Abstract:The simultaneous determination of emodin, aloe-emodin and rhein, in Semen Cassiae, Qinghai Wild Dahuang tea, Ningxia Juemingzi tea, Lanzhou Juemingzi tea and Surong Juerningzi tea, has been investigated by micellar electrokinetic capillary chromatography for the first time. With an electrolyte containing 15 mm borax, 30 mm SDS, 10% (v/v) ethanol, at pH 9.60 and 20 kV applied voltage, the three analytes were completely separated within 12 min. The effects of the concentration of borax, electrolyte pH, the concentrations of SDS and organic modifier and the applied voltage on electropheoretic behavior and separation were studied. The linear calibration range was 4-120 mug/mL (r = 0.9921) for emodin, 10-200 mug/mL (r = 0.9970) for aloe-emodin and 2-100 mug/mL (r = 0.9971) for rhein, respectively. Under the optimum conditions, the relative standard deviation (RSD) values (n = 6) of the migration time and the peak area of each peak was 0.59-0.80% and 1.30-3.22%, respectively. The contents of the analytes in Rheum, Qinghai Wild Dahuang tea, Semen Cassiae and three other kinds of teas were easily determined with recoveries ranging from 95.3 to 104.4%. Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.
A Validated Stability-Indicating RP-HPLC Method for the Simultaneous Estimation of Aloe Emodin, Aloin, and Emodin in a Marketed Formulation of Aloe Vera

Abstract:The present analytical study focuses on the simultaneous estimation of Aloe emodin, aloin, and emodin as chemical markers commonly found in herbal formulations using the reversed-phase high-performance liquid chromatography ( RP-HPLC ) method. Aloin, Aloe emodin, and emodin are active components present in Aloe species, rhubarb, and cascara. A novel method based on RP-HPLC was developed for the simultaneous estimation of these chemical markers. A Waters Alliance-e2695 chromatograph with a 150 × 4.6 mm, 3.5 μm column, a mobile phase with acetonitrile, 0.1% formic acid, and methanol (20 : 50 : 30) were used for this analytical work. The flow rate was 1.0 mL/min, and detection was carried out by a PDA detector with an absorption wavelength of 237 nm at an ambient temperature of 25°C. The linearity ranges for Aloe emodin, aloin, and emodin were found to be 25–150, 12.5–75, and 2.5–15 μg/mL, respectively. The mean recoveries of Aloe emodin, aloin, and emodin were within the acceptance limit (98–102%). The relative standard deviation of precision and robustness was found to be less than 2%. Forced degradation studies were carried out for chemical markers with the help of the developed RP-HPLC method, and no considerable degradants were found. The developed stability-indicating method was applied for the quantification of the three markers in the Ayurvedic formulation of Aloe vera.

Determination of anthraquinones in Rhamnus purshiana using high-performance liquid chromatography coupled to diode array detector and simple ultraviolet spectroscopic analysis

Abstract:A new method based on Ultraviolet spectrophotometry was developed and compared with that based on high-performance liquid chromatography for the determination and quantification of anthraquinones in the extracts of Rhamnus purshiana bark. A validated quantitative analysis of cascaroside A, cascaroside B, emodin, and aloe-emodin in these herbal products has been previously performed using high-performance liquid chromatography coupled with a diode array detector. In the high-performance liquid chromatography analysis, all the anthraquinones showed satisfactory regression (r2 > 0.98) within the test ranges, and the recovery was in the range of 94-117%. The limits of detection and quantification were 0.008-0.010 and 0.029-0.035 μg/mL, respectively. Hierarchical cluster analysis and principal component analysis showed differences in the anthraquinones determined from herbal samples. Subsequently, a simple and low-cost ultraviolet spectrophotometric methodology for the quantitative analysis of the same compounds in the extracts was applied, and all the contents were determined. A paired t-test confirmed that there were no significant differences between the two methods. Our results revealed that the developed method is simple and provides the ability to discriminate and control the quality of anthraquinones in herbal products.
Simultaneous HPLC Determination of Amino Acids in Tea Infusion Coupled to Pre-column Derivatization with 2,4-Dinitrofluorobenzene

