Abstract:The blue-green algae Anabaena flos-aquae and Nostoc muscorum may reduce acetylene to ethylene most actively at p O 2 levels below 0.2 atm. High p O 2 levels inhibit acetylene reduction, nitrogen fixation, respiration and 14 CO 2 fixation in A . flos-aquae . The effect is not solely via an inhibition of nitrogenase activity because inhibition of 14 CO 2 fixation by a species of Phormidium which does not fix nitrogen, and by A . flos-aquae grown on combined nitrogen also occurs. The inhibition of acetylene reduction in Anabaena is reversible in short-term experiments, the rate of recovery being rather similar irrespective of the p O 2 level to which the alga was subjected initially. 3(3,4-dichlorophenyl)-1-1-dimethyl urea (DCMU) at a concentration of 3 x 10 -5 M, which completely inhibits oxygen evolution and acetylene reduction but not respiration by aerobically grown Anabaena cultures also inhibits acetylene reduction in Na 2 S-grown cultures. Salicylaldoxime at a concentration of 10 -4 M which partially inhibits electron flow from photosystem II inhibits acetylene reduction to a greater extent under aerobic conditions than in the presence of Na 2 S. The presence of Na 2 S also results in the removal of free oxygen from the medium in the pH range at which Anabaena normally grows. The data suggest that under our conditions (1) the photolysis of water, or photosystem II is essential for the growth of A. flos-aquae in the presence of H 2 S ; (2) H 2 S may provide electrons when reducing power from the photolysis of water is reduced but not inhibited completely; (3) H 2 S prevents an accumulation of oxygen in the medium during photosynthesis. These physiological findings from the laboratory may help to explain why blue-green algae live not only in aerobic environments but also in habitats where reducing conditions may prevail.

Abstract:Shake-flask cultures are widely used for screening of high producing strains. To select suitable strains for production scale, cultivation parameters should be applied that provide optimal growth conditions. A novel method of measuring respiratory activity in shake-flask cultures was employed to analyze Escherichia coli fermentation under laboratory conditions. Our results suggest that the length of fermentation, choice of medium, and aeration do not normally satisfy the requirements for unlimited growth in shake flasks. Using glycerol rather than glucose as a carbon source greatly reduced the accumulation of overflow and fermentative metabolites when oxygen supply was unlimited. A rich buffered medium, Terrific Broth (TB), yielded 5 times more biomass compared to LB medium but also caused oxygen limitation in standard shake-flask cultures at shaking frequencies below 400 rpm. These results were used to optimize the production of benzoylformate decarboxylase from Pseudomonas putida in E. coli SG13009, resulting in a 10-fold increase in volumetric enzyme production. This example demonstrates how variation of medium composition and oxygen supply can be evaluated by the measurement of the respiratory activity. This can help to efficiently optimize screening conditions for E. coli.

Evolution and Inhibition of Carbon Dioxide in the High Cell Density Culture of Ralstonia Eutropha

Abstract:More CO2 would be evolved with the high cell density during fermentation processes. It is found that the dissolved carbon dioxide will affects the growth of many microorganisms. The evolution of carbon dioxide in fed-batch high cell density culture of Ralstonia eutropha was determined in a 5-L fermentor, and the maximum CO2 concentration of 15% in effluent gas was detected. CO2 pulsating additions with different pulse durations (3 or 6 hours) and concentrations (10%, 22%) were employed to study the CO2 inhibition on cell growth. It is found that the inhibitory effect of CO2 on cell growth is stronger with the increase of CO2 pulse duration and/or CO2 concentration. In the case of 6-hour 22% CO2 pulse, the final cell concentration (DCW) decreased 31% as compared to the results of without CO2 addition. The amount of CO2 evolved mainly dependents on the cell culture concentration, so it is necessary to pay more attention on the large-scale high cell density culture for its occurrence of CO2 inhibition.
Interactive effects of CO2, temperature, and nitrate limitation on the growth and physiology of strain CCMP 1334 of the marine cyanobacterium Synechococcus (Cyanophyceae)

