Abstract:We investigate the field-effect control of ions in nanofluidic transistors (NFTs) with characteristic channel size (~100 nm) significantly larger than the system's Debye Length (~10 nm). These 100 nm NFTs achieve an ionic current modulation ratio of ~2.5, demonstrating better performance than the state-of-the-art 20 nm NFTs. The result attests a new operating regime beyond the Debye-screening limit. The relaxed constraint on channel size offers advantages in device manufacturing, testing, and reliability. It also opens up new applications in biological sensing and sample preparation.

Abstract:Applications of electric field, using either dc or high-frequency ac field, have shown many advantages in emulsification. We further develop this technique by a detailed study on low-frequency ac electro-flow-focusing (EFF) microfluidic emulsification. Counter-intuitively, the droplet size variation is not monotonic with the electric field, in contrary to the dc-EFF emulsification. This phenomenon originates from a relaxation oscillation of flow rate through the Taylor cone. Particularly, a continuous droplet size decrease was obtained at the voltage ramp-up stage. This emulsification process was modeled in analog to the accumulation and release of charges in an RC electric circuit with an adjustable resistor.
On controlling the flow behavior driven by induction electrohydrodynamics in microfluidic channels

Abstract:In this study, we develop a nondimensional physical model to demonstrate fluid flow at the micrometer dimension driven by traveling-wave induction electrohydrodynamics (EHD) through direct numerical simulation. In order to realize an enhancement in the pump flow rate as well as a flexible adjustment of anisotropy of flow behavior generated by induction EHD in microchannels, while not adding the risk of causing dielectric breakdown of working solution and material for insulation, a pair of synchronized traveling-wave voltage signals are imposed on double-sided electrode arrays that are mounted on the top and bottom insulating substrate, respectively. Accordingly, we present a model evidence, that not only the pump performance is improved evidently, but a variety of flow profiles, including the symmetrical and parabolic curve, plug-like shape and even biased flow behavior of quite high anisotropy are produced by the device design of "mix-type", "superimposition-type" and "adjustable-type" proposed herein as well, with the resulting controllable fluid motion being able to greatly facilitate an on-demand transportation mode of on-chip bio-microfluidic samples. Besides, automatic conversion in the direction of pump flow is achievable by switching on and off a second voltage wave. Our results provide utilitarian guidelines for constructing flexible electrokinetic framework useful in controllable transportation of particle and fluid samples in modern microfluidic systems.

Numerical investigation of field-effect control on hybrid electrokinetics for continuous and position-tunable nanoparticle concentration in microfluidics

Abstract:We introduce herein an effective way for continuous delivery and position-switchable trapping of nanoparticles via field-effect control on hybrid electrokinetics (HEK). Flow field-effect transistor exploiting HEK delicately combines horizontal linear electroosmosis and transversal nonlinear electroosmosis of a shiftable flow stagnation line (FSL) on gate terminals under DC-biased AC forcing. The microfluidic nanoparticle concentrator proposed herein makes use of a simple device geometry, in which an individual or a series of planar metal strips serving as gate electrode (GE) are subjected to a hybrid gate voltage signal and arranged in parallel between a pair of 3D driving electrodes. On the application of a DC-biased AC electric field across channel length direction, all the GE are electrochemically polarized, and the action of imposed hybrid electric field on the multiple-frequency bipolar counterions within the composite-induced double layer generates two counter-rotating induced-charge electroosmotic (ICEO) micro-vortices on top of each GE. Symmetry breaking in ICEO flow profile occurs once the gate voltage deviates from natural floating potential of corresponding GE. The gate voltage offset not only results in an additional pump motion of working fluid for enhanced electroosmotic transport but also directly changes the location of FSL where nanoparticles are preferentially collected by field-effect HEK. Our results of field-effect control on HEK are supposed to guide an elaborate design of flexible electrokinetic frameworks embedding coplanar metal strips for a high degree of freedom analyte manipulation in modern micro-total-analytical systems.
An Enhanced Electroosmotic Micromixer with an Efficient Asymmetric Lateral Structure.

