Abstract:The authors designed a chemical genomics screen with the aim of understanding genes and pathways that modulate neural stem/precursor cell differentiation. Multipotent mouse neural precursor cells isolated from cortices of embryonic day 12 (E12) embryos were subjected to spontaneous differentiation triggered by growth factor withdrawal. A quantitative whole-well immunofluorescence assay was set up to screen tool compound sets to identify small molecules with potent, dose-dependent, and reproducible effects on increasing neural stem cell differentiation toward neuronal lineage. Among the pro-neuronal compounds, kinase inhibitors were shown to exert pro-neuronal effect via a signaling pathway associated with the kinase. The global effect of hit compounds on modulating neuronal differentiation was confirmed by an in vivo mouse study and human neural stem cells culture. This study demonstrates that a phenotypic assay using cell type-specific antibody markers can be used for a large-scale compound screen to discover targets and pathways with impacts on differentiation of lineage-restricted precursor cells toward specific lineages.
(+)4-Cholesten-3-one promotes differentiation of neural stem cells into dopaminergic neurons through TET1 and FoxA2

Abstract:In this paper, we report the results of treating cells with an effective small molecule, (+)4-cholesten-3-one (PubChem CID: 91477), which can promote neural stem cell(NSC) differentiation into dopaminergic neurons. This study used rat neural stem cells stimulated with two different concentrations (7.8 μM and 78 μM) of (+)4-cholesten-3-one. Cell phenotypic analysis showed that (+)4-cholesten-3-one induced NSC differentiation into dopaminergic neurons, and the level of tyrosine hydroxylase(TH), which is specific for dopaminergic cells, was significantly increased compared with that of the drug-free control group. Furthermore, in this study, we found that this effect may be related to the transcription factor fork-head box a2 (FoxA2) and ten-eleven translocation 1 (TET1). The expression of TET1 and FoxA2 was upregulated after treatment with (+)4-cholesten-3-one. To verify the relationship between (+)4-cholesten-3-one and these genes, we found that the binding rate of TET1 and FoxA2 increased after the application of (+)4-cholesten-3-one, as confirmed by a coimmunoprecipitation (Co-IP) assay. With a small interfering RNA (siRNA) experiment, we found that only when Tet1 and Foxa2 were not silenced was the mRNA level of Th increased after (+)4-cholesten-3-one treatment. Taken together, these data show that (+)4-cholesten-3-one can promote the differentiation of NSCs into dopaminergic neurons by upregulating the expression of TET1 and FoxA2 and by increasing their binding. Thus, (+)4-cholesten-3-one may help address the application of neural stem cell replacement therapy in neurodegenerative diseases.

Neuron-Like Differentiation of Mesenchymal Stem Cells Mediated by Nurr1 and Mash1 in Vitro

Abstract:Cellular therapies using stem cells are promising approaches for the treatment of neurological diseases, as neuron has limited potential to self-renew or repair. Thus, mesenchymal stem cells (MSCs), a kind of multipotent progenitors, can represent an alternative source of cells for neuron replacement therapies. Here we show that forced co-expression of neurogenic factors Nurr1 and Mash1, two transcriptional factors specific to dopamine neuron development and treatment with Neurobasal-A medium containing nerve growth factor and GSK-3 inhibitors (TWS119) could directly and effectively induce mesenchymal stem cells differentiation into morphologically, phenotypically and functionally neuron-like cells. The differentiated cells significantly increased expression of neuron specific transcription factors (Pitx3, Lmx1a, Ngn2 and En2); increased various neuron markers such as tyrosine hydroxylase, microtubule associated protein-2 (MAP2), dopamine transporter (DAT), neuron-specific neuronal protein (NeuN), neurofilament medium (NF-M), beta-III Tubulin (Tuj1) and neuron specific enolase (NSE). Our results showed characteristic dopamine neurotransmitter release in response to depolarization stimuli and specific dopamine reuptake properties in vitro. Nurr1 and Mash1 expression remained the number of dopamine neuron-like cells stable in the cultures for 15 days, implying that Mash1 and Nurr1 modulate neuron-like differentiation of mesenchymal stem cells. Altogether, the combined expression of specific transcription factors Nurr1 and Mash1 enhances mesenchymal stem cells differentiation in vitro to generate neuron-like cells as a promising source of cell for therapy of neurological diseases.
Induced Neural Stem/precursor Cells for Fundamental Studies and Potential Application in Neurodegenerative Diseases

