Abstract:Saline-sodic soil is widely distributed around the world and has induced severe impacts on ecosystems and agriculture. Plant microbial desalination cell (PMDC) and soil microbial desalination cell (SMDC) were constructed to migrate excessive salt in the soil in this study. Compared with SMDC, PMDCs generated higher voltage ranging from 150 mV to 410 mV (500Ω) and the maximum power density reached 34 mW/m2. Higher desalinization efficiency was obtained by PMDCs, the soil conductivity reduced from initial 2.4 mS/cm to 0.4 ± 0.1 mS/cm and pH decreased from initial 10.4 to 8.2 ± 0.1. Soils desalination in PMDCs was achieved through multiple pathways, including ion migration in PMDCs driven by electrokinetic process, plant absorption and bioremediation by plant roots and anode microorganism activity. Geobacter was the dominant electrogenic bacteria at the PMDC anode. The electrochemical and desalinating performance of PMDCs was enhanced by plants and provided a new method for remediation of saline-sodic soil.

Abstract:Agricultural productivity is threatened worldwide because of salt-affected soils. Various remediation techniques have been successfully developed and are being utilized, but still there is no proper technical method under different conditions. In different situations, phytoremediation technically as well as economically is the best available option. More focussed efforts are required to measure the contribution of phytoextraction to the remedial procedure because the main mechanism behind salt phytoremediation is still not known. To improve the effectiveness and quality of the treatment of salt-affected soils, many new methods are used like mixing of treatment types, mixed plant cultures, biostimulation, etc. which can be extended to new methods like co-treatment and salt flow control measures. The new methods are in preliminary stages that require further research.
Laboratory investigation on solutes removal from artificial amended saline soil during the electrochemical treatment

Abstract:Soil salinity has become one of the environmental issues around the world, compromising thus land ecosystems as well as civil engineering infrastructures and facilities. This hazard is further aggravated, primarily when dealing with low permeable soils, where conventional remediation techniques are inadequate and mostly ineffective. Electrochemical treatment is an innovative and green technology for restoration of saline soils. This process has been proven to be the most efficient, promising technique, offering an optimal and sustainable remediation of salt-affected soils. The originality of this method involves application of an electric current through inserted electrodes into the soil matrix, inducing mobilization and transport of salts toward the electrodes of opposite charge via electro-migration process. The electrical current also induces a net water flow by electro-osmosis mechanism, allowing better draw and removal of solutes within the porous medium. This paper investigates the performance of the electrochemical treatment in remediation of salt-affected soil. Sodium, potassium and calcium were used as selective contaminants with targeted concentration of 0.5 M. In this aim, a laboratory bench scale was designed. It consists of soil box, anolyte and catholyte reservoirs, current monitoring devices, pH and electrical conductivity probe controller. The highest removal was for sodium and potassium with an extraction of 88 and 82%, respectively. Calcium exhibits lower removal rate of 57%, due to the development of the pH gradient, hindering therefore their mobility. This study demonstrated that electrochemical treatment can be advantageous in remediation of salt-affected soils, for enhancing agricultural productivity as well as soil constructional uses.

Abstract:Salination poses serious hazards to farmland soil. For the purpose of solving soil salination, desalination of water sources, and other problems faced by arid areas, a separation and desalination process for farmland saline-alkaline water is proposed; a separation and desalination device based on this process is also presented and tested. Results indicate that water associated with the pretreatment device satisfied the working conditions of the composite nanofiltration (NF)-reverse osmosis (RO) membrane system. The composite NF-RO membrane system produced a better filtering effect than either the NF membrane or the RO membrane. When used for filtering saline-alkaline water, the composite NF-RO membrane system achieved a desalination rate of 96.06%, a total hardness removal rate of 98.93%, and a Cl- removal rate of 99.32%, adhering to the standard for irrigation water quality. The flashing-condensation process realized a fresh water recovery rate greater than 70%. During brine evaporation using solar salt making processes, the primary compound of crystals precipitated was NaCl (with a relative content of 93%), suggesting that the precipitates have the potential values of industrial salts. These findings offer new technical references for solving the problem of farmland irrigation water faced by saline-alkaline areas worldwide.