Abstract:The marine cyanobacterium Synecococcus sp. (CCMP 1334) was grown in a continuous culture system on a 12:12 h light:dark cycle at all combinations of low and high pCO2 (400 and 1000 ppmv, respectively), nutrient availability (nitrate‐limited and nutrient‐replete conditions), and temperatures of 21, 24, 28, 32, and 35°C. The maximum nutrient‐replete growth rate was ~1.15 day−1 at 32–35°C. Median nutrient‐replete growth rates were higher at 1000 ppmv than at 400 ppmv pCO2 at all temperatures. Carbon:nitrogen ratios were independent of pCO2 at a fixed relative growth rate (i.e., growth rate ÷ nutrient‐replete growth rate) but decreased with increasing temperature. Carbon:chlorophyll a ratios were decreased monotonically with increasing temperature and were higher under nitrate‐limited than nutrient‐replete conditions. Ratios of phycoerythrin to chlorophyll a were independent of growth conditions. Productivity indices were independent of temperature and nutrient limitation but were consistently higher at 1000 ppmv than 400 ppmv pCO2. Both growth rates and dark respiration rates were positively correlated with temperature, and the associated Q10 values were 2.2 and 2.3, respectively. A model of phytoplankton growth in which cellular carbon is allocated to structure, storage, or the light or dark reactions of photosynthesis accounted for the general patterns of cell composition and growth rate. This strain of Synechococcus appears well suited to changes in environmental conditions that are expected as the climate warms in response to anthropogenic emissions of CO2.

Effect of diethylsulphoxide on growth, survival and ion exchange of Escherichia coli

Abstract:Aims: To study the effect of diethylsulphoxide (DESO) on Escherichia coli growth, survival and ionic exchange in comparison with dimethylsulphoxide (DMSO). Methods and results: Bacterial survival was estimated by counting colony-forming units and by the most probable number (five-tube) technique; the K+ and H+ transport and H(2) formation were determined electrochemically. Diethylsulphoxide at concentrations between 0.01 and 0.5% (w/v) stimulated and above 5% decreased the anaerobic growth rate and survival. 2H+ : K+ exchange and H(2) formation were lost at 5% DESO. At 0.05% DESO the kinetic characteristics of H+ : K+ exchange and H(2) formation were typical for Delta micro (H(+)) -dependent TrkA uncoupled with F(0)F(1) under respiration. Conclusions: Diethylsulphoxide at low concentrations serves as an electron acceptor for an anaerobic respiratory chain stimulating bacterial growth and survival through the modulation of H+ : K+ exchange and H(2) formation activity. The effects of DESO were more pronounced than those of DMSO. Significance and impact of the study: Diethylsulphoxide determines essential biological and therapeutic properties that make its application preferable.
CO2 uptake in ethanol-driven chain elongation system: Microbial metabolic mechanisms

Abstract:CO2 as a byproduct of organic waste/wastewater fermentation has an important impact on the carboxylate chain elongation. In this study, a semi-continuous flow reactor was used to investigate the effects of CO2 loading rates (Low = 0.5 LCO2·L-1·d-1, Medium = 1.0 LCO2·L-1·d-1, High = 2.0 LCO2·L-1·d-1) on chain elongation system Ethanol and acetate were utilized as the electron donor and electron acceptor, respectively. The results demonstrate that low loading rate of CO2 has a positive effect on chain elongation. The maximum production of caproate and CH4 were observed at a low CO2 loading rate. Caproate production reached 1.88 g COD·L-1·d-1 with a selectivity of 62.55 %, while CH4 production reached 129.7 ml/d, representing 47.4 % of the total. Metagenomic analysis showed that low loading rate of CO2 favored the enrichment of Clostridium kluyveri, with its abundance being 3.8 times higher than at of high CO2 loading rate. Metatranscriptomic analysis revealed that high CO2 loading rate induced oxidative stress in microorganisms, as evidenced by increased expression of heat shock proteins and superoxide dismutase genes. Further investigation suggested that genes associated with the reverse β-oxidation pathway, CO2 uptake pathway and hydrogenotrophic methanogenesis pathway were reduced at high CO2 loading rate. These findings provide insight into the underlying mechanisms of how CO2 affects chain elongation, and it could be a crucial reason for the poor performance of chain elongation systems with high endogenous CO2 production.

