Abstract:OBJECTIVE:To analyse the induction of apoptosis and reactive oxygen species (ROS) in human skin fibroblast by ultraviolet A (UVA). METHODS:Human skin fibroblasts were irradiated with UVA, apoptosis was determined with flowcytometry and fluorescent microscopy, the intracellular ROS production was measured with a spectrofluorometer, and the free radicals were determined with electron spin resonance. RESULTS:A significant increase of apoptosis was observed in human skin fibroblast after exposure to UVA with a dosage at 110-120 kJ.m-2, mannitol could counteract apoptosis; a higher level of intracellular ROS and the hydroxyl free radical was detected in the irradiated samples. CONCLUSION:A lower dose of irradiation of UVA(0.6 mW.cm-2) can induce apoptosis in human skin fibroblast, and ROS might play a role in the induction of apoptosis.
Activation of caspase- 3 during bacterial redox protein azurin -induced apoptosis in U2OS cells

Abstract:AIM: To study whether caspase-3,8 is activated during azurin-induced apoptosis in U2OS cells. METHODS: AnnexinV /PI method was used to detect apoptosis. The changes of procaspase-3 were analyzed by Western blot, the changes of caspase-3 mRNA were detected by semi-quantitative RT-PCR, and caspase-3 relative activity was determined by colorimetric assay. RESULTS: After U2OS cells were treated with 0, 25, 50, 100, 200, 500 mg/L azurin for 24 h, respectively, the level of procaspase-3 protein decreased and the level of caspase-3 mRNA increased as azurin concentration increased. When the cells were treated with 100 mg/L azurin for 6, 12, 24, 48 h , respectively, the caspase-3 activity began to rise from 6 h,reached the peak at 24 h,and was still higher than the control group at 48 h ( P 0 01). After the cells were treated with azurin at different concentrations for 24 h, caspase-3 activity increased in a concentration -dependent manner. CONCLUSION: Caspase-3 is activated and plays an important role in azurin-induced apoptosis in U2OS cells. [
THE EXPRESSION OF BAX AND BCL-2 IN RAT EPIDERMAL CELL INDUCED BY ULTRAVIOLET B

Abstract:[Objective] To investigate regulative effects of ultraviolet B(UVB) for normal apoptosis of epidermal cell in mice and apoptosis-related gene Bax and Bcl-2.[Methods] Normal ICR mice were divided into control group(sham irradiation group) and ultraviolet irradiation groups with dosage of 1 000 J /m2,1 500 J /m2,2 000 J /m2 for 2 hours,and 1 500 J /m2 for 4,8,12,24,48,72 hours groups with 10 mice each group.Protein expression of apoptosis-related gene Bax and Bcl-2 was detected by immunohistochemical method with streptavidin-peroxidase.[Results] Bax expression increased and Bcl-2 expression decreased in mouse skin after UVB irradiation with different dosages for 2 hours and both appeared dosage-dependant.Bax expression increased to peak value after UVB irradiation with 500 J /m2 dosage for 12 hours whereas Bcl-2 expression decreased.Bax expression began to decreased and Bcl-2 expression increased after 24 hours,and returned to normal value after 72 hours.Bax expression and Bcl-2 expression were related to time after UVB irradiation.[Conclusion] Bax expression increases and Bcl-2 expression decreases in mouse skin after UVB irradiation,so that it promotes apoptosis of epidermal cells injured by ultraviolet.
In Vitro Enhancement of Radiation-Induced Apoptosis in Human Hepatoma Cell Line HepG2 by a Selective Inhibitor of Cyclooxygenase-2 Enzyme NS -398

