Abstract:In the present work, a disposable microextraction device with a polyamide 6 nano-fibrous supported liquid membrane (SLM) is employed for the pretreatment of minute volumes of biological fluids. The device is placed in a sample vial for an at-line coupling to a commercial capillary electrophoresis instrument with UV-Vis detection (CE-UV) and injections are performed fully automatically from the free acceptor solution above the SLM with no contact between the capillary and the membrane. Up to 4-fold enrichment of model basic (nortriptyline, haloperidol, loperamide, and papaverine) and acidic (ibuprofen, naproxen, ketoprofen, and diclofenac) drugs is achieved by optimizing the ratio of the donor to the acceptor solution volumes (16 to 4 μL, respectively). The actual setup enables SLM extractions from less than a drop of sample and is suitable for pretreatment of scarce human body fluids. Two unique methods are reported for efficient clean-up and enrichment of the basic and acidic drugs from capillary blood (formed as dried blood spot), serum, and urine samples, which enable their determination at therapeutic and/or toxic levels. The hyphenation of the SLM extraction with CE-UV analysis provides good repeatability (RSD, 2.4–14.9%), linearity (r2, 0.988–1.000), sensitivity (LOD, 0.017–0.22 mg L−1), and extraction recovery (ER, 20–106%) at short extraction times (10 min) and with minimum consumption of samples and reagents.

Abstract:This contribution describes properties and utilization of free liquid membranes (FLMs) in micro-electromembrane extraction (μ-EME) of analytes from samples with complex matrices. An FLM was formed as a plug of a selected organic solvent, 1-ethyl-2-nitrobenezene (ENB) or 2-nitrophenyloctyl ether, in a narrow bore polymeric tubing and was sandwiched between a plug of aqueous donor and aqueous acceptor solution. The FLM acted as a phase interface that enabled selective transfer of analytes from donor into acceptor solution. Acceptor solution after μ-EME was analysed by capillary electrophoresis (CE). Fundamental characteristics of FLMs were depicted and discussed by presenting experimental data on their performance for various basic operational parameters, such as composition and volume of donor/acceptor solution, applied extraction voltage, thickness of FLM and extraction time. Positively charged basic drugs (nortriptyline, haloperidol and loperamide) and their solutions in water, urine and blood serum served as model samples. It was shown that FLMs may offer fast, efficient and selective pretreatment of crude biological samples providing that basic operational parameters of μ-EME are set properly. At optimised conditions, basic drugs in 1.5μL of a biological sample were transferred across 1.5μL of FLM (ENB) into 1.5μL of acceptor solution in about 5min at an extraction voltage of 100V. Repeatability values of μ-EMEs and CE-UV analyses of the three basic drugs were better than 7.7% for peak areas, recoveries ranged between 19 and 52% and linear relationship was obtained for analytical signal vs. concentration in 1-50mgL(-1) range (r(2) better than 0.996). Limits of detection, defined as 3×S/N, were below 1mgL(-1) for all examined matrices.

Abstract:A two-phase micro-electromembrane extraction (μ-EME) using a floating drop of an organic solvent was presented for rapid and efficient pretreatment of complex biological samples. The μ-EME system consisted of a glass vial containing aqueous sample (donor solution) and a small drop of a water-immiscible organic solvent (4-nitrocumene), which was floating on the surface of the aqueous solution in form of a free liquid membrane (FLM). The vial geometry and the optimized volume ratios of the donor and the FLM ensured a stable position of the FLM in the center of the vial during μ-EME, and one electrode of a d.c. power supply was inserted directly into the FLM while the other electrode was placed into the aqueous sample. The active surface area of the floating drop FLM contacting the sample was considerably larger in comparison to formerly reported μ-EME formats employing FLMs and resulted in a faster and a more efficient transfer of target analytes from the sample to the FLM. Four basic drugs (nortriptyline, papaverine, loperamide, and haloperidol) were selected as model analytes and were extracted from physiological solution, human urine, and dried blood spot samples. At the optimized μ-EME conditions (250 V, 15 min, 300 rpm, acidic donor) and the optimized ratio of the sample to the FLM volume (500:14 μL), extraction recoveries between 49 and 100% and enrichment factors up to 35.7 were achieved. Quantitative analyses of the basic drugs in the resulting FLMs (diluted with methanol) were performed by capillary electrophoresis with ultraviolet detection and demonstrated excellent repeatability (RSD ≤ 4.9%) and linearity (r2 ≥ 0.9997), and low limits of detection (5–28 ng/mL) of the method.

