Abstract:Armillaria root disease is among the largest causes of mortality and lost productivity of widely ranging horticultural, urban, and forest trees/shrubs in diverse boreal, temperate, and tropical regions around the world (Kim et al. 2022). Damage from Armillaria root disease will likely increase in response to changing climate and extreme weather because environmental stress can predispose host trees to Armillaria root disease (Murray and Leslie 2021; Kim et al. 2021). On August 14, 2021, a mature black oak (Quercus velutina) ca. 29 m in height and 64 cm DBH experienced a complete structural failure originating at the root plate, falling onto a major highway within the Pisgah National Forest near Brevard, North Carolina (35°16'50.876"N, 82°42'51.785"W, elevation 680 m) during sunny, calm conditions. No above-ground indications of predisposing disturbance, injury, or insect/disease activity were observed. Advanced wood decay, however, was present in many structural roots of the exposed root plate; abundant rhizomorphs attached to the surfaces of most roots were noted (Supplemental Figure 1). One large primary root (ca. 23 cm in diameter) exhibited clear signs of Armillaria root disease at the point of failure as evidenced by an extensive decay column, abundant black rhizomorphs on the root surface, and white mycelial fans (a sign of pathogenicity) within the root itself (Supplemental Figure 1). Rhizomorph samples were established in culture, two Armillaria isolates (PNF#001R-1 and PNF#001R-2) were examined, and pairing tests showed that both isolates belonged to the same genet (PNF#001R-1 = PNF#001R-2). Based on translation elongation factor 1α (tef1) sequences, both isolates (one genet) were identified as A. solidipes (GenBank accession no. OP823701), showing 98% similarity with A. solidipes tef1 sequences (e.g., MH879015) in GenBank. Also, nine replications of somatic incompatibility tests of the isolate with five North American Armillaria spp. [A. solidipes, A. mellea, A. gallica, A. mexicana, and Desarmillaria caespitosa (= North American A. tabescens)] showed 67% compatibility with A. solidipes, compared with 0-22% for A. mellea, D. caespitosa, A. gallica, and A. mexicana. All test species but A. mexicana are reported to occur in the eastern and/or southern USA, while A. mexicana occurs in a similar environment in Mexico (Elías-Román et al. 2018). To our knowledge, this is the first confirmed report of A. solidipes in North Carolina, USA, where it was causing disease on black oak, and this is the most southerly confirmed case of A. solidipes in eastern North America. Although Armillaria inoculation tests are impractical, black oak was previously reported as a host of A. solidipes in Massachusetts, USA (Brazee and Wick 2009). Armillaria solidipes is considered as the most important Armillaria root disease pathogen of conifer forests in western USA (Lockman and Kearns 2016), but it has been suggested that A. solidipes can thrive in northern hardwood forests that reside near conifer forests (Brazee and Wick 2009). Climate change is predicted to increase damage caused by A. solidipes on conifers in the western USA (Kim et al. 2021); however, it is undetermined if the occurrence of A. solidipes-caused disease in North Carolina is related to climate change or how climate change could influence this disease across the region. More surveys are needed to assess the impact of Armillaria root disease on health of mixed forests in the eastern USA.

First report of Cryptostroma corticale causing sooty bark disease in California and first worldwide report of silver maple as a host

Abstract:In November 2022 and February 2023, CAL FIRE tree health experts examined four maples (Acer spp.) planted decades earlier in a residential setting in Elk Grove, Sacramento Co., California (USA). Three of the trees were silver maples (Acer saccharinum ) and one was a Norway maple (A. platanoides); both species are exotic to California. The trees were in an irreversible state of decline, with the canopy substantially thinned and browning. Extensive bark cankers elongating longitudinally along the stem were visible on all trees (Fig. S1). Cankers were filled by fungal stromata protruding through the bark and producing masses of elliptical dark brown conidia (Fig. S2), approximately 5.5 x 3.7 um in size, giving the cankers a sooty appearance. The cankered bark could be peeled off easily, revealing dark and discrete lesions in the phloem and xylem. Samples from the three trees were shipped to the U.C. Berkeley Forest Pathology and Mycology Laboratory and to the CDFA PPDC in Sacramento, CA. In the laboratories, small wood chips were taken from the margins of the lesions, surface sterilized by dipping them for 30 seconds in 70% Ethanol, rinsed for 30 seconds in sterile water and plated onto 2.5% Malt Extract Agar amended with 0.3g/L Streptomycin or onto one-half strength acidified potato dextrose agar (APDA). Two morphologically identical cultures were obtained, one (T2) from a silver maple and one (T6) from the Norway maple. Cultures were then grown in liquid 2.5% malt extract broth and, after one week, DNA was extracted using the Qiagen Plant DNeasy DNA extraction kit. The ITS sequences are diagnostic for this fungus (Li et al. 2021) and those of the two cultures (GB OR064033 and OR933565) were 100% homologous to GenBank sequences of Cryptostroma corticale ( e.g. GB OP474010-11). The RPB2 sequence of T2 ( GB OR992132) was 100% homologous to that of C. corticale (GB HG934116.1). The isolate obtained from silver maple was inoculated in four potted silver maples by removing a bark disk 50 mm in diameter with a cork borer in three spots staggered at different heights and sides on the stem, placing a colonized agar plug of C. corticale in contact with the phloem, replacing the bark disk and wrapping with parafilm. Two control trees were mock inoculated using sterile agar plugs. Trees were in 57 L pots, had an average stem caliper of 2.7 cm, an average height of 3.5 m and were kept in a lath house at average high temperatures of 18-24 degrees C. After ten weeks, average lesion length was 15.4 cm (SE= 4.6) and 4.3 cm (SE=2.3) in the fungus-inoculated and control trees, respectively. An ANOVA test, nesting lesions sizes within tree, determined lesions lengths were different between inoculated and mock trees (P= 0.04). The fungus was reisolated from all points in all inoculated trees but never from control trees. C. corticale was first described in the UK from sycamore maple (Acer pseudoplatanus) (Gregory et al. 1949) and is an emerging problem in Europe (Muller et al. 2023). In North America, it has been reported from A. negundo, A. campestre, A. macrophyllum and Cornus nuttallii (Worral 2020), and it appears to be present in the Pacific Northwest (Brooks et al. 2023, Goree 1969). Norway maple is included in the European Plant Protection Organization list of hosts for C. corticale (EPPO 2023), however our finding of C. corticale on silver maple is a first report of this host worldwide and of this pathogen in California. This report is noteworthy, given that C. corticale is also a human pathogen infecting the respiratory system (Braun et al. 2021).
First report of Bretziella fagacearum infecting chestnut in Michigan

