Abstract:Peanut (Arachis hypogaea L.) is a major species of the family, Leguminosae, and economically important not only for vegetable oil but as a source of proteins, minerals and vitamins. It is widely grown in the semi-arid tropics and plays a role in the world agricultural economy. Peanut production and productivity is constrained by several biotic (insect pests and diseases) and abiotic (drought, salinity, water logging and temperature aberrations) stresses, as a result of which crop experiences serious economic losses. Genetic engineering techniques such as Agrobacterium tumefaciens and DNA-bombardment-mediated transformation are used as powerful tools to complement conventional breeding and expedite peanut improvement by the introduction of agronomically useful traits in high-yield background. Resistance to several fungal, virus and insect pest have been achieved through variety of approaches ranging from gene coding for cell wall component, pathogenesis-related proteins, oxalate oxidase, bacterial chloroperoxidase, coat proteins, RNA interference, crystal proteins etc. To develop transgenic plants withstanding major abiotic stresses, genes coding transcription factors for drought and salinity, cytokinin biosynthesis, nucleic acid processing, ion antiporter and human antiapoptotic have been used. Moreover, peanut has also been used in vaccine production for the control of several animal diseases. In addition to above, this study also presents a comprehensive account on the influence of some important factors on peanut genetic engineering. Future research thrusts not only suggest the use of different approaches for higher expression of transgene(s) but also provide a way forward for the improvement of crops.

Abstract:Peanut ( Arachis hypogaea L.), also known as groundnut, is cultivated globally and is a widely consumed oilseed crop. Its nutritional composition and abundance in lipids, proteins, vitamins, and essential mineral elements position it as a nutritious food in various forms across the globe, ranging from nuts and confections to peanut butter. Cultivating peanuts provides significant challenges due to abiotic and biotic stress factors and health concerns linked to their consumption, including aflatoxins and allergens. These factors pose risks not only to human health but also to the long-term sustainability of peanut production. Conventional methods, such as traditional and mutation breeding, are time-consuming and do not provide desired genetic variations for peanut improvement. Fortunately, recent advancements in next-generation sequencing and genome editing technologies, coupled with the availability of the complete genome sequence of peanuts, offer promising opportunities to discover novel traits and enhance peanut productivity through innovative biotechnological approaches. In addition, these advancements create opportunities for developing peanut varieties with improved traits, such as increased resistance to pests and diseases, enhanced nutritional content, reduced levels of toxins, anti-nutritional factors and allergens, and increased overall productivity. To achieve these goals, it is crucial to focus on optimizing peanut transformation techniques, genome editing methodologies, stress tolerance mechanisms, functional validation of key genes, and exploring potential applications for peanut improvement. This review aims to illuminate the progress in peanut genetic engineering and genome editing. By closely examining these advancements, we can better understand the developments achieved in these areas.

Abstract:Abiotic stresses are major limiting factors that pose severe threats to agricultural production. Conventional breeding has significantly improved crop productivity in the last century, but traditional breeding has reached its maximum capacity due to the multigenic nature of abiotic stresses. Alternatively, biotechnological approaches could provide new opportunities for producing crops that can adapt to the fast-changing environment and still produce high yields under severe environmental stress conditions. Many stress-related genes have been identified and manipulated to generate stress-tolerant plants in the past decades, which could lead to further increase in food production in most countries of the world. This review focuses on the recent progress in using transgenic technology and gene editing technology to improve abiotic stress tolerance in plants, and highlights the potential of using genetic engineering to secure food and fiber supply in a world with an increasing population yet decreasing land and water availability for food production and fast-changing climate that will be largely hostile to agriculture.

Abstract:<h3 class="a-plus-plus">Key message</h3>This review summarizes the recent advances in legume genetic transformation and provides an insight into the critical factors that play a major role in the process. It also sheds light on some of the potential areas which may ameliorate the transformation of legumes.Legumes are an important group of dicotyledonous plants, highly enriched in proteins and minerals. Majority of the legume plants are cultivated in the arid and semi-arid parts of the world, and hence said to be climate resilient. They have the capability of atmospheric nitrogen fixation and thus play a vital role in the ecological sphere. However, the worldwide production of legumes is somehow not up to the mark and the yields are greatly affected by various biotic and abiotic stress factors. Genetic engineering strategies have emerged as a core of plant biology and remarkably facilitate the crop improvement programmes. A significant progress has been made towards the optimization of efficient transformation system for legume plants over the years but this group is still underutilized in comparison to other crops. Among the variety of available DNA delivery systems, Agrobacterium-mediated and particle bombardment have been primarily deployed for optimization and trait improvement. However, recalcitrance and genotype-dependence are some of the major bottlenecks for successful transformation. In this context, the present review summarizes the advances taken place in the area of legume transformation and provides an insight into the present scenario. The challenges and future possibilities for yield improvement have also been discussed.

