Abstract:Our initial in vitro (HL-60 cells) and in vivo (B16-F10 murine) studies showed nanosecond pulsed electric fields (nsPEFs) caused intracellular changes and melanoma involution, respectively. We wanted to describe the morphologic changes in cell ultrastructure and investigate the mechanism for change due to nsPEFs in B16-F10 melanoma tumors in SKH-1 mice. We injected B16-F10 cells into 120 female SKH-1 mice to derive melanoma tumors. After multiple nsPEF treatments (40 kV/cm field strength; 5 ns rise time; 300 ns duration), morphologic changes of melanoma size, shape, tumor nesting pattern, blood vessel structure and cell ultrastructure were observed. Nuclear changes were recorded with light-and transmission electron-microscopy (TEM). Apoptosis in situ was detected by several immunohistochemistry (IHC) methods: (1) terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling (TUNEL); (2) caspase 3, 6 and 7; and (3) histone H2AX. After multiple nsPEF treatments the melanoma blood supply was disrupted and melanomas shrank an average of 90% within two weeks. TEM showed increased rough endoplasmic reticulum and condensed dark-staining nucleoli; TUNEL presented fluorescent increase. Together these suggest post-treatment apoptotic activity, which was concurrently allied with increased caspase 3, 6 and 7, and histone H2AX fluorescence (DNA damage) after pulsing. Our study shows that blood flow and apoptotic changes play a central role in the biological effects caused by nsPEFs. Therefore nsPEFs may have application potential in cancer therapy, gene regulation and biophysical research by non-invasively disrupting intracellular components and inducing apoptosis in malignant tumors.

Abstract:Objective : Nanosecond pulsed electric fields (nsPEFs) can affect the intracellular structures of cells in vitro. Abstract:A recent study has shown that nanosecond pulsed electric fields (nsPEF) can affect the intracellular structures of melanoma within weeks. nsPEF is a non-drug, non-thermal treatment using ultrashort, intense pulsed electric fields with nanosecond durations. In the current study we followed up melanoma histopathology and metastasis with tissue micro-array 5 months post-nsPEF. After nsPEF treatment, tumor growth, tumor histology, metastasis, peri-tumor vessel and micro-vessel density were examined for the effect of nsPEF treatment on melanoma in vivo. The 17 nsPEF-treated mice were tumor-free for 169 days, significantly longer than those 19 control mice bearing melanoma without nsPEF. Histopathology follow-up showed that melanoma did not recur to the primary injection place after complete elimination. Compared with the control tumor, nsPEF-treated tumors present decreased micro-vessel density in a time-course manner in this survival study. Treatment with nsPEF caused continuous histopathological changes in melanomas, eliminated melanoma without recurrence at the primary site and prolonged animal survival time by inhibiting tumor blood supply and leading to tumor infarction. Thus, nsPEF could be applied in a non-ionizing therapeutic approach, without other agents, to locally treat tumors within a defined boundary.

Abstract:Microsecond pulsed electric fields (PEF) have previously been used for various tumour therapies, such as gene therapy, electrochemotherapy and irreversible electroporation (IRE), due to its demonstrated ability. However, recently nanosecond pulsed electric fields (nsPEF) have also been used as a potential tumor therapy via inducing cell apoptosis or immunogenic cell death to prevent recurrence and metastasis by interacting with intracellular organelles. A large proportion of the existing in-vitro studies of nsPEF on cells also suggests cell necrosis and swelling/blebbing can be induced, but the replicability and potential for other effects on cells suggesting a complicated process which requires further investigation. Therefore, this study investigated the effects of pulse width and intensity of nsPEF on the murine melanoma cells (B16) and normal murine fibroblast cells (L929) through electromagnetic simulation and in-vitro experiments. Through examining the evolution patterns of potential difference and electric fields on the intracellular compartments, the simulation has shown a differential effect of nsPEF on normal and cancerous skin cells, which explains well the results observed in the reported experiments. In addition, the modelling has provided a clear evidence that a few hundreds of ns PEF may have caused a mixed mode of effects, i.e. a 'cocktail effect', including cell electroporation and IRE due to an over their threshold voltage induced on the plasma membrane, as well as cell apoptosis and other biological effects caused by its interaction with the intracellular compartments. The in-vitro experiments in the pulse range of the hundreds of nanoseconds showed a possible differential cytotoxicity threshold of electric field intensity between B16 cells and L929 cells.
Antitumor Effect and Immune Response of Nanosecond Pulsed Electric Fields in Pancreatic Cancer