Abstract:Homogeneous and rapid mixing in microfluidic devices is difficult to accomplish, owing to the low Reynolds number associated with most flows in microfluidic channels. Here, an efficient electroosmotic micromixer based on a carefully designed lateral structure is demonstrated. The electroosmotic flow in this mixer with an asymmetrical structure induces enhanced disturbance in the micro channel, helping the fluid streams' folding and stretching, thereby enabling appreciable mixing. Quantitative analysis of the mixing efficiency with respect to the potential applied and the flow rate suggests that the electroosmotic microfluidic mixer developed in the present work can achieve efficient mixing with low applied potential.

Abstract:Accurate and fast control of fluid flow velocity and mixing is an essential technique for in vitro cell or microbial culture. Here, we propose a cross microchannel structure based on electroosmotic drive, which enables efficient cooperative dynamic control of fluid velocity and concentration. In this study, the effects of channel width, applied voltage, and waveform on fluid mixing and flow behavior in microchannels are investigated experimentally and numerically. We find that the flow velocity and the fluid mixing ratio can be efficiently regulated by the applied voltage. Moreover, reducing the width of one of the microchannels can significantly improve the control accuracy of the fluid mixing ratio in the system. We also find that various concentration and velocity waveforms can be generated by controlling the voltage waveform. Our results reveal some important features of electroosmotic-driven flow that could facilitate the design of electrically operated fluid devices.

Abstract:<p>We study herein numerically the use of induced‐charge electrokinetic phenomena (ICEK) to enable a flexible control of ion transport of dilute electrolyte in a straight ion concentration polarization (ICP) system. The effect of three convection modes of ICEK, including induced‐charge electroosmosis (ICEO), flow‐field effect transistor (flow‐FET) and alternating‐current electroosmosis (ACEO), on convective arrestment of diffusive wave‐front propagation is investigated by developing a cross‐scale and fully‐coupled transient numerical simulation model, wherein multiple frequency electrochemical polarization and nonlinear diffuse charge dynamics in spatiotemporally varying solution conductivity are taken into account. We demonstrate by detailed comparative simulation studies that, ACEO vortex flow field above a metal strip array arranged along the anodic chamber's bottom surface serves as the most efficient way for adjusting the salt density distribution at micrometer and even millimeter dimension, due to its high flexibility in controlling the stirring flow state with the introduction of two extra electrical parameters. The specific operating status is determined by whether the electrode array is floating in potential (ICEO), or biased to ground (flow‐FET) or forced to oscillate at another Fourier mode (ACEO). Abstract:External field driven fluid manipulation, in particular electric field, offers the advantages of real-time control and exceptional flexibility, rendering it highly promising for applications in microfluidic devices, liquid separation and energy catalysis. However, it is still challenging for controlled liquid transport and fine control of droplet splitting. Herein, we demonstrate a strategy to achieve direction-controlled liquid transport and fine droplet splitting on an anisotropic groove-microstructured electrode surface via an electrically switched asymmetric interface. The balance of asymmetric capillary force generated by microstructures and electro-capillary force is critical in determining directional liquid transport and fine droplet splitting. Asymmetric bubbles generated by liquid electrolysis form an asymmetric liquid–gas–solid interface and result in gradient liquid wetting behavior on the two neighboring electrode surfaces. The electric field further enhances the asymmetric wetting of a liquid droplet on the electrode surface, exhibiting electric field direction-dependent motion. Moreover, the groove-microstructured electrode surface can strengthen the liquid droplet anisotropic wetting and correspondingly refine the volume range of the splitting sub-droplet. Even unidirectional/bidirectional liquid droplet transport can be controlled in collaboration with the asymmetric groove-microstructure and electric field. Thus, this work provides a new route for liquid transport and droplet splitting, showing great potential in controllable separation, microreaction and microfluidic devices.