Abstract:An HPLC method coupled to pre-column derivatization with 2,4-dinitrofluorobenzene (DNFB) was developed for simultaneous determination of 19 amino acids in tea infusion. The amino acid-derived samples were separated on a reversed-phase C18 column (4.6 × 250 mm) at 35 °C and monitored by an ultraviolet detector at 360 nm. Separation analysis showed a good resolution (Rs > 1.5) for most amino acids with a gradient elution of mobile phases at a flow rate of 1 ml min -1 . Precision analysis revealed low repeatability errors with relative standard deviation (RSD) ranging from 0.11 to 1.53 % for retention times and 0.60 to 2.84 % for peak areas, and recovery rate ranged from 70.60 to 108.80 %. Linear calibration curves were well established at the concentrations ranging from 0.57 to 57.38 μg ml −1 , with correlation coefficients R 2 ≥ 0.9918 for 19 detected amino acids. The superior stability of the amino acid derivatives at room temperature gave an advantage to simultaneous preparation of many samples and continuous analysis using auto sampler. The established method was successfully applied to separate and quantify amino acids in three tea cultivars, and the results indicated that the albino tea cultivars contained higher contents of total amino acid, theanine, γ-aminobutyric acid (GABA), glutamic acid (Glu), aspartic acid (Asp), and serine (Ser) than normal tea cultivar.

Abstract:OBJECTIVE:To develop an HPLC method to determine the contents of danshensu, hydroxysafflor yellow A, rosmarinic acid, lithospermic acid, salvianolic acid B in the water extract of mixed Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos simultaneously.METHOD:The separation were carried out at 30 degrees C on a ZORBAX Eclipse Plus C18 column (4.6 mm x 100 mm, 1.8 microm) with formic acid-500 mmol x L(-1) ammonium formate-water solution (0.5:10:90) as mobile phase A and acetonitrile-formic acid solution (100: 0.5) as mobile phase B in gradient mode at a flow rate of 0.5 mL x min(-1). Detection wavelengths were 280 nm for danshensu, rosmarinic acid, lithospermic acid, salvianolic acid B, and 380 nm for hydroxysafflor yellow A.RESULT:The 5 components were separated well with a good linearity (R2 > 0.999 3) in the range of the test concentration. The average recoveries of danshensu, hydroxysafflor yellow A, rosmarinic acid, lithospermic acid, and salvianolic acid B were 99.1%, 102%, 102%, 98.5% and 101%, respectively.CONCLUSION:This method is simple, accurate, and repeatable.

Abstract:Abstract Smilax china L. and Salix alba are the medicinal plants of interest due to the presence of various therapeutic activities of these plants such as anti‐inflammatory, anti‐diabetic, anti‐psoriatic and digestive properties. Smilax china and Salix alba consists of bioactive marker Quercetin and Salicin, respectively. In the present study, a simple, rapid, reliable and accessible high‐performance thin‐layer chromatography method has been established for simultaneous quantification of quercetin and salicin. High‐performance thin‐layer chromatography fingerprinting was carried out on pre‐coated Silica G60 F254 plates with toluene: ethyl acetate: formic acid (3.5:6.0:1.0 v/v/v) as mobile phase. The developed plates were scanned and quantified at two different λ max 380 and 275 nm. The drug samples were satisfactorily resolved with Rf 0.61 ± 0.02 for quercetin and 0.07 ± 0.03 for salicin. The method was validated for linearity, accuracy, precision, and specificity. The limits of detection and quantification for quercetin and salicin were 2.0 and 6.00 µg per band and 6.00 and 30.0 µg per band respectively. The developed high‐performance thin‐layer chromatography method can be implemented for the concurrent estimation of quercetin and salicin for routine quality control of herbal drugs containing these phytoconstituents.

Abstract:Moringa oleifera Lam. has been used as a traditional medicine for the treatment of numerous diseases. A simultaneous high-performance liquid chromatography (HPLC) analysis was developed and validated for the determination of the contents of crypto-chlorogenic acid, isoquercetin and astragalin, the primary antioxidative compounds, in M. oleifera leaves. HPLC analysis was successfully conducted by using a Hypersil BDS C18 column, eluted with a gradient of methanol-1% acetic acid with a flow rate of 1 mL/min, and detected at 334 nm. Parameters for the validation included linearity, precision, accuracy and limits of detection and quantitation. The developed HPLC method was precise, with relative standard deviation < 2%. The recovery values of crypto-chlorogenic acid, isoquercetin and astragalin in M. oleifera leaf extracts were 98.50, 98.47 and 98.59%, respectively. The average contents of these compounds in the dried ethanolic extracts of the leaves of M. oleifera collected from different regions of Thailand were 0.081, 0.120 and 0.153% (w/w), respectively. The developed HPLC method was appropriate and practical for the simultaneous analysis of crypto-chlorogenic acid, isoquercetin and astragalin in the leaf extract of M. oleifera. This work is valuable as guidance for the standardization of the leaf extracts and pharmaceutical products of M. oleifera.

