Abstract:G protein-coupled receptors (GPCRs) are seven integral transmembrane proteins that are the primary targets of almost 30% of approved drugs and continue to represent a major focus of pharmaceutical research. All of GPCR targeted medicines were discovered by classical medicinal chemistry approaches. After the first GPCR crystal structures were determined, the docking screens using these structures lead to discovery of more novel and potent ligands. There are over 360 pharmaceutically relevant GPCRs in the human genome and to date about only 30 of structures have been determined. For these reasons, computational techniques such as homology modeling and molecular dynamics simulations have proven their usefulness to explore the structure and function of GPCRs. Furthermore, structure-based drug design and in silico screening (High Throughput Docking) are still the most common computational procedures in GPCRs drug discovery. Moreover, ligand-based methods such as three-dimensional quantitative structure-selectivity relationships, are the ideal molecular modeling approaches to rationalize the activity of tested GPCR ligands and identify novel GPCR ligands. In this review, we discuss the most recent advances for the computational approaches to effectively guide selectivity and affinity of ligands. We also describe novel approaches in medicinal chemistry, such as the development of biased agonists, allosteric modulators, and bivalent ligands for class A GPCRs. Furthermore, we highlight some knockout mice models in discovering biased signaling selectivity.

Abstract:G protein-coupled receptors (GPCRs), the largest family of human membrane proteins and an important class of drug targets, play a role in maintaining numerous physiological processes. Agonist or antagonist, orthosteric effects or allosteric effects, and biased signaling or balanced signaling, characterize the complexity of GPCR dynamic features. In this study, we first review the structural advancements, activation mechanisms, and functional diversity of GPCRs. We then focus on GPCR drug discovery by revealing the detailed drug-target interactions and the underlying mechanisms of orthosteric drugs approved by the US Food and Drug Administration in the past five years. Particularly, an up-to-date analysis is performed on available GPCR structures complexed with synthetic small-molecule allosteric modulators to elucidate key receptor-ligand interactions and allosteric mechanisms. Finally, we highlight how the widespread GPCR-druggable allosteric sites can guide structure- or mechanism-based drug design and propose prospects of designing bitopic ligands for the future therapeutic potential of targeting this receptor family.

Abstract:G protein-coupled receptors (GPCRs) represent the largest family of proteins targeted by drug design and discovery efforts. Of these efforts, the development of GPCR agonists is highly desirable, due to their therapeutic robust utility in treating diseases caused by deficient receptor signaling. One of the challenges in designing potent and selective GPCR agonists lies in the inability to achieve combined high binding affinity and subtype selectivity, due to the high homology between orthosteric sites among GPCR subtypes. To combat this difficulty, researchers have begun to explore the utility of targeting topographically distinct and less conserved binding sites, namely "allosteric" sites. Pursuing these sites offers the benefit of achieving high subtype selectivity, however, it also can result in a decreased binding affinity and potency as compared to orthosteric agonists. Therefore, bitopic ligands comprised of an orthosteric agonist and an allosteric modulator connected by a spacer and allowing binding with both the orthosteric and allosteric sites within one receptor, have been developed. It may combine the high subtype selectivity of an allosteric modulator with the high binding affinity of an orthosteric agonist and provides desired advantages over orthosteric agonists or allosteric modulators alone. Herein, we review the recent advances in the development of bitopic agonists/activators for various GPCR targets and their novel therapeutic potentials.

Abstract:G protein-coupled receptors (GPCRs) are versatile and vital proteins involved in a wide array of physiological processes and responses, such as sensory perception (e.g., vision, taste, and smell), immune response, hormone regulation, and neurotransmission. Their diverse and essential roles in the body make them a significant focus for pharmaceutical research and drug development. Currently, approximately 35% of marketed drugs directly target GPCRs, underscoring their prominence as therapeutic targets. Recent advances in structural biology have substantially deepened our understanding of GPCR activation mechanisms and interactions with G-protein and arrestin signaling pathways. This review offers an in-depth exploration of both traditional and recent methods in GPCR structure analysis. It presents structure-based insights into ligand recognition and receptor activation mechanisms and delves deeper into the mechanisms of canonical and noncanonical signaling pathways downstream of GPCRs. Furthermore, it highlights recent advancements in GPCR-related drug discovery and development. Particular emphasis is placed on GPCR selective drugs, allosteric and biased signaling, polyphamarcology, and antibody drugs. Our goal is to provide researchers with a thorough and updated understanding of GPCR structure determination, signaling pathway investigation, and drug development. This foundation aims to propel forward-thinking therapeutic approaches that target GPCRs, drawing upon the latest insights into GPCR ligand selectivity, activation, and biased signaling mechanisms.