Abstract:Salinization has become one of the main environmental problems affecting the worldwide ecological balance. The effluent discharge of saline industrial effluents, such as marine aquaculture, and its rapid increase in global production in the last decades has become an important part of this problem. The halophyte Sarcocornia neei (Lag.) has proven its ability to remove and assimilate nutrients from saline wastewater, making it a promising candidate for application in phytoremediation systems. In this study, its ability to reduce water salinity was evaluated. For this purpose, the plants were reared for 53 days in artificial effluents at four different salinities: A) 0, B) 10, C) 20, and D) 30 g L-1. A significant salt reduction was obtained in all treatments with NaCl addition. The maximum decrease was achieved in treatment C, reducing 6.91 ± 1.52 g L-1, approximately 35% of the total added at the beginning of the trials. Plants tolerated all salt content ranges; no mortality or salt stress symptoms were observed in any treatment. The results obtained are a first approach suggesting that S. neei could be used for bio-desalination of saline wastewater. Further studies are needed to evaluate its application in pilot systems integrated into saline industrial effluents.

Abstract:Due to the high salt content and pH value, the structure of saline-sodic soil was deteriorated, resulting in decreased soil fertility and inhibited soil element cycling. This, in turn, caused significant negative impacts on crop growth, posing a major challenge to global agriculture and food security. Despite numerous studies aimed at reducing the loss of plant productivity in saline-sodic soils, the knowledge regarding shifts in soil microbial communities and carbon/nitrogen cycling during saline-sodic soil improvement remains incomplete. Consequently, we developed a composite soil amendment to explore its potential to alleviate salt stress and enhance soil quality. Our findings demonstrated that the application of this composite soil amendment effectively enhanced microbial salinity resistance, promotes soil carbon fixation and nitrogen cycling, thereby reducing HCO 3 − concentration and greenhouse gas emissions while improving physicochemical properties and enzyme activity in the soil. Additionally, the presence of CaSO 4 contributed to a decrease in water-soluble Na + content, resulting in reduced soil ESP and pH by 14.64 % and 7.42, respectively. Our research presents an innovative approach to rehabilitate saline-sodic soil and promote ecological restoration through the perspective of elements cycles.

The accumulation of salts not only hampers crop growth and yield but also jeopardizes the livelihoods of millions who depend on agriculture for sustenance. Elevated salt levels in saline soils induce osmotic, ionic, oxidative, and water stress in plants. Implementing biological solutions offers the most dependable and sustainable method to safeguard food security while reducing reliance on agrochemicals which hampers various physiological and metabolic processes in plants. To ensure optimal plant growth under such changing conditions, Implementing biological solutions (Rhizobacteria) offers the most dependable and sustainable method to safeguard food security while reducing reliance on agrochemicals must be integrated into agricultural practices. This chapter concisely explores the mechanisms and utilization of beneficial microorganisms in both plants and soil to mitigate salt stress. Leaching efficiency and plant growth response in an integrated use of saline water for coastal saline soil reclamation