Interface dynamics in electroosmotic flow systems with non-Newtonian fluid frontiers

Abstract:Microfluidic applications involving liquid manipulation, selective membranes, and energy harvesting strongly em-phasize the importance of the electrokinetic phenomenon, which is widely used at multiple fluid and electrochemi-cal interfaces. However, critical scientific issues that address multifield coupling and multiscale physics have not been well addressed in non-Newtonian fluids. In this paper, electrical field–fluid flow–ion transport coupling is numerically implemented in two mainstream problems, i.e., induced electroconvection phenomena at ion-selective interfaces and induced charge electroosmosis in polarized cylinders. The effects of different non-Newtonian rheo-logical properties, which are absent in Newtonian fluids, on the interfacial dynamics, instability and ion transport are examined. The results reveal that the non-Newtonian rheology significantly modulates the statistical data and interfacial phenomena. Generalized power-law fluids alter velocity and interfacial charge profiles, with shear thin-ning enhancing ion transport to lower overlimiting current thresholds and shear thickening broadening the limiting current region (with hindered ion transport). In Boger-type Oldroyd-B fluids, the addition of polymer decreases the velocity amplitude and increases the interface resistance. At low voltages, polymer viscoelasticity minimally affects the ohmic and limiting regions, but under convection-dominated flow, different rheological parameters, such as the viscosity ratio, Weissenberg number, anisotropic parameter, and electrohydrodynamic coupling con-stants, enable controllable regulation of ion transport behavior across a wide range. Finally, this paper states that modulated electroosmosis by complex charged polymers is the future cutting edge. The relevant results supple-ment the non-Newtonian physics of electrokinetic systems and provide guidance for the design and operation of microfluidic devices.

LATTICE BOLTZMANN SIMULATION OF ELECTROOSMOTIC FLOWS IN MICROCHANNEL WITH A SUDDEN EXPANSION

Abstract:Great progress has been carried out on the Micro Total Analysis System (μTAS) in recent years, in which the chemical analysis system is miniaturized by micromachining. This system can reduce reagent consumption, dispersion effects and analysis time. Nowadays, μTAS devices are beginning to make significant contributions to biotechnology, biomedicine and drug discovery. Meanwhile, due to the difficulties of controlling fluid motion in such a small scale, fluid flow driven in microfluidic devices has received increasing attention. So far electroosmotic flow (EOF) is one of the most popular and important fluid driven techniques in microfluidic devices because in an EOF the velocity profile is plug-like and the mean velocity is independent of the cross-sectional area, as well as its valveless control of fluid flow. Therefore, electro-osmotic flow can offer significantly less deleterious dispersive effects than pressure-driven flow and provide a favored high-performance sample separation technique. Due to its importance, electroosmotic flow in microchannels has received considerable attentions recently and been investigated both numerically and experimentally. However, fluid flow in microfluidic devices is often through channels with changes in cross-sectional area, and/or channels with bends and bifurcations, which make a great difference to velocity profile, pressure distribution and so on. A systematic study of these different configurations will provide valuable insight into the flow physics. The goal of the present study is to undertake a numerical study of EOF in microchannels with a sudden expansion in order to identify the factors affecting the flow. The present work is in the use of an incompressible lattice-Boltzmann model with a forcing term which is perhaps better suited to microflows than the conventional Navier-Stokes solvers. The impact of expansion ratio (ER), ionic molar concentration (c ∞), external electric potential and zeta potential on the flows are investigated. Numerical results including velocity profile, pressure distribution and external electrical potential distribution have been presented. It has been shown that boundary layer separation has been suppressed by the electroosmotic force exerted on the fluid near walls. Moreover, a sudden change in the channel has a great influence on the electroosmotic flows. A jump has been found in fluid velocity, pressure drop at the junction, as well as electrical potential due to EDL. The external electrical potential is linear along the axis of the channel but a turning point emerges at the junction. Non-linear pressure drop in the narrow channel becomes more obvious as ER increases. Results also show that velocity profile at the outlet depends on ER and c ∞. The convex velocity profile turns to concave one as ER or c ∞ increases, resulted from interactions between adverse pressure gradient and electric force exerted on the fluid in the middle of channel.
Modulation of electroosmotic flows in electron-conducting microchannels by coupled quasi-reversible faradaic and adsorption-mediated depolarization