Abstract:Nanosecond pulsed electric fields (nsPEFs) have emerged as a novel and effective strategy for the non-surgical and minimally invasive removal of tumors. However, the effects of nsPEFs treatment on the tumor immune microenvironment remain unknown. In this study, the changes in the morphology and function of pancreatic cancer cells after nsPEFs were assessed and the modifications in the immune profile in pancreatic cancer models were investigated. To this end, electrodes were inserted with different parameters applied to ablate the targeted tumor tissues. Tumor development was found to be inhibited, with decreased volumes post-nsPEFs treatment compared with control tumors (P < 0.05). Hematoxylin and eosin staining showed morphological changes in pancreatic cancer cells, Ki-67 staining confirmed the effects of nsPEFs on tumor growth, and caspase-3 staining indicated that nsPEFs caused apoptosis in the early stages after treatment. Three days after nsPEFs, positron emission tomography demonstrated little residual metabolic activity compared with the control group. Gene expression profiling identified significant changes in immune-related pathways. After treatment with nsPEFs, CD8+ T lymphocytes increased. We showed that nsPEFs led to a significant decrease in immune suppressive cells, including myeloid derived suppressor cells, T regulatory cells, and tumor-associated macrophages. In addition, the levels of TNF-α and IL-1β increased (P < 0.05), while the level of IL-6 was decreased (P < 0.05). NsPEFs alleviated the immunosuppressive components in pancreatic cancer stroma, including hyaluronic acid and fibroblast activation protein-α. Our data demonstrate that tumor growth can be effectively inhibited by nsPEFs in vivo. NsPEFs significantly altered the infiltration of immune cells and triggered immune response.
Nanosecond Pulsed Electric Fields (nspefs) Inhibit Melanoma Angiogenesis

Abstract:In our initial study, a major change found in the melanomas treated by nsPEFs was an immediate reduction in blood flow to the tumor and up to 90% tumor shrinkage within two weeks following a cumulative PEF exposure time of 120 mus. The reduction of existing vessels and the lack of new blood vessel formation into and within the melanoma tumors stimulated the present research on angiogenesis. We investigated the vascular formation and the expression of angiogenic growth factors in melanomas treated by multiple nsPEFs: 40 kV/cm; 5 ns rise time; 300 ns duration to identify mechanisms inducing tumor involution. Samples of B16-F10 melanoma cells injected into 66 female SKH-1 mice were analyzed by tissue microarray technology. Tumor blood vessels were studied by confocal microscopy with double fluorescent staining for endothelial cells (CD31) and pericytes (smooth muscle actin). Intra-tumoral microvessel density (IMD) was assessed by immunohistochemistry with CD31 (marks large vessels), CD34 (medium vessels) and CD105 (microvessels). Vascular endothelial growth factor (VEGF) and platelet-derived endothelial cell growth factor (PD-ECGF) expressions were also determined by Western-blot to show change in up-stream control factors. Double fluorescence staining showed decreased tumor blood network formation. IMD density assessed by CD31, CD34 and CD105 had significantly lower expression in treated compared to control groups (Table 1, p<0.05). Nanosecond PEFs down-regulated protein expression of VEGF and PD-ECGF. Expression of VEGF and PD-ECGF statistically different between treated and control groups. We propose nsPEFs have relatively rapid direct effects and long-term effects to inhibit expression of VEGF and PD-ECGF thus breaking the pro-angiogenic and anti-angiogenic balance. Angiogenic inhibition restrains vessel formation and as a result can cause melanomas to involute.