Abstract:Circulation Research, Volume 135, Issue 1, Page 174-197, June 21, 2024. GPCRs (G protein-coupled receptors), also known as 7 transmembrane domain receptors, are the largest receptor family in the human genome, with ≈800 members. GPCRs regulate nearly every aspect of human physiology and disease, thus serving as important drug targets in cardiovascular disease. Sharing a conserved structure comprised of 7 transmembrane α-helices, GPCRs couple to heterotrimeric G-proteins, GPCR kinases, and β-arrestins, promoting downstream signaling through second messengers and other intracellular signaling pathways. GPCR drug development has led to important cardiovascular therapies, such as antagonists of β-adrenergic and angiotensin II receptors for heart failure and hypertension, and agonists of the glucagon-like peptide-1 receptor for reducing adverse cardiovascular events and other emerging indications. There continues to be a major interest in GPCR drug development in cardiovascular and cardiometabolic disease, driven by advances in GPCR mechanistic studies and structure-based drug design. This review recounts the rich history of GPCR research, including the current state of clinically used GPCR drugs, and highlights newly discovered aspects of GPCR biology and promising directions for future investigation. As additional mechanisms for regulating GPCR signaling are uncovered, new strategies for targeting these ubiquitous receptors hold tremendous promise for the field of cardiovascular medicine.

Abstract:G protein-coupled receptors (GPCRs) remain a major domain of pharmaceutical discovery. The identification of GPCR lead compounds and their optimization are now structure-based, thanks to advances in X-ray crystallography, molecular modeling, protein engineering and biophysical techniques. In silico screening provides useful hit molecules. New pharmacological approaches to tuning the pleotropic action of GPCRs include: allosteric modulators, biased ligands, GPCR heterodimer-targeted compounds, manipulation of polypharmacology, receptor antibodies and tailoring of drug molecules to fit GPCR pharmacogenomics. Measurements of kinetics and drug efficacy are factors influencing clinical success. With the exception of inhibitors of GPCR kinases, targeting of intracellular GPCR signaling or receptor cycling for therapeutic purposes remains a futuristic concept. New assay approaches are more efficient and multidimensional: cell-based, label-free, fluorescence-based assays, and biosensors. Tailoring GPCR drugs to a patient's genetic background is now being considered. Chemoinformatic tools can predict ADME-tox properties. New imaging technology visualizes drug action in vivo. Thus, there is reason to be optimistic that new technology for GPCR ligand discovery will help reverse the current narrowing of the pharmaceutical pipeline.

Abstract:G-protein-coupled receptors (GPCRs) are the largest family of proteins in the human genome. Within the last year, we have witnessed a relative explosion in the amount of structural information available for the GPCR family with two new structures of opsin in the presence and absence of transducin peptide, four new structures of beta-adrenergic receptors, and a recent structure of the human adenosine A2A receptor. The new biological insight being gained, such as the highly divergent extracellular loops and areas of structural convergence within the transmembrane helices, allows us to chart a course for further investigation into this important class of membrane proteins.

Abstract:G protein-coupled receptors (GPCRs) are transmembrane proteins that participate in many physiological processes and represent major pharmacological targets. Recent advances in structural biology of GPCRs have enabled the development of drugs based on the receptor structure (structure-based drug design, SBDD). SBDD utilizes information about the receptor–ligand complex to search for suitable compounds, thus expanding the chemical space of possible receptor ligands without the need for experimental screening. The review describes the use of structure-based virtual screening (SBVS) for GPCR ligands and approaches for the functional testing of potential drug compounds, as well as discusses recent advances and successful examples in the application of SBDD for the identification of GPCR ligands.

Abstract:G protein-coupled receptors (GPCRs) are the target of one-third of all approved drugs; however, these drugs only target about one-eighth of the human repertoire of GPCRs. GPCRs regulate a diverse range of critical physiological processes including organ development, cardiovascular function, mood, cognition, multicellularity, cellular motility, immune responses and sensation of light, taste, and odor. However, many GPCRs are expressed poorly, and a significant proportion have unknown ligands and unclear signaling pathways. GPCRs are better suited to be targeted by monoclonal antibodies (mAbs) because of the challenges encountered in small-molecule discoveries such as druggability, selectivity, and distribution. mAbs have better drug-like properties in these respects. Herein, the authors review previously discovered functional mAbs that target GPCRs that are in the clinic and/or in development. They also review the biophysical considerations that make GPCRs so challenging to work with but also provide opportunities for biologic druggability. GPCRs are proven targets of small molecules yet remain an under-represented target of biologics. We believe that antibody drugs that target GPCRs have the potential to unlock new therapeutic avenues and also uncover previously unappreciated receptor biology, particularly when harnessing next-generation biologic modalities.

