Abstract:The swift and stable detection of pathogens in urine samples holds significant implications for the immediate clinical diagnosis and treatment of urinary tract infections (UTIs). In this study, we propose a detection strategy utilizing a hybrid substrate composed of graphene oxide (GO) and silver nanoparticles (Ag NPs) for the detection of pathogens subjected to amoxicillin-mediated (amo-mediated) treatment. This strategy employs dynamic surface-enhanced Raman spectroscopy (D-SERS) for stable and rapid detection, capturing signal variations induced by amo-mediated changes in pathogen morphology. During the 5 min D-SERS detection time window, stable SERS signals were detected for three types of pathogens and four types of pathogens were successfully distinguished using principal component analysis (PCA). In comparison to conventional nanosubstrates, the GO/Ag NP hybrid substrate exhibits outstanding stability and enhancement effects. This approach enables the dual detection of the pathogen cell structure and metabolites, facilitating specific identification of pathogens in the urinary tract, with a detection limit for Escherichia coli reaching 1 × 104 colony-forming units (CFU)/mL, meeting the clinical microbiology laboratory diagnostic standards for UTIs (105 CFU/mL). Testing of 188 clinically collected urine samples using this strategy yielded a sensitivity (SENS) of 86.4% and a specificity (SPC) of 89.7%. This introduces a novel method for diagnosing UTIs, offering broad applications in the field of clinical pathogen detection.

Abstract:Salmonella should be absence in pharmaceutical preparations and foods according to regulations in many countries. Up to now, rapidly detecting Salmonella at 1 CFU[10 g (mL) ](1) in pharmaceutical preparation or 1 CFU[25 g (mL) ](1) in food samples is still a challenge. Herein, we present an aptamer-based surface enhanced Raman spectroscopy (SERS) method for rapidly detecting Salmonella Enteritidis by using a handheld Raman instrument. The aptamer could specifically recognize S. Enteritidis, and 4-MBA self assembled on the surface of Au@Ag NPs was used as a Raman reporter molecule. The method was validated to be high specific with no interference from other five pathogenic bacteria. It could identify S. Enteritidis contaminant at similar to 1 CFU(10 g)(1) spiked level in a real sample (Wenxin granule, a botanical drug) after 6 h of enrichment. The detection time was much shorter than that of the methods (more than 54 similar to 96 h) in the standards of pharmaceutical preparations and foods. In addition, the method could quantitatively determinate S. Enteritidis with satisfactory results. The SERS peak intensities of 4-MBA at 1072 cm1 showed a good linear correlation (R2 = 0.9873) with the logarithms of S. Enteritidis concentrations ranging from 4.17 x102 to 1.39x107 CFUmL1. T-test result (P = 0.425) revealed that there was no significant difference between the determination results obtained by the SERS method and the plate counting method. Therefore, the study indicated that the method was practical and reliable, and it could be a promising alternative for the on-site detection of S. Enteritidis.(c) 2021 Elsevier B.V. Abstract:Sensitive and convenient sensing of urease and its inhibitors is exceptionally urgent in clinical diagnosis and new drug development. In this study, the gold nanoclusters (AuNCs) and hydroxyl double salt (HDS) were composited by a simple confinement effect to prepare highly fluorescent AuNCs@HDS composites to monitor urease and its drug inhibitors. HDS was used as a matrix to confine AuNCs (AuNCs@HDS), facilitating the emission intensity of AuNCs. However, acidic conditions (low pH) can disrupt the structure of HDS to break the confinement effect, and quench the fluorescence of AuNCs. Therefore, a sensing platform for pH-related enzyme urease detection was constructed based on the sensitive response of AuNCs@HDS to pH. This sensing platform had a linear response range of 0.5–22.5 U/L and a low limit of detection (LOD) of 0.19 U/L for urease. Moreover, this sensing platform was also applied to monitor urease inhibitors and urease in human saliva samples. Additionally, a portable hydrogel kit combined with a smartphone was developed for urease detection to achieve portable, low-cost, instrument-free, and on-site monitoring of urease.