Abstract:Nanosecond pulsed electric field (nsPEF) has emerged as a promising approach for inducing cell death in melanoma, either as a standalone treatment or in combination with chemotherapeutics. However, to date, there has been a shortage of studies exploring the impact of nsPEF on the expression of cancer-specific molecules. In this investigation, we sought to assess the effects of nsPEF on melanoma-specific MAGE (Melanoma Antigen Gene Protein Family) expression. To achieve this, melanoma cells were exposed to nsPEF with parameters set at 8 kV/cm, 200 ns duration, 100 pulses, and a frequency of 10 kHz. We also aimed to comprehensively describe the consequences of this electric field on melanoma cells' invasion and proliferation potential. Our findings reveal that following exposure to nsPEF, melanoma cells release microvesicles containing MAGE antigens, leading to a simultaneous increase in the expression and mRNA content of membrane-associated antigens such as MAGE-A1. Notably, we observed an unexpected increase in the expression of PD-1 as well. While we did not observe significant differences in the cells' proliferation or invasion potential, a remarkable alteration in the cells' metabolomic and lipidomic profiles towards a less aggressive phenotype was evident. Furthermore, we validated these results using ex vivo tissue cultures and 3D melanoma culture models. Our study demonstrates that nsPEF can elevate the expression of membrane-associated proteins, including melanoma-specific antigens. The mechanism underlying the overexpression of MAGE antigens involves the initial release of microvesicles containing MAGE antigens, followed by a gradual increase in mRNA levels, ultimately resulting in elevated expression of MAGE antigens post-experiment. These findings shed light on a novel method for modulating cancer cells to overexpress cancer-specific molecules, thereby potentially enhancing their sensitivity to targeted anticancer therapy.

Abstract:Purpose: The application of nanosecond pulsed electric fields (nsPEFs) could be an effective therapeutic strategy for peritoneal metastasis (PM) from colorectal cancer (CRC). The aim of this study was to evaluate in vitro the sensitivity of CT-26 CRC cells to nsPEFs in combination with chemotherapeutic agents, and to observe the subsequent in vivo histologic response. Methods: In vitro cellular assays were performed to assess the effects of exposure to 1, 10, 100, 500 and 1000 10 ns pulses in a cuvette or bi-electrode system at 10 and 200 Hz. nsPEF treatment was applied alone or in combination with oxaliplatin and mitomycin. Cell death was detected by flow cytometry, and permeabilization and intracellular calcium levels by fluorescent confocal microscopy after treatment. A mouse model of PM was used to investigate the effects of in vivo exposure to pulses delivered using a bi-electrode system; morphological changes in mitochondria were assessed by electron microscopy. Fibrosis was measured by multiphoton microscopy, while the histological response (HR; hematoxylin-eosin-safran stain), proliferation (KI67, DAPI), and expression of immunological factors (CD3, CD4, CD8) were evaluated by classic histology. Results: 10 ns PEFs exerted a dose-dependent effect on CT-26 cells in vitro and in vivo, by inducing cell death and altering mitochondrial morphology after plasma membrane permeabilization. In vivo results indicated a specific CD8+ T cell immune response, together with a strong HR according to the Peritoneal Regression Grading Score (PRGS). Conclusions: The effects of nsPEFs on CT-26 were confirmed in a mouse model of CRC with PM.
Modulation of radiochemoimmunotherapy-induced B16 melanoma cell death by the pan-caspase inhibitor zVAD-fmk induces anti-tumor immunity in a HMGB1-, nucleotide- and T-cell-dependent manner

Abstract:One prerequisite that radiotherapy (RT) and chemotherapy (CT) result in anti-tumor immune responses is triggering of immunogenic cell death forms such as necroptosis. The latter is inducible by inhibition of apoptosis with the pan-caspase inhibitor zVAD-fmk. The design of multimodal therapies that overcome melanoma's resistance to apoptosis is a big challenge of oncoimmunology. As hints exist that immune stimulation by hyperthermia (HT) augments the efficacy of melanoma therapies and that tumors can be sensitized for RT with zVAD-fmk, we asked whether combinations of RT with dacarbazine (DTIC) and/or HT induce immunogenic melanoma cell death and how this is especially influenced by zVAD-fmk. Necroptosis was inducible in poorly immunogenic B16-F10 melanoma cells and zVAD-fmk generally increased melanoma cell necrosis concomitantly with the release of HMGB1. Supernatants (SNs) of melanoma cells whose cell death was modulated with zVAD-fmk induced an upregulation of the activation markers CD86 and MHCII on macrophages. The same was seen on dendritic cells (DCs), but only when zVAD-fmk was added to multimodal tumor treatments including DTIC. DCs of MyD88 KO mice and DCs incubated with SNs containing apyrase did not increase the expression of these activation markers on their surface. The in vivo experiments revealed that zVAD-fmk decreases the tumor growth significantly and results in a significantly reduced tumor infiltration of Tregs when added to multimodal treatment of the tumor with RT, DTIC and HT. Further, a significantly increased DC and CD8+ T-cell infiltration into the tumor and in the draining lymph nodes was induced, as well as an increased expression of IFNγ by CD8+ T cells. However, zVAD-fmk did not further reduce tumor growth in MyD88 KO mice, mice treated with apyrase or RAG KO mice. We conclude that HMGB1, nucleotides and CD8+ T cells mediate zVAD-fmk induced anti-melanoma immune reactions in multimodal therapy settings.
Effect of Nanosecond Pulsed Electric Fields in Combination with Everolimus on Melanoma.