Abstract:Genetic engineering technique offers myriads of applications in improvement of horticultural crops for biotic and abiotic stress tolerance, and produce quality enhancement. During last two decades, a large number of transgenic horticultural crops has been developed and more are underway. A number of genes including natural and synthetic Cry genes, protease inhibitors, trypsin inhibitors and cystatin genes have been used to incorporate insect and nematode resistance. For providing protection against fungal and bacterial diseases, various genes like chitinase, glucanase, osmotin, defensin and pathogenesis-related genes are being transferred to many horticultural crops world over. RNAi technique has been found quite successful in inducing virus resistance in horticultural crops in addition to coat protein genes. Abiotic stresses such as drought, heat and salinity adversely affect production and productivity of horticultural crops and a number of genes encoding for biosynthesis of stress protecting compounds including mannitol, glycine betaine and heat shock proteins have been employed for abiotic stress tolerance besides various transcription factors like DREB1, MAPK, WRKY, etc. Antisense gene and RNAi technologies have revolutionized the pace of improvement of horticultural crops, particularly ornamentals for color modification, increasing shelf-life and reducing post-harvest losses. Precise genome editing tools, particularly CRISPR/Cas9, have been efficiently applied in tomato, petunia, citrus, grape, potato and apple for gene mutation, repression, activation and epigenome editing. This review provides comprehensive overview to draw the attention of researchers for better understanding of genetic engineering advancements in imparting biotic and abiotic stress tolerance as well as on improving various traits related to quality, texture, plant architecture modification, increasing shelf-life, etc. in different horticultural crops.

Abstract:Although the availability of effective antimicrobial therapy, both otitis media with effusion (OME) and acute bacterial meningitis (ABM) are still important infections for children, leading serious health problems. The most frequently isolated bacteria from cerebrospinal fluid (CSF) of ABM patients are Haemophilus influenzae type b, Neisseria meningitidis and Streptococcus pneumoniae, and middle ear effusion (MEE) samples of OME patients are H. influenzae, S. pneumoniae, Moraxella catarrhalis, respectively. Since they are fastidious bacteria, various problems may arise in the rapid diagnosis in both ABM and OME settings. In this study, the diagnostic value of polymerase chain reaction (PCR) has been searched for the detection of bacterial DNA in CSF and MEE specimens and evaluated in comparison to conventional culture method accepted as the "gold standard". A total of 75 samples (53 CSF, 22 MEE) collected from meningitis and OME suspected children were included in the study. With the conventional culture method, one S. pneumoniae strain was isolated from a CSF sample, and one H. influenzae (non-type b) and two M. catarrhalis strains were isolated from three of MEE samples (total isolation rate: %5.3; 4/75). Standard PCR protocol was applied for the detection of H. influenzae, while multiplex PCR protocol was used for M. catarrhalis and S. pneumoniae, since H. influenzae and S. pneumoniae amplification products were of similar size. PCR revealed genomic DNA sequences of S. pneumoniae from five of the CSF samples, while three H. influenzae, three M. catarrhalis and two S. pneumoniae+M. catarrhalis were detected from MEE samples (total detection rate: %17.3; 13/75). Sensitivity and specificity rates of PCR method were found as 100% and 92.3% for CSF samples, and 100% and 73.7% for MEE samples, respectively, with a total sensitivity of 100%, specificity of 87.3%, positive predictive value of 30.8%, and negative predictive value of 100%. As a result it was concluded that PCR method could be considered as a rapid, reliable and feasible method for the detection of the most common fastidious bacteria that lead to meningitis and OME.

Abstract:Abiotic stresses like drought, heat, and salinity are major causes of agricultural production losses worldwide. However, progress in developing genetically modified crops with better tolerance to these stresses has been slow. Modern agriculture faces various challenges, such as the complex field environment, diverse abiotic stress combinations, and global climatic changes. A combination of approaches is required to improve crop tolerance to abiotic stress significantly. These include understanding stress response and acclimation networks, testing under different growth conditions, using innovative techniques considering genetic and physiological variations in crops, and incorporating enzymes and proteins from other organisms.