Abstract:In the summer of 2021, a 20-year-old 'Colossal' (Castanea sativa × C. crenata hybrid) tree in a commercial chestnut orchard in northwest Michigan suddenly declined. Until 2023, an additional 26 adjacent trees declined, suggesting the occurrence of root-graft transmission of the pathogen. The initial wilting of leaves progressed to complete tree death in about 10 days. Symptoms included wilting, and bronzing, followed by tanning starting at leaf apex and margins, with significant defoliation. Sometimes black-to-brown streaks of discoloration appear in the sapwood, with no signs of mycelial mat production on dead trees. Branches from symptomatic trees in two different areas of the orchard were submitted to Plant and Pest Diagnostics at Michigan State University. Bretziella fagacearum (Bretz) Z.W. de Beer, Marinc., T.A. Duong & M.J. Wingf. was detected in both samples using nested PCR (Wu et al. 2011) and qPCR (Bourgault et al. 2022). The products of the nested PCR were sequenced (GenBank accession nos. OR522695-OR522696) and BLASTn search results showed 100% identity to an ex-type strain of B. fagacearum (MH865866). Surface-sterilized discolored sapwood chips were plated on acidified potato dextrose agar (aPDA). Bretziella fagacearum was consistently recovered; colony and endoconidia morphology aligned with the description of the pathogen (De Beer et al. 2017). A pure culture (BF277) was obtained for inoculation experiments. To confirm pathogenicity, 10 'Colossal' chestnut seedlings (average stem diameter of 9 mm) were inoculated in the greenhouse with a 14-day old culture of BF277. Using a conical drill bit, two 0.4 mm diameter holes were drilled, one was 5 cm above the soil line at a 45° angle and the other was on the opposite side of the stem at least 10 cm above the soil line. A 50-µl conidial suspension (1 × 107 conidia per ml) was applied and the holes were sealed with Parafilm. Five 'Colossal' seedlings were inoculated with sterile water. Leaf epinasty with bent petioles was observed 14 days later. Leaf wilting and necrosis similar to natural infection in the orchard were observed at 24 and 34 days after inoculation, respectively. Water-inoculated control plants showed no symptoms. Bretziella fagacearum was reisolated from symptomatic plants by surface sterilizing leaf petioles with 75% ethanol (30 s), followed by 10% (v/v) bleach (1 min), and two rinses with sterile deionized water (>1 min). Petiole pieces (~1 cm) were plated on aPDA. The pathogen was reisolated from six symptomatic plants and detected using qPCR in the remaining four seedlings. Bretziella fagacearum was not detected in control plants. The identity of the recovered fungus was confirmed following the amplification of the internal transcribed spacer (ITS) from extracted genomic DNA, as described in Chahal et al. 2022. The resulting PCR product was sequenced and assembled into a consensus sequence using Geneious Prime. The consensus sequence (accession no. OR515809) revealed 100% identity to the ex-type of B. fagacearum (KU042044). This is the first record of B. fagacearum infecting chestnut trees in Michigan. Previously, B. fagacearum has been reported infecting Chinese chestnut (C. mollissima) in Missouri (Bretz and Long, 1950). Oak wilt is widely distributed in Michigan and is the predominant disease afflicting red oaks in the Midwestern U.S. Consequently, constant vigilance and monitoring are essential in chestnut orchards to promptly detect and effectively manage potential infections.
The Effect of Multipath on the Detection of Symbol-Rate Spectral Lines by Delay and Multiply Receivers

Abstract:The effect of a secondary path signal on the detectability of the symbol rate spectral line of a binary phase-shift keyed (BPSK) signal by a delay and multiply receiver is investigated. The measure of detectability used is the rate line signal-to-noise ratio. The parameters that define the secondary path signal are initially modeled as deterministic quantities; the phase of the secondary path signal is then taken to be a random variable independent and uniform on (0, 2 pi ). It is found that, with deterministic path parameters, the presence of a secondary path can seriously affect the detectability of BPSK rate lines. When the phase is taken to be uniform on (0, 2 pi ) over the receiver observation time, then the delay and multiply receiver performs as if no multipath is present. In this case, a noncoherent power addition of the primary and secondary path signals occurs. >
First report of Apple bitter rot caused by Colletotrichum godetiae in Ontario

Abstract:Bitter rot is an emerging disease of apple (Malus domestica) fruit in Ontario in part due to changing weather conditions. The disease was mostly documented in warm and humid regions such as the southern USA, and Central and South America. Thirteen Ontario orchards in the fall of 2019 and 15 in 2020 were scouted for bitter rot based on their previous history of the disease. 100 fruit were collected from ten asymptomatic trees per cultivar and two susceptible cultivars, 'Empire' and 'Ambrosia' were scouted per orchard. If an orchard did not have either one of these cultivars, 'Honeycrisp' or 'Gala' were used. The fruit was stored at 4-5 oC for five months and then left at 22 oC for two weeks and assessed thereafter for bitter rot symptoms. Monoconidial cultures of Colletrotrichum spp. were established from the symptomatic fruit using potato dextrose agar media with antibiotics at 22 oC 14-hour light cycles. The fungal isolates were divided into two groups based on colony morphology observations seven days after culturing. The first group showed light grey, cottony, aerial mycelium on top and a pink to dark red color on the reverse of the plate. The second group showed light to dark grey, cottony mycelium on top and dark green colonies on the reverse of the plate. It is difficult to identify Colletrotricuhum species solely based on morphology, therefore the representative isolates from each group were used for multilocus gene sequencing and species identification. Genomic DNA was extracted using the Qiagen DNeasy Plant Mini Kit according to the manufacturer's protocols. The ITS region was amplified and sequenced using the primers ITS-1F (Grades & Bruns 1993) and ITS-4 (White et al. 1990). Primers T1 and T2 (O'Donnell & Cigelnik 1997) were used to amplify and sequence the 750 bp region of the TUB gene. Primers GDF1 and GDR1 (Guerber et. al. 2003) were used to amplify a 280 bp region of the GADPH gene. Following an NCBI nucleotide blast search, the isolates from group 1 were identified as C. fioriniae.The ITS sequences of group 2 isolates were matched 100% to the Colletotrichum godetiae type strain CBS133.44 and they were 99% matched to the closest species C. Johnstonii CBS128532. The TUB sequences matched 100% identity to 20 sequences belonging to C. godetiae, 99.59 % identity to C. godetiae type strain CBS133.44, and 97.75% to C. Johnstonii CBS128532. The GADPH sequences matched with 100% identity to C. godetiae ON241087.1 or MT816329.1 and 99-99.5% identity to the type strain CBS133.44 and 98.61-99% identity to CBS128532. Based on the blast results the group 2 isolates were identified as C. godetiae and their sequences were submitted to GenBank with ID OP702962 for ITS and OP972240 and OP972241 for GADPH and OP972242 for the TUB gene. Out of 50 isolates collected in this work, 94% belonged to group 1 and 6% belonged to group 2. Koch's postulates were performed on selected isolates by artificial inoculation of 5 healthy detached fruits of the cultivar, 'Empire.' Fruit surfaces were wiped with 70% ethanol, dried, wounded with a sterile needle, and then inoculated with a 10 µl of spore suspension containing 1x10^6 spores /ml. Inoculated fruits were incubated in a humid chamber at 22°C in dark. Symptoms started to appear at 5 days post-inoculation and looked like small brown circular lesions which developed orange spore masses as they grew into larger lesions. The non-inoculated control fruit did not develop lesions. Fungal colonies were established from the spores collected from the inoculated fruit and found to have identical morphological characteristics to the original isolates. C. godetiae has previously been reported to cause bitter rot in apples from various countries in Europe (Baroncelli et al. 2014; Munda, 2014; Wenneker et al. 2016). C. fioriniae has previously been reported as the dominant species causing bitter rot in apples although other species including C. chrysophilum and C. noveboracense have also been reported as causal agents from the Eastern USA (Khodadadi et al. 2020). To the best of our knowledge, this is the first report of Colletotrichum godetiae causing the bitter rot of apples in Ontario, Canada.
First Report of Colletotrichum Plurivorum Causing Anthracnose on Pecan (carya Illinoinensis) in China