Abstract:Manual handling of microliter volumes of samples and reagents is usually prone to errors and may have direct consequence on the overall performance of microextraction process. Direct connection of a syringe pump and a disposable microextraction unit using flexible polymeric tubing was employed for semi-automated liquid handling in micro-electromembrane extraction (μ-EME). A three-phase μ-EME system was formed by consecutive withdrawal of microliter volumes of donor solution, free liquid membrane (FLM) and acceptor solution into the unit. Excellent repeatability and accuracy of the withdrawal sequence was achieved for solution volumes typically used in μ-EME (1-5 μL) as well as excellent correlation between the initially withdrawn and the finally collected solution volumes. μ-EMEs were initiated by application of d.c. electric potential to the terminal aqueous solutions and specific μ-EME parameters were optimized in order to ensure complete transfer of model analytes from donor to acceptor solution. Exhaustive μ-EMEs of three basic drugs, nortriptyline, papaverine and haloperidol, were achieved from 1.3 μL of acidified donor solution (10 mM HCl) across 2.5 μL of FLM (1-ethyl-2-nitrobenzene) into 1.3 μL of acidified acceptor solution (25 mM HCl) in 10 min at 150 V. The three drugs were also exhaustively extracted from salt- and protein-containing standard solutions, human urine and human plasma with extraction recoveries ranging from 79 to 102%. Resulting acceptor solutions were analysed by capillary electrophoresis with ultraviolet detection (CE-UV) and the μ-EME-CE-UV method was characterized by good linearity (coefficients of determination ≥ 0.992), high repeatability (RSD values ≤ 6.5%) and limits of detection ≤ 0.15 mg/L.

Abstract:PURPOSE To investigate the effects of the application of mitomycin C (MMC) after photorefractive keratectomy (PRK) on the ciliary body and intraocular pressure (IOP) in an experimental model. METHODS Forty eyes of 20 rabbits underwent PRK to correct -8.00 diopters (D) in a 5-mm optical zone. Sterile surgical sponges (5 mm) soaked with 0.02% MMC were applied to corneas with the epithelium removed. Eyes were randomly chosen to receive MMC for 2 minutes (study group), and sponges soaked with balanced salt solution were applied on the contralateral eye (control group) for 2 minutes. Histological evaluation of the ciliary body was performed in a masked fashion by light and transmission electron microscopy. RESULTS Histological examination of the morphology of the ciliary body structures revealed no differences between the two groups. Tonometric measurements of the IOP remained stable during the follow-up period with no significant changes (P=.75). CONCLUSIONS Adjuvant application of MMC after PRK does not appear to induce alterations on the ciliary body or changes in IOP measurements.
Polyaniline-polycaprolactone electrospun nanofibrous mat: new polymeric support with anion exchange characteristic for immobilizing liquid membrane in efficient on-chip electromembrane extraction of polar acidic drugs