Abstract:G protein-coupled receptors (GPCRs) are a superfamily of transmembrane signal transducers that facilitate the flow of chemical signals across membranes. GPCRs are a desirable class of drug targets, and the activation and deactivation dynamics of these receptors are widely studied. Multidisciplinary approaches for studying GPCRs, such as downstream biochemical signaling assays, cryo-electron microscopy structural determinations, and molecular dynamics simulations, have provided insights concerning conformational dynamics and signaling mechanisms. However, new approaches including biosensors that use luminescence- and fluorescence-based readouts have been developed to investigate GPCR-related protein interactions and dynamics directly in cellular environments. Luminescence- and fluorescence-based readout approaches have also included the development of GPCR biosensor platforms that utilize enabling technologies to facilitate multiplexing and miniaturization. General principles underlying the biosensor platforms and technologies include scalability, orthogonality, and kinetic resolution. Further application and development of GPCR biosensors could facilitate hit identification in drug discovery campaigns. The goals of this review are to summarize developments in the field of GPCR-related biosensors and to discuss the current available technologies.

Abstract:<p>Given their extensive role in cell signalling, GPCRs are significant drug targets; despite this, many of these receptors have limited or no available prophylaxis. Novel drug design and discovery significantly rely on structure determination, of which GPCRs are typically elusive. Progress has been made thus far to produce sufficient quantity and quality of protein for downstream analysis. As such, this review highlights the systems available for recombinant GPCR expression, with consideration of their advantages and disadvantages, as well as examples of receptors successfully expressed in these systems. Additionally, an overview is given on the use of detergents and the styrene maleic acid (SMA) co-polymer for membrane solubilisation, as well as purification techniques.</p>

Abstract:Structures of 70 unique G protein-coupled receptors (GPCRs) have been determined, with over 370 structures in total bound to different ligands and the receptors in various conformational states. Structure-based drug design has been applied to an increasing number of GPCR targets over the past decade and now a few of these drug candidates have entered clinical trials. Given the length of time required for a drug to reach the market, there are no documented examples of licensed drugs being developed with the aid of a structure, but this is likely to change as current efforts come to fruition.

