Abstract:A plant expression vector pBIA9-AMF containing an antisense fragment of the CYP86MF gene and the tapetum-specific A9 promoter was constructed. Plasmid vectors were introduced by floral-dipping and pollen-tube transformation methods to Chinese cabbage pak-choi (Brassica campestris ssp. chinensis (L.) Makino var. communis Tsen et Lee, syn. B. rapa ssp. chinensis (L.) Makino var. communis Tsen et Lee) and flowering Chinese cabbage (B. campestris ssp. chinensis (L.) Makino var. parachinensis (Bailey) Tsen et Lee). Results showed that Kan(R) seedlings could be obtained by the pollen-tube method through germination tests of T-1 progeny seeds, but not by the floral-dipping method. One of the two Kan(R) seedlings proved that the antisense fragment of the CYP86MF gene was integrated into the Chinese cabbage genome by PCR amplification and Southern blotting. Northern hybridization indicated that the CYP86MF gene, under the A9 promoter, was inhibited in the transformant, and self-infertility was found in the transgenic plantlets after self-pollination. Comparison of floral organs and the morphology of microspores by scanning electron microscopy of transformant and wild-type plantlets, showed no significant differences except that microspores from the transgenic plant were covered in a layer of an unknown substance. We suggest this layer might be the cause of the male-sterile phenotype induced by the antisense fragment of the CYP86MF gene.
Production of Brassica Tri-Genomic Vegetable Germplasm by Hybridisation Between Tuber Mustard ( Brassica Juncea ) and Red Cabbage ( B. Oleracea )

Abstract:Brassica is a key agricultural genus, containing oilseed and vegetable crops sharing three major genomes (A, B and C) distributed in diploid (genome: AA, BB and CC) and allotetraploid (genome: AABB, AACC and BBCC) species. Interspecific hybridisation between Brassica species is an important way to combine valuable characters of different species. In the present study, interspecific hybrids between tuber mustard (AABB) and red cabbage (CC) were obtained with the aid of embryo culture. The results showed that ovules at five different maturity levels (7, 9, 10, 13 and 14 days after pollination), which were cultured on Murashige and Skoog medium supplemented with 3 % sucrose, 0.2 % active carbon and 400 mg/L glutamine, showed varying seed survival rates. A total of 15 (5 %) seeds were obtained, with ovules cultured at 14 days after pollination yielding the most seeds. Subsequently, eight (53 %) of the seeds did not germinate into normal plants. Furthermore, the putative hybrids were confirmed by random amplified polymorphic DNA (RAPD) and flow cytometry analysis, and only one (14 %) of the seven plants was confirmed to be a true tri-genomic hybrid (genome: ABC). Then the nodal segments of the true hybrid were treated with liquid medium supplemented with colchicine to obtain an allohexaploid, as confirmed by flow cytometry analysis and morphological observation. The synthesized hybrid showed unique phenotypes in comparison with the parents, such as in stem colour and leaf margin. The synthesized Brassica tri-genomic vegetable germplasm with A, B and C genomes has potential as a source of new crops for agriculture as well as a basis for further investigation of the mechanisms of Brassica evolution and adaptation.
Study on Transformation of Cotton Pollen Using Agrobacterium Tumefaciens

Abstract:The pollen-mediated transformation approach could not only circumvent the tedious tissue culture procedures like in Agrobacterium infection, but also circumvent the incertitude of the pollen tube path-way. So,its transformants combined from the transformed pollen and eggs would not be chimera.At last this approach is simple, easy to operate, and could be widely used in practice. For the transformation happened in the pollen culture medium, the solution with a suitable solute has to be selected. This study showed the medium consisting of (0.1%) H_3BO_3 、(0.3%) Ca(NO_3)_2、(0.2%) MgSO_4·7H_2O、(0.1%) KNO_3 and 45% sucrose could maintain normal turgidity of the pollens and facilitate them to be germinated. There are more than 95% pollens don't dispose in this medium and the germinating rate was up to 58.5%. A transient GUS expression assay and the exploded pollen rate proved that the transformation method with 1‰ Tween-20 in the medium using vacuum-infiltration was a better method. Finally, four transformed seedlings were confirmed by Kanamycin selection, GUS expression and Northern blot. Therefore this transformation approach is available.
Identification and preliminary analysis of a new PCP promoter from Brassica rapa ssp. chinensis