Abstract:Recent research has shown that defined sets of exogenous factors are sufficient to convert rodent and human somatic cells directly into induced neural stem cells or neural precursor cells (iNSCs/iNPCs). The process of transdifferentiation bypasses the step of a pluripotent state and reduces the risk of tumorigenesis and genetic instability while retaining the self-renewing capacity. This iNSC/iNPC technology has fueled much excitement in regenerative medicine, as these cells can be differentiated into target cells for re placement therapy for neurodegenerative diseases. Patients' somatic cell-derived iNSCs/iNPCs have also been proposed to serve as disease models with potential value in both fundamental studies and clinical applications. This review focuses on the mechanisms, techniques, and app lications of iNSCs/iNPCs from a series of related studies, as well as further efforts in designing novel strategies using iNSC/iNPC technology and its potential applications in neurodegenerative diseases.

Abstract:The success of stem cells therapy to treat neurodegenerative diseases is currently restricted by the lack of suitable stem cells. Mesenchymal stem cells (MSCs) have demonstrated several advantages as seed-cells for the stem cells therapy. In particular, the low immunogenicity and multiple lineages differentiation capability enables the possibility of using MSCs to treat neurodegenerative diseases. However, a more potent neuronal differentiation capacity of MSCs is required during a success treatment against neurodegenerative diseases. Bioengineering using small molecules to boost the neuronal differentiation of MSCs has been proposed as a promising strategy. Herein, we developed a new series of (2-phenylthiazol-4-yl)urea derivatives and one of them, 18g were observed to successfully promote neuronal differentiation of MSCs after culturing MSCs with 18g for 4 days. In addition, neither significant cytotoxicity nor cell cycle altering were found after the incubation. Interestingly, the osteogenic differentiation potential of MSCs was not affected after 18g treatment. The present study provides a promising small molecule to boost the innate neuronal differentiation capacity of MSCs with no serious detrimental effects.
Nano‐Enabled Intracellular Bursting of Calcium and Retinoic Acid Regulates Dopaminergic Neuronal Differentiation of NSCs for Parkinson's Disease Therapy

Abstract:A new method is proposed to regulate dopaminergic neuronal differentiation of neural stem cells (NSCs) through nano‐enabled intracellular bursting of calcium ions and retinoic acid. Calcium ions act as accelerators, promoting differentiation, while retinoic acid guides differentiation toward dopaminergic neurons. The transplanted NSCs with endocytosed calcium‐retinoic acid (Ca‐RA) nanoparticles can achieve dopaminergic differentiation in vivo and improve motor and memory abilities of parkinson's disease (PD) mice. Parkinson's disease (PD) is a debilitating neurodegenerative disorder characterized by dopaminergic neuron degeneration. Neural stem cell (NSC) therapy offers promise for replacing these neurons and restoring neural function. However, directing NSCs to become dopaminergic neurons is challenging. Retinoic acid (RA) is a potential regulator, but its insolubility in water limits its use in PD therapy. Herein, nanonizing RA with calcium acetate to create calcium‐retinoic acid nanoparticles (Ca‐RA NPs) is proposed. These nanoparticles can be internalized by NSCs and then dissociated in the acidic environment of lysosomes to lead to a burst of Ca2+ and RA. In vitro results showed that the intracellular bursting of Ca2+ and RA accelerated neuronal differentiation and maturation by 5–10 days compared to spontaneous NSC differentiation. Importantly, Ca‐RA NPs uniquely directed NSCs to dopaminergic neurons, involving the interaction between the calcium ion‐mediated MAPK signaling pathway and the RA‐mediated RA signaling pathway. Animal experiments further validated the efficacy of Ca‐RA‐coated NSCs in restoring motor and cognitive functions in PD mice by rapidly forming dopaminergic neural circuits. Given that both RA and calcium acetate are approved by the FDA, this strategy has the potential for translation into a clinical treatment approach for stem cell therapy of PD.

