Abstract:Abstract Microbe-mediated ammonia oxidation is a key process in soil nitrogen cycle. However, the effect of maize straw mulching on the ammonia oxidizers in the alkaline purple soil remains largely unknown. A three-year positioning experiment was designed as follows: straw mulching measures as the main-plot treatment and three kinds of nitrogen application as the sub-plot treatment. We found the contents of soil organic carbon (SOC), total nitrogen (TN), available potassium (AK), available nitrogen (AN), available phosphorus (AP), and NH 4 + -N were increased after straw mulching and nitrogen application in alkaline purple soil, so did the amoA genes abundance of ammonia-oxidizing archaeal (AOA) and bacterial (AOB). Terminal restriction fragment length polymorphism (T-RFLP) analysis revealed that Thaumarchaeote (448-bp T-RF) was dominated the AOA communities, whereas Nitrosospira sp (111-bp T-RF) dominated the AOB communities. The community compositions of both AOA and AOB were altered by straw mulching and nitrogen application in alkaline purple soil, however, the AOB communities was more responsive than AOA communities to the straw mulching and nitrogen application. Further analysis indicated that SOC and AP were the main factors affecting the abundance and community compositions of AOA and AOB in alkaline purple soil. The present study reported that straw mulching and nitrogen strategies differently shape the soil ammonia oxidizers community structure and abundance, which should be considered when evaluating agricultural management strategies regarding their sustainability and soil quality.

Abstract:Straw return can be used to reduce fertilizer input and improve soil sustainability, but how straw return and reduced fertilizer application affect beneficial soil microbes, particularly arbuscular mycorrhizal (AM) fungi, remain poorly understood. Here, we conducted a five years of field experiment in a rainfed maize field on the Loess Plateau of northwestern China. The experiment included four treatments with straw return plus four nitrogen (N) application rates (100%, 80%, 60%, and 0% of the common N application rate (225 kg N ha -1 year -1 ) in this region), and two reference treatments with only full or zero N application. We quantified mycorrhizal colonization and AM fungal communities colonizing maize roots were characterized using Illumina sequencing. Forty virtual taxa (VTs) of AM fungi were identified from root samples, of which VT113 (related to Rhizophagus fasciculatus ) and VT156 ( Dominikia gansuensis ) were the two most dominant taxa. Both root length colonization and VT richness of AM fungi were sensitive to N fertilization but not to straw return, and both were gradually increased with the decreasing of N fertilizer rate. The VT composition of AM fungal community was also affected by N application but not by straw return, and the community variation could be well explained by soil available N and P concentrations. In addition, 60%, 80% and full N fertilization produced similar maize yields. This study reveals the response patterns of AM fungi to straw return and N fertilizer reduction, and suggests that straw return with N fertilizer reduction may be a promising practice to synchronously maintain mycorrhizal symbiosis and crop productivity.

Abstract:Straw return is an effective conservation tillage measure that improves soil properties, increases crop yields, and is an efficient means of reusing agrobioenergy by-products. Straw return has been shown to affect nitrogen (N) cycling and related functional microbes in agricultural soils. However, the relationship between nitrous oxide (N2O) emissions and N-related functional genes under different straw return strategies in wheat-corn double cropping systems remains elusive. Here, we conducted a short-term field study to investigate the response of N2O emissions driven by N cycling functional genes (bacterial and archaeal amoA genes and nirS, nirK, and nosZ genes) to different straw return patterns in a wheat-corn double cropping system in the North China Plain. Three treatments were established, namely, both wheat and corn straw return (CWSR), only wheat straw mulch return (WSR), and no straw return (NSR). Over the wheat-corn season, the total N2O emissions significantly increased by 31.74% and 12.37% in CWSR and WSR-treated soils, respectively. Straw return increased nitrifying and denitrifying functional gene abundances, thus stimulating N2O emissions. Correlation analysis showed that AOA (r = 0.262, p < 0.05) and AOB (r = 0.703, p < 0.01) were strongly and positively correlated with NO3−-N concentrations, indicating that AOA and AOB were involved in nitrification under straw return. Both nirS (r = 0.287, p < 0.05) and nirK genes (r = 0.252, p < 0.05) were positively correlated with N2O emissions, while nosZ genes (r = −0.266, p < 0.05) were negatively correlated with N2O emissions. Structural equation model (SEM) analysis further revealed that straw return indirectly affected N2O emissions by influencing the abundances of AOB-amoA (r = 0.324, p < 0.05), nirK (r = 0.417, p < 0.05), and nosZ (r = −0.388, p < 0.001) genes. Overall, short-term straw return increased N2O production in a wheat-corn double cropping system, mainly through increased AOB-amoA and nirK gene abundance, which stimulated N2O production, and reduced nosZ gene abundance, which inhibited N2O reduction.