Abstract:The potential of application of an electrospun nanofiber sheet as new polymeric support for immobilizing the liquid membrane, instead of a common commercial polypropylene sheet, in on-chip electromembrane extraction (EME) of some acidic polar drugs followed by HPLC with ultraviolet detection is presented. The nanofiber sheet was prepared by electrospinning a mixture of polycaprolactone and polyaniline. The successful synthesis of the electrospun nanofiber sheet was confirmed by field emission-scanning electron microscopy, energy-dispersive X-ray spectroscopy, elemental mapping, and atomic force microscopy. Several parameters affecting the efficiency of the microextraction method, including pHs of the donor and acceptor phases, applied voltage, sample flow rate, phosphate content of the acceptor phase, and sample volume, were investigated and optimized. After optimization, the linearity range of 0.5-250.0 µg L-1 and detection limits of 0.2-1.0 µg L-1 were obtained for the analytes. The extraction recovery values and preconcentration factors were 10.7-55.3% and 16-83, respectively. The presence of polyaniline in the composition of the nanofibers significantly improved the extraction efficiency of the polar acidic drugs due to providing the possibility of various interactions with the target analytes such as hydrogen bonding, π-stacking, and anion exchange. The obtained results demonstrate the excellent efficiency of the synthesized electrospun nanofibrous mat as a novel support membrane for immobilizing 1-octanol and as an interactive substrate for electromembrane extraction of acidic polar drugs. Eventually, the proposed on-chip EME method exhibits acceptable precision (relative standard deviations less than 9.7% (n = 3)) and good accuracy (86-112%) for determining the target analytes in the plasma samples.
Capillary electrophoresis of neurotransmitters using in-line solid-phase extraction and preconcentration using a methacrylate-based weak cation-exchange monolithic stationary phase and a pH step gradient

Abstract:A novel approach for in-line solid-phase extraction capillary electrophoresis (SPE-CE) for basic analytes was developed. The method is based on the use of a weak cation-exchange monolith synthesised in situ in the front end of the CE capillary via photoinitiated polymerization to form poly(methacrylic acid-co-ethylene glycol dimethacrylate), which was used to create the SPE phase in-line with the CE separation capillary. The monolithic SPE material exhibited a surface area of 23.1 m2/g and a capacity of 403 nM for dopamine. Adsorption of the analytes as protonated, cationic species onto the SPE phase was achieved using an electrolyte of 6 mM phosphate and 12 mM sodium ion, buffered at pH 7.0, which is above the pKa of the monolith but below the pKa of the analytes. Elution of the analytes from the SPE phase was achieved using an electrolyte with a pH below that of the pKa of the monolith, namely 12 mM phosphate and 12 mM sodium ion, buffered at pH 3.0. Due to the discontinuous electrolyte combination, analytes were simultaneously eluted and focused as the electrophoretically mobilised pH step boundary moved through the SPE monolith, after which the analytes were separated by conventional CZE in the remainder of the capillary. Quantitative extraction from a solution of 0.5 microg/ml dopamine and epinephrine was achieved when flushing up to 15 column volumes of sample through the capillary. The limits of detection (S/N=3) for dopamine and epinephrine were 3.7 and 4.3 ng/ml, and this method provided a sensitivity enhancement for dopamine of 462 times compared to CZE using hydrodynamic injection. The developed method was used to preconcentrate a test mixture of neurotransmitters comprising dopamine, epinephrine, 5-hydroxytryptamine, metanephrine and also histamine. The applicability of this approach to real life samples was demonstrated by using a urine sample from a healthy person to detect dopamine at sub-ppm levels.
On-chip electromembrane extraction of some polar acidic drugs from plasma samples by the development of an active and efficient polymeric support for liquid membrane based on electrospinning process

Abstract:Electromembrane extraction (EME), despite its high performance in the extraction of highly polar basic analytes, has challenges in extracting polar acidic compounds. This is the result of a lack of access to the suitable supported liquid membrane (SLM) for this group of analytes. Therefore, it would be valuable to provide a suitable solution for this problem. Accordingly, in the present study, a new method based on on-chip EME followed by HPLC with UV detection was developed for the extraction and determination of some polar acidic drugs in order to provide high extraction efficiency. Here, a new polymeric sheet was introduced as a support for SLM immobilization, which is not only used to impregnate 1-octanol as SLM but also enhances extraction recovery by exerting effective interactions with target acidic analytes. The polymeric sheet was composed of nanofibers prepared by electrospinning polycaprolactone blended with a composite of graphene oxide and aluminum polycations. Encapsulation of the composite in polycaprolactone nanofibers improved the extraction efficiency of polar acidic compounds by creating additional interactions with the target analytes, including hydrogen bonding, dipole-dipole, π-π stacking, and anion exchange process. The electrospun nanofiber-based sheet was characterized by FE-SEM, EDX, elemental mapping, TEM, and AFM. The extraction parameters were further optimized with an orthogonal-rotatable central composite design (CCD). Applying CCD determined optimal conditions by minimum experiments, and interactions between the parameters were clarified. Under optimized conditions, the proposed method provided extraction recoveries from 36.5 to 64.1%, relative standard deviations less than 5.7% (n = 4), and detection limits of 0.3-0.5 μg L-1. Furthermore, the proposed method was successfully used for the determination of target analytes in plasma samples, providing good accuracy (87-110%) and precision (3.2-8.8%).
Combination of Electromembrane Extraction and Liquid-Phase Microextraction in a Single Step: Simultaneous Group Separation of Acidic and Basic Drugs