Abstract:AIM: To investigate the possible role of NS-398,a selective inhibitor of cyclooxygenase-2 enzyme,in radiation-induced apoptosis of human hepatoma cell line HepG2 in vitro.METHODS: Hepatoma cell line HepG2 was treated with various concentrations(25,50,100,200 μmol/L) of NS-398 before MTT assay was used to evaluate the cytotoxicity of NS-398.Transmission electron microscopy(TEM) was used to observe the changes of apoptosis in morphology.FCM was performed to quantify the apoptotic percentage.Real-time PCR was used to detect the expression of bcl-2,bax and caspase-3 mRNA,Western blotting was used to measure the expression of Bcl-2 and bax protein,and colorimetric method was provided to analyze the change of caspase-3 activity.RESULTS: The cytotoxicity of NS-398 increased in time-dependent and dose-dependent manners.NS-398 significantly enhanced radiation-induced apoptosis(P0.01),increased the expression of bax mRNA,Bax protein,caspase-3 mRNA and enhanced caspase-3 activity,whereas no significant change in Bcl-2 expression was found(P0.05).CONCLUSION: NS-398 enhances radiation-induced apoptosis in hepatoma cell line HepG2.The mechanism may be associated with the up-regulation of the expression of Bax,caspase-3 and enhancement of the activity of caspase-3,which ultimately induce apoptosis in HepG2.
[Selective Inducement Effect of Bacterial Redox Protein Azurin on Apoptosis of Human Osteosarcoma Cell Line U2OS].

Abstract:BACKGROUND & OBJECTIVE Bacterial redox protein azurin can selectively inhibit proliferation, and induce apoptosis in various human cancer cells. These in vitro findings have been confirmed in vivo in nude mice bearing tumor xenograft. Furthermore, azurin could diminish tumor volume in nude mice bearing tumor xenograft with no evidence of toxicity. The mechanism of its action is unknown. This study was designed to explore the effects of azurin on apoptosis of osteosarcoma cell line U2OS, and its molecular mechanism. METHODS U2OS cells were cultured with different concentrations of azurin. Cell viability was detected by MTT assay. The 50% inhibitory concentration (IC(50)) of azurin was calculated by Bliss method. Apoptotic morphology and apoptotic body of U2OS cells were observed under fluorescent microscope, and transmission electron microscope. DNA ladder was observed through agarose gel electrophoresis. Cell apoptosis was determined by flow cytometry (FCM). Protein levels of Bax, Bcl-2, and Caspase-3 were quantified by Western blot. RESULTS Azurin inhibited growth and induced apoptosis of U2OS cells in a dose-dependent manner 48 h after treatment, with the IC(50) of(114.54+/-7.65) mg/L. Moreover, its inhibitory effect was significantly higher on U2OS cells than on MG63 cells or L-02 cells under the same concentrations (P<0.05). When treated with 100 or 200 mg/L of azurin for 24 h, U2OS cells showed typical apoptotic morphology, typical DNA ladder bands were observed. No apoptotic feature was observed in control cells. Sub-G1 peak and apoptotic peak of U2OS cells were observed when exposed to 200 mg/L of azurin for 48 h with the apoptosis index of 35.8%, cell cycle was arrested in G1 phase. Bcl-2 was down-regulated when U2OS cells were treated with azurin for 24 h, while Bax and caspase-3 were up-regulated. CONCLUSIONS Azurin could selectively induce apoptosis of human osteosarcoma U2OS cells. The induction of apoptosis by azurin may be closely associated with down-regulation of Bcl-2, up-regulation of Bax, and activation of Caspase-3.

Abstract:In this study, we compared the cellular morphology over time under different culture conditions and the effect of UV irradiation on cells elucidated the morphological characteristics of UV induced apoptosis. It was shown that UV radiation could penetrate the culture medium to reach the irradiated cells and induce DNA mutations at a small dose. Culture medium might contribute to maintain physiological responses of adherent cell. After irradiation, cells responded differently under different culture conditions. Apoptosis in the medium was dose-dependent on UV, and morphological changes began with darkening of the nucleus and then the appearance of membrane blebs at the cell edges. When there was no medium, the organelles of irradiated cells migrated from the periphery of the nucleus to the edge of the cell, and irradiation of cells containing medium did not show migration of the organelles. In temperature experiments, the cell membrane of high temperature treated cells broke, and the cells maintained the physiological activity at low temperature for a period of time.