Abstract:Carbon dioxide (CO 2 ) serves as the primary substrate for the photosynthesis of phytoplankton, forming the foundation of marine food webs and mediating the biogeochemical cycling of C and N. We studied the effects of CO 2 variation on the Michaelis-Menten equations and elemental composition of Skeletonema dohrnii and Heterosigma akashiwo . CO 2 functional response curves were conducted from 100 to 2000 ppm. The growth of both phytoplankton was significantly affected by CO 2 , but in different trends. The growth rate of S. dohrnii increased as CO 2 levels rose up to 400 ppm before reaching saturation. In contrast to S. dohrnii , the growth rate of H. akashiwo increased with CO 2 increasing up to 1000 ppm, and then CO 2 saturated. In addition, H. akashiwo showed a slower growth rate than S. dohrnii for all CO 2 concentrations, aside from 1000 ppm, and the Michaelis-Menten equations revealed that the half-saturation constant of H. akashiwo was higher than S. dohrnii . An increase in CO 2 concentration was seen to significantly affected the POC: Chl- a of both S. dohrnii and H. akashiwo , however, the effects on their elemental composition were minimal. Overall, our findings indicate that H. akashiwo had a more positive reaction to elevated CO 2 than S. dohrnii , and with higher nutrient utilization efficiency, while S. dohrnii exhibited higher carbon fixation efficiency, which is in line with their respective carbon concentrating mechanisms. Consequently, elevated CO 2 , either alone or in combination with other limiting factors, may significantly alter the relative relationships between these two harmful algal blooms (HAB) species over the next century.

Abstract:ABSTRACT Carbon dioxide is an antimicrobial gas commonly used in modified atmosphere packaging. In the present study, the effects of carbon dioxide on the growth of and neurotoxin production by nonproteolytic Clostridium botulinum type E were studied during the growth cycle. Quantitative reverse transcription-PCR and an enzyme-linked immunosorbent assay were used to quantify expression of the type E botulinum neurotoxin gene ( cntE ) and the formation of type E neurotoxin. The expression levels of cntE were similar in two strains, with relative expression peaking in the transition between exponential phase and stationary phase. In stationary phase, cntE mRNA expression declined rapidly. The cntE mRNA half-life was calculated to be approximately 9 minutes. Neurotoxin formation occurred in late exponential phase and stationary phase. High carbon dioxide concentrations delayed growth by increasing the lag time and decreasing the maximum growth rate. The effects of carbon dioxide concentration on relative neurotoxin gene expression and neurotoxin formation were significant. Expression of cntE mRNA and the formation of extracellular neurotoxin were twofold higher with a headspace carbon dioxide concentration of 70% (vol/vol) compared to 10% (vol/vol). This finding sheds a new, cautionary light on the potential risks of botulism associated with the use of modified atmosphere packaging.