Abstract:High-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) were compared to simultaneously determine and separate 11 anthraquinones from rhubarb, including emodin, chrysophanol, rhein and their glucosides, aloe-emodin, sennoside A, and sennoside B. A UV-diode array detector (DAD) at 254 nm with a gradient elution of acetonitrile/water (method A: 0 min 6:94, 12 min 12:88, 15 min 20:80, 40 min 25:75, 53 min 55:45, 55 min 100:0; method B: 0 min 5:95, 2 min 15:85, 5 min 20:80, 12 min 25:75, 15 min 50:50, 19 min 98:2) at 28(+/-1) degrees C (method A) and 30-60 degrees C (method B) in HPLC or with 0.03 M borate buffer (pH 10.0) containing 25% (v/v) acetonitrile with 0.002 M 2,6-di-O-methyl-beta-cyclodextrin (CD) and 0.005 M alpha-CD in CE effectively detected this separation in 25 min. The detection limits of anthraquinones from rhubarb were in the 0.02-0.2 microg/mL and 0.1-0.8 microg/mL ranges for HPLC and CE, respectively. The established HPLC and CE methods are suitable for quantitative determination of emodin, chrysophanol, aloe-emodin, emodin-1-beta-d-glucoside, emodin-8-beta-d-glucoside, chrysophanol-1-beta-d-glucoside, chrysophanol-8-beta-d-glucoside, and rhein-8-beta-d-glucoside.

Quantification of two biomarker compounds by a validated high-performance thin-layer chromatographic method from different extracts of Pluchea dioscoridis growing in Saudi Arabia

Abstract:The aim of this study was to develop a validated high-performance thin-layer chromatography (HPTLC) procedure for resolution of chemical constituents and identification and quantification of two selected natural anticancer compounds, stigmasterol (PD-1) and cinnamic acid (PD-2), in Pluchea dioscoridis chloroform fraction (PDCF). The chromatographic estimations were conducted on normal HPTLC (20 cm × 10 cm glass-backed silica gel 60 F254) plates with chloroform-methanol-acetic acid (93:5:2, V/V) used as the mobile phase. para-Anisaldehyde was used for the derivatization of the developed plate, and compact spots were scanned at λmax = 513 nm. A well resolved, compact, and intense peaks of PD-1 and PD-2 were recorded at RF = 0.57 and 0.19, respectively. The proposed analytical method for both biomarker compounds was found to be handy, simple, precise, linear (%RSD = 1.03–1.45), accurate (98.91–99.14%), reliable, and sensitive for the analysis of both biomarkers. The LOD/LOQ (ng) for PD-1 and PD-2 were recorded as 38.73/117.37 and 42.58/129.04, respectively, in the linearity range of 200–1400 ng per spot. The obtained result showed maximum quantities of PD-1 and PD-2 (5.36 and 16.98 μg mg-1, respectively). The developed HPTLC was found to be suitable for the routine analysis of these 2 biomarkers in the chloroform fraction of Pluchea dioscoridis and can be further employed in the process quality control of herbal formulations containing the said biomarkers.

Abstract:In this study, an HPLC-DAD method with high separation efficiency, detection sensitivity, and selectivity was developed and validated for the simultaneous qualitative and quantitative determination of anthraquinones (aloe-emodin, rhein, emodin, chrysophanol, and physcion) in Rumex acetosella L. and Rumex crispus L. plants collected at different regions and times. Optimal method conditions were determined by studying different mobile phases, mobile phase compositions, flow rates, column, column temperatures, injection volume, and wavelength of the compounds. Validation studies were carried out to prove the validity of the developed method. The correlation coefficients for all compounds were found to be in the range of 0.9991–0.9997, and the calibration curves were linear in the concentration range of 0.1–1.0 μg/mL. The limits of detection and quantification values were calculated in the range of 0.014–0.017 and 0.017–0.057 μg/mL, respectively. The developed and validated method indicated that it is effective and suitable for determining aloe-emodin, rhein, emodin, chrysophanol, and physcion in extracts of Rumex plants. In addition, the antioxidant activities of the plant extracts were determined using the Cupric Ion Reducing Antioxidant Capacity and 2,2-diphenyl-1-picrylhydrazil radical assays, revealing their antioxidant properties.