Abstract:This study evaluated the combined effects of biochar and straw on N2O flux and the community compositions of nitrifiers and denitrifiers in the maize season in an intensively farmed area in northern China. The experiment consisted of four treatments: (1) CK (only chemical fertilizer application); (2) C (biochar application); (3) SR (straw application to the field); and (4) C+SR (the application of both biochar and straw). The results indicated that during the maize growing season, N2O flux decreased by 30.3% in the C treatment and increased by 13.2% and 37.0% in the SR and C+SR treatments compared with CK, respectively. NO3−-N, NH4+-N, and microbial biomass carbon (MBC) were the main soil factors affecting N2O flux, and they were positively correlated with NO3−-N and negatively correlated with MBC in the C treatment and positively correlated with NH4+-N in the SR and C+SR treatments. Both biochar addition and straw return shifted the community compositions of nitrifiers and denitrifiers. N2O production was mainly reduced by promoting the ammonia-oxidizing bacteria (AOB) gene abundance and inhibiting the nirK gene abundance in the C treatment but promoted by inhibiting the AOB and nosZ gene abundances in the SR and C+SR treatments. Nitrosospira (AOB) and Rhizobium (nirK) were the main contributors among the treatments. NO3−-N, NH4+-N, and MBC were the main soil factors affecting the denitrifier communities. The predominant species associated with the nirK, nirS, and nosZ genes were positively correlated with NO3−-N and MBC and negatively correlated with NH4+-N. These results provide valuable information on the mechanism of N2O production and reduction in biochar- and straw-amended soil under field conditions.

Abstract:In order to explore the alteration of N transformation and N2O emissions in acid soil with the co-application of straw and different types of nitrogen (N) fertilizers, an incubation experiment was carried out for 40 days. There are totally five treatments in the study: (a) without straw and N fertilizer (N0), (b) straw alone application (SN0), (c) straw with NH4Cl (SN1), (d) straw with NaNO3 (SN2), and (e) straw with NH4NO3 (SN3). N2O emissions, soil physicochemical properties, and abundance/activity of ammonia-oxidizing archaea (AOA) were measured. The results showed that the combined application of straw and N enhanced N2O emissions, particularly, SN2 and SN3 treatments. Moreover, the soil pH was lower in co-application treatments and the average decreasing rate was 9.69%. Specially, the pH was lowest in the SN1 treatment. The results of correlation analysis indicated a markedly negative relationship between pH and N2O, as well as a negative relationship between pH and net mineralization rate. These findings suggest that pH alteration can affect the N transformation process in soil and thus influence N2O emissions. In addition, the dominant AOA at the genus level in the SN2 treatment was Nitrosopumilus, and Candidatus nitrosocosmicus in the SN3 treatment. The reshaped AOA structure can serve as additional evidence of the changes in the N transformation process. In conclusion, as the return of straw, the cumulation of N2O from arable acid soil depends on the form of N fertilizer. It is also important to consider how N fertilizer is applied to reduce the possibility of N being lost in the soil as gas.