Abstract:The promoter of Brassica campestris Male Fertile 5 ( BcMF5 ), a pollen coat protein member, class A ( PCP-A ) gene family, was isolated from Brassica rapa L. ssp. chinensis Makino (Chinese cabbage-pak-choi) by Thermal Asymmetric Interlaced Polymerase Chain Reaction (TAIL-PCR). Sequence analysis suggested that the 605-bp promoter of BcMF5 appears to be a pollen promoter. In an attempt to confirm the promoter activity of BcMF5 promoter, −609 to +3 bp and −377 to +3 bp fragments of the upstream sequence of BcMF5 were inserted at the site upstream of the coding region of the uid A gene in the sense orientation to construct two deletion expression vectors. Transient expression analysis in onion epidermal cells by particle bombardment showed that both −609 to +3 bp and −377 to +3 bp fragments of BcMF5 promoter were capable of driving β-glucuronidase gene expression. Furthermore, by Agrobacterium-mediated genetic transformation method, Arabidopsis transgenic Kan R plants were obtained. GUS assay analysis revealed that the promoter of BcMF5 induced gene expression at the early stage of anther development and drove high levels of GUS expression in anther walls, upper regions of petals, pollen, and pollen tubes in the middle and late stage of anther development, but did not drive any expression in sepals and pistils.
Improvement of the Transformation System in Chinese Cabbage(brassica Campestris Ssp. Chinensis Makino)

Abstract:The binary plasmid vector pBI35S-AMF was transferred into cv.Shanghaiqing of Chinese cabbage pak-choi(Brassica campestris ssp. Mokino chinensis var.communis Tsen et Lee) and cv.Youqing of flowering Chinese cabbage(B.campestris ssp. chinensis Makino var.parachinensis(Bailey) Tsen et Lee) by Agrobacterium-mediated method.The pivotal factors,which influenced the transformation frequency,were compared,and the parameters of transformation system were optimized.It was showed that selective medium containing 300 mg/L ampicillin,which inhibited growth of Agrobacterium,led to production of more Kan~R shoots;the time required for preculture and co-culture was three days and two days respectively;and the efficiency of inoculation with LBA4404/pBI35S-AMF was significantly higher than with EHA105/pBI35S-AMF.Molecular analysis to Kan~R seedlings showed that the frequency of Southern blotting was 82.2% and most of transformants were multi-copies integration.Moreover,the results indicated that the efficiency of transformation system was stable.

Abstract:In our earlier work, a cytochrome P450 CYP86MF gene was isolated from floral bud of Chinese cabbage ( Brassica campestris L. ssp. chinensis Makino, syn. B. rapa L.) by mRNA differential display PCR (DD-PCR) and rapid amplification of cDNA ends (RACE). To unravel the biological function of CYP86MF gene, the antisense fragment from the CYP86MF gene was transferred into Chinese cabbage pak-choi ( B. campestris ssp. chinensis var. communis Tsen et Lee). Out of 22 plants transformed with the antisense gene constructed from the CYP86MF , 20 reached to flowering stage. Morphological investigations showed that the transgenic plants developed the normal floral organ. However, they remained self-infertile, even when artificial self-pollination was performed in the bud stage. Pollen germination test indicated that the pollen from the transgenic line TB-2 could not germinate normally. Further physiological, biochemical and cytological analyses showed that only significant difference was detectable in contents of the endogenous hormones, and a layer of unknown material adhered to the surface of microspore. The present studies thus provided valuable clues for understanding the biological function of the CYP86C subfamily genes. Furthermore, our studies also demonstrate a novel method for obtaining artificial male sterility line of Chinese cabbage.
Comparative Transcriptome Analysis between Fertile and CMS Flower Buds in Wucai (Brassica campestris L.)