Abstract:Recently, we have experienced cerebrovascular embolic events in 2 consecutive patients in our outpatient clinic. Accordingly, we want to share our comments with literature on these 2 patients. Both patients had newly diagnosed left ventricular (LV) dysfunction, sinus rhythm, and cerebrovascular event within the first week after initiation of heart failure treatment. Although, our cases are not enough to make a general statement or conclusion, we can recommend that patients with newly diagnosed severe LV dysfunction with normal sinus rhythm and without echocardiographically visible thrombus should also be closely followed up for thromboembolic complications at least during the first weeks of congestive heart failure treatment.
Oxygen tolerance estimates in Campylobacter species depend on the testing medium

Abstract:Oxygen tolerance of the microaerophile Campylobacter jejuni subsp. jejuni varied with different brands of complex media which were used for plating the dilute cell suspensions. The tryptone component was one factor. With some tryptones growth occurred at 21% oxygen whereas with others there was no growth at oxygen levels of 15% or higher. A chemically-defined, agar-solidified plating medium was used to estimate the oxygen tolerance of Camp. jejuni subsp. jejuni, Camp. coli and Camp. fetus subsp. fetus, and also to assess the effect of added scavengers of reactive oxygen intermediates on the oxygen tolerance. Some scavengers such as allopurinol, azelaic acid, caffeine, cimetidine, TEMPOL and pyruvate enhanced oxygen tolerance markedly whereas others such as carnosine, dimethyl thiourea, spermidine and superoxide dismutase had little effect.
Study on Effects of Different Carbon Sources on Growth of Synechocystis Sp.pcc6803 and Synechococcus Elongates PCC7942

Abstract:Effect of carbon source conditions on growth of Synechocystis sp. PCC6803 and Synechococcus elongates PCC7942 were studied. According to results of single carbon source, the two strains occurred the highest growth rate under 0.1 mol/L initial NaHCO3; However, PCC6803 occurred lower growth rate than that of PCC7942 under 0.2 or 0.3 mol/L initial NaHCO3. Furthermore, the OD685 nmof PCC6803 even was decreased during later stages. Under the mix carbon sources condition,there are different highest growth rates to occur between the two strains. Combined 0.1 mol/L initial NaHCO3with CO2aeration, the highest growth rate of PCC6803 was observed. Whereas, the highest growth rate of PCC7942 was studied in company with 0.1 mol/L initial NaHCO3and air aeration.
Effect of reduced nutritional supply on the metabolic activity and survival of cariogenic bacteria in vitro

Abstract:Sealed cariogenic bacteria are deprived from dietary carbohydrate, but could be provided with nutrients by pulpal fluids, with adaptive strain-specific activities being possible. We investigated survival and metabolic activity of the cariogenic bacteria Streptococcus sobrinus, Actinomyces naeslundii and Lactobacillus rhamnosus in different carbohydrate-limited media without carbon source (CLM), or containing glucose (CLM-G), albumin (CLM-A), or α1-acid glycoprotein (CLM-AGP) in vitro. Bacterial metabolite concentrations (lactate, pyruvate, oxaloacetate, citrate, acetate, formate, ethanol, acetoin) after 20 and 4 hours incubation, and bacterial numbers (CFU) after 24 hours incubation were analyzed using multivariate-analysis-of-variance (MANOVA). The medium (p = 0.02/MANOVA), strain and incubation-time (both p < 0.001) had significant impact on metabolite concentrations. Bacteria secreted mainly lactate (80.3 µg/106 bacteria S. sobrinus) and acetate (54.5 µg/106 bacteria A. naeslundii). Nearly all metabolites were produced in higher concentrations in S. sobrinus than in A. naeslundii or L. rhamnosus (p < 0.05/HSD). Metabolite concentration was significantly higher in CLM-G than in other media for most metabolites (p < 0.05). L. rhamnosus showed significantly lower survival than S. sobrinus and A. naeslundii (p < 0.05/HSD) regardless of the media, while S. sobrinus and A. naeslundii showed medium-specific survival. Survival of carbon starvation was strain- and medium-specific. Sustained organic acid production was found for all strains and media.

Abstract:In this paper, it is shown that a stepped-impedance open-stub can be used to design a reflection type dispersive delay line with peak delays more than tens of nano seconds. A loss less transmission line model is used for analysis. Finally, a delay line is fabricated that provides a measured delay of 22.23 ns at 2.5 GHz. Total rectangular area of the unit is 60 mm × 40 mm on a 0.5 mm thick substrate with dielectric constant 3.55.