Abstract:The control of nitrous oxide (N 2 O) emissions through nitrification and the optimization of maize yield are important in agricultural systems. However, within the semi-arid region, the impact of fertilization on the function of nitrification communities and its connection with N 2 O emissions in the rhizosphere soil is still unclear. Our study investigates the influence of fertilization treatments on the communities of ammonia-oxidizing bacteria (AOB) and the complete ammonia oxidizers of the Nitrospira known as comammox (CAOB) in a maize agroecosystem. Nitrous oxide production, potential nitrification activity (PNA), maize yield, and nitrogen use efficiency (NUE) were determined for the same samples. The fertilizer treatments included a control group without fertilization (NA), inorganic fertilizer (CF), organic fertilizer (SM), combined inorganic and organic fertilizer (SC), and maize straw (MS). The SC treatment indicated a lower cumulative N 2 O emission than the CF treatment in the 2020 and 2021 cropping seasons. The AOB community under the CF, MS, and SM treatments was predominantly composed of Nitrosospira cluster 3b, while the SC treatment was associated with the comammox Nitrospira clade A.1 lineage, related to key species such as Ca. Nitrospira inopinata and Ca. Nitrospira nitrificans . Network analysis demonstrated a positive potential for competitive interaction between hub taxonomy and distinct keystone taxa among AOB and comammox Nitrospira nitrifiers. The structural equation model further revealed a significant positive association between AOB nitrifiers and N 2 O emission, PNA, soil pH, SOC, NO3− -N, and DON under organic fertilization. The keystone taxa in the comammox Nitrospira nitrifier and network Module II exhibited a positive correlation with maize productivity and NUE, likely due to their functional activities stimulated by the SC treatment. It is noteworthy that the AOB community played a more significant role in driving nitrification compared to the composition of comammox Nitrospira . Collectively, combined inorganic and organic fertilizer (SC) treatment exhibits high potential for reducing N 2 O emissions, enhancing maize productivity, increasing NUE, and increasing the sustainability of the nitrogen dynamics of maize agroecosystems in the semi-arid Loess Plateau.

Abstract:Context Crop straw is a significant byproduct of agricultural production and a rich source of nutrients, but it is carelessly disposed of, potentially harming the environment and wasting valuable resources. In China, returning straw to the soil as a means of disposing of agricultural waste is a crucial management technique that not only makes use of agricultural waste but also enhances soil quality and grain yield. Objective A five-year (2018–2022) field experiment was designed to study the effect of straw treatment methods and optimal nitrogen (N) application rate on the photosynthesis process and grain yield of maize. Methods The different straw methods (straw return (SR) and traditional practice (TP) — no straw return) were assigned to main plots while the N levels such as N100 (100 kg ha−1), N150 (150 kg ha−1), N200 (200 kg ha−1), N250 (250 kg ha−1), and N300 (300 kg ha−1) were allotted to sub-plots. Results The results showed the photosynthesis rate (Pn), intercellular carbon dioxide concentration, stomatal conductance, transpiration rate, relative chlorophyll content (SPAD), leaf area index (LAI), dry matter (DM), soil nitrogen content, maize grain nitrogen accumulation, and grain yield were increased with an increase in N levels. Similarly, SR also showed a promotion effect on the aforementioned parameters compared with TP. After five years, SR with an optimum N level (250 kg ha−1) increased the SPAD values (2%) and Pn (8%) which resulted in higher LAI (27%) compared with TPN300. As a result, more DM (15%) was formed and finally increased the grain yield (7%). It was revealed that the SPAD, Pn, LAI, DM, and grain yield were maximum at a higher 300 kg N ha−1 level but it was found statistically similar with 250 kg N ha−1. So in this study, SR with 250 kg N ha−1 is optimum for better photosynthetic efficiency, dry matter production, and grain yield. The correlation analysis also showed a positive correlation (i) of SPAD values with Pn (ii) LAI with SPAD values and Pn, (iii) DM with SPAD values, Pn, and LAI, and (iv) grain yield with DM under SR compared with TP. Conclusion and implication In , SRN250 resulted in better chlorophyll content and photosynthesis process that yielded more photosynthetic area for higher DM production and subsequently higher maize grain yield. The SR reduced the amount of N fertilizer and improved the grain yield and it is the best approach to improve grain yield and minimize agricultural waste production. Long-term trials are needed to understand the effects of straw incorporation and nitrogen application in different regions and locations.

Abstract:Organic material (OM) applied to cropland not only enhances soil fertility but also profoundly affects soil nitrogen cycling. However, little is known about the relative contributions of soil ammonia-oxidizing archaea (AOA) and bacteria (AOB) to nitrous oxide (N2O) production during ammonia oxidation in response to the additions of diverse types of OMs in the tropical soil for vegetable production. Herein, the soils were sampled from a tropical vegetable field subjected to 4-year consecutive amendments of straw or manure. All the soils were amended with ammonium sulfate ((NH4)2SO4, applied at a dose of 150 mg N kg−1) and incubated aerobically for four weeks under 50% water holding capacity. 1-octyne or acetylene inhibition technique was used to differentiate the relative contributions of AOA and AOB to N2O production. Results showed that AOA dominated N2O production in soil managements of unfertilized control (CK), chemical fertilization (NPK), and NPK with straw (NPKS), whereas AOB contributed more in soil under NPK with manure (NPKM). Straw addition stimulated AOA-dependent N2O production by 94.8% despite the decreased AOA-amoA abundance. Moreover, manure incorporation triggered both AOA- and AOB-dependent N2O production by 147.2% and 233.7%, respectively, accompanied with increased AOA and AOB abundances. Those stimulating effects were stronger for AOB, owing to its sensitivity to the alleviated soil acidification and decreased soil C/N ratio. Our findings highlight the stimulated N2O emissions during ammonia oxidation by historical OM amendments in tropical vegetable soil, with the magnitude of those priming effects dependent on the types of OM, and appropriate measures need to be taken to counter this challenge in tropical agriculture ecosystems.