Abstract:Background: Wucai (Brassica campestris L. ssp. chinensis var. rosularis Tsen) is a variant of nonheading Chinese cabbage (Brassica campestris L.), which is one of the major vegetables in China. Cytoplasmic male sterility (CMS) has been used for Wucai breeding in recent years. However, the underlying molecular mechanism of Wucai CMS remains unclear. In this study, the phenotypic and cytological features of Wucai CMS were observed by anatomical analysis, and a comparative transcriptome analysis was carried out to identify genes related to male sterility using Illumina RNA sequencing technology (RNA-Seq). Results: Microscopic observation demonstrated that tapetum development was abnormal in the CMS line, which failed to produce fertile pollen. Bioinformatics analysis detected 4430 differentially expressed genes (DEGs) between the fertile and sterile flower buds. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were performed to better understand the functions of these DEGs. Among the DEGs, 35 genes (53 DEGS) were implicated in anther and pollen development, and 11 genes were involved in pollen cell wall formation and modification; most of these showed downregulated expression in sterile buds. In addition, several genes related to tapetum development (A6, AMS, MS1, MYB39, and TSM1) and a few genes annotated to flowering (CO, AP3, VIN3, FLC, FT, and AGL) were detected and confirmed by qRT-PCR as being expressed at the meiosis, tetrad, and uninucleate microspore stages, thus implying possible roles in specifying or determining the fate and development of the tapetum, male gametophyte and stamen. Moreover, the top four largest transcription factor families (MYB, bHLH, NAC and WRKY) were analyzed, and most showed reduced expression in sterile buds. These differentially expressed transcription factors might result in abortion of pollen development in Wucai. Conclusion: The present comparative transcriptome analysis suggested that many key genes and transcription factors involved in anther development show reduced gene expression patterns in the CMS line, which might contribute to male sterility in Wucai. This study provides valuable information for a better understanding of CMS molecular mechanisms and functional genome studies in Wucai.
Factors Affecting Transformation Efficiency by Micro-Injecting Agrobacterium into Flower Bud of Chinese Cabbage

Abstract:Three main parameters were selected to study their importance in transformation by bud microinjection in non-head Chinese cabbage [Brassica campestris ssp. Chinensis (L.) Makino var. Communis Tsen et Lee]. The results showed that the developmental stage of floral bud, the concentrations of sucrose and surfactant Silwet L-77 were critical for the successful transformation by this new method. The suitable bud size is 2- 3 mm in length, the favorable concentration of sucrose and surfactant Silwet L-77 are 8 and 0.02% respectively. When the sucrose concentration was greater than 10% or that of Silwet L-77 was above 0.1%, the treated buds became yellow and finally blighted. 4/6 T1 seedlings resistant to kanamycin were positive by PCR analysis, and T2 progeny of all these positive T1 plants have one or more hybridizing bands by Southern blot. Under 5% sucrose, 0.02% Silwet L-77 and grade 2 bud (2- 3 mm in its length) parameters, the most favorable transformation efficiency is about 0.56%, and mean efficiency reaches 0.16% in all experiments indicating that bud microinjection is potential transformation way in non-head Chinese cabbage.