Abstract:OBJECTIVE To present results from HIV testing, knowledge of HIV status and socioeconomic factors associated with the probability of having a HIV positive result among transwomen (TW) in Mexico. MATERIALS AND METHODS In 2012, we conducted an HIV seroprevalence survey to 585 TW in Mexico City in three strata: gathering places, the Condesa HIV Clinic and in four detention centers. We estimated the prevalence of HIV in each strata and applied a probit model to the overall sample to analyze factors associated with the probability of a HIV positive result. RESULTS The prevalence of HIV was 19.8% in meeting places; 31.9% in detention centers and 64% among the participants of the clinic. Age, low education and number of sexual partners was positively associated with HIV. CONCLUSIONS Results from the study provide relevant information to design HIV prevention interventions tailored to the needs of the TW population.

Abstract:Abiotic stress conditions such as drought, heat, or salinity cause extensive losses to agricultural production worldwide. Progress in generating transgenic crops with enhanced tolerance to abiotic stresses has nevertheless been slow. The complex field environment with its heterogenic conditions, abiotic stress combinations, and global climatic changes are but a few of the challenges facing modern agriculture. A combination of approaches will likely be needed to significantly improve the abiotic stress tolerance of crops in the field. These will include mechanistic understanding and subsequent utilization of stress response and stress acclimation networks, with careful attention to field growth conditions, extensive testing in the laboratory, greenhouse, and the field; the use of innovative approaches that take into consideration the genetic background and physiology of different crops; the use of enzymes and proteins from other organisms; and the integration of QTL mapping and other genetic and breeding tools.

Abstract:Crop improvement under changing climatic conditions is required to feed the growing global population. The development of transgenic crops is an attractive and conceivably the most effective approach for crop improvement with desired traits in varying climatic situations. Here, we describe a simple, efficient and robust in planta Agrobacterium-mediated genetic transformation method that can be used in most crops, including rice, wheat and cotton, and particularly in tissue culture recalcitrant crops, such as chickpea and pigeon pea. The protocol was successfully used for the development of transgenic chickpea and pigeon pea lines for resistance against pod borer. Transgenic lines in chickpea, pigeon pea and wheat were also developed for salt stress tolerance. These lines exhibited improved salt tolerance in terms of various physio-biochemical parameters studied. Since the protocol is rapid, as no tissue culture step is involved, it will significantly contribute to the improvement of most crops and will be of interest for plant biologists working with genetic engineering or genome editing.

Abstract:The recent rapid climate changes and increasing global population have led to an increased incidence of abiotic stress and decreased crop productivity. Environmental stresses, such as temperature, drought, nutrient deficiency, salinity, and heavy metal stresses, are major challenges for agriculture, and they lead to a significant reduction in crop growth and productivity. Abiotic stress is a very complex phenomenon, involving a variety of physiological and biochemical changes in plant cells. Plants exposed to abiotic stress exhibit enhanced levels of ROS (reactive oxygen species), which are highly reactive and toxic and affect the biosynthesis of chlorophyll, photosynthetic capacity, and carbohydrate, protein, lipid, and antioxidant enzyme activities. Transgenic breeding offers a suitable alternative to conventional breeding to achieve plant genetic improvements. Over the last two decades, genetic engineering/transgenic breeding techniques demonstrated remarkable developments in manipulations of the genes for the induction of desired characteristics into transgenic plants. Transgenic approaches provide us with access to identify the candidate genes, miRNAs, and transcription factors (TFs) that are involved in specific plant processes, thus enabling an integrated knowledge of the molecular and physiological mechanisms influencing the plant tolerance and productivity. The accuracy and precision of this phenomenon assures great success in the future of plant improvements. Hence, transgenic breeding has proven to be a promising tool for abiotic stress improvement in crops. This review focuses on the potential and successful applications, recent progress, and future perspectives of transgenic breeding for improving abiotic stress tolerance and productivity in plants.