Simultaneous determination and pharmacokinetic studies of aloe emodin and chrysophanol in rats after oral administration of Da-Cheng-Qi decoction by high-performance liquid chromatography

Abstract:A validated high-performance liquid chromatography (HPLC) method was developed for simultaneous determination and pharmacokinetic study of aloe emodin and chrysophanol in rats. It was performed on a reverse-phase C(18) column and a mobile phase made up of methanol and 0.2% acetic acid (83:17, v/v). The ultraviolet detection was 254 nm. 1,8-dihydroxyanthraquinone was used as the internal standard. The assay was linear over the range 28-2800 ng/mL (r(2) = 0.9993) for aloe emodin and 25.6-2560 ng/mL (r(2) = 0.9991) for chrysophanol. The average percentage recoveries of three spiked plasmas were 98.8-104.8% and 97.7-103.2% for aloe emodin and chrysophanol, respectively. Their RSD of intra-day and inter-day precision at concentrations of 56, 280 and 1400 ng/mL for aloe emodin and 51.6, 258 and 1290 ng/mL for chrysophanol were less than 3.5%. This method was applied for the first time to simultaneously determinate aloe emodin and chrysophanol in rats following oral administration of traditional Chinese medicine of Da-Cheng-Qi decoction. The pharmacokinetic parameters showed that chrysophanol was better absorbed with higher concentrations in plasma than aloe emodin did. They both eliminated slowly in male rats. The assay is suitable for identifying the plasma and tissue levels of aloe emodin and chrysophanol in preclinical investigations.
Development and Validation of a SPE-HPLC Method for Quantification of Rhein, Emodin, Chrysophanol and Physcion in Rhamnus petiolaris Boiss. & Balansa

Abstract:Anthraquinones exhibit a significant group of natural and synthetic quinone derivatives because of their biological activities and industrial applications. Rhamnaceae is one of the families known to contain different kinds of anthraquinones. In this study, it was aimed to quantify rhein, emodin, chrysophanol and physcion in fruits of Rhamnus petiolaris Boiss. & Balansa belonging to Rhamnaceae by solid phase extraction and high performance liquid chromatography with ultraviolet detection. The anthraquinones were separated using a C18 analytical column. Gradient elution was performed using a mobile phase consisted of 0.1% o-phosphoric acid solution and methanol. Analytes were detected at 254 nm. Calibration curves were prepared in the range of 0.25-5.00 μg/mL for rhein, chrysophanol, physcion, 1.00-50.00 μg/mL for emodin. Limits of detection and quantification were between 0.07-0.11 and 0.20-0.34 μg/mL, respectively. Relative standard deviations were ≤ 5.78% in repeatability and intermediate precision studies. Accuracy was determined as relative mean error (8.17-12.06%). Extraction was achieved by maceration with acetone and ethanol, followed by hydrophilic-lipophilic balance solid phase extraction. Recoveries were between 96.2 and 109.6%. The developed and validated method was successfully performed to quantify rhein, emodin, chrysophanol and physcion in R. petiolaris fruit extracts. Only physcion was not detected above limit of detection.
Comparison of three different extraction methods and HPLC determination of the anthraquinones aloe-emodine, emodine, rheine, chrysophanol and physcione in the bark of Rhamnus alpinus L. (Rhamnaceae)

Abstract:Introduction: Rhamnus alpinus L. (Rhamnaceae), a traditional plants in the flora of the Abruzzo region, is known to contain active anthraquinone secondary metabolites. However, the content of anthraquinones varies among R. alpinus samples depending on collection season and site. Thus, using simple, reliable and accurate analytical methods for the determination of anthraquinones in R. alpinus extracts allows comparative study of different methods of extraction. Objective: After a partial validation of an HPLC method for the simultaneous determination of five anthraquinones, aloe-emodine, rheine, emodine, chrysophanol and physcione, in the bark of R. alpinus, we compared three different methods of extraction. Methodology: Anthraquinones were extracted from the bark of R. alpinus using different techniques (methanol maceration, ultrasonic and supercritical CO(2) extraction). Separation and quantification of anthraquinones were accomplished using a reversed-phase C(18) column with the mobile phase of H(2)O-methanol (40 : 60, v/v, 1% formic acid) at a wavelength of 254 nm. The qualitative analyses were also achieved at wavelength of 435 nm. Results: All calibration curves were linear over the concentration range tested (10-200 mM) with the determination coefficients >or=0.991. The detection limits (S/N = 3) were 5 mM for each analytes. All five anthraquinones were found in the samples tested at concentrations reported in experimental data. Conclusion: The described HPLC method and optimised extraction procedure are simple, accurate and selective for separation and quantification of anthraquinones in the bark of R. alpinus and allow evaluation of the best extraction procedure between the tested assays.
High-performance thin-layer chromatography (HPTLC) method development and validation for the estimation of curcumin and cineole