Abstract:Pecan (Carya illinoinensis) is an economically important nut crop worldwide (Xiao et al 2021). Anthracnose symptoms were found on pecan fruits and leaves in plantations in Anhui and Jiangsu provinces, China in August 2019. Irregular, dark brown or black spotted lesions firstly appeared on the surface and inside of fruits, and spread to all leaves. The symptoms resulted in 30% to 50% leaf drop and nearly a half of fruit decay in almost all trees of the susceptible cv. Wichita. The causal agent were isolated from fruits with target symptoms following the steps: surface disinfected with 75% ethanol (2×, 30 s), rinsed with sterile deionized water (3×), ~ 0.5 cm small fragments of the fruits excised and plated on potato dextrose agar (PDA) medium and incubated at 28 °C in dark for 3-d. Mycelium of each colony was picked and incubated on fresh PDA at 25 °C with a 12-hour light/dark cycle for 6-d to induce conidia formation. One 5-mm hyphal plug produced from each single spore isolate was transferred onto fresh PDA to obtain the pure cultures. Koch's postulates was employed for pathogenicity determination of the isolates. Non-wounded healthy leaves from seedlings of the disease susceptible cv. Pawnee were disinfected with 1% NaClO and inoculated with 5-mm 5-d hyphal of each isolate at 25 ℃. Tiny lesion spots were visible on the leaves after 2 days post inoculation (DPI) with isolate W-6 (the only pathogenic one among all isolates), and expanded over time until to the leaves withered, while the control leaves and leaves inoculated with other isolates remained asymptomatic. The pathogenicity of W-6 were confirmed using leaves and fruits of living Pawnee trees growing in Linglong Mountain Plantation, Lin'an, Hangzhou, China (119⁰38'51″E, 30⁰12'39″N, elevation: 119m). Three experimental replicates were conducted separately with three bio-replicates for all pathogenetic testing. The same symptoms were observed on both detached leaves and leaves and fruits of living trees.. The colony of W-6 have round cottony mycelium with complete edges and showed the fastest growth rate 3 - 4 DPI. After 7 DPI, white aerial mycelium turned yellowish brown and formed Acervulus in the mycelium. Conidia (n=50) one-celled, 12.0 - 20.0 μm × 3.5 to 6.0 μm width. Hyaline cylindrical with slightly rounded ends and two or three large guttulate at the centre. Most Acervulus dark brown and slightly irregular in shape, 12.70 × 18.79 μm (n=10). Setae were dark brown in color with average length around 34.10 μm (n=10). These characteristics matched previous descriptions of Colletotrichum orchidearum species complex, including C. plurivorum (Damm et al 2019). The identity of W-6 was confirmed by multi-locus phylogenetic analysis using the internal transcribed spacer (ITS) rDNA region and partial sequences of the conserved genes glyceraldehyde-3-phosphate dehydrogenase (GAPDH), actin (ACT), beta-tubulin 2 (TUB2), and chitin synthase (CHS). The sequences of W-6 were used for Basic Local Alignment Search Tool (BLAST) against NCBI GenBank and the sequences with 100% identity to that of W-6 were achieved, respectively. The concatenated sequences of the ACT-CHS-GAPDH-ITS-TUB2 was used for building a phylogenetic tree. The molecular analyses allowed the identification of the pathogen as C. plurivorum. It was known that 9 of the 11 Colletotrichum species causing pecan anthrax worldwide were reported in southern China (Brenneman 1989; Oh et al 2021). This is the first report of C. plurivorum as causal agent of pecan in China.
Automatic detection of cardiac cycle and measurement of the mitral annulus diameter in 4D TEE images

Abstract:Mitral regurgitation is a wide spread problem. For successful surgical treatment quantification of the mitral annulus, especially its diameter, is essential. Time resolved 3D transesophageal echocardiography (TEE) is suitable for this task. Yet, manual measurement in four dimensions is extremely time consuming, which confirms the need for automatic quantification methods. The method we propose is capable of automatically detecting the cardiac cycle (systole or diastole) for each time step and measuring the mitral annulus diameter. This is done using total variation noise filtering, the graph cut segmentation algorithm and morphological operators. An evaluation took place using expert measurements on 4D TEE data of 13 patients. The cardiac cycle was detected correctly on 78% of all images and the mitral annulus diameter was measured with an average error of 3.08 mm. Its full automatic processing makes the method easy to use in the clinical workflow and it provides the surgeon with helpful information.
First Report of Diplodia quercivora and Neofusicoccum vitifusiforme Associated with Cankers and Necrosis of Holm Oak (Quercus ilex) in Declining Stands in Southern Italy