Abstract:Non-heading Chinese cabbage, a variety of Brassica campestris, is an important vegetable crop in the Yangtze River Basin of China. However, the immaturity of its stable transformation system and its low transformation efficiency limit gene function research on non-heading Chinese cabbage. Agrobacterium rhizogenes-mediated (ARM) transgenic technology is a rapid and effective transformation method that has not yet been established for non-heading Chinese cabbage plants. Here, we optimized conventional ARM approaches (one-step and two-step transformation methods) suitable for living non-heading Chinese cabbage plants in nonsterile environments. Transgenic roots in composite non-heading Chinese cabbage plants were identified using phenotypic detection, fluorescence observation, and PCR analysis. The transformation efficiency of a two-step method on four five-day-old non-heading Chinese cabbage seedlings (Suzhouqing, Huangmeigui, Wuyueman, and Sijiu Caixin) was 43.33%–51.09%, whereas using the stout hypocotyl resulted in a transformation efficiency of 54.88% for the 30-day-old Sijiu Caixin. The one-step method outperformed the two-step method; the transformation efficiency of different varieties was above 60%, and both methods can be used to obtain transgenic roots for functional studies within one month. Finally, optimized ARM transformation methods can easily, quickly, and effectively produce composite non-heading Chinese cabbage plants with transgenic roots, providing a reliable foundation for gene function research and non-heading Chinese cabbage genetic improvement breeding.

Abstract:Abstract Chinese chestnut ( Castanea mollissima ) is an important germplasm resource for the breeding of Castanea species worldwide with vital ecological and economic value. Biotechnology overcomes the limitations of traditional breeding and accelerates germplasm improvement. However, a genetic transformation system for Chinese chestnut has not yet been established. In this study, a stable and efficient Agrobacterium -mediated genetic transformation method for Chinese chestnut is described. Embryogenic calli of C. mollissima cv. ‘Yanshanhongli’ were used as the target material. The sensitivity of embryogenic calli to kanamycin was determined, whereby the proliferation of non-transformed calli was completely inhibited at 180 mg/L. Antibiotic inhibition results for Chinese chestnut embryogenic calli showed that 50 mg/L cefotaxime and 500 μM timentin completely inhibited the growth of Agrobacterium tumefaciens but did not affect the normal growth of Chinese chestnut embryogenic calli. When embryogenic calli were co-cultured for 2 days with Agrobacterium tumefaciens strain AGL1 harboring the PBI121-EGFP plasmid, an embryogenic callus transformation efficiency of 4.55% was obtained, and two transgenic chimera were acquired. This Agrobacterium -mediated transformation system for Chinese chestnut provides a fundamental platform for genetic improvement of core germplasm and for further verification of gene function.

Abstract:Many genes in the genic male sterile A/B line (Bajh97-01A/B) of Chinese cabbage pak choi ( Brassica campestris L. subsp. chinensis Makino) are expressed differentially, and some play critical roles in the formation of pollen walls. In this study, one of these genes, Brassica campestris Male Fertility 16 ( BcMF16 ), has been isolated and characterized. The BcMF16 gene shares approximately 85% nucleotide sequence homology with two exopolygalacturonase (EC3.2.1.67) genes of Arabidopsis thaliana . Cluster analysis of polygalacturonase peptides indicate that BcMF16 belongs to the pollen polygalacturonase clade. Quantitative real-time PCR analysis has revealed that BcMF16 is specifically expressed in reproductive tissues of the fertile line of genic male sterile A/B line of Chinese cabbage pak choi, and that expression levels dramatically increased during later stages of pollen development. In situ hybridization has demonstrated that BcMF16 is specifically and transiently expressed in both tapetum and pollen following microspore separation at the tetrad stage.
Identification and Functional Characterisation of a Novel Anther-Specificltppromoter Frombrassica Campestrisssp.chinensis

Abstract:The pollen development-related gene msLTP from Brassica campestris ssp. chinensis belongs to the lipid transfer protein (LTP) gene family. The promoter of the msLTP gene was isolated by thermal asymmetric interlaced PCR (TAIL-PCR). The functional elements of the promoter (named Bc15) were analysed. Some anther/pollen-specific elements, such as the GGTT box and the GTGA box, were identified in the sequence. In an attempt to confirm the promoter activity, a series of 5'-truncated constructs was fused with the GUS reporter gene to functionally analyse the key regulatory regions of the promoter. Transient expression analysis showed high GUS activity in onion epidermal cells containing the region from -731 to -1 bp. The truncated construct bearing the -731 to -1 bp fragment was stably introduced into Arabidopsis. Histochemical analysis of the transgenic plants showed distinct and specific GUS expression at different stages of anther/pollen development. Transverse sections of flower buds further indicated that the Bc15 promoter activates reporter gene expression in the anther specifically. These findings elucidated the molecular mechanisms of LTP gene regulation during pollen development, and suggested a potential application of the Bc15 promoter in engineering male sterility for hybrid production.
Genome-wide identification, transcript profiling and functional analyses of PCP gene family in Wucai (Brassica campestris)