Abstract:The application of straw and nutrients to agricultural subsoils is of significant interest to enhance carbon (C) sequestration and soil fertility. However, little research has explored the effect straw application on microbial stoichiometry, soil organic carbon (SOC) mineralization and accumulation and their relationships, in subsoil. In order to address these knowledge gaps, we examined the soil characteristics (organic carbon mineralization, available nutrient contents and microbial stoichiometric ratio) under straw addition (maize leaf and stem) with and without nitrogen (N) supplementation (no addition, 30 mg N kg(-1) dry soil(-1), 60 mg N kg(-1) dry soil(-1)) during an 80-day incubation experiment. The microbial stoichiometric ratio imbalance (C:N and C:Phosphorus (P)) and extracellular enzyme stoichiometry were measured as indicators of the systematic relationship between soil resource availability and the mineralization of organic carbon. Our study demonstrated that the addition of straw significantly enhanced CO2 emissions and led to an increase in the C:N imbalance, while simultaneously decreasing microbial carbon use efficiency (CUE). In addition, stem addition showed 5.6% lower CUE, but 8.2% higher SOC compared with leaf addition. We also found that nitrogen addition to subsoil alleviated microbial nitrogen limitation. 60 mg N kg(-1) dry soil rates of nitrogen application had a positive effect on reducing C:N imbalance and promoting the accumulation of SOC. Extracellular enzyme activity and microbial stoichiometric ratio were the most important controlling factors of SOC mineralization and microbial CUE, respectively. In conclusion, the application of straw alongside N to balance stoichiometric ratios can significantly increase SOC content, indicating the potential for carbon sequestration in agricultural subsoils.
Effects of straw returning combined with earthworm addition on nitrification and ammonia oxidizers in paddy soil

Abstract:Introduction: Soil ammonia oxidation, which acts as the first and rate-limiting step of nitrification, is driven by ammonia-oxidizing bacteria (AOB), ammonia-oxidizing archaea (AOA) and complete ammonia oxidizer (comammox, amoA gene of clade-A and clade-B). Straw returning, widely used ecological technology in China, is an effective measure for promoting straw decomposition and soil nutrient cycling when combined with earthworm addition. However, the effects of straw returning combined with earthworm addition on soil ammonia oxidizers remain poorly understood. Methods: A 2-year plot experiment was conducted with 5 treatments: no fertilizer (CK); regular fertilization (RT); straw returning (SR); earthworm addition (W); straw returning + earthworm addition (SRW). The AOA, AOB, comammox clade-A and clade-B community microbial diversities and structures were investigated by high-throughput sequencing. Results: The results showed that (1) compared to RT treatment, W, SR, and SRW treatments all significantly increased the richness of AOA and comammox clade-A and clade-B (p < 0.05), and the richness of AOB was only significantly promoted by SRW treatment (p < 0.05). However, only SRW had a higher comammox clade-B diversity index than RT. (2) The ammonia oxidizer community structures were altered by both straw returning and earthworm addition. Soil NH4 +-N was the critical environmental driver for altering the ammonia oxidizer community structure. (3) Compared with RT treatment, the soil potential nitrification rate (PNR) of W and SRW treatments increased by 1.19 and 1.20 times, respectively. The PNR was significantly positively correlated with AOB abundance (path coefficient = 0.712, p < 0.05) and negatively correlated with clade-B abundance (path coefficient = -0.106, p < 0.05). Discussion: This study provides scientific support for the application of straw returning combined with earthworm addition to improve soil nitrification with respect to soil ammonia-oxidizing microorganisms.