Abstract:Curcumin and cineole are phytochemicals which have shown synergistic effects in allergic rhinitis (AR). The aim of the current work was to develop a simple, rapid, and accurate method for the simultaneous estimation of curcumin and cineole using high-performance thin-layer chromatographic (HPTLC) technique. Aluminum plates pre-coated with silica gel 60F254 were used as the stationary phase. The solvent system ratio of n-hexane–ethyl acetate (6:3.5, V/V) produced sharp, symmetrical, and well-defined peaks for curcumin and cineole, with RF values of 0.31 ± 0.02 and 0.71 ± 0.02, respectively. The absorbance mode at 242 nm was used for densitometric analysis. Curcumin's linear range was observed between 1 and 6 µg/spot (R2 = 0.995), while cineole's was 10–60 µg/spot (R2 = 0.997). The limit of detection (LOD) and limit of quantification (LOQ) were found to be 0.045 and 0.135 µg/spot, and 0.034 and 0.102 µg/spot for curcumin and cineole, respectively. The developed method was validated as per the International Council for Harmonisation (ICH) guidelines. The developed method was found to be accurate and sensitive and can be used for the simultaneous estimation of curcumin and cineole in formulations.
Simultaneous Quantification of Two Bioactive Flavonoids, Homoeriodictyol and Persicogenin, in the Methanol Extract of the Aerial Parts of Two Different Species of Genus Rhus by a Validated High-Performance Thin-Layer Chromatographic—Densitometric Method

Abstract:A simple and sensitive high-performance thin-layer chromatographic (HPTLC)—densitometric method was developed for the simultaneous quantification of two flavonoid compounds, persicogenin and homoeriodictyol, in the methanol extracts of the aerial parts of two species (Rhus retinorrhoea and Rhus tripartita) of the genus Rhus grown in the Kingdom of Saudi Arabia. Chromatography was performed on glass-backed silica gel 60 F254 HPTLC plates using toluene-ethyl acetate-methanol (8:2:0.5, v/v) as the mobile phase. Scanning and quantification were done at 293 nm. The system was found to give compact spot for homoeriodictyol and persicogenin at, RF = 0.30 ± 0.01 and 0.48 ± 0.01, respectively. The linearity ranges for homoeriodictyol and persicogenin were found to be the same (100–800 ng spot−1) with correlation coefficients (r2 values) of 0.9989 and 0.9983, respectively. The limit of detection (LOD) for homoeriodictyol and persicogenin was found to be 26 and 31 ng band−1, respectively, while the limit of quantification (LOQ) was found to be 77 and 92 ng band−1, respectively. Homoeriodictyol (7.06%) and persicogenin (2.33%) were only found in R. retinorrhoea. The developed method was found to be accurate and precise; hence, it can be used as an important tool to assure the therapeutic dose of homoeriodictyol and persicogenin in herbal formulations as well as for the standardization and quality control of bulk drugs.