Abstract:Electromembrane extraction (EME) and liquid-phase microextraction (LPME) were combined in a single step for the first time to realize simultaneous and clear group separation of basic and acidic drugs. Using 2-nitrophenyl octyl ether as the supported liquid membrane (SLM) for EME and dihexyl ether as the SLM for LPME, basic and acidic drugs were extracted and separated simultaneously from a low pH sample by EME and LPME, respectively. After 15 min of extraction, basic drugs (citalopram and sertraline) were exhaustively extracted, whereas the recoveries for acidic drugs (ketoprofen and ibuprofen) were in the range of 76%-86%. Longer extraction time provided higher recoveries for the acidic drugs, but this somewhat deteriorated the group separation. Matrices effects from the coexisting acidic drugs/basic drugs were tested, and we observed that simultaneous EME/LPME was not affected by coexisting drugs at high concentration. This approach was further investigated from human plasma. Extraction recoveries were strongly dependent on dilution of plasma with buffer and on extraction time. Finally, this simultaneous EME/LPME approach was evaluated in combination with liquid chromatography (LC)-MS. The linearity ranges for the basic and acidic drugs were 10-600 ng/mL and 1-60 μg/mL, respectively, with R(2) > 0.997 for all analytes. The repeatability at three different levels for all analytes was less than 15%. The limits of quantification (LOQ, S/N = 10) were found to be 4.0-6.3 ng/mL and 0.6-0.9 μg/mL for basic and acidic drugs, respectively. Simultaneous EME/LPME enabled efficient group separation of basic and acidic analytes under optimum experimental conditions for both EME and LPME.
Direct Analysis of Free Aqueous and Organic Operational Solutions as a Tool for Understanding Fundamental Principles of Electromembrane Extraction

Abstract:Aqueous and organic phases in microelectromembrane extraction (μ-EME) were formed as adjacent plugs of free immiscible solutions in narrow-bore polymeric tubing, and each single phase was recovered and analyzed after μ-EME. A three-phase μ-EME setup was employed for investigation of time-dependent distribution of model basic drugs among aqueous and organic phases. Exact concentrations of nortriptyline and papaverine in donor solution, acceptor solution, and free liquid membrane (FLM) were determined by capillary electrophoresis with ultraviolet detection (CE-UV). At typical μ-EME conditions (acceptor, 1 μL of 25 mM HCl; FLM, 1 μL of 4-nitrocumene; donor, 1 μL of basic drugs in 10 mM HCl; and extraction potential, 250 V), experimentally determined distribution of the drugs confirmed the kinetic model for electrically mediated transfer of charged analytes. Rapid depletion of the drugs from donor solution (0-180 s) and rapid saturation of FLM with the drugs (15-60 s) were followed by gradual transfer of the drugs across FLM and gradual liberation into acceptor solution (30-240 s). Exhaustive transfer of the drugs from donor to acceptor solution was obtained in 15 min. A good correlation between the analytes' distribution and μ-EME electric currents was observed; the currents increased during drugs' transfer across FLM, were concentration dependent, and demonstrated transfer of the drugs across FLM in their ionized forms. Proper understanding of the fundamental principles of μ-EME transfer enabled further fine-tuning of the μ-EME process. Transfer of the drugs across FLM was controlled by optimizing the composition and pH of acceptor solution, and quantitative fractionation of nortriptyline into aqueous acceptor (96%) and of papaverine into organic FLM (95%) was achieved based on their different pKa values. μ-EME fractionation of the two drugs was compatible with raw human urine and excellent repeatability (RSD ≤ 3.9%), linearity (r2 ≥ 0.9989), and limits of detection (≤ 0.15 μg/mL) were achieved for μ-EME-CE-UV of the drugs in standard solutions and urine samples.
Microdialysis Hollow Fiber As a Macromolecule Trap for On-Line Coupling of Solid Phase Microextraction and Capillary Electrophoresis