Abstract:Straw return with nitrogen (N) fertilizer can generally regulate soil N cycling by affecting the microbial community, thereby influencing crop productivity. Bacteria and fungi play a key role in nitrification, while the effects of soil microbial interactions that include protists under the condition of straw return and N fertilizer remain uncertain. Here, we evaluated the importance of the soil microbiome, particularly protists and ammonia‐oxidizing bacteria (AOB), in regulating the soil N transformation potential and wheat yields under different fertilization practices. The results showed that the activity of β‐1,4‐N‐acetylglucosaminidase (NAG) and the potential nitrification rate (PNR) increased significantly by 156.4% ~ 312.3% and 183.1% ~ 402.6% with the addition of N fertilizer and straw, respectively. The effect on PNR was greater for straw than fertilizer addition. The fertilization practices changed the microbial community structure, and there were stronger responses from the protists and AOB. The soil microbiome formed three closely co‐occurring modules (Modules 0, 1 and 2). Among them, Module 0 showed strong positive correlations with the soil total N, NAG, PNR and crop yield and may be the key ecological cluster. Moreover, the predatory protists Cercozoa, Conosa and Ochrophyta in Module 0 were significantly positively associated with the activity of NAG and the PNR, yields and AOB. The predatory protists in Module 0 may change the soil potential nitrification rate and enhance the yield of wheat by preying on the competitors of AOB. The results suggest that the incorporation of straw promotes the N transformation potential and wheat yields linked to trophic interactions between predatory protists and AOB in fluvo‐aquic soil.

Abstract:To determine the reasonable rate of straw return and nitrogen (N) fertilizer use which may maintain soil ecosystem health, we analyzed their soil microbial biomass and composition in a 10-year field experiment with different rates of straw return (50%, 100%) and N fertilizer (270, 360, 450, 540 kg N ha−1 yr−1) by phospholipid fatty acid (PLFA) analysis and high-throughput sequencing. A rate of 50% straw return combined with 450 or 540 kg N ha−1 yr−1 effectively increased the soil available nutrient contents mainly for total nitrogen, available potassium, and available phosphorus. Total PLFAs indicated that straw return combined with N fertilizer promoted soil microbial growth and increased biomass. A rate of 100% straw return with 450 kg N ha−1 yr−1 was not conducive to the stability of the soil ecosystem according to the ratio of fungi to bacteria (F:B). The similar rate of straw returning and the similar level of nitrogen fertilizer application will be divided into the same cluster using a heatmap analysis. Some saprophytic fungi or pathogens became the dominant fungi genera, such as Gibberella, Sarocladium, Pseudallescheria, and Mycosphaerella, in the treatments with 100% straw returning combining higher N fertilizer (>450 kg ha−1 yr−1 yr−1 added). The relative abundances of some heavy metal-tolerant bacteria, such as those in Proteobacteria and Chlorobi, increased in the soils in the 100% straw return treatments. Therefore, the combined application of 100% straw returning and higher N fertilizer (>450 kg ha−1 yr−1) added long-term was not appropriate for soil health, which will lead to the risk of disease and pollution in soil.

Abstract:Enhancement of microbial assimilation of inorganic nitrogen (N) by straw addition is believed to be an effective pathway to improve farmland N cycling. However, the effectiveness of differently pretreated straws on soil N 2 O emissions and soil N-acquiring enzyme activities remains unclear. In this study, a pot experiment with four treatments (I, no addition, CK; II, respective addition of maize straw, S; III, composted maize straw under no fungi inoculation, SC; and IV, composted maize straw under fungi inoculation, SCPA) at the same quantity of carbon (C) input was conducted under the same amount of inorganic N fertilization. Results showed that the seasonal cumulative N 2 O emissions following the SCPA treatment were the lowest at 4.03 kg N ha −1 , representing a significant reduction of 19 % compared with the CK treatment. The S and SC treatments had no significant effects on N 2 O emissions. The decrease of soil N 2 O emissions following the SCPA treatment was mainly attributed to the increase of microbial N assimilation and the increased abundance of functional genes related to N 2 O reductase. The SCPA treatment significantly decreased soil alkaline phosphatase (ALP) activity and increased leucine aminopeptidase (LAP) activity at the basal fertilization, while increased soil ALP and LAP activity, decreased soil N-Acetyl-β-D-Glucosidase (NAG) activity at harvest. Compared with the CK treatment, the S, SC, and SCPA treatment significantly increased soil β-glucosidase (β-GC) activity at harvest. The decrease in the (NAG+LAP)/ALP ratio following the SCPA treatment indicated that the composted maize straw under fungi inoculation alleviated microbial N limitation at harvest. Moreover, PICRUSt analysis also suggested that the SCPA treatment increased the abundance of bacterial genes associated with N assimilation and N 2 O reduction, whereas the S and SC treatment did not significantly affect the abundance of N 2 O reduction genes compared with the CK treatment. Our results suggest that the composted maize straw under fungi inoculation would reduce the risk of N 2 O emissions and effectively mitigate the microbial N limitation in dryland wheat system.