Abstract:Surface-enhanced Raman scattering (SERS) technology was combined with nanotechnology to detect the Raman intensity of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in saliva and of three kinds of populations (healthy volunteers, patients with gingivitis, and patients with periodontitis). The Raman intensities of IL-1β and TNF-α in the saliva of the periodontitis group were significantly higher than those of the control and gingivitis groups (P = 0.00). The Raman intensities of the two inflammatory factors in the gingival crevicular fluid (GCF) of the periodontitis group were also significantly higher than those of the other two groups (P = 0.00). In the same group, the Raman intensities of IL-1β and TNF-α showed no significant difference between the saliva and GCF samples, respectively (P > 0.05). Combining SERS technology and nanotechnology can aid in detecting the Raman intensity of inflammatory factors using saliva and GCF more effectively than traditional methods.

Abstract:Early diagnosis and monitoring of SARS-CoV-2 virus is essential to control COVID-19 outbreak. In this study, we propose a promising surface enhanced Raman scattering (SERS)-based COVID-19 biosensor for ultrasensitive detection of SARS-CoV-2 virus in untreated saliva. The SERS-immune substrate was fabricated by a novel oil/water/oil (O/W/O) three-phase liquid-liquid interfaces self-assembly method, forming two layers of dense and uniform gold nanoparticle films to ensure the reproducibility and sensitivity of SERS immunoassay. The detection was performed by an immunoreaction between the SARS-CoV-2 spike antibody modified SERS-immune substrate, spike antigen protein and Raman reporter-labeled immuno-Ag nanoparticles. This SERS-based biosensor was able to detect the SARS-CoV-2 spike protein at concentrations of 0.77 fg mL-1 in phosphate-buffered saline and 6.07 fg mL-1 in untreated saliva. The designed SERS-based biosensor exhibited excellent specificity and sensitivity for SARS-CoV-2 virus without any sample pretreatment, providing a potential choice for the early diagnosis of COVID-19.

Abstract:Saliva could be an optimal sample for non-invasive cancer detection, as it contains plenty of proteins and metabolites which can reflect the health status of an individual. Moreover, pairing it with high-sensitivity, label-free detection techniques could prove successful for early cancer diagnosis. In this study, we explore the enhancement of salivary characteristic Raman bands by using label-free, ultrasensitive surface-enhanced Raman scattering (SERS) based on gold nanoparticles. SERS maps were acquired from dry samples of saliva supernatant mixed with Au colloidal nanoparticles, which was then pipetted on clean glass slides. The SERS spectra presented a high variability of signal intensities and frequency shifts. However, several reproducible SERS spectra showing well-resolved bands were obtained at certain locations on the maps, where Au nanoparticles clustered together during the air-drying. The healthy and oral cancer saliva could be differentiated using principal components analysis based on several SERS bands assigned mainly to amino acids and proteins. Moreover, thiocyanate Raman modes were detected in saliva samples of both smoking and non-smoking volunteers and cancer patients. The analysis indicated that the cancer group displayed an overall higher level of the 2126 cm−1 band area assigned to C–N stretching vibrations of thiocyanate.

Abstract:Abnormal levels of uric acid (UA) in urine serve as warning signs for gout and metabolic cardiovascular diseases, necessitating the monitoring of UA levels for early prevention. However, the current analytical methods employed suffer from limitations in terms of inadequate suitability for home-based applications and the requirement of non-invasive procedures. In this approach, creatinine, a metabolite with a constant excretion rate, was incorporated as an endogenous internal standard (e-IS) for calibration, presenting a rapid, pretreatment-free, and accurate strategy for quantitative determination of UA concentrations. By utilizing urine creatinine as an internal reference value to calibrate the signal fluctuation of surface-enhanced Raman spectroscopy (SERS) of UA, the quantitative accuracy can be significantly improved without the need for an external internal standard. Due to the influence of the medium, UA, which carries a negative charge, is selectively adsorbed by Au@Ag nanoparticles functionalized with hexadecyltrimethylammonium chloride (CTAC) in this system. Furthermore, a highly convenient detection method was developed, which eliminates the need for pre-processing and minimizes matrix interference by simple dilution. The method was applied to the urine detection of different volunteers, and the results were highly consistent with those obtained using the UA colorimetric kit (UACK). The detection time of SERS was only 30 s, which is 50 times faster than UACK. This quantitative strategy of using urinary creatinine as an internal standard to correct the SERS intensity of uric acid is also expected to be extended to the quantitative detection needs of other biomarkers in urine.