Abstract:On-line coupling of solid phase microextraction (SPME) and capillary electrophoresis (CE) is highly desirable due to the apparent advantages of the two techniques particularly in the context of microanalysis. However, the hyphenation is a significant challenge, because of band broadening and analyte carryover caused by the slow kinetics of analyte desorption in liquid phase. A novel strategy was presented in this study to overcome these problems. Analytes desorbed from an SPME fiber, which was held by an adapter, were first transferred by electrophoretic migration into a short piece of microdialysis hollow fiber, which was located at the inlet of a CE system. Analytes with molecular weights greater than the molecular weight cut-off of the microdialysis material were trapped in the microdialysis hollow fiber due to the dialysis effect. Then, under another electric field with different electrode polarity, the analytes trapped in the microdialysis hollow fiber migrated into the separation capillary and were separated. In the coupling approach, the microdialysis hollow fiber functioned as a macromolecule trap and a sample pre-concentrator as well. Band broadening was eliminated because the initial sample volume was very small (at nL level). Meanwhile, analyte carryover was eliminated because the desorption time could be as long as needed. Coupling of SPME with CE including two modes, capillary zone electrophoresis (CZE) and capillary isoelectric focusing (CIEF), was successfully demonstrated with proteins as test analytes. High efficiency and high resolution were obtained. The detection limits with UV absorbance whole-column imaging detection were 3.0 x 10(-7) and 3.0 x 10(-8) M (S/N = 3) for beta-lactoglobulin A and ovalbumin, respectively.
Rapid and simple pretreatment of human body fluids using electromembrane extraction across supported liquid membrane for capillary electrophoretic determination of lithium

Abstract:Electromembrane extraction was used for simultaneous sample cleanup and preconcentration of lithium from untreated human body fluids. The sample of a body fluid was diluted 100 times with 0.5 mM Tris solution and lithium was extracted by electromigration through a supported liquid membrane composed of 1-octanol into 100 mM acetic acid acceptor solution. Matrix compounds, such as proteins, red blood cells, and other high-molecular-weight compounds were efficiently retained on the supported liquid membrane. The liquid membrane was anchored in pores of a short segment of a polypropylene hollow fiber, which represented a low cost, single use, disposable extraction unit and was discarded after each use. Acceptor solutions were analyzed using capillary electrophoresis with capacitively coupled contactless conductivity detection (CE-C(4) D) and baseline separation of lithium was achieved in a background electrolyte solution consisting of 18 mM L-histidine and 40 mM acetic acid at pH 4.6. Repeatability of the electromembrane extraction-CE-C(4) D method was evaluated for the determination of lithium in standard solutions and real samples and was better than 0.6 and 8.2% for migration times and peak areas, respectively. The concentration limit of detection of 9 nM was achieved. The developed method was applied to the determination of lithium in urine, blood serum, blood plasma, and whole blood at both endogenous and therapeutic concentration levels.

Continuous flow membrane microextraction as a clean method for detecting codeine and papaverine in biological samples using HPLC-UV

Abstract:In this study, continuous-flow membrane microextraction (CFMME) was connected online with a high-performance liquid chromatography-UV detector (HPLC-UV) for the pre-concentration and high clean-up of papaverine and codeine in biological samples. The extraction cell was designed with donor and acceptor chambers separated by a sheet membrane. By adjusting the pH of the donor phase (10 mL, pH 11), the analyte molecules were extracted into the supported liquid membrane (SLM) (15 μL of 1-octanol). The donor solution was circulated through the donor chamber using a peristaltic pump and magnetically agitated by a bar stirrer placed near the membrane, enhancing the diffusion and convection flow of the targeted drugs from the donor solution to the SLM. Subsequently, by adjusting the acceptor solution to an acidic pH of 2 (100 μL), the drugs in ionic form were reversely extracted into the acceptor phase. The procedure exhibited desirable relative standard deviation (RSD%) of less than 3.70%, linear ranges of 7-600 ng mL-1 for codeine and 2.0-600 ng mL-1 for papaverine, and limits of detection of 2.0 ng mL-1 for codeine and 0.6 ng mL-1 for papaverine. The design of the extraction cell significantly improved the performance for determining targeted drugs in complex matrices such as plasma and urine.