Abstract:Under a full straw returning system, the relationship between soil bacterial community diversity and straw decomposition, yield, and the combined application of slow-release nitrogen and urea remains unclear. To evaluate these effects and provide an effective strategy for sustainable agricultural production, a 2-year field positioning trial was conducted using maize as the research object. Six experimental treatments were set up: straw returning + no nitrogen fertilizer (S1N0), straw returning + slow-release nitrogen fertilizer:urea = 0:100% (S1N1), straw returning + slow-release nitrogen fertilizer:urea = 30%:70% (S1N2), straw returning + slow-release nitrogen fertilizer:urea = 60%:40% (S1N3), straw returning + slow-release nitrogen fertilizer:urea = 90%:10% (S1N4), and straw removal + slow-release nitrogen fertilizer:urea = 30%:70% (S0N2). Significant differences (p < 0.05) were observed between treatments for Proteobacteria, Acidobacteriota, Myxococcota, and Actinobacteriota at the jointing stage; Proteobacteria, Acidobacteriota, Myxococcota, Bacteroidota, and Gemmatimonadota at the tasseling stage; and Bacteroidota, Firmicutes, Myxococcota, Methylomirabilota, and Proteobacteria at the maturity stage. The alpha diversity analysis of the soil bacterial community showed that the number of operational taxonomic units (OTUs) and the Chao1 index were higher in S1N2, S1N3, and S1N4 compared with S0N2 at each growth stage. Additionally, the alpha diversity measures were higher in S1N3 and S1N4 compared with S1N2. The beta diversity analysis of the soil bacterial community showed that the bacterial communities in S1N3 and S1N4 were more similar or closely clustered together, while S0N2 was further from all treatments across the three growth stages. The cumulative straw decomposition rate was tested for each treatment, and data showed that S1N3 (90.58%) had the highest decomposition rate. At the phylum level, straw decomposition was positively correlated with Proteobacteria, Actinobacteriota, Myxococcota, and Bacteroidota but significantly negatively correlated with Acidobacteriota. PICRUSt2 function prediction results show that the relative abundance of bacteria in soil samples from each treatment differed significantly. The maize yield of S1N3 was 15597.85 ± 1477.17 kg/hm2, which was 12.80 and 4.18% higher than that of S1N1 and S0N2, respectively. In conclusion, a combination of slow-release nitrogen fertilizer and urea can enhance the straw decomposition rate and maize yield by improving the soil bacterial community and structure within a full straw returning system.
[Effects of Straw Addition on N2O and CO2 Emissions from Red Soil Subjected to Different Long-Term Fertilization]