Abstract:Due to the high sensitivity, specificity, efficiency and cost-effectiveness, surface-enhanced Raman spectroscopy (SERS) has been extensively used for the detection and evaluation of biological substances in body fluids, such as blood, tissues and cells. However, collecting these bodily species is not very convenient because patients have to suffer from skin irritation pain and contacted injection risks. Urine and saliva, which carry abundant metabolic information, can also reflect the health conditions about the human body and seems to be more practicable and convenient for truly non-invasive disease diagnosis. In particular, a number of cancers, diseases and illicit drugs have been successfully recognized and analyzed based on the SERS spectra of urine and saliva, enabling a screening of certain diseases and a tracing of disease development. In this review, we will introduce the state-of-the-art regarding the clinical applications of SERS analysis on urine and saliva, and briefly discuss the future development of SERS based non-invasive disease diagnosis.

Abstract:Surface Enhanced Raman Spectroscopy (SERS) technique is a fast, convenient and nondestructive optical diagnose method that can provide specific fingerprints of biomolecular compositions and structures. Human saliva contains many biomarkers that can reflect health condition. In this paper, we employed SERS methods based on nanometer silver colloid to obtain high quality SERS of 208 saliva samples from 46 patients with nasopharyngeal carcinoma (NPC), 30 patients with rhinitis and 48 healthy volunteers. The results showed there were great differences in SERS pattern among these three subjects. Principal component analysis-linear discriminant analysis (PCA-LDA) was utilized to analyze and classify the saliva spectra from NPC, rhinitis and healthy subjects. The diagnostic sensitivity was 89. 5%, 93. 0% and 100% respectively, and meanwhile the specificity was 94. 6%, 96. 7% and 97. 7% respectively. The research showed that saliva SERS combined with PCA-LDA can rapidly and effectively distinguish three saliva groups, and this technique has great potential to be a non-destructive and clinical screening method for nasopharyngeal carcinoma detection.

Abstract:Surface-enhanced Raman scattering (SERS) is a flexible tool for biological analysis due to its excellent properties for detecting wide varieties of target biomolecules including pathogenic bacteria. In the present work, a highly selective and sensitive SERS-based aptasensor for the detection of Vibrio parahaemolyticus has been developed. An effective SERS substrate was designed and fabricated by synthesis of SiO2@Au core/shell nanoparticles (NPs). To selectively detect V parahaemolyticus, we employed aptamer 1 (apt 1) immobilized on the surface of SiO2@Au core shell NPs. The Raman signal intensity,is from a cyanine dye 3 (Cy3) modified aptamer 2 (apt 2), which binds to the target the same as SiO2@Au-apt1. In the presence of the target, SiO2@Au-apt 1-target-apt 2-Cy3 sandwich-like complexes form due to the high affinity and specificity of the aptamer for V. parahaemolyticus. Under optimal conditions, the correlation between the SERS signal and the V. parahaemolyticus concentration is linear within a range of 10-10(6) cfu mL(-1), with a limit of detection of 10 cfu mL(-1). Novel Portable Sensing System with Integrated Multifunctionality for Accurate Detection of Salivary Uric Acid

Abstract:Uric acid, as the terminal product of purine metabolism in the body, is an important marker of many diseases. Uric acid is abundant in saliva, offering the possibility of its non-invasive detection. However, it is sensitive to interference in saliva by a variety of factors. A reliable method of processing saliva is centrifugation (CF), but the cost and size of equipment limit its use in everyday life. In this study, a novel portable salivary-sensing system (PSSS) with integrated suction filtration (SF) and temperature insulation was proposed to obtain more accurate salivary uric acid levels through a simple procedure. The PSSS includes a saliva container, a high-sensitive uric acid sensor (UAS), an accompanying printed circuit board (PCB), and a mobile application. The responses produced by the UAS presents excellent linearity (4.6 μA/mM with R2 = 0.9964), selectivity, reproducibility, and stability for the detection of low levels of uric acid. The difference in detection values between the UAS and the commercial sensor is only ~4%. The primary feature of the saliva container is the processing of saliva by SF instead of CF. Samples from CF and SF showed no significant differences regarding uric acid levels, and both exhibited approximately 50% deviation from the untreated samples, while the difference in uric acid levels between the samples after SF and after applying both treatments was ~10%. Besides, insulation of the saliva container can partially eliminate sources of error induced by the environment during uric acid level testing. The PSSS provides a novel strategy for the immediate detection of specific markers in saliva. We believe that the PSSS has promising potential for future application in the rapid saliva testing.
An enzymatic reaction-based SERS saliva analysis microporous array chip for chiral differentiation and high-throughput detection of D-amino acids