Abstract:A field experiment was carried out to evaluate an integrated use of saline water for saline soil reclamation around Bohai Bay in China. A landscape shrub (Caryopteris × clandonensis 'Worcester Gold') was cultivated and irrigated with five water salinities (ECi): 0.8, 3.1, 4.7, 6.3, and 7.8 dS·m−1. Drip irrigation was scheduled by evolving soil matric potential thresholds of −5, −10, −15, and −20 kPa in four years. A gravel-sand layer as subsurface drainage was installed at 1.0 m depth. After four years, the extremely saline soil (ECe of 27.8 dS·m−1) for 0–1.0 m depth was reclaimed to moderately saline soil (ECe of 4.1–7.2 dS·m−1). The decreases in survival rate, growth, and shoot dry weight of the plants as increasing ECi were observed. To achieve a relative survival rate of >50%, the ECi thresholds for plants were 7.8, 6.9, 5.5, and 5.3 dS·m–1 for the four years, respectively. The quantificationally estimating root zone drainage and salt leaching efficiency indicated that more efficient salt leaching offered by ECi up to 3.1 dS·m–1 in comparison with freshwater with ECi of 0.8 dS·m–1 and less efficiency occurred at ECi > 6.3 dS·m–1. The plants strongly responded to the different soil water and salinity regime in the dynamic leaching process, as thus it is recommended to use an inter-seasonal evolving matric potential threshold of −10 kPa for dry season of the third year, −15 kPa for rainy season and following dry season for providing optimized soil water and salinity condition.This article is protected by copyright. Abstract:Salinity, resulting from various contaminants, is a major concern to global crop cultivation. Soil salinity results in increased osmotic stress, oxidative stress, specific ion toxicity, nutrient deficiency in plants, groundwater contamination, and negative impacts on biogeochemical cycles. Leaching, the prevailing remediation method, is expensive, energy-intensive, demands more fresh water, and also causes nutrient loss which leads to infertile cropland and eutrophication of water bodies. Moreover, in soils co-contaminated with persistent organic pollutants, heavy metals, and textile dyes, leaching techniques may not be effective. It promotes the adoption of microbial remediation as an effective and eco-friendly method. Common microbes such as Pseudomonas , Trichoderma , and Bacillus often struggle to survive in high-saline conditions due to osmotic stress, ion imbalance, and protein denaturation. Halophiles, capable of withstanding high-saline conditions, exhibit a remarkable ability to utilize a broad spectrum of organic pollutants as carbon sources and restore the polluted environment. Furthermore, halophiles can enhance plant growth under stress conditions and produce vital bio-enzymes. Halophilic microorganisms can contribute to increasing soil microbial diversity, pollutant degradation, stabilizing soil structure, participating in nutrient dynamics, bio-geochemical cycles, enhancing soil fertility, and crop growth. This review provides an in-depth analysis of pollutant degradation, salt-tolerating mechanisms, and plant-soil-microbe interaction and offers a holistic perspective on their potential for soil restoration.

Abstract:Salt-affected soils have detrimental effects on agriculture and ecosystems. However, these soils can still be used for halophyte (salt-tolerant plants) cultivation using brackish and/or saline water. In this study, we employed soil technologies and mutualistic microorganisms as a sustainable strategy to improve the growth and reproduction of the halophyte Limonium algarvense Erben’s growth and reproduction under saline conditions. A microcosm assay was conducted under controlled greenhouse conditions to cultivate L. algarvense using a saline Fluvisol (FLU) amended—or not—with a Technosol (TEC). Plants were inoculated with the arbuscular mycorrhizal fungus (AMF) Rhizoglomus irregulare and/or a consortium of plant growth-promoting bacteria (PGPB), and they were irrigated with estuarine water. Soil enzyme analysis and physicochemical characterisation of the soils, collected at the beginning and at the end of the assay, were carried out. The physiological status of non-inoculated and inoculated plants was monitored during the assay for 4 months, and AMF root colonisation was evaluated. In FLU, only plants inoculated with the AMF survived. These plants had lower number of leaves, and shoot and root dry biomass than the ones grown in the TEC by the end of the assay. In the TEC, PGPB inoculation led to higher NDVI and PRI values, and AMF inoculation promoted higher reproductive development but not pollen fertility. The findings show that the combined use of soil and microbial technologies can be successfully applied to cultivate L. algarvense, suggesting their generalized use for other Limonium species with economic interest, while contributing to the sustainable use of marginal lands.