Abstract:Escherichia coli varies the synthesis of many of its respiratory enzymes in response to oxygen availability. These enzymes include cytochrome o oxidase (cyoABCDE) and cytochrome d oxidase (cydAB), used during aerobic cell growth, and a fumarate reductase (frdABCD), dimethyl sulfoxide/trimethylamine oxide reductase (dmsABC), and nitrate reductase (narGHJI), used during anaerobic respiratory conditions. To determine how different levels of oxygen affect the expression of each operon, strains containing cyo-lacZ, cyd-lacZ, frdA-lacZ, dmsA-lacZ, and narG-lacZ fusions were grown in continuous culture at various degrees of air saturation. The basal-level expression of the anaerobic respiratory genes, frdABCD, dmsABC, and narGHJI, occurred when the air saturation of the medium was above 20%; as the saturation was reduced to below 10% (ca. 2% oxygen), the expression rapidly increased and reached a maximal level at 0% air. In contrast, cyoABCDE gene expression was lowest under anaerobic conditions while cyd-lacZ expression was about 40% of its maximum level. When the oxygen level was raised into the microaerophilic range (ca. 7% air saturation) cyd-lacZ expression was maximal while cyo-lacZ expression was elevated by about fivefold. As the air level was raised to above 20% saturation, cyd-lacZ expression fell to a basal level while cyo-lacZ expression was increased to its maximum level. The role of the Fnr and ArcA regulatory proteins in this microaerophilic control of respiratory gene expression was documented: whereas Fnr function as an aerobic/anaerobic switch in the range of 0 to 10% air saturation, ArcA exerted its control in the 10 to 20% range. These two transcriptional regulators coordinate the hierarchial control of respiratory pathway gene expression in E. coli to ensure the optimal use of oxygen in the cell environment.

Abstract:Numerous insect species and their associated microbial pathogens are exposed to elevated CO 2 concentrations in both artificial and natural environments. However, the impacts of elevated CO 2 on the fitness of these pathogens and the susceptibility of insects to pathogen infections are not well understood. The yellow mealworm, Tenebrio molitor , is commonly produced for food and feed purposes in mass-rearing systems, which increases risk of pathogen infections. Additionally, entomopathogens are used to control T. molitor , which is also a pest of stored grains. It is therefore important to understand how elevated CO 2 may affect both the pathogen directly and impact on host-pathogen interactions. We demonstrate that elevated CO 2 concentrations reduced the viability and persistence of the spores of the bacterial pathogen Bacillus thuringiensis . In contrast, conidia of the fungal pathogen Metarhizium brunneum germinated faster under elevated CO 2 . Pre-exposure of the two pathogens to elevated CO 2 prior to host infection did not affect the survival probability of T. molitor larvae. However, larvae reared at elevated CO 2 concentrations were less susceptible to both pathogens compared to larvae reared at ambient CO 2 concentrations. Our findings indicate that whilst elevated CO 2 concentrations may be beneficial in reducing host susceptibility in mass-rearing systems, they may potentially reduce the efficacy of the tested entomopathogens when used as biological control agents of T. molitor larvae. We conclude that CO 2 concentrations should be carefully selected and monitored as an additional environmental factor in laboratory experiments investigating insect-pathogen interactions.

Abstract:The adaptive proliferation of bacteria, or cell division in the absence of an exogenous substrate, is controlled by density-dependent mechanisms with the participation of AHL- and AI-2-dependent quorum sensing systems. Along with the signaling molecules of these bacterial communication systems, bacterial metabolites that are permanently released during microbial metabolism, for example, CO 2 , can also participate in the regulation and can serve as biomarkers of cell density. It has been found that carbon dioxide is necessary to the adaptive proliferation initiation, and the elevated atmospheric CO 2 content causes this process to terminate prematurely. Thus, CO 2 is able to regulate bacterial adaptive reactions and is probably one of the signals involved in the initiation and termination of the process of adaptive proliferation. It was shown that CO 2 in the form of the bicarbonate ion can activate the cAMP-dependent signaling cascade and is also included in the bacterial cell mass.