Abstract:e20102 Background: Nanosecond pulsed electric field (nsPEF) is a bioelectrical technology generated high voltage electric field in nanosecond duration to destroy tumor cells with non-thermal effect. It is a potential technique for cancer therapy. PI3K/mTOR/AKT pathway is activated in most melanomas, but single agent activity of mTOR inhibitors is limited in advanced melanomas. The aim of the study is to evaluate the anti-growth efficacy of nsPEF in combination with everolimus in melanoma. Methods: Human A375 melanoma cells were exposed to everolimus for 48 hours, followed by 25 pulses of nsPEF with 100 ns duration at the electric field strength of 15 and 20 kV/cm. Cell viability was measured by CellTilter-Glo luminescent cell viability assay 24 hours later. Cell apoptosis and cell cycle were analyzed by flow cytometry. BALB/c mice were inoculated subcutaneously with A375 cells, and randomly divided into four groups: group I as control, group II treated with nsPEF in a single treatment (30 kV/cm, 100 ns, 100 pulses), group III given everolimus (4 mg/kg) by oral gavage daily for 2 weeks, and group IV treated with nsPEF plus everolimus. Results: With the combination of nsPEF, cell growth was decreased gradually according to the nsPEF intensity. nsPEF could induce melanoma cell apoptosis, and the ratio of apoptotic cells was correlated with nsPEF intensity. Cell cycle analysis showed that both everolimus and nsPEF alone could induce G2/M phase arrest, and combination of everolimus with nsPEF could further increase the percentages of G2/M-arrested cells. In vivo experiments showed that the skin lesions caused by nsPEF all healed in one week, and no adverse effect were observed. When compared to control group, the everolimus group, the nsPEF group and the combination group suppressed tumor growth to 32%, 44% and 10%, respectively. Conclusions: nsPEF and everolimus have synergistic anti-tumor effect on melanoma cells through inducing G2/M phase arrest and apoptosis. Our findings indicate that nsPEF in combination with mTOR inhibitor could be used as a potential treatment for advanced melanoma.

Abstract:As a novel therapeutic tool, nanosecond pulsed electric field（nsPEF） has shown promising applications in melanoma treatment. This paper reviews the research progress of nsPEF in the treatment of melanoma in cell experiments, animal experiments and human experiments, and points out that nanosecond pulsed electric field can induce melanoma ablation through various mechanisms and produce long-term anti-tumor memory to inhibit its recurrence and metastasis, while having highly targeted effect without additional thermal damage. These unique advantages have broad clinical application prospects for melanoma treatment and provide new ideas for clinical treatment of melanoma.
Vaccine and abscopal effects result from in situ vaccination with nanosecond electric pulse treatment for breast cancer in mice

Abstract:Abstract An electrical engineering technology, nanosecond electric pulse (nsEP), has been studied to ablate local tumor in a non-thermal and minimally invasive manner. Recently, several groups including our lab discovered that animals with complete tumor ablation following nsEP treatment were protected from a second tumor challenge. Therefore, nsEP tumor ablation is also a novel in situ vaccination approach. Breast cancer is considered a low immunogenic tumor with only a small subset of patients responsive to immune checkpoint inhibitors. We found nsEP treatment resulted in not only a potent in situ vaccine effect in a poorly immunogenic 4T1 mouse breast cancer model but also an abscopal effect, a rejection of distant untreated tumor lesion. We further examined the impact of nsEP on cancer cell death, tumor microenvironment (TME) modification, the dynamic changes of local and systemic immune response. 4T1 cells treated with nsEP did not show apoptotic death markers but released damage-associated molecular patterns, including calreticulin, high mobility group protein B1, and ATP. In contrast to abundant immune suppressor cells in untreated breast TME, nsEP treatment led to a significant reduction of both myeloid-derived suppressor cells and regulatory T cells. Conversely, the ratio of tumor associated macrophages M1-like verse M2-like was reversed. An increase of tissue-resident memory T cells was observed in draining lymph nodes. In summary, our results suggest that nsEP is a novel non-drug immunogenic cell death inducer and in situ vaccination approach. Further elucidation of its mechanisms and the development of an in vivo nsEP delivery system suitable for patients is critical to warrant this novel technology toward a clinical trial.