Abstract:Straw return, as an important measure for soil fertility improvement in farmland, significantly affects the emissions of greenhouse gases N2O and CO2. Thus, the collected soil samples from five long-term (30-year) fertilization treatments (no fertilization, CK; recommended chemical fertilizer, F; 200 % of recommended chemical fertilizer, 2F; pig manure, M; and chemical fertilizer combined with pig manure, FM) were amended with and without straw and incubated under constant temperature and humidity conditions (25 ℃ and 65 % maximum field water holding capacity) for 20 days so as to investigate the key factors influencing N2O and CO2 emissions in response to straw addition in long-term fertilization treatments. The results showed that fertilization significantly increased N2O emissions. Compared to those under the unfertilized treatment[(22.05 ±2.09) μg·kg-1, calculated as nitrogen, the same as below], cumulative N2O emissions from the chemical fertilizer treatments significantly increased by 119 %-195 %[(48.38 ±20.81) μg·kg-1 and (65.13 ±12.55) μg·kg-1 from the F and 2F treatments, respectively], and those from the manure treatments increased by 275 %-399 %[(82.72 ±12.73) μg·kg-1 and (1 101.99 ±425.71) μg·kg-1 from the M and FM treatments, respectively]. Soil NO3--N, DOC, and DTN were the main factors influencing N2O emissions from fertilized treatments in the absence of straw addition. Straw addition significantly increased cumulative N2O emissions by 345 % and 247 % in the 2F and M treatments, respectively, compared to those in the corresponding fertilized treatments without straw addition, with no significant effect on N2O emissions in the CK, F, and FM treatments. Straw addition increased DOC content and microbial activity and decreased soil NO3--N and DTN contents, thereby increasing N2O emissions. Fertilization also significantly increased CO2 emissions. Compared to those from the unfertilized treatment, cumulative CO2 emissions from the manure treatments significantly increased by 120 %-130 %[(122.11 ±4.3) mg·kg-1 (calculated as carbon, the same as below) and (116.47 ±4.55) mg·kg-1 from the M and FM treatments, respectively], and those in the 2F treatment increased by 28 %[(65.13 ±12.55) mg·kg-1]. In the absence of straw addition, soil MBC, DOC, and DTN were the main factors influencing CO2 emissions. Compared to those in the treatments without straw addition, straw addition significantly increased cumulative CO2 emissions by 660 %-1132 % among fertilization treatments, due to increased DOC and MBC contents and enhanced microbial activity. In conclusion, straw addition significantly increased N2O and CO2 emissions through increased soil DTN consumption and DOC content among fertilization treatments. In soils treated with manure amendment, straw return should be rationally considered for the purpose of balancing the comprehensive trade-offs between fertility improvement and greenhouse gas emissions.

Abstract:Under the system of full straw returning, the relationship between soil fungal community diversity and soil physiochemical properties, and the combined application of slow-release nitrogen and urea is unclear. To evaluate its effect and provide an effective strategy for sustainable agricultural production, a 2-year field positioning trial was conducted using maize as the research object. The experiment was designed with two factors: straw treatment(S) and nitrogen fertilizer treatment(N),Six experimental treatments were set up,S1N0,S1N1,S1N2,S1N3,S1N4,S0N2,respectively.Analysis of 54 soil samples revealed 15 fungal phyla and 49 fungal classes. The composition of fungal communities in each treatment was basically the same, but there were significant differences in species abundance. Under total straw returning conditions, the combined application of slow-release nitrogen fertilizer and normal nitrogen fertilizer significantly increased the relative abundance of Ascomycota. During the jointing stage, tasseling stage and maturity stage, S1N4, S1N3 and S1N2 increased by 25.76%, 22.97%, 20.74%; 25.11%, 30.02%, 23.64% and 22.47%, 28.14%, 22.71% respectively compared with S0N2.The relative abundance of Basidiomycota was significantly reduced. Alpha diversity analysis showed that the straw returning mode significantly increased the Shannon index and decreased the Simpson index, which was obvious in the jointing stage and tasseling stage. The principal coordinate analysis analysis results showed that the fungal communities formed different clusters in the horizontal and vertical directions at the three growth stages of corn jointing, tasseling and maturity. At the jointing stage and tasseling stage, the communities of the straw return treatment and the straw removal treatment were separated, and the community distribution of each treatment was not significantly different in the mature stage. Total straw returning combined with slow-release fertilizer significantly (P＜0.05) increased the soil organic carbon, nitrate nitrogen and ammonia nitrogen content in each growth period, and increased the soil total nitrogen and hydrolyzable nitrogen content (P＞0.05).After the straw was returned to the field, the combined application of slow-release nitrogen fertilizer and common urea had a significant impact on soil urease, catalase, and sucrase activities. Among them, the three enzyme activities were the highest in the S1N3 treatment at the jointing stage and maturity stage, and the S1N4 treatment at the tasseling stage had the highest enzyme activity. Fungal community composition is closely related to environmental factors. Soil organic carbon, urease and catalase are positively correlated with Ascomycota and negatively correlated with Basidiomycota.