Abstract:The emergence of new plant diseases is an increasingly important concern. Climate change is likely to be among the factors causing most of the emerging diseases endangering forest and tree heritage around the world. Such diseases may be caused by latent pathogens or microorganisms cryptically associated with plants. The shift from a non-pathogenic to a pathogenic stage may depend on physiological alterations of the host, environmental changes, and/or stress factors. In some woods of the Salento Peninsula (Apulia Region, Italy), sudden declines of holm oak plants (Quercus ilex L.) have been observed since 2016. The morphological and molecular characterization of representative fungal isolates associated with cankers and necrosis in declining plants indicated that these isolates belong to the Botryosphaeriaceae family, and the most frequent species were Diplodia corticola and Diplodia quercivora, followed by Neofusicoccum vitifusiforme. In artificially inoculated young holm oak plants, both D. corticola and D. quercivora species produced intense and severe subcortical and leaf margin necrosis. N. vitifusiforme, although less aggressive, induced the same symptoms. Our research, in addition to confirming the involvement of D. corticola in olm oak decline, represents the first report of D. quercivora as a new pathogen of Q. ilex in Italy. Furthermore, to the best of our knowledge, we also found N. vitifusiforme as a new pathogen of Q. ilex.

First Report of Fusarium Concentricum As a Causal Agent of Fusarium Leaf Blotch on Pecan (carya Illinoinensis) in Southeast China.

Abstract:The economically important nut crop pecan (Carya illinoinensis (Wangenh.) K. Koch) is seriously affected by increasing incidence of fungal disease worldwide (Xiao et al 2021). The top leaves of the pecan variety 'Pawnee' in the orchard of Zhejiang A&F University, Zhejiang, China were damaged by massive dark brown plaques in summer to autumn 2021. The causal agent was isolated from leaves with target plaques following the steps: sterilized with 70% alcohol (30 s × 2), rinsed with sterilized water (3 ×) before and after 5% sodium hypochlorite (30 s), excised the plaques, and placed on PDA medium at 28℃ in a dark incubator for 3-d. The mycelium on the edge of each colony was transferred to fresh SNA medium in dark for 2 weeks to induce conidia formation. A few conidia-germinated mycelia were transferredand inoculated on new plates containing fresh PDA medium to obtain the purified cultures. Koch's postulates were applied to validate the pathogenicity of the purified isolates. Non-woundedly healthy leaves (disinfected with 5% sodium hypochlorite) of 'Pawnee' seedlings were inoculated with 5 mm 7-d old purified cultures. Dark-brown spots appeared on the leaves 2 days post inoculation at 25℃. The spots became larger accompanied by partially cracking and slight deformation of inoculated leaves from day 2 to day 4, while the control leaves remained asymptomatic. A re-isolated strain ZJ-6 from these infected leaves was identified as the pathogenic isolate with the same symptom as the previous one. Morphologically, aerial mycelia of the pathogenic isolate ZJ-6 cashmere and white. The reverse of colony orange. The edge of the colony appeared gradually thinner, the aerial mycelia loose and flocky, and the matrix mycelium whitened. Hyphae were septate, translucent with smooth wall and 1.47-7.14 µm in width. Microconidia (n = 20) obovoid to fusoid, mainly with 0-septate, 4.45-7.78×4.79-16.25 µm. Macroconidia (n = 20) sickle, mainly with 3-5 septa, 5.56-10.28×56.67-114.54 µm. Simultaneous of monophialidic and polyphialidic conidiophores. Conidiophore width 1.47-3.68 µm, slightly smaller than vegetative hyphae. The morphological characteristics matched with previous descriptions of Fusarium species (Nirenberg and O'Donnell 1998; Wang et al 2013). The identity of ZJ-6 was confirmed by phylogenetic reconstruction using the concatenated sequences of the ATP citrate lyase (ACL1), Calmodulin (CaM), the internal transcribed spacer (ITS) rDNA region, ribosomal RNA gene (LSU), the largest subunit of DNA-dependent RNA polymerase II (RPB1), partial translation elongation factor-1 alpha (TEF) and β-Tubulin (TUB). To this end, the genomic DNA of ZJ-6 was extracted by the M5 hipermix-MF859 (Mei5 Biotechnology) and submitted to GenBank under the accession numbers OP933646, OP933647, OP925890, OP925889, OP933396, OP933648, and OP933397, respectively. The obtained sequences of ZJ-6 were used for nucleotide BLAST against thetandard databases, respectively, and the strains with sequence identification values above 98% were selected to construct multiple alignment for building a phylogenetic tree. This analyses allowed the identification of ZJ-6 as Fusarium concentricum Nirenberg & O'Donnell, a species with few reports that can cause serious damage to the fruits and branches of other hosts (Hasan et al 2020; Huda-Shakirah et al 2020; Wang et al 2013). This is the first report of pathogenic F. concentricum on pecan in Southeast China that caused no harvest of infected plants.
First Report of Botryosphaeria dothidea Causing Stem Canker and Dieback of Apple Trees in Ontario, Canada