Induction and differentiation of bone marrow mesenchymal stem cells into dopaminergic neurons

Abstract:BACKGROUND: Recent studies have found that neurotrophic factor plays important roles in differentiation of bone mesenychymal stem cells (BMSCs). However, few reports have addressed whether neural stem cells (NSCs) with regeneration capacity in the brain tissue in vitro can induce the differentiation of BMSCs into dopaminergic neurons. OBJECTIVE: To investigate the possibility of rat BMSCs differentiation into dopaminergic neurons under the induction condition of glial cells-derived neurotrophic factor (GDNF) and NSCs coculture. METHODS: BMSCs from Sprague Dawley rats were isolated and cultured. The third passage of BMSCs was divided into two groups. Group A: Medium was MEM containing GDNF; group B: BMSCs marked with Brdu and then cocultured with NSCs which had formed primary neurospheres. Three days later, BMSCs from each group were suffered immunohistochemistry staining to examine the tyrosine hydroxylase of the differentiated neuron to observe the differentiation of BMSCs. RESULTS AND CONCLUSION: At 24 hours postinduction, BMSCs of group A were cone-shaped and had the prolonged processes. The amount of processes increased, with the presence of neuron-like morphology. The web connection among the cells can also be observed. Three days later, some cells expressed tyrosine hydroxylase. In group B, NSCs attached to culture dish immediately. The attached cells proliferated and showed neuronal phenotype. A majority of attached cells expressed tyrosine hydroxylase, and a few cells presented double staining: Brdu/tyrosine hydroxylase. Results have exhibited that BMSCs have the potential of differentiating into dopaminergic neurons. The GDNF proves to be more effective in the experiment compared with NSCs.
Human neural stem cells promote mitochondrial genesis to alleviate neuronal damage in MPTP-induced cynomolgus monkey models

Abstract:Currently, there is no effective treatment for Parkinson's disease (PD), and the regenerative treatment of neural stem cells (NSCs) is considered the most promising method. This study aimed to investigate the protective effect and mechanism of NSCs on neurons in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) induced cynomolgus monkey (Macaca fascicularis) model of PD. We first found that injecting NSCs into the subarachnoid space relieved motor dysfunction in PD cynomolgus monkeys, as well as reduced dopaminergic neuron loss and neuronal damage in the substantia nigra (SN) and striatum. Besides, NSCs decreased 17-estradiol (E2) level, an estrogen, in the cerebrospinal fluid (CSF) of PD cynomolgus monkeys, which shows NSCs may provide neuro-protection by controlling estrogen levels in the CSF. Furthermore, NSCs elevated proliferator-activated receptor gamma coactivator-1 alpha (PGC-1a), mitofusin 2 (MFN2), and optic atrophy 1 (OPA1) expression, three genes mediating mitochondrial biogenesis, in the SN and striatum of PD monkeys. In addition, NSCs suppress reactive oxygen species (ROS) production caused by MPTP, as well as mitochondrial autophagy, therefore preserving dopaminergic neurons. In summary, our findings show that NSCs may preserve dopaminergic and neuronal cells in an MPTP-induced PD cynomolgus monkey model. These protective benefits might be attributed to NSCs' ability of modulating estrogen balance, increasing mitochondrial biogenesis, and limiting oxidative stress and mitochondrial autophagy. These findings add to our understanding of the mechanism of NSC treatment and shed light on further clinical treatment options.

Abstract:Neural stem cells (NSCs) are currently considered powerful candidates for cell therapy in neurodegenerative disorders such as Parkinson's disease. However, it is not known when and how NSCs begin to differentiate functionally. Recent reports suggest that classical neurotransmitters such as acetylcholine (Ach) are involved in the proliferation and differentiation of neural progenitor cells, suggesting that neurotransmitters play an important regulatory role in development of the central nervous system (CNS). We have shown by calcium imaging and immunochemistry that proliferation and differentiation are enhanced by M2 muscarinic Ach receptors (mAchR) expressed on the NSC surface and on their neural progeny. Moreover, atropine, an mAchR antagonist, blocks the enhancement and inhibits the subsequent differentiation of NSCs. Further understanding of this neural-nutrition role of Ach might elucidate fetal brain development, the brain's response to injury, and learning and memory.
Ginkgolide B promotes neuronal differentiation through the Wnt/β-catenin pathway in neural stem cells of the postnatal mammalian subventricular zone

Abstract:Chinese herbal medicines (CHMs) have been used to treat human diseases for thousands of years. Among them, Ginkgo biloba is reported to be beneficial to the nervous system and a potential treatment of neurological disorders. Since the presence of adult neural stem cells (NSCs) brings hope that the brain may heal itself, whether the effect of Ginkgo biloba is on NSCs remains elusive. In this study, we found that Ginkgo biloba extract (GBE) and one of its main ingredients, ginkgolide B (GB) promoted cell cycle exit and neuronal differentiation in NSCs derived from the postnatal subventricular zone (SVZ) of the mouse lateral ventricle. Furthermore, the administration of GB increased the nuclear level of β-catenin and activated the canonical Wnt pathway. Knockdown of β-catenin blocked the neurogenic effect of GB, suggesting that GB promotes neuronal differentiation through the Wnt/β-catenin pathway. Thus, our data provide a potential mechanism underlying the therapeutic effect of GBE or GB on brain injuries and neurodegenerative disorders.