Abstract:Background: The advancement in gene tagging and mapping in model crops such as Arabidopsis, rice andmaize has outpaced progress in major legume crops like groundnut, chickpea, redgram, greengram andsoybean, even in the post-genome release period of legume databases. To address this gap, a study investigated the transferability of gene tagged markers (GTMs) associated with rice yield, grain size andmicronutrient content (Fe and Zn) to groundnut. Methods: This study utilized 17 diverse groundnut genotypes for molecular analysis, targeting 45 GTMs spanning 24 known functional genes. Result: Remarkably, 31 out of the 45 markers (76%) demonstrated high transferability to groundnut, indicating significant conservation in functional regions across species. The amplification efficiency of rice GTMs varied among groundnut genotypes, ranging from 79.17% to 91.67%. Notably, 17 markers targeting 14 rice genes showed amplification across all 17 groundnut genotypes (100%) studied. These findings underscore the potential of leveraging knowledge from functionally characterized genes in molecular model crops and annotated orthologous genes in ‘Omics’ databases to enhance gene tagging efforts in crops like groundnut. By harnessing this approach, there is an opportunity to expand the molecular-level tagging of genes and improve crops like groundnut, which have seen limited progress in gene tagging. This strategy opens avenues for pyramiding desirable genes with precision, thereby enhancing crop traits with greater accuracy.

Abstract:Mungbean (Vigna radiata L. Wilczek) is an important short-duration grain legume of the genus Vigna that has wider adaptability across agro-climatic regions and soil types. Significant strides have been made towards the development of superior, high-yielding, and climate resilient cultivars in mungbean. A number of donors for various traits to have been deployed in introgression breeding. However, the use of common sources of resistance to different biotic stresses may lead to boom and bust cycles due to the appearance of new races or biotypes. Therefore, broadening the genetic base using wild and exotic plant genetic resources may offer a better quality of durable resistance. Many crop wild relatives (CWRs) confer a high degree of resistance against multiple diseases. Recently, several agronomically important genes have been mapped using inter-specific populations, which are being deployed for the improvement of mungbean. In such a situation, tagging, mapping, and exploiting genes of interest from cross-species donors for stress tolerance will offer novel genetic variations. This will also provide increased opportunities for the selection of desirable types. Advances in genomics and transcriptomics have further made it easy to tag the cross-compatible resistance loci and study their expression for delineating the mechanism of resistance. The comparative analysis of omics technology also helps in understanding the evolution and offers a scope for using cross-specific target genes for mungbean improvement. This review focuses on the effective utilization of cross-species cultivated and wild relatives as well as their omics resources for breeding multiple disease-resistant mungbean cultivars.

Abstract:Legumes have played an essential part as food and feed, but their productivity is inadequate to deliver the protein needs of a rapidly growing human population and livestock sectors. In numerous places around the world, legumes are the peculiar source of protein which essential for the human consumption. They are typically used as a supplement to other dietary proteins. As legumes have the richest protein source, the health experts and consumers demand increased rapidly. Simultaneously, the inhabitants of humans in the universe increased promptly. Meanwhile, fluctuation in biotic and also abiotic components is an extensive constraint for crop growth. It may provoke stress which results in the reduction of yield. However, the legume shows recalcitrant nature, these are the major root cause of the shortage of food. The biotechnology provides a solution to rectify all those issues and genetic transformation plays a consequential impact on enhancement of the legume productivity. This review clearly emphasizes the constraints of legume productivity, the strategy used, skill requirement and current advances in biotechnology for legume improvement.

Abstract:Advancements in gene transfer technology have indeed opened up exciting possibilities for more effectively manipulating the genetic makeup of live organisms, ranging from microorganisms to plants and animals. Direct and indirect transformations are the two basic types of gene transfer techniques. Indirect method comprises Agrobacterium mediated method as it involves intermediate host between gene of interest and target and this method is most opted one out of all present. Direct gene transformation methods, on the other hand, do not involve the use of an intermediate host organism. Instead, they rely on physical means to transfer genes between cells. Biolistic transformation uses high-velocity particles to deliver DNA into target cells, while microinjection and macroinjection involve the direct injection of DNA into cells. Protoplast fusion combines the genetic material of two different cells by fusing their protoplasts. Natural methods for gene transfer encompass mechanisms that occur naturally in various organism, includes transposition, conjugation, phage and retroviral transductions and bacterial transformation. Chemical techniques utilize chemical agents to facilitate gene transfer, such as calcium phosphate-mediated transformation, polyethylene glycol (PEG)-mediated transformation, DEAE (Diethylethanolamine)-Dextran-mediated transformation. Genes can be also being transferred using electrical techniques such as electroporation and electrofusion. Crop improvement and trait improvement are now being hastened by the fast-rising number of sequenced plant genomes, information from functional genomics data to understand gene function, innovative gene cloning, and tissue culture techniques. Despite being indispensable, its progress is still hindered by the fact that many plant species and agricultural genotypes exhibit low transformability or are resistant to established tissue culture and regeneration conditions. Here, we review the techniques employed in plant transformation and provide a concise overview of their evolution in agricultural crops, from their first inception to present time.
Sources of peanut bud necrosis disease resistance in groundnut with desirable yield parameters and pod features