Abstract:Apple trees (Malus domestica L. Borkh.) exhibiting extensive and unknown tree fruit decline symptoms were observed in commercial orchards over the past 5 years in Ontario, Canada. The trees exhibited shoot dieback, attached wilted leaves and cankers on the main trunk. Trees with rapid development of cankers upward from the graft union developed extensive vascular discoloration that resulted in sudden collapse of the entire tree. In 2018-19, up to 42% mortality was observed on 2- to 8-year old apple trees. Nine symptomatic trees were collected from two orchards located in southwestern and one in southcentral Ontario. Samples (1 cm length) were collected from symptomatic trunk and shoot tissue, surface sterilized with 70% ethanol for 30 sec, followed by 1% NaClO for 20 min and three rinses in sterile water. The samples were air-dried and placed on a 2% potato dextrose agar (PDA, Difco) culture media supplemented with kanamycin (50 mg L-1). The PDA plates were incubated at 22°C for 5 days in the dark. All fungal colony-forming units that developed were hyphal-tip transferred to individual PDA plates and incubated at 22°C for 7 days in the dark. Purified mycelial isolates were classified into morphotypes prior to molecular identification. One morphotype showed white to olive-green colonies that developed on a moderately dense mycelial mat with aerial hyphae. Several solitary and globose black pycnidia that contained a single ostiole were produced on pine needles on PDA after incubation at 22°C for 14 days in the dark. Conidia were hyaline, fusiform, aseptate with an average size of 3.9 - 5.2 x 21.4 - 26.6 μm (N=50). Genomic DNA was extracted from 5-day old culture of a representative isolate, #M68-17, grown on PDA using the Plant/Fungi DNA Isolation Kit (Norgen Biotech Corp., Thorold, ON, Canada). The rDNA internal transcribed spacer (ITS), translation elongation factor 1- alpha gene (TEF-1α), and β-tubulin gene (TUB2) were amplified using the primer pairs ITS1/ITS4 (White et al. 1990), EF1-728F/EF1-986R (Carbone and Kohn 1999), and Bt2a/Bt2b (O'Donnell et al. 1998), respectively, and sequenced. The nucleotide sequences obtained (GenBank # MZ926850, MZ934654, MZ934655) were 100.00% similar to those of Botyrosphaeria dothidea (Moug. ex Fr.) Ces. & De Not. isolates from other hosts in several countries in the NCBI database (MT111097, MT309401, MN515421, respectively). Randomized Accelerated Maximum Likelihood (RAxML) analysis using the three gene sequence data was completed (Stamatakis et al. 2008). The isolate #M68-17 was clustered with high bootstrap support values with the B. dothidea isolates from the fungal biodiversity centre (CBS) collection, including the ex-epitype (CBS 115476, CBS 110302) (Fig. 1). A living culture of the representative isolate, #M68-17, was deposited in the Canadian Collection of Fungal Cultures (DAOMC 252246). Pathogenicity tests were conducted in the lab on wood cuttings and in planta. Ten 20 cm apple cuttings and five potted one year old apple seedlings were surface sterilized, wounded and inoculated with 4 mm mycelium agar plug from a 5-day old culture of isolate #M68-17 and wrapped with Parafilm. Three control apple cuttings and seedlings were inoculated with PDA plugs and incubated in the same environment. Cuttings were placed inside a clear plastic container with moist paper towels and incubated at room temperature in the dark. Seedlings were placed in an open-air area between two greenhouses and watered as needed. Twelve days post-inoculation, the average length of the developed necrotic lesions on cuttings was 8.8 ± 0.4 cm. Necrotic and sunken canker symptoms appeared at 10 days, spread upward from the inoculation point and by 6 weeks the upper portion of the seedling was dead (Fig. 2). B. dothidea was isolated from all the inoculated cuttings and seedlings, thus fulfilling Koch's postulates. Control cuttings and seedlings showed no symptoms, and the fungus was not isolated from the wood. B. dothidea was reported as a pathogen causing cankers on a wide range of woody crop plants including apples, almonds, pistachios, hazelnut, walnut, olive and grapes in the United States, China, Uruguay, Spain, Tunisia and Turkey (Chebil et al. 2014; Delgado-Cerrone et al. 2016; Moral et al. 2019; Tang et al. 2012; Türkölmez et al. 2016). However, this is the first report of B. dothidea causing stem canker and death of young apple seedlings in Ontario, Canada. The findings suggest that B. dothidea has the potential to severely affect apple production in Ontario. Accurate identification of pathogen(s) associated with apple decline will support management of the disease.

Abstract:Nitrogen-13 is the longest lived radioisotope of nitrogen, and compounds labeled with this isotope can be detected in vivo. In this study nitrogen-13 labeled amino acids are prepared by enzymatic reactions. The acids prepared are glutamine, glutamic acid, valine, alanine and leucine. In all cases the nitrogen-13 labeled substrate is ammonia, which must be prepared with a high specific activity for the enzyme reactions to proceed with high yield. The methods available for preparing nitrogen-13 ammonia have been adapted to prepare a high specific activity product.
[Blood pressure change and syncope during leg phlebography].

Abstract:Although syncope attacks such as black-out, faint consciousness, and cold sweat are sometimes experienced during leg phlebography, no study of their incidence and mechanism has been reported. We measured blood pressure noninvasively by using a Finapress with ECG monitor during overall examinations (21 cases, 33 limbs; male 8, female 13) following anamnesis. Age, sex, and past history of drug, syncope, leg phlebography, and other diseases were determined. All examinations were done in the upright position. Three cases (14.3%) and four limbs (12.1%) showed syncope attacks during leg phlebography. Syncope occurred after steps taken for the evaluation of venous return in two limbs, during infusion of contrast medium in one, and after infusion in the other. In all cases, the systolic blood pressure measurement during syncope was below 80 mmHg, and the sudden decrease of both systolic blood pressure (-83.0 +/- 22.0 mmHg) and heart rate (-29.5 +/- 5.0/min) suggested vasovagal reaction as a mechanism of syncope. Other causes of syncope including anaphylaxy, hyperventilation syndrome, seizure, and arrhythmia (except for bradycardia) were not found. There were also significant changes in blood pressure and heart rate in the nonsyncope group during leg phlebography that seemed to trigger vasovagal excitation. Premedication, contrast media, and position might be important factors and should be discussed further.

In July 2018, an unknown disease occurred on 5 to 10% of 5,000 postharvest Cuiguan pear fruit (P. pyrifolia) after 15 days in our laboratory storage room (20°C, 85 to 90% relative humidity) in NanChang City (28.68° N, 115.85° E), Jiangxi Province. Initial symptoms were shallow black spots on the fruit surface that later enlarged into circular brown lesions with water-soaked margins and a lesion diameter of 20 to 40 mm. White hyphae grew on the pear lesions and, slowly, infected fruit became soft and necrotic. To isolate the suspected pathogen, small sections (5 × 3 mm) of diseased fruit were sterilized with 75% ethanol for 30 s followed by 0.1% mercuric chloride solution for 5 min, rinsed three times with sterile water, and then transferred onto potato dextrose agar (PDA) and incubated at 28°C for 5 days. Finally, five isolates were obtained and purified on PDA. The isolates produced abundant aerial mycelium, which was initially white and then became purple over time at 28°C. The average growth of mycelium was 5.4 mm per day (n = 5). On carnation leaf agar (Fisher et al. 1982), macroconidia were three to five septa, moderately curved to straight, and 28.7 to 53.2 × 3.2 to 5.3 µm in size (38.4 × 4.2 µm on average, n = 25). On PDA, the microconidia were almost straight, aseptate, and 6.3 to 11.4 × 2.1 to 4.0 µm (8.3 × 2.9 µm on average, n = 25), and no chlamydospores were observed. The morphological features were consistent with Fusarium proliferatum (Leslie and Summerell 2006). In addition, molecular phylogenetic analyses were used to define the isolate identity. The universal primers ITS1/ITS4 (White et al. 1990) and F. proliferatum-specific primers PRO1/PRO2 (Mulè et al. 2004) were used to amplify the internal transcribed spacer (ITS)-5.8S ribosomal DNA region and calmodulin gene. The PCR amplicons (accession nos. MK158221 and MK144327) aligned with GenBank showed 100% similarity with F. proliferatum (accession nos. KU891521.1 and LT841258.1). Thus, the pathogen was identified as F. proliferatum based on morphological and molecular characteristics. Pathogenicity tests were conducted by inoculating nine disease-free pear fruit after disinfesting in 75% ethanol. A prepared spore suspension (10 μl, 1.0 × 106 spores/ml) was dropped on the wounds (a depth of 1 mm with a 1-mm diameter) on the pear surface, which were made by a sterilized needle. Sterile water (10 μl) was dropped on the wounded sites of nine pear fruits as negative controls, and 10 μl of a prepared spore suspension was dropped onto nine nonwounded control pears. All fruits were incubated in a growth chamber at 30/26°C (day/night, 12-h photoperiod, 90% relative humidity). After 3 days, the first symptom of shallow black spots was observed on nine wounded inoculated pears, and after 5 days mean lesion diameter was 19.8 mm, whereas the symptom of nonwounded pears was mild. The water controls remained healthy. To complete Koch’s postulates, F. proliferatum was again isolated from infected pears. To our knowledge, this is the first report of F. proliferatum causing rot on Cuiguan pear in China and around the world. First Report of Leaf Rot on Catalpa bungei caused by Fusarium verticillioides in China