Abstract:— Saline and sodic soil sample was collected from salty beach. Polyvinyl alcohol borate hydrogel due to its cation attracting ability was utilized as an adsorbent for the removal of excess cations responsible for the salinity and sodicity of soil. Experimental data were collected before treatment and after each treatment using 1 : 2 by weight soil to water extract method. Electrical conductivity and pH were observed. The amounts of sodium, potassium, magnesium, and calcium ions were determined by flame photometry and atomic absorption spectrometry. From data it was evaluated that there is the decrease in electrical conductivity and amounts of cations after desalinating soil that was previously categorized as saline. Sodium adsorption ratio and exchangeable sodium percentage were also determined and found to decrease after treatment of soil sample with polyvinyl alcohol borate hydrogel. After treatment, soil becomes nonsaline but remains sodic. This was due to the fact that it has the higher sodium ion content in comparison with other cations. Thus, further remediation to decrease the sodicity of soil was suggested before its application for agricultural purposes.

Abstract:Salinity affects over 33% of irrigated farmland globally. Developing a low-cost, safe, and effective material as a soil salinity mitigation option would be of significant importance. This study proposed to synthesize a hydrogel using liquefied biomass from sugarcane bagasse, polyvinyl alcohol, and sodium tetraborate decahydrate. The effectiveness of the produced hydrogel in mitigating soil salinity was evaluated based on an incubation experiment at two salinity levels (5 and 10 dS m-1). The experiment was conducted by mixing liquefied hydrogel with soil at four application rates (0, 1, 2, and 3% w/w) with three replications. Porewater and soil samples were tested for pH and electrical conductivity (EC). Soil samples were also analyzed for selected cations and anions. The results demonstrated that hydrogel significantly reduced porewater EC at both 5 and 10 dS m-1 salt solutions. In addition, hydrogel reduced Cl-, P, Ca2+, and Al3+ concentrations in soil samples with maximum reductions observed from 3% hydrogel treatment. However, pH of porewater showed a consistent increase with hydrogel application. The application of hydrogel also increased NH4–N at high salt level. Overall, hydrogel has shown promising results in reducing soil salinity and could potentially be used as a soil amendment for saline soils.

Exploring the growth and phytoremediation efficacy of Suaeda fruticosa in agricultural soil contaminated by shrimp aquaculture

Abstract:Plants face numerous environmental challenges from biotic and abiotic stressors, with soil salinization emerging as a significant global concern. The coastal regions of Tamil Nadu, face severe environmental challenges due to discharge of saline water from shrimp farms exacerbates this issue, compromising the viability of paddy and other crops in the vicinity. This study explores the phytoremediation potential of Suaeda fruticosa in addressing soil salinity resulting from shrimp farming activities under field conditions over a 120-day period to restore soil health in salt affected soil. This research demonstrates Suaeda fruticosa's exceptional salt tolerance and bioaccumulation potential in facilitating soil restoration. Significant enhancements were observed in various growth parameters, including 466% increase in plant height, 338% in fresh weight and 387% in dry weight. Biochemical parameters also showed substantial enhancements with total chlorophyll, protein, proline, phenol, and glycinebetaine levels increasing by 655%, 588%, 690%, 153%, and 531%, respectively. Enzymatic activities exhibited notable elevations as well, with catalase, peroxidase, and polyphenol oxidase activities escalating by 258%, 587%, and 121% respectively, indicating robust adaptation to saline environments. Moreover, Suaeda fruticosa exhibited remarkable bioaccumulation capabilities, accumulating 461 kg NaCl ha −1 . This led to substantial improvements in soil characteristics, including a reduction in pH from 8.8 to 6.49, electrical conductivity from 5.7 to 1.53 dSm −1 , and sodium adsorption ratio from 16.1 to 4.4 mmol L −1 . The successive cultivation of Suaeda fruticosa in this study, has proven to be a viable strategy for reclaiming salt-affected lands, thereby alleviating a significant constraint on crop productivity. In this phytoremediation study, Suaeda fruticosa can effectively bioaccumulate considerable amounts of NaCl from the soil, leading to marked improvements in soil quality. This includes reductions in soil pH, electrical conductivity, and sodium adsorption ratio. A salt marsh halophyte, Suaeda fruticosa not only accumulates salt but also adapts to such conditions by synthesizing substantial amounts of organic solutes and enhancing its antioxidant machinery to mitigate toxicity induced by ionic stress. This research underscores a promising and sustainable approach for the reclamation of saline-affected agricultural soils, offering a viable solution to enhance crop productivity in coastal regions impacted by shrimp aquaculture.