Abstract:INTRODUCTION:Aralia elata leaves are known to have several biological activities, including anti-arrythmia, antitumor, anti-inflammatory, anti-fatigue, antimicrobial and antiviral effects. Our previous study found that triterpenoid saponins from the leaves of A. elata had antitumor effects. Quantification of the triterpenoids is important for the quality control of A. elata leaves.OBJECTIVE:To establish high-performance liquid chromatography coupled with evaporative light scattering detection (HPLC-ELSD) for the simultaneous determination of four major triterpenoid saponins, including Aralia-saponin IV, Aralia-saponin VI, 3-O-β-d- glucopyranosyl-(1 → 3)-β-d-glucopyranosyl-(1 → 3)-β-d-glucopyranosyl oleanolic acid 28-O-β-d-glucopyranoside (Aralia-saponin TTP)and Aralia-saponin V.METHODOLOGY:The separation was carried out on a Dikma Diamonsil C18 column (4.6 mm × 250 mm, 5 μm) efficiently with gradient elution consisting of acetonitrile and water.RESULTS:All calibration curves showed good linear regression (R2 > 0.9996) within the ranges of tested concentrations. This validated method was applied to determine the contents of the four major triterpenoid saponins in 53 samples from different regions of northeast China. Hierarchical clustering analysis was first used to classify and differentiate Aralia elata leaves.CONCLUSION:The method developed was successfully applied to analyse four major triterpenoid saponins in Aralia elata leaves which is helpful for quality control of the herb. Abstract:A new, simple, selective, sensitive, precise, and robust high-performance thin-layer chromatography (HPTLC) method for the simultaneous quantification of 6-gingerol, guggulsterone E, and guggulsterone Z in coded formulation AYUSH SG-5 (for rheumatoid arthritis) was developed and validated. Analysis of 6-gingerol, guggulsterone E, and guggulsterone Z was performed on thin-layer chromatography (TLC) aluminum plates pre-coated with silica gel 60F254 as the stationary phase. Linear ascending development was carried out in CAMAG twin-trough glass chamber saturated with a mobile phase consisting of toluene–acetone (9:1, V/V) at room temperature (25 ± 2 °C). CAMAG TLC scanner was used for spectrophotometric scanning and analysis in absorbance mode at 280 nm. The system was found to give compact spots for 6-gingerol (RF value 0.149 ± 0.022) scanned at 545 nm and guggulsterone E (RF value 0.192 ± 0.025) and guggulsterone Z (RF value 0.249 ± 0.028) scanned at 254 nm. The linear regression analysis data for the calibration plots exhibited a good linear relationship with r2 = 0.9992 ± 0.02 in the concentration range 100–1200 ng spot–1 with respect to the peak area. The method was validated as per the International Council for Harmonisation (ICH) guidelines to examine the validity, precision, robustness, ruggedness, limits of detection and quantification. The quantified and estimated amount of 6-gingerol, guggulsterone E, and guggulsterone Z in AYUSH SG-5 as well as in raw drugs was found to be well within limits (5% of the labeled content). Statistical analysis of the data manifested that the method is reproducible and sensitive for the simultaneous estimation and quantification of 6-gingerol, guggulsterone E, and guggulsterone Z.

Abstract:This research focused on investigating biologically active and pharmaceutically important phytochemicals using reliable and reproducible analytical techniques specifically designed for traditional medicinal plants. Considering the complex chemical diversity present in plant extracts, reliable identification methods are crucial. The goal of this study was to develop and validate a robust UHPLC-ESI-MS/MS method for the qualitative and quantitative analysis of organic acids (OAs) and phenolic compounds (PCs). With gradient elution of water containing 0.1 % formic acid and acetonitrile containing 0.1 % formic acid as the mobile phase, OAs and PCs were separated chromatographically on a C18 reversed-phase column. Through the use of an electrospray ionization source operating in negative ionization mode, these compounds were identified on a triple quadrupole tandem mass spectrometer. The validation results clearly indicate that the method exhibited linearity with a squared correlation coefficient ( R 2 ≥ 0.9986), demonstrating sensitivity (LOQ: 1.08 μg/L-10.79 μg/L), precision (RSD%≤2.73), and trueness (RE% ≤ 1.53) for the OAs and PCs found in the plants. The developed UHPLC-ESI-MS/MS method was successfully applied to the qualitative and quantitative analysis of OAs and PCs in Turkish plants, which are commonly used in traditional medicine. Our study identified a total of 12 OAs and PCs in F. vulgaris , 12 in P. divaricatum , 9 in H. linarioides , and 11 in R. ribes . In particular, gallic acid had the highest content in F. vulgaris (925.67 μg/g extract) and R. ribes (1358.78 μg/g extract), while H. linarioides (730.66 μg/g extract) and P. divaricatum (4003.02 μg/g extract) had relatively high levels of quinic acid. In conclusion, the developed UHPLC-ESI-MS/MS method allows the simultaneous analysis and quantification of 2 OAs and 33 PCs in plants and proves to be a valuable tool for evaluating the efficacy of traditional medicinal plants.