Abstract:A Raman-active boronate modified surface-enhanced Raman scattering (SERS) microporous array chip based on the enzymatic reaction was constructed for reliable, sensitive, and quantitative monitoring of D-Proline (D-Pro) and D-Alanine (D-Ala) in saliva. Initially, 3-mercaptophenylboronic acid (3-MPBA) was bonded to Au-coated Si nanocrown arrays (Au/SiNCA) via Au-S bonding. Following this, H2O2 obtained from D-amino acid oxidase (DAAO)-specific catalyzed D-amino acids (D-AAs) further reduced 3-MPBA to 3-hydroxythiophenol (3-HTP) with a new Raman peak at 882 cm-1. Meanwhile, the original characteristic peak at 998 cm-1 remained unchanged. Therefore, the I882/I998 ratio increased with increasing content of D-AAs in the sample to be tested, allowing D-AAs to be quantitatively detected. The Au/SiNCA with large-area periodic crown structure prepared provided numerous, uniform "hot spots," and the microporous array chip with 16 detection units was employed as the platform for SERS analysis, realizing high-throughput, high sensitivity, high specificity and high-reliability quantitative detection of D-AAs (D-Pro and D-Ala). The limits of detection (LOD) were down to 10.1 µM and 13.7 µM throughout the linear range of 20-500 µM. The good results of the saliva detection suggested that this SERS sensor could rapidly differentiate between early-stage gastric cancer patients and healthy individuals.

Abstract:The absorption spectroscopy of silver colloids and the mixture of silver colloids with urine were measured. The urine' s Raman spectroscopy were tested and analyzed before and after the adding of silver colloids.Based on the technology of Surface Enhanced Raman Spectroscopy,the intensity of the urine's Raman signal in the silver colloids was increased obviously and the SERS signal of uric acid in urine was successfully detected.The characteristic Raman bands of urine were tentatively assigned.The SERS of the urine of the morning and the night were compared and analyzed.The detection of morning urine bears higher credibility and SNR.The results showed that the surface-enhanced Raman spectroscopy might provide a new kind of method for the clinic detection of urine.

Abstract:For the progress of point-of-care medicine, where individual health status can be easily and quickly monitored using a handheld sensor, saliva serves as one of the best-suited body fluids thanks to its availability and abundance of physiological indicators. Salivary biomarkers, combined with rapid and highly sensitive detection tools, may pave the way to new real-time health monitoring and personalized preventative therapy branches using saliva as a target matrix. Saliva is increasing in importance in liquid biopsy, a non-invasive approach that helps physicians diagnose and characterize specific diseases in patients. Here, we propose a proof-of-concept study combining the unique specificity in biomolecular recognition provided by surface-enhanced Raman spectroscopy (SERS) in combination with molecular dynamics (MD) simulations, which give leave to explore the biomolecular absorption mechanism on nanoparticle surfaces, in order to verify the traceability of two validated salivary indicators, i.e., interleukin-8 (IL-8) and lysozyme (LYZ), implicated in oropharyngeal squamous cell carcinoma (OSCC) and oral infection. This strategy simultaneously assures the detection and interpretation of protein biomarkers in saliva, ultimately opening a new route for the evolution of fast and accurate point-of-care SERS-based sensors of interest in precision medicine diagnostics.

Abstract:A highly structured SERS biosensor was proposed for Staphylococcus aureus ( S. aureus , ATCC 29213) detection. Polydimethylsiloxane (PDMS) film was chemically modified and gold and silver core–shell nanoparticles (Au@Ag NPs) were self-assembled on it. Sulfhydryl-modified aptamer of S. aureus was immobilized on the SERS substrate to form a capture substrate and the aptamer modified with 6-carboxy-x-rhodamine (ROX) was used as a signal molecular probe. The aptamer and target constructed a sandwich structure "capturing substrate-target-signal probe". Concurrently, the bacterial surface was covered with Au@Ag NPs which resulted in more "hot spots" and improved the detection sensitivity. The SERS intensity was plotted against the logarithmic concentration of S. aureus (3.6 × 10 to 3.6 × 10 8 cfu/mL) and achieved an ultra-low detection response of 16 cfu/mL. S. aureus was detected in tap water, milk and fish samples with satisfactory recoveries (93.25%∼106.42%) and validation results ( p > 0.05). Significantly, the method offered a speedy and accurate S. aureus examining in food/environmental settings.