A biomimetic interfacial solar evaporator for heavy metal soil remediation

Abstract:Interfacial solar evaporation is a sustainable and efficient way to enhance water evaporation during seawater desalination and wastewater treatment. In this work, this concept is applied for heavy metal (HM) soil remediation, which is a major global challenge currently limiting food production and endangering human health. Inspired by natural transpiration remediation processes in plants, here a tree inspired evaporator structure is used to accelerate the extraction of HM containing liquids from soil. Coating of simple biomimetic leaf structures with reduced graphene oxide (rGO) can realize a high evaporation rate (3.1 kg m−2h−1) under 1.0 sun illumination. Four-week indoor solar evaporation tests of the evaporator shows that evaporation performance is generally stable and HMs (As, Cd, Cr, Pb, and Zn) in soil pore water are reduced to varying degrees. After solar driven evaporative remediation (SDER) a plant assay in the treated soil shows that relative to an untreated control soil, HM content in broccoli roots decreases by 37.2, 56.4, 25.9, 46.3 and 26.9% for As, Cd, Cr, Pb and Zn, respectively. This work successful demonstrates a new direction in solar evaporator design by enhancing water evaporation to achieve efficient soil contaminant remediation.

Enhanced Salt Tolerance in Synechocystis sp. PCC 6803 Through Adaptive Evolution: Mechanisms and Applications for Environmental Bioremediation

Abstract:Salt stress is common in natural environments, where elevated salt levels in brackish water and saline soil can hinder the growth of organisms, thereby exacerbating environmental challenges. Developing salt-tolerant organisms not only uncovers novel mechanisms of salt tolerance but also lays the groundwork for managing and utilizing saline environments. Cyanobacteria, which are widely distributed in hydrosphere and soil, serve as ideal models for studying salt stress. In this study, the model cyanobacterium Synechocystis sp. PCC 6803 was selected, whose salt (NaCl) tolerance improved from 4.0% to 6.5% (m/v) through adaptive laboratory evolution. Genome re-sequencing and mutant analysis identified six key genes associated with salt tolerance. Notably, the deletion of slr1670, which encodes glycerol glucoside hydrolase, improved the strain's salt tolerance. In addition, slr1753 encodes a membrane protein that may enhance salt tolerance by facilitating ion transport to the extracellular space. Further analysis revealed that overexpression of slr1753 significantly accumulates Na+ on the cell surface, enabling effective seawater treatment using the engineered strain, resulting in a 6.35% reduction of Na+ in the seawater. Moreover, the adapted bacteria can be used for the remediation of saline soil samples, leading to a 184.2% and 43.8% increase in the germination rate and average height of Brassica rapa chinensis, respectively, along with a 25.3% rise in total organic carbon content and reductions in both total salt content by 1.82% and pH by 1.91% in soil. This study provides novel insights into salt tolerance mechanisms and the bioremediation of high-salinity environments.