Abstract:Nitric oxide (NO) from (Z)-1-[N-(2-aminoethyl)-N-(2-ammonioethyl)amino]diazen-1- ium-1,2-diolate (NOC-18) induces apoptosis in human leukemia HL-60 cells. This effect was prevented by the pan-caspase inhibitor benzyloxycarbonyl-Val-Ala-Asp-fluoromethyl ketone (Z-VAD-FMK), thereby implicating caspase activity in the process. NOC-18 treatment resulted in the activation of several caspases including caspase-3, -6, -8, and -9(-like) activities and the degradation of several caspase substrates such as nuclear lamins and SP120 (hnRNP-U/SAF-A). Moreover, release of cytochrome c from mitochondria was also observed during NOC-18-induced apoptosis. This change was substantially prevented by Z-VAD-FMK, thereby suggesting that the released cytochrome c might function not only as an initiator but also as an amplifier of the caspase cascade. Bid, a death agonist member of the Bcl-2 family, was processed by caspases following exposure of cells to NOC-18, supporting the above notion. Thus, NO-mediated apoptosis in HL-60 cells involves a caspase/cytochrome c-dependent mechanism.

UVB induces apoptosis via downregulation of CALML3-dependent JNK1/2 and ERK1/2 pathways in cataract

Abstract:The aim of the current study was to understand the mechanisms of apoptosis occurring in cultured human lens epithelial cells (HLECs) following ultraviolet B (UVB) irradiation. The investigations intended to confirm the presence of apoptosis and to reveal the roles of oxidative stress, calcium (Ca2+), c‑Jun NH2‑terminal kinase (JNK)1/2, and extracellular signal‑regulated kinase (ERK)1/2 signaling pathway in these progresses. Cell apoptosis, ROS generation and intracellular Ca2+ concentration was measured by flow cytometry. The expression of CALML3, caspase-3, Bax, Bcl-2, p-JNK1/2, JNK1/2, p-ERK1/2 and ERK1/2 was measured by RT-qPCR and western blot analysis. Annexin V‑fluorescein isothiocyanate/propidium iodide staining demonstrated that UVB irradiation increased the apoptotic rate, reactive oxygen species (ROS) production and intracellular Ca2+ concentration of HLECs in dose‑ and time‑dependent manners. Overexpression of calmodulin like 3 (CALML3) reversed the effects of UVB irradiation on apoptosis, ROS production and Ca2+ concentration of HLECs, and decreased expressions of caspase‑3 and Bax, with increased expressions of Bcl‑2. Notably, silencing of CALML3 had similar effects to UVB irradiation and inhibited the activation JNK1/2 and ERK1/2 pathways. Nimodipine, a Ca2+‑channel antagonist, significantly attenuated the damages induced by CALML3 downregulation. In conclusion, UVB irradiation induced increase in apoptosis, ROS production and Ca2+ concentration of HLECs, in part, by downregulating the expression of CALML3 and involved oxidative stress, Ca2+, JNK1/2 and ERK1/2 signaling pathways, suggesting that investigating CALML3 may useful for developing cataract treatment.

Abstract:Cataract is the most common eye disease that causes blindness in patients. Ultraviolet B (UVB) irradiation is considered an important factor leading to cataract by inducing apoptosis in human lens epithelial cells (HLECs), but the mechanism is currently unclear. In this study, we investigated HLECs under different intensities of UVB irradiation and different exposure time. The annexin V-FITC/propidium iodide staining results showed that UVB irradiation could efficiently lead to HLECs apoptosis in time- and dose-dependent manner. The expression of pro-apoptotic Bax gene was promoted by UVB irradiation, while anti-apoptotic Bcl-2 gene expression was inhibited at both transcript and protein levels. Notably, the ratio of Bax/Bcl-2 displayed a high and positive correlation to the proportion of apoptotic HLECs. Mitochondrial dysfunction was also observed with rapid loss of potential (∆Ψ m), as well as changes of the levels of reactive oxygen species, malondialdehyde, total antioxidative capabilities, and superoxide dismutase. In caspase pathway, the level of caspase-3 protein increased after UVB irradiation. All these discovered changes may play important roles in UVB-induced HLECs apoptosis, and would be helpful in understanding the mechanism of UVB-induced cataract and providing potential prevention and treatment strategies.
Efficacy and Mechanism of Antibiotic Resistance Gene Degradation and Cell Membrane Damage during Ultraviolet Advanced Oxidation Processes