Abstract:Catalpa bungei originates from China. Because of its well-developed root system and strong resistance to wind and soil, it is one of the top ten recommended species of ecological management (Jian et al., 2022). In September 2023, a severe leaf rot of C. bungei was observed at Lanlake farm (500 acres) in Nanyang (33°3'23" N, 112°28'50" E), China. The incidence rate of leaf rot reached 45% (n = 100). The pale-yellow spots initially appeared on the adaxial surface of leaf margins, subsequently enlarging to form irregular black rot lesions, with the yellow halos around the necrotic area of the lesion, ultimately causing the entire leaves to wither. Diseased leaves (20) were collected, cut into pieces, sterilized, and then placed on potato dextrose agar (PDA). A total of 25 purified fungal strains were isolated, and three strains (QS2-1, QS2-2, QS2-3) from distinct areas were selected for further analysis. Each strain produced abundant aerial mycelium, initially white, which later developed purple pigments. The aerial conidiophores were sparsely branched, ending with verticillate phialides. The strains generally produced many more microconidia than macroconidia on PDA media. Microconidia were clavate and measured 3.9 to 6.6 × 1.1 to 2.4 μm (n = 50). To produce macroconidia, we used YPG liquid medium (0.3% yeast extract, 1% peptone, and 2% glucose) with shaking (200 r.p.m.) for 5 days. Macroconidia were slender, straight, and measured 19.5 to 27.1 × 1.9 to 3.5 μm, with 3 to 5 septa (n = 50). The morphological characteristics matched the species description of Fusarium verticillioides (Sacc.) Nirenberg 1976 (Leslie and Summerell, 2006). The rDNA internal transcribed spacer (ITS), β-tubulin gene (tub2), translation elongation factor 1-alpha gene (tef1), calmodulin (cmdA), RNA polymerase II largest subunit (rpb1), and RNA polymerase II second largest subunit (rpb2) were amplified for molecular identification (O'Donnell et al., 2022). The sequences were submitted to GenBank with accession numbers OR741762, OR741763, OR741765 (ITS), OR762222, OR762223, OR939807 (tub2), OR939799, OR939800, PQ035927 (tef1), OR778611, OR939808, OR939809 (cmdA), PQ035921, PQ035922, PQ035923 (rpb1), and PQ035924, PQ035925, PQ035926 (rpb2). BLASTn analysis of QS2-1 sequences exhibited 99% similarity with F. verticillioides sequences (strains CBS 576.78) MT010888 of cmdA (711/713, 99%), MT010956 of rpb1 (1790/1791, 99%), and MT010972 of rpb2 (868/870, 99%). A phylogenetic tree was constructed using concatenated sequences along with the sequences of the type strains employing the neighbor-joining method, showing the three strains formed a clade with the type strain CBS 576.78. Pathogenicity was tested on 10 healthy potted seedlings by spraying them with a conidial suspension (106 conidia ml-1), while 5 seedlings were sprayed with sterilized water as a control. The plants were placed in climate incubators. Ten days after inoculation, typical lesions were observed on the treated plants, but not on the control group. The reisolated strains were identified as F. verticillioides through morphological characterization, thus fulfilling Koch's postulates. F. verticillioides is known to cause Fusarium ear rot on maize, and other plants including Brassica rapa (Akram et al., 2020) and Schizonepeta tenuifolia (Li et al., 2024). This is the first documented instance of F. verticillioides causing leaf rot on C. bungei globally. Identifying the pathogen is critical to implementing effective disease management strategies, especially in choosing proper pesticide agents and screening disease-resistant varieties.
First Report of Diplodia intermedia Causing Canker and Dieback Diseases on Apple Trees in Canada