Abstract:A theoretical model is proposed for the description of steady electroosmotic flows within a cylindrical electron-conducting microchannel that is depolarized by faradaic and adsorption-mediated processes. The bipolar electron-transfer (e.t.) reactions are examined in the general situation where the electrolyte contains a quasi-reversible redox couple. The rate of the e.t. reactions is governed by transversal convective diffusion of the electroactive species to/from the surface and a position-dependent degree of reversibility. The nonuniform distribution of the electric field in solution, that is intimately coupled to that of the local faradaic current density, alters the double layer composition along the conducting surface via the occurrence of simultaneous electronic and ionic double layer charging processes. This in turn generates a nonlinear distribution of the zeta potential, which affects the electroosmotic flow. The highly coupled spatial profiles for the concentrations of the electroactive species, the faradaic current density, the electrokinetic potential, the electric field and the electroosmotic velocity in/along the metallic channel are solved by consistent numerical analysis of (i) the convective-diffusion equation, (ii) the generalized Butler-Volmer expression that includes mass transport and electron-transfer kinetic contributions, (iii) the continuity and Navier-Stokes equations, and (iv) the Poisson equation for finite currents. The results reported as a function of the surface properties of the channel and the kinetic characteristics of the e.t. reaction illustrate the deviations of the electroosmotic flow profiles as compared to the typical pluglike distribution predicted by Smoluchowski's equation and encountered for homogeneous and dielectric channels. Manipulation of the flow patterns by bipolar electrochemical means is a promising way to control and optimize the local detection and separation of electroactive molecules or molecules dyed with electroactive elements.
Reconfigurable liquid pumping in electric-field-defined virtual microchannels by dielectrophoresis

Abstract:Dielectrophoresis (DEP), widely used to generate body forces on suspended particles, is investigated to provide surface forces at the liquid-medium interfaces and pump a high-permittivity liquid in a low-permittivity medium along a virtual microchannel defined by an electric field between parallel plates. Because the pumping pressure is proportional to the square of the intensity of the electric field and independent of the channel width, DEP pumping is advantageous as the dimension of the microchannel shrinks down. The absence of the channel walls simplifies the fabrication processes and further increases its feasibility in nanofluidic applications. We demonstrate water pumping in an immiscible silicone oil medium at adjustable velocities by applying voltages above the threshold value whose square is linearly proportional to the cross-sectional aspect ratio (AR), i.e., the height to width ratio, of the microchannel. With a properly designed AR, liquid valve is achieved by appropriate voltage applications. Without the barriers of channel walls, merging multiple streams and capillary filling of the spacing between electric-field-defined virtual microchannels are observed and studied. Moreover, in situ reconfigurable liquid pumping is demonstrated by a four way switching valve on a programmable crossing electrode set.
A universal design of field-effect-tunable microfluidic ion diode based on a gating cation-exchange nanoporous membrane

Abstract:Based on the continuum mechanics theory, we propose herein a universal design of microfluidic ionic diode based on external concentration polarization of a gating ion-selective medium embedded in the microfluidic network with four power terminals. This micro/nanofluidic hybrid chip employs a cation-exchange nanoporous membrane (CEM) coupled with both a control and output microfluidic channel. Under the action of a vertical electric field throughout the CEM, nanoscale surface conduction of excessive counterions within the charged nanopores is converted to the propagation of either enriched or depleted boundary toward the opposing electrode-terminal in phase with the electroconvective flow, thereby making an adjustment in the electrical conductance of output microchannel for achieving high-flux field-effect current control and diode functionality. Three basic working states, including the “on,” “transition,” and “off” statuses, are distinguished in different ranges of source voltage magnitude. The rectification factor of the proposed ionic circuit platform can attain one hundred-fold even at small source and gate voltages. The presented field-effect-tunable microfluidic ion diode is easily scalable, permits appreciable fluid flow due to an intrinsically small hydrodynamic resistance, and holds promise for producing high-flux ion current rectification in next-generation integrated circuits.