Nanosecond Pulsed Electric Field Ablation‐induced Modulation of Sphingolipid Metabolism is Associated with Ly6c2+ Mononuclear Phagocyte Differentiation in Liver Cancer

Abstract:Preclinical studies have proven that nanosecond pulsed electric field (nsPEF) ablation can be a safe and effective treatment for humans with unresectable liver cancer that are ineligible for thermal ablation. The concomitant activation of antitumor immunity by nsPEF can also potentially prevent tumor recurrence. However, whether nsPEF exhibits similar efficacy in a clinical setting remains to be investigated. A prospective clinical trial (clinicaltrials.gov identifier: NCT04039747) was conducted to evaluate the safety and efficacy of ultrasound (US)‐guided nsPEF ablation in 15 patients with unresectable liver cancer that were ineligible for thermal ablation. We found that nsPEF ablation was safe and produced a 12‐month recurrence‐free survival (RFS) and local RFS of 60% (9/15) and 86.7% (13/15), respectively, in the enrolled patients. Integrative proteomic and metabolomic analysis showed that sphingolipid metabolism was the most significantly enriched pathway in patient sera after nsPEF without recurrence within 8 months. A similar upregulation of sphingolipid metabolism was observed in the intratumoral mononuclear phagocytes (MNPs), rather than other immune and nonimmune cells, of an nsPEF‐treated mouse model. We then demonstrated that lymphocyte antigen 6 complex, locus C2‐positive (Ly6c2+) monocytes first differentiated into Ly6c2+ monocyte‐derived macrophages with an increase in sphingolipid metabolic activity, and subsequently into Ly6c2+ dendritic cells (DCs). Ly6c2+ DCs communicated with CD8+ T cells and increased the proportions of IFN‐γ+ CD8+ memory T cells after nsPEF, and this finding was subsequently confirmed by depletion of liver Ly6c2+ MNPs. In conclusion, nsPEF was a safe and effective treatment for liver cancer. The alteration of sphingolipid metabolism induced by nsPEF was associated with the differentiation of Ly6c2+ MNPs, and subsequently induced the formation of memory CD8+ T cells with potent antitumor effect.
Nanosecond Pulsed Electric Fields As a Novel Drug Free Therapy for Breast Cancer: an in Vivo Study.

Abstract:Nanosecond pulsed electric fields (nsPEFs) is a novel non-thermal approach to induce cell apoptosis. NsPEFs has been proven effective in treating several murine tumors, but few studies focus on its efficacy in treating human tumors. To determine if nsPEFs is equally effective in treatment of human breast cancer, 30 human breast cancer tumors across 30Balb/c (nu/nu) mice were exposed to 720 pulses of 100ns duration, at 4pulsespersecond and 30kV/cm. Two weeks after treatment, the growth of treated tumors was inhibited by 79%. Morphological changes of tumors were observed via a 3.0T clinical magnetic resonance imaging (MRI) system with a self-made surface coil. Pulsed tumors exhibited apoptosis evaluated by TUNEL staining, inhibition in Bcl-2 expression and decreased blood vessel density. Notably, CD34, vascular endothelial growth factor (VEGF) and VEGF receptor (VEGFR) expression in treated tumors were strongly suppressed. To evaluate the might-be adverse effects of nsPEFs in healthy tissues, normal skin was treated exactly the same way as tumors, and pulsed skin showed no permanent damages. The results suggest nsPEFs is able to inhibit human breast cancer development and suppress tumor blood vessel growth, indicating nsPEFs may serve as a novel therapy for breast cancer in the future.