Abstract:A dieback of apple trees (Malus domestica (Suckow) Borkh.) associated with cankers was observed in commercial orchards in southwestern Ontario, Canada, in 2019. Fifteen 2 to 10-year-old symptomatic trees were collected from three orchards exhibiting up to 37% disease incidence. Small sections of diseased wood (1 cm long) were surface sterilized with 70% ethanol for 30 sec and 1% NaClO for 20 min, rinsed thrice in sterile water, placed on 2% PDA (Difco) amended with kanamycin (50 mg liter-1), and incubated at 22°C for 5 days in the dark (Ilyukhin et al. 2023). Fungal colonies that were consistently isolated were hyphal-tipped, transferred to individual PDA plates and incubated at 22°C for 7 days in the dark. Purified isolates with same characteristics were classed into morphotypes. One morphotype was initially white and turned dark olivaceous with dense aerial mycelium. Pycnidia were produced on pine needles on PDA (Fig. S2) after incubation at 22°C for 17 days in the dark. Conidia were brown, aseptate, ovoid, and measured 27.9 to 31.3 μm x 12.1 to 14.2 μm (mean ± S.D. of 15 conidia = 29.9 ± 0.9 μm × 13.2 ± 0.6 μm), the typical morphology of a Diplodia sp. (Phillips et al. 2012). Genomic DNA was extracted from a 7-day-old culture of a representative isolate M45-28, using the Plant/Fungi DNA Isolation Kit (Norgen Biotech, Canada). The internal transcribed spacer (ITS), translation elongation factor 1-α (EF1-α) and β-tubulin gene regions were amplified and sequenced with primers ITS1/ITS4, EF1-728F/EF1-986R and Bt2a/Bt2b and deposited in GenBank with accession numbers MZ970605, MZ995430 and MZ995431, respectively. Based on the sequence, the fungus was identified as Diplodia intermedia A.J.L. Phillips et al. and matched isolates from different hosts and countries (ITS: 100%, MG220378; EF1-α: 100%, MG220385; β-tubulin: 99.24%, MT592502). The maximum likelihood-based phylogenetic analysis of ITS, EF1-α and β-tubulin concatenated sequences was performed using IQ-Tree 2.2.2.7 (Minh et al. 2020). M45-28 was clustered with high bootstrap support values with D. intermedia isolates from the Westerdijk Fungal Biodiversity Institute collection, including the ex-holotype (CBS 124462) (Fig. S1). A living culture of M45-28 was deposited in the Canadian Collection of Fungal Cultures (DAOMC 252253). Pathogenicity assay was conducted by inoculating mycelial plugs from a 7-day-old culture of M45-28 into wounds made on the trunk of 5 eight-month-old potted healthy 'Royal Gala' apple seedlings. Five control seedlings were inoculated with sterile plugs. Canker symptoms appeared 15 days after inoculation, spread around, up and down the main stem from the inoculation point, and by 7 weeks the upper portion of the seedling was dead (Fig. S2). Diplodia intermedia was re-isolated from all inoculated seedlings and species identity was confirmed by sequencing as described above, fulfilling Koch's postulates. Control seedlings remained symptomless and the fungus was not isolated from the wood. Diplodia intermedia was reported to cause cankers on apple in Uruguay (Delgado-Cerrone et al. 2016), wild apple (Malus sylvestris) in Portugal (Phillips et al. 2012), grapevines in France (Comont et al. 2016) and forest trees in Iran (Kazemzadeh Chakusary et al. 2019). To the best of our knowledge, this is the first report of D. intermedia causing canker and dieback diseases on apple trees in Canada. Further studies are required to better understand the epidemiology involved in the dynamic spread of the disease in order to recommend an adequate phytosanitary program for its control.
Fungi from Galleries of the Emerald Ash Borer Produce Cankers in Ash Trees

Abstract:The emerald ash borer (EAB, Agrilus planipennis) is a devastating invasive pest that has killed millions of ash trees in the United States and Canada. EAB was discovered in the US in 2002 and first reported in Minnesota in 2009. It attacks ash trees that are native to the United States, including Fraxinus americana (white ash), F. nigra (black ash) and F. pennsylvanica (green ash). It also attacks Chionanthus virginicus (white fringe tree). Seven species of fungi isolated and identified only from EAB-infested trees in a previous study as having the potential to cause cankers were used to test their pathogenicity in F. americana (white ash). The fungi used were Cytospora pruinosa, Diplodia mutila, Diplodia seriata, Paraconiothyrium brasiliense, Phaeoacremonium minimum, Phaeoacremonium scolyti, and Thyronectria aurigera. Two field experiments that used F. americana used two inoculation methods: woodchip and agar plug inoculations. Results indicated that all of the fungi tested caused cankers in varying amounts, as compared to the controls. The largest cankers were caused by D. mutila (270 mm2), C. pruinosa (169 mm2), and D. seriata (69 mm2). All fungi except for T. aurigera were re-isolated and sequenced to confirm Kochs’ postulates. Canker-causing fungi found in association with EAB galleries have the potential to contribute to tree dieback and mortality.

Abstract:This paper presents a comparative study of the power consumption of two methods to cancel the offset capacitance in high-resolution capacitive displacement sensors. One method is applied based on correlated-double-sampling (CDS) technique and the other method is applied using oversampling technique. The analytical comparison predicts similar power consumption, if the same resolution for the same conversion time need to be reached. Experimental results with implementations of the two methods confirmed the analytical prediction.
First Report of Alternaria alternata Causing Leaf Spot on Catalpa bungei in China

Abstract:Catalpa bungei originates from China. It is fast-growing and possesses a vertically aligned trunk, rendering it a commendable construction material and a significant economic species. In July 2022, a serious leaf spot occurred in the LanLake farm (surveyed area of about 700 acres) in Nanyang (33°3'23" N, 112°28'50" E), Henan Province, China. The incidence rate of leaf disease reached 54% (n=100). The disease initially manifested as irregular round spots with a yellowish-brown hue, subsequently extending in all directions. Later, the lesion periphery exhibited a darkening effect, leading to yellowing. Twenty diseased leaves were randomly collected and cut into small pieces at the interfaces between infected and healthy tissues. The tissues were sterilized in a solution of 75% ethanol and 1% NaClO for 30 seconds and 1 minute, respectively. After rinsing in sterile water, the pieces were placed on potato dextrose agar (PDA) plates and incubated at 25°C for 5 days. A total of 29 purified fungal strains were acquired, exhibiting comparable phenotypes in terms of morphological characteristics. Three strains (QS1-1, QS1-2, and QS1-3) were isolated for subsequent investigations. The colony exhibited abundant aerial mycelium with shades ranging from dark green to grey-brown on the reverse side. To analyze the morphological characteristics of conidia, potato carrot agar (PCA) was used as the culture medium and incubated at 25°C with a 12-hour light/dark cycle. Conidia were obclavate or spheroidal, dark brown, with 3 to 5 transverse septa, and 1 to 4 longitudinal septa, measuring 12.4 to 36.7 × 4.4 to 9.0 μm (n=100), with conical beak lengths ranging from 0 to 4.3 μm. These morphological traits suggested that the pathogen shares similarities with the Alternaria species. The rDNA internal transcribed spacer (ITS), translation elongation factor 1-alpha gene (tef1), glyceraldehyde 3-phosphate dehydrogenase gene (gapdh), and RNA polymerase II second largest subunit (rpb2) were amplified for further molecular identification. The resultant sequences were submitted to GenBank with the following accession numbers: OR733559, OR742124, OR761873 (ITS), OR939796, OR939797, OR939798 (tef1), OR939801, OR939802, OR939803 (gapdh), and PP054846, PP054847, PP054848 (rpb2). A Phylogenetic tree was constructed of combined genes (ITS, tef1, gapdh, and rpb2) of sequences, alongside the sequences of the type strains by the neighbor-joining method. The three strains formed a clade with the strains CBS 121456 of Alternaria alternata in phylogenetic trees, being separated from other Alternaria spp. The morphological features and molecular analyses supported the strains as members of Alternaria alternata (Woudenberg et al. 2015). To validate pathogenicity, a conidial suspension (106 conidia ml-1) of all three strains was inoculated onto three healthy leaves of five seedlings, with 50 μl of inoculum absorbed with cotton balls. Another group of five plants received sterile water as a control. All plants were incubated in a climate chamber at 28°C and 90% relative humidity. Four days post-inoculation, lesions resembling natural phenomena were observed, whereas control plants showed no symptoms. Subsequent reisolation produced cultures that were morphologically and molecularly identical to the original strains, fulfilling Koch's postulates. Stem canker of C. bungei caused by Phytophthora nicotianae has been reported in China (Chang et al. 2022). This is the first report of A. alternata causing leaf spots on C. bungei in China. Further research is required on management options to control this disease and the host range still needs to be clarified for accurate disease management.
Impact of cytogenetic abnormalities in adults with Ph-negative B-cell precursor acute lymphoblastic leukemia.