Abstract:G protein-coupled receptors (GPCRs) constitute a superfamily of membrane proteins which have in common the presence of 7 transmembrane α-helices linked by several intra- and extracellular loops. This particular structure is important for extracellular stimulus recognition through the amino acids forming the extracellular loops (class B, C and F of GPCRs) or by the transmembrane α-helices (class A and B of GPCRs). Their structure is also essential for signal transduction via intracellular proteins (G proteins, arrestins and kinases – GRKs-) [ Citation 1 ]. These proteins are attached to the intracellular loops and/or to the intracellular regions of the α-helices of the GPCRs, even when they are not activated by extracellular ligands or drugs. When GPCRs are activated, there is a modification of their protein structure that allosterically transmits the signal from the binding pocket to those intracellular effector proteins [ Citation 2 , Citation 3 ]. Importantly, GPCRs participate in more than 80% of the signal transduction processes that occur across plasma membrane. As more than 800 GPCRs have been identified in the human genome, this superfamily of receptors constitutes the largest protein superfamily codified in our genome. Most of these receptors are activated by sensory stimuli, mainly smell, and around 360 are stimulated by extrasensorial ligands, from small molecules to large proteins. Furthermore, more than 90% of GPCRs are expressed in the CNS and, thereby, are involved in the vast majority of neuronal functions [ Citation 4 ]. Their protein structure alteration by polymorphisms, mutations and posttranslational modifications, among others, can affect their ability to be activated by extracellular signals and/or their capacity to transduce these signals intracellularly. Therefore, structural alterations have been associated with many pathologies, including degenerative, neurological, inflammatory and cancerous [ Citation 5 ]. Moreover, alterations in protein structure can be associated with a decrease in drug therapeutic efficacy and with the appearance of patient-related side effects [ Citation 6 ]. Given the aforementioned, currently, GPCRs constitute the most important superfamily of membrane proteins pharmacologicaly targeted in clinical practice. Interestingly, among all the drugs approved by the Food and Drug Administration (FDA), around 35% target GPCRs [ Citation 7 , Citation 8 ]. Until 2017, the FDA and the European Medicines Agency (EMA) had approved 475–704 drugs for 108–134 different GPCRs, mainly aminergic and opioid receptors (e.g. cariprazine, brexpiprazole, naloxegol, or naldemedine) [ Citation 7 , Citation 8 ]. Moreover, there are 321 drugs targeting GPCRs in clinical trials (e.g. LJPC-501, INT-767, or RX-10045), 66 of which acting on GPCRs that are not currently a target of any approved drug [ Citation 7 ]. Between 2011 and 2015 the sales of drugs acting on GPCRs amounted to 917 billion USD [ Citation 9 ]. The development of such a great number of dugs has been achieved applying classical and modern techniques and methodologies along with in-silico and experimental approaches or a combination of both. These drugs activate, block or modulate (increasing or decreasing) the function of GPCRs targeting their orthosteric or allosteric centers (see section 4). In 2017, Gloriam and collaborators [ Citation 7 ] published a study about drugs acting on GPCRs approved or in clinical trials. These authors pointed out that there is a tendency to search for drugs more selective than classical drugs that usually have a polypharmacology with an average of 4–10 possible targets. They also state that aproximately 56% of non-sensory GPCRs are clinically unexplored. Among them, a substantial proportion (27%) are orphan GPCRs, which are receptors lacking known endogenous ligands. Therefore, the study of function and pharmacology of orphan GPCRs could potentially unravel new targets for the treatment of numerous pathologies. Trends in Pharmacological Sciences

Abstract:G protein-coupled receptors (GPCRs) are targets for ∼35% of approved drugs but only ∼15% of the ∼800 human GPCRs are currently such targets. GPCRomics, the use of unbiased, hypothesis-generating methods [e.g., RNA-sequencing (RNA-seq)], with tissues and cell types to identify and quantify GPCR expression, has led to the discovery of previously unrecognized GPCRs that contribute to functional responses and pathophysiology and that may be therapeutic targets. The combination of GPCR expression data with validation studies (e.g., signaling and functional activities) provides opportunities for the discovery of disease-relevant GPCR targets and therapeutics. Here, we review insights from GPCRomic approaches, gaps in knowledge, and future directions by which GPCRomics can advance GPCR biology and the discovery of new GPCR-targeted drugs.

Abstract:G-protein-coupled receptors (GPCRs) mediate many important physiological functions and are considered as one of the most successful therapeutic targets for a broad spectrum of diseases. The design and implementation of high-throughput GPCR assays that allow the cost-effective screening of large compound libraries to identify novel drug candidates are critical in early drug discovery. Early functional GPCR assays depend primarily on the measurement of G-protein-mediated 2nd messenger generation. Taking advantage of the continuously deepening understanding of GPCR signal transduction, many G-protein-independent pathways are utilized to detect the activity of GPCRs, and may provide additional information on functional selectivity of candidate compounds. With the combination of automated imaging systems and label-free detection systems, such assays are now suitable for high-throughput screening (HTS). In this review, we summarize the most widely used GPCR assays and recent advances in HTS technologies for GPCR drug discovery.

Abstract:G protein-coupled receptors (GPCRs) represent one of the largest families of cell surface receptors, and are the target of more than half of the current therapeutic drugs on the market. When activated by an agonist, the GPCR undergoes conformational changes that facilitate its interaction with heterotrimeric G proteins, which then relay signals to downstream intracellular effectors. Although GPCRs were thought to function as monomers, many studies support the hypothesis that G protein coupling involves the formation of GPCR homo- and/or hetero-complexes. These complex systems have been suggested to exhibit specific signaling cascades, pharmacological, internalization, and recycling properties. In this review, we summarize recent advances in our understanding of the structure, function and dynamics of GPCR complexes, as well as the findings obtained in animal models.