Oleanolic Acid Induces Differentiation of Neural Stem Cells to Neurons: an Involvement of Transcription Factor Nkx-2.5

Abstract:Neural stem cells (NSCs) harbor the potential to differentiate into neurons, astrocytes, and oligodendrocytes under normal conditions and/or in response to tissue damage. NSCs open a new way of treatment of the injured central nervous system and neurodegenerative disorders. Thus far, few drugs have been developed for controlling NSC functions. Here, the effect as well as mechanism of oleanolic acid (OA), a pentacyclic triterpenoid, on NSC function was investigated. We found OA significantly inhibited neurosphere formation in a dose-dependent manner and achieved a maximum effect at 10 nM. OA also reduced 5-ethynyl-2′-deoxyuridine (EdU) incorporation into NSCs, which was indicative of inhibited NSC proliferation. Western blotting analysis revealed the protein levels of neuron-specific marker tubulin-βIII (TuJ1) and Mash1 were increased whilst the astrocyte-specific marker glial fibrillary acidic protein (GFAP) decreased. Immunofluorescence analysis showed OA significantly elevated the percentage of TuJ1-positive cells and reduced GFAP-positive cells. Using DNA microarray analysis, 183 genes were differentially regulated by OA. Through transcription factor binding site analyses of the upstream regulatory sequences of these genes, 87 genes were predicted to share a common motif for Nkx-2.5 binding. Finally, small interfering RNA (siRNA) methodology was used to silence Nkx-2.5 expression and found silence of Nkx-2.5 alone did not change the expression of TuJ-1 and the percentage of TuJ-1-positive cells. But in combination of OA treatment and silence of Nkx-2.5, most effects of OA on NSCs were abolished. These results indicated that OA is an effective inducer for NSCs differentiation into neurons at least partially by Nkx-2.5-dependent mechanism.
Including Memory Friction in Single- and Two-State Quantum Dynamics Simulations.

Abstract:We present a simple computational algorithm that allows for the inclusion of memory friction in a quantum dynamics simulation of a small, quantum, primary system coupled to many atoms in the surroundings. We show how including a memory friction operator, F̂, in the primary quantum system's Hamiltonian operator builds memory friction into the dynamics of the primary quantum system. We show that, in the harmonic, semi-classical limit, this friction operator causes the classical phase-space centers of a wavepacket to evolve exactly as if it were a classical particle experiencing memory friction. We also show that this friction operator can be used to include memory friction in the quantum dynamics of an anharmonic primary system. We then generalize the algorithm so that it can be used to treat a primary quantum system that is evolving, non-adiabatically on two coupled potential energy surfaces, i.e., a model that can be used to model H atom transfer, for example. We demonstrate this approach's computational ease and flexibility by showing numerical results for both harmonic and anharmonic primary quantum systems in the single surface case. Finally, we present numerical results for a model of non-adiabatic H atom transfer between a reactant and product state that includes memory friction on one or both of the non-adiabatic potential energy surfaces and uncover some interesting dynamical effects of non-memory friction on the H atom transfer process.
Inducible motor neuron differentiation of human induced pluripotent stem cells in vivo