Abstract:Multiple cytogenetic subgroups have been described in adult Philadelphia chromosome (Ph)-negative B-cell precursor (BCP) acute lymphoblastic leukemia (ALL), often comprising small numbers of patients. In this study, we aimed to reassess the prognostic value of cytogenetic abnormalities in a large series of 617 adult patients with Ph-negative BCP-ALL (median age, 38 years), treated in the intensified Group for Research on Adult Acute Lymphoblastic Leukemia (GRAALL)-2003/2005 trials. Combined data from karyotype, DNA index, fluorescence in situ hybridization, and polymerase chain reaction screening for relevant abnormalities were centrally reviewed and were informative in 542 cases (88%), allowing classification in 10 exclusive primary cytogenetic subgroups and in secondary subgroups, including complex and monosomal karyotypes. Prognostic analyses focused on cumulative incidence of failure (including primary refractoriness and relapse), event-free survival, and overall survival. Only 2 subgroups, namely t(4;11)/KMT2A-AFF1 and 14q32/IGH translocations, displayed a significantly worse outcome in this context, still observed after adjustment for age and after censoring patients who received allogeneic stem cell transplantation (SCT) in first remission at SCT time. A worse outcome was also observed in patients with low hypodiploidy/near triploidy, but this was likely related to their higher age and worse tolerance to therapy. The other cytogenetic abnormalities, including complex and monosomal karyotypes, had no prognostic value in these intensive protocols designed for adult patients up to the age of 60 years.

Abstract:Fusarium wilt is one of the most devastating diseases of cotton ( Gossypium spp.). It is caused by the soil-borne pathogen Fusarium oxysporum Schlechtend. f. sp. vasinfectum (Atk.) Snyd. & Hans (Atkinson 1892). To date, eight races of F. oxysporum f. sp. vasinfectum have been reported worldwide based on their diverse genetics and reactions to the host (Cianchetta et al. 2015). Fusarium wilt symptoms appear at all development stages. The fungus enters through the roots and colonizes the vascular system, leading to discoloration and wilting (Davis et al. 2006). During a survey in the July 2021 growing season, cotton plants showing typical wilting symptoms, stem discoloration, and root rot were observed in two cotton fields in Sumner County, Kansas. To confirm the causal agent, root tissues and lower parts of the stems were collected from 25 diseased cotton plants and cleaned as described by Larran et al. (2001). Small segments (1 to 2 cm) of roots and stems were incubated on potato dextrose agar (PDA) at room temperature. The isolates were purified with the single spore method and 23 of 25 isolates (92%) showed typical morphology of F. oxysporum , with a white-peach pigmentation as reported previously (Leslie and Summerell 2008). Two isolates, CK13B and CK18A, one from each field, were selected, and genomic DNA was extracted with E.Z.N.A Fungal DNA Mini kit (Omega Bio-tek, Norcross, GA). PCR assays were performed as reported previously (Ortiz et al. 2017) to amplify and sequence a portion of three nuclear genes: translation elongation factor ( EF-1α ), phosphate: H * symporter ( PHO ), and β-tubulin ( BT ). BLASTn analysis of CK13B sequences showed 93 to 98% identity with F . oxysporum f. sp. vasinfectum isolates from Australia, with 93.43% (1,706/1,826 bp) identity of EF-1α to isolate AuSeed14 (KT323873), 95.11% (1,849/1,944 bp) of PHO to isolate AuK24232 (KT323909), and 97.56% (1,840/1,886 bp) identity of BT to isolate AusSeed14 (KT323833). Sequences of CK13B for EF1-α , PHO , and BT genes were submitted to GenBank (accession no. ON754247, ON754248, and ON754249, respectively). The CK18A isolate showed high identity with F . oxysporum f. sp. vasinfectum races 1, 2, and 6 lineage isolates (Ortiz et al. 2017), with 99.89% (1,822/1,824 bp) identity of the EF1-α gene to isolate CDR238 (KT323838) from Arkansas, 98.36% (1,916/1,948 bp) identity of the PHO gene to isolate CDR1131 (KT323887) from Louisiana and 98.78% (1,864/1,887 bp) identity of the BT gene to isolate ATCC36198 (KT323799) from Brazil. Sequences of the CK18A isolate were submitted to GenBank (accession no. ON651444, ON725043, and ON725044, respectively). Pathogenicity of both isolates was tested on G . hirsutum as previously described (Kim et al. 2005), with three replicates per isolate and a control treatment in a growth chamber. Briefly, cotton seedlings were maintained with a 12 h light/dark cycle at 25 to 20°C. At the first true leaf stage, seedlings were uprooted and roots were cleaned by rinsing with sterile water and placed in a conidial suspension (1 × 10 6 conidia/ml) for 2 h while healthy control seedlings were dipped in water only. Plants were put into sterile soil, and the results were observed and recorded every 7 days for 30 days. The inoculated plants showed the same symptoms as those observed in the fields: wilting, leaf chlorosis, and necrosis. Both isolates were reisolated from the roots of the inoculated plants. DNA was extracted and used for PCR with the specific gene primers listed above. The sequences matched those of the original isolates, completing Koch's pustulates. To our knowledge, this is the first report of F. oxysporum f. sp . vasinfectum causing Fusarium wilt disease of cotton in Kansas. These results will help growers select cultivars and design disease control strategies (Davis et al. 2006). Further work is needed to determine the specific races in the area.