Abstract:Groundnut (Arachis hypogea L.) is an important oilseed, food and fodder crop which is one of the five most important oil seed crops of the world. In India, groundnut is grown in an area of 5.47 million ha with a production of 5.51 million tones (2). Peanut bud necrosis disease (PBND) caused by peanut bud necrosis tospovirus (PBNV) vectored by Thrips palmi Karny (9) is a major disease of groundnut in South East Asia especially in India causing an yield loss of 89 million US dollars (1). The yield loss due to PBND in groundnut mainly depends on the time of infection. If the infection occurs on young plants (<60 days) pod yield loss will be 100% (6). On the basis of severity of PBNV incidence, a number of “hotspots” such as Mainpuri (Uttar Pradesh), Rajendranagar, Kadiri and Palem (Andra Pradesh), Latur (Maharashtra), Tikamgarh (Madhya Pradesh) and Raichur (Karnataka) have been identified (3). Host plant resistance is an economical as well as desirable component in the management of viral diseases. Since, PBND is destructive disease, identification of genotypes that canresist/tolerate the disease during early stages of crop growth are useful in minimizing yield loss due to the disease. Although, some varieties are identified as resistant/tolerant to PBND but most of them do not have the desirable pod features. Screening procedures to identify groundnut genotypes with resistance to PBND have relied mainly on natural field infection (4). Abstract:Salinity stress is a major concern in regions where irrigation relies on saline water. This study aimed to investigate the relative water content (RWC), electrolytic leakage (EL), total chlorophyll content, free amino acid content, and total soluble sugar content were analyzed in different groundnut species subjected to various salinity treatments. The results showed that salinity stress significantly reduced the RWC in groundnut leaves, with A. duranensis (wild type) exhibiting higher RWC values compared to the Arachis hypogaea species. RNA sequencing was performed to identify differentially expressed genes (DEGs) during salt stress. A total of 9079 DEGs were identified, with 1372 genes upregulated and 2509 genes downregulated. Genes belonging to transcription factor families, such as WRKY, MYB, bHLH, E2F, and Auxin efflux carrier proteins, were induced under salt stress in the tolerant genotype. Conversely, genes encoding NADH dehydrogenase, glutathione S-transferase, protein kinases, UDP-glycosyltransferase, and peroxidase were downregulated. Gene ontology and pathway analyses revealed several enriched categories and metabolic pathways associated with salt stress response, including catalytic activity, response to salt stress, ATP-dependent activity, and oxidative phosphorylation. The findings of this study provide insights into the physiological and molecular responses of groundnut to salinity stress. A. duranensis exhibited better salinity tolerance than Arachis hypogaea, as indicated by higher RWC values, lower electrolytic leakage, and differential gene expression patterns. These results contribute to our understanding of the mechanisms underlying salt stress tolerance in groundnut and may guide future efforts to develop salinity-tolerant groundnut species, ultimately improving crop yield in saline-affected regions.

Abstract:Mungbean ( Vignaradiata L. Wilczek) is an important grain legume widely cultivated in tropical and subtropical regions of the Indian subcontinent and in South East Asian countries. Protein and carbohydrate of Mungbean are easily digestible and create less flatulence than proteins derived from other legumes.Mungbean is very sensitive to salinity, drought, high and low temperature during the flowering and seed/pod development stages resulting in heavy losses to productivity. The development of genetically engineered plants by the introduction and/or over expression of selected abiotic stress tolerant genes seems to be a viable option for obtaining improved plants. Stable transformation and expression of transgene ( codA gene) was achieved in mungbean through Agrobacterium tumefaciens mediated system using cotyledonary node explants, under the optimized conditions. Molecular analysis of transgenic plants was done by using PCR, DOT-BLOT, ELISA and Western blotting. The primary transformants were checked for salt tolerance by the leaf disc test.