Abstract:Generation of hiPSCs with multi‐gene modifications, including motor neuron‐specific transcription factors Ngn2, Isl1, and Lhx3 controlled by Tet‐On system and one anti‐apoptosis gene Bcl‐xL. The genetically modified hiPSCs survived and directionally differentiated into motor neurons after transplantation in vivo. The genetically modified hiPSCs partially converted into neural progenitors, serving as a spare cell resource. Objectives Transplantation of neural progenitor cells (NPCs) derived from human‐induced pluripotent stem cells (hiPSCs) is one of the promising treatment strategies for motor neuron diseases (MNDs). However, the inefficiency in committed differentiation of NPCs in vivo limits its application. Here, we tried to establish a potential therapeutic strategy for MNDs by in vivo directional differentiation of hiPSCs engineered with motor neuron (MN) specific transcription factors and Tet‐On system. Materials and Methods We engineered hiPSCs with three MN‐specific transcription factors and Tet‐On system. The engineered cells were directly transplanted into immunodeficient mice through subcutaneous, intra‐spinal cord and intracerebroventricular injections. Following doxycycline (Dox) induction, teratoma formation, and motor MN differentiation were evaluated. Results We generated genetically engineered hiPSCs, in which the expression of Ngn2, Isl1, and Lhx3 was controlled by a drug‐inducible transgenic system. These cells showed normal pluripotency and proliferative capacity, and were able to directionally differentiate into mature motor neurons (MNs) and NPCs with high efficiency in spinal cords and cerebral lateral ventricles under the induction of Dox. The grafts showed long‐term survival in the recipient mice without formation of teratoma. Conclusions The induced mature MNs and NPCs were expected to replace the damaged endogenous MNs directly, and play a role of de novo stem cell stock for long‐term neuron damage repair, respectively. Therefore, in vivo directional differentiation of the hiPSCs engineered with MN‐specific transcription factors and Tet‐On system via Dox induction could be a potential therapeutic strategy for MNDs with high efficacy and safety.

Abstract:We varied the promoter strength of xylose reductase (XR) gene and the copy number of xylulokinase (XK) gene to determine how XR and XK activities affect the xylose-fermenting abilities of recombinant Saccharomyces cerevisiae expressing xylitol dehydrogenase (XDH). The most enhanced ethanol yield and lowered xylitol yield occurred in strain I-PGK/AUR, which has high activity of both XR and XDH and moderate XK activity.
Enhanced yield of cholinergic neurons from induced pluripotent stem cells (iPSC): A two-step induction protocol

Abstract:Background: Cholinergic neurons, a type of neurons found in central nervous system, play a vital role in muscle movement and activities. Cholinergic neurons degeneration is the main pathological symptom of neurodegenerative diseases. Among a variety of stem cells, iPSCs have emerged as a promising candidate for transplantation to improve the repair of neuronal lesion sites. However, the establishment of an appropriate induction method to yield large numbers of cholinergic neurons has yet to be determined. Here, we studied the differentiation potential of iPSCs to generate cholinergic neurons by developing a new optimized differentiation protocol. Methods: The iPSCs were harvested on 6-well matrigel-coated plate and incubated with serum‑free DMEM/F12 with 2 % B27 supplement, 20 ng/ml the basic fibroblast growth factor and 20 ng/ml epidermal growth factor for 48 hours. Then, the pre-induced cells were treated in neuronal induction medium supplemented with all-trans retinoic acid, sonic hedgehog, 100 ng/ml glial-derived neurotrophic factor and 200 ng/ml brain-derived neurotrophic factor for 7 days. Cell viability during induction stages was tested by MTT assay. Differentiated cells were evaluated with crystal violet staining, immunocytochemistry and real‑time PCR. Results: Our results showed that the survival rate of iPSCs leveled out and was similar to that in the control group following the differentiation process. Immunochemistry results revealed that the expression of ChAT was observed in cells in both pre‑induction and induction stages with a significantly higher expression level at the induction stage as compared to the pre-induction stage. However, none of these markers was expressed in the iPSCs. Cresyl violet staining confirmed the neuronal phenotype of differentiated cells. The induction group significantly expressed the higher levels of Islet1, Olig2 and HB9, whereas pluripotency markers including those of Oct4 and Nestin plunged. Conclusion: Our investigation represents a highly efficient protocol for iPSCs differentiation toward cholinergic neurons which could be used for further preclinical transplantation studies (Tab. 1, Fig. 5, Ref. 35). Abstract:Neural stem cells(NSCs) is one specific type of multipotential stem cells that have the ability to proliferate for a long time,to differentiate into multiple lineages,including neurons,astrocytes and oligodendrocytes,and to migrate toward damaged sites.In all mammalian species,these NSCs mainly exist in most regions of embryonic brain and at least two regions of the adult brain,the subgranular zone(SGZ) of the hippocampal dentate gyrus,and the subventricular zone(SVZ) of the lateral ventricle wall.Because of their properties,NSCs are currently considered to be promising components of cell-replacement strategies aimed at treating Parkinson's disease(PD).There are two therapeutic channels,exogenous and endogenous.For the former,cell is transplanted directly or as a gene carrier for cell replacement and gene therapy.For the latter,neurogenesis is induced by regulating growth factors and cytokines.Exogenous and endogenous NSCs undergo proliferation,migration,and differentiation into dopaminergic neuron in brain,and improve the motor function in animal models of PD.Moreover,the both channels are synergistic.The transplantation of exogenous NSCs can enhance the response of endogenous NSCs in PD models,and the both NSCs are correlated with a host microenvironment which supports to repair and protect damaged neuron.
Secretome of Undifferentiated Neural Progenitor Cells Induces Histological and Motor Improvements in a Rat Model of Parkinson's Disease