Abstract:Coastal salt-affected soils account for a large area all around the world. Soil salinity and pH are two important parameters affecting soil quality. Investigating the correlation of electrical conductivity (EC) and pH at different soil depths in saline soil was useful for quickly assessing the saline–alkaline characteristics. During the natural desalination process in the field area of reclaimed lands, the phenomena of pH increase and nitrogen accumulation may occur. A field sampling experiment was conducted in slightly saline soil affected by natural desalination and newly reclaimed heavily saline soil. A series of soil–water ratio extracts consisting of 1:2.5, 1:5, 1:10, 1:20, and 1:40 was designed to measure the EC and pH for simulating the saline–alkaline characteristics during the soil desalination process. Meanwhile, for reasonable utilization of the naturally ameliorated slightly saline soil which consists of a high content of nitrogen, a plastic mulching (PM) accompanied with nitrogen (N) fertilizer addition experiment in maize cultivation plots was designed. Results showed that a significant correlation of EC and/or pH existed in all ratios of soil extracts, and the slightly saline soil had a higher nitrogen content (1.06 g kg−1). The EC was negatively correlated with pH at a depth of 0~100 cm in the coastal saline soil, which indicated the increase of pH value and alkalization during its natural desalination. Furthermore, PM treatments showed no significant difference with N treatments in soil bulk density and soil water content in the slightly saline soil. The PM and N treatments obtained similar grain yield, which was between 6.2 and 6.5 t ha−1. The soil salinity decreased in all treatments and the harvest index was largest in PM treated plots. Our study was beneficial for rapidly monitoring saline–alkaline characteristics and sustainable utilization of coastal saline soil resources. In addition, we should focus far more on pH improvement during the desalination process and rational utilization of chemical fertilizer for obtaining sustainable benefits in the coastal saline soil.
Ensifer sp. GMS14 enhances soybean salt tolerance for potential application in saline soil reclamation

Abstract:Rhizosphere microbiomes play an important role in enhancing plant salt tolerance and are also commonly employed as bio-inoculants in soil remediation processes. Cultivated soybean ( Glycine max ) is one of the major oilseed crops with moderate salt tolerance. However, the response of rhizosphere microbes me to salt stress in soybean, as well as their potential application in saline soil reclamation, has been rarely reported. In this study, we first investigated the microbial communities of salt-treated and non-salt-treated soybean by 16S rRNA gene amplicon sequencing. Then, the potential mechanism of rhizosphere microbes in enhancing the salt tolerance of soybean was explored based on physiological analyses and transcriptomic sequencing. Our results suggested that Ensifer and Novosphingobium were biomarkers in salt-stressed soybean. One corresponding strain, Ensifer sp. GMS14, showed remarkable growth promoting characteristics. Pot experiments showed that GMS14 significantly improved the growth performance of soybean in saline soils. Strain GMS14 alleviated sodium ions (Na + ) toxicity by maintaining low a Na + /K + ratio and promoted nitrogen (N) and phosphorus (P) uptake by soybean in nutrient-deficient saline soils. Transcriptome analyses indicated that GMS14 improved plant salt tolerance mainly by ameliorating salt stress-mediated oxidative stress. Interestingly, GMS14 was evidenced to specifically suppress hydrogen peroxide (H 2 O 2 ) production to maintain reactive oxygen species (ROS) homeostasis in plants under salt stress. Field experiments with GMS14 applications showed its great potential in saline soil reclamation, as evidenced by the increased biomass and nodulation capacity of GMS14-inoculated soybean. Overall, our findings provided valuable insights into the mechanisms underlying plant-microbes interactions, and highlighted the importance of microorganisms recruited by salt-stressed plant in the saline soil reclamation.

Abstract:Solar-driven interfacial evaporation is an efficient, green, and low-carbon footprint way to separate heavy metals from contaminated soil. A biomimetic mushroom-like evaporator based on polypyrrole-modified cotton fabric is designed, which uses the mechanism of evaporative diffusion and gravity to drive the separation of heavy metal salt crystals, achieving sustainable remediation heavy metal contaminated soil (HMCS). This stereo biomimetic evaporator not only achieves excellent evaporation performance under solar energy drive but also enhances evaporation efficiency by capturing environmental energy from the side and then using natural convection. At an air velocity of 6 m s −1 , the evaporation rate reaches 2.32 kg m −2 h −1 . After continuous operation for 192 h, the biomimetic evaporator maintains stable evaporation performance and successfully separates metal salt pollutants from the washing solution of HMCS. The removal rates of heavy metal ions Pb 2+ , Cr 3+ , Cd 2+ , Zn 2+ , and Hg 2+ in polluted soil are 53 %, 59 %, 60 %, 62 %, and 84 %, respectively. Compared to untreated control soil, the concentrations of Pb 2+ , Hg 2+ , Cr 3+ , Zn 2+ , and Cd 2+ in the roots of pea treated with solar evaporation remediation decreased by 66.4 %, 31.6 %, 31.1 %, 29.0 %, and 8.3 %, respectively. This study provides a potential new approach and method for the sustainable, low-cost, and low-energy remediation of HMCS using solar evaporation technology.

