers select cultivars and design disease control strategies (Davis et al. 2006). Further work is needed to determine the specific races in the area.

Abstract:Artemisia argyi is a perennial herb native to East Asia. It is an important traditional Chinese medicinal plant known for its strong flavor and medicinal effects. It is rich in active ingredients and has a wide range of biological activities, including anti-inflammatory, antioxidant, and immune regulation properties. From May to July in 2023, a serious leaf rot outbreak occurred on A. argyi in several farms (approximately 200 acres) in Tanghe county (32°46'44" N, 112°43'13" E), Henan Province, China. The incidence rate reached 65% (n=200). Pale yellow spots (1-2 cm in diameter) first appeared on the leaves, then expanded to form irregular yellowish-brown lesions, eventually causing the entire leaves to wither. Diseased leaves (30) were collected and cut into 5 x 5 mm2 pieces in the areas between infected and healthy tissues. The excised plant tissues were sterilized in 75% ethanol and 1% sodium hypochlorite solution for 30 seconds and one minute, respectively. The tissues were then rinsed with sterile water and placed on potato dextrose agar (PDA) followed by incubating at 25 °C for 3 days. The isolated strains belonged to the genera Fusarium and Alternaria. After pathogenicity verification, 25 purified Fusarium strains were obtained. Three representative strains (AC-Q, AC-X, AC-Y) from different regions were used for further studies. Each strain formed abundant aerial mycelium that was initially white and later developed into purple pigments. Aerial conidiophores were sparsely branched, terminating with verticillate phialides. Macroconidia were slender, straight, and measured 21.8 to 47.5 × 3.1 to 4.4 μm, with two to four septa. Microconidia were clavate and measured 8.31 to 11.6 × 2.1 to 3.5 μm. Morphological characteristics were consistent with the species description of Fusarium verticillioides (Sacc.) Nirenberg 1976 (Leslie and Summerell, 2006). The rDNA internal transcribed spacer (ITS), β-tubulin gene (tub2), translation elongation factor 1-alpha gene (tef1), calmodulin (cmdA), RNA polymerase II largest subunit (rpb1) and RNA polymerase II second largest subunit (rpb2) were amplified for molecular identification (O'Donnell et al., 2022). The sequences were deposited in GenBank with accession Nos. OR960548, OR960552, OR960555 (ITS), OR972413, OR972414, OR972415 (tub2), OR797685, OR797686, OR797687 (tef1), OR972410, OR972411, OR972412 (cmdA), PP035106, PP035107, PP035108 (rpb1), and PP035109, PP035110, PP035111 (rpb2). BLASTn analysis of AC-Q sequences exhibited 99 to 100% similarity with F. verticillioides sequences (strains CBS 576.78) MT010888 of cmdA, MT0109566 of rpb1, and MT010972 of rpb2. A phylogenetic tree was constructed with concatenated sequences (tub2, tef1, cmdA, rpb1, rpb2), alongside the sequences of the type strains using the neighbor-joining method. The three strains formed a clade with the type strain CBS 576.78 of F. verticillioides, and were separated from other Fusarium spp. These morphological and molecular identifications indicated that the pathogen was F. verticillioides. Pathogenicity was tested on 10 healthy 2-month-old potted seedlings by spraying them with a conidial suspension (106 conidia ml-1), and 5 seedlings were sprayed with sterilized water as a control. The plants were placed in a climate incubator at 28°C and a relative humidity of approximately 90%. Ten days after seedling inoculation, typical lesions were observed on the treated plants, except in the control group. The reisolated strains were identified as F. verticillioides by morphological and molecular characterization, fulfilling Koch's postulates. F. verticillioides is known to cause Fusarium ear rot on maize, as well as diseases on other plants in China such as Brassica rapa (Akram et al., 2020) and Schizonepeta tenuifolia (Li et al., 2024). This is the first report of F. verticillioides causing leaf rot on A. argyi worldwide. Identification of the pathogen is crucial for implementing management approaches to reduce yield losses.

Abstract:Root rot is a major disease that causes decline of alfalfa production, and Fusarium is a major pathogen associated with root rot. In this study, 13 Fusarium isolates were obtained from alfalfa with root rot in Gansu Province, the major alfalfa production region in China. The isolates were characterized by molecular genotyping ( ITS , TEF 1-α and RPB2 fragments) and identified as six species, which included the F. acuminatum , F. incarnatum , F. oxysporum , F. proliferatum , F. redolens , and F . solani . We found that their morphology varied significantly at both the macro- and micro-levels, even for those from the same species. We developed a low cost and fast pathogenicity test and revealed that all isolates were pathogenic to alfalfa with typical root rot symptoms such as leaf yellowing and brown lesions on the root and stem. However, the virulence of the isolates differed. We also found that the conidia of all isolates germinated as early as 24 hours post inoculation (hpi), while hyphae colonized the root extensively and invaded the xylem vessel by 48 hpi. Together our results reveal that different virulent Fusarium isolates use a similar invasion strategy in alfalfa. This natural plant-fungus pathosystem is intriguing and warrants further examination, particularly with regard to efforts aimed at mitigating the impact of multiple similar vascular pathogens on infected alfalfa plants.