Abstract:Abstract Parkinson's disease (PD) is a progressive neurodegenerative movement disorder that results from the death of dopamine (DA) neurons. Over recent years, differentiated or undifferentiated neural stem cells (NSCs) transplantation has been widely used as a means of cell replacement therapy. However, compelling evidence has brought attention to the array of bioactive molecules produced by stem cells, defined as secretome. As described in the literature, other cell populations have a high-neurotrophic activity, but little is known about NSCs. Moreover, the exploration of the stem cell secretome is only in its initial stages, particularly as applied to neurodegenerative diseases. Thus, we have characterized the secretome of human neural progenitor cells (hNPCs) through proteomic analysis and investigated its effects in a 6-hydroxidopamine (6-OHDA) rat model of PD in comparison with undifferentiated hNPCs transplantation. Results revealed that the injection of hNPCs secretome potentiated the histological recovery of DA neurons when compared to the untreated group 6-OHDA and those transplanted with cells (hNPCs), thereby supporting the functional motor amelioration of 6-OHDA PD animals. Additionally, hNPCs secretome proteomic characterization has revealed that these cells have the capacity to secrete a wide range of important molecules with neuroregulatory actions, which are most likely support the effects observed. Overall, we have concluded that the use of hNPCs secretome partially modulate DA neurons cell survival and ameliorate PD animals’ motor deficits, disclosing improved results when compared to cell transplantation approaches, indicating that the secretome itself could represent a route for new therapeutic options for PD regenerative medicine.

Abstract:PURPOSE This study compared the effectiveness of a CD-ROM self-study program with a workshop and scoring videotapes as methods for learning to score the Test of Infant Motor Performance. METHODS Twenty-three therapists participated. Subjects in the videotape group (N = 11) were trained by attending a four-hour workshop and scoring 14 training videotapes, whereas subjects in the CD-ROM group (N = 12) used a CD-ROM self-study program. All subjects scored four videotapes for rater reliability analysis after completing the training procedures. Student t tests were used to compare the means of the two groups on three reliability variables (percentage of aberrant ratings, rater consistency, and rater severity) obtained from Rasch analysis and to compare average time spent on learning. RESULTS Seven therapists (58.3%) in the CD-ROM group and six (54.5%) in the videotape group met the preestablished criteria for achieving satisfactory rater reliability. No statistically significant difference between learning methods was found in percentage of aberrant ratings or rater consistency, but raters in the videotape group were more lenient in scoring than those in the CD-ROM group. The CD-ROM group spent significantly less time learning than the videotape group. CONCLUSIONS Therapists can learn how to score the Test of Infant Motor Performance by using the CD-ROM self-study program in about half the time and with similar reliability when compared with a workshop and videotape practice training procedure. About 40% of therapists will need more training to attain acceptable rater reliability.

Abstract:Midbrain dopaminergic (mDA) neurons play a critical role in regulating postural reflexes and movement as well as modulating psychological processes. Dysfunction or degeneration of mDA neurons is involved in a number of neurological disorders including Parkinson's disease. Availability of large quantities of human mDA neurons would greatly enhance our ability to reveal pathological processes underlying mDA neuron degeneration and to identify treatments for these neurological conditions. Human pluripotent stem cells (PSCs), including embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs), provide an unlimited source for mDA neurons. Here we describe a chemically defined protocol for mDA neuron differentiation. PSCs are first converted to neuroepithelia in a chemically defined medium without any growth factors, followed by patterning the neuroepithelia to midbrain progenitors with fibroblast growth factor 8 (FGF8) and sonic hedgehog (SHH) and subsequent differentiating to functional mDA neurons. This protocol typically yields about half of the neuronal population being mDA neurons, determined by expression of mDA markers, electrophysiological recordings, and the ability to reverse functional deficit in a rat model of Parkinson's disease.