Abstract:Combinations of UV with oxidants can initiate advanced oxidation processes (AOPs) and enhance bacterial inactivation. However, the effectiveness and mechanisms of UV-AOPs in damaging nucleic acids (e.g., antibiotic resistance genes (ARGs)) and cell integrity represent a knowledge gap. This study comprehensively compared ARG degradation and cell membrane damage under three different UV-AOPs. The extracellular ARG (eARG) removal efficiency followed the order of UV/chlorine > UV/H2O2 > UV/peracetic acid (PAA). Hydroxyl radical (•OH) and reactive chlorine species (RCS) largely contributed to eARG removal, while organic radicals made a minor contribution. For intracellular ARGs (iARGs), UV/H2O2 did not remove better than UV alone due to the scavenging of •OH by cell components, whereas UV/PAA provided a modest synergism, likely due to diffusion of PAA into cells and intracellular •OH generation. Comparatively, UV/chlorine achieved significant synergistic iARG removal, suggesting the critical role of the RCS in resisting cellular scavenging and inactivating ARGs. Additionally, flow cytometry analysis demonstrated that membrane damage was mainly attributed to chlorine oxidation, while the impacts of radicals, H2O2, and PAA were negligible. These results provide mechanistic insights into bacterial inactivation and fate of ARGs during UV-AOPs, and shed light on the suitability of quantitative polymerase chain reaction (qPCR) and flow cytometry in assessing disinfection performance.
U(VI) exposure induces apoptosis and pyroptosis in RAW264.7 cells

Abstract:U(VI) pollution has already led to serious harm to the environment and human health with the increase of human activities. The viability of RAW264.7 cells was assessed under various U(VI) concentration stress for 24 and 48 h. The reactive oxygen species (ROS), mitochondrial membrane potential (MMP), and superoxide dismutase (SOD) activities of RAW264.7 cells under U(VI) stress were measured. The results showed that U(VI) decreased cell activity, induced intracellular ROS production, abnormal MMP, and increased SOD activity. The flow cytometry with Annexin-V/PI double labeling demonstrated that the rate of late apoptosis increased with the increase of U(VI) concentration, resulting in decreased Bcl-2 expression and increased Bax expression. The morphology of RAW264.7 cells dramatically changed after 48 h U(VI) exposure, including the evident bubble phenomenon. Besides, U(VI) also increased the proportion of LDH releases and increased GSDMD, and Ras, p38, JNK, and ERK1/2 protein expression, which indicated that the MAPK pathway was also involved. Therefore, U(VI) ultimately led to apoptosis and pyroptosis in RAW264.7 cells. This study offered convincing proof of U(VI) immunotoxicity and established the theoretical framework for further fundamental studies on U(VI) toxicity.

Fluorescence Resonance Energy Transfer Analysis of Bid Activation in Living Cells During Ultraviolet-Induced Apoptosis

Abstract:Ultraviolet (UV) irradiation is a DNA-damaging agent that triggers apoptosis through both the membrane death receptor and mitochondrial apoptotic signaling pathways. Bid, a pro-apoptotic Bcl-2 family member, is important in most cell types to apoptosis in response to DNA damage. In this study, a recombinant plasmid, YFP-Bid-CFP, comprised of yellow and cyan fluorescent protein and a full length Bid, was used as a fluorescence resonance energy transfer analysis (FRET) probe. Using the FRET technique based on YFP-Bid-CFP, we found that Bid activation was initiated at 9 +/- 1 h after UV irradiation, and the average duration of the activation was 75 +/- 10 min. Bid activation coincided with a collapse of the mitochondrial membrane potential with an average duration of 50 +/- 10 min. When cells were pretreated with Z-IETD-fmk (caspase-8 specific inhibitor) the process of Bid activation was completely inhibited, but the apoptosis was only partially affected. Z-DEVD-fmk (caspase-3 inhibitor) and Z-FA-fmk (non asp specific inhibitor) did not block Bid activation. Furthermore, the endogenous Bid activation with or without Z-IETD-fmk in response to UV irradiation was confirmed by Western blotting. In summary, using the FRET technique, we observed the dynamics of Bid activation during UV-induced apoptosis and found that it was a caspase-8 dependent event.
Apoptosis of EMT-6 Cells Caused by Low-power Low-frequency Ultrasound and Nanoparticles