Abstract:Pollen coat proteins (PCPs) are cysteine-rich small-molecule proteins, which exhibit high levels of polymorphism and are expressed in gametocytes. Previous investigations have revealed that PCP genes are involved in pollen wall synthesis, pollen-stigma recognition, and pollen development and germination. However, gene expression and function of PCP family in pollen development is not well understood in Wucai ( Brassica campestris L.). In this study, genome-wide identification and expression analysis of the BcPCP gene family members were conducted, including their physical and chemical properties, chromosome localization, phylogenetic relationships, gene structure, and tertiary structure. A total of 20 BcPCP genes were identified and classified into three subfamilies showing high homology to Arabidopsis thaliana. Expression pattern analysis indicated that the BcPCP gene family exhibits higher expression levels in reproductive organs, suggesting their potential involvement in the reproductive development. Notably, BraA02g002400.3 C , potentially associated with male sterility, was identified through multiple transcriptomic and proteomic datasets. Subsequently, sequence analysis revealed its homology with the Arabidopsis GRP20 gene, and thus it was named BcGRP20 . Functional analysis of this gene showed that overexpression of BcGRP20 gene in the Arabidopsis grp20 mutant could restore anther fertility. Overall, our findings indicate that BcGRP20 plays a critical role in pollen development and may be the causative gene for male sterility in Wucai. This study provides candidate genes for further functional identification of BcPCP genes in Wucai, which are crucial for anther development.

Abstract:In our earlier work, a pollen-expressed polygalacturonase gene BcMF6 was isolated from floral bud of Chinese cabbage ( Brassica campestris L. ssp. chinensis Makino) by cDNA-amplified fragment length polymorphism (cDNA-AFLP) transcript profiling and rapid amplification of cDNA ends (RACE). To unravel the biological function of BcMF6 gene, the antisense fragment from the BcMF6 gene with A9 promoter and CaMV35S promoter was transferred into flowering Chinese cabbage ( B. campestris ssp. chinensis var. parachinensis ). Out of transgenic plants transformed with the antisense gene constructed from the BcMF6 , transgenic line with A9 promoter have a similar appearance to that with CaMV35S promoter. Morphological investigations showed that the transgenic plants developed the smaller floral organ with thin anther and less pollen. Pollen germination test indicated that only near 50% the pollen from the transgenic line could normally germinate. Further scanning electron microspore analysis of transgenic plants confirmed that half of pollen was abnormal. Cytological comparisons of microspore development also demonstrated that process of microsporogenesis was held up, microspores maturation was disrupted and pollen grain fail to separate, finally. In a word, the present study revealed that BcMF6 , as a polygalacturonase gene, has a role in pollen maturation and pollen tube growth.
Stable expression of exogenous imported sporamin in transgenic Chinese cabbage enhances resistance against insects

Abstract:Cultivating insect pest-resistant varieties is one of the most effective ways to prevent or mitigate pest infestation in Chinese cabbage ( Brassica campestris ssp. chinensis ). Via the agrobacterium tumefaciens-mediated transformation method, this study introduced the protease inhibitor encoding gene sporamin into two widely cultured cultivars ‘Youdonger’ and ‘Shanghaiqing’, of the common variety of Chinese cabbages ( B. campestriss ssp. chinensis var. communis ), getting transgenic plants with high sporamin expression. In vitro insect bioassays indicated that, compared with the wild type plants, the transgenic plants exhibited improved resistance to diamondback moth ( Plutella xylostella L.) The analysis of inheritance pattern of exogenous sporamin in the progenies of single copy insertion transgenic lines demonstrated that sporamin could be inherited and expressed stably in transgenic progenies. Field survey of the insect resistance under the normal culture condition confirmed the enhanced resistance in transgenic progenies to diamondback moth. Our results strongly suggest that sporamin is an efficient candidate gene for insect-resistant genetic engineering in Chinese cabbage.