Abstract:Coupling capillary electrophoresis (CE) to mass spectrometry (MS) is a powerful strategy to leverage a high separation efficiency with structural identification. Traditional CE-MS interfacing relies upon voltage to drive this process. Additionally, sheathless interfacing requires that the electrophoresis generates a sufficient volumetric flow to sustain the ionization process. Vibrating sharp-edge spray ionization (VSSI) is a new method to interface capillary electrophoresis to mass analyzers. In contrast to traditional interfacing, VSSI is voltage-free, making it straightforward for CE and MS. New nanoflow sheath CE-VSSI-MS is introduced in this work to reduce the reliance on the separation flow rate to facilitate the transfer of analyte to the MS. The nanoflow sheath VSSI spray ionization functions from 400 to 900 nL/min. Using the new nanoflow sheath reported here, volumetric flow rate through the separation capillary is less critical, allowing the use of a small (i.e., 20 to 25 μm) inner diameter separation capillary and enabling the use of higher separation voltages and faster analysis. Moreover, the use of a nanoflow sheath enables greater flexibility in the separation conditions. The nanoflow sheath is operated using aqueous solutions in the background electrolyte and in the sheath, demonstrating the separation can be performed under normal and reversed polarity in the presence or absence of electroosmotic flow. This includes the use of a wider pH range as well. The versatility of nanoflow sheath CE-VSSI-MS is demonstrated by separating cationic, anionic, and zwitterionic molecules under a variety of separation conditions. The detection sensitivity observed with nanoflow sheath CE-VSSI-MS is comparable to that obtained with sheathless CE-VSSI-MS as well as CE-MS separations with electrospray ionization interfacing. A bare fused silica capillary is used to separate cationic β-blockers with a near-neutral background electrolyte at concentrations ranging from 1.0 nM to 1.0 μM. Under acidic conditions, 13 amino acids are separated with normal polarity at a concentration ranging from 0.25 to 5 μM. Finally, separations of anionic compounds are demonstrated using reversed polarity under conditions of suppressed electroosmotic flow through the use of a semipermanent surface coating. With a near-neutral separation electrolyte, anionic nonsteroidal anti-inflammatory drugs are detected over a concentration range of 0.1 to 5.0 μM.
A demountable nanoflow nebulizer for sheathless interfacing nano-high performance liquid chromatography with inductively coupled plasma mass spectrometry

Abstract:For direct analyses of micro-samples and the hyphenation with nanoflow separation techniques, a low-cost demountable nanoflow nebulizer was fabricated for inductively coupled plasma mass spectrometry (ICPMS). A small-bore capillary (20 mm) with a tapered tip served as the sample capillary of the nebulizer. The gas orifice diameter, the internal diameter and the wall thickness at the tip of the sample capillary were 160, 20, and 5 mm, respectively, which ensured stable generation of aerosol by the nebulizer at nanoflow rates down to 50 nL min(-1) and an ultra-low aspiration rate (80 nL min(-1)). Narrower aerosol size distribution and higher analyte transport efficiency (nearly 100%) were obtained with the proposed nebulizer than with the previous demountable capillary microflow nebulizer. The sensitivity and detection limit of the proposed nebulizer at 500 nL min(-1) were about one third of those obtained by using the demountable capillary microflow nebulizer at 5 mL min(-1), whereas the precision of the former at 500 nL min(-1) was even to some extent superior over that obtained with the latter at 5 mL min(-1). Furthermore, the ICP-MS instrument could tolerate direct introduction of 100% organic solvents by using the developed nebulizer operating at nanoflow rates. It was utilized to sheathless interface nano-high performance liquid chromatography with ICP-MS for arsenic speciation analysis with good accuracy and precision, proving its strong robustness and good suitability.
Analysis of basic drugs in biological samples using dynamic single-interface hollow fiber liquid-phase microextraction combined with fast electromembrane extraction