Abstract:Free-surface electrokinetic flows have been attracting increasing attention from the research community over recent times, as attributable to their diverse fields of applications ranging from fluid mixing, particle manipulation to bio-chemical processing on a chip. Traditional electrokinetic processes governing free surface flows are mostly effective in manipulating fluids having characteristically low values of the electrical conductivity (lower than 0.085 S/m). Biological and biochemical processes, on the other hand, typically aim to manipulate fluids having higher electrical conductivities. To circumvent the problem of traditional electrokinetic processes in manipulating highly conductive fluids mediated by free surface flows, here we demonstrate a novel on-chip methodology for the same by exploiting the interaction between an alternating electric current and a thermal field. The consequent local gradients in electrical properties can be tuned to direct the flow towards a specific direction on the interface, opening up a new realm of on-chip process control without necessitating the creation of a geometric confinement. Similar concept of free-surface flow can be used to trigger the fluid along a open-channel flow

A Lectin from Platypodium elegans with Unusual Specificity and Affinity for Asymmetric Complex N-Glycans*

Abstract:Background: Legume lectins with new glycan specificity can be discovered in the Dalbergieae tribe. Results: We determined the sequence, specificity, and crystal structures of a new Man/Glc lectin (PELa) from Platypodium elegans seeds. Conclusion: The unusual affinity of PELa for asymmetrical complex N-glycans is related to the extended binding site and conformational constraints on oligosaccharides. Significance: Dalbergieae lectins are of interest for biotechnological applications. Lectin activity with specificity for mannose and glucose has been detected in the seed of Platypodium elegans, a legume plant from the Dalbergieae tribe. The gene of Platypodium elegans lectin A has been cloned, and the resulting 261-amino acid protein belongs to the legume lectin family with similarity with Pterocarpus angolensis agglutinin from the same tribe. The recombinant lectin has been expressed in Escherichia coli and refolded from inclusion bodies. Analysis of specificity by glycan array evidenced a very unusual preference for complex type N-glycans with asymmetrical branches. A short branch consisting of one mannose residue is preferred on the 6-arm of the N-glycan, whereas extensions by GlcNAc, Gal, and NeuAc are favorable on the 3-arm. Affinities have been obtained by microcalorimetry using symmetrical and asymmetrical Asn-linked heptasaccharides prepared by the semi-synthetic method. Strong affinity with Kd of 4.5 μm was obtained for both ligands. Crystal structures of Platypodium elegans lectin A complexed with branched trimannose and symmetrical complex-type Asn-linked heptasaccharide have been solved at 2.1 and 1.65 Å resolution, respectively. The lectin adopts the canonical dimeric organization of legume lectins. The trimannose bridges the binding sites of two neighboring dimers, resulting in the formation of infinite chains in the crystal. The Asn-linked heptasaccharide binds with the 6-arm in the primary binding site with extensive additional contacts on both arms. The GlcNAc on the 6-arm is bound in a constrained conformation that may rationalize the higher affinity observed on the glycan array for N-glycans with only a mannose on the 6-arm.

Abstract:Recently, single-molecule manipulation techniques in micro- and nanofluidic channels have attracted significant attention. To precisely control the transport velocity, the dynamics of the surrounding liquid must be understood in addition to the behavior of the target particles. Some unknowns about interactions between electrolyte ions and solvents remain to be clarified from a microscopic viewpoint. Herein, we propose a technique to generate a liquid flow driven by ion transport phenomena, the so-called electrohydrodynamic (EHD) flow, where electrolyte ions are dialyzed using a cation-exchange membrane. With this method, it is possible to apply an electric body force in liquids, which is different from electroosmotic flows that are limited to ion transport in electric double layers, and is expected to be a good candidate for detailed control of liquid flows in micro- and nanofluidic channels. To collect basic design data based on the knowledge of microscopic fluid dynamics of the present technique, a mathematical model of an EHD flow dragged by electrical carriers in an ionic current is developed and results are compared with experimental data. In our experiments, EHD flows are efficiently driven by applied electric fields in a cation dominant current. To induce such an EHD flow, the externally applied electric potential can be drastically reduced to 2.0 V in comparison with previous methods because we do not need an excessively high voltage to inject electrical charges into liquids. This method enables us to induce EHD flows in aqueous solutions and is expected to open the door to low-voltage driven liquid flow control.