Abstract:Cytological features of 'Aijiaohuang' chinese cabbage-pak-choi (Brassica campestris ssp. chinensis) Bcajh97-01A/B genic male-sterile AB line were examined to determine phenotypic reasons for male sterility. The sterile line Bcajh97-01A was found to undergo aberrant cytokinesis during male meiosis. Transcriptional profiling of the flower buds of both fertile and sterile plants was performed at the periods preceding meiosis, at the tetrad to uninucleate pollen period, and at the binucleate to mature pollen period. Transcript-derived fragments (TDFs) from corresponding genes that were expressed in flower buds at these three different stages could be divided into nine classes. We sequenced a total of 14 new TDFs that were differentially displayed at particular pollen developmental stages, including eight genes with unknown or hypothetical functions and six genes showing significant homology with known genes. This characterization of the Bcajh97-01A genic male-sterile line allowed the identification of candidate genes underlying genic male sterility.

Abstract:Results of PCR amplification and restriction digestion showed that the fragment of antisense gene CYP86MF was introduced into pBI121 plasmid and the expression vector pBI35S-AMF was transferred into Agrobacterium successfully. Then, the antisense gene fragment was transferred into Chinese cabbage (Brassica campestris L. ssp. chinensis Makino) cv. ^Shanghaiqing^ and more than 130 Kan^{R} plantlets were obtained.
Male-Sterile Plants Induced by Anti-gene CYP86MF in Brassica oleracea var. Italica

Abstract:An anti-gene CYP86MF was introduced into hypocotyls of broccoli(Brassica oleracea L.var. italica Plenck) with Agrobacterium tumefaciens, and transgenic plants were obtained by kanamycin selection. It was proved that anti-CYP86MF was integrated with chromosome of transgenic plant, according to the results of PCR, Southern blot, Northern blot. Plants with hypogenetic stamina or ungerminated pollen were observed. The transgenic male-sterile plants could set seeds if artificially pollinated with normal pollen, so the pistil of male-sterile plant was normal, and its sterility was induced by anti-CYP86MF.
Comparative Morphological, Physiological, and Transcriptomic Analyses of Diploid and Tetraploid Wucai (Brassica campestris L.)

Abstract:Polyploid plants often exhibit superior yield, stress resistance, and quality. In this study, homologous tetraploid wucai (Brassica campestris L.) was successfully obtained by spraying seedling growth points with colchicine. The morphological, cytological, and physiological characteristics of diploid and tetraploid wucai were analyzed, and transcriptomic sequencing was performed at three stages of development. Tetraploid seedings grew slowly but exhibited darker leaves, enlarged organs and cells, increased stomatal volume, decreased stomatal density, improved nutritional content, and enhanced photosynthesis. Differentially expressed genes (DEGs) identified in diploid and tetraploid plants at three stages of development were enriched in different pathways. Notably, DEGs identified in the tetraploid plants were specifically enriched in starch and sucrose metabolism, pentose and glucuronate interconversions, and ascorbate and aldarate metabolism. In addition, we found that the light green module was most relevant to ploidy, and DEGs in this module were significantly enriched in the glycolysis/gluconeogenesis and TCA cycle pathways. The differential expression of key glycolysis-associated genes at different developmental stages may be the driver of the observed differences between diploid and tetraploid wucai. This study lays a technical foundation for the development of polyploid wucai germplasm resources as well as the breeding of new varieties with improved quality, yield, and stress resistance. It also provides a good empirical reference for the genetic breeding of closely related Brassica species.