Abstract:In this study, a combination of single-interface hollow fiber liquid-phase microextraction and electromembrane extraction was applied for the extraction of some basic drugs from biological samples. The extraction process was followed by the HPLC-UV instrument. In this dynamic system, 1-octanol was impregnated into the pores of hollow fiber wall as an acceptor phase, the sample solution (pH = 12) was pumped into the lumen of hollow fiber by a syringe pump, the extraction efficiency was improved by filling and emptying (25 times) of sample solution (step 1). The extracted analytes in the organic acceptor phase were back-extracted by a fast electromembrane extraction procedure (step 2). Effective parameters on the extraction efficiency of both methods were investigated and optimized. Under optimized conditions (organic acceptor phase: 1-octanol, sample solution volume: 1500 µL, flow rate: 2.0 mL min-1, the number of filling and emptying cycles: 25, voltage: 100 V, back-extraction time: 2 min, and aqueous acceptor phase: 100 mM HCl), the proposed method provided good linearity with determination coefficients ranging from 0.992 to 0.996 over a concentration range of 2.5-1000 ng mL-1. Extraction recoveries were obtained in the range of 65.1 and 88.8%, which resulted in preconcentration factors in the range of 32.6-44.4. The limits of detection were found to be within the range of 0.12-0.36 ng mL-1, while the corresponding repeatability ranged from 3.7 to 9.3% (n = 3). Finally, the optimized method was applied for the quantification of propranolol, diltiazem and, lidocaine in urine and, plasma samples with relative recoveries ranged between 94.1 and 105.4%, indicating the reliability of the method.

Abstract:Background: A novel supported liquid extraction approach using small polymeric nanofibrous discs was demonstrated and applied to the analysis of real river water. Nanofibrous discs were tested to extract model mixture of 9 common water contaminants 4-nitrophenol, various chlorophenols, bisphenol A, permethrin, and fenoxycarb featuring a wide range of log P values (1.9-6.5). Polyacrylonitrile, polyhydroxybutyrate, and polylactic acid nanofibers were selected as adsorptive materials. One-step desorption was performed directly in HPLC vials, to avoid time-consuming evaporation and reconstitution steps. The discs were allowed to sediment to the bottom of the vial before injection into the chromatographic system. Results: Various parameters affecting the extraction efficiency including 1-octanol volume, extraction time, ionic strength, and sample volume were investigated and optimized. Wetting the nanofiber discs with 1-octanol resulted in up to 20-fold increase in enrichment factor when compared to non-wetted polymer counterparts. The highest enrichment factors were observed for analytes with a log P range of 3.3-4.5. Our developed method showed good linearity in the range 20-200 μg/L for all analytes tested. Satisfactory repeatability with RSD <13 % were achieved covering all steps including disc preparation, wetting, extraction/elution, and chromatography analysis, and recoveries ranged from 58.93 to 121.43 %. Significance: This work represents novel simple supported liquid extraction approach using impregnated polymer nanofiber discs. Using only 50 μL 1-octanol, we reduced the organic solvent compared to other extraction methods. There was no need for any plastic cartridge to hold the sorbent and direct in-vial desorption reduced the unnecessary, time-consuming steps and simplified the sample preparation protocol.