Abstract:To observe the nanoparticles enhanced EMT-6 apoptosis by low-power low-frequency ultrasound,after loading some nanoparticles,EMT-6 cell suspension was exposed to low-power low-frequency ultrasound.Investigating the apoptosis effects through microscope images of adherent cells and counting the digestive cells before and after ultrasonic exposure.Results Low-power low- frequency ultrasound could significantly increase the apoptosis of EMT-6 cells under aid of the nanoparticles.With the increase of concentration of the carbon nano-tubes,improvement of the ultrasound intensity,or prolonging of the irradiation time of ultrasound,the effects of apoptosis would become even much evident.Nanoparticles can help enhance EMT-6 apoptosis,and thusserve as a potential cancer therapy.
A Study on the Apoptosis in HL-60 Cells Induced by Cyclosporine A

Abstract:AIM:To investigate if cyclosporine A(CsA) induces the apoptosis in HL-60 cells.METHODS:Apoptosis was measured by the morphology of apoptotic cells,the quantitation of DNA fragmentation with the diphenylamine reaction,the agarose-gel electrophoresis of DNA fragment,and flow cytometric analysis.RESULTS:When the cells were incubated with CsA 50 mg/L for 4h, the percentage of DNA fragmentation increased to (28.2±5 8)% compared to the control group (12.5±1 7)% without CsA ( P 0 01, n =10).Chromatin condensation,nuclear fragmentation and production of apoptotic bodies were seen in the cells treated with CsA utilizing light and electron microscopy.DNA electrophoresis of CsA-treated HL-60 cells showed nuclear fragmentation in oligonucleosomal fragments (DNA ladder).Flow cytometric analysis revealed that the percentage of apoptotic cells was 49.7% and 9.1% after the cells were incubated with or without CsA 50 mg/L respectively.Induction of DNA fragmentation by CsA was both dose-and-time dependent.CONCLUSION:CsA can induce apoptosis in HL-60 cells.
Mechanism of Rat Osteosarcoma Cell Apoptosis Induced by a Combination of Low-Intensity Ultrasound and 5-Aminolevulinic Acid in Vitro.

Abstract:We investigated the killing effect of low-intensity ultrasound combined with 5-aminolevulinic acid (5-ALA) on the rat osteosarcoma cell line UMR-106. Logarithmic-phase UMR-106 cells were divided into a control group, ultrasound group and 5-ALA group. The cell apoptotic rate, production of reactive oxygen species, and the change in mitochondrial membrane potential were analyzed by flow cytometry; ultrastructural changes were observed by transmission electron microscopy. Using low-intensity ultrasound at 1.0 MHz and 2.0 W/cm(2) plus 5-ALA at a concentration of 2 mM, the apoptotic rate of the sonodynamic therapy group was 27.2 +/- 3.4% which was significantly higher than that of the control group, ultrasound group, and 5-ALA group (P < 0.05). The production of reactive oxygen species was 32.6 +/- 2.2% and the decrease in mitochondrial membrane potential was 39.5 +/- 2.5%. The 33342 staining showed nuclear condensation and fragmentation in the ultrasound group and 5-ALA group. Structural changes in the cell membrane, mitochondria, Golgi apparatus, and other organelles observed by transmission electron microscopy included formation of apoptotic bodies. The killing effect of low-intensity ultrasound combined with 5-ALA on UMR-106 cells was significant. Cell apoptosis played a vital role in the killing effect, and the mitochondria pathway contributed to the apoptosis of UMR-106 cells.