Abstract:In this paper, we present a lab-in-a-droplet bioassay strategy for a centrifugal microfluidics or lab-on-a-disc (LOAD) platform with three important advancements including density difference pumping, power to disc and bidirectional flow control. First, with the water based bioassay droplets trapped in a micro-channel filled with mineral oil, centrifugal force due to the density difference between the water and oil phases actuates droplet movement while the oil based medium remains stationary. Second, electricity is coupled to the rotating disc through a split-core transformer, thus enabling on-chip real-time heating in selected areas as desired and wireless programmable functionality. Third, an inertial mechanical structure is proposed to achieve bidirectional flow control within the spinning disc. The droplets can move back and forth between two heaters upon changing the rotational speed. Our platform is an essential and versatile solution for bioassays such as those involving DNA amplification, where localized temperature cycling is required. Finally, without the loss of generality, we demonstrate the functionality of our platform by performing real-time polymerase chain reaction (RT-PCR) in a linear microchannel made with PTFE (Teflon) micro-tubing.
Analysis of Sequential Micromixing Driven by Sinusoidally Shaped Induced-Charge Electroosmotic Flow

Abstract:Multi-fluid micromixing, which has rarely been explored, typically represents a highly sought-after technique in on-chip biochemical and biomedical assays. Herein, we propose a novel micromixing approach utilizing induced-charge electroosmosis (ICEO) to implement multicomplex mixing between parallel streams. The variations of ICEO microvortices above a sinusoidally shaped floating electrode (SSFE) are first investigated to better understand the microvortex development and the resultant mixing process within a confined channel. On this basis, a mathematical model of the vortex index is newly developed to predict the mixing degree along the microchannel. The negative exponential distribution obtained between the vortex index and mixing index demonstrates an efficient model to describe the mixing performance without solving the coupled diffusion and momentum equations. Specifically, sufficient mixing with a mixing index higher than 0.9 can be achieved when the vortex index exceeds 51, and the mixing efficiency reaches a plateau at an AC frequency close to 100 Hz. Further, a rectangle floating electrode (RFE) is deposited before SSFE to enhance the controlled sequence for three-fluid mixing. One side fluid can fully mix with the middle fluid with a mixing index of 0.623 above RFE in the first mixing stage and achieve entire-channel mixing with a mixing index of 0.983 above SSFE in the second mixing stage, thereby enabling on-demand sequential mixing. As a proof of concept, this work can provide a robust alternative technique for multi-objective issues and structural design related to mixers.

Abstract:Ionic field effect transistors provide flexible and fast control of electrokinetic transport in nanofluidics, but most research has focused on Newtonian fluids. In this study, we use the finite element method (FEM) in COMSOL Multiphysics to solve the electro-osmotic flow of power-law fluids in a nanochannel using the Poisson-Nernst-Planck (PNP) and Navier-Stokes (NS) equations. The changes in wall surface charge density, ion concentration, fluid velocity, axial potential, and ionic current in the nanochannel under different gate electrode voltages and power-law indices were studied. The results show that under the same gate electrode voltage, the change of the shear-thinning fluid is more obvious, and the difference between the shear-thickening fluid and the Newtonian fluid is relatively small. The shear-thinning fluid with a power-law index of 0.8 has a velocity about 10 times higher than the Newtonian fluid, and its ionic current is also much greater than the Newtonian fluid.