Abstract:A ratiometric SERS aptasensor based on catalytic hairpin self-assembly (CHA) mediated cyclic signal amplification strategy was developed for the rapid and reliable determination of Escherichia coli O157:H7. The recognition probe was synthesized by modifying magnetic beads with blocked aptamers, and the SERS probe was constructed by functionalizing gold nanoparticles (Au NPs) with hairpin structured DNA and 4-mercaptobenzonitrile (4-MBN). The recognition probe captured E. coli O157:H7 specifically and released the blocker DNA, which activated the CHA reaction on the SERS probe and turned on the SERS signal of 6-carboxyl-x-rhodamine (ROX). Meanwhile, 4-MBN was used as an internal reference to calibrate the matrix interference. Thus, sensitive and reliable determination and quantification of E. coli O157:H7 was established using the ratio of the SERS signal intensities of ROX to 4-MBN. This aptasensor enabled detection of 2.44 × 102 CFU/mL of E. coli O157:H7 in approximately 3 h without pre-culture and DNA extraction. In addition, good reliability and excellent reproducibility were observed for the determination of E. coli O157:H7 in spiked water and milk samples. This study offered a new solution for the design of rapid, sensitive, and reliable SERS aptasensors.

Abstract:A method for saliva analysis combining membrane protein purification with silver nanoparticle-based surface-enhanced Raman spectroscopy (SERS) for non-invasive nasopharyngeal cancer detection was present in this paper. In this method, cellulose acetate membrane was used to obtain purified whole proteins from human saliva while removing other native saliva constituents and exogenous substances. The purified proteins were mixed with silver nanoparticle for SERS analysis. A diagnostic accuracy of 90.2% can be achieved by principal components analysis combined with linear discriminate analysis, for saliva samples obtained from patients with nasopharyngeal cancer (n = 62) and healthy volunteers (n = 30). This exploratory study demonstrated the potential for developing non-invasive, rapid saliva SERS analysis for nasopharyngeal cancer detection.

Abstract:The common quantification methods of purines, including high-performance liquid chromatography, micellar electrokinetic chromatography, reverse phase ion pair chromatography, and capillary zone electrophoresis, have been used for the analysis of purines. These conventional techniques are limited by long operation times, large sample volumes, requirements for pretreatment and separation processes, or sophisticated, bulky and expensive equipment. Here, we have developed a simple method to prepare a probe for the quantitative analysis of purines using the surface-enhanced Raman scattering (SERS) method. The SERS probe is a rough Ag wire obtained with simple electrochemical treatment, which is optimized to reach the maximum SERS signal enhancement. The Raman enhancement factor (EF) of Ag wire after roughening can reach ∼106. Under optimized conditions, the rough Ag wire is capable of recognizing four purine bases (namely, guanine, adenine, hypoxanthine, and xanthine) in the range of 10−5 to 10−1 mg/mL with a linear relationship between SERS intensity of signature peak and purine concentration. The recovery rate of each purine base is higher than 80% in the mixture of four purines. The overall method was successfully applied to the monitoring of the level of purines in marketplace beer beverages. The novel wire-based SERS sensor used with portable Raman spectroscopy is prospective in diet safety and personalized point-of-care medical detection.

Abstract:Rapid identification and diagnosis of bacteria and other microorganisms is a great challenge for drinking water safety due to the increasing frequency of pathogenic infections. Raman spectroscopy is a non-destructive tool to characterize the biochemical fingerprints of bacterial cells and its signal can be improved by surface-enhanced Raman scattering (SERS). Thus, Raman scattering has a huge potential in fast diagnosis of pathogens in drinking water, with low cost and high reproducibility. In this work, we developed a novel fast diagnosis method to detect aquatic pathogens via magnetic SERS assay. With chemical co-precipitation synthesis and surface glucose reduction, the silver-coated magnetic nanoparticles (Ag@MNPs) had a well-developed core-shell structure and high efficiency to capture bacterial cells. Ag@MNPs achieved 103 enhancement factor for rhodamine 6G and the limit of detection was 10-9 M. The magnetic SERS assay also successfully detected various bacteria (A. baylyi and E. coli) with high sensitivity (105 CFU/mL). This platform provided a promising and easy-operation approach for pathogen detection for food and drinking water safety.