Abstract:Zinc ions inhibit the morphological and DNA fragmentation features of apoptosis in a number of systems. HL-60 cells pretreated with zinc and exposed to UV irradiation maintained their normal morphology for up to 8 h, whereas non-zinc-treated cells underwent extensive apoptosis. Zinc pretreatment also inhibited both single and double-stranded DNA fragmentation, which is characteristic of apoptosis. The most effective zinc concentration that blocked apoptosis over short incubation periods (up to 8 h) was also the most toxic over extended time periods (> or = 12 h). The mechanism of cell death at these longer time periods was akin to necrosis, but occurred in the absence of any DNA fragmentation. The effects of the nuclease inhibitor aurintricarboxylic acid (ATA) was also examined on UV-induced apoptosis in HL-60 cells. ATA had no toxic effects over the concentration range tested, but also failed to prevent DNA fragmentation in whole cells. Further analysis showed that it effectively inhibited DNA fragmentation in isolated nuclei.
Far-Ultraviolet Light at 222 nm Affects Membrane Integrity in Monolayered DLD1 Colon Cancer Cells

Abstract:222 nm far-ultraviolet (F-UV) light has a bactericidal effect similar to deep-ultraviolet (D-UV) light of about a 260 nm wavelength. The cytotoxic effect of 222 nm F-UV has not been fully investigated. DLD-1 cells were cultured in a monolayer and irradiated with 222 nm F-UV or 254 nm D-UV. The cytotoxicity of the two different wavelengths of UV light was compared. Changes in cell morphology after F-UV irradiation were observed by time-lapse imaging. Differences in the staining images of DNA-binding agents Syto9 and propidium iodide (PI) and the amount of cyclobutane pyrimidine dimer (CPD) were examined after UV irradiation. F-UV was cytotoxic to the monolayer culture of DLD-1 cells in a radiant energy-dependent manner. When radiant energy was set to 30 mJ/cm2, F-UV and D-UV showed comparable cytotoxicity. DLD-1 cells began to expand immediately after 222 nm F-UV light irradiation, and many cells incorporated PI; in contrast, PI uptake was at a low level after D-UV irradiation. The amount of CPD, an indicator of DNA damage, was higher in cells irradiated with D-UV than in cells irradiated with F-UV. This study proved that D-UV induced apoptosis from DNA damage, whereas F-UV affected membrane integrity in monolayer cells.
Ultraviolet B radiation induces oxidative stress and apoptosis in human lens epithelium cells by activating NF-κB signaling to down-regulate sodium vitamin C transporter 2 (SVCT2) expression

Abstract:Ultraviolet B (UVB) exposure is reported to cause cataract formation by inducing excessive reactive oxygen species (ROS) and apoptosis in human lens epithelial cells (HLECs). Sodium-dependent Vitamin C transports-2 (SVCT2) is a ascorbic acid (AsA) transporter for that can protect cells and tissues from oxidative stress. Here, we focus on the functional characterization and mechanism analysis of SVCT2 in UVB-treated HLECs. The results showed a significant reduction of SVCT2 expression in HLECs treated with UVB. SVCT2 abated apoptosis and Bax expression and increased Bcl-2 expression. Moreover, SVCT2 decreased ROS accumulation and MDA level, but increased the activities of antioxidant enzymes (SOD and GSH-PX). NF-κB inhibitor (PDTC) alleviated ROS production and apoptosis, and promoted SVCT2 expression in UVB-treated HLECs. Additionally, ROS inhibitor (NAC) suppressed oxidative stress, apoptosis, and induced SVCT2 expression in UVB-treated HLECs, while these effects were significantly abated due to the activation of NF-κB signaling. Furthermore, SVCT2 facilitated 14 C-AsA absorption in UVB-treated HLECs. Together, our findings demonstrated that UVB exposure-induced ROS generation, which further activated NF-κB signaling to down-regulate SVCT2 expression in HLECs. Then, downregulated SVCT2 promoted ROS accumulation and induced apoptosis by decreasing AsA uptake. Our data reveal a novel NF-κB/SVCT2/AsA regulatory pathway and suggest the therapeutic potential of SVCT2 in UVB-induced cataract.