Abstract:G protein–coupled receptors (GPCRs) are important pharmaceutical targets, working as entry points for signaling pathways involved in metabolic, neurological, and cardiovascular diseases. Although small molecules remain the major GPCR drug type, biologic therapeutics, such as peptides and antibodies, are increasingly found among clinical trials and Food and Drug Administration (FDA)-approved drugs. Here, we review state-of-the-art technologies for the engineering of biologics that target GPCRs, as well as proof-of-principle technologies that are ripe for this application. Looking ahead, inexpensive DNA synthesis will enable the routine generation of computationally predesigned libraries for use in display assays for the rapid discovery of GPCR binders. Advances in synthetic biology are enabling the increased throughput of functional GPCR assays to the point that they can be used to directly identify biologics that modulate GPCR activity. Finally, we give an overview of adjacent technologies that are ripe for application to discover biologics that target human GPCRs.

Abstract:Cells in a living organism must communicate with each other through continuously sending and receiving messages. G-protein coupled receptors (GPCRs) are the largest family of communicating molecules at the cell surface. They transmit diverse extracellular signals, ranging from light and small chemical hormones to large peptide and protein hormones, and as such they play crucial roles in numerous physiological and pathological processes. More importantly, GPCRs are the most successful class of drug targets that are relevant to many major diseases, including cancer, heart failure, and inflammatory diseases. Over 50% of currently used drugs are targeted to GPCRs. However, these drugs target only 50–60 GPCRs, leaving the majority of human GPCRs, exceeding 800, unexplored for drug discovery. Given the prominent roles of GPCRs in biology and their successful track records as drug targets, GPCRs have become a hot frontier in basic research of life science and therapeutic discovery of translational medicines. In this special issue, there are nine exciting reviews that cover a broad scope of GPCR structures, biology, diseases, and drug discovery. Among them, four reviews are dedicated to GPCR structures. Over the last few years, we have witnessed a revolution in Class A GPCR structural biology. Rhodopsin is the founding member of the GPCR family and its signaling mechanism is a paradigm for many other GPCRs. The crystal structure of bovine rhodopsin was first solved in 2000 and since then structures of rhodopsin have been solved in several functional states, including the inactivated dark state, partially active opsin, and the fully active state that is bound with a Gαt peptide. ZHOU, MELCHER, and XU review the signaling mechanisms gained from rhodopsin structures and compare these mechanisms to other members of Class A GPCRs, most notably β2-adrenergic receptor (β2AR)1. Class B and C GPCRs distinguished themselves from Class A GPCRs by having a large extracellular domain for binding of ligands. Compared to Class A GPCRs, structure determination for the full-length receptors of other families of GPCRs is lagging behind. However, structures for a half dozen of Class B and C GPCR extracellular domains (ECDs) in complex with their respective ligands have been determined. The structures and ligand binding mechanisms of Class B and Class C GPCRs will be reviewed by the XU and LIU groups, respectively2, 3. The rapid explosion of Class A GPCR structures is owed to technology development in various stages of structure determination; including protein expression, purification, crystallization, and X-ray diffraction, for which a thorough review is provided by ZHAO and WU4. Following the reviews of GPCR structures, five reviews are focused on GPCR biology, diseases, and drug discovery. β2AR is a prototype of GPCRs, and in parallel to rhodopsin, it has been serving as a model for studying other GPCRs. WOO and XIAO review the signaling mechanisms by β-adrenergic receptor subtypes that provide a basis for developing selective biased β2 agonists for the treatment of heart failure5. Moving from heart to immune systems, YE and SUN review the role of GPCRs in inflammation processes from chemotaxisis to transcriptional regulation of inflammation programs6. Lappano and Maggiolini provide a systematic review on the relationship of cancers with various subfamilies of GPCRs, which may serve as the basis for developing novel pharmacological interventions for cancers7. A large number of GPCRs are orphan receptors whose ligand remains unknown. Despite missing cognate ligands, many orphan GPCRs are known to play important physiological functions, mostly from genetic knockout studies. LIU and colleagues provide a thorough review on different class of orphan nuclear receptors, their functions, and possible strategies for identification of endogenous ligands8. Finally, as one of the most important drug targets, GPCRs are widely pursued by both academic and industrial research for drug discovery. ZHANG and XIE provide a comprehensive review on methods of GPCR drug discovery, including various ligand binding and cell-based functional assays, many of which are tool kits for basic research as well as drug discovery for GPCRs9. Followed the waves of technological advances and inter-disciplinary approaches in modern biological research, the field of GPCRs is evolving rapidly into a new phase of exciting progresses. With a rich history of biology and drug discovery, GPCR research is expected to continue to be a dominant source of innovative medicines for the 21st Century. We hope that the collection of these nine exciting reviews will provide a window into this new era of GPCR research and drug discovery.