Abstract:Abstract Studies were performed using pure cultures of A. acrogenes and E. coli and a heterogeneous microbial population growing in carbon‐limited chemostats with glucose as the sole carbon and energy source. A two‐level factorial experimental design was employed to test the hypothesis that the concentration of growth‐limiting substrate in a chemostat is controlled by the growth rate alone and is independent of the concentration of substrate entering the reactor. The pure culture experiments showed that the conclusions depend upon the measurement employed for growth‐limiting substrate. When the concentration of glucose was measured directly, the hypothesis was found to be true within the limits of the study (500–1500 mg/liter). However, if the chemical oxygen demand (COD) test was used as the measure of growth‐limiting substrate the hypothesis was found to be false. When heterogeneous cultures were employed the hypothesis was false regardless of the technique used to measure the concentration of growth‐limiting substrate. Nevertheless, it was possible to generate regression equations which described the interactions among influent COD, growth rate, and effluent COD with a high level of correlation.

Changes in gene expression, cell physiology and toxicity of the harmful cyanobacterium Microcystis aeruginosa at elevated CO2

Abstract:Rising CO2 concentrations may have large effects on aquatic microorganisms. In this study, we investigated how elevated pCO2 affects the harmful freshwater cyanobacterium Microcystis aeruginosa. This species is capable of producing dense blooms and hepatotoxins called microcystins. Strain PCC 7806 was cultured in chemostats that were shifted from low to high pCO2 conditions. This resulted in a transition from a C-limited to a light-limited steady state, with a ~2.7-fold increase of the cyanobacterial biomass and ~2.5-fold more microcystin per cell. Cells increased their chlorophyll a and phycocyanin content, and raised their PSI/PSII ratio at high pCO2. Surprisingly, cells had a lower dry weight and contained less carbohydrates, which might be an adaptation to improve the buoyancy of Microcystis when light becomes more limiting at high pCO2. Only 234 of the 4691 genes responded to elevated pCO2. For instance, expression of the carboxysome, RuBisCO, photosystem and C metabolism genes did not change significantly, and only a few N assimilation genes were expressed differently. The lack of large-scale changes in the transcriptome could suit a buoyant species that lives in eutrophic lakes with strong CO2 fluctuations very well. However, we found major responses in inorganic carbon uptake. At low pCO2, cells were mainly dependent on bicarbonate uptake, whereas at high pCO2 gene expression of the bicarbonate uptake systems was down-regulated and cells shifted to CO2 and low-affinity bicarbonate uptake. These results show that the need for high-affinity bicarbonate uptake systems ceases at elevated CO2. Moreover, the combination of an increased cyanobacterial abundance, improved buoyancy, and higher toxin content per cell indicates that rising atmospheric CO2 levels may increase the problems associated with the harmful cyanobacterium Microcystis in eutrophic lakes.

Abstract:13C NMR was used to study the effect of oxygen on methanol oxidation by a type II methanotrophic bacterium isolated from a bioreactor in which methane was used as electron donor for denitrification. Under high (35-25%) oxygen conditions the first step of methanol oxidation to formaldehyde was much faster than the following conversions to formate and carbon dioxide. Due to this the accumulation of formaldehyde led to a poisoning of the cells. A more balanced conversion of (13)C-labelled methanol to carbon dioxide was observed at low (1-5%) oxygen concentrations. In this case, formaldehyde was slowly converted to formate and carbon dioxide. Formaldehyde did not accumulate to inhibitory levels. The oxygen-dependent formation of formaldehyde and formate from methanol is discussed kinetically and thermodynamically.