Remediation of organic amendments on soil salinization: Focusing on the relationship between soil salts and microbial communities

Abstract:Soil salinization has emerged as a major factor with an adverse influence on agricultural green development worldwide. It is necessary to develop high-efficiency and ecologically beneficial management measures for alleviating soil salinization. The experiment of application for cow manure (CM), biochar (BC), and bio-organic fertilizer (BIO) in soil with light salinity was conducted to investigate the remediation of organic materials on soil salinization with melon (Cucumis melo L.) by reducing the availability of saline ions and shifting the soil microbial community. Results showed that BC treatment significantly decreased the EC values of the soil and soil solution by 19.23% and 27.02% and the concentrations of Na+, K+, and Cl- by 13.28%, 13.08%, and 15.21%, respectively, followed by CM and BIO treatments. High-throughput sequencing identified that organic amendments significantly improved the richness of the soil bacterial community and increased the relative abundances of Acidobacteria and Firmicutes by 33.11% and 111.2%, respectively, and the beneficial salt-tolerant bacterial genera Flavobacterium, Bacillus and Arthrobacter by 32.04%, 38.92% and 35.67%, respectively. Additionally, soil Na+, Ca2+, K+ and Cl- were significantly negatively correlated with Acidobacteria and Flavobacterium and were also the most important factors driving the changes in the structure of the soil bacterial communities. The bacterial networks were more complex in the organic amendments treatments than in CK, reflecting through more nodes and links and a higher average clustering coefficient, density and modularity. This study provided a comprehensive understanding of the application of organic amendments in alleviating soil salinization and improving soil bacterial and fungal communities and provides scientific support for agriculture green development.
A sustainable approach for bioremediation of secondary salinized soils: Studying remediation efficiency and soil nitrate transformation by bioaugmentation

Abstract:Nitrate is the main nitrogen source for plant growth, but it can also pollute the environment. A major cause of soil secondary salinization is the rising level of nitrates in the soil, which poses a threat to the sustainability and fertility of global greenhouse soils. Herein, Bacillus megaterium NCT-2 was used as a microbial agent to remove nitrate by bioaugmentation, and the remediation efficiency of secondary salinized soil in different degrees was evaluated. The findings showed that the highest nitrate removal rate of 62.76% was in a medium degree of secondary salinized soil. Moreover, the results of 16S rRNA high-throughput sequencing and quantitative real-time PCR (qPCR) demonstrated that NCT-2 agent reduced the microbial diversity, increased the microbial community stability, and changed the composition and function of the microbial community were changed by NCT-2 agent in all districts soil. Further analysis demonstrated that the NCT-2 bacterial agent significantly increased the key enzyme genes of the assimilation pathway (nitrite reductase gene NasD, 87–404 times, and glutamine reduction enzyme gene GlnA, 13–52 times) and dissimilatory reduction to ammonium (DNRA) (nitrate reductase gene NarG, 14–56 times) in different degrees of secondary salinized soils. This proved that NCT-2 agent could promote the nitrate assimilation and the dissimilation and reduction to ammonium in secondary salinized soil. Thus, the current findings suggested that the NCT-2 agent has a significant potential for reducing excessive nitrate levels in secondary salinized soil. The remediation efficiency was related to the microbial community composition and the degree of secondary salinization. This study could provide a theoretical basis for the remediation of secondary salinized soil in the future.