Abstract:An incubation experiment with equivalent N rates was conducted for 56 days using a typical black soil amended with chemical fertilizer with or without straw amendment using a 15N cross-labeling technique. Compared with the chemical fertilizer treatment (15NCF), chemical fertilizer combined with straw treatment (CF + S) showed a significantly higher (P < 0.05) contribution from applied N to microbial biomass N (BN) in the first 14 days and to particulate organic N (PON) and mineral-associated total N (MON) throughout the incubation. Straw application in the CF + S treatment significantly (P < 0.05) decreased the recovery of chemical fertilizer N as soil inorganic N except at day 3 but increased the recovery of chemical fertilizer N as BN before day 14 and as PON and MON from day 14 to the end of the incubation period. At the end of the incubation period, the total N2O-N emissions in the CF + S treatment increased significantly (P < 0.05) compared with the CF treatment, and the increase in N2O-N emissions was 73% from chemical fertilizer and 27% from straw N individually. Our results suggest that the combined application of chemical fertilizer and straw increased soil fertility together with an increase in N2O emissions in the typical black soil and the N2O emissions from straw cannot be ignored.

Abstract:We investigated the linkages between composition and abundance of ammonia –oxidizing bacteria (AOB) and denitrifiers and emissions of N2O in a summer maize (Zea mays L.) field in a calcareous, intensively managed, agricultural soil. The treatments were zero‐N (control), NH4+–based fertilizer N (favoring nitrification), NO3−–based fertilizer N + glucose + soil compaction (NO3−+Glu+Com, favoring denitrification), and urea + straw (U+S, representing conventional farming practices). We observed a substantial period (approximately 1 wk) of high N2O emissions that commenced after N fertilization and irrigation. The highest emissions occurred in the NO3−+Glu+Com treatment. A significant positive relationship was observed between soil NH4+–N concentrations and N2O emissions in the NH4+ and U+S treatments. In general, a positive relationship between N2O emissions and soil water‐filled pore space (WFPS) existed when WFPS reached >55%. Community structure of AOB in the NH4+ treatment during the high N2O emission stage, based on terminal restriction fragment length polymorphism, was similar to that in the control treatment. There were no changes in the denitrifier community structure under NO3−+Glu+Com when 53 times higher N2O emissions occurred than in the control. High abundance of amoA gene copies for AOB and nir gene copies for denitrifiers were detected, however, during the high N2O emission period in the NH4+ and NO3−+Glu+Com treatments, respectively. The results indicate that N2O emissions were related to the abundance of AOB or denitrifiers. It remains unclear, however, whether their community composition can also have an effect on N2O emissions.
Effects of Straw Amendment in Combination with Synthetic N Fertilizer Addition on N2O, N2, and Their Stoichiometric Ratios in Three Different Agro-Ecosystems

Abstract:Nitrogen (N) fertilizer and crop residue amendments are important agricultural practices that could increase soil health, fertility, and crop yield. Such practices may also change soil denitrification processes where contradictory observations have been reported on soil N2O emissions with fewer studies on N2 emissions due to its large atmospheric background concentrations limiting its soil-borne measurement. This study aims to investigate N2O production and reduction of N2 emissions under a conducive denitrifying environment (like anaerobic microsites, 80% WFPS, available N and C) after rice straw amendment and KNO3 application to three different soil types (fluvo-aquic, black, and paddy soils). In this regard, three treatments for three different soil types were set consisting of (a) a non-amended treatment (control), (b) a KNO3 treatment (KNO3, 20 mM KNO3), and (c) a straw plus KNO3 treatment (2.5 g rice straw kg−1 dry soil and 20 mM KNO3), which were incubated under 80% WFPS. Moreover, direct N2O and N2 fluxes were measured over 17 days in the current incubation experiment with a robotized incubation system using a helium atmosphere. Results showed that rice straw amendment combined with N fertilizer increased both N2O and N2 fluxes compared with control or KNO3 treatments in all three soil types. Overall, compared with the black and paddy soils, the N2O and N2 fluxes were higher in the fluvo-aquic soil, with a maximum of 234.2 ± 6.3 and 590.1 ± 27.3 g N ha−1 from F_SK treatment, respectively, during the incubation period. The general trends in three soil types of both N2O and N2 emissions were control < KNO3 < rice straw plus KNO3 treatments. Straw amendment in combination with KNO3 can stimulate a high denitrification rate (less N2O and higher N2), whereas their effect on stoichiometric ratios of N2O/(N2O + N2) highly depends on soil nitrate concentration, oxygen level, soil moisture content, and labile C. The current study underscores that the rice straw amendment in combination with N fertilizer can trigger denitrification with less increment on soil N2O but higher N2 emissions under conditions favoring denitrification.