Abstract:Nanosecond pulsed electric field (nsPEF) treatment has emerged as a promising and effective approach for pancreatic cancer. Tumor-infiltrating myeloid cells are crucial tumor regulators and potential immunotherapy targets. Understanding the effect of nsPEF on the myeloid cells in tumors is necessary for grasping the anti-tumor impact of nsPEF therapy. This study describes the phenotype and function of myeloid cells in Panc02 pancreatic cancer mouse models on day three after nsPEF using single-cell RNA sequencing (scRNA-Seq). Defining comparable myeloid cells in Panc02 tumors enabled characterization of their response to nsPEF treatment. Treatment with nsPEF increased infiltration by monocytes/macrophages, which participated in forming a immunosuppressive tumor microenvironment. NsPEF also promoted the recruitment of dendritic cells to tumors. Our comprehensive investigation of crucial myeloid subsets and significant cellular interactions regulating tumor immunity indicated that the nsPEF induced a compartmental remodeling of tumor-infiltrating myeloid cells in pancreatic cancer. These results provide information for interpreting the complex immune changes after nsPEF treatment in pancreatic cancer and may guide future therapeutic interventions.

Abstract:Cancer remains a leading cause of death worldwide and total number of cases globally is increasing. Novel treatment strategies are therefore desperately required for radical treatment of cancers and long survival of patients. A new technology using high pulsed electric field has emerged from military application into biology and medicine by applying nsPEF as a means to inhibit cancer. However, molecular mechanisms of nsPEF on tumors or cancers are still unclear. In this paper, we found that nsPEF had extensive biological effects in cancers, and clarified its possible molecular mechanisms in vitro and in vivo. It could not only induce cell apoptosis via dependent-mitochondria intrinsic apoptosis pathway that was triggered by imbalance of anti- or pro-apoptosis Bcl-2 family proteins, but also inhibit cell proliferation through repressing NF-κB signaling pathway to reduce expressions of cyclin proteins. Moreover, nsPEF could also inactivate metastasis and invasion in cancer cells by suppressing Wnt/β-Catenin signaling pathway to down-regulating expressions of VEGF and MMPs family proteins. More importantly, nsPEF could function safely and effectively as an anti-cancer therapy through inducing tumor cell apoptosis, destroying tumor microenvironment, and depressing angiogenesis in tumor tissue in vivo. These findings may provide a creative and effective therapeutic strategy for cancers.
Pulsed Electric Field Ablation versus Radiofrequency Thermal Ablation in Murine Breast Cancer Models: Anticancer Immune Stimulation, Tumor Response, and Abscopal Effects

Abstract:Purpose: To compare the immune response and survival after size-matched radiofrequency (RF) ablation and a proprietary form of pulsed electric field (PEF) ablation in murine tumors. Material and methods: Orthotopically inoculated EMT6 or 4T1 murine tumors received sham, RF ablation, or PEF ablation. 4T1 tumor ablations included subgroups with intraperitoneal checkpoint inhibition immunotherapy (αPD-1). Blood was collected for cytokine profiling and flow cytometry. Tumor size was measured and survival was monitored. Tumor samples were processed for histology, immunohistochemistry, flow cytometry, and cytokine profiling. Lungs were collected from 4T1-bearing mice for hematoxylin and eosin histology to assess metastatic spread and abscopal effect induced by ablation. Results: PEF elicited distinct immunomodulatory effects, with clear differences in serum and tumor cytokine profiles compared with RF ablation, including intratumoral downregulation of vascular endothelial growth factor, hypoxia-inducible factor 1α, c-MET, interleukin-10, Ki67, and tumor necrosis factor-α (all P < .05). PEF increased innate immune activation, with enhanced recruitment of dendritic cells, M1 macrophages, and natural killer cells coupled with a reduction in M2 macrophages and myeloid-derived suppressor cells (all P < .05). Concurrently, PEF strengthened adaptive immunity compared with RF ablation, characterized by increased antigen-specific T cells and decreased regulatory T cells (all P < .05). PEF stalled tumor growth and increased survival at the end of the study (≥4× versus RFA). Finally, PEF promoted an abscopal effect of clearing metastases in the lungs, which was stronger in combination with αPD-1 than with PEF alone. Conclusions: The proprietary form of PEF used in this study evoked a preferential immunostimulatory profile versus RF ablation thermal ablation in mice, with implications for enhancing the therapeutic effectiveness of checkpoint inhibition immunotherapy for immunotherapy-unresponsive tumors.