Abstract:Electromembrane extraction (EME) is a miniaturized technique in which charged molecules are extracted from biological samples, through a porous membrane, into an acceptor solution. This process occurs in the presence of an electric field that enables increased analyte recovery in short extraction times. Studies are being carried out to establish recommended extraction conditions for different groups of compounds based on their physicochemical properties. In this work, pre-established generic liquid membranes for the extraction of basic compounds were tested using synthetic cathinones within a wide log P range as basic model analytes. First, recommended organic solvents for the chosen analytes were evaluated in terms of recovery. The best candidates were included in an optimization study with other relevant EME conditions (voltage, extraction time, and agitation rate). All analyses were performed using a UPLC-MS/MS with the multiple reaction monitoring mode. 2-Undecanone was chosen as the liquid membrane providing the best extraction efficiency in combination with 40 V, 35 min, and 725 rpm. Validation showed r 2 > 0.99 in the 1–500 ng/mL range, accuracy of ±12%, imprecision of ≤19%, 1 ng/mL as limit of quantitation, 0.1–0.5 ng/mL as limit of detection, matrix effect 97–118%, and recovery 63–110%. Among the generic liquid membranes studied in this work, 2-Undecanone was the best choice for the target basic analytes. High recoveries and low matrix effects were achieved, thus demonstrating the applicability of recommended generic liquid membranes in the 96-well plate setup and with whole blood samples. Our findings contribute to further establishing EME as an actual sample preparation technique for routine analyses that could be further explored in a commercial format.

Abstract:In the present study, a new chip was designed for simultaneous electromembrane extraction (EME) of acidic and basic analytes. The method followed by the HPLC-UV analysis, was used for simultaneous extraction and determination of trace amounts of target analytes with different properties. Ibuprofen (IBU), as an acidic model analyte, and disopyramide (DIS) and lidocaine (LID), as basic model analytes, were selected. The chip was composed of three polymethylmethacrylate (PMMA) plates. The chamber for the sample solution was set in the middle plate. Two other chambers were carved on the other plates and they were acted as two acceptor phases’ reservoirs. Two stainless steel electrodes, which were responsible for applying a DC electrical potential on both sides of the supported liquid membranes (SLMs), were placed in the bottom of acceptor phases chambers and connected to a power supply. The two porous polypropylene membrane sheets, which were impregnated with different organic solvents, were used to separate the plates. Effective parameters such as SLM composition (1-octanol for acidic analyte and NPOE+5%DEHP for basic analytes), pH of donor phase and acceptor phases, applied voltage across SLM, and extraction time were optimized. Under the optimized conditions, the calibration curves were linear with coefficient of determinations (R2) higher than 0.991 in the water, urine, and blood plasma samples. The obtained LODs were less than 5.0 μg L−1 and 1 μg L−1 for the basic and acidic analytes, respectively. The results demonstrated that on-chip EME would be used most effectively for extraction and determination of IBU, DIS, and LID in biofluids.

Abstract:Capsule phase microextraction (CPME) is an efficient bioanalytical technique that streamlines the sample preparation by integrating the filtration and stirring mechanism directly into the device. A novel composite sorbent designed to be selective towards the target analytes consisting of mixed-mode sorbent chemistry synthesized by sol–gel technology is found promising and superior to the conventional C 18 sorbents. Herein we describe the encapsulation of an ionic liquid (IL)/Carbowax 20M-functionalized sol–gel sorbent (sol–gel IL/Carbowax 20 M) in the lumen of porous polypropylene tubes for the capsule phase microextraction of three phosphodiesterase-5 inhibitors namely avanafil, sildenafil, and tadalafil in human serum and urine samples. The CPME device was characterized by Scanning Electron Microscopy (SEM) and Fourier-Transform Infrared Spectroscopy (FT-IR). The experimental parameters of CPME procedure (e.g. sample pH and ionic strength, extraction time, stirring rate, elution solvent and volume) were carefully optimized to achieve the highest possible extraction efficiency for the analytes. Method validation was conducted in terms of precision, linearity, accuracy, matrix effect, lower limits of quantification, and limits of detection (LOD). The method linearity was investigated in the range of 50–1000 ng mL −1 for all analytes while the precision was less than 11.8 % in all cases. For all analytes, the LOD values were 17 ng mL −1 . The IL/CW 20M-functionalized microextraction capsules could be reused at least 25 times both for urine and serum samples. The green character and the applicability of the proposed method were evaluated using the ComplexGAPI and BAGI indexes. The optimized CPME protocol exhibited reduced consumption of organic solvent and generation of waste, cost-effectiveness, and simplicity. Finally, the proposed method was successfully applied to the analysis of sildenafil in human urine after administration of drug-containing formulation.
